In vitro induction of tetraploid ginger (Zingiber officinale Roscoe) and its pollen fertility and germinability were investigated. The growth of shoot tip cultures on agar MS medium containing 2.0 mg l−1 BA was greater than that of similar cultures in liquid MS medium with the same BA concentration. In liquid medium, the combinations of 0.5, 1.0, or 2.0 mg l−1 BA with 0.05 mg l−1 NAA tended to enhance the growth of the cultures. The efficiency of tetraploid induction was assessed by treating shoot tip explants on agar or in liquid MS medium containing 2.0 mg l−1 BA, 0.05 mg l−1 NAA, and 0.2% (w/v) colchicine for 4, 8, 12, and 14 days. The total number of tetraploids induced on solid medium was 18, but only five in liquid medium. On both media, the colchicine treatment for 8 days gave the maximum level of tetraploid induction. Therefore, it was found that the treatment of shoot tip explants on agar medium containing 2.0 mg l−1 BA, 0.05 mg l−1 NAA, and 0.2% (w/v) colchicine for 8 days was the most efficient way of inducing tetraploid ginger. Induced tetraploid strains, ‘4× Kintoki’, ‘4× Sanshu’, and ‘4× Philippine cebu 1’, had higher pollen fertility and germinability than the diploid counterparts, i.e., in the diploid strains, pollen fertility ranged from 0.3 to 6.2% and the germination rate from 0.0 to 0.1%, while in the tetraploid strains, pollen fertility ranged from 27.4 to 74.2% and the germination rate from 4.8 to 12.9%.  相似文献

Fresh rhizomes of 18 diploid and tetraploid genotypes of Indian ginger, including commercial cultivars and experimental genotypes, were assayed for gingerol content and antioxidant activity. The tetraploid ginger type was derived from the respective diploid ginger by shoot tip culture. [6]-Gingerol was identified as the major pungent phenolic compound in all samples (mean 1354.78 ± 470 μg/g), while [8]- (mean 180.94 ± 45 μg/g) and [10]-gingerol (mean 234 ± 79 μg/g) occurred in lower concentrations. The total gingerol content of the tetraploid type was much higher than that of the respective diploid type and especially the [10]-gingerol. Both diploid and tetraploid types of the cv. Meghalaya Local contained the highest level of gingerol. At both ploidy levels, the genotypes showed significant differences in phenols and antioxidant activities. Correlation of total phenolics with different antioxidant assay at both diploid and tetraploid level was linear and strong to moderate. For genotypes at both ploidy levels, antioxidant activity (BCO) was determined especially by [6]-gingerol while [8]- and [10]-gingerol did not affect BCO. These results demonstrated that in the tetraploid types of ginger, the gingerol content and antioxidant activity were higher than in the diploid ones.  相似文献

Leaf area estimation is an important biometrical observation one has to do for comparing plant growth in field and pot experiments. In this study, a leaf area estimation model was developed for ginger (Zingiber officinale Roscoe), using linear measurements of leaf length (L) and maximum width (W). Leaves from five ginger varieties (Varada, Rejatha, Mahima, Maran and Himachal) were used to develop the model in 2006–2007. The actual leaf area (LA) was measured with a leaf area meter (LI-3100, LI-COR, Lincoln, NE, USA) and taken as reference LA. The linear measurements were used to build linear (LA = a + b × L × W) and power models (LA = α × (L × W)β) for each variety, as the modeling among variety were not different from each other, data for all five varieties have been pooled and compared with earlier models by graphical procedures and statistical criteria such as Mean Square Error (MSE), Root Mean Square Error (RMSE) and Chi-square (χ2). The selected model was validated during 2007–2008. The validation data set was used to produce a validation model for each variety by re-estimating the model parameters to develop the estimation model and the models were compared for consistency. The predicted LA (PLA) was compared with observed LA (OLA) by graphical procedures and lack of agreement was evaluated by calculating the relative bias, estimated by the mean of differences (d) and the standard deviation (SD) of the differences. Normality test was carried out by Spearman's rank correlation coefficient (rs) and residuals were normally distributed. Finally, the proposed model for leaf area estimation of ginger is LA = −0.0146 + 0.6621 × L × W, R2 = 0.997. This model can be reliably used for estimating leaf area of ginger non-destructively. The same equation can be extrapolated to all varieties and land races of ginger as it is vegetatively propagated crop with narrow genetic variability.  相似文献

