Potato progenies in a line x tester mating design and the clonal parents were screened for field resistance to potato leafroll virus (PLRV) to determine the heritability of this trait. Twelve advanced potato clones or varieties were crossed as pistillate parents to two pollen testers. The seedling progenies and clonal parents were exposed to aphid-transmitted potato leafroll virus for two growing seasons. Cumulative infection by potato leafroll virus was determined by post-season sero-logical indexing of foliage grown from sprouted tubers after 2 years of exposure. Narrow-sense heritability was estimated from regression of mid-parent on progeny as h = 0.72. This estimate indicates a high level of useabie genetic variance for PLRV resistance in advanced breeding materials. Although variation in resistance to PLRV appears to be a quantitative trait in susceptible and moderately resistant clones, per-formance of the most resistant parents suggests that genes with major effects may be present. These results are similar to the conclusions of other researchers who found one or two genes controlling the pheno-types of extreme resistance, resistance to infection, or suppression of virus titre.  相似文献

Summary Four clones of the Chilota Potato Collection from the germplasm bank of the Universidad Austral de Chile, with potential combined resistance to PVX, PVY and PVS, were mechanically inoculated with these viruses in order to demonstrate wether they were truly resistant or not. Virus coat protein in the inoculated clones was determined by means of NCM-ELISA, DAS-ELISA, Western Blot and NASH.Two of the four inoculated clones were resistant to both PVY and PVS. Dry rot is a postharvest fungal disease affecting potato (Solanum tuberosum L.) tubers. The disease, caused by several species of Fusarium such as F. solani var. coeruleum, F. sambucinum, F. oxysporum, F. avenaceum, F. culmorum, results in significant yield losses. Some Fusarium species associated with the disease produce toxins, which are implicated in mycotoxicoses of humans and animals. The pathogens cannot penetrate the tuber through the lenticels or in the absence of wounds and cause infection only if the potato skin is ruptured. The seed tuber is considered as the main source of inoculum although soil infested by Fusarium spp. also constitutes a source of inoculum. Control of the disease, once provided by the fungicide thiabendazole, is now difficult due to the appearance of thiabendazole-resistant strains and the lack of potato cultivars with high levels of resistance to dry rot. An integrated disease management program including detection strategies, appropriate cultural practices and storage conditions (including a wound healing period) along with the use of synthetic chemical fungicides as seed tuber and/or postharvest treatment is recommended to reduce incidence and severity of dry rot. Recent studies also indicate the possibility that generally recognized as safe (GRAS) compounds and microbial antagonists could eventually be integrated into dry rot management strategies.  相似文献

A considerable number of highly diverse species exist in genus Solanum. Because they can adapt to a broad range of habitats, potato wild relatives are promising sources of desirable agricultural traits. Potato taxonomy is quite complex because of introgression, interspecific hybridization, auto- and allopolyploidy, sexual compatibility among many species, a mixture of sexual and asexual reproduction, possible recent species divergence, phenotypic plasticity, and the consequent high morphological similarity among species. Recent researchers using molecular tools have contributed to the identification of genes controlling several types of resistance as well as to the revision of taxonomical relationships among potato species. Historically, primitive forms of cultivated potato and its wild relatives have been used in breeding programs and there is still an enormous and unimaginable potential for discovering desirable characteristics, particularly in wild species Different methods have been developed to incorporate useful alleles from these wild species into the improved cultivars. Potato germplasm comprising of useful alleles for different breeding objectives is preserved in various gene banks worldwide. These materials, with their invaluable information, are accessible for research and breeding purposes. Precise identification of species base on the new taxonomy is essential for effective use of the germplasm collection.  相似文献

The cultivated potato is a major crop worldwide. It is a high input crop with complex quality requirements at harvest and during storage. Potato breeders are fortunate to have access to a very diverse and accessible germplasm resource. Wild Solanum relatives provide genetic diversity as well as genes for valuable production and quality traits. In most cases, crossing success can be predicted based on endosperm balance number (EBN), or effective ploidy, of the parents. Crossing barriers between most wild species and the cultivated potato are the consequence of differences in EBN and can be easily overcome using ploidy manipulations and bridge crosses. The most common ploidy manipulations include haploid extraction to reduce EBN and 2n gamete production to increase EBN. Additional methods to produce fertile interspecific hybrids include mentor pollination, embryo rescue, hormone treatments, reciprocal crosses, selection of cross‐compatible genotypes and somatic fusion. Knowledge of crossing barriers and mechanisms to overcome them allows potato breeders access to the rich gene pool in the genus Solanum.  相似文献

