Recently, 2 novel neuropeptides were discovered, both derived from the same precursor by proteolytic processing, which bind and activate 2 closely related orphan G protein-coupled receptors, Named orexin-A and -B (Sakurai et al., 1998). Both stimulate food intake when administered centrally to rats. 2. Our aim was to elucidate whether central injection of mammalian orexin-A or -B stimulates food intake in the chick. 3. Under conditions of free access to food, orexin-A did not alter the food intake of chicks, but cumulative food intake was significantly suppressed by orexin-B. 4. The orexin-B was then administered to chicks deprived of food for 3 h to confirm its suppressive effect. No significant effect of orexin-B on food intake was detected. 5. Central injection of orexin-B did not modify food intake when appetite was stimulated by fasting. 6. Neither of these orexins appears to stimulate feeding in chicks.  相似文献

Orexin is a hypothalamic neuropeptide that regulates feeding behavior in rats. Orexin-B has recently been cloned in pigs and was shown to stimulate food intake after intramuscular injection. This study was designed to determine whether intracerebroventricular (ICV) and intravenous injections of orexin could regulate appetite in sheep. Suffolk wethers were moved to indoor facilities, adapted to diets for 6 wk, and trained to stand in stanchions for 3 to 6 h each day for 2 wk before indwelling ICV cannulas were installed. These sheep were provided water and they consumed feed ad libitum. On the day before an experiment, each sheep was cannulated in a jugular vein. On the day of an experiment, sheep were placed in stanchions and allowed to stand for 1 h before use. Sheep were then monitored over a 2-h control period before i.v. injection with saline or porcine orexin-B (3 micrograms/kg BW) or ICV injection with artificial cerebrospinal fluid (CSF), orexin (0.03, 0.3, or 3 micrograms/kg BW) or in a second experiment with either orexin B (0.03, 0.3, 3 micrograms/kg BW), neuropeptide-Y (NPY; 0.3 microgram/kg BW), or orexin plus NPY. Food intake was monitored for consecutive 2-h periods. The i.v. injections of orexin did not affect food intake or metabolite or hormone concentrations. In ICV sheep, orexin increased food intake at 2 (P < 0.04) and at 4 h (P < 0.02). Food intake was greatest with the 0.3 microgram/kg BW dosage of orexin (P < 0.05). In the first 2 h after injection, orexin had an effect similar to that of NPY (0.23 kg for orexin and 0.2 kg for NPY). The combination of NPY and orexin had a greater effect on food intake (to 0.34 kg) than did either orexin (P < 0.05) or NPY (P < 0.008) alone. Differences were not apparent in the subsequent 2-h interval. No differences were noted in free fatty acid, glucose, growth hormone, luteinizing hormone, or insulin concentrations following orexin injection. There was an effect of ICV orexin treatment on plasma cortisol concentrations (P < 0.002). Cortisol was increased by orexin at the 0- to 2-h (P < 0.008) and in the 2- to 4-h (P < 0.009) intervals after orexin injection. These data indicate that central administration of orexin stimulates feed intake in sheep.  相似文献

To assess the effects of creep feed consumption on individual feed intake characteristics and performance of group-housed weaned pigs, 16 litters (149 piglets) were fed a commercial creep feed (3,040 kcal NE/kg, 15.2 g lysine/kg) supplemented with 1% chromic oxide. Another five litters (48 piglets) were not given access to creep feed (no-feed). Piglets were weaned at 28 d after birth. On d 18, 22, and 27 of age, fecal samples from all the piglets were taken using fecal loops. A green color of the feces indicated that the piglet had eaten creep feed. Piglets that had green-colored feces three times were considered as eaters. Piglets that never showed green-colored feces were considered as non-eaters. At weaning 22 piglets of each type (no-feed, non-eaters, and eaters) were selected based on BW, litter origin, and sex. These 66 pigs were assigned to six pens equipped with computerized feeding stations. Eaters, non-eaters, and no-feed pigs were equally divided over all six pens. After weaning a prestarter (d 0 to 13) and a starter diet (d 14 to 34) were offered for ad libitum consumption. The individual feed intake characteristics of latency time (interval between weaning and first feed intake) and initial feed intake (intake during the first 24 h following first feed intake) and performance traits were determined for all piglets. The pigs that were designated as eaters needed less time between weaning and first feed intake than the pigs that were designated as non-eaters and no-feed pigs (P = 0.04 and P = 0.06, respectively). Initial feed intake was not affected (P > 0.1) by feed intake prior to weaning. However, during d 0 to 8 the eaters had more visits per day during which feed was consumed than both the non-eaters and no-feed pigs. Averaged over the first 8 d after weaning, the ADFI and ADG of the eaters were higher than that of the non-eaters and no-feed pigs (P < 0.05). Averaged over the total 34-d period the effect of creep feed intake on postweaning ADFI was much less pronounced (P = 0.20), whereas ADG of the eaters was the highest (P < 0.05). Creep feed intake during the sucking period stimulates early postweaning feed intake as well as postweaning performance.  相似文献