Zingiber officinale, Rosc., one of the major tropical spices in the world, belongs to the family Zingiberaceae. Bacterial contamination is a major constraint on the cryopreservation of in vitro shoot tip explants. The main objective of this study was to identify the nature of this bacterial contamination and its response to various antimicrobial agents (e.g. the antibiotics cephotaxime and streptomycin sulphate, or copper sulphate) for more effective control. The bacteria isolated from ginger plantlets were identified by amplification and sequencing of their 16S ribosomal DNA, followed by partial sequence analysis. Luteibacter yeojuensis was found in all contaminated cultures. L. yeojuensis is found mainly in soil and as a human pathogen. We believe this is the first report of L. yeojuensis contaminating in vitro cultures of ginger. Among the antimicrobial agents tested in the shoot multiplication medium [i.e. 1.0× Murashige and Skoog salts + 2.5 mg ll 6-benzylaminopurine + 3% (w/v) sucrose + 0.45% (w/v) ClarigarTM], 100 mg l1 cephotaxime was most effective at reducing bacterial growth. It also gave the maximum number of shoots per shoot bud explant compared to the same medium supplemented with streptomycin sulphate, or CuSO4, or control medium. No further bacterial contamination was observed when 8-week-old cultures were then subcultured on the same medium without added antibiotic or CuSO4.  相似文献

【目的】从核桃生殖生物学角度出发,研究雌花状态和不同授粉方式对新疆早实核桃花粉萌发及花粉管生长的影响,旨在了解核桃授粉受精过程的基本规律,以期为提高核桃坐果率及果实品质提供科学依据,对于核桃品种选育、品种改良以及生殖隔离研究等均具有重要意义。【方法】以乌什县主栽核桃品种‘新新2号’(Juglans regia‘Xinxin 2’)为试材,运用荧光显微技术研究核桃雌花状态附着花粉的能力和不同授粉方式(自然授粉、人工辅助自花授粉、人工辅助异花授粉)下花粉粒在柱头上的萌发及花柱中花粉管的生长状况。【结果】(1)各时期雌花柱头附着花粉数量之间存在极显著差异(P<0.01),显花期和始花期柱头不具备附着花粉的能力,V字期柱头附着花粉的数量明显低于小八字期、大八期字和翻卷期。小八字期和翻卷期的花粉附着力之间差异不显著(P>0.05),但花粉萌发率之间存在较大差异。(2)核桃不同授粉方式下,花粉在柱头上的萌发时间格局、花粉管生长延伸完成受精的时间格局存在极显著差异(P<0.01)。花粉在柱头上8~36 h完成萌发,其中人工辅助异花授粉所需时间最短(8 h),自然授粉所需时间最长(36h)。授粉后36 h,各授粉方式之间花粉萌发率差异不显著(P>0.05)。核桃花粉从萌发到完成受精至少需要10 d,其中人工辅助异花授粉所需时间最短。在授粉受精过程中,花粉管从柱头穿过引导组织所需时间最长,为76~108 h。授粉后240 h,人工辅助异花授粉花粉管长度占花柱长度的比例极显著高于人工辅助自花授粉(P<0.01),与自然授粉之间差异显著(P<0.05)。【结论】核桃大八字期柱头的花粉附着力最强,且可授性最强,为核桃最佳授粉期。人工辅助异花授粉比自然授粉、人工辅助自花授粉先形成受精核。  相似文献

The pollen germination and pollen tube growth among nine Pistacia genotypes was quantified in order to identify differences in the tolerance of pollen to temperature variations. The effect of temperature on in vitro pollen germination and pollen tube growth was investigated in Pistacia vera (Uygur, Atli, Kaska, Sengel, Kavak), P. atlantica, P. khinjuk, P. terebinthus and P. palaestina. When pollen was incubated in a germination medium for 24 h in darkness, distinct differences were observed in pollen germination and pollen tube growth at different temperatures.  相似文献

