The importance of Anoplocephala perfoliata in horses with colic was studied in 139 horses referred for colic and 139 control horses with no signs of colic for at least three years. The serodiagnostic method of Proudman and Trees, which measures the level of A. perfoliata antibody, was used to detect A. perfoliata infection. Thirty-two horses were examined at necropsy, to determine whether the presence of A. perfoliata in the ileocaecal region was associated with the A. perfoliata antibody level. The mean A. perfoliata antibody level was significantly higher in horses with colic than in horses without colic (P < 0.001), indicating a relationship between A. perfoliata infection and colic in general. There was no relation between age and A. perfoliata antibody level. The mean A. perfoliata antibody level in 12 horses with ileocaecal disorders was significantly higher than that in control horses (P < 0.001). Of the 32 horses examined at necropsy, 7 horses with tapeworms in the ileocaecal region had a significantly higher mean A. perfoliata antibody level than the 25 horses without the parasite (P = 0.030). Lastly, examination of faeces to detect the presence of A. perfoliata infection was not useful in the present study.  相似文献

The serological changes in two groups of horses known to be harbouring Anoplocephala perfoliata were studied; 12 were treated with 1.5 mg/kg praziquantel and 200 microg/kg ivermectin, and 14 were treated with 200 microg/kg ivermectin. Serological and faecal analyses were carried out on each animal at intervals for 758 days. The titres of antibodies specific for A perfoliata decreased from the day of treatment to day 28 in both groups, and continued to decrease in the group treated with praziquantel and ivermectin, with the first significant decrease from the other group at day 121. From day 151 to day 295 the first significant increase in antibody levels in the group treated with both drugs was observed; no A perfoliata eggs were detected in the faeces of these animals until day 295 when five of the 10 were positive.  相似文献

OBJECTIVE: To determine whether specific feeding practices were associated with development of colic in horses. DESIGN: Prospective matched case-control study. ANIMALS: 364 horses examined by veterinarians in private practice in Texas because of colic (cases; n = 182) or any other reason (controls; 182). PROCEDURE: Participating veterinarians were sent forms at the beginning of the study to collect information on signalment, feeding management practices, farm management practices, and preventive medical treatments. Case and control horses were compared by use of conditional logistic regression to identify factors associated with colic. RESULTS: Risk factors for colic were a recent change in batch of hay, decreased exposure to pasture, a recent change in type of grain or concentrate fed, feeding > 2.7 kg (6 lb) of oats/d, feeding hay from round bales, and Thoroughbred breed. Recent anthelmintic administration decreased the risk of colic. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that certain changes in diet (eg, change in batch of hay, change in type of grain or concentrate, feeding hay from round bales) and management (eg, decreased availability of pasture) increase the risk of colic in horses.  相似文献

Objective To compare the sensitivities of three coprological techniques for the diagnosis of Anoplocephala perfoliata infection in horses and to assess the value of the methods for diagnosis of horses at risk of clinical cestodiasis.
Design Faecal samples were collected from necropsied horses with or without A perfoliata infection and examined using one sedimentation and two different flotation methods. The coprological results were compared with worm counts performed at necropsy of the horses and the degree of mucosal damage. In addition, the efficiency of recovery of A perfoliata eggs from faeces was tested.
Results The overall sensitivities of the methods ranged from 22.5 to 37.5%, and the capacity of the methods to diagnose infection increased with the intensity of infection. A simple flotation method achieved a better sensitivity (37.5%) at all intensities of infection compared with the other two methods (22.5 to 25%). That method was also more sensitive in detecting eggs in 'negative' faecal samples spiked with known numbers of A perfoliata eggs.
Conclusion The results indicated that, despite the low sensitivities of present methods, faecal flotation is likely to be of value in detecting horses at risk of clinical disease.  相似文献

