试验用OPS法对小鼠未成熟卵母细胞进行玻璃化冷冻保存研究,并对解冻后小鼠未成熟卵母细胞的成熟情况进行观察。结果表明:小鼠未成熟卵母细胞在10%EG、10%DMSO前处理后,在EFS30、EFS40、EDFS30和EDFS40中平衡15~45s进行冷冻保存,使卵母细胞解冻后的形态正常率最高达92.4%,与对照组无显著差异(P>0.05),成熟率达40.5%。  相似文献

①用EFS30、EFS40、EDFS30、EDFS40四种玻璃化冷冻液对MⅡ期水牛卵母细胞进行毒性试验,结果表明:试验组卵母细胞形态正常率与对照组均无显著性差异(P>0.05);对卵母细胞孤雌激活后EDFS30、EDFS40组的卵裂率与对照组(75.28%)及EFS30、EFS40组差异显著(P<0.05);利用4种冷冻保护剂采用OPS法冷冻保存MⅡ期水牛卵母细胞,其中以EDFS40作为冷冻液时,卵母细胞冷冻解冻后孤雌激活卵裂率最高,达31.60%;以EDFS40作为冷冻液,比较了GMP法和OPS法的冷冻效果,结果表明GMP法冷冻效果好于OPS法。②采用不同预处理时间和平衡时间使用细管法常规冷冻G V期卵母细胞,结果表明预处理5 min、平衡15min组的形态正常率和极体排出率相对较高,分别为72.73%、27.27%。  相似文献

5-6周龄雌性昆明小鼠的成熟卵母细胞,在不同前处理液(10% EG或10% EG 10% DMSO)中平衡5 min,然后在冷冻溶液(EFS30、EFS40、EDFS30或EDFS40)中平衡30 s后进行开放式拉长麦管法(OPS,open pulled straw)和固体表面冷冻法(SSV,solid-surface vitrification)玻璃化冷冻保存研究.试验结果表明:玻璃化冷冻中,多种抗冻保护剂组合使用的效果优于使用单一抗冻保护剂,而且EDFS冷冻液冷冻保存小鼠卵母细胞的效果好于EFS,尤其是EDFS30;SSV法冷冻保存小鼠成熟卵母细胞效果优于OPS法.  相似文献

试验选用5-6周龄雌性昆明小鼠的成熟卵母细胞,在不同前处理液(10%EG或10%EG+10%DMSO)中平衡5min,然后在冷冻溶液(EFS30、EFS40、EDFS30或EDFS40)中平衡30s后进行OPS法和SSV法玻璃化冷冻保存。试验结果表明:(1)小鼠成熟卵母细胞的OPS法冷冻保存,用EFS冷冻的卵母细胞解冻后形态正常率为75.8%,激活后卵裂率为47.8%;在EDFS30中冷冻保存,解冻后形态正常率为87.2%,卵裂率为62.0%;在EDFS40中冷冻保存,解冻后形态正常率为86.6%,卵裂率为57.4%,与对照组(99.0%和80.5%)相比,差异极显著(P〈0.01)。(2)小鼠成熟卵母细胞的SSV法冷冻保存,用EFS冷冻的卵母细胞解冻后形态正常率为82.7%,卵裂率为47.1%;在EDFS30中冷冻保存,解冻后形态正常率为91.5%,卵裂率为57.5%;在EDFS40中冷冻保存,解冻后形态正常率最高为85.1%,卵裂率为55.9%,与对照组(99.0%和80.5%)相比,差异极显著(P〈0.01)。(3)OPS法和SSV法冷冻小鼠成熟卵母细胞解冻后形态正常率为86.5%和91.6%,卵裂率为53.8%和55.6%.与对照绢相比。差异极显著(P〈0.01)。  相似文献

试验采用EFS30、DFS30和EDFS30冷冻液对小鼠MⅡ期卵母细胞进行玻璃化冷冻,并对冷冻-解冻后卵母细胞存活率、纺锤体形态正常率、孤雌激活胚胎的发育能力进行研究。结果表明:利用EDFS30冷冻-解冻卵母细胞后的存活率(98.3%)显著高于EFS30组(88.2%)和DFS30组(89.5%)(P0.05);DFS30组纺锤体形态正常率(44.9%)显著低于其他各组;而孤雌激活后EFS30、DFS30、EDFS30和对照组之间卵裂率(59.1%、56.3%、59.3%和60.0%)、囊胚率(52.0%、53.7%、61.2%和62.1%)和囊胚细胞数(49、48、50、50)均无显著性差异(P0.05)。综上所述,3种冷冻液冷冻卵母细胞孤雌激活后均可获得较好的体外发育能力,其中EDFS30较适宜小鼠MⅡ期卵母细胞冷冻保存。  相似文献

