The role of caffeine as a chemical defense of coffee against the berry borer Hypothenemus hampei was investigated. No positive correlation was observed between resistance and caffeine content in experiments in which seeds from several coffee species presenting genetic variability for the alkaloid were exposed to adult insects. The same was observed in an experiment with coffee seeds that had their caffeine content doubled by imbibition with caffeine aqueous solutions. Other experiments showed that the attractiveness to insects was not related to the caffeine content of mature fruits. These results indicate that H. hampei has evolved an adaptation to handle the toxic effects of caffeine.  相似文献

The anthocyanin composition and antioxidant activity of the crowberry ( Empetrum nigrum) were studied. High-performance liquid chromatography (HPLC) combined with a diode array detector and electrospray ionization mass spectrometry were used for identification and quantification of individual anthocyanins. Freeze-dried crowberry powder was extracted with 80% methanol containing 0.5% acetic acid and subjected to HPLC. Thirteen kinds of anthocyanins were identified. The major anthocyanins were cyanidin-3-galactoside and delphinidin-3-galactoside, at 8.04 and 8.62 mg/g extract, respectively. The HPLC profile of crowberry extract was similar to bilberry and blueberry. The total content of anthocyanins in crowberry was 41.8 mg/g extract, higher than the other nine major berry species (2.5-38.8 mg/g extract). The antioxidant activity was also evaluated using the 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis(3-ethybenzothiazoline-6-sulfonic acid) radical quenching assays and the ferric reducing activity power assy. Crowberry extract exerted the strongest antioxidant activity. In conclusion, individual anthocyanins in crowberry were identified and then quantified in this study. Additionally, crowberry is suggested to be associated with a reduction in the risk of developing chronic diseases because of its strong antioxidant activity.  相似文献

The use of chlorophyll fluorescence measurements to noninvasively evaluate degrees of ripeness was investigated in berries at various stages of ripening from two white grapevine cultivars (Vitis vinifera L. Cv. Bacchus and Silvaner). Berries were characterized by diameter, weight, and density and by concentrations of fructose, glucose, sucrose, and total sugars, as well as fructose/glucose ratios, and also by chlorophyll fluorescence at F(0) and F(M) levels and the fluorescence ratio F(V)/F(M). Pearson product moment correlation analysis on data from both cultivars revealed clear negative associations between F(0) and concentrations of fructose, glucose, and total sugars, and fructose/glucose ratios (correlation coefficient < -0.89). Curvilinear trend-lines were established for plots of F(0) versus concentrations of fructose, glucose, and total sugars, but a linear relationship between F(0) and fructose/glucose ratios was found: the corresponding coefficients of determination were always >0.82. Therefore, chlorophyll fluorescence measurements are well-suited to determine noninvasively sugar accumulation in grape berries during ripening.  相似文献

Euterpe oleraceae is a large palm tree indigenous to the Amazon River and its tributaries and estuaries in South America. Its fruit, known as acai, is of great economic value to native people. In this study, a standardized freeze-dried acai fruit pulp/skin powder was used for all analyses and tests. Among many findings, anthocyanins (ACNs), proanthocyanidins (PACs), and other flavonoids were found to be the major phytochemicals. Two ACNs, cyandin 3-glucoside and cyanidin 3-rutinoside were found to be predominant ACNs; three others were also found as minor ACNs. The total content of ACNs was measured as 3.1919 mg/g dry weight (DW). Polymers were found to be the major PACs. The concentration of total PACs was calculated as 12.89 mg/g DW. Other flavonoids, namely, homoorientin, orientin, isovitexin, scoparin, and taxifolin deoxyhexose, along with several unknown flavonoids, were also detected. Resveratrol was found but at a very low concentration. In addition, components including fatty acids, amino acids, sterols, minerals, and other nutrients were analyzed and quantified. Total polyunsaturated fatty acid, total monounsaturated fatty acid, and total saturated fatty acids contributed to 11.1%, 60.2%, and 28.7% of total fatty acid. Oleic acid (53.9%) and palmitic acid (26.7%) were found to be the two dominant fatty acids. Nineteen amino acids were found; the total amino acid content was determined to be 7.59% of total weight. The total sterols accounted for 0.048% by weight of powder. The three sterols B-sitosterol, campesterol, and sigmasterol were identified. A complete nutrient analysis is also presented. Three-year-old citrus trees were grown in the greenhouse to study the effects of fertilizer concentration and root herbivory on plant growth and mineral concentration. In separate experiments, sour orange (Citrus aurantium L.) and Swingle citrumelo (C. paradisi Macf. × Poncirus trifoliate L.) plants were treated with a complete fertilizer diluted to provide 25, 100, 200, or 400 ppm N and grown for 7 weeks with or without Diaprepes abbreviatus L. larvae. Increased fertilizer concentration increased the shoot mass and the shoot:root ratio of both sour orange and Swingle citrumelo. Root herbivory also increased the shoot:root ratio by depressing root growth more than shoot growth. Effects of root herbivory on growth were consistent across the four levels of fertilizer concentration, indicating that tolerance is not a function of nutrient status. For both rootstocks, concentrations of nitrogen in roots and leaves increased with fertilizer concentration, and C:N ratios decreased. In sour orange, root herbivory most strongly affected the concentration of carbon in roots, whereas in Swingle citrumelo, root herbivory most strongly affected leaf nitrogen. In general, herbivory reduced mineral concentrations of roots but the strength, and sometimes the direction, of herbivore effects varied significantly among fertilizer treatments. This research indicates that application of excess, balanced fertilizer is unlikely to offset growth reductions due to root herbivory by D. abbreviatus, and suggests that supplementation of specific nutrients may be of value.  相似文献