A 12/13 kDa antigen, tapeworm ELISA test, developed for use in horses, was used to detect parasite-specific serum antibody, IgG(T), in the serum of donkeys. In a pilot study the 12/13 kDa antigen was tested and proved to detect the antibody, IgG(T), in donkey sera. Blood samples from 797 donkeys, naturally exposed to cestode infection, from four geographical localities were collected and sera were prepared and analysed. There was substantial serological evidence that donkeys were potentially infected with A. perfoliata. A range of ELISA OD values were obtained from the serological assay. Over 26% and 7.5% of the donkeys were moderately and highly infected, respectively, showing at least a 34% sero-prevalence. The rest, 66.1%, were either with low infection intensity or negative for A. perfoliata infection. The risk of infections, both in sero-prevalence and intensity, as determined by ELISA optical density (OD), were highest in the highland areas of Ethiopia where pastures are low-lying and wet, and permanent pasture management is regularly practised. Sex, age and body condition of the donkeys had no significant effect either on prevalence of the infection or on the serum antibody level. These results indicate a risk of intestinal disorders, particularly, colic, associated with A. perfoliata infection in donkeys.  相似文献

Intestinal helminths are an important cause of equine disease. Of these parasites, the Cyathostominae are the commonest group that infect horses. These nematodes consist of a complex tribe of 51 species, although individual horses tend to harbour 10 or so common species, in addition to a few rarer species. The Cyathostominae can be extremely pathogenic, and high levels of infection result in clinical symptoms ranging from chronic weight loss to colic, diarrhoea and death. As part of their life cycle, immature cyathostomins penetrate the large intestinal wall, where they can enter a state of inhibited larval development. These larvae can exist in this state for months to years, after which they subsequently re-emerge. If larvae re-emerge in large numbers (i.e. several million), severe pathological consequences ensue. The inhibited larvae are also relatively refractory to several of the currently available anthelmintics, so that horses treated previously with anthelmintics can still carry life-threatening burdens of these parasitic stages. Little is known about the cyathostomin larvae during their mucosal phase, and current research efforts are focused on investigating the biology of these stages. Much of the research described here highlights this area of research and details studies aimed at investigating the host immune responses that the mucosal larvae invoke. As part of this research effort, molecular tools have been developed to facilitate the identification of larval and egg stages of cyathostomins. These molecular tools are now proving very useful in the investigation of the relative contributions that individual, common cyathostomin species make to the pathology and epidemiology of mixed helminth infections. At the more applied level, research is also in progress to develop an immunodiagnostic test that will allow numbers of mucosal larvae to be estimated. This test utilises antigen-specific IgG(T) serum antibody responses as markers of infection. As anthelmintic resistance will be the major constraint on the future control of the Cyathostominae, researchers are now actively investigating this area and studies aimed at elucidating the molecular mechanisms of drug resistance are described. Another parasite which has assumed a clinically important role in horses is the tapeworm, Anoplocephala perfoliata. This parasite is prevalent world-wide and has been shown to be a significant cause of equine colic. Because previous methods of estimating the infection intensity of tapeworm were inaccurate, recent research has been directed at developing an immunodiagnostic ELISA for these cestodes. Specific IgG(T) responses to antigens secreted by adult tapeworms have been shown to provide a reasonable indication of infection intensity. An ELISA based on these responses is now commercially available. The steps involved in the development of this ELISA are described here. In addition to these recent advances in research, this review also outlines the principle areas for future research into these important equine parasites.  相似文献

Sourcing of horses naturally infected with parasites such as Anoplocephala perfoliata and Gasterophilus intestinalis for antiparasitic efficacy slaughter studies is often difficult, expensive, and usually excessive numbers of animals must be slaughtered before significant data can be obtained. To overcome this problem a "Modified Critical Treatment Method" was developed. This method does not require the horses to be sacrificed, but relies on the collection and processing of all faecal material containing expelled parasites from test animals. Efficacy is assessed by comparing, in the same horses, the numbers of parasites expelled with the test compound with the total numbers removed with test and reference endectocides. This, for the most part, means that farms that were previously unavailable, e.g. studs, can now be used for the collection of efficacy data.  相似文献