采用OPS管和GMP管对GV期的牛的卵母细胞进行玻璃化冷冻.在不同的前处理液中平衡5 min,然后在冷冻液(EFS30,EFS40,EDFS30或EDFS40)中平衡30 s,进行OPS法和GMP法玻璃化冷冻保存.结果显示,OPS法用EFS40液和EDFS40液冷冻后形态正常卵率为69.6%和76.1%,2组差异显著(P<0.05),成熟率最高达19.2%和33.3%,2组差异显著(P<0.05);GMP法用EFS40液和EDFS40液冷冻后形态正常卵率最高达75.6%和80.8%,2组差异显著(P<0.05),成熟率最高达15.6%和34.9%,2组差异显著(P<0.05).而采用EDFS40液,OPS法和GMP法对GV期卵母细胞体外发育的影响差异均不显著,但GMP法的冷冻效率较高.表明采用EDFS40液GMP法对GV期卵母细胞的冷冻效率优于OPS法.  相似文献

为了提高玻璃化冷冻保存山羊卵母细胞的效果,试验用不含Ca2+的玻璃化冷冻液1[30%乙二醇(EG)]、玻璃化冷冻液2[27.5%EG+2.5%二甲基亚砜(DMSO)]和玻璃化冷冻液3(25%EG+5%DMSO)研究抗冻保护剂DMSO和EG的不同浓度配比对山羊卵母细胞形态正常率、孤雌激活率、体外受精卵裂率及胚胎发育能力的影响。结果表明:山羊卵母细胞经玻璃化冷冻液1,2,3处理后细胞形态正常率(97.1%、97.3%、97.3%)与对照组(100%)相比有所降低,但差异不显著(P>0.05),且对卵母细胞均不产生具有化学毒性作用的孤雌激活;山羊卵母细胞经3种玻璃化冷冻液冷冻保存后,玻璃化冷冻液2冷冻保存卵母细胞的囊胚率(13.8%)显著高于玻璃化冷冻液1(5.7%)和玻璃化冷冻液3(5.1%)(P<0.05)。说明采用玻璃化冷冻液2冷冻保存山羊卵母细胞可获得较为理想的效果。  相似文献

本实验旨在探讨玻璃化冷冻保存对猪MⅡ期卵母细胞皮质颗粒分布和孤雌激活后早期胚胎发育能力的影响。实验将卵母细胞随机分为对照组、毒性实验组和冷冻组。采用EFS40和EDFS40两种玻璃化冷冻液处理,卵母细胞经恢复后对其进行染色,观察皮质颗粒的分布;并对另一部分卵母细胞实施孤雌激活,观察早期胚胎的发育。结果表明:毒性实验组和冷冻组卵母细胞皮质颗粒部分释放、完全释放的比例无显著差异,但均显著高于对照组(P<0.05)。不同毒性处理组和不同冷冻组间对皮质颗粒的分布无显著差异。与毒性实验组相比,冷冻处理组显著降低皮质颗粒在皮质区分布的比例(P<0.05)。EFS40毒性实验组孤雌激活后的存活率、卵裂率、囊胚发育率均显著高于EDFS40毒性实验组(86.6%vs.75.0%)、(61.8%vs.40.7%)、(30.2%vs.23.5%)(P<0.05)。EFS40冷冻组存活率显著高于EDFS40冷冻组,但均显著低于对照组。结果显示,抗冻保护剂处理和玻璃化冷冻均导致猪卵母细胞皮质颗粒释放,与EDFS40相比采用EFS40较适合猪MⅡ期卵母细胞冷冻保存。  相似文献

以冷冻保存后的小鼠卵母细胞为实验材料,研究其膜对体外受精效果的影响。在室温(25±0.5)℃条件下,以10%乙二醇(EG)+10%二甲基亚砜(DMSO)为预处理液,EDFS30为玻璃化溶液。将卵母细胞于10%EG+10%DMSO溶液中预处理30s,然后再移入EDFS30中处理25s,以开放式拉长细管(OPS)为承载器投入液氮中,即两步法玻璃化冷冻保存。结果表明:冷冻卵母细胞受精后的卵裂率(46.67%)显著低于新鲜组的(86.06%)(P<0.05)。冷冻卵母细胞去透明带后质膜上的平均精子结合数(10.70)与对照组(10.81)无显著性差异(P>0.05),形成雌雄原核数也无显著性差异(2.49vs.2.59)(P>0.05)。冷冻卵母细胞透明带打孔后,其体外受精后卵裂率与新鲜组差异不显著(84.73%vs.91.19%)(P>0.05)。因此玻璃化冷冻保存小鼠卵母细胞透明带的变化是影响体外受精效果的主要因素之一。  相似文献

