An intensive vertically integrated monitoring program for broiler breeders and their offspring was set up in a Belgian poultry integration between 1993 and 1997. Serology for anti-reovirus antibodies was performed by enzyme-linked immunosorbent assay on blood samples taken from the broiler parents throughout rearing and production and also on blood samples taken at day-old and at slaughter from the broilers. Furthermore, production parameters of the birds were registered. All data were used two by two in a simple correlation study to calculate the degree to which these variables were linearly correlated. The reovirus antibody pattern indicated frequent field infections breaking through vaccinal immunity in the broiler parents. Under the epidemiologic conditions of this study, high antibody titers in the parents or in the broilers at day-old were significantly correlated with poor feed conversion, increased mortality, increased slaughterhouse condemnation, and low production score in the broilers. These correlations strongly support the view that reovirus infections can be economically important under European conditions of broiler production. Improvement of reovirus vaccines or the vaccination scheme in broiler parents or both may lead to better production results in the broiler offspring.  相似文献

In this study, a follow-up was made between 1993 and 1997 from broiler breeders at birth down to offspring broilers at processing, through vertically integrated registration of infectious bronchitis virus (IBV) antibody titers and performance data. All measurements were used two by two in a simple correlation study to calculate the degree to which they were linearly correlated. The antibody patterns in the broiler breeders indicated frequent field infections breaking through vaccinal immunity. Significant correlations measured between antibody titers and production parameters within and between the generations strongly suggested negative effects of IBV infections on laying percentage in the breeders and on mortality and daily weight gain in the broilers. Economic losses associated with IBV infections in the broilers occurred predominantly in flocks hatched with low and erratic maternal antibody titers. We concluded that IBV vaccination strategies should aim at high and uniform antibody titers in the broiler breeders.  相似文献

In recent years inclusion body hepatitis (IBH) has emerged as an economically important disease in Western Canada. Historically, infections with infectious bursal disease virus (IBDV) and chicken anemia virus (CAV) have been known to suppress the immune system of broilers and make them more susceptible to a secondary disease such as IBH. Recently it has been reported that virulent adenoviruses are able to cause IBH as a primary disease in broilers without apparent involvement of IBDV and CAV. The objectives of this study were to examine the possible association of IBH with IBDV and CAV infections in Western Canada and to identify adenoviruses involved in outbreaks. Serum samples from 17 broiler-breeder flocks and their progeny were collected when broilers were hatched and then again from broilers at the time of slaughter, and these samples were tested for IBDV and CAV antibodies by enzyme-linked immunosorbent assay (ELISA). Based on the ELISA titers the antibody response to vaccination against IBDV and CAV was at an expected level in all broiler flocks. Therefore, IBH outbreaks in these flocks were not due to inadequate levels of antibodies against IBDV and CAV. Moreover, there was no correlation found between occurrences of IBH outbreaks in broilers and their IBDV or CAV titers at the time of processing. Viruses that were isolated from livers of birds suffering from IBH could be classified into four different genotypes. Their hexon gene loop 1 sequences showed high percentages of identity to FAdV-7, FAdV-8a, FAdV-8b, and FAdV-11. The results of this study could not demonstrate an association of IBH with IBDV and CAV infections, but they supported the hypothesis that IBH in broilers in Western Canada is a primary disease with no apparent immunosuppressive involvement.  相似文献

A survey of antibodies against infectious bursal disease virus (IBDV), infectious bronchitis virus (IBV), Newcastle disease virus (NDV), and reovirus (RV) was conducted in broiler-breeder flocks and selected progeny broiler flocks utilizing the enzyme-linked immunosorbent assay. Marked differences in antibody titers between different breeder flocks were related to differences in vaccination programs. Poor performance in some progeny broiler flocks was related to low antibody titers against IBDV in the source breeder flocks. Progeny broiler flocks in which there was a high incidence of condemnations for airsacculitis had elevated antibody titers against IBV. A few progeny broiler flocks that experienced high mortality due to gangrenous dermatitis had no antibody titers against IBDV at processing. Antibody titers against RV were very variable and could not be related to any production problems.  相似文献

