Sings of myopathy were recorded by means of electron microscopy from the skeletal muscles of 20 broiler chickens, aged five and six weeks and suffering from spontaneous nutritional encephalomalacia. Focal deposits of osmiophilic, granular or amorphous matter, intermixed also with membrane fragments and condensed to homogenous osmiophilic bodies, were found quite often in the sarcoplasm. Those deposits were interpreted as lipoproteid complexes (pigmentation due to vitamin E deficit). Also observed were degenerative disorders of compensatory reactions of membraneous fibre organelles (sarcoplasmic reticulum, mitochondria) as well as decomposition of myofibrils and fibre hyalinization. Fibre regeneration and vascular damage occurred less often. Pathogenetically, the changes are interpreted particularly with regard to a membrane-stabilising anti-oxidant action of vitamin E and compared to the selenium-dependent myopathies of ruminants from which primary myofibrillar damage was recordable but no pigment accumulation.  相似文献

An electronmicroscopical study has been carried out on the mitochondriae of transversely striated muscles in birds at experimentally induced hypergravitation. In the matrix of some mitochondriae sporadic small light focuses were observed located mainly at the periphery of the organelle. Thereby, however, no greater damage of the outer membranes was seen. The stress state of hypergravitation resulted in more important and more marked alterations on the inner membranes and the matrix of the mitochondriae of the femoral muscle.  相似文献

The morphogenesis of duck enteritis virus (DEV) and distribution in vivo were observed by electron microscopy after ducks were infected experimentally with DEV virulent strain. The investigation showed that a few typical herpesvirus virions and nucleocapsids were first observed in the spleen, thymus, and bursa of Fabricius (BF), and many nucleocapsids, mature viruses, and viral inclusion bodies could be found in the nucleus and cytoplasm of infected liver, small intestine, spleen, thymus, and BF when the ducks died. Nucleocapsids assembled both in nucleus and cytoplasm and could be divided into four different types according to their structures. Typical herpesvirus, light particles (L-particles), and virions without tegument could be observed at the same time. With the replication, assembly, and maturation of the viruses, intracytoplasmic and intranuclear inclusion bodies, electron-density particles, microtubules, hollow tubes, and coated electron-density bodies were observed in infected cells.  相似文献

Histochemical studies of normal and adenomatous intestinal mucosa of the pig showed the adenomatous epithelium to be deficient in enzymes normally found in mature absorptive cells. Electron microscopic studies have been carried out on the mitochondria of the transversely striated muscles with regard to experimental hypokinasis. As compared to the control group the mitochondria of m. pectoralis thoracicus and the m. iliotibialis posterior in hypokinetic birds reveal marked changes. In filamentous and ovoid mitochondria vacuoles can be observed which in some cases produced larger light formations with following disappearance of the cristae and destruction of mitochondria. Fat particles located at the poles of the altered mitochondria, sporadically occurring also laterally, presented another finding. The Z-lines of the sarcomer did not form a continuous line, but were somewhat shifted.  相似文献

The distribution patterns of the intensity of negative charge on the free surfaces (glycocalyx of the plasma membrane) of endothelial cells (ECs) in blood vessels and reticular cells (RCs) in the splenic cord of the rat spleen were studied by an electron microscopic cytochemical method using polyethyleneimine (PEI) as a cationic probe. Spleens from adult male rats were perfusion-fixed with 0.5% glutaraldehyde -4% paraformaldehyde containing 0.05% cetylpyridinium chloride and then perfused with 0.5% PEI at pH 7.4. On the free surfaces (glycocalyx of the plasma membrane) of the ECs examined, distinct PEI-positive reactions were observed in blood vessels, such as trabecular arteries, central arteries, arterial capillaries, pulp veins and trabecular veins. These PEI-positive electron-dense substances in the trabecular arteries, central arteries, and trabecular veins took the shape of a band of 170-250 nm in thickness. On the other hand, the corresponding ultrastructure of the ECs lining the splenic sinuses and the RCs in the splenic cord showed exceedingly weak PEI reactions. The PEI-reactive deposits were significantly thinner than those in the above blood vessels. As the thickness of the electron-dense substances can be related to the density of the negative charge, these results suggest that there is a high intensity of negative charge on the free surfaces (glycocalyx of the plasma membrane) of ECs in blood vessels where blood cells and plasma pass into the red pulp or are discharged from the red pulp. In contrast, the splenic sinuses and RCs, which are the main components of the red pulp, contain weakly negative-charged sites. This may contribute to the microcirculation of the splenic blood vessels and elucidate the possible physiological functions of the spleen, such as blood storage.  相似文献