In order to optimise production of the edible flower buds produced by myoga (Zingiber mioga Roscoe) a greater understanding of the growth and development of the plant and factors influencing flower initiation and development are required. The vegetative growth phase in myoga was characterised by a distinct period of pseudostem development from the planted rhizome piece followed by an extended period of rhizome growth from both the base of pseudostems and the planted rhizome piece. The transition from pseudostem formation to the initiation of rhizome growth occurred at the same time as the dry weight of the planted rhizome piece ceased to decline. Flower bud initiation and development occurred over an extended period, beginning soon after the commencement of new rhizome growth and ending prior to foliage senescence. Flower buds were observed at the terminal meristem of first, second, third and fourth order rhizomes. Increasing temperatures stimulated both increased vegetative growth and flower bud initiation and development. Low flower bud yields recorded under conditions of low temperature were the result of reduced rate of floral initiation and not abortion of flower buds. Increased shading of plants grown under glasshouse conditions resulted in reduced flower bud yield and similarly the response resulted from decreased initiation rather than abortion.  相似文献

【目的】探讨培养基中的钙、镁、钾浓度及光照、温度对西番莲花粉体外萌发及花粉管生长的影响。【方法】设置钙、镁、钾及光照、温度梯度,测量在该梯度下的花粉萌发率及花粉管延伸长度。【结果】钙、镁、钾非西番莲花粉离体萌发的必需培养基成分,钙对西番莲花粉离体萌发及花粉管生长具有显著影响,在质量浓度为0~300 mg·L-1时,萌发率及花粉管长度均随浓度的增加而显著增加,在300~1200 mg·L-1时最佳,大于1200 mg·L-1时,萌发率及花粉管长度均显著降低。镁、钾对花粉萌发及花粉管生长均无显著影响。光照、温度对花粉萌发及花粉管生长均具有显著影响,黑暗条件下最佳,随着光照度的增加,萌发率及花粉管长度均显著降低;二次方程能较好地拟合温度对花粉萌发及花粉管生长的影响。【结论】培养基中钙浓度及光照、温度对西番莲花粉体外萌发及花粉管生长具有显著影响,镁、钾无显著影响,最适的C(aNO3)2·4H2O质量浓度为300~1200 mg·L-1,西番莲...  相似文献

为进一步研究活体条件下精胺对梨花粉萌发及花粉管生长的影响,以幸水、新雪、长-二十世纪为材料,采用荧光显微镜观测了不同浓度精胺处理的花粉萌发及花粉管生长情况。试验结果表明:较低浓度的精胺能促进花粉萌发,而超过一定浓度时则表现抑制作用,适宜于花粉萌发的精胺浓度是0-0.025mmol/L。精胺对花粉管生长的促进作用因品种而异,0-0.05mmol/L,精胺促进幸水、新雪花粉管生长的作用主要表现在花柱中上部;而到达基部的花粉管仍较少,与对照间差异不显著。在0-0.025mmol/L的适宜浓度下,精胺可促进长-二十世纪花粉管生长并到达花柱基部。不同浓度的精胺对幸水、新雪坐果影响不大;而适宜浓度的精胺可促进长-二十世纪坐果。  相似文献

Emergence of multiple pollen tubes from single pollen grains occurred both in vitro and in vivo in sponge gourd (Luffa cylindrica (L.) Roem). The frequency with which pollen grains produced multiple pollen tubes in vivo (7.2%) was lower than that under in vitro conditions (14.9%). In pollen grains germinated in vitro, the total length of the multiple pollen tubes was greater than that of single pollen tubes, but individual tubes among the multiple tubes did not reach the same length as single tubes. Moreover, the growth of the single pollen tubes continued for a longer period in vitro than that of the multiple tubes. Fluorescence microscopy showed that callose was present throughout the pollen tube wall except in the apical part of growing pollen tubes, and nuclei moved into the longest of the multiple tubes. Results of Fourier transform infrared microspectroscopy indicated that abnormal cell wall components (peaks at 800–1000 cm−1) were more frequent in multiple pollen tubes lacking nuclei, and the pectin content (1733 cm−1) in multiple pollen tubes was much lower than that in single pollen tubes. These findings suggested that there were significant differences in pollen tube growth and wall composition between single and multiple pollen tubes, and that multiple pollen tubes had much less opportunity than single pollen tubes to reach the embryo sac and achieve double fertilization.  相似文献

