A trial based on progesterone radioimmunoassay of milk samples is described. Samples from 2274 cows in 14 herds were collected on the day of insemination and 7, 23 and 30 days later, unless a return to service occurred before the designated sample date. Two additional samples were collected from cows which returned to service more than 35 days after the first service, one on the day of the return and another seven days later. All six samples from these cows were assayed for progesterone concentration. Late returns, defined as returns to oestrus 36 or more days after mating, occurred in 8.6% of the cows. Milk progesterone assay results indicate that of these apparently late-returning cows, 55.9% suffered a loss of the conceptus, 22.1% had not been detected in oestrus around 21 days after first insemination, 11.8% were in anoestrus at insemination, 5.6% conceived to the insemination and were pregnant when thought to have had a late return, 2.6% were in prooestrus or dioestrus at insemination and 2.1% went into anoestrus after an oestrus insemination. The average prevalence of late returns after the 35th day (8.6%) and the estimated incidence of losses of concepta (4.8%) are lower than those reported in comparable studies elsewhere. Although many of the late-return cows were mated again and conceived, this syndrome nevertheless contributes significantly to the wastage rate, at least in New Zealand dairy herds with their strictly seasonal calving pattern. On the Slusovice co-operative farm, embryo transfer has been practised since 1984 as a method of controlled reproduction of the heifer and cow herd. The effect of superovulation, embryo recovery and administration of 500 micrograms Oestrophan Spofa upon the donors' conception rate and upon their resuming a new reproduction cycle was studied on this farm. The effect of superovulation, induced by FSH-P (Folicotropin, Spofa), was evaluated in 162 cows, out of which 121 (74.69%) got in calf after three inseminations. Fifty-nine (48.76%) of the pregnant cows got in calf after the first insemination, 38 (31.40%) after the second insemination, and 24 (19.83%) after the third insemination. Having been subjected to embryo collection and luteolysis, the donors conceived within 67.6 days, on an average, and the animals superovulated within 60 days post partum conceived within 59.11 days. The average SP of the superovulated animals was about 148 days but in the donors from which the embryos had been recovered within 60 postpartal days the SP did not exceed 110 days. Hence the earlier postpartal superovulation of cycling cows speeds up the resumption of reproduction in the donors and increases the numbers of recovered embryos. Repeated superovulation had no influence on the fertility of the cows but adequately prolonged the service period.  相似文献

The calving season of a 550-cow dairy herd was shortened by the induction of premature parturition in 231 cows and heifers, using the synthetic or natural prostaglandins cloprostenol or PGF2 alpha as the sole treatment, or these agents 8 and 12 days after dexamethasone trimethyl acetate (DTMA), depending on estimated prematurity at the time of first injection. A series of synchronised batch-calvings was produced in the cows, with 85% of the PG group calving within 96 hours of treatment, 97% of the DTMA — 8 days — PG group calving within 48 hours, and 100% of the DTMA — 12 days — PG group calving within 72 hours. PGF2 alpha was associated with very rapid deliveries and dystocia due to incomplete cervical dilation. Cow and calf survival was high. Eighty-three percent of those mated conceived, 78% of them in the first 8 weeks of mating, despite an incidence of incomplete membrane-expulsion that exceeded 40%. Some conceptions to first service did not progress beyond 6–7 weeks of gestation. In nearly all these cases, the animals conceived at a later service, usually at the first oestrus after expulsion.

The results indicate that acceptable results can be obtained in cows within 2 weeks of term using cloprostenol alone. Treatment regimes that allow a priming dose of depot corticosteroid to precede cloprostenol should be used for terminating less advanced pregnancies.  相似文献

