The present research was conducted to determine the relationship between the degradation of rimsulfuron and soil microbial biomass C in a laboratory-incubated clay loam soil (pH=8.1; organic matter=2.1%) under different conditions and at different initial dosages (field rate, 10 and 100 times the field rate). The half-life values varied between 0.4 and 103.4 days depending on temperature, soil moisture and initial dose. Evidence suggested that rimsulfuron could pose environmental risks in cold and dry climatic conditions. Significant decreases in microbial biomass C content in rimsulfuron-treated soil, compared to untreated soil, were observed initially, especially at higher temperatures and low moisture levels, but never exceeded 20.3% of that in control soil. The microbial biomass C content then returned to initial values at varying times depending on incubation conditions. The relationship between herbicide degradation and microbial biomass C content gave parabolic curves (P<0.005 in all cases) under all conditions tested. Generally, maximum biomass C decrease coincided with the decrease in the concentration of rimsulfuron to about 50% of the initial dose, except at 10  °C and 100×, when biomass began to recover as early as 65–70% of the initial dose. The final equations could be useful to deduce the decrease of soil microbial biomass in relation to herbicide concentration. From the degradation kinetics of the herbicide, the time required to reach this decrease can also be calculated. Received: 19 July 1999  相似文献

The effect of long-term waste water irrigation (up to 80 years) on soil organic matter, soil microbial biomass and its activities was studied in two agricultural soils (Vertisols and Leptosols) irrigated for 25, 65 and 80 years respectively at Irrigation District 03 in the Valley of Mezquital near Mexico City. In the Vertisols, where larger amounts of water have been applied than in the Leptosols, total organic C (TOC) contents increased 2.5-fold after 80 years of irrigation. In the Leptosols, however, the degradability of the organic matter tended to increase with irrigation time. It appears that soil organic matter accumulation was not due to pollutants nor did microbial biomass:TOC ratios and qCO2 values indicate a pollutant effect. Increases in soil microbial biomass C and activities were presumably due to the larger application of organic matter. However, changes in soil microbial communities occurred, as denitrification capacities increased greatly and adenylate energy charge (AEC) ratios were reduced after long-term irrigation. These changes were supposed to be due to the addition of surfactants, especially alkylbenzene sulfonates (effect on denitrification capacity) and the addition of sodium and salts (effect on AEC) through waste water irrigation. Heavy metals contained in the sewage do not appear to be affecting soil processes yet, due to their low availability. Detrimental effects on soil microbial communities can be expected, however, from further increases in pollutant concentrations due to prolonged application of untreated waste water or an increase in mobility due to higher mineralization rates. Effects of DDT and its metabolites on soil algae and enzymatic activity   总被引:1,自引:0,他引:1
The persistence of DDT [1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane] and its metabolites in soil, their toxicity to soil algae, and effects on microbial activities were studied in laboratory microcosms for 45 days. In non-sterile soils, removal of DDD [1,1-dichloro-2,2-bis(p-chlorophenyl)ethane] and DDE [1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene] was less than 3%, while 4–8% of applied DDMU [1-chloro-2,2-bis(p-chlorophenyl)ethylene], DDA [2,2-bis(p-chlorophenyl)acetic acid] and DDT were lost. Added DDOH [2,2-bis(p-chlorophenyl)ethanol] was more labile, as 60% was degraded during the same period. Soil microalgae were not measurably affected by the compounds tested at 10–50 mg kg–1, but at 100 mg kg–1 soil, DDD, DBP (p,p′-dichlorobenzophenone) and DDA significantly reduced their growth. Phosphatase activity was not affected by DDT and its metabolites at the concentrations tested (≤50 mg kg–1), but all compounds inhibited dehydrogenase activity at concentrations of 50 mg kg–1 soil. The toxic effects of DDT and its metabolites were dose-related.

