We evaluated the efficacy of buparvaquone in eliminating Babesia equi of European origin in carrier horses and in experimentally infected splenectomized ponies. When administered at the rate of 2.5 mg/kg of body weight, IM, 4 times at 96-hour intervals, buparvaquone was effective in eliminating B equi carrier infection in 1 horse. Such results could not be repeated at the same dosage or at 3.5 or 5 mg/kg, IM. Buparvaquone given at the rate of 4 to 6 mg/kg IV and/or IM was therapeutically effective in 4 of 5 acute B equi infections in splenectomized ponies. The treated ponies became carriers.  相似文献

We evaluated the efficacy of buparvaquone in eliminating infection with Babesia equi of European origin in carrier horses and in splenectomized horses with experimentally induced acute infection. When administered at the rate of 5 mg/kg of body weight, IV, 4 times at 48-hour intervals, buparvaquone prompted rapid abatement of parasitemia. However, secondary and tertiary recrudescent parasitemias invariably returned with establishment of the carrier state. Buparvaquone, at the dosage evaluated, had transitory therapeutic efficacy against acute B equi infection in splenectomized horses, but was unable alone to clear carrier infection.  相似文献

The controlled test method was used to evaluate the antiparasitic efficacy of IM inoculated 22,23-dihydroavermectin B1 (ivermectin) against gastrointestinal parasites of horses (ponies). Parasite infections were naturally acquired in southern Louisiana. Dose levels of the drug tested were 0.2 mg/kg, 0.3 mg/kg, and 0.5 mg/kg. Ivermectin at all dose levels tested had an efficacy greater than 97% (P less than 0.05) against Gasterophilus intestinalis larvae, Trichostrongylus axei, Oxyuris equi larvae, Strongylus vulgaris, S edentatus, 15 species of small strongyles, and small strongyle larvae. Ponies were less uniformly infected with Habronema sp larvae, G nasalis larvae, Parascaris equorum, O equi adults, Anoplocephala perfoliata, S equinus, and 11 small strongyle species, and statistical analysis was not possible to do. However, observations indicate that the drug was also highly effective against these species. There were no gross or clinical reactions observed in treated animals. Dissections of the injection sites revealed spindle-shaped lesions, 3 to 5 cm long, in a few ponies in all treatment groups, including those given the placebo injection.  相似文献

The efficacy of 3 antibiotic formulations was measured in the treatment of artificially induced anaplasmosis in the early stages of an ascending parasitemia (1% to 4%) in 23 splenectomized calves. Group 1, consisting of 5 calves, served as nontreated controls. Four calves (group 2) were treated 1 time with 10 mg of oxytetracycline (T-50)/kg of body weight IM; 5 calves (group 3) were treated 3 times with 10 mg of T-50/kg IM; 5 calves (group 4) were treated 1 time with 20 mg of an experimental oxytetracycline (T-200)/kg IM; and 4 calves (group 5) were treated 1 time with 10 mg of a synthetically derived antibacterial agent, doxycycline (D-100)/kg IM. All control calves died and 1 of 4 calves died that was treated 1 time with T-50. Other deaths did not occur. All treatments were effective in moderating the infective process, but T-50 given 3 times and T-200 given 1 time were markedly more effective than T-50 and D-100 given 1 time. There appeared to be little or no difference in therapeutic efficacy between T-50 and D-100 given 1 time and between T-50 given 3 times and T-200 given 1 time.  相似文献