183 Holstein cows were studied during and after administration of ch lormadinone acetate (CAP) postpartum. CAP was either fed orally (10mg/d ay for 15 or 20 days) or applied parenterally twice (50 mg) or once (100 mg). By oral administration, estrus was completely (=100%) and ovulation mostly (greater than 95%) suppressed, but follicle growth and estrogen production in growing follicles was not inhibited. Out of 39 closely observed animals, 89.7% ovulated after CAP withdrawal, 79.5% of them within 96 hours. With all animals inseminated, 37.5% conceived at this 1st ovulation. In 144 cows, of which 101 were orally treated with CAP and 39 by injection, shortening of the calving interval and a reduction in fertility problems was achieved. A partly significant improvement of the overall conception rate, the curtailing of the interval between parturition and conception as well as a marked reduction of the standard deviations from the mean values of those intervals were observed. Parenterally treated cows responded in a similar way. But since their 1st ovulations were spread over several weeks and since at these 1st ovulations silent heat was a considerable problem, no prediction of the timing of the 1st ovulation could be made. On the basis of the data presented on uterine contractility, ovarian functions, vestibular hyperaemia, and mucus secretion during and after CAP administration are discussed.  相似文献

Summary Of 1500 examined Holstein cows in milk, there were 315 cows with mucopurulent discharges at the time of insemination (21%). Twenty-four hours after insemination, the infected animals randomly received no treatment (group I, control, n = 93), intrauterine infusion of 1 g oxytetracycline (group II, n = 63) and intrauterine infusion of 5 million IU procain penicillin G sodium (group III, n = 159). First service conception rate following treatment was 48.3, 49.2 and 47.7% in groups I, II and III, respectively. In conclusion, treatment with either antibiotic had no advantage relative to the control on first service conception rate.  相似文献

This study was designed to evaluate if the immunosuppression typically observed during the immediate periparturient period (3 weeks before and after calving) in dairy cows influences the effectiveness of diagnostic tests for the detection of Johne's disease; and, if providing additional energy to the cows during this period would minimize any immunosuppressive effects. Twelve dairy cows naturally infected with Mycobacterium paratuberculosis were fitted with rumen cannulas in late gestation and assigned to treatment groups: control, n = 6; or stuffed, n = 6. Cows in the control group were allowed to consume feed ad libitum. Cows assigned to the stuffed treatment group were also fed ad libitum but received additional total mixed ration by manually stuffing their rumens with refused feed to maintain a dry matter intake of 2% body weight/day before calving and 2.5% body weight/day after calving. Parturition had a significant impact on immune function with significant reductions in M. paratuberculosis-specific antibodies detected in the serum and milk regardless of treatment group. Similarly, in vitro immunoglobulin production was decreased at calving for both treatment groups. In addition, stuffing cows modulated cell-mediated immune function by reducing antigen-specific lymphocyte proliferation and interferon-gamma production after calving. Shedding of M. paratuberculosis in the milk was apparent in 58% (7/12) of cows after parturition with no difference noted between control and stuffed animals. Parturition had no major effect on fecal shedding of cows regardless of treatment. These data suggest that parturition had a significant effect on immune function parameters including diagnostic tests for paratuberculosis. Furthermore, providing additional energy to cows with Johne's disease did not preclude immunosuppressive effects during the periparturient period.  相似文献

A clinical trial was carried out in two provinces of Canada to investigate the effect of treatment with eprinomectin at calving on production parameters in adult dairy cattle in 1999-2000. One of the objectives of this study was to evaluate the impact of treatment on reproductive performance as measured by: calving-to-conception interval, calving-to-first service interval and number of services per conception. The ability of an indirect ELISA using a crude adult Ostertagia ostertagi antigen to predict response to treatment also was evaluated. All lactating cows in 20 dairy herds were allocated randomly to receive either eprinomectin pour-on or placebo at calving. Information on reproductive parameters was obtained from computerised cow records. Survival models were used to evaluate the effect of treatment on the two intervals and a Poisson model was used to evaluate the number of services to conception. A total of 549 cows were included in these analyses. A marginally significant treatment effect on calving-to-conception interval was observed (hazard ratio=1.24, P=0.06) but not on calving-to-first service interval. A significant reduction in the number of breedings to conception for treated animals also was observed with a longer effect in cows with short interval to first service. Milk samples from a subset of 109 late-lactation cows were tested for antibodies against O. ostertagi. The ELISA optical-density ratio (ODR) values obtained between 120 days before calving and drying off were categorised as high ODR (>or=0.5) and low ODR (<0.5). Among untreated animals, the hazard of conception was lower (hazard ratio=0.38, 95% CI=[0.19,0.75]) for high-ODR cows compared to low ODR cows suggesting that higher parasite burdens had an adverse effect on reproductive performance. Treated high-ODR cows had a hazard of conception equivalent to the hazard for all cows in the low-ODR group (indicating that treatment prevented the negative effect associated with these higher parasite burdens).  相似文献

