Haemophilus somnus and reproductive disease in the cow: A review   总被引:2,自引:2,他引:2       下载免费PDF全文
Haemophilus somnus causes inflammatory disease in the genital tract of cows as reported in several field surveys and experimental trials. This organism can also innocuously colonize the healthy genital mucosa of the cow, which indicates its dual relationship with the host, that of pathogen and commensal. Experimental data indicate embryocidal capability of this pathogen suggesting a possible role in early embryonic death. Haemophilus somnus also causes sporadic abortions after a bacteremia in the dam. Retrograde infection of the pregnant uterus from the lower genital tract appears unlikely; however, this process can account for post-parturient endometritis. Detection of high homologous IgG2 serum antibody titers using an ELISA test may be useful for the diagnosis of current or recent genital inflammation. Experimental laboratory data indicate that a proportion of genital strains of H. somnus are pathogenic and capable of causing thrombotic meningoencephalitis and perhaps pneumonia. In vivo testing of the pathogenicity of genital strains remains to be conducted.  相似文献

Deformability of erythrocytes and hematological parameters in white leghorn and broiler chickens were measured at age 7, 14, 21, 28, and 35 days. For deformability testing, a simple vertical apparatus containing a polycarbonate membrane with 5-microns pores was used. This technique assesses erythrocyte deformability by measuring the filtration time of an erythrocyte suspension through the pores. There was a significant difference in filtration time between the leghorns and broilers at all sampling times. These results indicate that reduced erythrocyte deformability in broilers may be one of the predisposing factors that increase resistance to blood flow and alter the rheology of blood in the microcirculation in the lung. Increased resistance to flow may result in pulmonary hypertension, heart failure, and ascites in broiler chickens.  相似文献

A pilot study of 10 Dutch dairy herds was performed to investigate possible causes of stillbirth/perinatal weak calf syndrome in heifers. Possible causes and advice for further investigations on farms affected by this syndrome are discussed based on the results of investigations of blood and urine from pregnant young stock, postmortem examination of still-born calves, and a questionnaire held among farmers. Infections with Neospora caninum, Salmonella spp. or Leptospira hardjo, or a deficiency of iodine could be excluded.  相似文献

Results of necropsy examinations of 297 bovine carcasses with naturally occurring disease caused by Haemophilus somnus infection, and necropsied at the Western College of Veterinary Medicine in the period from 1970 to 1990, were analyzed with reference to the ages of affected cattle, seasonal occurrence of disease, the relative frequency of encephalitis, pneumonia, and myocarditis, and interrelationships among these conditions. The cattle came from 177 separate herds in the province of Saskatchewan. There was a range in age from one week to 10 years, and a mean age of 38 weeks. The diseases had a marked seasonal fall and winter incidence. The data indicated a trend characterized by an increasing percentage of cattle with pneumonia and myocarditis and a decreasing percentage with encephalitis during the period.  相似文献

The commercially available API ZYM microbiological identification system was evaluated for the rapid identification of Haemophilus somnus. Eighty-seven isolates of the organism had API ZYM profiles which were characteristic. The API ZYM profiles demonstrate clear differences between H. somnus and other genera but suggest a close association to three related organisms. Enzyme activity of H. somnus isolates were similar to organisms identified as Histophilus ovis, Haemophilus agni and strains UQV of Actinobacillus actinoides and Actinobacillus seminis but was clearly different from isolates of Pasteurella haemolytica, Pasteurella multocida, Bordetella bronchiseptica and group EF4. The API ZYM system allowed more rapid identification of H. somnus than conventional biochemical tests and may be a useful adjunct to conventional methods used for identification of H. somnus isolates. The test did not reveal obvious differences between isolates from various anatomic locations.  相似文献

Several cases of elevated mortality with neurologic signs in 14-to-16-day-old broilers were presented to the Poultry Diagnostic and Research Center from one local integrated company. Suspected of "spiking mortality" associated with hypoglycemia, blood glucose levels were <150 mg/dl overall, with several birds with blood glucose levels as low as 30 mg/dl. Tissues, submitted for histopathology, revealed rickets in 50% of the birds. Virus isolation and serology for reovirus and infectious bursal disease virus were negative. After evaluation of these cases, a brief investigation was conducted to determine the effects of feed restriction on the induction of rickets and resulting hypoglycemia. One-hundred 1-day-old broilers were randomly assigned to three treatment groups of: 1) ad libitum feed, 2) 25% restriction, and 3) 50% restriction. Restriction began on the fifth day of age and continued to 21 days of age. Samples collected during the course of the study included whole blood for blood glucose measurements and proximal tibiotarsus for histopathologic examination for rickets. We were unable to reproduce the clinical signs of spiking mortality, neurologic changes, or hypoglycemia experimentally. Histopathology of the growth plates of the proximal tibiotarsus did indicate mild changes consistent with rickets, but the changes were not significant between treatment groups.  相似文献