In this study, a Chinese field strain of subgroup J avian leukosis virus (ALV-J), NX0101, was studied for its immunosuppressive effects in both commercial broilers and SPF white Leghorn chickens infected at 1 day of age. Our data demonstrated that NX0101 induced much more significant body and immune organ weight loss in the infected commercial broiler chickens in an earlier age than that in the SPF white Leghorn chickens. At the same time antibody responses to vaccinations of Newcastle disease virus (NDV) and infectious bursa disease virus (IBDV) in the NX0101-infected chickens were also evaluated and compared between the commercial broiler chickens and the SPF white Leghorn chickens. Compared with the control group of chickens, the hemagglutination inhibition (HI) antibody response to NDV vaccines was significantly reduced in the NX0101-infected commercial broiler chickens from as early as 20 days after vaccination. However, no significant difference in HI antibody response was seen when HI titers reached their peaks in the NX0101-inoculated and control SPF white Leghorn chickens, except it declined significantly faster in infected birds. Neither of these two types of chickens showed significant decrease of antibody response to IBDV vaccination. Herein, we conclude that this NX0101 strain of ALV-J could selectively suppress humoral immune reactions to NDV, especially in broilers. But challenge experiments were not conducted and, therefore, it cannot be known if decreased antibody levels correlated with decreased protection against NDV in this case.  相似文献

The purpose of the present cross-sectional study was to evaluate the health status of organic broiler chickens and the contamination rate with Salmonella and Campylobacter in organic broiler production in Belgium. The broilers were screened for antibodies against routinely monitored poultry diseases at 1 day old and at slaughter. Fecal examination for the presence of worm eggs was done at slaughter. Bacteriological examination for the detection of Salmonella and Campylobacter was performed at day 1, week 2, week 4, week 7, week 10, and slaughter. Conventional broilers of the same poultry integration and reared in the same geographic area were also screened and served as reference. Serologic data indicated lower antibody titers against infectious bronchitis and Newcastle disease in organic flocks. No significant differences could be found in prevalence of Salmonella between organic and conventional broilers at slaughter. In contrast, Campylobacter infections at slaughter were significantly higher in organic flocks. Organic flocks most probably become infected with Campylobacter between week 7 and week 10. Worm eggs were found in neither the organic flocks nor the conventional flocks. In conclusion, there are indications that the respiratory health status is better in organic broilers but that organic flocks are more often infected with Campylobacter than are conventional flocks.  相似文献

Different infectious bursal disease virus (IBDV) live vaccines (intermediate, intermediate plus) were compared for their immunosuppressive abilities in specific-pathogen-free (SPF) layer-type chickens or commercial broilers. The Newcastle disease virus (NDV) vaccination model was applied to determine not only IBDV-induced immunosuppression but also bilateral effects between IBDV and NDV. None of the IBDV vaccines abrogated NDV vaccine-induced protection. All NDV-vaccinated SPF layers and broilers were protected against NDV challenge independent of circulating NDV antibody levels. Sustained suppression of NDV antibody development was observed in SPF layers, which had received the intermediate plus IBDV vaccine. We observed a temporary suppression of NDV antibody development in broilers vaccinated with one of the intermediate, as well as the intermediate plus, IBDV vaccines. Different genetic backgrounds, ages, and residual maternal antibodies might have influenced the pathogenesis of IBDV in the different types of chickens. Temporary suppression of NDV antibody response in broilers was only seen if the NDV vaccine was administered before and not, as it was speculated previously, at the time the peak of IBDV-induced bursa lesions was detected. For the first time, we have demonstrated that the NDV vaccine had an interfering effect with the pathogenesis of the intermediate as well as the intermediate plus IBDV vaccine. NDV vaccination enhanced the incidence of IBDV bursa lesions and IBDV antibody development. This observation indicates that this bilateral effect of an IBDV and NDV vaccination should be considered in the field and could have consequences for the performance of broiler flocks.  相似文献

A flock of broiler parents was hyperimmunised against Newcastle disease (ND) by subcutaneous injection of an experimental high-potency inactivated oil emulsion (IOE) vaccine in order to achieve high and long-lasting passive protection in broilers. During the egg production period mean log2 HI ND titre of the parents slowly decreased from 11.6 at 26 weeks of age to 10.0 at 53 weeks of age. Titres of day-old progeny paralleled those of their parents. Mean protection of broilers against mortality following challenge with Herts 33 virus at the ages of 1, 2, 3 and 4 weeks was 86%, 73%, 54% and 44%, respectively. As compared with the current Dutch situation, application of the high-potency IOE vaccine will significantly increase antibody titres in parent flocks and the level and duration of passive protection in broilers. It is concluded that passive ND protection of broilers by hyperimmunisation of the dams is realisable from an economic point of view and is suitable for low-risk areas.  相似文献