Two experiments were conducted to determine the voluntary feed intake and performance of lactating sows fed diets containing a sucrose/milk chocolate product (MCP) blend (Exp. 1) or dried porcine solubles (DPS; Exp. 2). Dried porcine solubles is a coproduct of heparin extraction from porcine small intestines. In Exp. 1, mixed-parity sows (n = 108) at two research centers were assigned to a corn-soybean-meal-based diet formulated to contain 0.9% total lysine or a similar diet that contained 4% sucrose and 2% MCP on an as-fed basis. Sows were allowed ad libitum access to dietary treatments from the day of farrowing until pigs were weaned at approximately 21 d postpartum. Diet had no significant effect on voluntary feed intake of sows during lactation, backfat depth, or postweaning interval to estrus, but it had variable effects on body weight changes. Inclusion of the sucrose/MCP blend in diets elicited a 2% improvement in litter weaning weight at one research center and a 6% depression in litter weaning weight at the other center (diet x research center, P < 0.05). Litter size throughout lactation was unaffected by dietary treatment. In Exp. 2, mixed-parity sows (n = 119) at two research centers were assigned to corn-soybean meal-based diets formulated to contain 0.9% total lysine with 0, 1.5, or 3.0% added DPS. Sows were assigned to dietary treatments within research center, farrowing group, and parity at parturition. Dried porcine solubles tended to increase (P < 0.10) total feed consumed in the first 9 d of lactation and average daily feed intake over the entire lactation (6.03, 6.53, and 6.30 kg) for sows fed 0, 1.5, and 3.0% DPS, respectively. Litter size and weight on d 18 of lactation were not affected by concentration of DPS in the diet. Days from weaning to estrus and percentage of sows displaying estrus were not influenced by diet. We conclude that inclusion of the sucrose/MCP blend in the diet for lactating sows had no consistent effect on voluntary feed intake of sows and weight gain of nursing pigs. Inclusion of DPS at 1.5 or 3.0% tended to improve feed intake of lactating sows but had no significant influence on litter performance.  相似文献

To test the hypothesis that orexin-B acts directly on the anterior pituitary to regulate LH and growth hormone (GH) secretion, anterior pituitary cells from prepuberal gilts were studied in primary culture. On day 4 of culture, 10(5) cells/well were challenged with 0.1, 10 or 1000 nM GnRH; 10, 100 or 1000 nM [Ala15]-hGRF-(1-29)NH2 or 0.1, 1, 10 or 100 nM, orexin-B individually or in combinations with 0.1 and 1000 nM GnRH or 10 and 1000 nM GRF. Secreted LH and GH were measured at 4 h after treatment. Basal LH and GH secretion (control; n = 6 pigs) was 183 +/- 18 and 108 +/- 4.8 ng/well, respectively. Relative to control at 4 h, all doses of GnRH and GRF increased (P < 0.0001) LH and GH secretion, respectively. All doses of orexin-B increased (P < 0.01) LH secretion, except for the 0.1 nM dose. Basal GH secretion was unaffected by orexin-B. Addition of 1, 10 or 100 nM orexin-B in combinations with 0.1 nM GnRH increased (P < 0.001) LH secretion compared to GnRH alone. Only 0.1 nM (P = 0.06) and 100 nM (P < 0.001) orexin-B in combinations with 1000 nM GnRH increased LH secretion compared to GnRH alone. All doses of orexin-B in combination with 1000 nM GRF suppressed (P < 0.0001) GH secretion compare to GRF alone, while only 0.1 nM orexin-B in combination with 10 nM GRF suppressed (P < 0.01) GH secretion compared to GRF. These results indicate that orexin may directly modulate LH and GH secretion at the level of the pituitary gland.  相似文献

