The binding processes of thearubigin, which is one of the two major polyphenols (the other one is theaflavin) that gives black tea its characteristic color and taste, to the bovine serum albumin (BSA) surface have been investigated by quartz crystal microbalance with dissipation monitoring (QCM-D). The mass and thickness of the thearubigin adlayer on BSA surfaces at various thearubigin concentrations, salt concentrations, and pH values have been determined by QCM-D using the Voigt model. Our results show that the adsorption isotherm of thearubigin on the BSA surface can be better described by the Langmuir model than the Freundlich model, suggesting that the thearubigin adsorption on the BSA surface is dominated by specific interactions, such as electrostatic interaction and hydrogen bonding, as evidenced by the stronger thearubigin adsorption at pH below the isoelectric point (pI) of BSA and shifts in the positions of both amide bands in the FTIR spectra of the BSA surface with and without thearubigin adsorption. The addition of salt can also influence the thearubigin binding to BSA surfaces. The salt concentration-enhanced effect at a salt concentration lower than 0.1 M is explained as that an increase of salt concentration can screen the electrostatic repulsion to a larger extent than the electrostatic attraction between thearubigin and BSA. On the other hand, when the salt concentration is higher than 0.1 M, both electrostatic repulsion and attraction can be significantly screened by the higher salt concentration, resulting in the salt concentration-reduced effect. However, when the salt concentration is further increased to 0.4 M, the addition of thearubigin may promote the formation of a certain type of complex with BSA, resulting in the increases of both thickness and mass of the thearubigin adlayer.  相似文献

The effect of ionic strength ( I) on the interfacial interactions between pectin and the bovine serum albumin (BSA) surface has been investigated using the quartz crystal microbalance with dissipation monitoring (QCM-D). As I increases from 0.01 to 0.02 M, the frequency shift (Delta F) decreases, whereas the energy dissipation shift (Delta D) changes toward a higher value. Further increase of I from 0.02 to 0.5 M causes both Delta F and Delta D to gradually return to almost zero. The adsorbed mass and thickness of the pectin adlayer estimated from the Voigt model confirm that the adsorption of pectin and the formation of thicker pectin adlayers on a BSA surface are favored by the increase of ionic strength at I = 0.01 approximately 0.02 M. An increase of I above 0.02 M hinders pectin adsorption and causes the formation of a thinner pectin adlayer. The ionic strength-enhanced effect at I values lower than 0.02 M is explained as an increase of ionic strength that can screen the electrostatic repulsion to a larger extent than the electrostatic attraction between pectin and BSA. However, when I is higher than 0.02 M, both electrostatic repulsion and attraction can be significantly screened by the increasing ionic strength, resulting in the ionic strength-reduced effect. On the other hand, the high viscoelasticity of the pectin adlayer revealed by the Voigt model suggests the formation of a network-structured pectin adlayer on the BSA surface, which contains two steps for higher pectin adsorptions at I = 0.0125 approximately 0.1 M by the indication of two slopes in Delta D-Delta F plots.  相似文献

Quartz crystal microbalance with dissipation monitoring (QCM-D) has been employed to study the interactions between (-)-epigallocatechin gallate (EGCG) and bovine serum albumin (BSA) surface. The adsorbed mass, thickness, and viscoelastic properties of EGCG adlayer on BSA surface at various EGCG concentrations, temperatures, sodium chloride concentrations, and pH values have been determined by QCM-D in combination with the Voigt model. The adsorption isotherm of EGCG on BSA surfaces can be better described by the Freundlich model than the Langmuir model, indicating that EGCG adsorption on BSA surfaces is dominated by nonspecific hydrophobic interactions, as supported by stronger EGCG adsorption at higher temperature. Shifts in the Fourier transform infrared spectra of the BSA surface with and without EGCG adsorption disclose that hydrogen bonding might also be involved in EGCG adsorption on BSA surfaces. The addition of salt and change of pH can also influence the EGCG adsorption on BSA surfaces. Usually, higher EGCG adsorption leads to higher values of viscosity and shear elastic modulus of EGCG adlayer, which can be explained by the aggregation of BSA through EGCG bridges. Compared with EGCG, nongalloylated (+)-catechin shows much lower adsorption capacity on BSA surfaces, suggesting the importance of the galloyl group in polyphenol/protein interactions.  相似文献

