Summary Overcoming self-incompatibility by application of three kinds of plant hormones, sucrose, 3 kinds of amino acids and 2 kinds of vitamines was tested in cvs. Honbashi-taibyo Minowase (H-Mino) and Minowase (Mino) of Raphanus sativus. Effects differed between the cultivars. In H-Mino, BA (100 mg/l) and glutamic acid, folic acid and nicotinic acid (500 mg/l) resulted in higher fruit set and higher number of seeds per pollinated flower. In Mino, BA and NAA (100 mg/l) and glutamic acid and glycine (500 mg/l) induced a high number of seeds per pollinated flower. These chemicals, however, induced parthenocarpic fruit set, especially GA3. From the observation of pollen on stigmas washed with glutamic acid, it appeared that the pollen-tube penetrated into a papilla cell after 1 hour and openings of papillae and detached pollen grains and tubes were found after 2 hours as the result of successful pollentube penetration of papillae. Pollen was heated at 50°C for 30, 45 or 60 minutes, at 60°C for 15, 30 or 45 minutes and at 70°C for 10, 20 or 30 minutes prior to self-pollination. In H-Mino, 60 and 70°C were effective, and expecially 60°C for 15 or 30 minutes resulted a higher percentage fruit set and more seeds per fruit. In Mino, although 50–70°C were effective, the mean number of seeds per pollinated flower was lower than in H-Mino.  相似文献

Summary Two self-incompatible Upper Amazon cacao clones, T85/799 and T79/501, were pollinated with compatible Amelonado pollen subjected to varying doses of gamma irradiation (10–100 Gy). The proportion of flat non-viable beans to fully formed, viable beans in the pods increased with an increase in dosage of gamma rays. At 60 Gy all the beans produced were flat and non-viable, beyond this dosage fruit set was zero. Pollinating the self-incompatible cacao clones with a 1 : 1 mixture of compatible mentor pollen irradiated at 60 Gy and normal self pollen produced a mixture of flat, non-viable beans and fully-formed viable beans. Similar experiments using irradiated pollen with a marker gene suggested that the fully-formed viable beans resulted from selfing. Increasing the proportion of the radiation-treated compatible pollen in the mixture increased the number of fully-formed beans. However, when compatible pollen which had been treated either at 80 Gy or with temperatures of 35° C, 40° C and 45° C for periods of five, ten and fifteen minutes in factorial combination were mixed with self pollen, no successful pollinations were achieved. Pollen viability tests indicated that, whilst pollen treated at 60 Gy were about 50% viable, those treated at either 80 Gy or with temperatures of 35–45° C were mostly not viable. This suggests that, to overcome the incompatibility in cacao, the tubes of the mentor pollen grains used should at least grow into the style. The possible causes for overcoming the self-incompatibility in cacao are discussed.  相似文献

Incompatible (self) pollen tube growth in styles of six cultivars of L. longiflorum is strikingly influenced by treatment of the styles for six minutes in 50°C distilled water prior to pollination. In pretreated detached styles the incompatible pollen tubes reach lengths normally attained by compatible pollen whereas in untreated styles they grow to only approximately two-thirds the length normally reached by compatible tubes. Pretreatment at either 25°C or 45°C for six minutes does not affect incompatible pollen tube growth. Both compatible and incompatible tubes grow only a few millimeters into styles treated at 55°C for six minutes. Stigmatoid cells in these styles appear necrotic. Pretreatment of attached styles at 50°C for six minutes resulted in fruit set following self-pollination. Seed number per capsule ranged from 6 to 114. Stylar pretreatment appears to be a highly promising approach to further investigation concerning the nature of the self-incompatibility reaction. Summary Ipomoea cairica Sweet (Convolvulaceae) exhibits sporophytic self-incompatibility. This has partially been overcome under in vitro conditions, by treating the pollen and/or stigma with 10-5–10-1M indole-3-acetic acid (IAA), the optimum being 10-2M. The self-pollen. which otherwise does not even stick to stigma. germinates after self-pollination provided only one or both the partners are treated. The pollen tubes not only penetrate stigmatic papillae but also traverse the whole length of the style, at least in optimum experimental combinations.  相似文献

Summary The influence of temperature on self-incompatibility of six S-homozygous clones of Brussels sprouts was studied. The clones were treated with constant temperatures of 14°, 17° and 20°C and alternating day and night temperatures of 17/14°, 20/14°, 23/14° and 26/14°C. To determine the degree of incompatibility the mean number of pollen tubes per style after selfing was calculated.The clone with the weak S-allele S-5 was less self-incompatible in the 23/14° and 26/14°C treatments than in the other treatments. The other clones with the weak S-alleles S-15 and S-45 remained sufficiently self-incompatible during all treatments. The clone with the rather strong S-39 showed the highest level of self-incompatibility at 26/14°C.In the two weakly self-incompatible clones with S-5 and S-39 a clear difference in self-incompatibility was found between young and old flowers. The strongly SI clones with S-39 showed no difference and in the two clones with S-15 the differences were small and significant in one case only.  相似文献

