The ability of Holothuria scabra to digest nutrients, such as organic matter (OM), protein and carbohydrate from animal and plant feed ingredients was investigated. Four test feeds prepared by mixing sand with single ingredients from animal sources (shrimp and mussel) and plant sources (diatom and seaweed) were fed to H. scabra to estimate apparent digestibility coefficient (ADC). The total assimilated nutrient (TAN) increased with ADC, whereas ingestion rate (IR) varied slightly among the feeds suggesting that ADC might be a good indicator of nutrient availability to H. scabra. The ADCOM of shrimp and mussel was significantly higher than that diatom and seaweed: 86.2%, 77.1%, 55.1% and 32.3% respectively. ADCprotein was similar for shrimp (88.7%), mussel (84.8%) and diatom (75.2%), but significantly lower in seaweed (34.4%). ADCcarbohydrate was similar in mussel (58.5%) and diatom (58.3%) as well as in seaweed (31.6) and shrimp (28.0%). ADCprotein was relatively higher than ADCcarbohydrate suggesting that H. scabra generally digests more protein than carbohydrate. Furthermore, results indicated that nutrients from animal‐based feeds are more efficiently digested by H. scabra; thus, animal ingredients rich in easily digestible protein could potentially provide an efficiently balanced diet for H. scabra fed with diatom containing high easily digestible carbohydrate.  相似文献

Floating hapas (fine mesh net enclosures) are a cost‐effective ocean nursery system to culture post‐metamorphic Holothuria scabra to release size. The growth of periphyton biofilm on hapas is a natural food source for early juveniles. This study investigated the effects of periphyton quality (i.e. chlorophyll‐a, phaeopigment, total biomass, autotrophic index or AI), water quality (nutrients, chlorophyll‐a) and environmental parameters (temperature, rainfall) on the temporal variation in the growth and survival of early juvenile (~3 mm) H. scabra reared in floating hapas. Five trials where the juveniles were reared for 60 days each in the eutrophic coastal waters of Bolinao, the Philippines were conducted during different months over 2 years. Significant differences in the growth and survival of juveniles among trials were found. Absolute growth rates (AGR) ranged widely (0.01–0.09 g/day). Trials with high AGR of juveniles (0.07–0.09 g/day) during the first 30 days of rearing had significantly higher chlorophyll‐a (chl‐a) in biofilm (15.9–27.5 mg/m2) and lower AI. Conversely, during the subsequent 30 days, trials with high AGR of juveniles (0.06–0.11 g/day) had significantly lower chl‐a and higher AI. Multivariate analyses showed that chl‐a in biofilm, AI and nutrients in the water column are good indicators of periphyton quality and juvenile growth rates in floating hapas. Further, this study validates the expansion of the feeding mode of juveniles from primarily grazing on microalgae, to feeding on detritus and heterotrophs as they grow. Field trials were conducted to determine the effects of size, grow‐out period and site on the growth and survival of Holothuria scabra. Juveniles were reared in sea cages in two sites for 2 months during two different times of the year. Juveniles from Experiment 1 were subsequently grown in sea pens for 8 months in one site. The significantly lower survival particularly of the small‐sized group (<0.1 g) in Pilar is likely due to reduced water and sediment quality because of proximity to fish‐farming activities. The importance of grow‐out period was evident in the significant differences in weight increments in all size groups and biomass in the small‐sized group (P < 0.10) in Lucero during Experiment 2 when water temperature range and incidence of rainfall were higher. The decrease in the average weight of adults during the second half of the grow‐out period may be due to the interplay of the reduction in salinity, onset of sexual maturity and biomass exceeding carrying capacity in the pens. The estimated biomass threshold in the sea cages and pens in Lucero were 300 g m?2 and 380 g m?2 respectively. Implications of the results on the viability of small‐scale pen grow‐out culture are discussed.  相似文献