Flower formation of most sweet potato clones in the collection at Manokwari was poor and when seed for breeding purposes was desired flowering had to be stimulated accordingly.Of various methods applied to stimulate the flowering some proved to be satisfactory. Summary In two experiments, using different testing methods, the number of newly formed cysts was determined on nine potato genotypes with resistance from various sources. Ten potato cyst nematode (PCN) populations were used in these experiments. Rank correlation between numbers of cysts over potato genotype-PCN population combinations for both experiments was high (rs = 0.90). Dendrograms for PCN populations and potato genotypes were constructed, based on a simultaneous hierarchical clustering procedure for potato genotype-PCN population interaction terms. Several virulence groups could be identified within Globodera rostochiensis as well as within G. pallida. Host genotypes, derived from the same sources of resistance, were clustered in different resistance groups.  相似文献

以10个不同类型的甘薯品种为试验材料,采用随机区组设计,以纯作为对照,研究小麦与甘薯行数比在3∶1和2∶1套作模式下甘薯地上部农艺性状和2种作物鲜产的变化。结果表明:参试甘薯最长蔓长比纯作减小;分枝数和茎粗因品种差异变化不尽一致;漯薯14号在3∶1模式种植时较纯作增产18.77%,漯薯15号在2∶1套作模式下较纯作增产3.99%,其余甘薯品种及小麦鲜产均不同程度下降;套作兼用型(漯薯11号和漯薯15号)甘薯品种产量更高;麦薯3∶1比2∶1套作模式更具优势。  相似文献

Sweet potato virus disease (SPVD), a result of the co-infection of whitefly transmitted Sweet potato chlorotic stunt virus (genus Crinivirus, family Closteroviridae) and the aphid transmitted Sweet potato feathery mottle virus (genus Potyvirus, family Potyviridae), is the most destructive disease of sweet potato in East Africa. A study was conducted to establish if genotypes identified as resistant or susceptible to SPVD in Kenya could be distinguished using molecular markers. A total of 47 unrelated sweet potato genotypes were selected from germplasm collections and classified into two phenotypic groups as resistant or susceptible to SPVD. Genotype selection was based on disease severity or days to symptom development in plants following graft inoculation. Amplified fragment length polymorphism (AFLP) marker profiles were generated for each individual and used in association studies to identify markers suitable for classifying the two pre-defined phenotypic groups. Analysis of molecular variance showed significant (P < 0.002) variation between the two groups using 206 polymorphic AFLP markers. Discriminant analysis and logistic regression statistical methods were used to select informative markers, and to develop models that would classify the two phenotypic groups. A training set of 30 genotypes consisting of 15 resistant and 15 susceptible were used to develop classification models. The remaining 17 genotypes were used as a test set. Four markers, which gave 100% correct classification of the training set and 94% correct classification of the test set, were selected by both statistical methods.  相似文献

彩色马铃薯较普通栽培马铃薯具有更丰富的营养物质,特别是富含抗氧化物质花青素,是近年来育种家研究的焦点。迄今为止开发的马铃薯SSR引物数量有限,特别是彩薯相关的SSR引物。本研究基于马铃薯全基因组序列利用MISA软件对SSR位点进行了分析。研究发现,在马铃薯全基因组序列中共获得218,997个SSR位点,平均3.39 kb出现一个SSR位点。单核苷酸是主要重复类型,占总SSR的62.05%,其次是二核苷酸和三核苷酸,所占比例为22.39%和13.11%。6种核苷酸类型的重复次数分布在5~746次,以5~10次为主,占总SSR的60.9%。在检测到的全部SSR中,共获得215种基元类型,除六核苷酸重复类型外,其他5种核苷酸重复类型的优势基元均以含有A/T-的基序为主。SSR基序长度主要分布在12~20bp之间,占全部SSR的43.19%。通过Primer5共设计出100对SSR引物,利用彩薯双亲的DNA初步筛选出48对可以扩增出条带的引物,有效扩增率达48%。进一步验证引物的有效性,随机选择F2群体的6个单株进行PCR扩增,筛选出条带清晰稳定,多态性较高的引物26对,平均多态性比率为72....  相似文献