Short-term effects of tillage systems on active soil microbial biomass   总被引:5,自引:0,他引:5
Conservation tillage, and especially no-tillage, induce changes in the distribution of organic pools in the soil profile. In long-term field experiments, marked stratification of the total soil microbial biomass and its activity have been observed as consequence of the application of no-tillage to previously tilled soils. Our objective was to study the evolution of the total and active soil microbial biomass and mineralized C in vitro during the first crop after the introduction of no-tillage to an agricultural soil. The experiment was performed on a Typic Hapludoll from the Argentinean Pampa. Remaining plant residues, total and active microbial biomass and mineralized C were determined at 0–5 cm and 5–15 cm depths, at three sampling times: wheat tilling, silking and maturity. The introduction of no-tillage produced an accumulation of plant residues in the soil surface layer (0–5 cm), showing stratification with depth at all sampling dates. Active microbial biomass and C mineralization were higher under no-tillage than under conventional tillage in the top 5 cm of the profile. The total soil microbial biomass did not differ between treatments. The active soil biomass was highly and positive correlated with plant residues (r 2=0.617;P<0.01) and with mineralized C (r 2=0.732;P<0.01). Consequently, the active microbial biomass and mineralized C reflected immediately the changes in residue management, whereas the total microbial biomass seemed not to be an early indicator of the introduction of a new form of soil management in our experiment.  The effects of 5 years of continuous grass/clover (Cont grass/clover) or grass (Cont grass) pasture or 5 years of annual grass under conventional (Ann grass CT) or zero tillage (Ann grass ZT) were compared with that of 5 years of continuous barley (LT arable) on a site which had previously been under arable crops for 11 years. For added comparison, a long-term grass/clover pasture site (LT past) nearby was also sampled. Soil organic C (Corg) content followed the order LT arable=Ann grass CT<Ann grass ZT<Cont grass=Cont grass/clover<LTpast. Trends with treatment for microbial biomass C (Cmic), basal respiration, flourescein diacetate (FDA) hydrolytic activity, arginine ammonification rate and the activities of dehydrogenase, protease, histidase, acid phosphatase and arylsulphatase enzymes were broadly similar to those for Corg. For Cmic, FDA hydrolysis, arginine ammonification and the activities of histidase, acid phosphatase and arylsulphatase, the percentage increase caused by 5 years of continuous pasture (in comparison with LT arable) was 100–180%, which was considerably greater than that for organic C (i.e. 60%). The microbial metabolic quotient (qCO2) was higher for the two treatments which were mouldboard ploughed annually (LT arable and Ann grass CT) than for the undisturbed sites. At the undisturbed sites, Corg declined markedly with depth (0–15 cm) and there was a similar stratification in the size and activity of Cmic and enzyme activity. The microbial quotient (Cmic/Corg) declined with depth whilst qCO2 tended to increase, reflecting a decrease in the proportion of readily available substrate with depth.  The interactive effects of vermi-compost from sewage sludge and either the sulfonylurea herbicide, rimsulfuron, or the imidazolinone herbicide, imazethapyr, on some soil biochemical and microbiological properties were investigated. The herbicides were applied at field and 10-fold field rates. Both herbicides exerted a detrimental effect on soil microbial biomass and its biochemical properties. Even though the effect of both herbicides on soil microbial biomass was not detectable at the field rate, some significant influences on acid and alkaline phosphatase were observed. The higher rate of herbicide application impaired the observed microbial parameters to a greater degree. The detrimental effects seemed to be reduced by organic amendments. Among the studied microbial characteristics, the specific respiration quotient was particularly reliable and sensitive in determining the influence of herbicides on the soil microbial biomass. In this paper a new synthetic index, specific hydrolytic activity (qFDA), for assessing microbial activity in reply to xenobiotic treatments is proposed. Received: 31 May 1999  相似文献

