The effects of soil disturbance and residue retention on the functionality of the symbiosis between medic (Medicago truncatula L.) and arbuscular mycorrhizal fungi (AMF) were assessed in a two-stage experiment simulating a crop rotation of wheat (Triticum aestivum L.) followed by medic. Plants were inoculated or not with the AMF, Glomus intraradices and Gigaspora margarita, separately or together. The contribution of the arbuscular mycorrhizal (AM) pathway for P uptake was determined using 32P-labeled soil in a small hyphal compartment accessible only to hyphae of AMF. In general AM colonization was not affected by soil disturbance or residue application and disturbance did not affect hyphal length densities (HLDs) in soil. At 4 weeks disturbance had a negative effect on growth and phosphorus (P) uptake of plants inoculated with G. margarita, but not G. intraradices. By 7 weeks disturbance reduced growth of plants inoculated with G. margarita or AMF mix and total P uptake in all inoculated plants. With the exception of plants inoculated with G. margarita in disturbed soil at 4 weeks, the AM pathway made a significant contribution to P uptake in all AM plants at both harvests. Inoculation with both AMF together eliminated the negative effects of disturbance on AM P uptake and growth, showing that a fungus insensitive to disturbance can compensate for loss of contribution of a sensitive one. Application of residue increased growth and total P uptake of plants but decreased 32P in plants inoculated with the AMF mix in disturbed soil, compared with plants receiving no residue. The AMF responded differently to disturbance and G. intraradices, which was insensitive to disturbance, compensated for lack of contribution by the sensitive G. margarita when they were inoculated together. Colonization of roots and HLDs in soil were not good predictors of the outcomes of AM symbioses on plant growth, P uptake or P delivery via the AM pathway.  相似文献

Plants are often grazed resulting in a sudden and significant removal of shoot tissue, which decreases photosynthesis and changes C allocation between within the plant. From results obtained in percolated sand it is possible to demonstrate an increase of rhizodeposition within few days after defoliation followed by a decrease of rhizodeposition. The aim of our study was to test if this pattern can be also observed for plants grown in soil. We grew Plantago arenaria in microcosms and defoliated half of them after 45 d. Half of the defoliated and non-defoliated microcosms were harvested 1.5 d, and the other half 8.5 d, after defoliation. We observed an increase of microbial biomass 1.5 d after defoliation followed by a decrease assessed 8.5 d after the treatment. In parallel, soil soluble C and the metabolic quotient of the microbial biomass first decreased and then increased at the second harvest reaching values equivalent to those of the non-defoliated treatment. Based on these results together with results obtained in artificial soil, we conclude that the defoliation of P. arenaria grown in soil leads to a transient peak of root exudation.  相似文献

It has been established that arbuscular mycorrhizal (AM) fungi are involved in the conservation of soil structure. However, the effect of ectomycorrhizal (EM) fungi alone or in interaction with AM fungi in soil structure has been much less studied. This experiment evaluated EM and AM fungi effects on soil aggregation and plant growth. Ash plants (Fraxinus uhdei) were grown in pots, and were inoculated with Glomus intraradices and Pisolithus tinctorius separately but also in combination. Our results showed that F. uhdei established a symbiotic association with EM and AM fungi, and that these organisms, when interacting, showed synergistic and additive effects on plant growth compared to singly inoculated treatments. EM and AM fungi prompted changes in root morphology and increased water-stable aggregates. AM fungi affect mainly small-sized macroaggregates, while EM and EM-AM fungi interaction mainly affected aggregates bigger than 0.5 mm diameter. These results suggest that ectomyccorrhizal as well as arbuscular mycorrhizal fungi should be considered in restoration programs with Fraxinus plants.  相似文献

