壳聚糖絮凝法回收鱼糜漂洗水中水溶性蛋白质的工艺研究   总被引:1,自引:0,他引:1  
采用壳聚糖作为絮凝剂进行鱼糜漂洗水中水溶性蛋白质的回收试验。分别通过体系的pH、回收温度和壳聚糖絮凝剂加入量的单因素试验及这3个因素的正交试验,研究其对蛋白质回收率、透光率和化学耗氧量(COD)去除率的影响,得出壳聚糖絮凝法回收40mL已知蛋白质质量浓度(9.0mg·mL-^1)的鱼糜漂洗水中水溶性蛋白质的最佳工艺条件为体系pH8.0,回收温度35℃,1%壳聚糖添加量为1.5mL。该工艺获得的蛋白质回收率为73.17%,透光率87.50%,COD去除率47.20%。  相似文献

本文研究了TG-K、TG-B、TG-AK三种酶制剂在不同的添加量、凝胶化时间下对冷冻蛇鲻鱼糜凝胶强度的影响。结果表明:三种酶制剂都有增强冷冻蛇鲻鱼糜凝胶强度的作用,以 0.15%的 TG-AK酶制剂,30℃时凝胶化 30分钟凝胶强度增强效果最好,此方案应用于鱼丸的生产,产品的凝胶强度弹性明显增强。  相似文献

为了综合利用小杂鱼资源,以冻杂鱼浆为原料,探讨了谷氨酰胺转氨酶(简称TGase)的添加量、作用时间、作用温度对鱼糜制品的弹性和凝聚性的影响。单因素试验和L9(34)正交试验结果表明,TGase的最佳工艺参数水平组合为TGae的用量为0.10%,TGase的作用温度为35℃,作用时间为1.5 h。  相似文献

研究了添加不同质量分数的水溶性壳聚糖对鱼糜制品冷藏保鲜效果的影响。以感官评分、菌落总数、挥发性盐基氮(TVB-N)、硫代巴比妥酸(TBA)和pH为指标,考察鱼糜制品在4℃冷藏过程中的品质变化。结果表明,水溶性壳聚糖能够明显抑制细菌的生长与繁殖,阻止感官品质的下降,延缓TVB-N和TBA的上升。其中,添加2.0%的水溶性壳聚糖保鲜效果最佳,与空白组相比,能将鱼糜制品的冷藏保鲜期从9 d延长至13~14 d。  相似文献

To evaluate the quality of frozen surimi, the ATPase assay system was applied to pH stat. Surimi homogenate was prepared without washing procedures as a material for the measurement. Among the possible compounds frozen surimi contains, Mg ion slightly inhibited Ca-ATPase activity, while sugars or polyphosphate did not. Thus, the surimi homogenate was directly used for ATPase assay. The quality of surimi samples with various commercial grades were analyzed by measuring Ca-ATPase activity and protein content to obtain ATPase total activity. SA grade surimi showed high total activity, and was clearly distinguished from the second grade ones by the total Ca-ATPase activity. The specific Ca-ATPase activity contributed to the total activity more than the protein content. These results demonstrated that employment of surimi homogenate and the pH stat method made the total activity measurement of surimi easy and rapid.  相似文献

为探讨超高压改善低盐鲢(Hypophthalmichthys molitrix)鱼糜凝胶品质的机制,利用低场核磁共振(low field nuclear magnetic resonance,LF-NMR)、差示扫描量热法(differential scanning calorimeter,DSC)、傅里叶红外光谱(fourier transform infrared spectroscopy,FT-IR)以及扫描电镜分析比较了超高压低盐鱼糜凝胶[300 MPa,1.5%氯化钠(NaCl)]与常压低盐鱼糜凝胶(0.1 MPa,1.5%NaCl)以及常压普通鱼糜凝胶(0.1 MPa,2.5%NaCl)在水分状态和微观结构上的差异。DSC结果表明超高压低盐鱼糜凝胶可冻结水的冰点降低,结合水含量(17.58%)较低盐对照组(10.89%)显著提高;LF-NMR表明超高压低盐鱼糜凝胶弛豫时间T21、T23和T24左移,不易流动水的含量(76.65%)较低盐对照组(67.29%)提高了9.39%;超高压处理能使低盐鱼糜凝胶形成光滑、连续、均匀的三维网络结构。因此,超高压处理(300 MPa,10 min)能够提高低盐鲢鱼糜凝胶结合水含量、改善微观结构。  相似文献