Quercetin is an abundant flavonoid in the human diet with numerous biological activities, which may contribute to the prevention of human disease but also may be potentially harmful. Quercetin is oxidized in cells to products capable of covalently binding to cellular proteins, a process that may be important for its biological activities. In the present study, using radiolabeled drug and quantifying the products after electrophoretic separation, proteins to which oxidized quercetin is binding irreversibly were identified. The binding of quercetin to human serum albumin (HSA) in human blood and the effect of stimulation of neutrophilic myeloperoxidase on this binding were also measured. The in vitro binding of quercetin to eight proteins in the presence of catalytic amounts of horseradish peroxidase and hydrogen peroxide was highly selective for HSA. For all proteins the binding was dramatically decreased by reduced L-glutathione. In the blood samples, the release of neutrophilic myeloperoxidase by phorbol ester caused a 3-fold increase in the binding of quercetin to HSA. This study shows that quercetin in the presence of peroxidase/hydrogen peroxide covalently links to proteins with a particularly high affinity for HSA and that this also may occur in vivo after exposure to quercetin. This provides further insights into the complex behavior of this major dietary flavonoid.  相似文献

The oxidation of methyl linoleate (LMe) in the presence of bovine serum albumin (BSA) was studied to analyze both the processes involved when lipid oxidation occurs in the presence of proteins and the relative progression of the several reactions implicated. The disappearance of LMe, the formation of primary and secondary lipid oxidation products, the loss of essential amino acids, and the production of oxidized lipid/amino acid reaction products (OLAARPs) were studied as a function of incubation time. During the first steps of lipid oxidation, LMe was converted quantitatively to methyl linoleate hydroperoxides, which were very rapidly degraded to either secondary products of lipid oxidation or OLAARPs. No significant differences were identified in the major lipid oxidation products formed in incubations with or without proteins, indicating that mechanisms for formation of these compounds are similar in both cases. In addition, no significant differences were observed between the time-courses of formation of secondary oxidation products and OLAARPs, suggesting that hydroperoxide decomposition and OLAARP formation occur simultaneously when the lipid oxidation process takes place in the presence of proteins. Furthermore, OLAARP formation seems to be an unavoidable process that should be considered as a last step in the lipid peroxidation process.  相似文献

The aqueous extract of Eucommia ulmoides leaves has been commonly known as Du-zhong tea as a functional health food for the treatment of hypertension, hypercholesterolemia, and fatty liver. This study developed a centrifugal ultrafiltration-high-performance liquid chromatography (HPLC) method for screening and identification of bioactive compounds in E. ulmoides leaves binding with bovine serum albumin (BSA). Six active compounds were screened, isolated, and elucidated by their ultraviolet (UV), electrospray ionization-mass spectrometry (ESI-MS), and nuclear magnetic resonance (NMR) data as geniposidic acid (1), caffeic acid (2), chlorogenic acid (3), quercetin-3-O-sambubioside (4), rutin (5), and isoquercitrin (6). The interaction between active compounds and BSA was investigated in the absence and presence of other compounds by quenching the intrinsic BSA fluorescence. The results indicated that the structures significantly affected the binding process. The values of binding constants for compounds 2-6 were in the range of 10(5)-10(6) mol L(-1), while geniposidic acid (1) hardly quenching the BSA intrinsic fluorescence. However, the quenching process of geniposidic acid was easily affected in the presence of other active compounds. The formation of the geniposidic acid-phenylpropanoid (flavonoid) complex could increase the binding affinity of geniposidic acid with BSA; however, the increased steric hindrance of the complex may make phenylpropanoid or flavonoid dissociate from BSA and then decrease their affinities.  相似文献