Mycorrhiza helper bacteria (MHB) can promote the formation and functioning of arbuscular mycorrhizal (AM) symbiosis, but their role and application potential in coping with soil-borne diseases are still unclear. A 14-week greenhouse pot experiment was conducted to obtain several actinomycete strains helping AM symbiosis in suppressing the Phytophthora blight of pepper (Capsicum annuum L.), using a soil inoculated with Phytophthora capsici after sterilization. Five Streptomyces strains, including S. pseudogriseolus, S. albogriseolus, S. griseoaurantiacus, S. tricolor, and S. tendae, as well as the AM fungus (Funneliformis caledonium) were tested. The Phytophthora blight severity reached 66% at full productive stage in the uninoculated control, and inoculation of F. caledonium, S. griseoaurantiacus, and S. tricolor alone significantly decreased (P < 0.05) it to 47%, 40%, and 35%, respectively. Compared to F. caledonium alone, additional inoculation of S. tricolor or S. tendae, which were isolated from the rhizosphere of a healthy individual in an infected field, significantly elevated (P < 0.05) root mycorrhizal colonization, root biomass, fruit yield, and total K acquisitions of pepper and further significantly decreased (P < 0.05) blight severity. According to the feature of enhancing disease-suppression by AM symbiosis, both S. tricolor and S. tendae were confirmed as MHB strains here. Specifically, S. tendae had a stronger performance in directly accelerating mycorrhization, while S. tricolor antagonist to the pathogenic P. capsici. Furthermore, S. griseoaurantiacus with the independent disease-suppression function was not an MHB strain here. The redundancy analyses demonstrated that when AM fungus was present, root mycorrhizal colonization replaced soil pH becoming the main factor affecting pepper Phytophthora blight. Thus, S. tricolor and S. tendae seemed to have the value of preparation and application in the future to help AM symbiosis against pepper Phytophthora blight.  相似文献

The effect of the soil yeast, Rhodotorula mucilaginosa LBA, on Glomus mosseae (BEG n°12) and Gigaspora rosea (BEG n°9) was studied in vitro and in greenhouse trials. Hyphal length of G. mosseae and G. rosea spores increased significantly in the presence of R. mucilaginosa. Exudates from R. mucilaginosa stimulated hyphal growth of G. mosseae and G. rosea spores. Increase in hyphal length of G. mosseae coincided with an increase in R. mucilaginosa exudates. No stimulation of G. rosea hyphal growth was detected when 0.3 and 0.5 ml per petri dish of yeast exudates was applied. Percentage root length colonization by G. mosseae in soybean (Glycine max L. Merill) and by G. rosea in red clover (Trifolium pratense L. cv. Huia) was increased only when the soil yeast was inoculated before G. mosseae or G. rosea was introduced. Beneficial effects of R. mucilaginosa on arbuscular mycorrhizal (AM) colonization were found when the soil yeast was inoculated either as a thin agar slice or as a volume of 5 and 10 ml of an aqueous solution. R. mucilaginosa exudates (20 ml per pots) applied to soil increased significantly the percentage of AM colonization of soybean and red clover.  相似文献

This study was carried out in a semiarid degraded area to assess the effectiveness of mycorrhizal inoculation with a mixture of native arbuscular mycorrhizal (AM) fungi or an allochthonous AM fungus (Glomus claroideum), on the establishment of Olea europaea subsp. sylvestris L. and Retama sphaerocarpa (L.) Boissier in this area. Associated changes in the soil microbiological properties and aggregate stability related to these AM inocula were also recorded. Eighteen months after planting, G. claroideum had increased available P in the rhizosphere of both shrub species. In general, both inoculation treatments increased water-soluble C and water-soluble and total carbohydrates, G. claroideum being the most effective inoculum, particularly in R. sphaerocarpa. The mixture of native AM fungi was the most effective treatment for increasing the aggregate stability of R. sphaerocarpa soil, while that of O. europaea was increased only by G. claroideum. Increased (dehydrogenase, urease, protease-BAA, acid phosphatase and -glucosidase) enzyme activities, in particular of dehydrogenase and acid phosphatase, were recorded in the rhizosphere of both mycorrhizal shrub species. The mixture of native AM fungi was the most effective treatment for stimulating the growth of O. europaea and R. sphaerocarpa (11.6-fold and 3.3-fold, respectively, greater than control plants). The establishment of mycorrhizal shrub species favoured the reactivation of soil microbial activity, which was linked to an increase in aggregate stability.  相似文献

