The fundamental role of coenzyme Q10 (CoQ10) in cellular bioenergetics is well established in humans and several other species, yet there is no information about its role in horses. In continuation of our study of serum CoQ10 in Thoroughbred race horses, we examined the effect of CoQ10 supplementation and exercise on changes in plasma CoQ10 and α- and γ-tocopherol concentrations. Two groups of horses, five per group, were used. One group received 1.9 g of CoQ10 daily and the second group 3.4 g as a dietary supplement. Both groups were subjected to graduated training sessions that involved low-intensity exercise (gallop) followed by high-intensity exercise (breeze). CoQ10 supplementation resulted in a significant increase in plasma CoQ10 during low-intensity exercise, whereas the increase following high-intensity exercise was markedly attenuated in both groups (P < .05). There were no changes in plasma α- and γ-tocopherol concentrations following the two exercise sessions. γ-Tocopherol constituted approximately 5% of α-tocopherol; this is the first report of plasma γ-tocopherol in horses. The attenuated response of plasma CoQ10 in horses receiving CoQ10 supplementation following high-intensity exercise may be attributed to its consumption during severe oxidative stress. This finding indicates that in horses not receiving any CoQ10 supplementation, high-intensity exercise would result in a marked depletion of endogenous CoQ10 in plasma, which could compromise their health and physical performance. This documents the importance of CoQ10 in horses. The study also establishes the safety and tolerability of high-dose CoQ10 in Thoroughbred race horses.  相似文献

Oxidative stress owing to an imbalance between reactive oxygen species and antioxidants, such as coenzyme Q10 (CoQ10), is a major contributor to male infertility. We investigated the effects of the reduced form of CoQ10 (ubiquinol) supplementation on semen quality in dogs with poor semen quality. Three dogs received 100 mg of ubiquinol orally once daily for 12 weeks. Semen quality, serum testosterone, and seminal plasma superoxide dismutase (SOD) activity were examined at 2-week intervals from 2 weeks before ubiquinol supplementation to 4 weeks after the treatment. Ubiquinol improved sperm motility, reduced morphologically abnormal sperm, and increased seminal plasma SOD activity; however, it had no effect on testosterone level, semen volume, and sperm number. Ubiquinol supplementation could be used as a non-endocrine therapy for infertile dogs.  相似文献

The coenzyme Q10 (CoQ10) is a potent antioxidant with critical protection role against cell oxidative stress, caused by the mitochondrial dysfunction. This study evaluated the effects of CoQ10 supplementation to in vitro maturation (IVM) or embryo culture media on the maturation, fertilization and subsequent embryonic development of pig oocytes and embryos. Maturation (Experiment 1) or embryo culture (Experiment 2) media were supplemented with 0 (control), 10, 25, 50 and 100 μM CoQ10. The addition of 10–50 μM CoQ10 to the IVM medium did not affect the percentage of MII oocytes nor the fertilization or the parameters of subsequent embryonic development. Exogenous CoQ10 in the culture medium neither did affect the development to the 2–4‐cell stage nor rates of blastocyst formation. Moreover, the highest concentration of CoQ10 (100 μM) in the maturation medium negatively affected blastocyst rates. In conclusion, exogenous CoQ10 supplementation of maturation or embryo culture media failed to improve the outcomes of our in vitro embryo production system and its use as an exogenous antioxidant should not be encouraged.  相似文献

ABSTRACT

1. This study was performed to evaluate the effects of dietary supplementation of coenzyme Q10 (CoQ10) on laying rate, body weight, plasma metabolites and some liver gene expression in broiler breeder hens.

2. A total of 128 broiler breeder hens (Arbor Acres Plus, 47 weeks of age) were randomly distributed to four dietary groups supplemented with different levels of CoQ10 (0, 300, 600 or 900 mg/kg diet) with four replicates of eight hens each. During 47–54 weeks of age, laying rate, egg mass and body weight were recorded weekly. To assay plasma biochemical indicators, blood samples were collected at 54 weeks of age. At the end of the experiment, for evaluating the abdominal fat weight, liver weight and expression of the adiponectin and proliferator-activated receptor-α (PPAR-α) genes in the liver, eight hens per treatment were selected, weighed and humanely killed by decapitation.