Chitin neoglycoconjugates (BSA-CO) were obtained by the conjugation of bovine serum albumin (BSA) with chitin oligosaccharides (CO) through the Maillard reaction (nonenzymatic glycation). CO produced by acid hydrolysis of chitin were fractionated using an ultrafiltration membrane system (1-3 kDa cutoff). The Maillard reaction was carried out by heating a freeze-dried mixture containing BSA and CO at 60 °C (under 43% relative humidity for 6 and 12 h). BSA-CO were characterized by available amino groups content, intrinsic tryptophan emission spectra, gel electrophoresis, and mass spectrometry. Biological assays included interaction with wheat germ agglutinin (WGA) and with bacterial adhesins of Escherichia coli K88+ and Salmonella choleraesuis. Glycation of BSA was revealed by reduction of available amino groups and fluorescence intensity and also retarded migration through SDS-PAGE. Conjugation of BSA with chitin oligomers appeared to be time dependent and was confirmed by mass spectrometry, by which molecular mass increase for monomers and dimers was observed. Monomers were estimated to contain either one or two glycation sites (at 6 and 12 h of treatment, respectively), with one or two tetrasaccharide units attached. Consequently, dimers showed two or four glycation sites. BSA-CO presented biological recognition by WGA and E. coli K88+ and S. cholerasuis adhesins. The strategy used in this work represents a simple method to obtain glycoconjugates to study applications involving protein-carbohydrate recognition.  相似文献

A new analytical method for measuring tannic acid (TA) using tannase was developed and applied to the investigation of TA metabolism in the rat following oral administration at a dose of 1.0 g/kg. The proposed method for TA determination was based on the enzymatic hydrolysis of TA to gallic acid (GA) and subsequent determination by HPLC. TA metabolites were determined by HPLC. 4-O-Methylgallic acid (4-OMGA), pyrogallol (PY), and resorcinol (RE) were detected in serum. TA was excreted into urine as GA (0.01%), 4-OMGA (0.10%), PY (0.24%), and RE (2.06%) and into feces as TA (62.74%), GA (0.19%), PY (0.02%), and RE (0.76%) within 54 h after oral administration. It was suggested that >60% of TA remained unchanged but that some was hydrolyzed to GA by tannase in the intestine and further metabolized to 4-OMGA, PY, and RE.  相似文献

The binding mechanism of molecular interaction between diosmetin and human serum albumin (HSA) in a pH 7.4 phosphate buffer was studied using atomic force microscopy (AFM) and various spectroscopic techniques including fluorescence, resonance light scattering (RLS), UV-vis absorption, circular dichroism (CD), and Fourier transform infrared (FT-IR) spectroscopy. Fluorescence data revealed that the fluorescence quenching of HSA by diosmetin was a static quenching procedure. The binding constants and number of binding sites were evaluated at different temperatures. The RLS spectra and AFM images showed that the dimension of the individual HSA molecules were larger after interaction with diosmetin. The thermodynamic parameters, ΔH° and ΔS° were calculated to be -24.56 kJ mol(-1) and 14.67 J mol(-1) K(-1), respectively, suggesting that the binding of diosmtin to HSA was driven mainly by hydrophobic interactions and hydrogen bonds. The displacement studies and denaturation experiments in the presence of urea indicated site I as the main binding site for diosmetin on HSA. The binding distance between diosmetin and HSA was determined to be 3.54 nm based on the F?rster theory. Analysis of CD and FT-IR spectra demonstrated that HSA conformation was slightly altered in the presence of diosmetin.  相似文献

The effects of 1:1 flavonoid-Cu(2+) complexes of four flavonoids with different C-ring substituents, quercetin (QU), luteolin (LU), taxifolin (TA), and (+)-catechin (CA), on bovine serum albumin (BSA) were investigated and compared with corresponding free flavonoids by spectroscopic analysis in an attempt to characterize the chemical association taking place. The results indicated that all of the quenching mechanisms were based on static quenching combined with nonradiative energy transfer. Cu(2+) chelation changed the binding constants for BSA depending on the structures of flavonoids and the detected concentrations. The reduced hydroxyl groups, increased steric hindrance, and hydrophilicity of Cu(2+) chelation may be the main reasons for the reduced binding constants, whereas the formation of stable flavonoid-Cu(2+) complexes and synergistic action could increase the binding constants. The changed trends of critical energy transfer distance (R(0)) for Cu(2+) chelation were contrary to those of binding constants.  相似文献