We investigated the feeding preferences of six species of mites and collembolans for three fungi commonly associated with roots of Acer saccharum (Glomus macrocarpum, Alternaria alternata and Trichoderma harzianum), from a maple-forest soil in southern Ontario, Canada. Experiments were also conducted in vitro to determine animal feeding responses to (1) increasing quantities of hyphal biomass, (2) the presence of root vs. litter fungal substrates, and (3) hyphae of different widths of Glomus macrocarpum. The results indicate that arthropods prefer to graze in the litter region rather than in the deeper soil layers. Under ideal moisture/temperature conditions, animals are forced to the lower regions by interspecific interactions. They prefer to graze on hyphae of conidial fungi rather than on those of arbuscular mycorrhizal fungi. When arbuscular mycorrhizal fungal hyphae are grazed, there is a clear preference for the narrower hyphae, which are those further away from the root. The thicker hyphal segments, commonly found connecting absorptive hyphal fans to roots, were less preferred. These data are not consistent with the hypothesis that microarthropods are detrimental to arbuscular mycorrhizal associations, and suggest that Glomalean fungi may have evolved mechanisms to deter grazing by microarthropods.  相似文献

Arbuscular mycorrhizal (AM) development in different soil types, and the influence of AM fungal hyphae on their original soil were investigated. Plantago lanceolata, which can grow in soils of a very wide pH range, was grown in two closely related limestone soils and an acid soil from rock habitats. Plants were colonised by the indigenous AM fungal community. The use of compartmented systems allowed us to compare soil with and without mycorrhizal hyphae. Root colonisation of P. lanceolata was markedly higher in the limestone soils (30-60%) than in the acid soil (5-20%), both in the original habitat and in the experimental study. Growth of extraradical AM fungal hyphae was detected in the limestone soils, but not in the acid soil, using the signature fatty acid 16:1ω5 as biomass indicator. Analysis of signature fatty acids demonstrated an increased microbial biomass in the presence of AM fungal hyphae as judged for example from an increased amount of NLFA 16:0 with 30 nmol g−1 in one of the limestone soils. Bacterial activity, but not soil phosphatase activity, was increased by around 25% in the presence of mycorrhizal hyphae in the first harvest of limestone soils. AM fungal hyphae can thus stimulate microorganisms. However, no effect of AM hyphae was observed on the soil pH or organic matter content in the limestone soils and the available P was not depleted.  相似文献

A laboratory experiment was carried out to test the hypothesis that the earthworms Lumbricus terrestris and Aporrectodea caliginosa are able to maintain their populations and reproduce in the acid forest soil of a deciduous forest where no lumbricids were found in the field. The experiment was conducted in 45-l containers in which layers of mineral subsoil, humus and organic topsoil collected from the site were established. Both species survived and at least L. terrestris reproduced during the 60 weeks’ incubation. Burrows and middens of L. terrestris were recorded and quantities of litter were consumed. The presence of lumbricids increased the organic matter content of humus, reduced the acidity of the topsoil and humus layers, and suppressed the population of the enchytraeid Cognettia sphagnetorum. A dense population of Enchytraeus albidus was found in L. terrestris middens. It is concluded that edaphic factors do not explain the absence of earthworms, but isolation from cultural landscapes and lack of opportunity to colonize the site from the surroundings is the decisive factor.  相似文献