Feed intake characteristics of 192, 27-d-old weanling pigs housed in groups and given ad libitum access to feed and water were measured individually with the use of computerized feeding stations. The groups were either homogeneous or heterogeneous as to BW distribution; pigs of three defined initial BW classes were used (mean BW of 6.7, 7.9, or 9.3 kg). The effects of BW distribution, BW class, and sex were studied with regard to average performance traits, latency time (interval between weaning and first feed intake), initial feed intake (intake during the first 24 h following first feed intake), and daily increase in feed intake during the interval between first feed intake and the day on which energy intake met or exceeded 1.5 times the maintenance requirement. Homogeneous and heterogeneous groups had similar latency times, initial feed intakes, and daily increases in feed intake. For the period 0 to 34 d after weaning, ADFI and ADG were also similar for homogeneous and heterogeneous groups, but gain:feed ratio was greater (P < 0.05) in the homogeneous groups. Gilts had higher (P < 0.05) initial feed intakes than barrows and also had greater (P < 0.05) ADFI and ADG during the period 0 to 13 d after weaning. Pigs with average BW of 6.7 kg had higher (P < 0.05) initial feed intakes than their counterparts with average BW of 7.9 kg and 9.3 kg, but the daily increase in feed intake was similar for the three groups. The lighter pigs had more daily visits and a lower feed intake per visit and tended to have a shorter postweaning latency to the onset of feeding than the heavier pigs. This study indicates that the high variability in early feeding behavior among group-housed weanling pigs may be related to BW and sex.  相似文献

To assess the effects of 2 high intensity sodium saccharine-based sweeteners on individual feed intake characteristics and performance of group-housed weaned pigs, one hundred ninety-eight 26-d-old weanling pigs were given ad libitum access to 3 dietary treatments containing: no additional sweetener (control), 150 mg of sweetener (Sucram C-150)/kg, or 150 mg of sweetener (Sucram 3D)/kg. At weaning, piglets were allocated to 18 pens (11 pigs/pen) based on BW, sex, and ancestry, and pens were randomly assigned to 3 treatments with 6 pens per treatment. The pens were equipped with computerized feeding stations. During the first 12 d, pigs were offered pelleted prestarter diets that were replaced at once by pelleted starter diets for the last 7 d of the 19-d experimental period. The individual feed intake characteristics consisting of latency time (interval between weaning and first feed intake), initial feed intake (intake during the first 24 h following the first feed intake), the number of total visits per day, and the number of visits in which feed was consumed, together with the time and the feed intake per visit, were determined for all piglets. Performance traits and fecal consistency were determined per pen for d 0 to 5, d 5 to 12, and d 12 to 19, as well as for the total period (d 0 to 19). The initiation of feed intake was not affected by the addition of high intensity sweeteners to the diet. From 12 d postweaning, dietary sweeteners caused the piglets to focus more on feed intake and less on exploratory behavior, as shown by the increased percentage of visits with feed intake in pigs fed the Sucram 3D diet compared with those fed the control diet (P = 0.002). The overall daily feed intake increased with time but was not affected by the addition of sweeteners. Nevertheless, dietary sweeteners prevented the depression of feed intake on d 8 and 10 postweaning (d 8, P = 0.013; d 10, P = 0.014), which seemed to coincide with an improved fecal consistency score (d 5 to 12, P = 0.11; d 12 to 19, P < 0.001). However, the changes in feed intake characteristics and fecal consistency only resulted in numerical effects on postweaning pig performance (ADFI, P = 0.126; ADG, P = 0.140). The results of the present study indicate that weanling pigs need a certain period of time before clear effects of dietary sweeteners on individual feed intake characteristics and pig performance can be observed.  相似文献

Suckling pigs were separated from their dam for 24 h on day 21 (1 × 24 h fasting, n = 10) or day 21 and 24 (2 × 24 h fasting, n = 10). Pigs in the control group (n = 10) were not fasted before weaning. All pigs were weaned on day 28 postpartum. Feed intake during the first 4 days post-weaning was higher (P < 0.05) for pigs exposed to 1 × 24 h fasting compared with controls. Water consumption was not affected by fasting prior to weaning. The difference in post-weaning feed uptake was not reflected in any clinical traits, intestinal morphology, or activity of digestive enzymes (maltase, dipeptidylpeptidase IV, aminopeptidases A and N; P > 0.15). In conclusion, a short period of fasting prior to weaning can increase post-weaning feed uptake, although this had no clinical impact under the present experimental conditions.  相似文献