The effect of vegetation composition on various soil microbial properties in abandoned arable land was investigated 2 years after agricultural practice had terminated. Microbial numbers and processes were determined in five replicate plots of each of the following treatments: continued agricultural practice (monoculture of buckwheat in 1997), natural colonization by the pioneer community (arable weeds), and manipulated colonization from low (four species, three functional groups: grasses, forbs and legumes) or high diversity (15 species, three functional groups) seed mixtures from plant species that are characteristic of abandoned fields in later successional stages. The results indicated that differences in above-ground plant biomass, plant species composition and plant species diversity had no significant effect on soil microbial processes (net N mineralization, short-term nitrification, respiration and Arg ammonification), microbial biomass C and N (fumigation-incubation) or colony-forming units of the major microbial groups. Hence, there were no indications that soil microbial processes responded differently within 2 years of colonization of abandoned arable land by later successional plants as compared to that by plants from the natural pioneer weed community. Therefore, it seems that during the first few years after arable field abandonment, plants are more dependent on the prevailing soil microbiological conditions than vice versa.  Microbial populations, biomass, soil respiration and enzyme activities were determined in slightly acid organic soils of major mountainous humid subtropical terrestrial ecosystems, along a soil fertility gradient, in order to evaluate the influence of soil properties on microbial populations, activity and biomass and to understand the dynamics of the microbial biomass in degraded ecosystems and mature forest. Although the population of fungi was highest in the undisturbed forest (Sacred Grove), soil respiration was lowest in the 7-year-old regrowth and in natural grassland (approximately 373 μg g–1 h–1). Dehydrogenase and urease activities were high in "jhum" fallow, and among the forest stands they were highest in the 7-year-old regrowth. Microbial biomass C (MBC) depended mainly on the organic C status of the soil. The MBC values were generally higher in mature forest than in natural grassland, 1-year-old jhum fallow and the 4-year-old alder plantation. The MBC values obtained by the chloroform-fumigation-incubation technique (330–1656 μg g–1) did not vary significantly from those obtained by the chloroform-fumigation-extraction technique (408–1684 μg g–1), however, the values correlated positively (P<0.001). The enzyme activities, soil respiration, bacterial and fungal populations and microbial biomass was greatly influenced by several soil properties, particularly the levels of nutrients. The soil nutrient status, microbial populations, soil respiration and dehydrogenase activity were greater in Sacred Grove, while urease activity was greater in grassland.  Soil organic matter level, mineralizable C and N, microbial biomass C and dehydrogenase, urease and alkaline phosphatase activities were studied in soils from a field experiment under a pearl millet-wheat cropping sequence receiving inorganic fertilizers and a combination of inorganic fertilizers and organic amendments for the last 11 years. The amounts of soil organic matter and mineralizable C and N increased with the application of inorganic fertilizers. However, there were greater increases of these parameters when farmyard manure, wheat straw or Sesbania bispinosa green manure was applied along with inorganic fertilizers. Microbial biomass C increased from 147 mg kg–1 soil in unfertilized soil to 423 mg kg–1 soil in soil amended with wheat straw and inorganic fertilizers. The urease and alkaline phosphatase activities of soils increased significantly with a combination of inorganic fertilizers and organic amendments. The results indicate that soil organic matter level and soil microbial activities, vital for the nutrient turnover and long-term productivity of the soil, are enhanced by use of organic amendments along with inorganic fertilizers. The aim of this work was to calculate indices of hydrolase production (Pr) and persistence (Pe) through simple arithmetical calculations. Changes in acid and alkaline phosphomonoesterase, phosphodiesterase, urease, protease, and β-glucosidase activities were monitored under controlled conditions in seven soils with a wide range of properties, in which microbial growth was stimulated by adding glucose and nitrogen. Glucose mineralization was monitored by CO2–C evolution, and microbial growth was quantified by determining the soil adenosine triphosphate (ATP) content. Hydrolase Pr and Pe indices were numerically quantified by the following relationships: Pr = H / t H and Pe = (r / Ht, respectively, where H indicates the peak value of each measured hydrolase activity, t H is the time of the peak value, r indicates the residual activity value, and Δt is the time interval t r − t H, where t r is the time of the residual activity value. Addition of glucose and N-stimulated soil respiration increased ATP content and stimulated the production of the measured hydrolase activities in all soils; the measured variable reached a maximum value