The rose of an isolate of the arbuscular mycorrhizal (AM) fungusGlomus mosseae in the protection ofMedicago sativa (+Rhizobium meliloti) against salt stress induced by the addition of increasing levels of soluble salts was studied. The interactions between soluble P in soil (four levels), mycorrhizal inoculum and degree of salinity in relation to plant growth, nutrition and infective parameters were evaluated. Salt stress was induced by sequential irrigation with saline water having four concentrations of three salts (NaCl, CaCl2, and MgCl2).15N-labelled ammonium sulphate was added to provide a quantitative estimate of N2 fixation under moderate to high salinity levels. N and P concentration and nodule formation increased with the amount of plant-available P or mycorrhizal inoculum in the soil and generally declined as the salinity in the solution culture increased from a moderate to a high level. The mycorrhizal inoculation protected the plants from salt stress more efficiently than any amount of plant-available P in soil, particularly at the highest salinity level applied (43.5 dS m–1). Mycorrhizal inoculation matched the effect on dry matter and nutrition of the addition in the soil of 150 mg P kg–1. Nevertheless the highest saline solution assayed (43.5 dS m–1) affected more severely plants supplemented with phosphorus than those with the addition of mycorrhizal inoculum. Such a saline-depressing effect was 1.5 (biomass), 1.4 (N) and 1.5 (P) times higher in plants supplied with soluble phosphate than with AM inoculum. Mechanisms beyond those mediated by P must be involved in the AM-protectioe effect against salinity. The15N methodology used allowed the determination of N2 fixation as influenced by different P applications compared to mycorrhizal inoculation. A lack of correlation between nodule formation and function (N2 fixation) was evidenced in mycorrhizal-inoculated plants. In spite of the reduced activity per nodule in mycorrhizal-inoculated In spite of the reduced activity per nodule in mycorrhizal-inoculated plants, the N contents determined indicated the highest acquisition of N occurred in plants with the symbiotic status. Moreover, N and P uptake increased while Ca and Mg decreased in AM-inoculated plants. Thus P/Ca ratios and cation/anion balance in general were altered in mycorrhizal treatments. This study therefore confirms previous findings that AM-colonized plants have optional and alternative mechanisms available to satisfy their nutritive requirements and to maintain their physiological status in stress situations and in disturbed ecosystems.  相似文献

Dendrobaena octaedra (Lumbricidae) and Cognettia sphagnetorum (Enchytraeidae) are the two most dominating soil invertebrates in terms of biomass in boreal coniferous forest soils. A microcosm experiment was set up in order to study the influence of pH, moisture and resource addition on D. octaedra and C. sphagnetorum when both species are simultaneously present. Two kinds of coniferous forest humus were used as substrate, pine stand humus (pH 4.2), and spruce stand humus (pH 4.6); in the third treatment the pine stand humus was adjusted with slaked lime (CaOH2) to the same initial pH as the spruce stand humus. Each substrate was adjusted to water contents of 25%, 42.5% and 60% of WHC (referred to as ‘dry’, ‘moist’ and ‘wet’). In the second part of the experiment, spruce needle litter and birch leaf litter were separately added into the pine stand humus (‘moist’, unlimed) and compared with a control without litter. The microcosms were plastic jars with 75 g (d.m.) of humus, into which 4 specimens of D. octaedra and 70 specimens of C. sphagnetorum were added. D. octaedra showed the highest biomass and C. sphagnetorum the lowest biomass in the spruce stand humus with higher pH. Moisture did not affect earthworms, while C. sphagnetorum thrived best at the highest moisture. Addition of both kinds of litter increased the numbers and biomass of D. octaedra, while on C. sphagnetorum resource addition had little effect. The results help to explain the abundance of these two species in coniferous forests differing in soil acidity, moisture and fertility.  相似文献