比较了不同冷冻方法和冷冻保护液对山羊卵母细胞冷冻-解冻后体外受精和激活后的发育效果.结果表明,开放式拉长塑料细管(OPS)法的效果好于玻璃化细管法和常规冷冻法;在OPS法中,20%EG 20%DMSO对卵母细胞的冷冻保护效果好于EDFS40,而在玻璃化细管法冷冻中,则是EDFS40好于20%EG 20%DMSO.对于常规冷冻法而言,则是1.5mol/L EG的冷冻效果好于1.5mol/L PROH.  相似文献

1-(2-Chloroethyl)3-cyclohexyl-1-nitrosourea (CCNU) is an alkylating agent in the nitrosourea subclass. A prospective evaluation of CCNU was done to determine the maximally tolerated dosage of CCNU in tumor-bearing cats. Response data were obtained when available. Twenty-five cats were treated with CCNU at a dosage of 50-60 mg/m3 body surface area. Complete hematologic data were available for 13 cats. Neutropenia was the acute dose-limiting toxicity. The median neutrophil count at the nadir was 1,000 cells/microL (mean, 2,433 cells/microL; range, 0-9,694 cells/microL). The time of neutrophil nadir was variable, occurring 7-28 days after treatment, and counts sometimes did not return to normal for up to 14 days after the nadir. Based on these findings, a 6-week dosing interval and weekly hematologic monitoring after the 1st treatment with CCNU are recommended. The nadir of the platelet count may occur 14-21 days after treatment. The median platelet count at the nadir was 43,500 cells/microL. No gastrointestinal, renal, or hepatic toxicities were observed after a single CCNU treatment, and additional studies to evaluate the potential for cumulative toxicity should be performed. Five cats with lymphoma and 1 cat with mast cell tumor had measurable responses to CCNU. Phase II studies to evaluate antitumor activity should be completed with a dosing regimen of 50-60 mg/m3 every 6 weeks.  相似文献

Human and equine athletes are reported to have a high prevalence of gastric disease, and anecdotal evidence suggests a similar phenomenon applies to racing sled dogs. To investigate the prevalence of gastric disease in racing sled dogs, we conducted 2 gastroscopy studies on dogs competing in the annual Iditarod Sled Dog Race. A pilot study of dogs that were either dropped from the 2000 Iditarod Sled Dog Race because of illness or that finished the race indicated that, approximately 5 days after competing, 10 of 28 dogs (35%) had endoscopic evidence of gastric ulceration, erosion, or hemorrhage. The next year, an endoscopic study of 73 dogs participating in the 2001 Iditarod race was performed in order to evaluate a larger population of dogs. Data from 70 of these dogs could be used; 34 (48.5%) had ulceration, erosion, gastric hemorrhage, or some combination of these findings. When this group of 70 dogs was compared retrospectively to a control group of 87 dogs presented to the Texas A&M University (TAMU) Veterinary Medical Teaching Hospital, the Iditarod sled dogs had a significantly higher prevalence (P = .049) of gastric lesions. These findings suggest that, similar to athletes of other species, elite canine athletes have an increased prevalence of gastric disease compared to the canine population at large.  相似文献

Continuous rate infusion (CRI) of furosemide in humans is considered superior to intermittent administration (IA). This study examined whether furosemide CRI, compared with IA, would increase diuretic efficacy with decreased fluid and electrolyte fluctuations and activation of the renin-angiotensin-aldosterone system (RAAS) in the horse. Five mares were used in a crossover-design study. During a 24-hour period, each horse received a total of 3 mg/kg furosemide by either CRI (0.12 mg/kg/h preceded by a loading dose of 0.12 mg/kg IV) or IA (1 mg/kg IV q8h). There was not a statistically significant difference in urine volume over 24 hours between methods; however, urine volume was significantly greater after CRI compared with IA during the first 8 hours ([median 25th percentile, 75th percentile]: 9.6 L [8.9, 14.4] for CRI versus 5.9 L [5.3, 6.0] for IA). CRI produced a more uniform urine flow, decreased fluctuations in plasma volume, and suppressed renal concentrating ability throughout the infusion period. Potassium, Ca, and Cl excretion was greater during CRI than IA (1,133 mmol [1.110, 1,229] versus 764 mmol [709, 904], 102.7 mmol [96.0, 117.2] versus 73.3 mmol [65.0, 73.5], and 1,776 mmol [1,657, 2.378] versus 1,596 mmol [1,457, 1,767], respectively). Elimination half-lives of furosemide were 1.35 and 0.47 hours for CRI and IA, respectively. The area under the excretion rate curve was 1,285.7 and 184.2 mL x mg/mL for CRI and IA, respectively. Furosemide CRI (0.12 mg/kg/h) for 8 hours, preceded by a loading dose (0.12 mg/kg), is recommended when profound diuresis is needed acutely in horses.  相似文献

