Many methods have been used to evaluate synthetic and natural products as hydroxyl radical scavengers. In this study a spectrophotometric method to evaluate the scavenger activity of the hydroxyl radical produced in Fenton reaction is proposed. The oxidation of l-epinephrine to adrenochrome permitted the detection of the hydroxyl radical by spectrophotometry and its possible scavenging by hydroquinone, a well known antioxidant. The antioxidative properties of bulb, leaf and stalk of Allium ursinum were investigated. Activities of antioxidant enzymes (superoxide dismutase, catalase, peroxidase, glutathione peroxidase), quantities of malonyldialdehyde, superoxide and hydroxyl radicals and reduced glutathione and also the contents of total flavonoids, chlorophylls a and b and carotenoids were determined. The extracts from all plant organs exhibited antioxidant activity, the highest having been observed in the leaves. Furthermore, ESR signal of PBN-OH radical adducts in the presence of leaves phosphate buffer (pH 7) extract was reduced for 87.61%. 相似文献

The aqueous extract of Phellodendron amurense Rupr.(Amur Cork Tree) provides a rich source of antioxidants and chemical compounds, and can be used for food and wood preservative materials.In this study, we characterized the chemical composition of this extract by GC and GC/MS.The antioxidant capacity was evaluated using a variety of antioxidant assays(superoxide radical,hydroxyl radical, nitric oxide radical, and DPPH radical scavenging activity).Additionally, total polyphenolic content was determined.Phenolic acids and acetone derivatives were major compounds of the extract capable of scavenging the DPPH free radical and reducing ferric ions.DPPH and ferric ion reduction results were strongly correlated with total phenolic content of the extract which also exhibited strong nitric oxide, hydroxyl radical scavenging and superoxide anion radical scavenging activities. 相似文献

The antioxidant activity of the methanol extract of Filipendula hexapetala flowers was assessed by the assay for ferric-reducing antioxidant power (FRAP), the assay for DPPH free radical scavenging ability (DPPH) and the assay for the influence of lipid peroxidation in liposomes, induced by Fe(2+)/ascorbate system and measured by the TBA test (LP). The activity of the investigated extract in all test-systems was found to be significant. Raspberry (Rubus idaeus) leaves, collected in different locations of Lithuania were extracted with ethanol and the extracts were tested for their antioxidant activity (AA) by using ABTS(.)(+) decolourisation and DPPH(.) scavenging methods. All extracts were active, with radical scavenging capacity at the used concentrations from 20.5 to 82.5% in DPPH(.) reaction system and from 8.0 to 42.7% in ABTS(.)(+) reaction. The total amount of phenolic compounds in the leaves varied from 4.8 to 12.0 mg of gallic acid equivalents (GAE) in 1 g of plant extract. Quercetin glucuronide, quercetin-3-O-glucoside and rutin were identified in the extracts. 相似文献

The antioxidative potential of Achillea alexandri-regis has been studied by measuring the influence of herb extracts on hydroxyl and superoxide radicals' quantity in different in vitro systems. The ethyl acetate extract exhibited hydroxyl radical scavenging activity in all tested biological system (liver homogenate, hemolyzed blood, serum and postmitochondrial liver fraction), whereas butanol extract reduced hydroxyl radicals significantly only in the postmitochondrial liver fraction. An ultrasonic technique was employed to extract polysaccharides from Ophiopogon japonicus. The ultrasonic extracted polysaccharides (POJ-U) were purified, and POJ-U1a (a homogeneous fraction) was obtained. The structural characteristics of POJ-U1a were investigated by infrared spectra, gas chromatography, high performance liquid chromatography, periodate oxidation, Smith degradation, methylation analysis, gas chromatography–mass spectrometry and nuclear magnetic resonance spectroscopy. The results indicated that the relative molecular weight of POJ-U1a was 4.02 × 103 Da. POJ-U1a was an α-configuration polysaccharide with a highly branched structure, and consisted of pyranoside and funanside. The backbone of POJ-U1a consisted of 1,6-α-d-glucopyranose and 1,3,6-α-d-glucofuranose in the molar ratio of 7:3, while the branched chains were mainly composed of 1,3-α-d-glucopyranose and 1-α-d-glucopyranose in the molar ratio of 1:3. The branched structure of POJ-U1a was proved intuitively by AFM. Significant antioxidant activity of POJ-U1a has been proved as shown by its DPPH radical scavenging, hydrogen radical scavenging and superoxide anion scavenging activities, which indicated that POJ-U1a showed strong antioxidant activity. 相似文献