Experiments were conducted to identify suitable methods for broodstock rearing, induced breeding and grow‐out culture of Holothuria scabra in Sri Lanka. Two hundred and seventy‐two brooders (500–600 g) collected from off Mannar were individually packed in oxygen‐filled polythene bags with and without sea water and transported to a sea cucumber hatchery at Kalpitiya. Lagoon pens, sand‐filled fibreglass tanks and bare tanks were used in triplicates to maintain brooders. Spawning was initiated using air dry, water jet and thermal‐stimulation methods. Hatchery produced juveniles with an average weight of 11 ± 5 g were reared (2 individuals m?2) in lagoon pens, mud ponds and fibreglass tanks in triplicates. The significantly high evisceration rate was observed when brooders were transported without sea water (t‐test, P < 0.05). Brooders maintained in bare tanks showed a significant weight reduction than the brooders in sand‐filled tanks and lagoon pens (anova , P < 0.05, d.f. = 2). Thermal stimulation (ambient temperature ± 3–5°C) was found to be the most successful method of spawning initiation of H. scabra. The mean (±SD) percentage males and females participated for spawning per trial was 9.2 (±10) and 4.6 (±5.6) respectively. On an average, 1.16 millions of eggs (±1.03 SD, n = 5) were obtained per spawning trial. H. scabra juveniles reared in tanks showed significantly lower growth rate than the juveniles in pens and ponds (anova , P < 0.05). Lagoon pens and sand‐filled tanks are suitable to maintain brooders and lagoon pens can be successfully used for mass rearing of juveniles.  相似文献

Holothuria scabra (sandfish) and Babylonia areolata were trialed in two large‐scale co‐culture experiments. Experiment 1 assessed co‐culture in 4 × 400 m2 earthen ponds where Babylonia were cultured in a central pen at a density of 400 individuals/m2 with sandfish occupying the remaining pond space at 1.1 individuals/m2. Sandfish grew from 18.20 ± 6.67 g to 119.03 ± 17.74 g in 92 days and Babylonia (fed trash fish in both experiments) grew from 0.90 ± 0.38 g to 4.93 ± 1.44 g, but Babylonia growth was not increased in co‐culture compared to monoculture. Water and sediment quality varied between co‐culture and monoculture ponds. Neither showed clear improvement due to sandfish culture. Experiment 2 compared non‐segregated sandfish‐Babylonia co‐culture with Babylonia monoculture in 20 m2 concrete raceways. Sandfish were cultured at 2 individuals/m2 and Babylonia at 300 individuals/m2. Sandfish grew up to 1.91 g.day?1 with 100% survival. Babylonia weight gain was significantly greater in co‐culture raceways (3.35 ± 0.64 g over 61 days), which was double that of Babylonia in monoculture. Substrate total N was reduced by 20% in co‐culture compared with monoculture (p = .032). This provisional study of commercial scale sandfish‐Babylonia co‐culture demonstrates culture compatibility, providing a basis for further system development.  相似文献

Stock density is one among the most important factors in aquaculture that directly influences the growth of organisms; however, there is limited information about the effects of stocking density on growth performance of sea cucumber Holothuria tubulosa (Gmelin, 1788) that has a commercial potential and represents a new species for aquaculture. In this study, we investigated the effect of stocking densities on the growth performance of juvenile H. tubulosa (40.3 ± 3.34 g) in laboratory conditions. Stocking densities were selected as 6, 15 and 30 ind m?2 with total biomass 253.3 ± 0.18, 601.2 ± 0.11 and 1201.4 ± 0.15 g m?2 respectively. We monitored the growth by wet weights and calculated the growth performance through weight gain, growth rate, specific growth rate, relative weight gain, coefficients of variation and survival rate during 8 weeks of research period. We found that individual mean weight gain in 6 ind m?2 group was approximately 29.53 g while it was 3.03 g and ?4.36 g for 15 ind and 30 ind m?2 groups respectively. Results have shown that the specific growth rate tends to decrease as the number of individuals in unit area increases. The final mean weight, growth rate, specific growth rate, relative weight gain and coefficients of variation were significantly different among treatment groups. In conclusion, a density of 6 ind m?2 is recommended for stocking juvenile H. tubulosa under rearing conditions. Stocking density of 15 ind m?2 is not favourable for long term as the growth rate is negligible where 30 ind m?2 should be avoided in tank‐based rearing units.  相似文献

