Reduction of plant height has played a significant role in improving wheat production and knowledge of dwarfing genes in Chinese wheat will be very important for developing high yielding cultivars. Molecular markers were used to detect the presence of genes Rht-B1b (Rht1), Rht-D1b (Rht2) and Rht8 in 220 wheat genotypes from autumn-sown wheat regions in China. They include landmark landraces, leading cultivars and core parents involved in wheat breeding from the 1950s to the present. Results indicated that Rht-D1b and Rht8 dominate with frequencies of 45.5% and 46.8%, respectively, followed by Rht-B1b with a frequency of 24.5%. The frequencies of Rht-B1b and Rht-D1b increased, from 8.6 to 32.2% and 36.2 to 53.4%, respectively, whereas the frequency of Rht8 has remained constant over time, when compared with cultivars released before and after 1990. This indicates that both the Rht-B1b and Rht-D1b were successfully used in wheat production in Chinese environments. Our study shows that Rht-B1b and Rht-D1b can be used in the post-anthesis heat stressed environments. Rht-B1b in Chinese wheats is derived from two sources, viz., Norin 10 and the Italian introduction St2422/464 (Rht-B1b and Rht8). The identity of Rht-B1b in these two sources still needs to be confirmed. Suwon 86 carrying both Rht-B1b and Rht-D1b, and Chinese cultivars, Huixianhong and Yaobaomai, are the primary sources of Rht-D1b in Chinese wheats. It is likely that Rht-D1b in Youbaomai derives from an unknown introduction. Italian introductions such as Funo and Abbondanza, and Lovrin 10 with the 1B/1R translocation, and Chinese landraces are the three major sources of Rht8. This information will be very valuable for wheat breeding in China, and internationally.  相似文献

Fusarium head blight (FHB), leaf rust and stem rust are among the most destructive wheat diseases. High‐yielding, native disease resistance sources are available in North America. The objective of this study was to map loci associated with FHB traits, leaf rust, stem rust and plant height in a “Vienna”/”25R47” population. DArT markers were used to generate a genetic map, and quantitative trait loci (QTL) analysis was performed by evaluating 113 doubled haploid lines across three environments in Ontario, Canada. FHB resistance QTL were identified on chromosomes 4D, 4B, 2D and 7A, while a QTL for leaf and stem rust resistance was identified on chromosome 1B. The dwarfing alleles of both Rht‐B1 and Rht‐D1 were associated with increased FHB index and DON content.  相似文献

The height-reducing gene Rht8 was introduced into Italian wheats by breeder Nazareno Strampelli from the Japanese landrace Akakomugi, and has been widely used in wheats adapted to southern and eastern European conditions. Following identification of a close linkage to Rht8, microsatellite marker Gwm261 has been used extensively to screen large numbers of diverse international germplasm. A 192bp allele at this locus has been taken as “diagnostic” for Rht8 and used to infer the international distribution of Rht8. In this paper, we report several instances of cultivars and mapping populations that vary for the presence of the 192bp allele at the Xgwm261 locus (Xgwm261 192 ), but with no associated reduction in height, suggesting a lack of association with Rht8. For instance, in the population derived from a cross between Sunco (Rht-B1b, Xgwm261 165 ) and Tasman (Rht-D1b, Xgwm261 192 ), there were significant height differences associated with the segregation of Rht-B1b and Rht-D1b, but no height differences between Xgwm261 genotypes. Similar results were obtained in a population derived from the cross between Molineux (Rht-B1b, Xgwm261 192 ) and Trident (Rht-D1b Xgwm261 208 ). In contrast, the cross between Trident and Chuanmai 18 (Xgwm261 192 ) gave significant height effects at both the Rht-D1 and Xgwm261 loci, with no epistatic interaction between loci. Chuanmai 18 is closely related to the Strampelli wheat Mara (ancestrally derived from Akakomugi) and is therefore likely to carry Rht8. The old Japanese cultivar Norin 10, used by Norman Borlaug to introduce Rht-B1b and Rht-D1b into Mexican wheats, also has a 192bp allele at the Xgwm261 locus, and the sequence of the amplified product is identical to that of Akakomugi. We suggest that the widespread use of Norin 10-derived germplasm during and after the Green Revolution introduced a second haplotype into international germplasm, in which Xgwm261 192 has no association with Rht8. Therefore, the presence of Xgwm261 192 is only indicative of Rht8 in wheat cultivars that have inherited this allele from Akakomugi or a Strampelli wheat ancestor.  相似文献