A controlled test was performed to titrate the anthelmintic dosage of dienbendazole in 24 mixed-breed ponies naturally infected with Strongylus vulgaris, S edentatus, and small strongyle species, as determined by parasitic egg and larval counts in feces. Comparison of results of treatment was made among 3 dienbendazole dosages--2.5, 5, and 10 mg/kg of body weight--and a gum (excipient) mixture given by nasogastric intubation. All ponies were euthanatized and necropsied at 7 or 8 days after treatment. Trichostrongylus axei, Habronema muscae, S vulgaris, S edentatus, small strongyles, and Oxyuris equi were efficaciously eliminated in response to all doses of dienbendazole; Gasterophilus spp were not affected by any dose. There were not sufficient numbers of Draschia megastoma, Anoplocephala spp, or Parascaris equorum in the ponies to evaluate drug effect. Changes in the appearance of the intestinal lining were dose-dependent; in the ponies treated with 5 and 10 mg of dienbendazole/kg, the mucosa appeared clean and smooth, though in ponies given 2.5 mg/kg, it appeared clean, but was nodular and moderately reactive to embedded immature small strongyles. In the gum mixture-treated ponies, the large intestinal mucosa was inflamed, with edematous areas, in response to infections caused by large and small strongyles. A limited clinical titration was done in 12 ponies that were fecal culture negative for S vulgaris larvae, although other strongyles were detected. Two ponies in each of 6 groups were given the following dosages: 0 (gum mixture only), 0.5, 1, 2.5, and 5 mg of dienbendazole/kg. The effect of T-2 toxin on host resistance to acute and latent Babesia microti infections was evaluated in mice and was compared with the effects of the immunosuppressive drugs dexamethasone and cyclophosphamide. Mice with acute or latent B microti infection were treated with 2 mg of T-2 toxin/kg of body weight, 0.2 mg of dexamethasone/kg, or 30 mg of cyclophosphamide/kg daily for 5 days. Treatment with dexamethasone or cyclophosphamide caused significant (P less than 0.05) increases in Babesia parasitemia during acute infection and significantly (P less than 0.05) prolonged the duration of parasitemia during acute babesiosis. Treatment with T-2 toxin caused a transient significant (P less than 0.05) increase in Babesia parasitemia on day 10 after acute infection and numerical, though statistically nonsignificant, increases in the maximal level and duration of parasitemia during acute babesiosis. Significant (P less than 0.005) recrudescence of parasitemia was observed in the dexamethasone- and cyclophosphamide-treated mice with latent Babesia infection. Treatment with T-2 toxin did not cause recrudescence of parasitemia in mice with latent Babesia infection.  相似文献

The pharmacokinetics of gentamicin were determined in male and female Ictalurus punctatus weighing between 0.3 kg and 1.7 kg. Plasma gentamicin concentrations were measured by radioimmunoassay technique. In the 1st experiment, an intracardiac bolus dosage of gentamicin (1 mg/kg) was given to 10 channel catfish. Samples of blood were obtained (by cardiac puncture) immediately before gentamicin was given and at various times over a 24-hour period. The gentamicin half-life was 770 minutes (beta = 0.0009 +/- 0.0003 minute-1). The maintenance therapeutic IV dosage of gentamicin was calculated to be 1.6 mg/kg given at 33.2-hour intervals. In the 2nd experiment, an IM dosage of the drug (1 mg/kg) was given to 9 channel catfish, and samples of blood were obtained at various times over a 24-hour period. The gentamicin half-life was 770 minutes (beta = 0.0009 +/- 0.0002 minutes-1). The maintenance therapeutic IM dosage of gentamicin was calculated to be 3.5 mg/kg given at 33.2-hour intervals. The IM bioavailability of gentamicin in channel catfish was estimated to be 60%. Other pharmacokinetic values were also determined. It was concluded that the therapeutic regimen of choice for gentamicin in channel catfish was 3.5 mg/kg given IM with 33-hour intervals between doses.  相似文献

Twenty ponies were allotted to 4 groups of 5 ponies each, and oxibendazole was orally administered at dose levels of 5, 10, 15, and 20 mg/kg. In the 3 groups of ponies given the largest doses, efficacy against 3 species of Strongylus was between 92 and 100% and that against small strongylids of the genera Cyathostomum, Cylicocyclus, Cylicodontophorus, and Cylicostephanus was more than 99%. All adults and 95 to 100% of larvae of the pinworm Oxyuris equi were removed. In the group of ponies given 5 mg/kg, 86 to 100% of the large strongylids and 84 to 100% of the small strongylids were removed, as were all larval and adult pinworms. Trichostrongylus axei was found only in 4 of the 5 ponies given 5 mg/kg; results were encouraging, but not consistent. Almost all of the Parascaris equorum were found in this group of ponies; no anthelmintic activity was detected at this dose level. Oxibendazole removed approximately 99% of small strongylid 4th-stage larvae. No efficacy against the larval stages of Gasterophilus intestinalis and against Habronema spp and Setaria equina was observed.  相似文献