Various research projects in Europe and North Africa have recently intended to breed temperate holothurians to alleviate fishing pressure on natural populations. However, to date little is known about the nutritional requirements of East Atlantic and Mediterranean species. In this study, we propose a “natural population”‐oriented approach to characterize food sources, digestive efficiency and resources allocation based on the composition of pigments and fatty acids (FA) in gut contents and tissues (muscles, gonads and digestive tract walls) of wild individuals of the species Holothuria (Panningothuria) forskali (Delle Chiaje, 1823) sampled in Brittany (France). Our study reveals that neither green nor red algae enter the diet of H. forskali in spring and that the only fresh vegetal material found in gut contents is brown algae (very likely diatoms). The high nutritional quality of gut contents however contrasts with the detrital nature of the ingested food sources, suggesting that a trophic upgrading of organic matter occurs before digestion. In addition, unusual FA (i.e. only present in a few groups of living species) such as long‐chain monounsaturated FA (especially the FA 23:1ω9) were found in large proportions in muscles and gonad and their effect on sea cucumber fitness needs further investigation.  相似文献

Stable isotope analyses were applied to explore the relative dietary nitrogen contributions from fish meal and pea meal (Pisum sativum) to muscle tissue of Pacific white shrimp postlarvae (141 ± 31 mg) fed low protein diets having different proportions of both ingredients as the sole dietary protein sources. A negative control diet was formulated to contain 100% pea meal and six more isoproteic diets to have decreasing levels of pea meal‐derived nitrogen: 95%, 85%, 70%, 55%, 40% and 0% of the initial level. Growth rates were negatively correlated to dietary pea protein inclusion due to progressive essential amino acid deficiencies (sulphur amino acids, threonine, lysine, histidine). The nitrogen turnover rate significantly increased in muscle tissue of shrimps fed diets having high levels of pea meal; however, contrary to observations from a previous study using soy protein, the relative contributions of dietary nitrogen from pea meal to shrimp muscle tissue were equal or higher than expected contributions established by the dietary formulations. Results highlight the effectiveness of stable isotope analysis in assessing the nutritional contributions of alternative ingredients for aquaculture feeds and the potential suitability of pea as a source of protein (provided the diets are nutritionally balanced).  相似文献

The culture of snakehead fish (Channa striata and Channa micropeltes) in Vietnam is limited, and snakehead culture has been banned in Cambodia, because traditional practices include capture of fingerlings from the wild as seed, as well as capture of small‐size (also known as trash fish or low‐value) fish. As hatchery breeding technology has improved, we investigated the optimal weaning practices for these two species. Both laboratory experiments and farm trials were conducted. For C. striata, the optimal weaning procedure is to begin at 17 days after hatch (dah) and wean the fish at 10% replacement of live feed with formulated feed per day. However, for C. micropeltes, the optimal procedure is to wait until 40 dah to begin weaning and then to wean the fish with a 10% replacement of live feed with formulated feed every 3 days. These results should enable farmers to domesticate snakehead culture in Vietnam and Cambodia and eliminate reliance on fish captured from the wild as both seed and feed.  相似文献

Anaesthetic efficacy of eugenol was investigated on iridescent shark, Pangasius hypophthalmus. Fish (2, 5, 10 and 20 g) subjected to 20–200 mg L?1 eugenol and behavioural response as well as induction and recovery times were recorded. Induction and recovery times were significantly affected by eugenol concentration as well as fish weight (< 0.05). Generally, 27–300 s after exposure to 20–200 mg L?1 eugenol, iridescent sharks reached stage 3 anaesthesia (suitable for general handling). Fish entered stage 4 anaesthesia (suitable for surgery and blood sampling) over 54–710 s exposure to such concentrations. Recovery time was 109–600 s in all weight classes as well as eugenol concentrations. Mortality (44–100%) was only observed in 2 g fish when subjected to 110–170 mg L?1 eugenol. This study, for the first time, showed behavioural response of iridescent shark to anaesthesia as well as effectiveness of eugenol as anaesthetic in this important aquaculture‐ornamental species. According to the models obtained in this study, minimum eugenol concentrations to induce anaesthesia over less than 3 min were 53.8–81.5 mg L?1 in 2–20 g fish. Likewise, maximum eugenol concentrations in which fish recovered over less than 5 min were 65.9–105.8 mg L?1 in 2–20 g fish.  相似文献