Near isogenic lines (NILs) varying for genes for reduced height (Rht) and photoperiod insensitivity (Ppd-D1a) in a cv. Mercia background (rht (tall), Rht-B1b, Rht-D1b, Rht-B1c, Rht8c + Ppd-D1a, Rht-D1c, Rht12) were compared at one field site but within contrasting (‘organic’ vs. ‘conventional’) rotational and agronomic contexts, in each of 3 years. In the final year, further NILs (rht (tall), Rht-B1b, Rht-D1b, Rht-B1c, Rht-B1b Rht-D1b, Rht-D1b + Rht-B1c) in both Maris Huntsman and Maris Widgeon backgrounds were added together with 64 lines of a doubled haploid (DH) population [Savannah (Rht-D1b) × Renesansa (Rht-8c Ppd-D1a)]. Assessments included laboratory tests of germination and coleoptile length, and various field measurements of crop growth between emergence and pre jointing [plant population, tillering, leaf length, ground cover (GC), interception of photosynthetically active radiation (PAR), crop dry matter (DM) and nitrogen accumulation (N), far red: red reflectance ratio (FR:R), crop height, and weed dry matter]. All of the dwarfing alleles except Rht12 in the Mercia background and Rht8c in the DHs were associated with reduced coleoptile length. Most of the dwarfing alleles (depending on background) reduced seed viability. Severe dwarfing alleles (Rht-B1c, Rht-D1c and Rht12) were routinely associated with fewer plant numbers and reduced early crop growth (GC, PAR, DM, N, FR:R), and in 1 year, increased weed DM. In the Mercia background and the DHs the semi-dwarfing allele Rht-D1b was also sometimes associated with reductions in early crop growth; no such negative effects were associated with the marker for Rht8c. When significant interactions between cropping system and genotype did occur it was because differences between lines were more exaggerated in the organic system than in the conventional system. Ppd-D1a was associated positively with plant numbers surviving the winter and early crop growth (GC, FR:R, DM, N, PAR, height), and was the most significant locus in a QTL analysis. We conclude that, within these environmental and system contexts, genes moderating development are likely to be more important in influencing early resource capture than using Rht8c as an alternative semi-dwarfing gene to Rht-D1b.  相似文献

In the final year, further NILs (rht (tall), Rht-B1b, Rht-D1b, Rht-B1c, Rht-B1b+Rht-D1b, Rht-D1b+Rht-B1c) in Maris Huntsman and Maris Widgeon backgrounds were added together with 64 lines of a doubled haploid (DH) population [Savannah (Rht-D1b) × Renesansa (Rht-8c+Ppd-D1a)]. There were highly significant genotype × system interactions for grain yield, mostly because differences were greater in the conventional system than in the organic system. Quadratic fits of NIL grain yield against height were appropriate for both systems when all NILs and years were included. Extreme dwarfing was associated with reduced PAR, RUE, AGB, HI, and increased weed prevalence. Intermediate dwarfing was often associated with improved HI in the conventional system, but not in the organic system. Heights in excess of the optimum for yield were associated particularly with reduced HI and, in the conventional system, lodging. There was no statistical evidence that optimum height for grain yield varied with system although fits peaked at 85 and 96 cm in the conventional and organic systems, respectively. Amongst the DH lines, the marker for Ppd-D1a was associated with earlier flowering, and just in the conventional system also with reduced PAR, AGB and grain yield. The marker for Rht-D1b was associated with reduced height, and again just in the conventional system, with increased HI and grain yield. The marker for Rht8c reduced height, and in the conventional system only, increased HI. When using the System × DH line means as observations grain yield was associated with height and early vegetative growth in the organic system, but not in the conventional system. In the conventional system, PAR interception after anthesis correlated with yield. Savannah was the highest yielding line in the conventional system, producing significantly more grain than several lines that out yielded it in the organic system.  相似文献