Equine piroplasmosis (EP) has not been considered indigenous in The Netherlands. However, following the detection of an apparently indigenous subclinical Babesia caballi infection in a horse on Schouwen-Duiveland (an island in the Zeeland Province), a survey was undertaken between May and September 2010 to assess the prevalence of the causative agents of EP in the South-West of The Netherlands. Blood samples from 300 randomly selected horses were tested for specific antibodies against Theileria equi and B. caballi using an indirect fluorescence antibody test (IFAT), and for parasite DNA using a specific polymerase chain reaction combined with reverse line blotting (PCR-RLB). Twelve of the horses (4%) were seropositive for EP. Of these, nine (75%) were positive (titre?1:160) for B. caballi alone and three (25%) were also positive for T. equi. PCR-RLB detected T. equi DNA in five horses (1.6%), two of which were seronegative. Four (1.3%) of the positive horses (three positive for T. equi and one for both B. caballi and T. equi) were considered truly indigenous. During the study, two indigenous ponies from a farm situated outside the sampling area were diagnosed with acute clinical piroplasmosis characterized by severe anaemia and pyrexia. Blood smears showed T. equi - like inclusions in red blood cells, and T. equi infection was confirmed in both ponies by PCR-RLB. The initial subclinical B. caballi infection, the survey results and the two acute clinical EP cases confirmed the autochthonous transmission of B. caballi and T. equi infections in The Netherlands.  相似文献

Tissue chambers, implanted subcutaneously in the neck in six ponies, were inoculated with Streptococcus equi subsp. zooepidemicus in order to determine the clinical efficacy of prophylactic administration of trimethoprim/sulfadiazine (TMP/SDZ) against this infection. The TMP/SDZ treatment consisted of one intravenous (i.v.) injection of 5 mg/kg TMP and 25 mg/kg SDZ and the same dose of TMP/SDZ per os (p.o.), both given 3 h before inoculation. The oral dose was then repeated every 12 h for 5 days. TMP/SDZ concentrations in tissue chamber fluid (TCF) were above 10 times MIC at the moment of inoculation, and they were maintained at this level or higher throughout the duration of treatment. Trimethoprim/sulfadiazine treatment resulted in a marked reduction of viable bacteria in the tissue chamber but did not eliminate the infection, resulting in abscessation from day 19 onwards in all six ponies. This shows that, even when TCF is not yet purulent, TMP/SDZ is unable to eliminate the streptococci. Therefore, TMP/SDZ should not be the antimicrobial treatment of choice in infections in secluded sites in horses.  相似文献

Tissue chambers, implanted subcutaneously on both sides of the neck in eight ponies, were inoculated with Streptococcus equi subsp. zooepidemicus in order to compare the clinical efficacy of trimethoprim/sulfadiazine (TMP/SDZ) and penicillin G treatment in a purulent infection. The TMP/SDZ treatment consisted of one intravenous (i.v.) injection of 5 mg/kg TMP and 25 mg/kg SDZ and the same dose of TMP/SDZ per os (p.o.), both given 20 h after inoculation. The oral dose was then repeated every 12 h for 21 days. The penicillin treatment consisted of one i.v. injection of 20 000 IU/kg sodium penicillin G and intramuscular (i.m.) injection of 20 000 IU/kg procaine penicillin G, both given 20 h after infection. The i.m. dose was then repeated every 24 h for 21 days. Eight ponies, each with two tissue chambers, were used in a cross over design; in the first experiment the left tissue chamber (TC) was infected and in the second experiment the right. TMP/SDZ treatment resulted in a limited reduction of viable bacteria in the TC but did not eliminate the infection, resulting in abscessation in 10-42 days in all eight ponies. However, penicillin treatment eliminated the streptococci in seven of eight ponies, and only one pony suffered abscessation on day 10. This constitutes a significantly better efficacy of the penicillin treatment in this model. The most probable cause of the failure of TMP/SDZ to eliminate the streptococci is inhibition of the action of TMP/SDZ in the purulent TCF. Therefore, TMP/SDZ should not be used to treat purulent infections in secluded sites in horses.  相似文献

Acute experimentally induced aflatoxicosis in the weanling pony   总被引:1,自引:0,他引:1  
Nineteen weanling ponies and 1 adult pony were given a single oral dose of aflatoxin B1 (AFB1). Dosages were: 0, 0.5, 1, 2, 4, 5, 6, and 7.4 mg of AFB1/kg of body weight. Vital signs were monitored, and whole blood and serum collected for analysis of serum enzymes, prothrombin time, blood cell counts, and serum urea nitrogen. Ponies that died were examined for gross lesions, and tissues were collected for histopathologic examination and analysis of AFB1 and AFM1 residues. Two of the 4 ponies given the 2 mg/kg dose and all ponies given the larger dosages died within 76 hours. Clinical signs included increased rectal temperature, faster heart and respiratory rates, abdominal straining, bloody feces, and tetanic convulsions. At necropsy, ponies that died of acute aflatoxicosis showed visceral petechiae and hepatic focal lesions. Histopathologic changes included severe hepatic necrosis, vacuolation, and bile duct hyperplasia. Aflatoxins B1 and M1 were recovered from liver, kidney, skeletal muscle, and gastrointestinal contents. One other pony given the 2 mg/kg dose died 32 days after dosing, and 1 control pony died after 70 days. Continuous elevations in prothrombin time and serum aspartate aminotransferase, alanine aminotransferase, and gamma-glutamyl transpeptidase levels were observed in ponies dosed at 4 mg/kg or more. Significant (P less than 0.05) elevations in these values, which peaked 2 to 3 days after dosing, were seen in ponies given the 2 mg/kg dose. This group also had significant increases over controls in PCV and hemoglobin concentration 5 days after dosing.  相似文献