The primary objective of this study was to evaluate the effects of foliar boron and calcium application after harvest on the quantity and activity of pollen in the ‘Housui’ and ‘Wonwhang’ pears on a subsequent year. Pollen grains of the ‘Housui’ pear were cultured on germination medium, to which had been added boric acid (0, 25, 75, 100, 200, 300, 400, and 500 mg L−1) and calcium nitrate (0, 10, 25, 50, 100, 150, 200, 250, 300, 400, and 500 mg L−1). Boric acid, which was added to the germination media, exerted a significant stimulatory effect on both pollen germination and pollen tube growth, although pollen tube growth was inhibited at higher concentrations than 300 mg L−1. Calcium nitrate addition stimulated pollen germination, except at concentration of 500 mg L−1. However, pollen tube growth was significantly inhibited with increasing concentrations of calcium nitrate. In the orchard experiment, boron and calcium were sprayed at concentrations of 0, 100, 200, 500 or 1000 mg L−1 onto leaves after harvest, respectively. Boron and calcium content in the tissues as well as pollen production and growth were determined after these treatments. The foliar application of boron mainly resulted in an increase of boron concentration in buds. It also induced an increase in the weight of the anther and pollen in the following year. On the other hand, the foliar application of calcium resulted in an increase of calcium concentration mainly in the leaves, but pollen weight was decreased at high concentration treatment in the following year. The germination rate and tube growth of collected pollen were highest in the trees which had received boron treatment at a concentration of 200 mg L−1. In contrast, the germination rate and tube growth of collected pollen were decreased by calcium application at concentrations of 500 and 1000 mg L−1 without significant increase at lower concentrations. Consequently, the accumulation of boron in the developed buds of pear trees subjected to post-harvest foliar boron application generated positive effects on both the quantity and quality of pollen in the following year.  相似文献

选用自交不亲和性程度不同的2个甜樱桃品种莫利和拉宾斯为试材,分别在开花前、开花当天和开花后进行莫利自花、拉宾斯自花和莫利×拉宾斯异花授粉,并于授粉后12、24、48、72、96h切取花柱,用FAA固定,荧光染色后压片观察。结果表明,3个组合的花粉萌发率都以开花当天授粉的最佳,拉宾斯自交和莫利×拉宾斯的花粉管生长状况也以开花当天授粉的最佳,但莫利自交的花粉管生长状况却以花前第4天最佳,花后第4天次之,而当天授粉的最差。因此认为,甜樱桃自交不亲和程度与其雌蕊的发育状态密切相关,蕾期授粉和延迟授粉都能够在一定程度上克服甜樱桃的自交不亲和性,但蕾期授粉的作用更加明显。  相似文献

Synseeds of ginger (Zingiber officinale) were produced using aseptically proliferated 2-week old encapsulating explants (microshoots) upon complexation of 4% sodium alginate prepared in Murashige and Skoog (1962) medium (MS) and 100 mM calcium chloride. Conversion of synseeds into plantlets (conversion) was recorded as 66% and 53% on MS (3% sucrose) and on MS (3% sucrose) + 2.5 mg/l BA media, respectively. However, shoots/synseed were significantly higher on MS (3% sucrose) + 2.5 mg/l BA medium. For short-term storage of germplasm, sucrose-dehydrated synseeds were found better than air-dehydrated or fresh synseeds. Synseeds dehydrated in 0.25 M sucrose liquid medium for 16 h and stored in cryovials (with out medium) at 25 °C for 8 weeks and 12 weeks exhibited 53% and 13% conversion, respectively, on MS (3% sucrose) + 2.5 mg/l BA medium. Plantlets obtained from stored synseeds were hardened, established successfully ex vitro and were morphologically similar to each other as well as their mother plants. This synseed protocol could be useful for short-term storage and exchange of germplasm of ginger between national as well as international laboratories.  相似文献