The impact of the Rht dwarfing genes on P utilization efficiency (PUTE = grain dry matter per kg P in above-ground biomass), total P uptake (Pt) and related traits was studied in the varietal backgrounds of two tall wheat cultivars, Maringa and Nainari 60. Four sets of near-isogenic lines carrying different combinations of the alleles Rht-B1b, Rht-D1b and Rht-B1c for gibberellin-insensitive dwarfism in the hexaploid wheat (Triticum aestivum L.) were compared with tall controls in two field trials under conditions of adequate nutrient supply and irrigation in Northwest Mexico. The yield-increasing effect of the dwarfing genes Rht-D1b and Rht-B1b led to improved PUTE in Maringa and total P uptake in both cultivars. Also, the double dwarf line of Maringa had larger grain yields and P uptake compared to the tall control. The Rht-B1c genotypes showed low PUTE, thick roots and high P concentration in vegetative biomass indicating a surplus of assimilates and P, which could not be translocated into the grains. A similar problem could be observed in Nainari 60 with Rht-B1b and Rht-D1b, which produced the largest grain dry matter with the lowest P concentrations in grains although they showed high P accumulation in straw. Most of the net P uptake occurred before anthesis. P absorption after anthesis was more critical for the dwarf genotypes. For double dwarfs and Rht-B1c, respectively, only 3% and 21% of the total accumulated P at maturity was absorbed at post-anthesis. The grain P of the dwarf lines came more from P accumulated at pre-anthesis and translocated from the vegetative biomass into the grain. The pre-anthesis P accumulation was positively correlated with spikes per m2 (r = 0.91), whereas post-anthesis P accumulation correlated better with grains per spike(r = 0.72), and thousand kernel weight (r = 0.51). P uptake efficiency played a secondary role under these non-P-limiting conditions, and differences in root length density were only slightly affected by Rht-genes. Semi‐dwarf wheat is an important prerequisite for releasing a successful commercial cultivar in high‐yielding environments. In Northern Europe, this aim is achieved by using one of the dwarfing genes Rht‐B1 (formerly known as Rht‐1) or Rht‐D1 (Rht‐2). Both genes, however, result in a higher susceptibility to Fusarium head blight (FHB). We analysed the possibility to use the two non‐adapted FHB resistance quantitative trait loci Fhb1 and Fhb5 (syn. QFhs.ifa‐5A) to counterbalance the negative effect of the dwarfing allele Rht‐D1b in a winter wheat population of 585 doubled‐haploid (DH) lines segregating for the three loci. All lines were inoculated with Fusarium culmorum at four locations and analysed for FHB severity, plant height, and heading date. The DH population showed a significant (< 0.001) genotypic variation for FHB severity ranging from 3.6% to 65.9% with a very high entry‐mean heritability of 0.95. The dwarfing allele Rht‐D1b reduced plant height by 24 cm, but nearly doubled the FHB susceptibility (24.74% vs. 12.74%). The resistance alleles of Fhb1 and Fhb5 reduced FHB susceptibility by 6.5 and 11.3 percentage points, respectively. Taken all three loci together, Fhb5 alone was already able to reduce FHB susceptibility to the same extent as Rht‐D1b increased it. This opens new avenues for selecting semi‐dwarf wheat by marker‐assisted introgression of Fhb5 without the enhancement of FHB susceptibility.  相似文献

Bulgarian common wheat cultivars released in the period 1925–2003 were studied using the gibberellic acid (GA) test and microsatellite analysis of the Xgwm261 locus on chromosome 2DS to identify the semi-dwarfing (Rht) genes. The old cultivars, isolated through selection from landraces, carried rare alleles (211- and 215-bp) at Xgwm261 locus, and those developed by hybridisation to foreign cultivars, carried the 165- and 174-bp alleles. Forty-two (55.3%) of 76 modern cultivars were GA-responsive. The 192-bp allele, diagnostic for Rht8, was observed in 64 (84.2%) modern cultivars, of which 37 carry Rht8 alone, and 27 possess a combination of Rht8 and a GA-insensitive allele viz. Rht-B1d (17); Rht-D1b (6) and Rht-B1b (4). The 174-bp allele is present in seven cultivars, only one of which is photoperiod-sensitive, and the rest are day-length insensitive. The 203-bp allele was found in six modern cultivars. Cultivars carrying the Rht8 allele are the most widespread and some of them have been cultivated for a long period. Cultivars with the `Saitama 27' allele (Rht-B1d) are the most productive and are second in distribution in the country. The recently observed trend for increasing the proportion of cultivars with GA-insensitive Rht genes is probably due to their combination with the 192-bp allele of Xgwm261 locus tightly linked to the Ppd-D1, to the break of the link between the 174-bp allele and ppd-D1, and to the introduction of other genes influencing flowering time.  相似文献