Two groups of lactating dairy cows in each of 4 dairy herds were injected with either 0.5 mg (219 cows) or 0.3 mg (118 cows) of Ooprostenol (I.C.I.) The first group of cows were injected from Day 6 to Day 17 of the oestrous cycle (Day 0 — oestrus) and the second group from Day 6 to Day 12. The four herds were selected specifically because oestrus-detection procedures were thorough and reliable. Of the cows receiving the 0.5 mg dose rate, 54.3% were observed in oestrus within 4 days of treatment. The remainder were in groups which were: (i) in oestrus from 5 to 10 days post-treatment (20.5% with prolonged response intervals), and 11 to 20 days post-treatment (15.5% with unaltered cycle lengths); or (ii) experienced a “silent oestrus” in the immediate post-treatment period and either did not show oestrus for a further 3 weeks (6.4% in oestrus from 21 to 29 days post-treatment) or conceived to a single set-time insemination 72 h after treatment (0.9%). Use of the lower dose rate resulted in fewer cows being observed in oestrus within 72 h (43.8% v 31.4%) and a corresponding rise in the percentage experiencing a “silent oestrus” and returning to service (6.4% v 13.6%) or conceiving to set-time insemination (0.9% v 3.4%). Most of the unaltered cycle lengths occurred with cows being treated on Day 6 of the oestrous cycle. The pregnancy rate for the 171 cows observed in oestrus within 4 days of treatment and inseminated at 72 h, or 72h and 96 h, was 57.3%.

Records from this trial, as well as previous trials in which cows were injected with 0.5 mg of Cloprostenol, showed that 446 of 524 cows (85%) injected at from Day 6 to Day 17 of the oestrous cycle were observed in oestrus within 5 days (144 h) of treatment. Amongthese 446 cows, the interval to post-treatment oestrus was influenced by the stage of the cycle at injection. Whereas 89% of the oestrous responses in cows treated on Days 6 to 9 were observed within 72 h of injection, the comparable figures for cows treated on Days 10 to 13 and Days 14 to 17 were 48% and 70% respectively.

Variation in the post-treatment interval to oestrus is greater in lactating dairy cows than in heifers and is not resolved by using double-injection regimes. The reasons for this variation have not been identified. The most effective use of prostaglandins for oestrus synchronisation in lactating dairy cows requires the treatment of groups of animals at similar stages of theiroestrous cycles and their post-treatment insemination at observed oestrus. These requirements may limit the usefulness of this technology in herds with poor breeding management.  相似文献

Clinical findings were confirmed by bacteriological tests. All animals conceived at the first insemination after treatment. The skim milk progesterone profile was assessed by radioimmunoassay, without extraction, from the day of insemination (day 0) until the cows were dried off on day 225 of gestation. A total of 418 samples were collected from 154 pregnant Holstein cows. The daily variation in skim milk progesterone was recorded from day 1 until day 45 of pregnancy to detect the commencement of progesterone secretion from the corpus luteum after insemination. Subsequent determinations were made every 2 weeks from day 46 until lactation ceased. On the day of artificial insemination and for the first 2 days after insemination, all the cows had a basal progesterone concentration <0.1 ng/ml. A rise in progesterone (0.2±0.1 ng/ml) was first detected on the third day after insemination. The progesterone values then increased significantly (p<0.001) until day 15.The values then remained nearly constant (2.5–3.5 ng/ml) until day 106 of pregnancy, when they began to decline. Between days 120 and 180 of gestation, progesterone was significantly decreased (2.2–2.9 ng/ml) before it rose again to the previous plateau (3.5–3.9 ng/ml) around day 180. The effect of serum glucose, ASAT and urea on reproductive performance was studied in 45 Finnish dairy cows from 8 different herds. Blood samples were taken a month before calving as well as 2 weeks and 2 months after calving. Serum urea, ASAT and glucose were used as indicators of metabolic and energy balance. Glucose and ASAT concentrations were significantly correlated with fertility 2 weeks after calving, but not before calving or 2 months after calving. Antepartum, 60% of the cows had a low urea level; at 2 weeks and at 2 months postpartum 89% and 65%, respectively had low levels of urea. The cows were divided into groups as follows: low or high (greater than or equal to 2.9 mmol/l) glucose level and low or high (greater than or equal to 2.5 mmol/l) urea level. Low levels of glucose and/or urea in puerperium were associated with poor fertility. The intervals from calving to first insemination and conception were shorter in the group with high glucose and high urea than in the group with low values. The cows with the shortest intervals from calving to first insemination and conception showed adequate metabolic balance (high levels of urea and glucose). Clinical ketosis had an adverse affect on intervals from calving to first insemination and on conception. Intervals from calving to first insemination and to conception were significantly (p less than 0.01) longer in ketotic cows than in non-ketotic ones. Liver damage, reflected as elevated activity of ASAT, was associated with poor fertility. If shorter calving intervals are desired, adequate energy balance in puerperium is essential.  相似文献