Condition index (CI) is a widely used parameter in ecological and physiological studies. For molluscan bivalves, no universal consensus exists for CI calculation. A literature review revealed that a total of 19 methods have been used for CI calculation in molluscan bivalves. To evaluate the CI calculation in northern quahogs Mercenaria mercenaria (also called hard clams), market‐sized clams at 12–14 months old (n = 300 from three populations) were collected for gravimetrical and volumetric measurements. Significant correlations were identified among body sizes (body length, height and width), body weights (total weight, wet meat weight, dry meat weight, wet shell weight, dry shell weight) and body volume (p < .0001, R values ≥ .800). Referencing previously used CI equations for molluscan bivalves, a total of 18 equations were evaluated the suitability, considering non‐lethal measurements, for northern quahogs through correlation analysis of the CIs from different equations. With equations constructed as the percentages of dry or wet meat weight out of total weight, dry shell weight, dry shell plus meat weight, body volume, body length*height*width, shell length or cubic of shell length, CIs were significantly correlated, meaning that these equations were exchangeable. Specifically, three equations with non‐lethal measurements (percentages of total body weight out of total body volume, body length*height*width or cubic of body length) were evaluated effective. Overall, this publication provided a review for CI calculations in molluscan bivalves and evaluated the CI equations for northern quahogs.  相似文献

Comparative evaluation of mahua (Bassia latifolia) oil cake (MOC) and castor bean (Ricinus communis) seed (CBS) was tested as piscicides for tilapia (Oreochromis mossambicus) and panchax (Aplocheilus panchax), and residual toxicity test was done on Labeo bata. Delayed response of CBS compared with MOC was evident as first and total mortality of tilapia was encountered at 4th and 10th hour of application, against 26 hr and 42 hr in panchax respectively. Susceptibility of the test fish to the piscicides was highly variable as tilapia was more susceptible than panchax under both the piscicides tested especially in CBS. Residual toxicity decreased polynomialy over time (R2 ≥ 0.99) and reduced to below 10% on day 13–14 in both MOC and CBS treatments. Both MOC and CBS contributed positively to the nutrient pool of water in which CBS provided more nitrogen compared with MOC. N: P ratio of water in CBS treatment increased exponentially to more than 18 indicating phosphorus limitation.  相似文献

Phosphoenolpyruvate carboxykinase (PEPCK) is known to exist as mitochondrial (PEPCK‐M) and cytosolic isoforms (PEPCK‐C). They are involved in glucose production and play vital roles in regulating glucose homeostasis. This study aimed to (1) clone and characterize PEPCK‐C cDNA from golden pompano fish (Trachinotus ovatus), which are known to have poor utilization of dietary carbohydrates, and (2) analyze the regulation of its expression by varying dietary carbohydrate‐to‐lipid ratios and nutritional status. A full length golden pompano PEPCK‐C cDNA fragment of 2,652 bp was cloned, which contains an open reading frame of 1,875 bp encoding 624 amino acids and shows a high homology to cobia (Rachycentron canadum) PEPCK‐C sequence (92% similarity). The analysis of tissue distributions of PEPCK‐C mRNA showed that a high abundance of PEPCK‐C was expressed in golden pompano liver, followed by kidney; while lower expression levels were found in the heart and intestine. In liver, PEPCK is under the regulation of dietary composition and nutritional status at the enzymatic and molecular levels. Our results indicate that PEPCK‐C plays a modulating role in the adaptation of hepatic gluconeogenesis to different nutrient conditions. Thus, the absence of molecular inhibition of gluconeogenic PEPCK gene expression in rainbow trout (Oncorhynchus mykiss) and gilthead sea bream (Sparus aurata), which can at least partially account for their poor utilization of dietary carbohydrate, is specific to these species and is not observed in golden pompano. Further studies that examine, at the enzymatic and molecular levels, other key enzymes involved in hepatic glucose metabolism are necessary to understand the poor utilization of carbohydrates in golden pompano.  相似文献