A field experiment was conducted to study and compare the effectiveness of two arbuscular mycorrhizal fungi (AMF), Glomus macrocarpum (GM) and Glomus fasciculatum (GF) on three accessions of Artemisia annua. The AM inoculation significantly increased the production of herbage, dry weight of shoot, nutrient status (P, Zn and Fe) of shoot, concentration of essential oil and artemisinin in leaves as compared to non-inoculated plants. The extent of growth, nutrient concentration and production of secondary plant metabolites varied with the fungus–plant accession combination. The mycorrhizal dependency of the three accessions was related to the shoot: root ratio. Comparing the two fungal inoculants in regard to increase in essential oil concentration in shoot, the effectiveness of GF was more than that of GM. While in two accessions, GM was more effective in enhancing artemisinin concentration than GF. Increase in concentration of essential oil was found to be positively correlated to P-status of the plant. Conversely, no correlation was found between shoot-P and artemisinin concentration.

Elevated aluminum (Al) availability limits plant growth on acidic soils. Although this element is found naturally in soils, acidic conditions create an environment where Al solubility increases and toxic forms of Al impact plant function. Plant resistance to Al is often attributed to organic acid exudation from plant roots and the chelation of cationic Al in the rhizosphere. The association of arbuscular mycorrhizal (AM) fungi with the roots of plants may alleviate Al toxicity by altering soil Al availability or plant exposure through the binding of Al to fungal structures or through the influence of fungi on exudation from roots. Diverse communities of AM fungi are found in soil ecosystems and research suggests that AM fungi exhibit functional diversity that may influence plant performance under varying edaphic environments. In the present study, we evaluated acidic isolates of six AM species in their responses to Al. Andropogon virginicus (broomsedge), a warm-season grass that commonly grows in a range of stressful environments including acidic soils, was used as a plant host for Acaulospora morrowiae, Glomus claroideum, Glomus clarum, Glomus etunicatum, Paraglomus brasilianum, and Scutellospora heterogama. Fungal spores were germinated and exposed to 0 or 100 μM Al on filter paper in sand culture or were grown and exposed to Al in sand culture in association with A. virginicus. Short- and long-term responses to Al were evaluated using direct measurements of fungal spore germination, hyphal elongation, and measurements of A. virginicus colonization and plant growth as a phytometer of AM function in symbio. Spore germination and hyphal elongation varied among AM species in response to Al, but patterns were not consistent with the influences of these AM species on A. virginicus under Al exposure. Exposure to Al did not influence colonization of roots, although large differences existed in colonization among fungal species. Plants colonized by G. clarum and S. heterogama exhibited the least reduction in growth when exposed to Al, produced the highest concentrations of Al-chelating organic acids, and had the lowest concentrations of free Al in their root zones. This pattern provides evidence that variation among AM fungi in Al resistance conferred to their plant hosts is associated with the exudation of Al-binding organic acids from roots and highlights the role that AM fungal diversity may play in plant performance in acidic soil environments.  相似文献

Summary Concentrations of phenamiphos ranging from 0 to 40 rg/g soil were established in a typical Oxisol (Tropeptic Eutrustox), inoculated or uninoculated with Glomus aggregatum. The effect of the nematicide on the development of vesicular-arbuscular mycorrhizal (VAM) symbiosis was evaluated in the greenhouse using Leucaena leucocephala as an indicator host plant. Treatment of soil with phenamiphos did not have a significant influence on the development of mycorrhizal activity measured in terms of subleaflet phosphorus concentrations. Similarly, the nematicide did not have an adverse effect on the level of mycorrhizal colonization or on the P content of shoots, as determined at the time of harvest. However, shoot dry weight was adversely influenced by phenamiphos when the chemical was applied to the uninoculated soil at 20 g/g soil or higher, and when it was applied to the inoculated soil at 40 g/g soil. It is concluded that phenamiphos is not likely to influence the growth of Leucaena or its symbiotic association with VAM fungi if the concentrations applied do not exceed levels known to suppress nematodes.Hawaii Institute of Tropical Agriculture and Human Resources Journal series No. 3146  相似文献