Thaumatin-like proteins and chitinases, which are pathogenesis-related (PR) proteins, were the major soluble protein components of grapes from five cultivars of Vitis vinifera. This dominance of PR proteins was apparent at berry softening (véraison) and then throughout berry development for the Muscat of Alexandria, Sultana, and Shiraz cultivars and in the berries of the Sauvignon Blanc and Pinot Noir cultivars examined at commercial maturity. The M(r) of the major thaumatin-like protein from Muscat of Alexandria grapes was 21 272, and those of the three major chitinases from this cultivar, ChitB, ChitC, and ChitD, were 25 588, 25 410, and 25 457, respectively. The vines in the study were irrigated and showed no obvious signs of disease. Shiraz vines that had not been irrigated throughout the season were clearly water stressed, but had levels of PR proteins in the berry similar to vines that had been fully irrigated. It appears that the production of PR proteins that cause protein instability in wines by grapes may be little influenced by environmental conditions.  相似文献

The antioxidative properties of coffee brew fractions were studied using electron spin resonance spectroscopy using 2,2,6,6-tetramethyl-1-piperidin-1-oxyl (TEMPO) and Fremy's salt (nitrosodisulfonate) as stabilized radicals. TEMPO was scavenged by antioxidants formed during roasting and not by chlorogenic acid, whereas Fremy's salt was scavenged by all antioxidants tested including chlorogenic acid. The stabilized radical TEMPO allowed the exclusive measurement of roasting-induced antioxidants. The roasting-induced antioxidant activity of coffee brews increased with increasing degree of roast, and most of these antioxidants were formed during the initial roasting stage. The majority of these roasting-induced antioxidants were present in the high molecular weight fractions, indicating that the formation of these antioxidants preferably occurs at specific high molecular weight structures, likely being arabinogalactan and/or protein moieties which might be part of the melanoidin complex. It was found that chlorogenic acids most probably do not lose their antioxidant activity and phenolic characteristics upon incorporation in coffee melanoidins. The parameter fast reacting antioxidants (FRA) was introduced as an alternative for the antioxidative potential. FRA levels showed that coffee fractions rich in roasting-induced antioxidants exposed their antioxidant activity relatively slowly, which must be a consequence of its complex structure. Finally, the melanoidin content and the roasting-induced antioxidant activity showed a positive and linear correlation for the coffee brew fractions, showing that roasting-induced antioxidants are present within melanoidins. This is the first time that the formation of roasting-induced antioxidants could be directly correlated with the extent of Maillard reaction and melanoidin formation in a complex product such as coffee.  相似文献