Salmonid fishes may reside within or migrate between stream and lake habitats, or undergo anadromous migrations between freshwater and the ocean. While the degree of anadromy of salmonids has been thoroughly compared, no analogous review has examined the degree of lake use. To assess the extent of reliance on lake habitat in this family, we considered 16 species of salmon, trout and charr from the genera Oncorhynchus, Salmo and Salvelinus, comparing their (a) use of lakes as spawning habitat, (b) rearing strategies in lakes, and (c) occurrence and diversity of lacustrine trophic polymorphism. In identifying the primary life‐history patterns of each species and exploring the lesser‐known lacustrine behaviours, we found that the extent of reliance on lakes exhibits a negative association with the degree of anadromy. Oncorhynchus rely least on lakes, Salmo to an intermediate level and Salvelinus the most, opposite of the general prevalence of anadromy among these genera. Lakes are critical to adfluvial and lake‐resident salmonids, but they also support anadromous and fluvial life histories by providing spawning, rearing, overwintering and/or summer refuge habitat. Adfluviality, although a non‐anadromous life history, consists of similar migration‐related traits and behaviours as anadromy, including the parr–smolt transformation, sex‐biased patterns of migration and residency, and the presence of precocious males. Lakes support life‐history variants, reproductive ecotypes and trophic morphs unique to lacustrine habitat. Therefore, conservation of salmonids is dependent on maintaining the diversity and quality of their habitats, including lakes.  相似文献

Members of the Iridoviridae family have been considered as aetiological agents of iridovirus diseases, causing fish mortalities and economic losses all over the world. Virus identification based on candidate gene sequencing is faster, more accurate and more reliable than other traditional phenotype methodologies. Iridoviridae viruses are covered by a protein shell (capsid) encoded by the important candidate gene, major capsid protein (MCP). In this study, we investigated the potential of the MCP gene for use in the diagnosis and identification of infections caused Megalocytivirus of the Iridoviridae family. We selected data of 66 Iridoviridae family isolates (53 strains of Megalocytivirus, eight strains of iridoviruses and five strains of Ranavirus) infecting various species of fish distributed all over the world. A total of 53 strains of Megalocytivirus were used for designing the complete primer sets for identifying the most hypervariable region of the MCP gene. Further, our in silico analysis of 102 sequences of related and unrelated viruses reconfirms that primer sets could identify strains more specifically and offers a useful and fast alternative for routine clinical laboratory testing. Our findings suggest that phenotype observation along with diagnosis using universal primer sets can help detect infection or carriers at an early stage.  相似文献

This study was conducted to evaluate nutritional values of selected commercial fish meals (FM) with two experiments in diets of turbot. In experiment I, five diets based on two Peruvian anchovy meals (one is a flame‐dried FM: Peru‐1 and the other steam‐dried FM: Peru‐2), a Danish FM (Danish LT), a domestic anchovy LT meal (Domestic LT) and an Alaskan white FM (WFM) as major protein source were formulated to contain 56% protein. The diets were fed to fish with an initial average weight of 119.4 ± 0.1 g for 9 weeks. Growth rate, feed utilization, whole body and visceral compositions, nitrogen, lipid and energy balances did not significantly change in Experiment 1 among the treatments. In Experiment 2, four diets based on three FMs (Domestic LT, Danish LT and Peru‐1) and their mixture (Mix FM) were formulated to contain 50% protein and fed to triplicated groups of fish with an average initial weight of 51.4 ± 0.1 g for 6 weeks. Once again, there were no significant differences in growth rate and feed consumption among the treatments. However, fish on Mix FM diet had significantly better FCR and PER than the others. The results of two experiment indicated that Danish LT and Domestic LT were better in terms of phosphorus (P) retention and losses than two Peruvian FMs and WFM, suggesting that although different sources of FMs were comparable in terms of growth and feed utilization, they could lead to substantial differences in environmental impacts.  相似文献