The influence of sucrose (0--40 wt %) on the thermal denaturation and gelation of bovine serum albumin (BSA) in aqueous solution has been studied. The effect of sucrose on heat denaturation of 1 wt % BSA solutions (pH 6.9) was measured using ultrasensitive differential scanning calorimetry. The unfolding process was irreversible and could be characterized by a denaturation temperature (T(m)), activation energy (E(A)), and pre-exponential factor (A). As the sucrose concentration increased from 0 to 40 wt %, T(m) increased from 72.9 to 79.2 degrees C, E(A) decreased from 314 to 289 kJ mol(-1), and ln(A/s(-1)) decreased from 104 to 94. The rise in T(m) was attributed to the increased thermal stability of the globular state of BSA relative to its native state because of differences in their preferential interactions with sucrose. The change in preferential interaction coefficient (Delta Gamma(3,2)) associated with the native-to-denatured transition was estimated. The dynamic shear rheology of 2 wt % BSA solutions (pH 6.9, 100 mM NaCl) was monitored as they were heated from 30 to 90 degrees C, held at 90 degrees C for either 15 or 120 min, and then cooled to 30 degrees C. Sucrose increased the gelation temperature due to thermal stabilization of the native state of the protein. The complex shear modulus (G) of cooled gels decreased with sucrose concentration when they were held at 90 degrees C for 15 min because the fraction of irreversibly denatured protein decreased. On the other hand, G of cooled gels increased with sucrose concentration when they were held at 90 degrees C for 120 min because a greater fraction of irreversibly denatured protein was formed and the strength of the protein-protein interactions increased.  相似文献

The interaction between ilaprazole and bovine serum albumin (BSA) has been investigated in the absence and presence of four popular flavonoids with different C-ring structures, quercetin, luteolin, taxifolin, and (+)-catechin, by means of fluorescence spectroscopy. The results indicated that ilaprazole had a strong ability to quench the intrinsic fluorescence of BSA, and site marker competitive experiments indicated that the binding of ilaprazole to BSA primarily took place in subdomain IIA. The quenching process of ilaprazole with BSA was easily affected by flavonoids,; however, they did not change the quchenching mechanism of ilaprazole with BSA, whereas all of the fluorescence quenching was initiated by a static quenching procedure combining with nonradiative energy transfer. The presence of flavonoids decreased the quenching constants of ilaprazole with BSA from 2.2 to 23.7% and decreased the binding constants from 73.7 to 98.3%, which depended on the different flavonoids' structures. The decreased binding constants and unchangeable spatial distance of ilaprazole with BSA by the introduction of quercetin, luteolin, and taxifolin may result from the competition of flavonoids and ilaprazole binding to BSA, whereas in the presence of (+)-catichin, decreased binding constants and increased spatial distance possibly resulted from the formation of a ternary ilaprazole-BSA-(+)-catechin complex. All of these results may have relevant consequences in rationalizing the interferences of common food to gastric ulcer treatments.  相似文献

Chlorination can significantly enhance the antioxidant and antitumor activity of genistein. In this paper, genistein, 8-chlorogenistein, and 3',8-dichlorogenistein were selected to investigate the binding to bovine serum albumin (BSA) using fluorescence spectroscopy and circular dichroism (CD). The results showed that chlorination, especially at position 3', had significant effects on the binding constant value of chlorinated genistein derivatives to BSA; however, the binding site and the binding number were slightly affected. The thermodynamic parameters indicated that hydrophobic and electrostatic forces played important roles in the binding process and the enhanced binding affinity mainly associated with the increase of the hydrophobicity caused by the chlorine atom substitution. Furthermore, the CD data demonstrated that the conformation of BSA was slightly altered in the presence of genistein, 8-chlorogenistein, and 3',8-dichlorogenistein, with different reduced α-helix contents. The results obtained herein will be of biological significance in toxicology investigation and genistein derivative drug design.  相似文献