The effects of aqueous extracts of Celastrus paniculatus (CP) seeds were shown to have antioxidant properties in rats. In the study reported here, we have investigated the free radical scavenging capacity of three aqueous extracts (WSEs) obtained from CP seeds: a room temperature extract (WF); a hot water extract (HF); an acid extract (AF). All the WSEs exhibited a dose-dependent free radical scavenging capacity for 1,1-diphenyl-2-picryl-hydrazyl radical (DPPH) and also for superoxide-generated assays (in vitro assays). In addition, we used enriched forebrain primary neuronal cell (FBNC) cultures to evaluate the neuroprotective effects of the three CP-WSE extracts on H(2)O(2)-induced toxicity. FBNC were pre-treated with the CP-WSE and then with H(2)O(2) to evaluate the protection afforded against H(2)O(2)-induced toxicity. The criteria for neuroprotection by the WSEs were based on a mitochondrial function test following the H(2)O(2)-induced neurotoxicity. All the WSEs significantly attenuated H(2)O(2)-induced neuronal death, and AF was the most effective in protecting the neuronal cells against oxidative injury caused by H(2)O(2). In 10 day FBNC, cellular superoxide dismutase activity was not affected by the WSEs or H(2)O(2), but catalase activity was decreased and levels of malondialdehyde were increased by H(2)O(2) treatment. When the neuronal cells were treated with WSEs prior to H(2)O(2) exposure, catalase activity was increased and levels of malondialdehyde were decreased significantly. The data presented here suggest that CP seed WSEs protected neuronal cells in part by their free radical scavenging properties, by reducing lipid peroxidation, and also by their ability to induce the antioxidant enzyme catalase. Our results indicate that WSEs might exert neuroprotective effects against increased oxidative stress resulting from free radical damage that is associated with a number of neurodegenerative diseases. 相似文献

以胆木叶为原料,通过不同极性溶剂依次萃取50%醇提取物,分别得到石油醚萃取物(PEE)、氯仿萃取物(CE)、乙酸乙酯萃取物(EAE)、正丁醇萃取物(BE)和水萃取物(WE),它们的收率依次为0.62%、3.90%、0.68%、1.78%和6.06%.采用过氧化氢体系、超氧阴离子(O-2·)体系、羟基自由基体系(·OH)和Oyaizu法,以Vc为阳性对照,进行5个不同萃取部分抗氧化活性的测定和比较.结果表明,5种萃取部分均具有一定的抗氧化活性,随着浓度的增加而提高;且在不同的自由基产生体系中,其抗氧化活性强弱不尽相同.在过氧化氢体系中,对H2O2的清除能力强弱顺序依次为Vc>BE>CE>EAE>PEE>WE;在超氧阴离子体系中,对O-2·抗氧化活性强弱顺序依次为Vc>EAE>BE>PEE>WE>CE,在羟基自由基体系中,对·OH抗氧化活性强弱顺序依次为Vc>BE>CE>EAE>PEE>WE;而其还原能力强弱顺序依次为 BE>EAE>WE>CE>PEE. 相似文献

from the methanol extract, and a hot water extract of the fruiting bodies of P. citrinopileatus. The biological activities such as melanin biosynthesis inhibition, tyrosinase inhibition, antioxidant activities [1,1-diphenyl-2-picrylhydrazyl
(DPPH) radical scavenging, oxygen radical absorbance capacity (ORAC), and superoxide dismutase (SOD)-like activity], antibacterial
activities, and antihyaluronidase activities of these extracts were evaluated. We found that the n-hexanesoluble, diethyl ether-soluble, and ethyl acetate-soluble fractions exhibited melanin biosynthesis inhibition in B16
melanoma cells, as well as antioxidant (ORAC) and antibacterial activities. However, the n-butanol-soluble and water-soluble fractions and the methanol and hot water extracts exhibited antioxidant (DPPH radical scavenging,

The ethanolic extract of Leucas aspera root was subjected to acetic acid induced writhing inhibition, 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging assay and brine shrimp lethality bioassay for screening of antinociceptive, antioxidant and cytotoxic activity, respectively. The extract produced significant writhing inhibition in acetic acid induced writhing in mice at the doses of 250 and 500 mg/kg. The extract showed a significant free radical scavenging activity with an IC(50) of 8 microg/ml. The extract showed significant lethality to brine shrimp with an LC(50) value. 相似文献