A total of 292 sows, at three experimental stations (Auburn, Texas A&M, Texas Tech), was used to determine the effects of depriving sows of feed and(or) water before weaning on reproductive performance. The four treatments were arranged as a 2 X 2 factorial to evaluate two periods of feed deprivation (0 and 48 h) and two periods of water deprivation (0 and 24 h) before weaning. Sows were allowed to consume feed ad libitum during lactation, and were fed 1.8 kg/d during the weaning to breeding interval. Only sows nursing six or more pigs at weaning were used. Pigs were weaned at approximately 42 d at Auburn and 28 d at Texas A&M and Texas Tech. Treatment responses were similar at each station with no evidence of a station X treatment interaction for any of the traits (P greater than .10). During the 48-h treatment period, sows receiving feed and water ad libitum lost 1 kg compared with weight losses of 16 kg for sows deprived of feed for 48 h, 13 kg for sows deprived of water for 24 h, and 17 kg for sows deprived of feed for 48 h and water for 24 h. Weight losses for sows deprived of water only, may have partially resulted from reduced feed intake during the 24-h treatment period; however, feed intake during this period was not measured. At breeding, sows deprived of feed and(or) water maintained their greater weight losses, suggesting that some tissue loss occurred during the 48- or 24-h treatment periods.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In a Hungarian large breeding unit, 481 weaned sows were assigned to three groups and were treated as follows. Sows in Group 1 (Control, n=161) were artificially inseminated (3.01 +/- 0.4 times) during their standing reflex; sows in Group 2 (n=160) were artificially inseminated 3 times at 12-hour intervals, independent of detection of oestrus and immediately after administration of a GnRH-agonist at 96 hours postweaning; and sows in Group 3 (n=160) were artificially inseminated 3 times at 12-hour intervals, beginning at their standing reflex after administration of a GnRH-agonist. Pre-trial daily average lactational feed intake, average daily feed intake from weaning to oestrus, oestrus within 6 days of weaning (%), ovulation within 6 days of weaning (%), wean-to-oestrus interval (h), duration of oestrus (h), follicle size (mm), interval from oestrus to ovulation (h), subsequent day 24 pregnancy rate (%), farrowing rate (%) and total number of pigs born were evaluated. Pre-trial average daily voluntary lactational feed intake was 7.1 +/- 0.5 kg in Group 1, 7.2 +/- 0.4 kg in Group 2, and 7.3 +/- 0.7 kg in Group 3 (P > 0.05). Average voluntary daily feed intake from weaning-to-oestrus was 4.3 +/- 0.9 kg in Group 1, 4.2 +/- 0.8 kg in Group 2, and 4.1 +/- 0.5 kg in Group 3 (P > 0.05). Oestrus was detected within 6 days of weaning in 143 (88.8%) sows in Group 1, 143 (89.4%) sows in Group 2, and in 142 (88.8%) sows in Group 3. Follicle size did not differ (P > 0.05) among the groups. In Group 1, 83.2%, in Group 2, 90.6%, and in Group 3,91.3% of the sows ovulated within 6 days of weaning (P < 0.05), but there were no significant (P > 0.05) differences in 24 Day pregnancy rates (81.4%; 91.3%; and 92.5%). Farrowing rates were in Group 1, 84.5%, in Group 2, 91.3%, in Group 3, 91.9% (P > 0.05). Wean-to-oestrus interval was 115.5 h in Group 1, 114.9 h in Group 2, and 115.7 h in Group 3 (P > 0.05). Duration of oestrus was significantly shorter in Group 2 (41.9 h) and Group 3 (42.1 h) than in Group 1 (68.3 h) (P < 0.001). Similarly, the interval from oestrus to ovulation was significantly different (P < 0.01) between the groups (Group 1, 49.0 h Group 2, 32.0 h, and Group 3, 31.1 h). Sows in Group 2 (12.7) and Group 3 (12.6) had a significantly higher (P < 0.01) number of pigs born than sows in Group 1 (n = 10.9). The interval between oestrus and ovulation was highly and positively correlated (r = 0.83) with the duration of oestrus.  相似文献