Chestnut (Castanea sativa) is an important basic food in rural diets and a major starch crop used in a similar way to potatoes. Chestnuts are a fundamental economic resource in the "chestnut regions" not only for the fruit but also for the chestnut wood. Chestnuts have become increasingly important with respect to human health, for example, as an alternative gluten-free flour source. Chestnuts are also a rich source of other beneficial compounds, but there have been few studies on the composition during processing. In this study, we analyzed the chemical composition of three Portuguese cultivars at different stages of industrial processing. The chestnut cultivars were Longal, Judia, and Martaínha. All three cultivars had high moisture contents but were low in ash, crude fat, and crude protein contents, with high starch and low fiber contents. The free amino acid contents, including various essential amino acids, varied depending on the cultivar. All three cultivars also had a significant content of polyphenolics with gallic acid; ellagic acid was predominant among hydrolyzable and condensed tannins. Many of these compounds are known to exert significant positive effects on human health. The one-way analysis of variance for fresh chestnut shows significant differences among the three cultivars for most of the studied parameters. The same statistical analysis applied to each one of the two cultivars (Judia and Longal) sampled for the four processing steps analyzed indicates a significant effect of this factor in practically all of the constituents. On the other hand, the two-way analysis of variance shows that, besides the residual, the processing step and the interaction cultivar x processing step were the factors that more contributed for the total variation observed in the constituents analyzed, while the contribution of cultivar was much less significant.  相似文献

Summary A study was undertaken to investigate arylsulphatase activity in 15 soils cropped to cocoa (Theobroma cacao) and coffee (Coffea canephora var. robusta) in Ghana. The arylsulphatase activity was correlated positively and significantly with organic C, total N, and cation exchange capacity, and correlated negatively with acetate soluble sulphate. The enzyme was deactivated at an incubation temperature of over 60°C. Preheating and oven-drying of soils decreased arylsulphatase activity. Addition of 1.0 ml toluene during the assay resulted in a sharp decline in arylsuphatase activity. The addition of trace elements at a concentration of 1 ppm caused a reduction in soil arylsulphatase activity compared with that of the untreated samples.  相似文献

The diagnosis and recommendation integrated system (DRIS) approach was used to interpret nutrient analyses of leaf tissues from pomegranate cv. Bhagwa orchards grown in southwestern Maharashtra, India. The DRIS norms were established for three growth stages,viz. 50% flowering, fruit development and first harvesting of pomegranate. Various nutrient ratios were obtained from high-yielding population and were used to compute DRIS indices for diagnosing nutrient imbalances and their order of limitation to yield. Nutrient sufficiency ranges at 50% flowering derived from DRIS norms were 1.32–2.15% nitrogen (N), 0.18–0.24% phosphorus (P), 1.29–1.99% potassium (K), 0.64–1.20% calcium (Ca), 0.23–0.45% magnesium (Mg), 0.16–0.26% sulfur (S), 103.04–149.12 mg kg?1 iron (Fe), 39.60–72.85 mg kg?1 manganese (Mn), 15.99–26.10 mg kg?1 zinc (Zn), 6.16–9.32 mg kg?1 copper (Cu), 23.38–39.88 mg kg?1 boron (B) and 0.29–0.47 mg kg?1 molybdenum (Mo). Similarly, the sufficiency range at fruit development and first harvesting was developed for computing DRIS indices. The requirement of Fe, Mg, S, Zn and N by the pomegranate plant was higher at 50% flowering and fruit development stages. According to these DRIS-derived indices, 87.85, 73.83, 70.09, 69.16 and 65.42% orchards were deficient in Fe, S, Mg, Zn, and N, respectively, at 50% flowering, while 70.03, 66.36, 63.55, 61.68, and 68.22% orchards were found to be deficient in respective nutrients during the fruit development stage.  相似文献

Soils at different developmental stages were sampled from eight sites on the slopes of Mt Etna, Sicily (Italy) and characterized for total C, microbial biomass and microbial respiration. The values of these parameters were greatest for the most developed soils, but differences in recent management and site characteristics limited analysis of trends with soil development across the eight sites. The decomposition kinetics of both intact leaf litter and the water-insoluble fraction of leaf litter from three common species on Etna [Etnean broom (Genista aetnensis), European chestnut (Castanea sativa), and Corsican pine (Pinus nigra)] were determined in four of the soils (the two with the smallest and the two with the largest organic C contents) in a laboratory experiment over 168 days to test two hypotheses. First, that the readily mineralized fraction of added plant C is greater when the plant material decomposes in well-developed soils compared to less developed soils, and second, that the microbial communities in less developed soils are less efficient at mineralizing C from low quality plant residues. The first hypothesis held for Genista and Pinus litter, but not Castanea litter. The second hypothesis was supported for the Castanea and Pinus litter, but not for the Genista litter. Thus, the general applicability of the hypotheses was dependent on the precise source and characteristics of the litter.  相似文献