Effect of polyamines on in vitro pollen germination at high temperatures in tomato (Lycopersicon esculentum Mill.) was investigated. Pollen germination and tube growth were significantly inhibited at 33°C and 35°C compared to those at 25°C. This inhibition was reversed by the addition of spermidine or spermine in the germination medium. Spermidine at 0.5 mM was slightly more effective than spermine at 0.05 mM. Spermidine at 0.05 and 0.5 mM and spermine at 0.05 mM slightly increased pollen germination rates at 25°C. Spermidine at 5 mM and spermine at 0.5 M were inhibitory to pollen germination, regardless of incubation temperatures. Spermidine also promoted germination of pollen grains incubated at 38°C for 1 and 3 h and then at 25°C for the rest of the 20 h incubation period. The effect was higher at 0.5 mM than at 0.05 mM. Treatment of spermidine to intact flowers 1 day before anthesis was also effective in ameliorating the high temperature inhibition of in vitro pollen germination on the polyamine-free medium. Here, the optimum concentration was 5 mM. These results demonstrate that polyamines can counteract the inhibitory effects of high temperature on pollen germination. They also suggest that the endogenous level of polyamines in germinating pollen grains is an important factor for the pollen germinability at high temperature.  相似文献

Due to the protogynous dichogamy of cherimoya and to the absence of proper pollinating vectors, hand-pollination with fresh pollen is a common practice for cherimoya commercial production. In order to optimize the process of hand-pollination, in this work we have studied the conservation of cherimoya pollen at −20, −80 and −196 °C for up to 3 months. In vitro pollen germination of fresh pollen was 57.1% and it was progressively reduced with conservation time at the three temperatures studied reaching a minimum after 3 months of storage of 10.4%, 14.2% and 13.6% at −20, −80 and −196 °C, respectively. Differences in germination among temperatures were only significant during the first 2 weeks of storage. Field pollinations with pollen stored for up to 3 months at the three temperatures show no yield differences compared to pollinations performed with fresh pollen. The results indicate that pollen collected and stored at sub-zero temperatures at the beginning of the cherimoya blooming season can be used along the whole blooming season avoiding the need of collecting fresh pollen daily.  相似文献

用荧光显微镜观察了G蛋白调节剂处理柱头后对梨花粉萌发和花粉管伸长的影响,结果表明:1)G蛋白激活剂霍乱毒素(CTX)处理柱头后进行自花授粉,授粉后24h和48h,可明显促进自花花粉的萌发;而G蛋白抑制剂百日咳毒素(PTX)处理柱头后异花授粉则抑制异花花粉萌发。2)CTX处理柱头后自花授粉对自花花粉管的生长具有促进作用;而PTX处理柱头后异花授粉,表现出对异花花粉管生长的抑制作用。  相似文献

This study was conducted at the Seongju Fruit Vegetable Experiment Station for 2 years, 2002–2003 to investigate the effect of pollination methods on development and sugar content of oriental melon fruits. Oriental melon fruit was pollinated by honeybees (Apis indica) and bumblebees (Bombus ignitus) and fruit setting growth regulators was used as a control treatment. Fruits pollinated by honeybees and bumblebees has lesser length and width of fruit compared to the control. Pollinated by bumblebees increased hardness and soluble solids of fruits by 27% and 4–5% and that of honeybees increased hardness and soluble solids of fruits by 12% and 5–10% compared to the control, respectively. Fermented fruit ratio (%) of oriental melon fruit pollinated by honeybees, bumblebees, and growth regulator was 6.7%, 9.1%, and 28.1%, respectively. Glucose, fructose and sucrose contents of fruits by HPLC (high performance liquid chromatography) were very low throughout the ripening stage, but abruptly increased at harvesting stage. In fructose content of fruits, pollination of bumblebees and honeybees was higher by 9% and by 13% than those of control, respectively. The amount of total sugar content in fully ripen fruits of oriental melon was not different between the treatments (P > 0.05). Total sugar content (%) of fruits pollinated by bumblebees was slowly decreased until 15 days after storage compared to that of other treatment. For the low fermented fruit ratio (%) and slowly reduced total sugar content at storage of fruits, the pollination of bees was more useful than fruit setting growth regulators in early cultivation of oriental melon under plastic houses. The economic analysis according to the pollination method will be needed in the near future.  相似文献