This study evaluated the influence of concomitant infections with porcine reproductive and respiratory syndrome virus (PRRSV) and Mycoplasma hyopneumoniae on growth performance, serum metabolite concentrations, and serum insulin-like growth factor-I (IGF-I) in growing pigs. Twenty-two barrows (10 weeks of age) were treated with either an intranasal administration of PRRSV and an intratracheal infusion of M. hyopneumoniae (treatment; n = 8) or a sham inoculation with medium (sham; n = 8), or were not treated (control; n = 6). The sham pigs were matched by body weight and pair-wise fed with treatment pigs. Pigs were weighed on the day of inoculation (day 0) and at 4 weeks postinoculation (day 28). Blood samples were collected prior to inoculation and at weekly intervals for 4 weeks. Pigs in the treatment group exhibited clinical signs consistent with PRRSV infection and M. hyopneumoniae pneumonia. Diagnostic procedures confirmed that treatment pigs were inoculated with PRRSV and M. hyopneumoniae and that sham and control pigs remained free of both pathogens. Average daily gain and feed conversion did not differ among the 3 groups. The IGF-I levels differed (P < 0.05) between control and treatment pigs, even after feed intake returned to similar levels among groups. At day 7, IGF-I concentrations were greater in sham pigs compared with treatment pigs, despite similar feed intake. Sham inoculation and decreased feed intake in sham pigs did not alter serum IGF-I concentrations. Evidently, IGF-I status of pigs affected with disease is influenced by nutritional and nonnutritional factors during the disease process.  相似文献

In order to prove the effect of 'fixed time insemination' and insemination at standing oestrus after post-weaning application of GnRH, in a Croatian large breeding unit, 502 sows were assigned to three groups and were artificially inseminated (AI) at their first post-weaning oestrus as many times as they stand, in 24-h intervals. The groups were treated as follows: group 1 (control, n = 160) were AI during their standing reflex; group 2 ['GnRH-fixed time insemination' (GnRH-FT-AI), n = 175] were AI, independent of detection of oestrus and following administration of GnRH-agonist at 96 h post-weaning; group 3 [GnRH insemination at standing oestrus (GnRH-OE-AI), n = 167] the animals were GnRH-agonist treated as group 2 and were AI at their standing reflex. Pre-trial daily average lactational feed intake, average daily feed intake from weaning to oestrus, oestrus within 6 days post-weaning (%), ovulation within 6 days post-weaning (%), weaning-to-oestrus interval (h), duration of oestrus (h), follicle size (mm), interval from oestrus to ovulation (h), subsequent day 24 pregnancy rate (%), farrowing rate (%) and total pigs born were evaluated. Pre-trial average daily lactational voluntary feed intake was 7.1 +/- 0.08 kg in group 1, 7.0 +/- 0.07 kg in group 2 and 7.1 +/- 0.17 kg in group 3 (p > 0.05). Average voluntary daily feed intake from weaning to oestrus was 5.1 +/- 0.3 kg in group 1, 5.2 +/- 0.5 kg in group 2 and 5.2 +/- 0.19 kg in group 3 (p > 0.05). Oestrus was detected within 6 days post-weaning in 134 (83.8%) in control, 164 (93.7%) in GnRH-FT-AI and 155 (92.8%) animals in GnRH-OE-AI groups (p = 0.05). Follicle size did not differ (p > 0.05) among the groups. In control 82.8%, in GnRH-FT-AI 91.5% and in GnRH-OE-AI 91.0% of the sows ovulated within 6 days post-weaning (p = 0.04), and had 80.6, 90.9 and 89.7% 24-day pregnancy rates (p = 0.16), respectively. In GnRH-FT-AI group 90.2%, in GnRH-OE-AI sows 89.7%, in control animals 79.9% farrowing rates were recorded (p = 0.17). Weaning to oestrus interval was 113.1 h in control, 114.1 h in GnRH-FT-AI and 112.6 h GnRH-OE-AI (p > 0.05). Duration of oestrus was significantly shorter in GnRH-FT-AI (44.9 h) and GnRH-OE-AI (48.1 h) animals, compared with the control (62.9 h) sows (p = 0.001). Similarly, the interval from oestrus to ovulation revealed significant (p = 0.004) differences between the groups (control 44.1 h, GnRH-OE-AI 34.1 h and GnRH-FT-AI 32.9 h). GnRH-FT-AI (12.5) and GnRH-OE-AI (12.6) sows had significantly higher (p = 0.01) number of total pigs born (n = 10.4) compared with control sows. GnRH-agonist-gel treatment to the sow shortens duration of oestrus, the interval from oestrus to ovulation, and may eliminate the need for oestrus detection in the hands of skilled personnel.  相似文献

