The present study was conducted to evaluate the benefits of indeterminate growth habit in breeding to improve yield potential of Japanese soybean varieties, which exclusively have determinate growth habit. Two populations of recombinant inbred lines (RILs) derived from crosses between determinate Japanese cultivars and indeterminate US cultivars were grown in Akita and Kyoto, and seed weight per plant (SW) and its components were compared between indeterminate and determinate RILs. The difference of SW between the two growth habits in RILs varied depending on maturation time. The SW of early indeterminate lines was significantly higher than that of early determinate ones in Akita, but not in Kyoto. Among yield components, the number of seeds per pod was constantly larger in indeterminate lines than that in determinate ones irrespective of maturation time. The number of seeds per plant and the number of pods per plant of the indeterminate lines were greater than those of the determinate lines in early maturation in Akita. These results suggest that the indeterminate growth habit is an advantageous characteristic in breeding for high yield of early maturing soybean varieties in the Tohoku region.  相似文献

An investigation was undertaken to develop new stable thermo‐sensitive genic male sterility (TGMS) rice lines in intermated progenies (IMPs) of TGMS lines by using an anther clearing technique. The results indicate that both pre‐ and post‐meiotic genetic systems operate during anther development for the expression of sterility in TGMS lines. In all the TGMS lines, sterile anthers were small with empty pollen grains of irregular shape, except for TS 16, which showed pollen‐free anthers. This indicates that the sensitive stage of TS 16 is around stage IV (stamen and pistil primordia) of panicle development. Distinct differences were observed between sterile and fertile phases with respect to anther size, shape and colour of the pollen grains in TS 18 and TS 29. The pollen grains at the sterile phase were small and irregular in shape while in the fertile phase they were plump and larger with a yellow colour, establishing that the occurrence of sterility in TS 18 and TS 29 is post‐meiotic. Three distinct classes of pollen fertility percentage viz. <20%, 50‐70% and >90% were observed in IMPs. Anther clearing in IMPs showed distinct developmental patterns of pollen production with respect to distinct classes of pollen fertility. Less than 20% pollen fertility was observed in hybrids such as TS 15 × TS 16, TS 15 × Co 47, TS 18 × TS 16 and TS 18 × Co 47 which hold promise for developing new TGMS lines with a good plant type and acceptable quality.  相似文献

Microspore response of three- way cross maize hybrid genotype 3AL/95 (Zea mays L.) was studied under simplified isolation and culture conditions. Fertile plant production was achieved through abundant plant regeneration. As a total, microspores of 160 tassels were inoculated and five sustainable microspore derived callus cultures (SMC) were obtained. Hybrid seeds (ML SC), which were produced by crossing of regenerates from two SMCs, gave rise to subsequent vigorous and fertile progeny. The response of the 3AL/95 and ML SC microspores was studied in three liquid culture media in order to improve the early viability of microspores. Them N6M medium provided better survival of cultured microspores (p = 5%) than the ppN6M/89 and the YPM-G media. The pH 5.8 in mN6M medium revealed significant increase (p = 1%) in microspore viability as compared to pH 3.0. The ML SC microspores showed higher viability(30%) on the first day of culture in the mN6M than those of the3AL/95 (19%) but without improved rate of callus formation and plant regeneration. Tobacco transgenic lines belonging to four cultivars modified with three genetic constructions providing the resistance to Potato virus Y (PVY) had been described as regards their agronomic and biological traits. The constructions included viral genes: the coat protein gene of Lettuce mosaic virus (LMV) and the replicase gene of PVY used in sense and antisense orientation. Evaluation of the chosen farm traits of the transgenic tobacco lines was performed by comparing them with their nontransgenic equivalents. The traits such as growth and development rate, height of stalks, leaf area, morphology of plants, course of meiosis and pollen viability were described. The cured leaves were analysed as regards to their physical traits and chemical composition. The individual transgenic lines differed from control cultivars to a little extent. Only insignificant changes, mainly in the quality of raw material were observed. Genetic modification did not cause any changes in the course of meiosis and pollen viability.  相似文献