The reason why cows carrying the mutation of complex vertebral malformation (CVM) show poor reproductive capability although they carry only one mutant allele is still not fully understood. Monitoring the progesterone profiles during oestrous cycle and early pregnancy in carrier cows might help explain their lowered reproductive capability. Progesterone concentration was measured in 19 CVM carrier cows and 21 control cows during oestrous cycle and early pregnancy. Milk samples were collected from all cows starting on the day of artificial insemination until day 45 post‐AI. Progesterone was measured in skim milk using enzyme‐linked immunosorbent assay (ELISA). Progesterone concentration was significantly reduced on day 7 (p < 0.05) and day 9 (p < 0.01) post‐insemination in conceived CVM carrier cows when compared with that in control conceived cows. The mean progesterone concentration during early pregnancy was significantly lower (p < 0.05) in conceived cows with CVM than that of control cows in the same period. However, the mean progesterone concentration did not differ significantly (p = 0.072) in CVM cows that showed fertilization failure or embryonic death than that of control cows. Additionally, of 13 conceived control cows, eight cows (61.5%) showed normal luteal function. In contrast, of nine conceived CVM cows, only four cows (44.4%) showed normal luteal function. The conception rate was 47.4% in CVM carrier cows and 61.9% in control cows, but this difference did not reach significance. In conclusion, progesterone concentration might be lowered during early pregnancy in conceived CVM cows compared with that in control cows.  相似文献

This experiment compared the reproductive performance of synchronised anoestrous dairy cows that were treated initially with a combination of progesterone and oestradiol benzoate and then with either gonadotrophin-releasing hormone (GnRH) or oestradiol benzoate to resynchronise returns to service. It was hypothesised that injecting anoestrous dairy cows with GnRH 12-15 days after insemination and coinciding with the time of insertion of a controlled intravaginal progesterone-releasing (CIDR) device would increase conception rates to the preceding 1st insemination compared with oestradiol benzoate treated cows; both GnRH and oestradiol benzoate would resynchronising the returns to service of those cows that did not conceive to the preceding insemination. Groups of cows in 11 herds were presented for a veterinary examination after they had not been seen in oestrus postpartum. Those cows diagnosed with anovulatory anoestrus (n = 1112) by manual rectal palpation and/or ultrasonography were enrolled in the trial. Each enrolled cow was injected with 2 mg oestradiol benzoate i.m. on Day -10, (where Day 0 was the 1st day of the planned insemination) concurrently with vaginal insertion of a CIDR device. The device inserted was withdrawn on Day -2 and then each cow injected i.m. with 1 mg of oestradiol benzoate on Day -1 unless it was in oestrus. Observation for oestrus preceded each insemination. Every cow that had been inseminated on Days -1,0,1 or 2 was presented for treatment for resynchrony on Day 14 (n = 891). They were divided into 2 groups; those with an even number were each injected i.m. with 250 microg of a GnRH agonist (Treatment group n = 477); each of the cows with an odd number injected i.m. with 1 mg of oestradiol benzoate (control group, n = 414). Each GnRH or oestradiol benzoate injection preceded reinsertion of a CIDR device previously inserted from Days -10 to -2. It was withdrawn on Day 22, 24 hours before injecting 1 mg oestradiol benzoate. Cows observed in oestrus were submitted for a 2nd insemination. Every enrolled cow still present in the herd was pregnancy tested by palpation of uterine contents per rectum about 6 weeks later and again at the end of a herd's seasonal breeding programme. The alternative use of GnRH instead of oestradiol benzoate did not affect the percentage of cows conceiving within 3 days of the mating start date (MSD) (35.6 % vs 35.3 %, P = 0.90), resubmission rates for a 2nd insemination among cows not pregnant to the 1st insemination (81.6 % vs 83.5 %, P = 0.41), 6-week pregnancy rate (59.3 % vs 60.6 %, P = 0.65), 21-weekpregnancy rate (86.6 vs 85.0, P = 0.36), mean interval from MSD to conception (32.5 +/- 1.8 days vs 29.9 +/- 1.8 days, P = 0.26) or conception rate of cows reinseminated by Day 28 (43.3 % vs 38.8 %, P = 0.39). When GnRH was compared with oestradiol benzoate, it did not increase conception rates to the 1st service; it was as effective as oestradiol benzoate in synchronising returns to service in previously treated anoestrous cows that did not conceive to the 1st service. Its use affected neither conception rates to the preceding 1st inseminations nor to the following 2nd inseminations.  相似文献