Syndyphalin-33 (SD-33) increases feed intake in sheep and recently weaned pigs. To assess the effects of SD-33 on hypothalamic gene expression, hypothalami were collected from unweaned pigs (n=19; 21±3 d of age) on day 0. Remaining pigs received an intramuscular injection of 0.5 μmole/kg SD-33 (SD) or saline (VEH) and weaned into individual pens. On days 1, 4, and 7 after weaning, hypothalami were collected from subsets of pigs (n=8 or 9) within each treatment group. Expression of μ-opioid receptor (MOR) was less in SD pigs than in VEH pigs on day 1 and day 4, suggesting down-regulation of the receptor by SD-33. Expression of hypothalamic melanocortin 4 receptor (MC4R) at 1 d after weaning was increased in VEH pigs (but not SD pigs) relative to levels before weaning. Expression of AGRP was not significantly altered by weaning or treatment at 1 d after weaning. At 4 d after weaning, expression of AGRP was greater in SD pigs than in VEH pigs, but at day 7 expression was less in SD pigs than in VEH pigs. A strong positive correlation was noted between expression levels of MOR and MC4R across treatment and time. Treatment with SD-33 appeared to partially abrogate the effects of weaning on expression of two key appetite-regulating genes within 24 h. Effects of SD-33 appear to be mediated at least in part by the μ-opioid receptor and include actions on the melanocortinergic pathway.  相似文献

Neonatal pigs were treated with lipolytic agents to determine whether this would cause a long-term decrease in their ability to deposit fat, with a consequent increase in muscle growth and feed efficiency. Groups of 25 female piglets were given clenbuterol (100 microg/kg BW), porcine somatotropin (pST; 100 microg/kg BW), pST plus clenbuterol, or saline injections from 3 d to 40 d of age. Five piglets from each group were then slaughtered to determine body composition. Clenbuterol and pST both increased ADG up to weaning when given separately (24%, P < 0.05; 20%, P < 0.1 respectively) but did not reduce fat deposition. In contrast, pigs given clenbuterol plus pST showed no increase in ADG and a 41% reduction in carcass fat (P < 0.05). Clenbuterol caused a marked decrease in beta2-adrenoceptor density in porcine adipose tissue (P < 0.001) and skeletal muscle (P < 0.01). This effect was attenuated by concurrent pST treatment, which helps to explain the synergistic effect of these drugs on fat deposition. Once the drugs were withdrawn at 40 d, the anabolic effect of pST gradually disappeared, so that the live weight of pST-treated and control pigs was identical at 168 d. Clenbuterol withdrawal caused the rapid loss of extra weight gained, plus an additional 4 to 5 kg live weight that was never recovered. During the 4-wk finishing period there was an increase in feed intake in pigs that had previously undergone treatment with pST (23%, P < 0.1), with no increase in ADG, and so feed efficiency was impaired (P < 0.05). Pigs that were treated with pST plus clenbuterol showed no marked increase in feed intake during this period. Carcasses from clenbuterol-treated pigs tended to be leaner at 168 d, but there was no long-term effect of pST or the combined treatment on carcass composition. Overall, the treatment of neonatal pigs with repartitioning agents was counter-productive, due to the withdrawal effects of the beta-adrenefgic agonist and the delayed long-term effect of pST on feed intake.  相似文献

The objective of this study was to determine whether intermittent suckling (IS) combined with an extended lactation can reduce postweaning growth check in pigs. Three weaning regimens [conventional weaning (CW), IS with 6-h separation intervals (IS6), and IS with 12-h separation intervals (IS12)] were compared. In CW (n = 17 litters), litters had continuous access to the sow until weaning (d 21, d 0 = farrowing). In IS6 and IS12, litters were separated from the sow for 12 h/d, beginning at d 14 and lasting until weaning (d 41 to 45). Litters were with the sow from 1400 to 2000 and from 0200 to 0800 (IS6, n = 14) or between 2000 and 0800 (IS12, n = 14). Litter size was standardized within 2 d after farrowing by crossfostering, resulting in an average litter size of 10.9 +/- 1.8 piglets. Piglets had ad libitum access to creep feed from d 7 onward. One week after the onset of IS (d 20), creep feed intake was increased in litters from both IS groups compared with CW litters (P < 0.05). Both IS groups consumed considerable amounts of creep feed before weaning (d 41 to 45). Total feed intake before weaning was greater (P = 0.004) in IS12 (3,808 +/- 469 g/piglet) than in IS6 (2,717 +/- 404 g/piglet). In comparison, CW litters consumed 18 +/- 9 g/piglet before weaning (d 21). Irrespective of weaning regimen, total feed intake of litters before weaning was highly correlated with post-weaning feed intake (P < 0.001). Furthermore, in all treatment groups, total preweaning feed intake was correlated with postweaning growth (P < 0.10). Irrespective of treatment, piglets suckling anterior teats grew faster than piglets suckling middle or posterior teats during the first 2 wk of lactation. Body weights at the end of the experiment (d 55) were similar among weaning regimens. Onset of IS induced a growth check in both IS groups (34% for IS12 and 22% for IS6). Only a mild growth check was observed after weaning of IS litters (14% for both IS groups). However, a serious growth check (98%) was observed after weaning of CW litters. Results of the current study indicate that IS stimulated feed intake during lactation, providing a more gradual transition to weaning. Because the IS6 regimen did not prevent the growth check after the onset of IS and is rather laborious, we suggest that IS12 might be preferable for a practical implementation of IS.  相似文献