Muscle proximate composition, amino acid profiles, fatty acid compositions and selected minerals of wild, pond and factory cultured Japanese flounder adults were compared in this study to elucidate their nutritive values and dietary nutrition requirements. Wild and pond cultured Japanese flounder flesh had higher crude proteins, but crude lipid contents of factory cultured fish were 3.8~4.0 fold greater. Major amino acids in Japanese flounder were aspartic acid, glutamic acid and lysine. Wild fish had higher levels of total amino acids, essential amino acids, nonessential amino acids and half‐essential amino acids and the contents in pond cultured samples were similar. Arachidonic acids (ARA), eicosapentaenoic acids (EPA), docosahexaenoic acids (DHA), polyunsaturated fatty acids (PUFA), DHA + EPA and n–3 PUFA in wild Japanese flounder were significantly higher than that in factory cultured fish, whereas fatty acid levels were close between pond cultured and wild fish. Japanese flounder were rich in zinc, iron and selenium, but poor in copper, chromium and nickel. Results indicated wild Japanese flounder had higher nutritional value and better meat quality, but the nutritional compositions of pond cultured fish were close to wild Japanese flounder, which suggested that pond culture of Japanese flounder offers broad application prospects.  相似文献

The experiment was conducted with three biofloc treatments and one control in triplicate in 500 L capacity indoor tanks. Biofloc tanks, filled with 350 L of water, were fed with sugarcane molasses (BFTS), tapioca flour (BFTT), wheat flour (BFTW) and clean water as control without biofloc and allowed to stand for 30 days. The postlarvae of Litopenaeus vannamei (Boone, 1931) with an Average body weight of 0.15 ± 0.02 g were stocked at the rate of 130 PL m?2 and cultured for a period of 60 days fed with pelleted feed at the rate of 1.5% of biomass. The total suspended solids (TSS) level was maintained at around 500 mg L?1 in BFT tanks. The addition of carbohydrate significantly reduced the total ammonia‐N (TAN), nitrite‐N and nitrate‐N in water and it significantly increased the total heterotrophic bacteria (THB) population in the biofloc treatments. There was a significant difference in the final average body weight (8.49 ± 0.09 g) in the wheat flour treatment (BFTW) than those treatment and control group of the shrimp. Survival of the shrimps was not affected by the treatments and ranged between 82.02% and 90.3%. The proximate and chemical composition of biofloc and proximate composition of the shrimp was significantly different between the biofloc treatments and control. Tintinids, ciliates, copepods, cyanobacteria and nematodes were identified in all the biofloc treatments, nematodes being the most dominant group of organisms in the biofloc. It could be concluded that the use of wheat flour (BFTW) effectively enhanced the biofloc production and contributed towards better water quality which resulted in higher production of shrimp.  相似文献

Sanguinarine (SA), with antimicrobial and antiparasitic activities against fish pathogens, exhibits great potential commercial use in aquaculture. However, little information on pharmacokinetics of SA restricts further application in aquaculture. In this study, pharmacokinetics of SA in common carp (Cyprinus carpio) following a single intraperitoneal administration [10 mg kg?1 BW (body weight)] was evaluated by high‐performance liquid chromatography (HPLC). The peak concentration (Cmax) of SA in kidney was 11.8 μg g?1, which was higher than in other tissues and plasma. The terminal half‐life in fish tissue and plasma was as follows: 42.3 h (kidney) > 37.2 h (liver) > 20.1 h (gill) > 18.8 h (muscle) > 10.9 h (spleen) > 10.0 h (plasma). Additionally, we determined the bacterial loads in tissues of common carp infected with Aeromonas hydrophila after i.p. administration of SA at 0, 5, 10 and 20 mg kg?1 BW. The results showed that i.p. administration of SA at 10 mg kg?1 BW significantly enhanced antibacterial efficacy against A. hydrophila, where the antibacterial ratio in the gill, kidney, spleen and liver on day 5 was 95.13%, 93.33%, 90.09% and 92.82%, respectively. Overall, these results suggested the potential of SA to treat A. hydrophila infection in common carp farming industry.  相似文献