Ototoxicosis was evaluated in 6 healthy ponies given 5 mg of gentamicin/kg of body weight, q 8 h, IM. Ponies 1, 2, and 3 were dosed for 7 days and ponies 4, 5, and 6 were dosed for 14 days. Serum peak and trough concentrations of gentamicin were measured by radioimmunoassay at regular intervals. Brain stem auditory-evoked responses were recorded every 5 days up to 60 days after the first dose to monitor auditory function. Although serum gentamicin concentrations were within or above the accepted clinical therapeutic range, loss of auditory function was not observed at the frequency range (1 to 4 kHz) tested. Serum chemical values remained within the accepted clinical range and no evidence of nephrotoxicosis was observed. Seemingly, gentamicin given IM to healthy ponies was safe and had minimal risk of side effects.  相似文献

The efficacy of closantel against experimentally induced Fascioloides magna infection in sheep was studied. In each of 3 experiments, closantel was administered 8 weeks after the sheep were given (oral inoculation) 100 metacercariae of F magna. In the 1st experiment, closantel was given orally to 5 groups of 6 sheep each at dosages of 0 (nontreated control), 5, 7.5, 10, and 15 mg/kg of body weight. In the 2nd and 3rd experiments, groups of 10 or 12 sheep were treated to confirm the efficacy of the previously determined optimal dosage of 15 mg/kg. An additional group of sheep (n = 10) was used in the 3rd experiment to evaluate the efficacy of closantel given IM at a dosage of 7.5 mg/kg. Closantel given orally at a dosage level of 15 mg/kg was highly effective (94.6% to 97.7%) in reducing F magna burdens. Also, pathologic scores associated with the F magna infection were reduced by 81.3% to 92.6% in sheep given this dosage of closantel. Efficacy of the IM administered dosage of 7.5 mg of drug/kg was equivalent to that of the 15 mg/kg oral dosage. Other than mild, transient lameness of the limbs which were injected with the drug (group 10), side effects were not observed.  相似文献

Halofuginone lactate, given once orally at a dosage rate of 1,2 mg/kg body mass on the 1st, 3rd or 5th days of fever, resulted in the recovery of only 1 out of 5 splenectomized cattle. Three splenectomized animals, treated on the 1st as well as the 4th day of fever, recovered and were then carriers. Six untreated controls all died. The potential value of a chemotherapeutic agent for Theileria parva lawrencei infections in South Africa is discussed.  相似文献

Eight ponies infected with Babesia equi were investigated for their serological response to B. equi schizont and piroplasm antigen with the indirect fluorescent antibody test (IFAT) and complement fixation test (CFT). Piroplasm antigen was prepared from an infected splenectomized pony, while schizont antigen was produced from cultured lymphoid cells which contained B. equi macroschizonts. The IFAT detected a rise in antibody titres to schizont antigen as well as to piroplasm antigen, but differences were obtained in the duration of antibody detection. Significant antibody titres to piroplasm antigen were detectable for a longer period post infection than to schizont antigen. The complement fixation test was not effective in detecting specific antibodies to schizont antigen in contrast to piroplasm antigen. The schizont antibody titres were in general extremely low and not detectable in 3 horses. Neither test showed any serological cross-reaction with B. caballi and B. bigemina antiserum using schizont antigen.  相似文献