Serological tests for the detection of antibodies against Haemophilus somnus were carried out in herds of beef and dairy cattle using three different techniques: agglutination, complement fixation and counterimmunoelectrophoresis. The agglutination test appeared to detect more seroreactors than the complement fixation and counterimmunoelectrophoresis tests. Results of the three tests indicated that there were more positive reactors in beef cattle and dairy cattle from infected herds than in dairy cattle from clinically normal herds.  相似文献

Immunogenicity of the lipid A component of Haemophilus somnus lipooligosaccharide in cattle and mice was examined after purification, detoxification, and covalent conjugation to a protein carrier. After 2 inoculations, a substantial antibody response was induced in most cattle to lipid A and the protein carrier. To determine whether antibodies to lipid A would be protective, 5 x 10(7) colony-forming units of H somnus strain 649 were administered IV to endotoxin-responsive (C3H/HeN) mice. In one study, 8 of 13 C3H/HeN mice aborted when inoculated. In contrast, abortion did not result when mice were inoculated with the same dose of an isolate of H somnus normally found in the prepuce or with the rough mutant Escherichia coli J5. In addition, endotoxin-nonresponsive (C3H/HeJ) mice were significantly (P = 0.03) more resistant to abortion by strain 649 than were C3H/HeN mice, but inoculated C3H/HeN mice were only slightly more resistant to H somnus abortion, compared with control mice. Although a large antibody response to lipid A was detected, there was no significant difference in the immunized group between mice that aborted and mice that delivered normally. Thus, lipooligosaccharide and other properties of virulent H somnus strains may contribute to abortion in mice.  相似文献

The effect of route and dosage of administration on the serological response to a vaccine containing genetically attenuated leukotoxin of Pasteurella haemolytica combined with bacterial extracts of P. haemolytica and Haemophilus somnus (Somnu-Star Ph, Biostar Inc., Saskatoon, Saskatchewan) was evaluated in a controlled field trial in 301 feedlot calves. Vaccination of calves on arrival at the feedlot with Somnu-Star Ph significantly (p < 0.05) increased P. haemolytica and H. somnus serum antibody titers and reduced bovine respiratory disease (BRD) morbidity. A single subcutaneous vaccination with Somnu-Star Ph was as effective in stimulating a humoral antibody response and in reducing BRD morbidity as double vaccination by the intramuscular or the subcutaneous route. Furthermore, there were no swellings or adverse reactions observed with either subcutaneous or intramuscular administration of Somnu-Star Ph.

These results suggest that feedlot calves can be immunized subcutaneously once on arrival with Somnu-Star Ph. Double vaccination was of no added value in this trial, because the majority of BRD morbidity occurred prior to revaccination fourteen days postarrival. Additional larger-sized field trials are needed to monitor the duration of immunity following vaccination and to test the effect of route and dosage of vaccination on mortality.

Perineal hernias almost exclusively affect male dogs (sexually intact or castrated). Factors that may contribute or predispose a dog to perineal hernias include tenesmus, pelvic musculature variations (male vs female), and gonadal hormone influence. In dogs with perineal hernias and tenesmus it is important to include rectal and prostatic diseases, including paraprostatic cysts, in the differential list of potential underlying causes. Surgical correction of the perineal hernia with a perineal herniorrhaphy is indicated. Successful treatment depends on the degree of preexisting neurologic alterations, meticulous surgical technique, and identification and correction of underlying contributing factors. Paraprostatic cysts develop predominantly in sexually intact medium to large breed dogs. These cysts are thin-walled structures often attached to the prostatic dorsal midline. Osseous metaplasia of paraprostatic cysts may occur. Clinical signs of tenesmus often result from compression on adjacent structures (urinary bladder and colon), and tenesmus may contribute to the development of perineal hernias. Preferred treatment of a paraprostatic cyst is surgical removal and castration. This report describes a dog with bilateral perineal hernias and a large mineralized paraprostatic cyst that was identified as a possible contributing factor to the hernias.  相似文献