3. Dietary supplementation of CoQ10 linearly decreased abdominal fat weight, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities by increased levels of CoQ10. The plasma levels of glucose, cholesterol and alkaline phosphatase (ALP) activity were quadratically decreased by increased levels of CoQ10. The best plasma levels of glucose, cholesterol and ALP activity were estimated at 562.5, 633.3 and 517.8 mg CoQ10/kg diet, respectively. Adiponectin and PPARα gene expression exhibited a linear increased by increased levels of CoQ10.

4. In conclusion, addition of CoQ10 to the diet influenced lipid metabolism and expression of the adiponectin and PPAR-α genes, which might be partially due to the improvement in mitochondrial metabolism and energy production. However, further studies are necessary to determine the effects of CoQ10 on these indicators in broiler breeder hens during ageing.  相似文献

Abstract

The influence of vitamin E supplementation on blood serum vitamin E concentration as well as the seasonal variation of serum vitamin concentration was investigated. Forty horses were divided into a control and three vitamin E supplementation (experimental) groups. The levels of the vitamin E supplementation in the three experimental groups were 1, 3 or 5 mg/kg body weight (bwt) per day. The vitamin was dosed orally. The supplementation was started either at the beginning or in the middle of the indoor (winter) feeding period. There was seasonal variation in the serum vitamin E content; the serum vitamin concentration increased during the grazing and decreased during the indoor feeding period. The basal feeding and a daily supplement of 1 mg/kg bwt were not adequate to maintain or increase the serum vitamin E concentration during the indoor feeding period in an exercising horse. The minimum daily intake of vitamin E would seem to be greater than 1.5 mg/kg bwt. It is suggested that a daily supplement of 3 to 5 mg/kg bwt may be required by horses in training to increase the serum vitamin E levels. This amounts to 1500–2500 mg/day for a horse weighing 500 kg. Supplementation should be started at the beginning of the indoor feeding period.  相似文献

研究日粮中添加辅酶Q10对腹水症肉鸡脂质代谢以及抗氧化能力的影响。将180只AA雄性肉仔鸡随机分为3组,每组6个重复,8日龄起在日粮中添加不同水平的辅酶Q10,分别为0、20、40mg/kg。15-21日龄关闭水暖进行较低温处理(15-18℃)以诱发腹水症。结果表明:日粮中添加40 mg/kg辅酶Q10显著降低腹水症肉鸡机体内TG、TC和LDL-C含量(P<0.05),而HDL-C没有变化;显著提高TAOC和降低MDA含量(P<0.05)。这表明辅酶Q10相对长期添加时能够改善肉鸡脂质代谢以及总抗氧化能力。  相似文献

Thirteen horses of Thoroughbred or Standardbred breeding were used to study the effect of dietary fish oil supplementation on blood lipid characteristics. Horses were assigned to either fish oil (n = 7) or corn oil (n = 6) treatment groups for 63 d. The fish oil contained 10.8% eicosapentaenoic acid (EPA) and 8% docosahexaenoic acid (DHA). Each horse received timothy hay and a mixed-grain concentrate at rates necessary to maintain BW. Oil (corn or fish) was top-dressed on the concentrate daily at a rate of 324 mg/ kg of BW. The n-6:n-3 ratio was approximately 3.6:1 for horses receiving the corn oil diet and 1.4:1 for horses receiving the fish oil diet. Horses were exercised 5 d/wk during the study. Before supplementation, there was no difference in the concentrations of any serum fatty acids between the 2 treatment groups. The mean basal concentrations of EPA and DHA on d 0 were 0.04 and 0.01 mg/mL, respectively. After 63 d, horses receiving the fish oil treatment, but not those receiving the corn oil treatment, had increased concentrations of EPA and DHA (P <0.05). Fish oil supplementation for 63 d also increased the concentrations of C22:0, C22:1, and C22:5 fatty acids (P <0.05). Overall, horses receiving fish oil had a decreased concentration of n-6 fatty acids (P <0.05) and a greater concentration of n-3 fatty acids (P <0.01), resulting in a lower n-6:n-3 fatty acid ratio after 63 d (P <0.05). Serum cholesterol concentrations increased (P <0.05) during the supplementation period in horses receiving the corn oil but not in horses receiving the fish oil. Compared with horses receiving corn oil, horses receiving fish oil had lower serum triglycerides at d 63 (P <0.05). These results demonstrate that 63 d of fish oil supplementation at 324 mg/kg of BW was sufficient to alter the fatty acid profile and blood lipid properties of horses receiving regular exercise.  相似文献