The main objective of the present study was to identify mitochondrial DNA based marker, which can distinguish male sterile and fertile counterparts of the cytoplasmic male sterile (CMS) lines used in production of rice hybrids. Amplified fragment length polymorphism (AFLP) analysis in CMS lines: IR58025A & IR62829A and their respective maintainers: IR58025B & IR62829B identified a polymorphic DNA fragment of about 510 bp size that was present in both CMS (A) and absent in their maintainer (B) lines. Sequencing followed by database analysis of the polymorphic fragment indicated about 97% similarity with mitochondrial NADH gene subunits of rice, maize and wheat. Based on the variable sequence regions, a site specific primer pair (BF-STS-401) was designed. PCR analysis showed that BF-STS-401 could amplify a strong band of 464 bp size in CMS and a faint band of the same size in maintainer line. To act as a positive control and avoid possible errors in PCR, BF-STS-401 was multiplexed with a new primer pair (BF-STS-402), derived from mitochondrial atp9 subunit of rice, producing monomorphic amplification indiscriminately in both CMS and maintainer lines. Both the primer pairs in combination clearly differentiated CMS lines from their corresponding maintainer lines. This primer combination was validated in a set of diverse genotypes consisting of different sources of CMS lines, restorer lines, hybrids, varieties and mixed samples from private seed companies. Our results suggested that the multiplex primer pairs developed in this study can be effectively utilized to assess the genetic purity in commercial seed lots of CMS lines and hybrids of rice.  相似文献

Elymus tsukushiense Honda (syn. Roegneria kamoji C. Koch) (2n = 6x = 42, StsStsHtsHtsYtsYts) is a hexaploid species, distantly related to bread wheat Triticum aestivum L. em Thell (2n = 6x = 42, AABBDD). Apart from the delineation of evolutionary relationships, this species is a potential source of resistance to scab, a devastating disease of wheat caused by Fusarium graminearum Schw. A standard C-banded karyotype was established identifying all 21 chromosome pairs of E. tsukushiense. By using C-banding and genomic in situ hybridization analyses, three wheat-E. tsukushiense chromosome addition lines, one ditelosomic addition line, and one disomic substitution line were identified in BC2 progenies from wheat × E. tsukushiense hybrids. Twenty DNA markers specific for the seven homoeologous groups of the Triticeae were used to determine the homoeology of the added E. tsukushiense chromosomes. The E. tsukushiense chromosomes in the addition lines NAU702, NAU703, and NAU701 were identified as belonging to homoeologous groups 1, 3, and 5, and thus, were designated as 1Ets#1, 3Ets#1, and 5Ets#1, respectively. NAU751 was identified as a disomic substitution line with chromosome 3A of wheat replaced by chromosome 3Ets#1. Line NAU702 has a high level of resistance to scab and will be used in chromosomal engineering and development of improved wheat germplasm for scab resistance breeding. Summary The difficulties in developing inter-varietal chromosome substitution lines in wheat are reviewed. The use of genetical, cytological and molecular markers is suggested as a way of overcoming them. These difficulties and the use of markers, as well as the need to develop duplicate lines to detect background variation, are described using the development and analysis of the Cappelle-Desprez (Bezostaya 1) chromosome substitution set as an example. The effects of substituting Bezostaya 1 chromosomes on final plant height and adult-plant resistance to yellow rust are reported. The large number of aneuploids and substitution lines available in wheat provides a tremendous, international resource, which should be exploited in the future.  相似文献

Wild-type Brassica oleracea L. have matte blue-green leaves caused by an interaction between leaf pigmentation and a waxy bloom coating the surface. Glossy mutants have reduced and/or altered epicuticular wax giving the leaves a shiny green appearance and have been identified in most B. oleracea crop varieties, including cauliflower, kale, broccoli, Brussels sprouts, cabbage, and collard. The genetic basis of glossy mutants has not been studied in B. oleracea. Glossiness can confer resistance to multiple herbivores mediated by modification of herbivore feeding behavior and foraging efficiency of their predators. The USDA-ARS-U.S. Vegetable Laboratory released two pairs of near-isogenic broccoli lines (NILs) that visibly differ only for glossiness, providing ideal germplasm for the genetic study of epicuticular wax in B. oleracea. Genotyping-by-sequencing resulted in hundreds of polymorphisms between each pair of NILs. Polymorphisms were identified in or near three different wax synthesis genes suggesting the two glossy mutants were caused by alteration of different steps of the pathway.  相似文献