The oxidation of methyl linoleate (LMe) and methyl linolenate (LnMe) in the presence of bovine serum albumin (BSA) in the dark at 60 degrees C was studied to analyze the role of the type of fatty acid and the protein/lipid ratio on the relative progression of the processes involved when lipid oxidation occurs in the presence of proteins. The disappearance of the fatty acid, the formation of primary and secondary products of lipid peroxidation, the loss of amino acid residues, the production of oxidized lipid/amino acid reaction products, and the development of color and fluorescence were studied as a function of incubation time in protein/lipid samples at 10:1, 6:1, and 3:1 w/w ratios. The incubation of LMe and LnMe in the presence of BSA at 60 degrees C rapidly produced lipid peroxidation and protein damage. Although reaction rates were much faster for LnMe than for LMe, both fatty acids had similar behaviors, and LnMe seemed to be only slightly more reactive than LMe for BSA by producing a higher increase of protein pyrroles in the protein and the development of increased browning and fluorescence. The protein/lipid ratio also influenced the relative progress of the reactions implicated. Thus, a lower protein/lipid ratio increased sample oxidation and protein damage. This also produced an increased browning, in accordance with the mechanisms proposed for browning production by oxidized lipid/protein reactions. On the contrary, browning of extracted lipids increased at higher protein/lipid ratios. This opposite tendency allowed evaluation of the overall significance of the different browning processes implicated in the final colors observed, concluding that color changes observed in BSA/lipid samples were mostly a consequence of oxidized lipid/protein reactions.  相似文献

Phenolic compounds are responsible for major organoleptic characteristics of plant-derived food and beverages; these substances have received much attention, given that the major function of these compounds is their antioxidant ability. In the context of this study, our major aim was study the binding of several phenolic compounds such as (+)-catechin, (-)-epicatechin, (-)-epicatechin gallate, malvidin-3-glucoside, tannic acid, procyanidin B4, procyanidin B2 gallate, and procyanidin oligomers to different proteins (bovine serum albumin and human alpha-amylase) by fluorescence quenching of protein intrinsic fluorescence. From the spectra obtained, the Stern-Volmer, the apparent static, and the bimolecular quenching constants were calculated. The structure of polyphenols revealed to significantly affect the binding/quenching process; in general, the binding affinity increased with the molecular weight of polyphenol compounds and in the presence of galloyl groups. For catechin monomer and procyanidin dimer B4, the K(SV) was 14,100 and 13,800 M(-1), respectively, and for galloyl derivatives, the K(SV) was 19,500 and 21,900 M(-1), respectively. Tannic acid was shown to be the major quenching molecule for both proteins. However, comparing different proteins, the same polyphenol showed different quenching effects, which are suggested to be related to the three-dimensional structure of the proteins studied. For (+)-catechin and BSA, the K(SV) was 8700 M(-1), and with alpha-amylase, it was 14,100 M(-1); for tannic acid, the K(SV) was 10,0548 and 11,0674 M(-1), respectively. From the results obtained, besides the main binding analysis performed, we conclude that this technique is more sensitive than thought because we can detect several interactions that have not been proven by other methods, namely, nephelometry. Overall, fluorescence quenching has proven to be a very sensitive technique with many potentialities to analyze the interaction between polyphenols and proteins.  相似文献

Oxidative deamination by various polyphenolic compounds is presumed to be due to the oxidative conversion of polyphenols to the corresponding quinones through autoxidation. Here we examined the oxidative deamination by the polyphenol-rich beverages green tea, black tea, and coffee at a physiological pH and temperature. Green tea, black tea, and coffee extracts oxidatively deaminated benzylamine and the lysine residues of bovine serum albumin to benzaldehyde and alpha-aminoadipic delta-semialdehyde residues, respectively, in sodium phosphate buffer (pH 7.4) at 37 degrees C in both the presence and absence of Cu2+, indicating the occurrence of an amine (lysyl) oxidase-like reaction. We also examined the effects of pH and metal ions on the reaction. The possible biological effects of drinking polyphenol-rich beverages on human are also discussed.  相似文献