以海水鱼糜(包括狭鳕、带鱼、鲷和混合鱼糜)以及淡水鱼糜(包括鲢和草鱼鱼糜)为研究对象,应用傅立叶近红外光谱技术结合化学计量学对鱼糜进行定性和定量分析。用聚类分析和线性判别的方法建立海水和淡水鱼糜的定性鉴别模型,并应用偏最小二乘法(PLS)分别建立海水鱼糜水分含量和蛋白质含量的近红外定量分析模型。结果表明,海水和淡水鱼糜的定性鉴别模型预测率达100%;建立的近红外定量分析模型中,海水鱼糜的水分和蛋白质的模型的相关系数分别达0.98和0.96以上,模型能较为准确、快速地预测定海水鱼糜中水分和蛋白质含量,有很大的潜力可应用于实际生产。  相似文献

研究以金线鱼和长蛇鲻的混合鱼糜生产下脚料为材料,利用钾法提取鱼油。结果表明,钾法提取鱼油的工艺参数为水解pH为7.5,水解温度70℃,水解时间55min,硝酸钾用量4.5%,盐析时间30min,在该条件下鱼油的提取率达到64.5%。鱼油的理化性质达到了SC/T3502-2000标准粗鱼油的二级要求。鱼油中饱和脂肪酸的含量较高,占总量的48.59%;单不饱和脂肪酸和多不饱和脂肪酸的含量相当,分别为24.48%和24.49%,多不饱和脂肪酸主要是DHA和EPA,含量分别为11.18%和5.17%。  相似文献

ABSTRACT:   This study examined the thermal gelation characteristics of myofibrillar protein (MP) and salt-ground horse mackerel surimi that had been mixed with hydroxypropylmethylcellulose (HPMC). Analysis, using thermal scanning rigidity monitor and differential scanning calorimetry, demonstrated that thermal gelation begins in the thermo-reversible HPMC gel used in this study at approximately 60°C. In the rescanning test, first scanning diagrams showed similar pattern, but second scanning diagrams were rather diverse between MP and MP/HPMC mixture. The addition of HPMC increased rigidity of salt-ground surimi and modified the thermal gelation during the heating process, suggesting that certain enhancing effect of gelation occurred when the mixture of MP and HPMC was heated. In the annealing test, the addition of HPMC lowered the breaking force, but increased the gel strength of surimi due to a substantial increase in deformation. Although the strong cross-linking between the two components might not occur during the gelling process, HPMC gel formed at high temperature occupies some space in thermal gel by interacting with proteins and resulted in a combinative gel with a higher rigidity and gel strength at higher temperature.  相似文献

ABSTRACT:   Changes in purine-related compounds of tilapia surimi product during processing were investigated. The washing step could result in about 60% decrease of total purine content in tilapia mince during processing. The main released purine substance was inosine monophosphate. The major reducing effect was conducted in the first 10 min during washing. No significant changes were observed after washing for 20 and 30 min. The lowest total purine content of tilapia surimi product was obtained with repeating the washing step twice. Thus, this procedure could reduce the purine content of tilapia mince from a high purine content level to a middle level. The gel strength of tilapia surimi product increased with increasing washing duration within 30 min. However, tilapia surimi product with a middle purine content and acceptable gel strength might be produced by washing twice in 10 min during processing.  相似文献

为了研究马铃薯基乳液(potato based emulsion, PE)对鱼糜凝胶品质的影响, 本研究将玉米油(corn oil,CO)或PE按不同添加量(0%~14%)与鱼糜混合,以鱼糜凝胶质构、水分、色泽、分子间相互作用、流变特性和微观结构为考察指标,探究PE作为脂肪替代物对鱼糜凝胶品质的影响及机理。 结果显示,在相同的油添加量下,与CO相比,PE能避免油滴对鱼糜凝胶质构特性和持水能力产生的劣化效果,使样品表现出更高的弹性模量和粘度,其中,6%油含量PE样品的硬度从1 073 g(0%油含量样品)提升到1 159 g,而6%油含量CO样品的硬度降低至837 g;另外,横向弛豫时间结果表明,PE抑制了不易流动水向由水的转变,使更多的不易流动水被束缚,这可能增强了光线漫反射,从而使PE样品白度更高;微观结构和分子间作用力测定结果表明PE样品中油滴分布更均匀,氢键、疏水相互作用和二硫交联更强,凝胶网络因淀粉颗粒的填充效应而更致密;十二烷基硫酸钠-聚丙烯酰胺凝胶电泳结果表明 ,PE组中油滴对鱼糜蛋白交联的干预程度更小。研究表明,PE可作为脂肪替代物提升鱼糜凝胶制品品质。本研究可为淀粉基脂肪替代物的开发及其在鱼糜凝胶产品中的应用提供理论基础。  相似文献