Serum selenium (Se), vitamin E, and resting thyroid hormone concentrations were measured in 201 horses in Prince Edward Island (PEI). Selenium concentrations were either marginal (0.0053 to 0.1200 ppm) or deficient (< 0.0053 ppm) in 79% of horses based on current reference ranges for Se in serum. Aged and young adult pleasure horses had a higher prevalence of inadequate Se concentrations compared to racehorses and broodmares (82% and 97% versus 45% and 72%, respectively). Overall, 13% of horses had inadequate (< 200 μg/dL) serum vitamin E concentrations; most of these were young pleasure horses. No horses were hypothyroid and, contrary to findings in other species, there was a positive relationship between serum thyroxine and Se concentrations (P < 0.05). We conclude that Se deficiency is widespread in PEI horse populations, especially in pleasure horses, and vitamin E deficiency is more common in young pleasure horses. Micronutrient supplementation practices employed by PEI horse owners appear inadequate to ensure sufficiency.  相似文献

Alpha-casozepine, a decapeptide derived from bovine milk α-S1 casein, has well-documented anxiolytic properties in several species. To evaluate potential benefit of alpha-casozepine to horses' compliance and comfort with routine management and health care procedures, we blindly compared behavior of alpha-casozepine–supplemented and control-supplemented horses with known specific aversions to specific health care procedures. Twenty-six light horse mares were first screened for aversions based on compliance and apparent comfort during a standard battery of 12 health care examinations and treatment procedures. Based on quantitative behavioral analysis of video-recorded sessions, baseline compliance and/or comfort with each procedure was scored from 0 (unable to progress) to 10 (excellent compliance and relaxed comfort). Based on those results, 10 of the 26 horses were selected as five pairs that were matched for breed and the same two specific aversions. One of each pair was randomly assigned to alpha-casozepine supplementation (2,000 mg PO daily) and the other to control supplement for 5 days. On day 5 of supplementation, the standard battery of 12 procedures was repeated. For the five alpha-casozepine–supplemented subjects, compliance and/or comfort scores improved for seven of their 10 aversions compared with one of 10 for the five matched controls (P < .01, Fisher Exact). Average score increased 1.5 (standard error [SE] = 0.87) points for alpha-casozepine aversions compared with an average decrease of 0.92 (SE = 0.61) points for matched control aversions (P < .05, Wilcoxon signed ranks). These results indicate a modest benefit of alpha-casozepine supplementation to horses for improvement of compliance and apparent comfort with mildly aversive routine health care procedures.  相似文献

The purpose of this study was to determine if ginseng fed at low levels enhances a horse's antibody response to vaccination against Equid herpesvirus 1 (EHV-1). For 28 d, 5 horses received ground, powdered ginseng (35 mg/kg body weight, 1.7 mg/kg total ginsenosides) in molasses as a carrier, and 5 received molasses only. On day 14, each horse was vaccinated against EHV-1. The time course of the antibody response to vaccination was significantly altered in the horses receiving ginseng, a clinically relevant increase in antibody titer being observed by postvaccination day 2 compared with day 6 in the control horses. The horses receiving ginseng also had a significant decrease in serum levels of sodium and a significant increase in serum levels of potassium. No adverse effects of ginseng treatment were identified by hematologic and blood biochemistry profiles. Thus, low-dose dietary supplementation with ginseng in healthy horses may be a useful adjunct to vaccination.  相似文献