Summary Production of mini-tubers as a source for seed potato was investigated by growing in soil micropropagated plants and micro-tubers produced from micropropagated plants. Cultures of several cultivars were initiated from indexed tubers and multiplied on modified MS medium. Cultures were micropropagated by using a modular system which allowed batch handling. Micropropagated plants produced mini-tubers in glasshouse after 70–115 days of growth in soil. A large proportion of the mini-tubers produced were between 9 and 15 mm diameter. Several factors, e.g., explant number, duration of in vitro culture and genotype influenced mini-tubers production. Micropropagated plants after culture of 86 days or longer produced micro-tubers ca. 2 to 10 mm diameter. Plants, which formed micro-tubers in vitro, produced less number of mini-tubers in soil. Micro-tubers produced 1 to 3 mini-tubers when grown in soil in chain-type paper pots, but produced conventional sized tubers when grown in soil under plastic polytunnel. Mini-tuber number varied widely between potato cultivars; cvs. Bintje and British Queen produced more mini-tubers than the other cultivars. Mini-tubers developed green hard skins when kept in light for 3 weeks, and could be stored in dark at 4° C upto 6 months. In a field trial, small mini-tubers ca. 5–10 mm diameter produced more but smaller tubers than mini-tubers ca. 15–20 mm diameter. The micropropagated plants and the plants grown from mini-tubers were genetically stable, and did not show any morphological aberrations except for one variegated plant among the several thousand produced. Summary Sixty-seven potato varieties/hybrids were grouped in 15 clusters on the basis of D2 values. The clustering pattern was not influenced by the geographic diversity of the varieties. However, a segregation between varieties of the Tuberosum and Andigena type varieties was observed. The exotic potato varieties and also the Indian varieties bred from Tuberosums showed a poor divergence. In contrast, the divergence in the varieties developed from Tuberosum-Andigena crosses was much greater. The inter-cluster distance of such varieties, with respect to Tuberosum and Andigena clusters, appeared to be influenced by the cytoplasm they carried. The varieties with Tuberosum cytoplasm were closer to clusters having Tuberosum varieties and those with Andigena cytoplasm were closer to clusters having Andigena type varieties. The characters least influenced by the selection during the course of evolution of the present day varieties were found to be mainly responsible for adding divergence to the population. A breeding plan involving varieties from different clusters has been outlined.  相似文献

Summary Traditional potato breeding is a laborious process in which, by intercrossing, valuable traits from different parental clones are combined in a progeny genotype. Depending on the availability of genes, molecular techniques can be used to add specific genes to existing cultivars that, although otherwise satisfactory, lack a lew commercially important traits. For virus resistance the gene for the coat protein of a given virus transplanted into the genome of the plant renders the plant resistant to that virus. In conferring such resistance to potato varieties it proved to be possible to preserve their intrinsic properties.  相似文献

Summary Screening sweet potato, Ipomoea batatas, L., for source potentials is more difficult than for sink capacities. Twenty clones were grafted to four tester varieties to screen them for source potentials in relation to sink capacities which are interdependent. Source potential of a clone was measured by its average scion effect when grafted on the four tester varieties. There were significant differences in source potentials among the twenty clones. Seven clones were selected for having high source potentials. Clone TIS 2498 was the best. Degrees of response of source to sink were estimated using the regression method for twenty clones showed significant differences and ranged from b–0.45 to b=2.06. Source potential showed significant relationship (r=0.62**) with response of source to sink. Implication of this approach to sweet potato breeding is discussed.  相似文献

Six potato genotypes including ‘Desiree’ and five breeding clones (2/14, 95C, 125B, 3589, 4056) were grown in the field under well-watered and stress conditions. The stress response of leaf tissue was assessed 8 and 12 weeks after plant emergence (WAE) by measuring canopy temperature and chlorophyll α fluorescence induction kinetics in detached leaves. Under drought-stress conditions, chlorophyll α fluorescence dropped in clones 2/14 and 4056, remained constant in 95C and 125B and increased in clone 3589. The canopy temperature exhibited little difference between drought and irrigated treatments in clones 95C and 125B, and significant effects in the other genotypes. This performance was interpreted in terms of tolerance to drought stress, and the potential use of canopy temperature and chlorophyll fluorescence as tools for drought screening of potato germplasm and segregant generations is discussed. The physiological properties of 95C and 125B highlighted by this study need further investigation and, if confirmed, they could be recombined with other useful agronomic characters by crossing in a breeding programme.  相似文献

The study of quantitative trait’s expression over time helps to understand developmental processes which occur in the course of the growing season. Temperature and other environmental factors play an important role. The dynamics of haulm senescence was observed in a diploid potato mapping population in two consecutive years (2004 and 2005) under field conditions in Finland. The available time series data were used in a smoothed generalized linear model to characterize curves describing the senescence development in terms of its onset, mean and maximum progression rate and inflection point. These characteristics together with the individual time points were used in a Quantitative trait loci (QTL) analysis. Although QTLs occurring early in the senescence process coincided with QTLs for onset of senescence, the analysis of the time points made it difficult to study senescence as a continuous trait. Characteristics estimated from the senescence curve allowed us to study it as a developmental process and provide a meaningful biological interpretation to the results. Stable QTLs in the two experimental years were identified for progression rate and year-specific QTLs were detected for onset of senescence and inflection point. Specific interactions between loci controlling senescence development were also found. Epistatic interaction between QTLs on chromosomes 4, 5 and 7 were detected in 2004 and pleiotopic effects of QTLs on chromosomes 3 and 4 were observed in 2005.  相似文献