Distal ileum, caecum and proximal colon of 470 horses were examined for helminths during 1 year at an abattoir in central Sweden. The infection levels of the horse tapeworm Anoplocephala perfoliata, their stage of development, site of attachment and gross pathological lesions caused by the worm were recorded. Faecal samples from 395 of the horses were examined specifically for tapeworm segments and eggs in order to correlate these findings with the numbers in the alimentary canal. In total 65% of the horses were infected with A. perfoliata and the mean intensity of infection was 79 worms per infected horse with a maximum of 912. The level of infection was significantly higher in (1) 3rd and 4th than in 1st and 2nd quarter of the year; (2) older horses than in yearlings; (3) females than in males and geldings; (4) thoroughbreed and cold-blooded horses than in Swedish standardbreeds and ponies. The level of infection was unaffected by the usage of anthelminthics against nematodes. Of the horses examined 51% had 1-100 worms whereas 14% were infected with more than 100 worms. Of the tapeworm positive horses 72% had mixed infections with both adult and juvenile worms, 20% solely juveniles, and 8% solely adults. The severity of intestinal lesions exacerbated by increasing numbers of A. perfoliata. About 11% of the intestines examined had severe lesions, but there was no history of acute abdominal distress in any of the horses included in this study. Although the number of detectable eggs was significantly higher for horses heavily infected with A. perfoliata, the egg recovery among infected horses was only 35%. An additional field survey comprising 218 horses on 88 premises in central and southern parts of Sweden showed that the prevalence of A. perfoliata egg positive horses was the same as found on faecal examination during the abattoir survey.  相似文献

Anoplocephala perfoliata (Cestoda, Cyclophyllidea), the commonest intestinal tapeworm of horses, can cause colic, intussusceptions, ileal impactions and intestinal perforations. Common diagnostic techniques for A. perfoliata infection, i.e. coprology and serology, show inherent limitations in terms of sensitivity and specificity and new approaches are thus required. Hence, the present study compared the reliability of coprological, serological (i.e. ELISA) and molecular (i.e. nested PCR) methods in detecting A. perfoliata infection in naturally infected horses and in horses treated with a combination of ivermectin and praziquantel. Of 42 horses subjected to coprological examination, 16 and 26 resulted negative and positive, respectively for the presence of A. perfoliata eggs at the coprological examination. The 26 coprologically positive animals were also positive by nested PCR. Fifteen out of the 16 horses coprologically negative were negative at the molecular assay, while one yielded a PCR product detectable on an agarose gel. Eighteen out of 26 positive horses were treated with a combination of ivermectin 18.7 mg/g and praziquantel 140.3mg/g and resulted subsequently negative by coprology and nested PCR performed 2 weeks after treatment. All infected and untreated animals had a high ELISA test optical density indicating high infection intensity and associated risk of colic. However, high optical density values were also obtained in four horses post-treatment and in three horses that were negative on molecular and coprological analysis. The results of the present work indicate that the nested PCR assay represents a valid method for the specific molecular detection of A. perfoliata in faecal samples collected from naturally infected horses and may have advantages over coprological and serological approaches for diagnosing A. perfoliata infection.  相似文献

OBJECTIVE: To determine whether dietary and other management factors were associated with development of colic in horses. DESIGN: Prospective matched case-control study. POPULATION: 2,060 horses examined by veterinarians in private practice in Texas for colic and noncolic emergencies. PROCEDURE: Each month for 12 months, participating veterinarians were sent forms to collect information on 1 horse with colic and 1 horse that received emergency treatment for a condition other than colic, information collected included signalment, farm management and characteristics, diet, medical and preventive medical factors, transport, and activity or use. Case and control horses were compared by means of conditional logistic regression to identify factors associated with colic. RESULTS: Recent change in diet, recent change in type of hay, history of previous episode of colic, history of abdominal surgery for colic, recent change in weather conditions, recent change in housing, Arabian breed, administration of an anthelmintic during the 7-day period prior to examination, failure to receive regular deworming, age > 10 years, and regular exercise (vs pastured at all times) were associated with increased risk of colic. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that changes in diet (particularly in type of hay fed) contribute to increased risk of colic. A regular program for administration of anthelmintics may reduce the overall frequency at which colic develops, but recent administration of anthelmintics may predispose some horses to colic. Arabian horses may have an increased risk of colic, and horses at pasture may have a decreased risk of colic.  相似文献

