Objectives of this study were to evaluate maternal and fetal influences on uterine and umbilical blood flows and nutrient fluxes of gravid uterine tissues of cows. Brahman cows with Brahman or Charolais fetuses and Charolais cows with Brahman or Charolais fetuses were used. Indwelling catheters were placed into a uterine artery and vein, an umbilical vein, and a fetal femoral artery and vein at 220 +/- .4 d after mating. Uterine and umbilical blood flows (liters/min) and net uptakes of oxygen, glucose, lactate, alpha-amino N, urea N, ammonia N, and estrone sulfate by the gravid uterus, fetus, and uteroplacenta were determined on d 227 +/- .4. Uterine blood flows in Brahman cows with Brahman (5.01) or Charolais (4.66) fetuses were similar but less (P less than .001) than in Charolais cows with Brahman (7.14) or Charolais fetuses (9.24), which differed (P less than .01). Umbilical blood flows of Charolais (3.78) were greater (P less than .01) than those of Brahman (2.29) fetuses. Rate of placental D2O clearance as well as net fetal uptake of oxygen, glucose, and alpha-amino N, gravid uterine uptake of alpha-amino N, and uteroplacental uptake of glucose and release of estrone sulfate were greater with Charolais than with Brahman fetuses. Gravid uterine oxygen uptake and estrone sulfate release and gravid uterine and uteroplacental lactate output were influenced by the interaction between cow and fetal breed. It is suggested that fetal growth may be limited by uterine blood flow and by function of the uteroplacenta, particularly in late gestation.  相似文献

Uterine and umbilical blood samples and blood flow (BF) measurements were obtained from one uterine horn and a single fetus of sows at d 70, 90 and 110 of gestation. Concentrations of O2 in blood and of estrogen (E), progesterone (P), glucose, alpha-amino nitrogen (N) and urea N in plasma were determined. Fetal weights were .267 +/- .018, .633 +/- .019 and 1.208 +/- .073 kg on d 70, 90 and 110, respectively. Uterine and umbilical BF remained constant with day of gestation, averaging 1.51 +/- .06 and .31 +/- .02 liters/min. Uptake of P by a uterine horn and fetus remained constant, but secretion of E by a uterine horn and uptake of E by a fetus increased (P less than .05) with stage of gestation. Uterine and fetal uptake of O2, glucose and alpha-amino N did not change and uptake of O2, glucose and alpha-amino N per kg fetus decreased (P less than .01) as gestation advanced. Secretion of urea N from the uterus was observed, with a concentration gradient from the fetal to the uterine circulations. Uterine and fetal respiratory quotients for glucose were .79 and .38, respectively. Thus, although glucose potentially served as a major energy source, a large portion of the energy requirements of the fetal pig were met by catabolism of other substrates. The decrease in umbilical BF and uptake of nutrients per kg fetus with day of gestation suggested that porcine fetal metabolism declined as pregnancy advanced.  相似文献

Acid-base balance and amino acid metabolism are intimately related. Changes in acid-base balance influence the metabolic fate of many amino acids. Also, acid-base homeostasis is achieved in part by alteration of amino acid metabolism, not only in the kidney, but also in liver, muscle and splanchnic tissue. Glutamine is the primary amino acid involved in renal ammonia-genesis, a process intimately related to acid excretion. The metabolism of other amino acids, such a serine, glycine and the branched-chain amino acids, also appears to be influenced by acid-base balance. Conversely, the metabolic fate of various amino acids will influence the daily acid load experienced by the animal. Oxidation of amino acids contributes to the total acid and base load imposed on the pig. The basic (cationic) amino acids (lysine, arginine and histidine) yield neutral end-products plus a proton; sulfur (methionine and cysteine) amino acids are also acidogenic because they generate sulfuric acid when oxidized. The dicarboxylic (anionic) amino acids (aspartate and glutamate, but not asparagine and glutamine) consume acid when oxidized and thus reduce the acid load of the diet. Acid-base balance and related phenomena are discussed in the context of practical and metabolic aspects of amino acid nutrition.  相似文献