and then decreased, returning to the value of the control soil. Apart from β-glucosidase activity, whose activity was not stimulated by glucose and N addition, the other measured hydrolase activities showed a trend that allowed us to calculate the Pr and Pe indices using the above-mentioned equations. Acid phosphomonoesterase and protease Pr values were significantly higher in soils under forest or set aside management; the alkaline phosphomonoesterase and phosphodiesterase Pr values were generally higher in the neutral and alkaline soils, and the urease Pr values showed no obvious relationships with soil pH or management. Concerning the persistence of enzyme activities, Pe values of the acid phosphomonoesterase activity were significantly higher in the acidic soils, and those of urease activity were higher in acidic soils and the Bordeaux neutral soil. No relationships were observed between Pe values of alkaline phosphomonoesterase, phosphodiesterase, or protease activities and soil pH or management. The different responses of hydrolases were discussed in relation to soil properties, microbial growth, and regulation at the enzyme molecular level.  相似文献

Mineralization of N from organic materials added to soil depends on the quality of the substrate as a carbon, energy and nutrient source for the saprophytic microflora. Quality reflects a combination of biochemical and physical attributes. We investigated how biochemical composition interacts with particle size to affect the soil microflora and N dynamics following incorporation of crop residues into soil. Four fresh shoot and root crop residues were cut into coarse and fine particle sizes, and incorporated into sandy-loam soil which was incubated under controlled environment conditions for 6 months. In the case of the highest biochemical quality material, potato shoot (C/N ratio of 10 : 1), particle size had no effect on microbial respiration or net N mineralization. For lower biochemical quality Brussels sprout shoot (C/N ratio of 15 : 1), reducing particle size caused microbial respiration to peak earlier and increased net mineralization of N during the early stages of decomposition, but reduced net N mineralization at later stages. However, for the lowest biochemical quality residues, rye grass roots (C/N ratio of 38 : 1) and straw (C/N ratio of 91 : 1) reducing particle size caused microbial respiration to peak later and increased net immobilization of N. For Brussels sprout shoot, reducing particle size decreased the C content and the C/N ratio of residue-derived light fraction organic matter (LFOM) 2 months following incorporation. However C and N content of LFOM derived from the other materials was not affected by particle size. For materials of all qualities, particle size had little effect on biomass N. We conclude that the impact of particle size on soil microbial activities, and the protection of senescent microbial tissues from microbial attack, is dependant on the biochemical quality of the substrate. Genetically modified crops, that produce Cry insecticidal crystal proteins (Cry) from Bacillus thuringiensis (Bt), release these toxins into soils through root exudates and upon decomposition of residues. The fate of these toxins in soil has not yet been clearly elucidated. Persistence can be influenced by biotic (degradation by microorganisms) and abiotic factors (physicochemical interactions with soil components, especially adsorption). The aim of this study was to follow the fate of Cry1Aa Bt toxin in contrasting soils subjected to different treatments to enhance or inhibit microbial activity, in order to establish the importance of biotic and abiotic processes for the fate of Bt toxin. The toxin was efficiently extracted from each soil using an alkaline buffer containing a protein, bovine serum albumin, and a nonionic surfactant, Tween 20. The marked decline of extractable toxin after incubation of weeks to months was soil-dependent. The decrease of extractable toxin with incubation time was not related to microbial degradation but mainly to physicochemical interactions with the surfaces that may decrease immunochemical detectability or enhance protein fixation. Hydrophobic interactions may play an important role in determining the interaction of the toxin with surfaces.  相似文献