A cross-sectional study was undertaken (October 2010 to August 2011) to determine the risk factors for dairy goat herds and dairy sheep flocks testing positive for paratuberculosis (PTB) in Ontario, Canada. A questionnaire was administered to 50 producers during a farm visit in which concurrently, 20 randomly selected, lactating animals over the age of 2 years underwent sampling for paratuberculosis testing. Only 1 of 50 farms (2.0%) was closed to animal movement, whereas 96.6% of dairy goat farms and 94.1% of sheep farms purchased livestock from other producers. Only 10.3% of dairy goat, and no dairy sheep farms used artificial insemination. Manure was spread on grazing pastures by 65.5% and 70.6% of dairy goat and dairy sheep farms, respectively. Because of the high true-prevalence of paratuberculosis infection detected, no risk factor analysis could be performed. This study demonstrates that biosecurity practices conducive to transmission of PTB are highly prevalent in Ontario small ruminant dairy farms.  相似文献

A total of 396 Thoroughbreds (86 males, 293 females, and 17 geldings) in Kentucky, 1 to 30 years of age, were examined at necropsy for presence of Anoplocephala perfoliata (2 female horses were not examined) in the cecum for lesions of Draschia megastoma in the stomach, during the 1-year period of Mar 1, 1982 through Feb 28, 1983. Prevalence of A perfoliata was 53% and of lesions of D megastoma was 63%. All lesions of D megastoma were partially or entirely in the glandular region of the stomach. Distances of the midpoint of these lesions from the margo plicatus varied from 0 to 290 mm. The center of about 88% of the lesions was within 50 mm of the margo plicatus . Measurements of the size of the lesions, exclusive of masses, indicated that about 95% were less than or equal to 50 mm in diameter. There appeared to be a higher infection rate of lesions of D megastoma in younger horses (81% in 1 to 7 year olds) than in older horses (41% in 8 to 30 year olds) with a few exceptions.  相似文献

The objective of this study was to investigate the associations between herd-level bovine leukemia virus (BLV) status and herd-level management and production variables. The study population consisted of 1330 cows sampled from 102 Ontario dairy herds. The individual-cow prevalence of BLV infection in the population (based on AGID testing) was 23%, with 69.6% of herds having one or more positive animals. The herd-level explanatory variables were divided into two datasets containing winter housing variables and all non-seasonal variables, and summer housing variables and all non-seasonal variables. In both datasets, multivariable analyses found a negative association between herd-level milk production and BLV status, and positive associations between weaning age and purchasing animals from outside sources, and BLV status. Housing pre-weaned calves in hutches or separate calf buildings in either season was associated with an increased risk of BLV. The model containing winter housing variables also included positive associations between contact with older animals and BLV status, and between BLV status and the facilities used to house dry cows in the winter.  相似文献

Cystotomy is a common surgical procedure in small animal veterinary medicine, yet common pre-, intra-, and post-operative practices have not been described. This survey evaluated cystotomy practices of 106 veterinarians in Ontario. The majority of respondents reported practices consistent with standard recommendations, but some deficiencies in antimicrobial and analgesic use, as well as intra- and post-operative practices, were identified. Some factors associated with the likelihood that practitioners reported recurrent urolithiasis or urinary tract infection are contrary to typical recommendations, such as the association of the use of absorbable, multifilament suture or a dorsal (versus ventral) incision and a lesser likelihood of reporting post-operative urinary tract infections. While care must be taken interpreting these statistical associations, the results suggest that objective assessment of common cystotomy recommendations (use of monofilament, absorbable suture) is required. Re-assessment of certain peri-operative practices, such as analgesic and antimicrobial administration, and post-operative testing, is required for a minority of practitioners.  相似文献