We hypothesized that the ability of the fetus to alter nutrient shunting and organ growth might be associated with uterine capacity. White crossbred gilts from a randomly selected control line, a line selected for ovulation rate, and a line selected for uterine capacity (UC) were unilaterally hysterectomized-ovariectomized at 160 d of age, mated at estrus, and slaughtered at 45, 65, 85, and 105 d of gestation (9 to 18 gilts for each line x day combination). Analysis of the data revealed that heart weights and fetal weights were decreased in the ovulation rate line. No significant differences were obtained in fetal, placental, or fetal organ weights between the control and UC lines. Allometric growth of organs was assessed by examination of the slopes of the relationships between fetal weights and fetal organ weights after natural log transformation. Only the relative growth of the liver differed between selection lines and was greater (P = 0.01) in the UC compared with the control line during early pregnancy (d 45 and 65). Allometric growth of the fetal brain, liver, and heart differed with day of gestation. A brain-sparing effect was greater (P < 0.01) on d 85 and 105 compared with d 45 and 65. By contrast, a heart-sparing effect was present during early gestation and disappeared in later gestation. Fetal liver weights were hypersensitive to differences in fetal weights on d 45, possibly associated with placental effects on fetal liver weight. Fetal spleen weights were proportional to fetal weights throughout gestation. These results indicate that selection for ovulation rate decreased total fetal and fetal heart weights, and that selection for UC altered the relationship between total fetal and fetal liver weights during early gestation. Results further indicate significant changes in allometric growth of organs during gestation.  相似文献

Consumption of Ponderosa pine (Pinus ponderosa) needles (PN) by beef cows during late gestation results in premature delivery in association with profound constriction of the caruncular arterial bed. Further, PN extracts and plasma from PN-fed cows increase uterine arterial tone in vitro. Uterine arterial tone is a measure of the arterial resistance to stretch and controls the baseline rate of flow through the vascular bed. Uterine arterial tone results from the uptake of extracellular Ca2+ into smooth muscle cells through specific membrane channels called potential sensitive channels. Functional potential sensitive channels remain open for prolonged periods after activation, allowing a continuous uptake of Ca2+ and the maintenance of uterine arterial tone. Recent evidence from our laboratory has demonstrated that a group of estrogen metabolites produced by the placenta and(or) endometrium, called catechol estrogens, inhibits Ca2+ uptake through the potential sensitive channels. During gestation, progressive decreases in uterine arterial tone are observed, with resultant increases in uterine arterial blood flow. Thus, the continuous production of catechol estrogens may be necessary to maintain the pronounced uterine vasodilation that is required for fetal survival. Ponderosa pine needle extracts exhibit antiestrogenic activity, as evidenced by their inhibition of estrogen-induced uterine hyperemia. Data from our laboratory show that after consumption of PN by beef cows during late gestation, uterine arterial blood flow progressively decreased to less than 50% of prefeeding rates before premature delivery of a live calf.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

