根据GenBank上公布的牛布氏杆菌外膜蛋白OMP31的基因序列设计特异性引物,对牛布氏杆菌样品进行PCR扩增并将产物克隆到pMD18-T载体后测序。结果表明,扩增的目的片段大小为602 bp,与预期扩增序列同源性为99.7%;该PCR检测体系的特异性强,不能在非布氏杆菌 DNA中扩增出条带;敏感性高,最低检测的DNA含量为0.9 pg/μL。该检测体系的成功构建为牛布氏杆菌病的检测、鉴定和流行病学调查提供了有力的技术支持。  相似文献

免疫组化法检测猪圆环病毒2型在人工感染猪体内的分布   总被引:3,自引:0,他引:3  
将猪圆环病毒2型(PCV2)BF株经滴鼻接种28日龄健康普通仔猪,于接种后不同时间剖杀,采集相关的器官组织制备组织切片,使用免疫组化方法检测病毒在猪体内的分布。结果显示,淋巴结、脾脏、扁桃体等淋巴组织均出现阳性信号,心、肝、肺、肾、胃、十二指肠、大脑及小脑同样存在阳性细胞,其他组织呈阴性反应。阳性信号主要位于感染细胞的胞质中,偶尔出现在细胞核内。结果表明,PCV2确已造成猪体组织感染。  相似文献

奶水牛布氏杆菌病是由布鲁氏杆菌感染而诱发的人畜共患传染病,多为慢性经过,临床症状表现为母牛流产、公牛睾丸炎、前列腺炎、精囊炎等,会导致奶水牛丧失繁殖功能,造成奶水牛养殖极大损失。因此,奶水牛布氏杆菌病的防控工作对奶水牛产业发展是非常重要的。多年来,笔者在灵山县负责奶水牛布氏杆菌病的防控工作,现总结提出奶水牛布氏杆菌病的一些防控措施供,大家参考。  相似文献

Seven of 18 elk on a deer farm were found by the official Rose‐Bengal agglutination test (RBT) and tube agglutination test to be brucellosis reactors/suspects. Evaluation with the competitive ELISA (C‐ELISA) and the fluorescence polarization assay (FPA) tests revealed that six and five sera were positive respectively. The seven reactors/ suspects were slaughtered and their blood and tissues were collected. Brucella species could be isolated from three of the slaughtered animals, with nine isolates being obtained from the popliteal, supramammary and submandibular lymph nodes, vaginal discharge, mammary tissue and spleen. Brucella genus‐specific PCR based on 16S rRNA and AMOS‐PCR, which is specific for differential Brucella species, revealed that all nine isolates were Brucella abortus. These nine were further confirmed to be B. abortus biovar 1 by classical biotyping scheme assays. This is the first report of an outbreak of brucellosis in domestic elk in Korea. Our observations suggest that deer should be included in the routine Brucella surveillance programme for the effective control and prevention of brucellosis in Korea.  相似文献

为证实改进的试管凝集试验(SAT)的使用价值,分别用微量试管凝集试验(MSAT)和SAT以国际标准血清为参比检测6份牛布鲁氏菌病阳性血清的抗体效价。结果表明:MSAT和SAT试验结果一致,而且MSAT具有稀释方法更简便、精确,省时省力,可节约血清、抗原等试验原材料,试验结果容易判定,重现性良好等优越性。  相似文献

本研究通过布氏流产杆菌(Brucella abortus)国际标准参考血清(ISABS)的使用,对目的应用的补体结合试验(CFT)、酶联免疫吸附测定法(ELISA)、虎红平板凝集试验(RBPT)及试管凝集试验(SAT)等四种血清学检测方法的测定结果进行了校正和临界点的设定。并对42份不同抗体效价血清进行了测定,同时通过计算和应用标准参考血清校正系数,对不同方法间的测定结果如何换算成国际标准单位进行了研究和探讨。结果表明,该校准方法可对不同实验室间的布氏杆菌病检测结果校准到统一的国际标准上来,可作为血清学测定国际标准化一条途径。  相似文献

对79只确诊为犬瘟热的病貉进行CDV在组织内的定位。采用HE染色检测到包涵体出现在肺(57只貉)、脑(19只貉)、肾(58只貉)、胃(63只貉)、脾(61只貉)、淋巴结(63只貉)、扁桃体(79只貉)。采用免疫组化方法检测出上述器官之外在足垫上皮细胞(70只貉)、鼻黏膜(69只貉)、支气管(71只貉)也有强阳性表达。结果表明,采用免疫组化方法能更加准确的对CDV在犬瘟热病貉组织内分布的定位,CDV分布最广泛的器官是扁桃体。  相似文献

One hundred and seven pregnant cows, which had been calfhood vaccinated with Brucella abortus strain 19 (S-19) were revaccinated with either S-19 or strain RB51 (S-RB51). All S-19-revaccinated animals seroconverted, while none of the RB51-revaccinated animals seroconverted. Two out of 25 (8%) S-19-revaccinated animals aborted, while none of the 57 RB51-revaccinated group aborted. Four of the S-19-revaccinated animals shed S-19 in the milk for at least 7 days, while only 1 cow shed S-RB51 for at least 3 days (but <7 days) post-parturition. Revaccination of strain 19 calfhood-vaccinated, pregnant cattle with S-RB51 appears to be a safe procedure with no diagnostically negative consequences.  相似文献

Brucella preferentially infects cattle, swine, sheep, and goats. However, some epidemiological surveys have been carried out to investigate nonruminants, such as horses. Horse brucellosis has been found in clinical cases, but there are few epidemiologic patterns. Between May 2008 and April 2009, a total of 120 horses were screened for brucella infections in Mashhad, Iran, by the rose bengal test and the tube agglutination test. Sera from three horses were found positive by rose bengal test and tube agglutination test, and therefore the prevalence rate was 2.5%. In horses, the highest individual seroprevalence was in an animal kept close under the intensive system, with other animals such as cattle, sheep, and goats. The zoonotic aspects of brucellosis from the horse must, therefore, be considered because the disease is important from a public health standpoint. The present study documents the first serological evidence of Brucella spp. infection in horses in Iran.  相似文献

A captive adult female bottlenose dolphin presented with stillbirth. The placenta appeared oedematous. No other gross lesions were evident in the placenta or the stillborn calf. Histopathology revealed mild multifocal placentitis and foetal encephalitis. Brucella sp. was isolated from lung, liver, spleen and kidney. Sequence and phylogenetic analysis demonstrated this organism to be most similar to Brucella ceti sequence type (ST) 27. Brucella sp. DNA was detected in formalin-fixed paraffin-embedded placenta and brain by real-time PCR using primers targeting the IS711 gene. Immunohistochemical staining revealed Brucella sp. antigen in placental inflammation. This is the first report of isolation of Brucella sp. from a marine mammal in the Southern Hemisphere and the first report of marine Brucella-associated disease in Australia.  相似文献

本研究在国内首次成功建立了辣根过氧化物酶标记的链霉亲和素-生物素(LSAB)免疫组化染色法检测猪生殖-呼吸道综合征病毒(PRRSV)抗原。应用LSAB染色技术检测12头人工感染PRRSV美洲株(ATCCVR-2332)或国内分离株(B96-4,B96-5)的SPF仔猪组织细胞内的PRRSV抗原,阳性检出率为100%。LSAB免疫组化染色法操作简单、灵敏度高、特异性强,结果清晰、稳定、重复性好,是一种检测组织细胞内PRRSV抗原的有效方法。  相似文献