The effect of early pregnancy failure on the release of prostaglandin F2 alpha (PGF2 alpha) in response to oxytocin (Ot) was examined in an abnormal breeder (AB) heifer that was not able to maintain a pregnancy beyond 21 days. This animal was used in three experiments: 1) She received one intravenous injection of 100 IU Ot 17 days after the onset of oestrus (Day 0). Frequent blood samples were taken for the measurement of 15-keto-13,14-dihydro-PGF2 alpha (PGFM) by radioimmunoassay. Daily samples for progesterone (P4) determinations were taken to monitor luteal function. This was then repeated using the same animal at either day 17 or 18 or 19 (day 17-19) of pregnancy. 2) Embryos from superovulated normal breeder (NB) donors were transferred at day 7 to the AB heifer as well as to NB control animals. 3) Seven day old embryos from the superovulated AB heifer were transferred to NB recipient animals. At day 17-19 of pregnancy all the recipient heifers (experiments 2 and 3) were subjected to the same protocol as in experiment 1. The results showed that the ability of Ot to stimulate PGF2 alpha release was reduced in the NB recipients bearing viable embryos when compared to cyclic animals. However, for the AB heifer, Ot stimulated PGF2 alpha release to the same extent whether the animal was cyclic or pregnant. Furthermore, the AB animal did not have the extended luteal function associated with removal of viable embryos on day 17-19. The data suggest that the embryonic loss might have been caused by failure of the embryos to prevent the luteolytic release of PGF2 alpha.  相似文献

Enterocolitis caused by Yersinia pseudotuberculosis was identified by bacterial culture and histopathology in a 5-month old suckler calf. No other significant enteric pathogens were detected. Diagnosis of this condition can be missed if selective media are not used for bacteriology and if histopathology is not attempted or the alimentary tract is unsuitable for examination. This is the first case report of Y. pseudotuberculosis infection in a calf in the United Kingdom.  相似文献

Transgenic animal models have been used in small numbers in gene function studies in vivo for a period of time, but more recently, the use of a single transgenic animal model has been approved as a second species, 6-month alternative (to the routine 2-year, 2-animal model) used in short-term carcinogenicity studies for generating regulatory application data of new drugs. This article addresses many of the issues associated with the creation and use of one of these transgenic models, the rasH2 mouse, for regulatory science. The discussion includes strategies for mass producing mice with the same stable phenotype, including constructing the transgene, choosing a founder mouse, and controlling both the transgene and background genes; strategies for developing the model for regulatory science, including measurements of carcinogen susceptibility, stability of a large-scale production system, and monitoring for uniform carcinogenicity responses; and finally, efficient use of the transgenic animal model on study. Approximately 20% of mouse carcinogenicity studies for new drug applications in the United States currently use transgenic models, typically the rasH2 mouse. The rasH2 mouse could contribute to animal welfare by reducing the numbers of animals used as well as reducing the cost of carcinogenicity studies. A better understanding of the advantages and disadvantages of the transgenic rasH2 mouse will result in greater and more efficient use of this animal model in the future.  相似文献

The efficacy of a treatment schedule using ivermectin, given subcutaneously at the rate of 200 micrograms kg-1, to control gastrointestinal parasitism and its effect on liveweight gains was assessed. Two herds with a total of 466 Hereford X Brangus cow/calf pairs were used. Each herd was on six pasture plots of comparable size, stocking rate, and quality and quantity of forage. Pasture groups were paired across the two herds. Pasture groups from one herd were randomly assigned to nonmedicated control and the other three to ivermectin treatment. Treatment assignments per pasture group were reversed in the other herd. The control group contained 118 cows and 112 calves and the medicated group 121 cows and 115 calves. Cows were treated in early July and the calves in late July or early August. Cattle from one herd were weighed on Days -28, 0 (day of cow treatment), 30 (day of calf treatment), 58, and 86, while the other herd was weighed on Days -29, 0 (day of cow treatment), 27 (day of calf treatment), 61, and 89. Day 0 was not the same calendar day for the two herds. Fecal samples for parasite egg counts were obtained from the rectum on weight days from five cows and six calves from each pasture; in most cases the same cattle were sampled each time. Calves treated with ivermectin gained (P less than 0.05) more weight than control calves between day of treatment for cows and the end of the trial.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