  【目的】  植物–微生物之间的互作反馈机制是果实类药材发育成熟过程中影响品质形成的重要因素之一。因此,我们研究了著名民族药用植物米槁果实发育成熟过程中3个关键时期根际微生物群落变化对其品质形成的影响。  【方法】  于贵州境内选择罗甸(LD)、坝碰(BP)、祥乐(XL) 3个典型米槁居群,在其幼果期、近成熟期和成熟期取果实和根际土壤样品,分别测定其果实化学成分和土壤中微生物群落、土壤化学性质的变化。采用多元统计方法分析米槁根际微生物群落动态变化与米槁果实成分积累的潜在关系。  【结果】  3个居群中共获得真菌有效优质序列870008条,可分为3026个OTU。获得细菌有效优质序列568739条,可分为4971个OTU;从不同果期微生物丰度较高的类群来看(去除未分类类群),根际真菌中幼果期以Saitozyma、被孢霉属(Mortierella)较高,近成熟期以灵芝属(Ganoderma)、小杯伞属(Clitocybula)较高,成熟期以被孢霉属(Mortierella)、Saitozyma较高。根际细菌中幼果期以鞘脂单胞菌属(Sphingomonas)、节杆菌属(Arthrobacter)较高,近成熟期以节杆菌属(Arthrobacter)、芽孢杆菌属(Bacillus)较高,成熟期以节杆菌属(Arthrobacter)、不动杆菌属(Acidibacter)较高;土壤养分中有效磷各个果期差异最为显著,其他养分差异相对较小;从果实化学成分组成情况来看,罗甸的米槁果实糖类和粗脂肪含量最高和挥发油含量中等,总体来说质量最好;RDA分析表明,优势菌属Saitozyma、被孢霉属(Mortierella)、节杆菌属(Arthrobacter)对米槁果实中α-松油醇、桧烯、可溶性多糖、粗多糖积累具有促进作用。土壤脲酶(S-UE)活性对米槁根际微生物群落组成影响最大,其次为有效磷和过氧化氢酶。  【结论】  不同果期米槁根际均以子囊菌门(Ascomycota,未鉴定属),Saitozyma、被孢霉属(Mortierella)、节杆菌属(Arthrobacter)等微生物类群对果实化学成分积累具有明显促进作用。但其中品质最佳的罗甸居群受子囊菌门(Ascomycota,未鉴定属)影响最大,坝碰、祥乐则受被孢霉属(Mortierella)影响最大,3个产地米槁根际真菌中伞菌属(Agaricus)、镰孢菌属(Fusarium)和瓶霉属(Phialophora)对果实松油醇、桧烯积累的影响不显著,而在不同果期微生物群落结构变化又主要受到土壤脲酶活性(S-UE)、有效磷含量和过氧化氢酶活性的影响。  相似文献

Abstract

White yam (Dioscorea rotundata cv. Olonko) was grown consecutively for three growing seasons, i.e. from February to October of 1975, 1976 and 1977, and treated with six levels of nitrogen, viz: 0, 40, 60, 120, 160 and 200 kg N/ha in the field. Leaf samples were taken at four stages of growth as follows: vegetative, tuber formation, tuber development and tuber maturation, and analyzed for NO3‐N, P, K, Ca and Mg.

Increasing nitrogen fertilization consistently increased leaf‐NO3‐N, particularly at the vegetative stages of growth, while no consistent trend was established for leaf‐P. Leaf‐K was increased at low rate of nitrogen fertilization during tuber formation and maturation whereas leaf‐Ca increased only at tuber maturation in the presence of higher rates of nitrogen fertilization. There was a marked increase in leaf‐Mg at all stages of growth when N treatment was increased to 200 kg N/ha. A positive correlation (r = 0.84???) was obtained for leaf‐K at the vegetative growth stage with tuber yield, while leaf‐Mg was positively correlated with tuber yield at vegetative (r = 0.46?), tuber formation (r = 0.50?) and tuber development (r = 0.67??) stages. All other elements were negatively correlated with yield at all stages of growth. Tuber yield was highest at the 200 kg N/ha treatment.  相似文献

Carabid larvae and adults were collected by pitfall trapping and soil sampling for a period of 1 year from an organic potato field in western Austria. The main results and conclusions were the following. (i) Larval catches enriched the total species spectrum (n = 49) by 6 %. Recording of pre-adult stages seems to be necessary for a complete registration of a beetle coenosis. (ii) Twenty species were caught in adult as well as in larval stages, representing apparently the autochthonous Carabidae of the investigated field. (iii) The use of soil sampling enriched the larval ground beetle spectrum and provided additional data on the life cycle of certain species. Therefore additional methods to pitfall trapping are stated to be important. (iv) Epigaeic activity of winter larvae surpassed that of adults during late autumn, winter and early spring, whereas summer larvae lived mainly hemiedaphicly. (v) Third instar larvae showed the highest pitfall catches in most species, which was probably caused by their higher epigaeic activity and greater activity radius. (vi) Life cycles of Nebria brevicollis, Loricera pilicornis, Calathus fuscipes, Amara aulica, Harpalus rufipes, Bembidion lampros and B. properans are presented and discussed. Data of larvae support the concept of species which reproduce a second time or show two breeding cohorts throughout the year. (vii) Carabid larvae may possess interesting predation value in the regulation of certain pests due to a phenology and habitat that differ from that of adult beetles.