A chloroform-fumigation extraction method with fumigation at atmospheric pressure (CFAP, without vacuum) was developed for measuring microbial biomass C (CBIO) and N (NBIO) in water-saturated rice soils. The method was tested in a series of laboratory experiments and compared with the standard chloroform-fumigation extraction (CFE, with vacuum). For both methods, there was little interference from living rice roots or changing soil water content (0.44–0.55 kg kg–1 wet soil). A comparison of the two techniques showed a highly significant correlation for both CBIO and NBIO (P<0.001) suggesting that the simple and rapid CFAP is a reliable alternative to the CFE. It appeared, however, that a small and relatively constant fraction of well-protected microbial biomass may only be lysed during fumigation under vacuum. Determinations of microbial C and N were highly reproducible for both methods, but neither fumigation technique generated NBIO values which were positively correlated with CBIO. The range of observed microbial C:N ratios of 4–15 was unexpectedly wide for anaerobic soil conditions. Evidence that this was related to inconsistencies in the release, degradation, and extractability of NBIO rather than CBIO came from the observation that increasing the fumigation time from 4 h to 48 h significantly increased NBIO but not CBIO. The release pattern of CBIO indicated that the standard fumigation time of 24 h is applicable to water-saturated rice soils. To correct for the incomplete recovery of CBIO, we suggest applying the k C factor of 2.64, commonly used for aerobic soils (Vance et al. 1987), but caution is required when correcting NBIO data. Until differences in fumigation efficiencies among CFE and CFAP are confirmed for a wider range of rice soils, we suggest applying the same correction factor for both methods.  A greenhouse experiment was conducted to compare effects of different C and N sources applied to a flooded soil on soil microbial biomass (SMB) C and N, extractable soil organic N (NORG), and NH4 +-N in relation to plant N accumulation of rice (Oryza sativa L.). In addition to a control without inputs (CON), four treatments were imposed receiving: prilled urea (PU), rice straw (RS), RS and PU (RS+PU), or Sesbania rostrata as green manure (SES). Treatments were arranged according to a completely randomized design with four replicates and further consisted of pots with and without transplanted rice. While plant effects on the SMB were relatively small, the application of organic N sources resulted in a rapid increase in SMB until 10 days after transplanting (DAT) followed by a gradual decline until 73 DAT. Plant N accumulation data in these treatments clearly indicated that the SMB underwent a transition from a sink to a source of plant-available soil N during the period of crop growth. Seasonal variation of the SMB was small in treatments without amendment of organic material (CON, PU) presumably due to a lack of available C as energy source. Extractable NORG was significantly affected by soil planting status and organic N source amendment, but represented only a small N pool with little temporal variation despite an assumed rapid turnover. Among the three treatments receiving the same amount of N from different sources, the recovery efficiency of applied N was 58% for PU and 28% for both RS+PU and SES treatments at 73 DAT. The N uptake of rice, however, was not driven by N availability alone, as most evident in the RS+PU treatment. We assume that root physiological functions were impeded after application of organic N sources. Received: 1 June 1999  相似文献

Soil microarthropods were sampled in plots centered on creosotebushes (Larrea tridentata) and in plots centered on mesquite (Prosopis glandulosa) coppice dunes. Nine plots in each area were covered by rain-out shelters with greenhouse plastic roofs which excluded natural rainfall and nine plots received natural rainfall. There were differences in the abundance of several mite taxa in soils from the mesquite coppice dune plots. Some taxa (Stigmaeidae, Nanorchestidae, and Entomobryidae) occurred in significantly lower numbers in the soils of the drought plots. Other taxa (Tarsonemidae and Cunaxidae) were more abundant in the drought plots in the mesquite coppice dunes. There were no significant differences in the abundance of any of the dominant taxa of soil microarthropods in the drought and control plots centered on creosotebush. In the creosotebush habitat, there were significantly fewer Prostigmata in the plots exposed to drought. In an area with both creosotebush and mesquite, there were no significant differences in microarthropod population responses to drought and in recovery from drought. The differences in responses of soil microarthropods to drought in creosotebush and mesquite habitats are attributed to the differences in soil stability, litter accumulations, and microclimate associated with the shrubs.  