Communities of intestinal helminths in horses are commonly studied post mortem. The study objectives were here to examine the species composition of the strongylid community in brood horses in Ukraine after deworming with an aversectin drug Univerm. The site distribution of the strongylid species was analysed according to dynamics of their expulsion in faeces. Forty-four horses of different ages from Poltavska oblast (22 horses), Kyivska oblast (17 horses) and Sumska oblast (5 horses) of Ukraine were included in the study. Horses were treated with Univerm anthelmintic (0.2% aversectin) at a dose rate of 0.5mg aversectin preparation per kg body weight. Faecal sampling (200 g each) was performed at 24, 36, 48 and 60 h post treatment, and all nematodes expelled were collected and identified. The largest numbers of strongylids were expelled at 24--36 h after treatment. Twenty-five nematode species from the subfamilies Strongylinae and Cyathostominae were identified. The number of strongylid species found per horse ranged from 7 to 20, on an average 11+/-3.6 (S.D.). The number of cyathostomin species found per horse ranged from 7 to 16, on an average 10+/-2.3 (S.D.). Cylicocyclus nassatus and Cyathostomum catinatum were the most dominant species were found in 100% of horses, amounting to 36.3% and 17.6% of the total number of strongylids collected, respectively. C. longibursatus, C. ashworthi, Cylicostephanus calicatus, C. leptostomus and C. minutus were identified in more than 80% horses and represented 39.9% of the total number of strongylids collected. The dynamics of the different strongylid species expelled was irregular. Correlation between the time of cyathostomin species expulsion in faeces and their predicted localisation inside the horse intestine was found. Species mainly localised in the caecum were found in faeces later than those species localised in the dorsal and ventral colons. Larvae and adult Parascaris equorum, Oxyuris equi and botfly larvae from the genus Gasterophilus were also found in horse faeces. This investigation shows that is possible to study the horse strongylid community after deworming with aid of an aversectin drug. The results obtained here correspond to those recorded in previous autopsy surveys in other countries.  相似文献

Failure of pH to predict ionized calcium percentage in healthy horses   总被引:2,自引:0,他引:2  
Blood, serum, and plasma total calcium concentrations and plasma and serum ionized calcium concentrations were anaerobically determined by use of a calcium-specific electrode for samples obtained from 39 healthy horses. Mean (+/- SD) serum ionized calcium concentration was 6.6 +/- 0.3 mg/dl (1.6 +/- 0.1 mmol/L) and the mean serum ionized calcium percentage was 58.2 +/- 3.4%. Serum ionized calcium percentage was not significantly correlated with serum pH. Plasma ionized calcium percentage was weakly correlated with plasma pH (r = -0.480; P less than or equal to 0.05). Ionized calcium concentration was determined in serum samples manipulated in vitro by additions of 1 to 80 microliters of 0.1N hydrochloric acid or sodium hydroxide to yield 6 to 10 pH values between 6.8 and 8.2. In all horses, the relationship between serum ionized calcium percentage and serum pH at these pH values was then examined by use of a repeated-measures multiple regression analysis. Correlations between serum ionized calcium percentage and adjusted serum pH value for each horse were highly significant (P less than or equal to 0.05); however, analysis of pooled data from all horses indicated that a statistically significant relationship between serum pH and ionized calcium percentage did not exist. Lack of a significant relationship between these variables was most likely attributable to heterogeneity of variance of ionized calcium percentage among horses, reflecting variation in undefined biochemical constituents of serum that affect the equilibrium of calcium binding. When it is essential to evaluate the calcium status of a horse, direct measurement of serum ionized calcium concentration is recommended.  相似文献