An AB programme restricted to 14 days through the use of mated, 135 were treated after selection on pre-mating oestrus dates or rectal examination. Treatment was administered on the 6th day of mating. Untreated cows were inseminated, on detection of oestrus, throughout the 14 days. Nine of 135 treated cows were inseminated within 48 hours, 50 at 72 hours only, 75 at both 72 and 96 hours and one at 96 hours after treatment. All cows in season were inseminated, irrespective of recent insemination. Prolonged and split heats, and short cycles (less than 14 days), occurred in treated and untreated cows.

AB conceptions confirmed in those cows retained in the herd were 623% of untreated cows (66.9% of those which were inseminated) and 69.1% of treated.

The conceptions achieved in this abbreviated AB programme are equal to those normally achieved after 7 weeks of AB. Mating management is discussed; the release of labour from AB involvement 4 weeks earlier than usual is a major advantage of this technique.  相似文献

One hundred and nine pure bred Holstein Friesians were injected intramuscularly with 0.125 mg of a synthetic Gn-RH analogue at the time of the first service to study the effect of Gn-RH on the fertility of inseminated cows. Sixty-four cows or 58.7 per cent were diagnosed as pregnant after one insemination compared to 54 animals or 49.5 per cent in the untreated control group. Total pregnancy rate of 81.65 per cent was 8.3 per cent higher than in the controls, while the services per conception rate improved from 1.49 to 1.39. Gn-RH administration at the time of oestrus could improve herd fertility through its including effect on follicular rupture and regulatory influence on ovarian function.  相似文献

The effect of two insemination policies after synchronization of oestrus on reproductive performance in two groups of cows was investigated. Oestrus was synchronized by two treatments with prostaglandin F2 alpha (PGF2 alpha) at a 14-day interval. Cows in Group 1 (n = 234) were inseminated twice on appointment 66 and 90 h after the second treatment. Cows in Group 2 (n = 222) were intensively watched for signs of oestrus after the synchronization protocol and inseminated on observed oestrus. Cows with abnormal discharge after synchronization were excluded from breeding and inseminated later during the study period. Service rate within 1 week after synchronization was higher in Group 1 than in Group 2 (89.3 versus 67.1%). Conception rates on first service did not differ between groups (33.2 versus 30.0%). Days to first service and days open were shorter in Group 1 (P < 0.05). The number of cows culled for infertility did not differ between the groups. Endometritis 14-20 days post-partum decreased the percentage of cows pregnant at the end of the study period in both groups but did not have a significant effect on conception rates and days open. It is concluded that additional inseminations required in the timed artificial insemination protocol were economically justified by the reduction in days open in comparison with insemination on observed oestrus after synchronization.  相似文献