To evaluate the influence of cow breed and number of fetuses on uterine and umbilical blood flows and nutrient fluxes or uterine tissues from gravid cows, surgery was performed on Charolais or Hereford cows with single or twin fetuses at 177 +/- .2 d (mean +/- SEM) after mating. Indwelling catheters were placed in a fetal femoral artery and vein, in an umbilical vein of each fetus, and in a uterine artery and vein of each gravid horn. Deuterium oxide (D2O) was infused into a fetal femoral vein at 183 +/- .3 and 190 +/- .5 d after mating to estimate uterine and umbilical blood flows (liters/minute). Blood oxygen and plasma glucose and lactate concentrations were determined and uterine arterio-venous (A-V) and umbilical venous-fetal arterial (v-a) differences and net uterine and fetal uptakes were calculated. Net utilization by the uteroplacenta was calculated as the difference between uterine and fetal net uptakes. Uterine blood flows were lower (P less than .01) in Hereford (4.80 +/- .28) than in Charolais (7.07 +/- .33) and lower (P less than .01) per fetus in cows with twin fetuses (5.22 +/- .34) than in cows with a single (6.65 +/- .28) fetus. Umbilical blood flows were greater for single than for twin fetuses. Fetal oxygen and glucose net uptakes averaged 57 and 12%, respectively, of net uteroplacental utilization. Lactate was released from uteroplacental tissues to fetal (42%) and maternal circulations (58%). The influence of fetal plasma substrate and insulin concentrations on the fractional synthesis rate or turnover of the mixed proteins in the organs of fetal lambs was calculated from measurements of the rate of uptake of L-14C lysine by the protein in the steady state in utero. The control values for protein synthesis rate in the fetal organs of Welsh mountain ewes, 130 to 140 days' pregnant, were similar to those previously observed in the fetuses of Border Leicester ewes: the mean turnover rates were 38, 72, 40 and 63 per cent per day for the brain, liver, cardiac muscle and the placenta respectively; that for skeletal muscles was 6.8 per cent per day; the corresponding half lives were 2.15, 1.27, 1.8 and 1.42 days respectively and 11.3 days for skeletal muscle. A fivefold increase in the plasma glucose levels had no influence on protein turnover rate over a six hour period. A fourfold increase in the concentration of some amino acids, following a mixed infusion, decreased turnover rate in brain, heart and liver. The half life was increased by 30 per cent, but a small reduction in half life was observed in skeletal muscle. Insulin caused a fall in intracellular lysine concentration in skeletal muscle and in plasma, which suggests that the hormone can reduce muscle protein catabolism in the fetal lamb in the last trimester of gestation. Enhancement of synthesis rate occurred in the presence of the amino acid infusions only; the half life of the proteins was reduced by a factor of three. Insulin also increased placental protein synthesis rate by 50 per cent but had no influence on the other tissues studied.  相似文献

Six horses were conditioned on a treadmill at a constant speed of 5.6 km/hr on a 12.5% grade for gradually increasing periods of time over 14 days in order to determine the effect of repeated submaximal exercise on the concentrations of plasma free amino acids, protein metabolism, and plasma volume. Following 14-days of training, plasma volume increased (29%, P<0.05), as did total circulating content of plasma protein, albumin and urea. Urinary urea nitrogen excretion decreased (P<0.05) with exercise training. After the first week of training, the concentration of glycine had decreased (P<0.05) and the concentrations glutamic acid, arginine and alanine were increased (P<0.05) when compared to their corresponding pre-training (control week) levels. Compared to pretraining levels, there were decreases (P<0.05) in aspartic acid, histidine, arginine, valine, phenylalanine, isoleucine, and lysine, following the second week of training. Following a week of recovery, all resting concentrations of plasma free amino acids; when compared to their pretraining control; had decreased, with the exception of three nonessential amino acids (glutamic acid,serine, and glycine). Based upon the results of the present study, it would appear that exercise training produced a significant change in the amino acid and protein metabolism of the horse.  相似文献

一氧化氮(NO)既是妊娠期母体和胎儿体内平衡的关键调节因子,也是促进母体心血管变化、胎儿发育和生长以及适应宫内外生活的重要物质。精氨酸(Arg)与高精氨酸(h-Arg)是一对同源氨基酸,h-Arg是非必需的阳离子氨基酸,其可能由赖氨酸分解代谢或其前体Arg的氨基转移合成。它们均为NO的前体物质,对孕体具有相似的功能。在母猪妊娠期补饲Arg或h-Arg,能够增加孕体NO的含量。NO通过调控激素、血管的生成与增加养分供给等增加胚胎附植、减少胚胎死亡、增强胎儿发育,进而改善胎儿宫内发育受限(IUGR)与母猪繁殖性能。随着妊娠的进行,孕体氧化代谢增加,会产生大量的自由基。Arg或h-Arg代谢产生的NO通过上调Nrf2通路刺激谷胱甘肽(GSH)代谢相关基因的表达,增强孕体抗氧化能力,保证孕体与胎儿的健康发育。作者针对母猪妊娠期补充Arg或h-Arg,通过其代谢产物NO改善母猪IUGR、提高繁殖性能、增强机体抗氧化能力的作用与可能存在的机制进行综述,旨在为Arg与h-Arg在妊娠母猪上的应用提供理论支持。  相似文献