The purposes of this study were to (1) prospectively establish serum IgM and IgG concentrations in normal, fit, adult horses over time and (2) determine the accuracy of serum IgM concentrations for diagnosing lymphoma. Serial IgM and IgG concentrations were measured with a radial immunodiffusion assay in 25 regularly exercised horses at 6-week intervals. Horses had serum IgM concentrations ranging from 50 to 242 mg/dL over 5 months, with 20% of horses having IgM < or = 60 mg/dL. The normal range for IgM in fit horses should be considered 103 +/- 40 mg/dL and a cut-point for an IgM deficiency, < or = 23 mg/dL. IgG concentrations ranged from 1,372 to 3,032 mg/dL. Retrospectively, medical records of adult horses (n = 103) admitted to the Cornell University Hospital for Animals for which serum IgM was measured were examined. Horses were categorized as "lymphoma negative" (n = 34) or "lymphoma positive" (n = 18). The sensitivity and specificity of a serum IgM concentration (< or = 60 mg/dL) for detecting equine lymphoma was 50 and 35%, respectively. At the new cut-point (< or = 23 mg/dL), the sensitivity was low at 28% and the specificity improved to 88%. The negative predictive values at various population prevalences indicate that a horse with a high serum IgM (> 23 mg/dL) is unlikely to have lymphoma, whereas the positive predictive value (70%) does not allow for reliable determination of lymphoma in a horse with serum IgM < or = 23 mg/dL. Therefore, serum IgM concentrations should not be used as a screening test for equine lymphoma.  相似文献

The pharmacokinetics of the anti-convulsant phenytoin were investigated in clinically healthy horses after oral (p.o.) and intravenous (i.v.) administration. A single dose of phenytoin (8.8 mg/kg body weight) was given i.v. as a bolus to nine horses and one horse received 13.2 mg/kg. A two-compartment open model was used to describe the disposition of phenytoin. Four of the horses that received an i.v. dose (three at 8.8 mg/kg and one at 13.2 mg/kg) were then given the same dose 3 days later by the oral route. Phenytoin achieved a peak concentration in serum within 1–4 h after p.o. administration and was poorly absorbed with a bioavailability of 34.5 ± 8.6%. Oral dosage regimens were calculated on the basis of a dosing interval of 8 h to provide average serum steady-state concentrations of 5 and 10 μg/ml for phenytoin.  相似文献

In 2004-2006, 322 brood horses from 11 horse farms were examined using the faecal egg count reduction test (FECRT) to determine the presence and distribution of anthelmintic resistance in strongylids in Ukraine. The anthelmintic drugs "Albendazole-7.5" (7.5mg of albenazole, Ukraine) at a dose of 5mg per kg body weight and "Univerm" (0.2% aversectin C, Russia) at a dose of 0.5mg per kg body weight were used. Seventy-one horses from six farms were examined in vivo to investigate the influence of anthelmintic treatment on the gastrointestinal strongylid community structure. Horses were treated with anthelmintics; faecal sampling (200 g in each sample) for strongylid expulsion was performed 24, 36, 48 and 60 h after treatment; and all strongylids expelled (25,292 specimens) were collected and identified. Fourteen horses from the Dubrovsky horse farm were also examined to determine the benzimidazole-resistant cyathostomin species; 5208 specimens of benzimidazole-resistant cyathostomins were collected and identified. According to the FECRT data, benzimidazole resistance in strongylids was observed only at the Dubrovsky horse farm (FECRT=68.7%). No resistance to macrocyclic lactones in strongylids or in Parascaris equorum was observed. Twenty-nine strongylid species were found in horses from six horse farms. The number of species per horse ranged from 4-9 (5.8+/-1.5) to 10-20 (14.4+/-2.9) and depended on horse anthelmintic treatment strategies. From 4 to 13 strongylid species predominated (prevalence>66.7%) in the strongylid community. Eleven cyathostomin species (Cylicocyclus nassatus, C. ashworthi, C. leptostomum, Cyathostomum catinatum, C. pateratum, Cylicostephanus calicatus, C. longibursatus, C. goldi, C. minutus, Coronocyclus coronatus and C. labiatus) were found to be resistant to benzimidazoles at the Dubrovsky horse farm. Ten of these were the dominant species in the strongylid community; only C. labiatus was a rare species (prevalence 29.4%). Species richness and species diversity were significantly higher in horses from farms without treatment or with occasional treatments than from farms with regular treatments. The shape of the prevalence frequency distribution of strongylid species from farms with regular treatments was bimodal ("core" and "satellite" mode). This distribution was multimodal at farms without treatment or with occasional anthelmintic treatments. The results of the current study indicated the possibility of the further spread of anthelmintic resistance on horse farms in Ukraine and the necessity of monitoring the development of resistance in horse parasitic nematodes.  相似文献