Fusarium head blight (FHB) is one of the major fungal diseases in wheat throughout the world. To control FHB severity, breeding genetically resistant varieties is thought to be the most promising strategy. In wheat breeding programmes, short cultivars predominantly carrying the Norin 10 derived semi-dwarfing allele Rht-D1b ( Rht2 ) are preferred worldwide because of higher achievable grain yields and lower risk of lodging. This study was conducted to determine the influence of different alleles at the Rht-D1 locus on FHB reaction. Three winter wheat populations were produced by crossing rather susceptible varieties 'Biscay', 'Pirat' and 'Rubens' carrying mutant-type allele Rht-D1b with the more resistant varieties 'Apache', 'Romanus' and 'History' containing the Rht-D1a wild-type allele ( rht2 ). The 190, 216 and 103 progeny of the F4-derived populations were assayed for the presence of Rht-D1a or Rht-D1b , plant height, and mean FHB rating after spray inoculation at flowering time with a highly aggressive isolate of Fusarium culmorum . Comparably, high mean FHB severities ranging from 28% to 49% for all population × environment combinations were achieved, with significant genotypic variation for FHB rating and plant height within all populations. Both traits were negatively correlated with r ranging from −0.48 to −0.61 in the complete populations. However, within the subpopulations homozygous for one or other height allele these correlations decreased considerably. The Rht-D1b semi-dwarfing allele resulted in 7–18% shorter plants, depending on the population, but a considerably increased FHB reaction of 22–53%. Nevertheless, significant genotypic variance for FHB resistance remained in all tested Rht-D1b subpopulations indicating that selection for moderately FHB resistant genotypes within agronomically beneficial Rht-D1b genotypes is still feasible.  相似文献

In order to characterise quantitative trait loci (QTLs) for Type I and Type II resistance against Fusarium head blight (FHB) in wheat, a population of recombinant inbred lines derived from the cross Cansas (moderately resistant)/Ritmo (susceptible) was evaluated in spray-inoculated field trials over three seasons. Map-based QTL analysis across environments revealed seven QTLs on chromosomes 1BS, 1DS, 3B, 3DL, 5BL, 7BS and 7AL (QFhs.whs-1B, QFhs.whs-1D, QFhs.whs-3B, QFhs.whs-3D, QFhs.whs-5B, QFhs.whs-7A, QFhs.whs-7B) associated with FHB resistance. They accounted for 56% of the phenotypic variance. QFhs.whs-1D primarily appeared to be involved in resistance to fungal penetration, whereas the other QTLs mainly contributed to resistance to fungal spread. FHB resistance was significantly correlated with plant height (PH) and heading date (HD). Including all single environments, corresponding overlaps of QTLs for FHB resistance and QTLs for PH/HD occurred at six loci, among them two consistently detected QTLs, QFhs.whs-5B and QFhs.whs-7A. When significant effects of PH and HD on FHB resistance were eliminated by covariance analysis, a second QTL analysis revealed possible escape mechanisms for the majority of the coincidental loci.  相似文献