In maize (Zea mays L.) breeding programs, selection among and within segregating progenies is based mainly on indirect selection criteria. A better understanding of the environment influence on physiological attributes of maize inbred lines is important to the identification and selection of superior inbred lines as well as to successful hybrid seed production. In this study, the size and form of genotype (G) and genotype × environment (G × E) interaction effects for plant grain yield (PGY) and several physiological attributes were examined for 12 maize inbred lines grown in four managed environments, represented by two seasons (Y) and two nitrogen levels. Mixed model analysis revealed that the G effect was relatively high for attributes related to light capture, phenology, early biomass production, and numerical components of PGY. The G × E interaction effect explained most of the variability for PGY, harvest index (HI), and biomass production at maturity. Three-mode principal component analysis allowed us to: (1) describe the associations among multiple attributes across environments, (2) reveal the form of the main patterns of G × E interaction, (3) establish the importance of the genotype × year (G × Y) interaction for kernel number, HI, and biomass at maturity in determining PGY, (4) identify promising genotypes of high-PGY across environments, and (5) detect genotypes of similar response patterns for PGY but with a contrasting relative behavior for other attributes, which may permit the simultaneous selection for grain yield and desired secondary traits. Such selection results would contribute greatly in the identification of superior inbreds than selecting for grain yield alone.

Summary A selected group of 42 guayule (Parthenium argentatum Gray) lines were evaluated on the basis of various morphological characteristics and compared to a total population of 234 lines. The objective of this study was to determine the amount of variation that existed both within and among guayule lines over a 2-year period. The parental lines all reproduced apomictically, and as expected, many lines exhibited a high degree of uniformity for every measured character. However, many other lines were extremely variable for varying numbers of characters. This appears to be due to varying degrees and combinations of apomeiosis, thus establishing a long-term source of genetic variation. The high amount of variability existing among and within these guayule lines indicates that significant progress by selection for both rubber percentage and yield is feasible.  相似文献

Reciprocal hybridization between four self-incompatible lines of Brassica napus: 271, 181, 184 and ‘White Flower’, revealed incompatibility. The reciprocal F1s obtained by bud pollination showed self-incompatible reactions, and no segregation for self-incompatibility was observed in all the reciprocal F2 populations, indicating that lines 271, 181, 184 and ‘White Flower’ were genetically identical with regard to self-incompatibility. Observations of self-incompatibility in 17 hybrids from crosses between line 271 and 17 varieties of B. napus showed 10 of the F1 hybrids to be self-compatible, while four were partially self-compatible and three were self-incompatible. Genetic analysis based on F2 and BC1 populations from five self-compatible F1 hybrids and two self-incompatible F1 hybrids suggested the existence of at least two loci controlling the self-incompatibility of line 271: one is the S locus, with dominant and recessive relationships between the S alleles, and the other is the suppressor (sp) of the S locus. The sp locus is genetically different from the S locus, and also shows dominant and recessive relationships between the sp alleles.  相似文献

Early-maturity helps chickpea to avoid terminal heat and drought and increases its adaptation especially in the sub-tropics. Breeding for early-maturing, high-yielding and broad-based cultivars requires diverse sources of early-maturity. Twenty-eight early-maturing chickpea germplasm lines representing wide geographical diversity were identified using core collection approach and evaluated with four control cultivars in five environments for 7 qualitative and 16 quantitative traits at ICRISAT Center, Patancheru, India. Significant genotypic variance was observed for days to flowering and maturity in all the environments indicating scope for selection. Genotypes × environment interactions were significant for days to flowering and maturity and eight other agronomic traits. ICC 16641, ICC 16644, ICC 11040, ICC 11180, and ICC 12424 were very early-maturing, similar to or earlier than control cultivars Harigantars and ICCV 2. The early-maturing accessions produced on average 22.8% more seed yield than the mean of four control cultivars in the test environments. ICC 14648, ICC 16641 and ICC 16644 had higher 100-seed weight than control cultivars, Annigeri and ICCV 2. Cluster analysis delineated three clusters, which differed significantly for all the traits. First cluster comprised three controls, ICCV 96029, Harigantars, ICCV 2 and two germplasm lines, ICC 16644 and ICC 16641, second cluster comprised 13 germplasm lines and control cultivar Annigeri, and third cluster comprised 13 germplasm lines. Maturity was main basis of delineation of the first cluster from others. Plot yield and its associated traits were the main basis for delineation of the second cluster from the others. Identification of these diverse early-maturing lines would be useful in breeding broad-based, early-maturing and high-yielding cultivars.  相似文献