通过胎儿尾部和脐带之间的生殖器结节定位进行马性别诊断,雄性胎儿准确率97.8%(182/186),雌性胎儿99.4%(160/161)。胎儿性别测定的最适时间为妊娠后59d和68d之间。妊娠中期胎儿性别诊断也非常可靠。超声波性别鉴定准确率达100%。在雄性,从100d到220d妊娠常规获得阴茎或包皮的图像;在雌性,从118d到227d妊娠马观察到胎儿乳腺和乳头。在马繁殖上常规使用的5MHz线性传感器,大多数情况下一直可用到妊娠160d。当胎位太高或其大小超过超声设备监测范围时,须使用3.5MHz探头。若掌握好性别鉴定的最适时间和认识正确的解剖结构,两种方法都是非常精确的。  相似文献

Splanchnic metabolism of nitrogenous nutrients and their uptake by the hind limb were studied in finishing lambs receiving ryegrass harvested at grazing stage with or without barley supplementation. Six multicatheterized lambs (40.2 +/- 1.5 kg) were fed with frozen ryegrass (RG) at 690 kJ of ME intake (MEI) x d(-1) x BW(-0.75) and 20.8 g of N intake (NI)/d successively without and with barley supplementation (RG + B), according to a crossover design. Barley supplementation represented 21% of DM intake and increased the MEI and the NI by 32 and 24% respectively, (P < 0.01). In the ruminal fluid, barley increased acetate and butyrate concentrations by 21.2 and 49.6%, respectively (P < 0.04), without any effect on the ammonia concentration. Consequently, the net portal appearance (NPA) of ammonia was not modified, but the NPA of total amino acids (TAA; +38%) and nonessential amino acids (NEAA; +45%) was increased (P < 0.05) by barley supplementation. Taken individually, the NPA of the essential amino acids (EAA) was increased for isoleucine (+32%; P < 0.05), threonine (+151%; P < 0.03), and lysine (+26%; P < 0.06), with no effect for the other EAA. In contrast to what was observed at the PDV level, no significant alteration in the net hepatic amino acid flux was observed for TAA, EAA, NEAA, branched-chain amino acids (BCAA), urea, and ammonia after barley supplementation, showing a relatively minor role of the liver in the regulation of the supply of amino acids to the peripheral tissues. However, taken individually, the net hepatic uptake of some NEAA involved in gluconeogenesis and/or ureagenesis was altered with barley supplementation: the alanine uptake was increased by 44% (P < 0.05), aspartate + asparagine (asx) uptake was decreased by 18% (P < 0.01), and glutamate + glutamine (glx) release tended (P < 0.10) to be increased by 208%. With barley supplementation, NI increased by 5 g of N/d, and net splanchnic release increased by 4.63 g of N/d. Consequently, the additional dietary N supply (together with energy supply) was nearly exclusively available to peripheral tissues as AA-N (N as amino acids), but no strong effect of this additional supply of AA to the hind limb could be demonstrated in terms of net AA hind limb fluxes. Consequently, barley supplementation of a ryegrass-based diet increased the net AA release by the splanchnic tissues, with little effect on the AA net uptake by the peripheral tissues.  相似文献