为了解沧麦6005叠氮化钠诱变群体中小麦重要功能基因的组成情况,利用高通量的KASP标记技术对小麦株高、抗病性、抗旱性、抗穗发芽、春化和品质等性状相关的基因进行了检测分析。结果表明:1)控制小麦株高的基因Rht-B1和Rht-D1在73份叠氮化钠诱变材料中出现6种组成类型,分别是Rht-B1a+197bp+Rht-D1a(1份)、Rht-B1a+197bp+Rht-D1b(39份)、Rht-B1a+Rht-D1a(1份)、Rht-B1a+Rht-D1b(13份)、Rht-B1b+Rht-D1a(13份)和Rht-B1b+Rht-D1b(3份),含有Rht-D1b的家系占全部突变家系的75.34%。2)在小麦抗病和抗逆性方面,73份诱变材料中发现含抗叶锈病基因Lr68的材料7份,含抗赤霉病基因Fhb1的材料5份,抗叶锈病基因Lr34在供试材料中未发现;在抗穗发芽方面,有3份材料含TaSdr-B1基因的TaSdr-B1a抗穗发芽单倍型,有53份材料含有PHS1基因的Rio Blanco type抗穗发芽单倍型,有16份材料含有TaMoc-A1基因的Hap-H抗穗发芽单倍型,在所有供试材料中含TaMFT-A1基因的Jagger-type和Zen/2174-type抗穗发芽单倍型的材料数分别为15份和22份;在抗旱性方面,有55份材料含有COMT-3B基因的3Ba单倍型,有28份材料含有Dreb-B1基因的TaDREB-B1a抗旱单倍型,有52份材料含有TaSST-4A基因的A2a抗旱单倍型;3)在小麦春化和早熟性方面均为一种单倍型,在春化方面,所有的诱变材料均为冬性类型。在早熟性方面,所有的供试材料在TaELF3-B1基因上均检测为晚开花的单倍型。4)在小麦品质方面,有45份材料含有高分子量麦谷蛋白亚基5+10,有18份材料含有Glu-A1基因的Ax1 or Ax2*强筋单倍型。综合表明,叠氮化钠诱变方法和KASP标记技术结合起来,可以作为一种小麦分子辅助育种的有效策略,能显著地提高小麦育种效率。  相似文献

Durum wheat is the most important tetraploid wheat mainly used for semolina and pasta production, but is notorious for its high susceptibility to Fusarium head blight (FHB). Our objectives were to identify and characterize quantitative trait loci (QTL) in winter durum and to evaluate the potential of genomic approaches for the improvement of FHB resistance. Here, we employed an international panel of 170 winter and 14 spring durum lines, phenotyped for Fusarium culmorum resistance at five environments. Heading date, plant height and mean FHB severity showed significant genotypic variation with high heritabilities and FHB resistance was negatively correlated with both heading date and plant height. The dwarfing gene Rht‐B1 significantly affected FHB resistance and the genome‐wide association scan identified eight additional QTL affecting FHB resistance, explaining between 1% and 14% of the genotypic variation. A genome‐wide prediction approach yielded only a slightly improved predictive ability compared to marker‐assisted selection based on the four strongest QTL. In conclusion, FHB resistance in durum wheat is a highly quantitative trait and in breeding programmes may best be tackled by classical high‐throughput recurrent phenotypic selection that can be assisted by genomic prediction if marker profiles are available.  相似文献

Fusarium head blight (FHB, scab), caused by Fusarium graminearum Schwabe, is a serious and damaging disease of wheat. Although some hexaploid wheat lines express a good level of resistance to FHB, the resistance available in hexaploid wheat has not yet been transferred to durum wheat. A germplasm collection of Triticum durum× alien hybrid lines was tested as a potential source of resistance to FHB under controlled conditions. Their FHB reaction was evaluated in three tests against conidial suspensions of three strains of F. graminearum at the flowering stage. Two T. durum×Thinopyrum distichum hybrid lines, ‘AFR4’ and ‘AFR5′, expressed a significantly higher level of resistance to the spread of FHB than other durum‐alien hybrid lines and a resistant common wheat line ‘Nyu‐Bay’. Genomic in situ hybridization using total genomic DNA from alien grass species demonstrated that ‘AFR5’ had 13 or 14 alien genome chromosomes plus 27 or 28 wheat chromosomes, while ‘AFR4’ had 22 alien genome and 28 wheat chromosomes. All of the alien chromosomes present in these two lines belonged to the J genome. ‘AFR5’ is likely to be more useful as a source of FHB resistance than ‘AFR4’ because of its relatively normal meiotic behaviour, high fertility and fewer number of alien chromosomes. ‘AFR5’ shows good potential as a source for transferring FHB resistance gene into wheat. The development of T. durum addition lines carrying resistance genes from ‘AFR5’ is underway.  相似文献