The incidence of early foetal loss is increasing under intensive management systems for dairy cattle. The aims of the present study were to determine whether there is any peak period of pregnancy loss during the early foetal period and to evaluate possible differences between single and twin pregnancies. The study population consisted of 1442 pregnant cattle from a single herd. Pregnancy was diagnosed by transrectal ultrasonography between 36 and 42 days after insemination, and then weekly until day 90 of gestation or until pregnancy loss. A total of 1310 cows (90.8%) bore single embryos and 132 (9.2%) carried twins. Pregnancy loss was registered in 139 (9.6%) cows before day 90 of pregnancy: 101 (7.7%) in single and 38 (28.8%) in twin pregnancies. The average time of pregnancy loss for all animals was 58.4 ± 12.6 days and ranged from 45 to 90 days. Seventy‐five per cent of the pregnancy losses were registered between 45 and 60 days of gestation. The average time of pregnancy loss for cows with singletons was 52.1 ± 4.1 days and ranged from 45 to 61 days and that for those with twins was 75.1 ± 12.4 days and ranged from 46 to 90 days. Seventy‐five per cent of the twin pregnancy losses were registered between 68 and 90 days of gestation. Our data show that the foetal loss in singleton pregnancies occurs earlier than in twin pregnancies. Assessment of normal development of gestation on days 60 and 90 after insemination is suggested.  相似文献

At 11 to 13 d after insemination, 1028 dairy cows on 19 farms were given an intramuscular injection of 10 micrograms of the GnRH analogue buserelin. Their reproductive performance was compared to 1022 control cows receiving a placebo. The GnRH analogue-treated cows had fewer short interoestrous intervals compared to the control cows, but there were no significant differences in pregnancy rates to either the insemination preceding or following treatment, in calving to conception interval, or to percent pregnant by the end of mating. There were no differences between treatments in herds of high, average or low pregnancy rate, in 2-year-old cows or cows greater than 2-years-old and in cows calved 40 d or less, or greater than 40 d. The use of 10 micrograms of the GnRH analogue, buserelin, 11 to 13 d after insemination, cannot be recommended as a method of improving reproductive performance in dairy herds.  相似文献

Maiden heifers and lactating cows of known ovarian status and of several breeds were treated with a synthetic prostaglandin, cloprostenol, or a synthetic progestagen, norgestomet, at the start of an artificial insemination (AI) program. Animals in the cloprostenol treatment received 2 injections 10 days apart. Over the next 26 days those animals that showed oestrous behaviour were inseminated. Synchronisation rates and calving rates to insemination over the first 7 days were calculated. Those in the norgestomet treatment received an implant of norgestomet plus an injection of norgestomet and oestradiol valerate. The implant was removed 10 days later and the animals were given an injection of pregnant mare serum gonadotrophin (PMSG). They were inseminated at 48 h (maiden heifers) or 56 h (lactating cows) after implant removal. Calving rates to fixed-time insemination were recorded. After completion of the AI program the animals in both treatments were joined with bulls. Overall calving rates (AI plus bulls) were calculated. By day 7 of the program, 82% of the maiden heifers and 76% of the lactating cows in the cloprostenol treatment had been detected in oestrus. By day 21 the respective figures were 99% and 81% Norgestomet treatment had an immediate and a prolonged effect on ovarian activity in those females classified as having inactive ovaries at the start of the AI program. Calving rates of those females to fixed-time AI and overall were similar to those of the females with active ovaries in both treatments. Their calving rates to fixed-time insemination, and overall calving rates for the lactating females, were significantly higher than the corresponding values of their contemporaries treated with cloprostenol and inseminated on observed oestrus over 7 days. For those females classified as having active ovaries at the start of the AI program, calving rates to first insemination and overall were similar for both treatments. Overall calving rates of lactating cows of each breed were, with one exception, higher in the norgestomet treatment than in the cloprostenol treatment. Although norgestomet treatment was more expensive than cloprostenol treatment, the advantage in calf crop resulted in an overall monetary advantage to the norgestomet treatment.  相似文献