Dans un champ de pommes de terre cultivé biologiquement dans l'ouest de l'Autriche, les larves et adultes de différentes espèces de carabes ont été récoltés par piégeage Barber et échantillonnage de sol pendant 1 an. Les principaux résultats et conclusions sont les suivantes. (i) La capture de larves a enrichi tout le spectre de l'espèce (n = 49) de 6 %. L'enregistrement des étapes pré-adultes semble donc être nécessaire pour obtenir une image complète d'un peuplement de coléoptères. (ii) Les vingt espèces de larves et d'adultes récoltées semblent bien représenter la composition de carabes autochtones de la zone étudiée. (iii) L'échantillonnage du sol a enrichi le spectre larvaire carabiques et a fourni des données supplémentaires sur le cycle de vie de certaines espèces. Des méthodes complémentaires au piégeage Barber semblent donc être importantes. (iv) Tandis que les larves en été sont principalement semi-édaphiques, l'activité épigée des larves en hiver a surpassé celle des adultes pendant l'automne, l'hiver et le début de printemps. (v) En raison probablement de leur activité épigée plus élevée et de leur rayon d'action plus grand, c'est dans les pièges Barber que le plus grand nombre de larves de troisième stade de la plupart des espèces a été observé. (vi) Les cycles de développement de Nebria brevicollis, Loricera pilicornis, Calathus fuscipes, Amara aulica, Harpalus rufipes, Bembidion lampros et B. properans sont présentés et discutés. Les données larvaires confirment l'idée que ces espèces se reproduisent deux fois par an ou que deux cohortes successives peuvent se reproduire chaque année. (vii) En raison d'une phénologie et d'un habitat qui diffèrent de celui des coléoptères adultes, les larves de carabes pourraient jouer un rôle important dans la régulation de certains parasites.  相似文献

Abstract

On air‐drying, the ATP contents of two moist soils fell to about one quarter of their original values. When a freshly‐sampled soil (field temperature 5.5°C) was stored moist (43% water holding capacity) for 7 days at 25°C the ATP content increased from 4.54 to 7.84 μg ATP g‐1 soil. Storage at 10°C caused a smaller increase; to 5.39 μg g‐1 soil. Microbial biomass C also increased on storage but the relative increase was less than that of ATP. Thus the biomass C/ATP ratio fell from 234 in the freshly sampled soil to 168 in the soil stored moist for 7 days at 25°C. The ATP content declined to less than half its starting value if storage was under waterlogged conditions.

The ATP method for determining microbial biomass in soil depends on the use of a constant factor (5.85 mg ATP g‐1 biomass C) for converting ATP content to biomass C. This factor came from work on soils that had been stored moist at 25°C for several days before biomass C and ATP measurements were made: it is only applicable to soils that have been stored in this way.  相似文献

Several red wines were elaborated to assess the effect of the degree of grape ripening on wine color and on the levels of flavanol and anthocyanin compounds, which are chiefly responsible for the wine color attributes. Two different cultivars and three different degrees of ripening were studied in two consecutive years. The wines were aged for 1 year in American oak barrels of medium-high char followed by 6 months in the bottle. The results showed that the wines made from more mature grapes had generally higher free anthocyanin content, and during aging the decrease of the free anthocyanins and flavanols took place in conjunction with an increase in the levels of the anthocyanin derivatives or "new pigments", which are responsible for maintaining color intensity and adding violet hues in aged wines. From the results obtained, it seems to be clear that the characteristics and composition of grapes harvested later (7-8 days in this region and for these varieties) than the usual time are quite beneficial to obtaining quality aged wines. The phenolic composition of wines made from the last harvested grapes is mainly responsible for the stability of their color, which is extremely important in product acceptance with a significant increase hence of product quality.  相似文献