Reciprocal crosses were made between resistant hexaploid spring wheat cultivars/lines Sumai 3, Ning8331, and 93FHB21, and susceptible tetraploids Stewart 63 and DT486 to generate 35 chromosome pentaploids. Four heads from each of five F1 pentaploid plants from each cross were screened with Fusarium graminearum for fusarium head blight (FHB) reaction. No pentaploid was as resistant to FHB as the resistant parents. Pentaploids derived from several crosses were more resistant than the susceptible parents, a few were more susceptible, and all plants from crosses with 93FHB21 failed to survive. Most viable seeds were obtained from the cross Sumai 3 × DT486. From this cross four of the five F1 pentaploid parents were fertile and 354F2 seeds derived from these four pentaploids were sown and evaluated for their FHB reaction. The majority of F2 plants from pentaploids 1 and 3had the visual appearance and level of resistance of Sumai 3, whereas progeny from pentaploids 4 and 5 were more varied morphologically and generally more susceptible. Forty-three of the screened F2 plants were tested for the presence of specific D chromosomes by wheat microsatellite analysis. There was no relationship between presence/absence of D chromosomes and FHB reaction. Twenty-four lines had all D chromosomes present of which 10 were intermediate-susceptible and 14 were resistant to FHB. Three lines, one resistant and two intermediate, had no D chromosomes. The remainder had between 1 and 6 of the D chromosomes present and ranged from resistant to susceptible in FHB reaction. It appears that FHB resistance is not conferred by the D genome of Sumai 3. Summary A doubled haploid (DH) wheat population derived from the cross Wangshuibai/Alondra‘s’ was developed through chromosome doubling of haploids generated by anther culture of hybrids. Fusarium head blight (FHB) was evaluated for three years from 2001 to 2003 in Jianyang, Fujian Province, China, where epidemics of FHB have been consistently severe. After 307 pairs of simple sequence repeat (SSR) primers were screened, 110 pairs were polymorphic between Wangshuibai and Alondra`s’, and used to construct a genetic linkage map for detection of quantitative trait loci (QTLs). A stable QTL for low FHB severity was detected on chromosomes 3B over all three years, and QTLs on chromosomes 5B, 2D, and 7A were detected over two years. Additional QTLs on chromosomes 3A, 3D, 4B, 5A, 5D, 6B and 7B showed marginal significance in only one year. Six QTLs were detected when phenotypic data from three years were combined. In addition, significant additive-by-additive epistasis was detected for a QTL on 6A although its additive effect was not significant. Additive effects (A) and additive-by-additive epistasis (AA) explained a major portion of the phenotypic variation (76.5%) for FHB response. Xgwm533-3B and Xgwm335-5B were the closest markers to QTLs, and have potential to be used as selectable markers for marker-assisted selection (MAS) in wheat breeding programs.  相似文献

利用矮秆基因Rht-B1、Rht-D1和千粒重功能基因TaCwi-A1、TaGW2-6A、TaSus2-2B的KASP标记,对云南省育成的42份小麦品种(系)进行单倍型检测,旨在筛选出含有目标基因的优异小麦种质,为云南省小麦产量相关性状的遗传改良提供材料和方法。结果表明,供试材料的株高基因组成分为5种类型,分别为Rht-B1a/Rht-D1a(40.48%)、Rht-B1a/Rht-D1b(23.81%)、Rht-B1a+197bp/Rht-D1a(4.76%)、Rht-B1b/Rht-D1a(28.57%)、Rht-B1b/Rht-D1b(2.38%)。供试材料中TaCwi-A1基因TaCwi-A1a高粒重单倍型的分布频率为42.86%,TaGW2-6A基因Hap-6A-A高粒重单倍型的分布频率为38.10%,TaSus2-2B基因Hap-H高粒重单倍型的分布频率为71.43%。5份品种(系)为3个千粒重基因的TaCwi-A1a/Hap-6A-A/Hap-H高粒重单倍型组合,频率为11.90%。研究表明,云南小麦品种(系)产量相关性状具有较好的遗传改良潜力。  相似文献