Growth rate in broiler birds has increased substantially in the last decade due to improvement in genetics, feed formulation, cleaner environment, and vaccine formulations. As a result, it has become necessary to review and revise prediction method for vaccination in chicks. This study was undertaken to determine the possible use of the rate of weight gain rather than age in predicting vaccination time. Two groups of 1-day-old broilers originating from old and young breeders, respectively, and with different levels of maternal antibodies against infectious bursal disease virus (IBDV) were used in this study. The chicks were divided into four groups and subjected to two feed regiments: groups A1 and B1 were fed broiler feed for normal growth rate, and groups A2 and B2 were fed breeder feed for slower growth rate. At 1, 4, 8, 12, 16, 22, 29, and 36 days of age, 22 chicks in each group were weighed, and blood samples were collected. Serum samples were tested for antibodies against IBDV by enzyme-linked immunosorbent assay (ELISA) and virus neutralization test. Maternal antibody decline curves for each group were plotted according to chick age and chick weight. Fast-growing birds in groups A1 and B1 showed a faster rate of antibody decline, whereas slow-growing birds in groups A2 and B2 had a slower rate of antibody decline. Based on the effect of weight gain on maternal antibody decline, a new way of predicting vaccination time for IBDV based on measuring maternal antibody titers at 4 days of age was proposed and tested. The predicted antibody decline was shown to correspond to the real ELISA titers measured in our experiments (R = 0.9889), whereas a lower correlation (R = 0.8355) was detected between real ELISA titers and the titers predicted by the current method using age-based Deventer formula.  相似文献

Broiler chickens infected at 3 weeks of age with infectious bursal disease virus (IBDV) were given Brucella abortus (BA) or sheep red blood cell (SRBC) antigens before, during, and after the acute phase of the infection. Gland of Harder (GH) extracts and serum samples were used to assay local and systemic antibody titer to each antigen 7 days after antigen was administered. Antibody titers to both BA and SRBC antigens were lower (P less than 0.05) in GH extracts and serum of IBDV-infected broilers than uninfected controls. The responses to BA, a thymus-independent antigen, took longer to become depressed than the responses to SRBC, a thymus-dependent antigen. The depression of antibody titers following IBDV inoculation suggests compromise of both local and systemic immune function, a finding of importance to the broiler industry.  相似文献

Two distinct serotypes of infectious bursal disease virus (IBDV) are recognized in chicken and turkey flocks in the United States. Serologic testing of chicken flocks for serotype 1 viruses is routinely performed to monitor disease status and vaccination. Earlier studies indicated that enzyme-linked immunosorbent assay (ELISA) test detects antibodies to both serotypes of the virus, while the virus neutralization (VN) test is serotype specific. It is useful to evaluate currently available commercial ELISA kits for their ability to differentiate between antibodies elicited by the two serotypes. Three trials were performed in which chickens were orally inoculated with either a high or a low dose of serotype 1 STC or serotype 2 OH strains of IBDV. Sera collected at 0, 7, 14, and 21 days from these chickens and antisera procured from naturally infected broiler (n=20) and layer (n=30) flocks were tested with five different commercial ELISA kits and by VN. All ELISA kits detected different levels of antibodies elicited against serotype 1 of the virus and moderate and high levels of antibodies against serotype 2 virus. A correlation existed between the ELISA and the VN titers of experimentally infected chickens. All serum samples tested from the commercial layer flocks and 65% of the broiler flocks had antibodies against the OH strain. However, no correlation between the VN titers and ELISA titers was observed for the commercial broilers and layers sera by the majority of the kits. The results indicated that currently available commercial ELISA kits detect antibodies elicited by the two serotypes of IBDV. Hence, the prevalence of serotype 2 antibodies in the flocks should be considered while determining antibody profiles of the flocks against serotype 1 viruses.  相似文献

A serologic survey in unvaccinated broiler parent and broiler progeny flocks demonstrated seroconversion against chicken infecrious anemia virus (CIAV) in all parent flocks before or around point of lay and in 38% of the broiler flocks examined at slaughter age. The presence of CIAV antibodies at slaughter of broilers was positively correlated with slaughterhouse condemnation rates. Results indicate that CIAV infections are highly prevalent in both broiler parent and broiler flocks and that CIAV infections in broilers are associated with increased slaughterhouse condemnation.  相似文献