【目的】阐明温度对枇杷花粉原位萌发、花粉管生长和胚囊寿命的影响,明确枇杷花粉萌发、花粉管生长的最适温度,为枇杷花期管理及生态区划提供新的科学依据。【方法】以‘大五星’为试材,将其离体花枝置于不同温度的恒温箱中进行培养,用‘龙泉5号’进行异花授粉,授粉后不同时间段分别采集花朵,采用荧光显微观察方法观察统计花粉萌发和花粉管生长和胚囊活力情况。【结果】在10.0、15.0和20.0℃的温度条件下花粉萌发率、被花粉管生长穿过的花柱比率和有花粉管的胚珠比率显著高于其他温度,当温度为5.0℃时花粉的萌发率仅为10%,温度升高至25.0、30.0℃时,萌发率又分别显著下降至65%和45%。当温度为5.0、30.0℃时,在授粉后120 h均未发现有花粉管穿过花柱生长至胚珠。【结论】温度对枇杷花粉萌发、花粉管生长和胚囊寿命有显著影响,枇杷花粉萌发的最适温度为19.9℃,花粉管最适宜的生长温度为18.0℃。  相似文献

A field experiment was conducted during 2005 and 2006 growing seasons at Ras Munif (32°22′N, 35°45′E), Ajloun, Jordan, to evaluate different pollination treatments and their effects on fruit set and quality of loquat fruit grown under Mediterranean conditions. Open, supplemental, rain, and covered pollination treatments were imposed on the selected trees. Results showed that under supplemental and open-pollination, extremely higher fruit set was obtained compared with rain and covered treatments. In spite of a heavy crop load, fruits obtained under open and supplemental pollination were also heavier and larger, this could be partly due to a significant increase in seeds and flesh weight. Sugar content was good for both open and supplemental pollination. Wind plays little or no role in loquat pollination process. Loquat flowers attracted very limited number of bee visitors, A. mellifera, Anthophora albigena and Xylocopa violocea of family Apidae and Halictus quadricinctus of family Halictidae were recorded the only loquat flower visitors. They showed a variation in their visiting time but loquat flowers attracted the highest rate and number during the mid of the day.  相似文献

Stands of summer cauliflower were grown within polyethylene-covered tunnels along which a temperature gradient was imposed. Two tunnels were maintained at either normal or elevated CO2 concentrations. At the last harvest (88 days from transplanting) no interaction between CO2 and temperature on total biomass was detected. The total dry weight of plants grown at 531 μmol mol−1 CO2 was 34% greater than those grown at 328 μmol mol−1 CO2, whereas a 1 °C rise reduced dry weight by 6%. From serial harvests the radiation conversion coefficient was 2.01 g MJ−1 and 1.42 g MJ−1 at 531 μmol mol−1 CO2and 328 μmol mol−1 CO2, respectively, but was not greatly affected by differences in temperature. No effect of either CO2 or temperature on the canopy light extinction coefficient was detected. The rate of progress towards curd initiation increased to a maximum at 15.5 °C, and declined thereafter. Provided the effect of temperature was accounted for, CO2 enrichment did not affect the time of curd initiation. From serial harvests after curd initiation, the logarithm of curd weight or diameter were negative linear functions of mean temperature from initiation. Increases in curd weight and diameter at 531 compared with 328 μmol mol−1 CO2 were greater at warmer temperatures (27% at 13 °C compared with 47% at 15 °C, 57 days after initiation). Effects of CO2 on curd diameter were less than those on curd dry weight because the curd dry matter content was greater at 531 compared with 328 μmol mol−1 CO2. Thus, the effects of elevated CO2 concentrations on fresh weight based yield parameters of cauliflower were less than the increase in total dry matter production.  相似文献