Itraconazole, a third-generation azole, was evaluated for treatment of resistant nasal mycotic infections in horses. Two horses with Aspergillus spp nasal granulomas and 1 horse with Conidiobolus coronatus nasal infection were treated with itraconazole (3 mg/kg PO bid). One of the horses with nasal aspergillosis was also treated by surgical resection of the nasal septum. The treatment time for the horses ranged from 3 to 4.5 months. No adverse effects were noted in any of the horses during the treatment period. Peak and trough serum itraconazole concentrations were < 0.5 μg/mL in all 3 horses. Itraconazole (3 mg/kg PO bid) appears to be effective in the treatment of nasal Aspergillus spp infections in horses because the fungal infection was eliminated in both horses. One horse still had excessive nasal sounds during exercise and was retired from training, whereas the other horse returned to normal. The nasal C. coronatus infection appeared resistant to itraconazole treatment in the affected horse because the granulomas were still present after 4.5 months of treatment.  相似文献

1. One hundred and sixty 1-d-old Arbor Acre male broiler chicks were fed with maize-soybean based diets for 6 weeks in a 2 x 2 factorial experiment. The factors were CoQ10 supplementation (0 or 40 mg/kg) and Escherichia coli lipopolysaccharide (LPS) challenge (LPS or saline). 2. CoQ10 was supplemented from d 1. From d 18, the chickens received three weekly i.p. injections of LPS (1.0 mg/kg BW) or an equivalent amount of sterile saline as control. From d 10 on, all chickens were exposed to low ambient temperature (12 to 15 degrees C) to induce ascites. 3. The blood packed cell volume and ascites heart index of broiler chickens were reduced by dietary CoQ10 supplementation. Mitochondrial State 3 and State 4 respiration, respiratory control ratio and phosphate oxygen ratio were not changed, but H+/site stoichiometry of complex II + III was elevated by dietary CoQ10 supplementation. 4. Cytochrome c oxidase and H+-ATPase activity were increased by CoQ10 supplementation, whereas NADH cytochrome c reductase and succinate cytochrome c reductase were not influenced. Mitochondrial anti-ROS capability was increased and malondialdehyde content was decreased by CoQ10 supplementation. 5. The work suggested that dietary CoQ10 supplementation could reduce broiler chickens' susceptibility to ascites, which might be the result of improving hepatic mitochondrial function, some respiratory chain-related enzymes activities and mitochondrial antioxidative capability.  相似文献