The effects on soil condition of increasing periods under intensive cultivation for vegetable production on a Typic Haplohumult were compared with those of pastoral management using soil biological, physical and chemical indices of soil quality. The majority of the soils studied had reasonably high pH, exchangeable cation and extractable P levels reflecting the high fertilizer rates applied to dairy pasture and more particularly vegetable-producing soils. Soil organic C (Corg) content under long-term pasture (>60 years) was in the range of 55 g C kg–1 to 65 g C kg–1. With increasing periods under vegetable production soil organic matter declined until a new equilibrium level was attained at about 15–20 g C kg–1 after 60–80 years. The loss of soil organic matter resulted in a linear decline in microbial biomass C (Cmic) and basal respiratory rate. The microbial quotient (Cmic/Corg) decreased from 2.3% to 1.1% as soil organic matter content declined from 65 g C kg–1 to 15 g C kg–1 but the microbial metabolic quotient (basal respiration/Cmic ratio) remained unaffected. With decreasing soil organic matter content, the decline in arginine ammonification rate, fluorescein diacetate hydrolytic activity, earthworm numbers, soil aggregate stability and total clod porosity was curvilinear and little affected until soil organic C content fell below about 45 g C kg–1. Soils with an organic C content above 45 g C kg–1 had been under pasture for at least 30 years. At the same Corg content, soil biological activity and soil physical conditions were markedly improved when soils were under grass rather than vegetables. It was concluded that for soils under continuous vegetable production, practices that add organic residues to the soil should be promoted and that extending routine soil testing procedures to include key physical and biological properties will be an important future step in promoting sustainable management practices in the area. The content levels and activities of the microbiota were estimated in topsoils and in one soil profile at agricultural and forest sites of the Bornhöved Lake district in northern Germany. Discrepancies between data achieved by fumigation-extraction (FE) and substrate-induced respiration (SIR), both used for the quantification of microbial biomass, were attributed to the composition of the microbial populations in the soils. In the topsoils, the active, glucose-responsive (SIR) versus the total, chloroform-sensitive microbial (FE) biomass decreased in the order; field maize monoculture (field-MM)>field crop rotation (field-CR) and dry grassland>beech forest. This ratio decreased within the soil profile of the beech forest from the litter horizon down to the topsoil. Differences between microbial biomass and activities suggested varying biomass-specific transformation intensities in the soils. The metabolic quotient (qCO2), defined as the respiration rate per unit of biomass, indicates the efficiency in acquiring organic C and the intensity of C mineralization, while biomass-specific arginine-ammonification (arginine-ammonification rate related to microbial biomass content) seems to be dependent on N availability. The qCO2, calculated on the basis of the total microbial biomass, decreased for the topsoils in the same order as did the ratio between the active, glucose-responsive microbial biomass to the total, chloroform-sensitive microbial biomass, in contrast to qCO2 values based on the glucose-responsive microbial biomass, which did not. There was no difference between the levels of biomass-specific arginine-ammonification in topsoils of the fertilized field-CR, fertilized field-MM, fertilized dry grassland and eutric alder forest, but levels were lower in the beech forest, dystric alder forest, and unfertilized wet grassland topsoils. Ratios between values of different microbiological features are suggested to be more useful than microbiological features related to soil weight when evaluating microbial populations and microbially mediated processes in soils.  相似文献

We studied the relationship between urease activity (UA) and soil organic matter (SOM), microbial biomass N (Nbiom) content, and urea-N fertilizer assimilation by maize in a Dark Red Latosol (Typic Haplustox) cultivated for 9 years under no-tillage (NT), tillage with a disc plough (DP), and tillage with a moldboard plough (MP). Two soil depths were sampled (0–7.5 cm and 7.5–15 cm) at 4 different times during the crop cycle. Urea was applied at four different rates, ranging from 0 to 240 kg N ha–1. The levels of fertilizer N did not affect the UA, SOM content, and Nbiom content. No significant difference between the treatments (NT, DP, and MP) was observed for SOM during the experiment, probably because the major part of the SOM was in recalcitrant pools, since the area was previously cultivated (conventional tillage) for 20 years. The Nbiom content explained 97% and 69% of the variation in UA in the upper and deeper soil layer, respectively. UA and biomass N were significantly higher in the NT system compared to the DP and MP systems. The highest maize productivity and urea-N recovery was also observed for the NT system. We observed that the increase in urea-N losses under NT, possibly as a consequence of a higher UA, was compensated for by the increase in N immobilized in the biomass. Received: 2 July 1999  相似文献