The aim of this work was to determine whether prenatal survival depends on the genotype of the mother or of the embryo and to identify the critical periods for prenatal mortality in two lines of rabbits divergently selected by high (H) and low (L) uterine capacity. Does from H (n = 124) and L (n = 115) lines were slaughtered at 72 h of gestation. Embryos recovered at 72 h of gestation were transferred to the oviducts of recipient does from the H (n = 23) and L (n = 19) lines. Each recipient does received eight embryos from the H line into one oviduct and eight embryos from the L line into the other. Recipient does were slaughtered on d 28 of gestation. No differences were found between lines in the embryo recovery either in ovulation rate (OR) or in fertilization rate of ova recovered. Recovery rate was higher for the H line (0.80 vs. 0.72, P < 0.01). The number of embryos recovered, fitting ovulation rate as a covariate, was also higher for the H line (9.74 vs. 8.78, P < 0.05). The H line showed a more advanced embryonic stage of development, having a higher percentage of blastocysts (PB) and a lower percentage of compact morulae (PCM) (38% vs. 20%, P < 0.001 for PB, and 51% vs. 64%, P < 0.01 for PCM). The percentage of early morulae was low and similar in both lines. Neither donor nor recipient lines affected embryonic survival from 72 h to 7 d of gestation. Fetal survival was affected by the recipient line (P < 0.05). An interaction between donor and recipient was found. Embryos from the H donor line had a better fetal survival rate than embryos from the L donor line (P < 0.05) in H recipient females. Within L recipient females, embryos from H and L donor lines showed similar fetal survival. Fetal survival was divided into early (from d 7 to 17 of gestation) and late (from d 17 to 28 of gestation). The high recipient line showed a higher early fetal survival than the L recipient line (P < 0.05). The same effect was observed for late fetal survival, but the difference between H and L recipient lines was lower (P < 0.10). Thus, fetal survival depends mainly on the maternal genotype, and the embryo genotype only affects fetal survival when embryo transfer is performed to a favorable maternal environment. Selection for uterine capacity in rabbits leads to modification of early embryonic survival and of early and late fetal survival, but differences are higher for early than for late fetal survival.  相似文献

Three sheep with a small isolated rumen (after Gridin et al., 1964) were studied for the passage of amino acids from the blood to the isolated rumen before feeding and 1, 2, 3, 4 and 5 hours after feeding. It was found that, on an average for all the time intervals mentioned above, the passage of glycine was the largest of all the amino acids studied (0.512 muMol per 100 ml), followed, in descending order, by lysine, alanine, glutamic acid, aspartic acid, serine, leucine, threonine, isoleucine, arginine, tyrosine, and phenylalanine (0.038 muMol per 100 ml). Before feeding and one hour after feeding, lysine shared the greatest proportion of all amino acids that had passed into the isolated rumen (0.565-0.43 muMol per 100 ml), followed, in descending order, by glycine, alanine, valine, aspartic acid, glutamic acid, leucine, serine, threonine, isoleucine, tyrosine, phenylalanine; arginine was, in this case, represented by the smallest proportion (0.042-0.030 muMol per 100 ml). It is inferred from the results that the amount of amino acids passing from the blood through the rumen wall changes with the time that has elapsed from feeding, and that before feeding this passage is more intensive than after feeding. These changes are held to be related with an increased passage of endogenous nitrogen to the rumen in the period of a relative deficiency of substances which are derived from the feed and are essential for maintaining homeostasis in the rumen.  相似文献

Hypothermia is directly linked to metabolism; however, it is still unknown how the overall metabolism is altered by oral administration of hypothermic agent, l -citrulline (l -Cit). The present study aimed to determine the characteristics of liver metabolites of chicks orally administered l -Cit to provide a greater understanding of its metabolism. Capillary electrophoresis–time-of-flight mass spectrometry (CE-TOFMS) and liquid chromatography–time-of-flight mass spectrometry (LC-TOFMS) were conducted on liver samples after oral administration of l -Cit. A total of 361 liver metabolites were identified. Although a small number of samples were used for each group, a principal component analysis and heatmap patterns confirmed that the composition of metabolites could be segregated from each other. Of the 361 compounds detected in the liver, 41 compounds, including amino acids related to the Cit-arginine (Arg) cycle, argininosuccinic acid, Arg, ornithine, and Cit, as well as gamma aminobutyric acid, glycine, histidine, and nicotinamide adenine dinucleotide were abundant in l -Cit-treated livers. In contrast, 24 compounds containing fatty acids, amino acids, and cyclic adenosine monophosphate were lower in the l -Cit group. These data imply that the active Cit-Arg cycle, TCA cycle metabolism, and a low activity in fatty acid metabolism occur in l -Cit-treated broiler chicks.  相似文献