采用同轴探针方法分别测定了30~80℃,频率320~2960MHz内罗非鱼鱼糜和阿拉斯加狭鳕鱼糜的介电常数和介电损失率。频率一定时,两种鱼糜的介电常数随温度升高而降低;在相同的温度下,频率增加,鱼糜的介电常数反而减小。频率升高使两种鱼糜的介电损失率先减少后增加,分别在890MHz(罗非鱼鱼糜)和1350MHz(狭鳕鱼糜)出现拐点。温度与介电损失率的相关性与频率有关,拐点频率之前,温度与介电损失率成正相关性,而大于拐点频率时,则成负相关性。本研究提出了两种鱼糜的介电常数和介电损大率关于温度和频率的预测方程,预测值与实验值具有良好的一致性。两种鱼糜的穿透深度在915MHz随温度升高而减小,在2450MHz时则正好相反。鱼种的差异对鱼糜介电特性和穿透深度有较大影响。SPSS软件可以对鱼糜的介电特性进行有效预测。  相似文献

利用木瓜蛋白酶、低温碱性酶、风味蛋白酶酶解红鱼(Sciaerlops ocellatus)鱼排,分别获得木瓜蛋白酶酶解物(PPH)、低温碱性酶酶解物(LPH)和风味蛋白酶酶解物(FPH)。对3种酶解物化学成分及氨基酸组成进行分析,结果显示,3种酶解物中总氨基酸含量都在70%以上,总氮含量为82.12%~84.61%。酶解物分别以5%(质量比)量加入鳙鱼(Aristichthys nobilis)鱼糜中,冻藏于-20℃。测定冻藏过程中肌原纤维蛋白质盐溶性和Ca^2+-ATPase活性的变化,同时测量鱼糜凝胶弹性的变化,并对冻藏样品进行扫描电镜观察,抗冻效果同商业抗冻剂(4%蔗糖、4%山梨醇、0.3%多聚磷酸盐混合剂)进行比较。结果表明,各酶解物能够在一定程度上抑制鱼糜蛋白质的冷冻变性,延缓肌原纤维蛋白质盐溶性和Ca^2+-ATPase活性的下降,鱼糜凝胶质量下降减少,其中木瓜蛋白酶酶解物具有最好的效果。  相似文献

The most important visual quality characteristic of Atlantic salmon is the red/pink flesh color. The primary source of this coloration in salmon is caused by deposition of relatively large amounts of pigments, such as astaxanthin, obtained from their diet. Astaxanthin is expensive, and in commercial farming practice, dietary color pigments comprises about 15-20% of the total feed cost. One important operational process in commercial fish farms is therefore to minimize pigment costs. Based on recent models on the effects of dietary pigment concentration and fish size on visual color perception of Atlantic salmon, this study has built a mathematical programming model designed to optimize dietary astaxanthin concentrations throughout the grow-out period that results in well-pigmented fish at minimum cost. We have applied a mixed-integer non-linear programming algorithm to solve this problem. Various managerial implications of applying optimization models in product quality management of farmed salmon are discussed.  相似文献

蛋白质对于鱼糜凝胶形成具有重要作用,通过研究鳊鱼鱼糜贮藏过程中肌球蛋白化学作用力、激光拉曼光谱等的变化探讨蛋白质结构对鱼糜凝胶形成的影响。结果发现,鱼糜在-80℃、-20℃冻藏过程中,肌球蛋白含量、离子键、氢键与疏水相互作用含量呈现先快后慢的二段式下降趋势,与其凝胶强度变化趋势一致;在0℃和5℃贮藏过程中,肌球蛋白含量、离子键和氢键含量呈不断下降的趋势,与凝胶强度变化趋势基本一致,而疏水相互作用为先增加后减少,由此可见,肌球蛋白含量、离子键和氢键含量在维持鱼糜蛋白高级结构上起着重要的作用。激光拉曼光谱结果显示α-螺旋结构是维持鳊鱼糜蛋白质网状结构的主要构象,在贮藏过程中,部分α-螺旋结构转变成无规卷曲,使包埋于肌球蛋白分子内的疏水性残基暴露于分子表面,进而发生蛋白变性;贮藏温度越高,无规卷曲结构越多,蛋白变性程度越大,表现在凝胶强度降低越多。鱼糜贮藏过程中蛋白质结构变化的研究对于阐明鱼糜凝胶形成机理和防止鱼糜蛋白变性具有一定意义。  相似文献