Eight calves were fed from the fourth to the twentieth week of age a milk powder diet containing 40 mg lead acetate per kg dry substance. This is twice the lead concentration legally permitted. In average, each animal has daily taken up 0.834 mg lead per kg body weight. Eight calves were used as controls. The animals gained the usual fattening weights and did not show clinical symptoms. Beginning with week 14, increased levels of blood urea were encountered in the animals which received lead in their diet. Morphologically, severe renal lesions were found in these calves. The kidneys were increased in weight, pale and of firm consistency. Histologically, fibrosis and periglomerular interstitial non-purulent nephritis were found. Light- and electronmicroscopically, numerous intranuclear inclusion-bodies typical for lead poisoning were demonstrated in epithelial cells of proximal tubules. The results show, that daily uptake of less than 1 mg lead per kg body weight must be considered as a toxic dose for calves, and not 5-7 mg as stated in literature.  相似文献

In vitro experiments of adhesion and colonisation of mucosal surface fragments (nose, trachea) from freshly killed rabbits with four strains of Pasteurella multocida subsp. multocida with different types of capsule antigen (A, D, without capsule) showed the following results: Above all the capsule serovar A strain, producing a capsule of hyaluronic acid and fimbria, were able to adhere and to form microcolonies on the mucosal surface. Microvilli of epithelial cells and mucus producing cells were recognized as the place of adhesion. The formation of microcolonies occurred with a destruction of the kinocilia, which was caused by a bacteria free culture filtrate as well.  相似文献

The time required for occurrence of estrogen-induced uterine tubal (oviductal) ciliogenesis and for differentiation of secretory cells was studied, utilizing electron microscopy procedures. Sixteen cycling gilts were ovariectomized; 3 to 4 months later, 12 principal gilts were each given subcutaneous injections of 17 beta-estradiol in 0.5 ml of corn oil at the rate of 200 mug/day, and 4 control gilts were given injections of corn oil only at the rate of 0.5 ml/day. Two principals each were killed on days 1, 2, 3, 4, 5, and 7 after start of treatment. The epithelial heights were low and completely atrophied 3 to 4 months after ovariectomy. Uterine tubal cilia were absent in all the control gilts. Cytologic changes were not seen in the atrophied epithelium of ovariectomized gilts 1 day after estradiol treatment, but definite proliferative elements consisting of an extensive fibrillar meshwork encrusted with granules (60 to 80 nm) were observed in close association with the nuclear envelope and in the apical cytoplasm after 2 days of estradiol treatment. By day 3, enlarged electron-opaque granules referred to as condensation forms, undergoing various stages of depletion, were closely associated with radially arranged procentrioles. These associations have been referred to as generative complexes. The presence of many generative complexes indicates that maximal production of basal bodies can be expected after 3 days of treatment with estradiol. The depletion of the condensation forms produced hollow spheres with thin walls as the procentrioles grew in length and assembled their microtubules. Enlarged mature-appearing basal bodies were abundant in the cytoplasm after 3 days of estradiol treatment. These bodies aligned themselves linearly along the luminal surface of the cell. Small ciliary buds were then formed above the cell surface, and ciliary filamentogenesis occurred in the bud. Motile cilia were observed on day 3, but cilia numbers increased markedly between day 4 and days 5 and 7. Procentrioles were generated from the diplosomal centriole after 2 days of estradiol treatment. These observations have provided evidence for both ancentriolar and centriolar basal body replication in the ciliated cells of uterine tube of the gilt. Maximal secretory cell differentiation occurred after 3 days of estradiol treatment. Hypertrophy of cytoplasmic organelles was evident on day 3, but the number of secretory granules and amount of rough endoplasmic reticulum increased markedly on days 5 and 7. Close association of secretory granules, Golgi apparatus, and endoplasmic reticulum was evident after estadiol treatment. These data indicate that both ciliated and secretory cells are sensitive to estrogen.  相似文献

Alkaline phosphatase activity as well as lipid, polysaccharide, glycogen, and acid mucopolysaccharide levels in the uteri of 79 gilts were histochemically tested during the first three sexual cycles. Intra-cyclic alkaline phosphatase activity exhibited clearly pronounced variations in the surface epithelium, with maximum values reached in metoestrus, and moderate variations in the endometrial stroma, with maximum values reached in dioestrus. Cycle-dependent variations were recordable also from the glycogen and lipid levels in the surface and glandular epithelia and from the acid mucopolysaccharide levels in the endometrial stroma. The activity of alkaline phosphatase as well as the levels of glycogen and acid mucopolysaccharides during the first cycle still were lower than those in the second and third. Biochemical examinations of the endometrium revealed significant cycle-dependent variations in the activities of alkaline phosphatase, inorganic pyrophosphatase, and succinate-dehydrogenase as well as in the concentrations of soluble protein and glycogen, with maximum values being recordable on the tenth day of cycle. No significant intracyclic variations were recordable from the activities of acid phosphatase as well as of aspartate alanine-amino transferase.  相似文献