4 groups of 3 growing Wistar rats each were orally given either 15N methionine, 15N lysine, 15N glycine or 15N ammonia sulphate over 10 days. By means of measuring 15N, the 15N accumulation in the amino acids (AA) of the body protein, statements were to be made on the transamination of the individual 15N substances and thus their suitability as tracer substances for studies of N metabolism. None of the tested 15N AA achieved a proportionate labelling of all AA of the body protein. The AA used as tracer in each case showed the highest 15N labelling of all AA in the body. Of the amino 15N detected in the animal body, ca. 19% were found in Met after a 15N Met application, ca. 88% in Lys after a 15N Lys application and ca. 50% in Gly after a 15N Gly application. After the application of 15N ammonia sulphate ca. 42% of the body amino 15N are apportioned to the essential and ca. 58% to the non-essential AA. Thus this substance produces a more proportional labelling of the essential and non-essential AA of the body protein than 15N Gly. The following quotas of the 15N amounts applied were found in the AA of the animal bodies: tracer substance lysine 52%, glycine 32%, ammonia sulphate 24%, methionine 21%. After summing up the amino acid 15N amounts in the animal body, eliminating in each case the tracer AA and taking into account the molecular weight of the AA, there was a good agreement of the intensity of the accumulation of 15N in the individual AA, irrespective of the applied tracer substance: arginine, glutamic acid, cysteine and aspartic acid highest, threonine, phenylalanine and lysine lowest accumulation.  相似文献

3 colostomized laying hybrids received orally with a conventional ration 1% urea with 96.06 atom-% 15N excess (15N'). over a period of 6 days. In the period of the experiment every hen consumed 2.87 g 15N'. After another 2 days, on which they received conventional feed urea, the animals were butchered. 15N' was determined in the total N and in 15 amino acids of the oviduct. Of the 15 amino acids the labelling of glutamic acid, glycine and serine was highest and on average amounted to 0.80, 0.66 and 0.67 atom-% 15N'. In lysine and arginine only 0.10 and 0.11 atom-% 15N' could be detected. The amino acid N with natural isotopic frequency amounted to a quarter for the basic amino acids, a tenth for the branched chain ones and for the non-essential ones (glutamic acid, aspartic acid, serine, glycine, alanine, proline) a third of the total oviduct 14N, The average quota of 15N' is only 3.6%, that of the branched chain amino acids 4.5 and that of the non-essential ones 21.1%. Consequently, the 15N' of the urea is mainly used for the synthesis of the non-essential amino acids of the oviduct.  相似文献

The crude protein digestibility and the amino acid absorption of six female pigs (average live weight 61 kg) with duodenal and ileocecal re-entrant cannulae, which were fed with a wheat + wheat gluten + lysine ration and an N-free ration, were determined in various segments of the intestines. Comparative experiments concerning the N-metabolism with pigs without and with cannulae showed that the animals renormalised their metabolism 14 days after narcosis and fistulation of the intestines. The extents of secretion and absorption of the various amino acids vary as it is shown from the values of the apparent and true digestibility resp. rate of passage through various segments of the intestines. While for methionine and glutamic acid absorption exceeds endogenous secretion already in the duodenum, the amino acids with a high endogenous quota (glycine, alanine, threonine, tryptophan) are, even at the terminal ileum, not as well absorbed as the others. Methionine is obviously synthesised on a large scale by the colon flora and excreted in feces. The fractionation of the duodenal and ileum chyme after feeding wheat + wheat gluten + lysine as well as N-free mixture, into the fractions "solid particles", "peptides-free amino acids" and "proteines precipitable by trichlor-acetic acid" supplies information on the degree of protein degradation in various segments of the intestines.  相似文献