A urine sucrose test has recently been reported to be a reliable method of detecting gastric ulcers in horses; however, technical difficulties associated with urine collection have limited the practical value of the test. The objective of this pilot study was to determine whether gastric sucrose permeability, as evaluated by serum sucrose concentration, could be used to detect gastric mucosal injury in horses. Twelve adult horses with naturally acquired gastric ulceration were studied. After a 20-hour nonfeeding period, each horse was dosed with 250 g of sucrose via nasogastric intubation. Blood samples were collected at 0, 15, 30, 45, 60, and 90 minutes, and horses underwent gastroscopy 4 hours later. The severity of gastric ulceration in each horse was defined by means of a 4-point ulcer-scoring system, and the relationship with serum sucrose concentration was analyzed by means of a linear mixed-effects model. Serum sucrose concentration was measured by liquid chromatography operating in tandem with electrospray mass spectrometry. After nasogastric administration of table sugar, horses with moderate to severe gastric ulceration had significant increase in serum sucrose concentration at 30, 45, 60, and 90 minutes, relative to earlier times (P < .05). Peak sucrose concentration was observed at 45 minutes, and was correlated with ulcer severity (Spearman's rank correlation coefficient = 0.898, P < .05). These data indicate that determination of sucrose concentration in equine serum may be a useful test for identifying horses with endoscopically visible gastric ulceration and has potential use as a noninvasive method for screening and monitoring horses engaged in racing training and other performance-related disciplines.  相似文献

At birth, 24 Standardbred foals were assigned at random to 1 of 2 groups and were given a placebo supplement (group 1) or an iron supplement (248 mg of iron/treatment; group 2). Foals were given iron supplement or placebo 4 times during the second and third weeks after birth. Hematologic variables and general health were monitored until foals were 4 months old. Mean PCV in foals of both groups decreased during the first 2 weeks after birth, but values remained within adult horse reference ranges. During the first 6 weeks after birth, foal erythrocytes were smaller than adult horse erythrocytes, but foal erythrocyte glucose-6-phosphate dehydrogenase activity was greater than that in adult horses. At every measurement, indices of anisocytosis were lower in foals, compared with adult horse reference values, suggesting that foals have a homogeneous population of microcytic erythrocytes during early foalhood. In 2-week-old foals of both groups and in 4-week-old placebo-treated foals, mean serum iron concentration was lower than that in adult horses. In foals at birth and during the first 4 months, total iron-binding capacity values were above the adult reference range. In newborn foals, transferrin saturation percentage values decreased to below the reference range in foals from 2 weeks to 4 months after birth. When foals were born, serum ferritin concentration values were above the adult horse reference range, but decreased to within the reference range by the time foals were 1 day old. OBJECTIVE: To validate use of high-performance liquid chromatography (HPLC) in determining imipramine concentrations in equine serum and to determine pharmacokinetics of imipramine in narcoleptic horses. ANIMALS: 5 horses with adult-onset narcolepsy. PROCEDURE: Blood samples were collected before (time 0) and 3, 5, 10, 15, 20, 30, and 45 minutes and 1, 2, 3, 4, 6, 8, 12, and 24 hours after IV administration of imipramine hydrochloride (2 or 4 mg/kg of body weight). Serum was analyzed, using HPLC, to determine imipramine concentration. The serum concentration-versus-time curve for each horse was analyzed separately to estimate pharmacokinetic values. RESULTS: Adverse effects (muscle fasciculations, tachycardia, hyperresponsiveness to sound, and hemolysis) were detected in most horses when serum imipramine concentrations were high, and these effects were most severe in horses receiving 4 mg of imipramine/kg. Residual adverse effects were not apparent. Value (mean +/- SD) for area under the curve was 3.9 +/- 0.7 h X microg/ml, whereas volume of distribution was 584 +/- 161.7 ml/kg, total body clearance was 522 +/- 102 ml/kg/h, and mean residence time was 1.8 +/- 0.6 hours. One horse had signs of narcolepsy 6 and 12 hours after imipramine administration; corrresponding serum imipramine concentrations were less than the therapeutic range. CONCLUSIONS AND CLINICAL RELEVANCE: Potentially serious adverse effects may be seen in horses administered doses of imipramine that exceed a dosage of 2 mg/kg. Total body clearance of imipramine in horses is slower than that in humans; thus, the interval between subsequent doses should be longer in horses.  相似文献