High African elephant stocking rates in nature reserves in southern Africa have been shown to have significant impacts on vegetation structure and diversity. However, the direct and indirect effects of elephants on fauna, particularly invertebrates, remain poorly known. The Maputaland Centre of Endemism, an area of southern Mozambique and northern KwaZulu-Natal in South Africa, is undergoing rapid rates of habitat conversion. Sand Forest, the rarest vegetation type in the Centre, is threatened by human disturbance outside of protected areas and by elephant browsing pressure in Tembe Elephant Park, one of the few protected areas for Sand Forest. Here we examine the effects of elephant- and human-induced disturbance to Sand Forest on a functionally important component of invertebrate diversity, i.e. the dung beetle fauna. The dung beetle assemblage associated with elephant-disturbed Sand Forest was transitional between undisturbed Sand Forest and Mixed Woodland, whereas that associated with human disturbance was markedly different to either habitat type. Differences are attributed to a change in habitat structure under both disturbance types, as well as to changes in the quality and distribution of dung under human disturbance. Therefore, grazing impacts by elephant not only affect plant diversity, but also implement changes in invertebrate fauna. The maintenance of the integrity of the Sand Forest–Mixed Woodland matrix is considered critical for biodiversity conservation in the Maputaland Centre of Endemism. Management of elephant populations, as well as the protection of sufficient Sand Forest inside conservation areas must, therefore, necessarily form part of the conservation strategy for the Centre.  相似文献

In addition to total organic carbon and nitrogen, potential organic carbon mineralization under controlled laboratory conditions and indicators such as the indicator of remaining organic carbon in soil (IROC), based on Van Soest biochemical fractionation and short-term carbon mineralization in soil, are used to predict the evolution of exogenous organic matter (EOM) after its application to soils. The purpose of this study was to develop near infrared reflectance spectroscopy (NIRS) calibration models that could predict these characteristics in a large dataset including 300 EOMs representative of the broad range of such materials applied to cultivated soils (plant materials, animal manures, composts, sludges, etc.). The NIRS predictions of total organic matter and total organic carbon were satisfactory (R2P = 0.80 and 0.85, ratio of performance to deviation, RPDP = 2.2 and 2.6, respectively), and prediction of the Van Soest soluble, cellulose and holocellulose fractions were acceptable (R2P = 0.82, 0.73 and 0.70, RPDP = 2.3, 1.9 and 1.8, respectively) with coefficients of variation close to those of the reference methods. The NIRS prediction of carbon mineralization during incubation was satisfactory and indeed better regarding the short-term results of mineralization (R2P = 0.78 and 0.78, and RPDP = 2.1 and 2.0 for 3 and 7 days of incubation, respectively). The IROC indicator was predicted with fairly good accuracy (R2P = 0.79, RPDP = 2.2). Variables related to the long-term C mineralization of EOM in soil were not predicted accurately, except for IROC which was based on analytical and well-identified characteristics, probably because of the increasing interactions and complexity of the factors governing EOM mineralization in soil as a function of incubation time. This study demonstrated the possibility of developing NIRS predictive models for EOM characteristics in heterogeneous datasets of EOMs. However, specific NIRS predictive models still remain necessary for sludges, organo-mineral fertilizers and liquid manures.  相似文献

Headspace sorptive extraction (HSSE) and stir bar sorptive extraction (SBSE), two recently introduced solventless enrichment techniques, have been applied to the analysis of the headspace of Arabica roasted coffee and of the headspace of the brew and of the brew itself. In both HSSE and SBSE enrichment is performed on a thick film of poly(dimethylsiloxane) (PDMS) coated onto a magnet incorporated in a glass jacket. Sampling is done by placing the PDMS stir bar in the headspace (gas phase extraction or HSSE) or by immersing it in the liquid (liquid phase extraction or SBSE). The stir bar is then thermally desorbed on-line with capillary GC-MS. The performance of HSSE and SBSE have been compared through the determination of the recoveries and relative abundances of 16 components of the coffee volatile fraction to classical static headspace (S-HS) and to headspace and in-sample solid phase microextraction (HS-SPME and IS-SPME, respectively) applying the fibers PDMS 100 microm, Carbowax/divinylbenzene 65 microm (CW/DVB), Carboxen/PDMS 75 microm(CAR/PDMS), polyacrylate 85 microm(PA), PDMS/divinylbenzene 65 microm(PDMS/DVB), and Carboxen/divinylbenzene/PDMS 50-30 microm(CAR/PDMS/DVB). In all cases, HSSE and SBSE gave higher recoveries, and this is entirely due to the high amount of PDMS applied.  相似文献