The epidemiology of H. placei and of other gastrointestinal nematodes in yearling dairy cattle was examined on two farms in Kiambu District, central Kenya during each of 13 one-month periods from April 1993 to April 1994. On each farm, 32 newly weaned dairy calves were given a single dose of albendazole and then placed on experimental pastures. Twelve of the animals were designated for bi-monthly slaughter (n = 2) and analysis of worm population characteristics and 20 were designated for blood and faecal collection and for weighing. Two parasite-free tracer calves were grazed alongside the weaner calves each month throughout the study period and were also slaughtered for analysis of worm populations. Faecal egg counts, haematological and serum pepsinogen determinations, herbage larval counts, and animal live weight changes were recorded monthly. The study revealed that Haemonchus placei, Trichostrongylus axei, Cooperia spp. and Oesophagostomum radiatum were responsible for parasitic gastroenteritis and that H. placei was the predominant nematode present in the young cattle on both farms. Faecal egg counts from resident cattle and necropsy worm counts revealed that pasture larval levels were directly related to the amount of rainfall. The total worm burdens in the animals were highest during the rainy season (March–June and October–December) and lowest during the dry seasons (July–September and January–February). The very low recovery of immature larvae of H. placei from the tracer calves indicated that arrested development is not a feature of the life cycle of this parasite in central Kenya. The maintenance of the parasite population depended on continuous cycling of infection between the host and the pasture. The agroclimatic conditions of the study area were such that, in general, favourable weather conditions for the development and survival of the free-living stages of gastrointestinal nematodes existed all year round.  相似文献

The occurrence of ectoparasites in sheep flocks is frequently reported but seldom quantified. Sheep production used to be a predominantly family activity in the state of Sa~o Paulo (Brazil), but it began to become a commercial activity in the past decade. Thus, information about the ectoparasites existing in sheep flocks has become necessary. The present data were obtained by means of questionnaires sent to all sheep breeders belonging to the `Associaça~o Paulista de Criadores de Ovinos' (ASPACO; Sa~o Paulo State Association of Sheep Breeders). Response reliability was tested by means of random visits paid to 10.6% of the respondents. Most of the properties (89.5%) reported the presence of one or more ectoparasites. Screw-worm (Cochliomyia hominivorax) was the most frequent ectoparasite (72.5%), followed by bot fly larvae (Dermatobia hominis, 45.0%), ticks (Amblyomma cajennense) and Boophilus microplus, 31.3%) and finally lice (Damalinia ovis, 13.8%). Combined infestations also occurred, the most common one being screw-worm with bot fly larvae (36.0%) followed by bot fly larvae with ticks (13.9%), screw-worm with ticks (9.3%), bot fly larvae with lice (6.9%), and ticks with lice (5.0%). The most common triple combination was screw-worm, bot fly larvae and ticks (12.8%). Breeds raised for meat or wool were attacked by bot fly larvae and ticks more often than other breeds. Lice were only absent from animals of indigenous breeds. The relationships among these ectoparasites are discussed in terms of sheep breeds, flock size, seasonality and the ectoparasitic combinations on the host.  相似文献

Our particular attention in this article was given to natural mediators for macrophages isolated from the sites of tissue injury. A number of chemotactic factors, which may satisfy many criteria making them acceptable as inflammatory leucocyte chemotactic factors, has been separated. Among them, our laboratory has isolated three macrophage (monocyte) chemotactic factors (MCF-a, -b and -c). Their purification, characterization and functional specificity are discussed.  相似文献

An experiment was conducted to study disappearance of C14 to C18 fatty acids, lag times and biohydrogenation (BH) rates of C18 fatty acids of ground soya bean and canola seeds in situ. Three ruminally fistulated Dallagh sheep were used to determine ruminal BH of unsaturated fatty acids (UFAs). Differences in the disappearance of fatty acids through the bags and lag times were observed between the oilseeds. We saw that the longer the incubation time of the oilseeds in the rumen, the lower the content of C18:2 and C18:3. Significantly higher lag times for both C18:2 and C18:3 were observed in ground canola compared to ground soya bean. BH rates of C18:2 and C18:3 fatty acids in soya bean were three times higher than those of canola. These results suggest that the fatty acid profile of fat source can affect the BH of UFAs by rumen micro‐organisms. So that UFAs of canola had higher ability to escape from ruminal BH. It seems that fatty acid profile of ruminant products is more affected by canola seed compared to soya bean seed.  相似文献

Serum biochemical values and the wet weight to dry weight ratios of tissues were determined in yaks in Shandan county of Gansu province. The liver, kidney, heart and muscle contents of seven elements in yaks were also analysed. Most serum biochemical values were similar to those of cattle, camels and sheep, but the calcium concentration was considerably above the normal range for other ruminants.