Necrotic enteritis (NE), a disease associated with high numbers of the intestinal bacterium Clostridium perfringens, is common in intensive broiler production. Antimicrobial feed additives may control the disease, but their use is now being questioned in many countries. A field study was undertaken at the end of 1997 to study the level of naturally occurring specific humoral immunity against phospholipase C (PLC; C perfringens alpha toxin) in Norwegian broiler flocks. Blood samples were collected at hatch from 61 study flocks, and the sampling was repeated for 56 of the same flocks at processing. The level of specific antibodies against PLC was analyzed in an enzyme-linked immunosorbent assay (ELISA) test. Data on production performance and weekly mortality were recorded. The relationship between the age of the hens and the level of specific maternal antibodies in the progenies was studied. The association between the level of the maternal antibodies and the production performance, including mortality, was analyzed. The level of specific antibodies against PLC in day-old broiler flocks was relatively high and varied considerably compared with the levels in the broilers at processing. The progenies from the oldest hens had significantly higher levels of specific antibodies than the chicks from younger hens. No outbreak of NE occurred during the study period, making it impossible to analyze the association between naturally occurring specific immunity against PLC and the occurrence of the disease. However, the results showed that the flocks with high titers of specific maternal antibodies against PLC had lower mortality during the production period than flocks with low titers.  相似文献

A computer-assisted single-serum-dilution indirect kinetic-based enzyme-linked immunosorbent assay (KELISA) was used for quantitating the natural decay rate of infectious bursal disease virus (IBDV) maternal antibody in progeny obtained from white leghorn breeders. The KELISA results were compared with those of a standard virus-neutralization (VN) test. Pullets were subjected to two IBDV immunization regimens. Group 1 was vaccinated at weeks 0, 2, and 10 with two live vaccines in drinking water and at week 20 with an oil-emulsified (SC) IBDV vaccine, group 2 received only the first and last immunizations, and group 3 served as the unvaccinated control. Pullets were artificially inseminated at 28 weeks of age. Progeny chicks from each group were bled every other day for 47 days. Both KELISA and VN test detected linear relationship in the decay of maternal antibodies. The VN test detected no significant differences (P greater than 0.05) in the IBDV maternal antibody titers at day 1 or in the rate of decay between the progeny from groups 1 and 2. The VN maternal antibody titers decreased at a rate of 0.16 log2 titer per day. In contrast, KELISA revealed higher IBDV maternal antibody titers in day-old progeny from pullets vaccinated 4 times (log2 = 14.3). However, KELISA titers of progeny from this group decreased at a faster rate than titers of progeny from pullets vaccinated twice (0.20 vs. 0.13 log2 titer per day).  相似文献

试验旨在研究红景天提取物对高原缺氧条件下肉鸡接种法氏囊疫苗和绵羊红细胞后产生的特异性抗体水平的影响。随机选取100只1日龄健康AA肉仔鸡,随机分为5组。其中,对照组只饲喂基础日粮,其余4组分别在基础日粮中添加0.10%、0.15%、0.20%和0.40%红景天水提物干粉。每个组取半数鸡只于13日龄腹腔注射接种绵羊红细胞,另一半鸡只于20日龄口服接种法氏囊疫苗。研究发现,免疫接种法氏囊疫苗后2 d和19 d,相比于对照组,0.20%和0.40%红景天处理组显著提高了法氏囊疫苗抗体水平(P<0.05)。在腹腔注射绵羊红细胞后5 d、8 d和12 d时,相比对照组,0.20%和0.40%红景天处理组显著提高了绵羊红细胞抗体水平(P<0.05)。该试验表明红景天提取物可以提高高海拔缺氧条件下肉鸡特异性抗体水平,因此其可能作为高海拔缺氧条件下肉鸡日粮的一种免疫调节性添加剂。  相似文献

Reoviruses are an important cause of suboptimum performance in commercial broilers worldwide. Integrators use the enzyme-linked immunosorbent assay against the S1133 antigen for monitoring serum of breeders for indicating pullet vaccine success. However, without correlating serology to reovirus challenge, it is difficult to determine whether titers reflect protective immunity. We developed a broiler challenge test against 2 common reovirus isolates (2408 and S1133) to evaluate the efficacy of reovirus pullet vaccine programs. Two reovirus serologic and challenge studies were undertaken using chicks from broiler integrators from the southeastern United States. Breeder flocks, from which the chicks were obtained, received at least 1 live and 2 inactivated reovirus vaccines during their pullet phase. One-day-old progeny were collected from 6 breeder flocks. At 1 d of age, 20 chicks from each broiler flock were bled, and serum was analyzed for antibodies. At 3 to 4 d of age, 20 progeny per flock were challenged with the 2408 reovirus by intratracheal route. At 10 to 14 d of age, another 20 birds per flock were challenged with the S1133 reovirus by footpad. Twenty birds per flock were used as nonchallenged controls. At 3 wk of age, all birds were killed and weighed. Percentage of protection was calculated for each flock based on the absence of gross lesions. Flocks with at least 50% protection were considered well protected. Most flocks were well protected against both viruses. The percentage of protection correlated with day-old enzyme-linked immunosorbent assay titers. Chicks from younger hens had higher titers and the best protection against challenge. Producers, whose hen flocks were monitored herein, were doing a good job of immunizing pullets against reovirus. They are now using reovirus progeny challenge studies along with breeder antibody titers to determine vaccination success of their pullets.  相似文献