A live vaccine based on a European isolate of porcine reproductive and respiratory syndrome virus (Porcilis PRRS) was tested in this study in order to determine the protection of fattening pigs against the respiratory form of the syndrome under field conditions. Ten thousand pigs in an infected farm were vaccinated against PRRS virus at the age of 6 weeks and were compared with non-vaccinated pigs with respect to their health status, mortality, performance parameters (average daily gain, average daily feed intake, feed conversion ratio) and the presence of certain pathogens in their lungs. The results showed that treated pigs became ill less frequently and demonstrated reduced mortality compared with untreated ones. As compared with non-vaccinated animals, PRRS-vaccinated pigs also performed in a better way with respect to the feed conversion ratio (P < 0.05) and average daily gain (P < 0.05), while feed intake was similar for both groups (P > 0.05). Bacteriological examinations of the lungs revealed increased incidence of respiratory bacterial infection in untreated pigs compared with treated ones. A tendency for a faster antibody response was also detected in the vaccinees. The results of the present study show that immunization with a live vaccine does protect fattening pigs against the respiratory manifestations of PRRS.  相似文献

Intestinal barrier function in pigs after weaning is almost exclusively determined in terminal experiments with Ussing chambers. Alternatively, the recovery in urine of orally administered lactulose can be used to assess intestinal permeability in living animals. This experiment was designed to study the barrier function of the small intestine of pigs over time after weaning. The aim was to relate paracellular barrier function (measured by lactulose recovery in the urine) with macromolecular transport [measured by horseradish peroxidase (HRP) using Ussing chambers] and bacterial translocation to assess whether lactulose recovery is related to possible causes of infection and disease. Forty gonadectomized male pigs (6.7 ± 0.6 kg) were weaned (d 0) at a mean age of 19 d, fitted with urine collection bags, and individually housed. Pigs were dosed by oral gavage with a marker solution containing lactulose (disaccharide) and the monosaccharides l-rhamnose, 3-O-methylglucose, and d-xylose at 2 h and at 4, 8, and 12 d after weaning. The recovery of sugars in the urine was determined over 18 h after each oral gavage. The day after each permeability test, the intestines of 10 pigs were dissected to determine bacterial translocation to the mesenteric lymph nodes and jejunal permeability for HRP in Ussing chambers. Recovery of l-rhamnose in urine was affected by feed intake and by the time after weaning (P ≤ 0.05). Recovery of lactulose from the urine was greater (P ≤ 0.05) at 4, 8, and 12 d after weaning compared with the first day after weaning and was negatively correlated with feed intake (r = -0.63, P ≤ 0.001). The mean translocation of aerobic bacteria to the mesenteric lymph nodes was greater at 5 and 13 d after weaning compared with d 1 (P ≤ 0.05). Lactulose recovery showed no correlation with permeability for HRP nor with bacterial translocation (P > 0.05). Although both lactulose recovery and bacterial translocation increased over time after weaning, lactulose recovery did not correlate with the permeability for HRP nor bacterial translocation within a pig (P > 0.05). Therefore, we conclude that lactulose recovery in the urine of pigs after weaning is not associated with risk factors for infections. However, it appears to be possible to measure paracellular barrier function with orally administered lactulose in pigs shortly after weaning. Further studies will reveal whether this variable is relevant for the long-term performance or health of pigs after weaning.  相似文献