SN224是从六倍体小黑麦与普通小麦杂种后代选育的矮秆小麦种质,为对其有效利用提供参考依据,本研究对其进行了细胞学和主要农艺性状的鉴定,对它矮秆性状的遗传特点进行了分析。结果表明,SN224平均株高68.6 cm,株型较紧凑,纺锤穗、有芒、白粒,千粒重42.0 g左右,中抗条锈病和白粉病,后期不早衰,综合农艺性状较好;SN224根尖细胞染色体数目为42条,花粉母细胞减数分裂MI可观察到21个二价体,为1BL?1RS易位系;SN224/辉县红杂种F1株高介于双亲之间,F2群体的株高分离表现连续变异。利用已知主效矮秆基因Rht-B1b、Rht-D1b和Rht8以及1RS的特异分子标记检测证明,SN224不含有3个矮秆主效基因,1RS对SN224矮秆性状的表达没有影响。利用SN224/辉县红F2群体,构建了含有134个标记的分子标记连锁遗传图谱,总长1332.1 cM。采用加性-完备区间作图法(ICIM-ADD)进行QTL分析,检测到2个降低株高的主效QTL QPh1B和QPh4B,分别位于1B染色体Xwmc719–Xgwm18和4B染色体Xgwm368–Xmag4284标记区间,它们可分别解释株高变异的20.0%和10.2%;检测到分别控制穗长、单株穗数和每穗小穗数的7个QTL;在4B染色体KSUM062–Xmag4284标记区间同时检测到降低株高、增加穗长和单株穗数的QTL。  相似文献

Fusarium head blight (FHB) is a destructive disease of wheat worldwide. FHB resistance genes from Sumai 3 and its derivatives such as Ning 7840 have been well characterized through molecular mapping. In this study, resistance genes in Wangshuibai, a Chinese landrace with high and stable FHB resistance, were analyzed through molecular mapping. A population of 104 F2-derived F7 recombinant inbred lines (RILs) was developed from the cross between resistant landrace Wangshuibai and susceptible variety Alondras. A total of 32 informative amplified fragment length polymorphism (AFLP) primer pairs (EcoRI/MseI) amplified 410 AFLP markers segregating among the RILs. Among them, 250 markers were mapped in 23 linkage groups covering a genetic distance of 2,430 cM. In addition, 90 simple sequence repeat (SSR) markers were integrated into the AFLP map. Fifteen markers associated with three quantitative trait loci (QTL) for FHB resistance (P < 0.01) were located on two chromosomes. One QTL was mapped on 1B and two others were mapped on 3B. One QTL on 3BS showed a major effect and explained up to 23.8% of the phenotypic variation for type II FHB resistance.  相似文献

In wheat, semidwarfism resulting from reduced height (Rht)‐B1b and Rht‐D1b was integral to the ‘green revolution’. The principal donors of these alleles are ‘Norin 10’, ‘Seu Seun 27’ and ‘Suwon 92’ that, according to historical records, inherited semidwarfism from the Japanese landrace ‘Daruma’. The objective of this study was to examine the origins of Rht‐B1b and Rht‐D1b by growing multiple seed bank sources of cultivars comprising the historical pedigrees of the principal donor lines and scoring Rht‐1 genotype and plant height. This revealed that ‘Norin 10’ and ‘Suwon 92’ sources contained Rht‐B1b and Rht‐D1b, but the ‘Seu Seun 27’ source did not contain a semidwarf allele. Neither Rht‐B1b nor Rht‐D1b could be definitively traced back to ‘Daruma’, and both ‘Daruma’ sources contained only Rht‐B1b. However, ‘Daruma’ remains the most likely donor of Rht‐B1b and Rht‐D1b. We suggest that the disparity between historical pedigrees and Rht‐1 genotypes occurs because the genetic make‐up of seed bank sources differs from that of the cultivars actually used in the pedigrees. Some evidence also suggests that an alternative Rht‐D1b donor may exist.  相似文献

Crown rot, caused by Fusarium pseudograminearum, is an important disease of wheat in Australia and elsewhere. In order to identify molecular markers associated with partial seedling resistance to this disease, bulked segregant analysis and quantitative trait loci (QTL) mapping approaches were undertaken using a population of 145 doubled haploid lines constructed from ‘2‐49’ (partially resistant) × ‘Janz’ (susceptible) parents. Phenotypic data indicated that the trait is quantitatively inherited. The largest QTLs were located on chromosomes 1D and 1A, and explained 21% and 9% of the phenotypic variance, respectively. Using the best markers associated with five QTLs identified by composite interval mapping, the combined effect of the QTLs explained 40.6% of the phenotypic variance. All resistance alleles were inherited from ‘2‐49’ with the exception of a QTL on 2B, which was inherited from ‘Janz’. A minor QTL on 4B was loosely linked (19.8 cM) to the Rht1 locus in repulsion. None of the QTLs identified in this study were located in the same region as resistance QTLs identified in other populations segregating for Fusarium head blight, caused by Fusarium graminearum.  相似文献