Anaplasmosis, caused by the tick-borne rickettsia, Anaplasma marginale, is an economically important disease of cattle in the United States and worldwide. Cattle that recover from acute infection become carriers in which low or microscopically undetectable A. marginale rickettsemia persists. Tetracycline antimicrobials are currently the only drug used in the US for treatment of acute anaplasmosis. There are currently no drugs specifically licensed for elimination of persistent infections. This study tested the efficacy of three oxytetracycline treatment regimens to clear A. marginale from cattle that were persistently infected. Forty Angus x Simmental steers, aged 6-12 months were experimentally infected with A. marginale. After the steers recovered from acute infection, seroconverted, and were confirmed infected using nested PCR followed by DNA hybridization, the carrier status of each animal was ascertained by sub-inoculation of blood into a separate, splenectomized Holstein calf. The steers were then blocked by bodyweight and randomly assigned as follows to four treatment groups: Treatment A, 300 mg/ml solution of oxytetracycline (Tetradure LA-300, Merial Canada Inc.) administered at 30 mg/kg, by intramuscular (i.m.) injection on day 0; Treatment B, the same 300 mg/ml solution of oxytetracycline administered at 30 mg/kg, i.m. on day 0 and again on day 5; Treatment C, a 200 mg/ml solution of oxytetracycline (Liquamycin LA-200, Pfizer Animal Health) administered at 22 mg/kg, intravenously (i.v.), q 24 h for 5 days (a treatment dose that corresponds with current Office International des Epizooties (OIE) recommendations for treatment prior to export). The fourth group consisted of untreated infected control cattle. All steers were still nested PCR and cELISA positive at 60 days after treatment. Infection was confirmed by subinoculation of blood into a splenectomized Holstein calf. These results demonstrated that the treatment regimens tested failed to clear A. marginale infections in carrier cattle.  相似文献

A controlled test was done in 30 naturally infected ponies to evaluate the antiparasitic activity of the dienbendazole analog VET 220-S given alone or with trichlorfon (TCF) by nasogastric intubation. Six ponies were nontreated; 6 were given VET 220-S (5.0 mg/kg); 6 were given TCF (40 mg/kg); 6 were given VET 220-S (2.5 mg/kg) and TCF (40 mg/kg); and 6 were given VET 220-S (5.0 mg/kg) and TCF (40 mg/kg). All ponies were euthanatized and necropsied 7 or 8 days after treatment. Draschia megastoma, Oxyuris equi, Strongylus vulgaris, S edentatus, and small strongyles were removed efficaciously by all doses of VET 220-S. Habronema muscae and microfilariae of Onchocerca cervicalis were not removed by VET 220-S or TCF. Gasterophilus intestinalis was 97.9% removed by TCF. Pregnant mares in all groups were not adversely affected by treatment, except for 1 mare that had diarrhea after TCF treatment. Parasite eggs per gram and larval culture data agreed with necropsy data.  相似文献

Gentamicin was administered parenterally for 6 days to 43 dogs with urinary tract infections. The daily dosage of 6.6 mg/kg (3 mg/lb) was divided into equal parts and given IM or SC at 8-hour intervals. Dogs selected for treatment with gentamicin had urinary infections that had not responded to treatment with other antimicrobial agents or had bacterial isolates from urine that were resistant to several antimicrobial agents on in vitro susceptibility tests. Response to treatment, defined as negative urine culture on the last day of therapy or 4 to 14 days after completion of the therapeutic course, included 20 of 22 (91%) infections caused by Escherichia coli, 8 of 9 (89%) infections caused by Kebsiella pneumoniae, 6 of 7 (86%) infections caused by Proteus spp, and 6 of 7 infections caused by Pseudomonas spp. These four species comprised 84% of the bacteria isolated from the dogs in this study.  相似文献

The analgesic and behavioral effects of butorphanol (0.22 mg/kg), flunixin (2.2 mg/kg), levorphanol (0.033 mg/kg), morphine (0.66 mg/kg), and xylazine (2.2 mg/kg), given IM were observed in 8 ponies. These ponies were instrumented to measure response objectively to painful superficial and visceral stimuli. Effects on the cardiopulmonary system and rectal temperature also were evaluated in 6 of these ponies. Observations were conducted before drug injection (base-line values) and after injection at 30, 60, 120, 180, and 240 minutes. Xylazine provided the highest pain threshold for the first 60 minutes and a sedative effect for 105 minutes. The effects for superficial pain and visceral pain persisted 3 hours and 4 hours, respectively. Morphine produced good analgesia for superficial pain (30 minutes), whereas butorphanol provided good effect for visceral pain (4 hours). A slight degree of analgesia for visceral pain was obtained after morphine (1 hour) and levorphanol (4 hours); flunixin did not induce analgesia. Butorphanol, levorphanol, and morphine stimulated motor activity. Behavioral effects did not occur after flunixin was given. Xylazine decreased systolic, diastolic, and mean blood pressures. Marked increases in these pressures, heart rate, and respiratory rate were observed after morphine was given. Changes of central venous pressure, rectal temperature, and blood gas values remained within base-line limits after both drugs were given. Butorphanol increased heart rates for 1 hour; flunixin and levorphanol did not alter any of the above values.  相似文献