利用小麦-黑麦异代换系H-13(1R/1D), H-24(5R/5A),H-33(6R/6A)与中国春-杀配子染色体二体异附加系CS-DA2C(来自山羊草Ae. cylindrica),CS-DA3C(来自山羊草Ae. triuncialis)配置杂交组合,对杀配子染色体的作用进行了研究。结果表明,杀配子染色体3C对H-13和H-24都是致死的,即有着完全的杀配子作用,但对H-33的杀配子作用则是不完全的,有着接近正常的结实率,说明不同的小麦-黑麦异代换系遗传背景对杀配子作用有很大影响,可能在品系H-33中存在对2C染色体杀配子作用的抑制基因。在以品系H-33为母本时,2个杀配子附加系F1的自交结实率差异显著,说明在相同的小麦-黑麦异代换系遗传背景下,染色体2C与3C的杀配子作用是不同的,在这2个杀配子染色体上可能带有不同的杀配子基因?这些研究结果为进一步创制小麦-黑麦易位系奠定了基础。  相似文献

In this study, doubled haploid lines generated from the durum wheat varieties, selections from Middle Anatolian landraces, ‘Çakmak-79’, ‘Berkmen-469’ and ‘Kunduru-1149’ (Savaskan et al., 1997), were analyzed using ten highly polymorphic microsatellite markers for genotyping and evaluation of genetic relationships between and within the doubled-haploid (DH) lines. The average PIC value was found to be 0.531. Populations of doubled-haploid lines of landrace selected cultivars ‘Çakmak-79’ and ‘Kunduru-1149’, were the two most distant populations with (δ μ)2 = 1.42. ‘Berkmen-469’ x ‘Çakmak-79’ and ‘Berkmen-469’ x ‘Kunduru-1149’ yielded similar genetic distances, (δμ)2 of 0.84 and 0.85, respectively. In addition, the genetic relationship between the progenitors of the DH lines together with other durum wheat varieties was analyzed. A meaningful relationship was obtained based on available pedigree information on the cultivars.  相似文献

Seeds from the aluminum sensitive cultivar ‘Anahuac’ were treated by gamma-ray radiation. Twenty eight selected Al+3 tolerant mutants were compared to the original Anahuac and the tolerant wheat cultivars IAC-24 and IAC-60 from 1994 to 1996 in acid (Capão Bonito) and limed (Monte Alegre do Sul and Tatui) soil field trials. Grain yield and agronomic characteristics were analyzed. Twenty six mutant lines yielded higher than the sensitive Anahuac cultivar in the acid soils of Capão Bonito. Under limed soil conditions, the mutants had a similar yield to the original sensitive cultivar. The majority of the mutants were similar in yield to the tolerant cultivars IAC-24 and IAC-60 under both conditions. Some of the mutants showed altered agronomic characteristics but even in these cases this will not limit its utilization. The results indicated that tolerant lines with good characteristics may be obtained from a susceptible cultivar by mutation induction, thus allowing cropping in conditions where Al+3 is a limiting factor.  相似文献

Growing numbers of candidate varieties, decrease of their variability for morphological traits, and internationalization of the national list all contribute to excessive increase of the trial costs, thus creating the need for the improvement of current variety evaluation procedures, especially regards their distinctness, Uniformity, and Stability (DUS) component. Due to rapid advancement in molecular techniques, the use of molecular markers in DUS testing as a complement to, or replacement of, morphological observations became the subject of great interest in scientific studies, and consequently topic for discussion within International Union for the Protection of New Varieties of Plants (UPOV). In order to explore the potential of simple sequence repeat (SSR) markers for distinctness tests, present study involved set of 41 maize inbred lines that were scored for 32 DUS characters prescribed by UPOV and genotyped at 28 SSR loci. Results were largely in favor of the use of molecular markers, revealing or confirming their already known advantages over morphological markers like better consistency with the pedigree, and relatively higher discriminative power. However, their integration into DUS testing protocols still depends upon resolving of several important issues.  相似文献

为了成功建立牛滋养层干细胞样细胞系,探讨牛滋养层干细胞样细胞(Bovine Trophoblastic Stem-like Cells)体外培养条件及相关影响因素,以胎牛子宫成纤维细胞条件培养液作为培养基,胶原铺被培养皿,利用体外受精牛囊胚培养得到牛滋养层干细胞样细胞,并调整培养体系,以探讨其最适生长条件。结果表明:利用添加FBS、β-巯基乙醇、肝素以及胎牛子宫成纤维细胞条件培养液的DMEM/F12,体外培养牛囊胚,可以初步建立牛滋养层干细胞样细胞系。  相似文献