In ovo vaccination against Marek's disease virus and infectious bursal disease virus (IBDV) in commercial broilers in the United States is common. Little information exists as to the safety and efficacy of intermediate IBDV vaccines given in ovo. Experiments were initiated to determine the safety and efficacy of three commercially available live intermediate IBDV vaccines by in ovo route. Commonly used vaccines were given at 18 days of embryonation to specific-pathogen-free (SPF) broiler embryos (first and second study) or to commercial broiler embryos (third study) that had maternal antibody against IBDV. When any of the antigenic standard vaccines was given at full dose to SPF embryos, embryonic and 3-wk posthatch mortality increased. Vaccines also caused significant microscopic lesions in the bursa of Fabricius at 1 and 3 wk posthatch. In contrast, there was no adverse effect on embryonic or posthatch mortality when vaccines were given at half dose to SPF or commercial broiler embryos. However, significant microscopic lesions were evident at 1 and 3 wk posthatch in the bursae of SPF embryos given the vaccines at half dose. When vaccines were given at half dose to commercial broiler embryos, lesions were evident at 1 but not 3 wk of age. In the third study, in ovo vaccinated chickens were challenged with either a virulent standard (APHIS) or antigenic variant (variant E) IBDV virus at 3 wk of age. All vaccines produced at least 87% protection against the standard and 60% protection against the variant challenge IBDV, as measured by bursal weight to body weight ratios. This study was the first to examine the safety and efficacy of the three commonly used intermediate IBDV vaccines given in ovo in protection against standard and antigenic variant IBDV challenge viruses.  相似文献

A field study was designed to determine the prevalence of subclinical infectious bursal disease (IBD) in broiler chickens from a commercial poultry company. Bursae of Fabricius (BF) from two vaccinated and three nonvaccinated broiler flocks were evaluated histologically, and antibody profiles of these broiler and matched parent breeder flocks were established. Lesions of IBD, including lymphoid necrosis, stromal edema, and infiltrates of heterophils and macrophages, were first detected in BF at 24 days of age in both vaccinated and nonvaccinated chickens. At 41 days, all BF had lesions characteristic of IBD, including severe lymphoid depletion, proliferation of epithelial cells, and mild fibroplasia. Although mean maternal antibody levels (measured by enzyme-linked immunosorbent assay) in broilers were apparently protective through day 12, IBD antibodies decreased to nonprotective levels (below 1,000) by day 16 or 20. Titers began to increase by day 28 or 32 because of field exposure. Sentinel birds, placed with broiler flocks, also developed IBD antibody titers. Broiler breeders had low and nonuniform antibody titers. Prevalence of field IBD exposure was high, and existing vaccination programs were not effective.  相似文献

To evaluate the economic effects of subclinical chicken anemia agent (CAA) infection on broiler performance, clinically normal broiler flocks were grouped into two categories: A) flocks in which none of 10 birds sampled at slaughter had antibody to CAA, and B) flocks in which six or more of 10 similarly sampled birds had CAA antibody. Production and performance parameters of 25 flocks in each category were compared. No statistically significant differences were found between category A and category B flocks in major production parameters such as sex, feed manufacturer, type of litter, stocking density, and age at slaughter. However, category A flocks achieved 13% (P less than 0.05) greater net income per 1000 birds, 2.0% better feed-conversion ratio (P less than 0.05), and 2.5% (P less than 0.05) greater average weight per bird than category B flocks. No significant differences in hockburn bonus per 1000 birds (a bonus payable to growers whose broilers have the lowest prevalence of contact dermatitis lesions on the hocks) and mortality were found between category A and category B flocks. These results show that subclinical CAA infection has a substantial, statistically significant effect on commercial broiler performance and profitability.  相似文献