Anthocyanins from the purple Solanum tuberosum newly cultivated by the Taian Academy of Agricultural Sciences were extracted and analyzed using high-performance liquid chromatography (HPLC) and UV-vis spectroscopy. Four individual anthocyanins were detected as the major components, and the total anthocyanin content was 273.5 ± 14.3 mg of cyanidin-3-glucoside equiv/100 g of dry seeds. Results of color stability showed that the purple S. tuberosum anthocyanins (PSTAs) are more stable under low pH and temperatures. Heat and general food additives have fine compatibility with PSTAs; however, they are very sensitive with oxidant and reduction. The influence of PSTAs on Cr(VI) targeted to bovine serum albumin (BSA) was also studied. The quenching of BSA fluorescence caused by Cr(VI) could be delayed by PSTAs. UV-vis and circular dichroism (CD) data suggested that PSTAs can protect the secondary and tertiary structures of BSA by probably interacting with Cr(VI) in advance.  相似文献

采用批实验研究了菲在单宁酸干扰下形成的不同晶形铝氧化物上的吸附现象,并用不同的吸附等温线方程对吸附平衡数据进行了拟合,重点比较了线性和非线性回归方法估计吸附等温线参数的差异。结果表明:菲在各种晶形的铝氧化物上都有明显的吸附,但并不是完全随着单宁酸含量和结晶度的变化而规律性变化。吸附平衡数据以Langmuir、Redlich-Peterson和Dubinin-Radushkevich吸附等温方程,用不同的回归方法估计的等温线参数值均有显著性差异。线性回归得到的参数有不确定性,表明用线性回归来判断吸附等温线能否对吸附平衡数据进行最优拟合是不可靠的。相反,非线性回归能较好地确定菲在不同晶形铝氧化物上的最佳吸附等温线及相应参数。采用R2和χ2共同检验发现,菲在4种不同结晶度(单宁酸与铝的摩尔比(MR)=0,10-3,10-2,10-1)铝氧化物上的最佳吸附等温线方程并不尽相同,分别为Freundlich,Freundlich,Dubinin-Radushkevich和Freundlich,反映了各种晶形的铝氧化物的表面异质性。修正的Freundlich方程比较不同晶形铝氧化物对菲的相对吸附容量顺序为:MR=10-3MR=10-2MR=0MR=10-1。由此,认为菲在无定形铝氧化物上的吸附是熵驱动的结果。  相似文献

Clay minerals have been shown to reduce the extent and rate of biodegradation of several compounds. Here, we investigated the ability of soil clays to protect proteins from biodegradation: the insecticidal protein from Bacillus thuringiensis subsp. tenebrionis (Btt toxin) and Bovine Serum Albumin (BSA). The two proteins adsorbed in large amounts (up to 0.24 g BSA g−1 clay and 0.74 g Btt toxin g−1 clay) and irreversibly to smectite clay particles from a vertisol. We measured the growth of a soil inoculum in the presence of each of proteins as the sole source of carbon. When clay was present in the medium, microbial growth was directly proportional to the amount of free protein (i.e. nonadsorbed). Hence, the two proteins were unavailable when adsorbed to clay. The clay had little influence on the ability of microorganisms to hydrolyse a soluble substrate. The inhibitory effect of clays on utilisation of BSA and Btt toxin was interpreted as being the result of the adsorption of the proteins to clay, which rendered the proteins unavailable for microbial utilisation.  相似文献

As endocrine-disrupting chemicals, a few frequently used phthalate plasticizers were banned or restricted for use as additives in food in some countries. The interaction mechanisms between three phthalate plasticizers with human serum albumin (HSA) were studied by fluorescence (quenching, synchronous, and three-dimensional), UV-vis absorption, circular dichroism (CD), and Fourier transform infrared (FT-IR) spectroscopy, in combination with molecular modeling under simulative physiological conditions, respectively. The results obtained from fluorescence quenching data revealed that the plasticizers-HSA interaction altered the conformational strcture of HSA. Meanwhile, the alterations of HSA secondary structure in the presence of phthalate plasticizers were investigated. The binding distances for the plasticizers-HSA system were provided by the efficiency of fluorescence resonance energy transfer. Furthermore, the thermodynamic analysis implied that hydrophobic forces were the main interaction for the plasticizers-HSA system, which agreed well with the results from the molecular modeling study.  相似文献