Summary Controlled hand pollinated pistils of rubber were observed using fluorescence microscopy to assess the efficiency of the universally-employed method for the production of progeny for plant breeding. The controlled hand pollination method conducted in the morning resulted in the deposition of a mean of 15.6 pollen grains on the stigma, with no stray pollination observed. Over 36% of the pistils had the potential to set fruit.Pollinations conducted in the afternoon at the normal time of anthesis, had double the fruit set potential of morning pollinations as measured by penetration of ovules by pollen tubes. Pollinator efficiency also varied, with excessive damage to stigmas resulting in reduced pollen germination and tube growth. There were differences between clones in both female and male fertility, in the proportion of pistils with more than three carpels and in the production of small abnormal stigmas. There was no difference in pollen tube growth following self- or cross-pollination, indicating that the self-sterility mechanism of rubber operates in the ovary. Pollen could be stored for 5 days at 5°C and 75% RH with a 22% loss of fertility.  相似文献

自交不亲和性(self-incompatibility,SI)是指植株的雌蕊对自身花粉和异体花粉进行识别从而抑制自身花粉萌发的特性, PUB (Plant U-Box)蛋白在植物抗逆及信号转导过程中起着重要的作用。本研究通过分析甘蓝自花授粉0~60min的柱头转录组数据,筛选到1个受自花授粉诱导上调表达的PUB蛋白编码基因BoPUB9。BoPUB9 cDNA序列长度为1368 bp, gDNA序列全长1720 bp,含有1个臂重复结构域和1个U-box结构域。荧光定量PCR结果显示, BoPUB9基因在甘蓝的不同组织中均有表达,在萼片中的表达量最高,花瓣、雄蕊、柱头表达量次之,在花粉花蕾中表达量相对较低,同GUS染色结果保持一致。BoPUB9基因在自花授粉0~30 min内快速上调表达, 15 min后为异花授粉后表达量的20多倍,在30min后达到差异最大,自花授粉后的表达量为异花授粉的40多倍。亚细胞定位结果显示,BoPUB9蛋白在细胞核和细胞质中均有表达,且BoPUB9蛋白在大肠杆菌中被成功诱导表达,相对分子质量为51 kD,与预测结果一致。酵母双杂交与poll-down结果显示...  相似文献

Summary The impact of cross pollination 30 h, 4 h, and immediately prior to self-pollination, and self-pollination 2, 4, 8, 12, 24 and 30 h prior to cross-pollination was assessed for pollen tube number per style, seed number per siliqua, and proportion of self-seed (sibs) per siliqua, in two inbred lines of brussels sprout. Pollination procedure had a marked effect on the amount of sib-and hybrid seed produced. Cross pollination 30 h prior to self-pollination produced significantly greater numbers of sib progeny, as did self-pollination 8–12 h before cross pollination. Prior application of self pollen reduces the number of pollen tubes in the style. The results of this experiment suggest that self-incompatibility in brussels sprout could better be assessed by self-pollination with a subsequent cross pollination 8 h later, and the determination of amounts of sib and F1 hybrid seed per siliqua.  相似文献

Summary Immersing the excised-prepollinated styles of self-incompatible Japanese pear, Pyrus serotina Rehd., into distilled water at 45°C for 1.5–2.5 min promoted the growth of self-pollen tubes; both the number of styles penetrating the pollen tubes through styles to cut ends and of pollen tubes protruding from cut ends were increased. The subsequent growth of the protruded pollen tubes on agar media was also enhanced by the treatments. Treatments at 40°C did not promote the growth of self-pollen tubes and those at 50°C led the styles to necrosis. In the stylar proteins analyzed by isoelectricfocusing polyacrylamide gel electrophoresis, several changes in banding pattern were found following the heat treatment; one of the bands was a glycoprotein that increases in quantity in developing styles as self-incompatibility of the style becomes stronger (Hiratsuka et al., 1986). This protein band was stained more weakly by Coomassie Brilliant Blue-G250 dye. Summary Pollen germination and tube growth were studied following compatible, incompatible and pseudo-compatible pollinations in chicory. Pollen germination begins 3 minutes after compatible pollinations. The earliest pollen tubes reach the ovary 17 minutes later. Many of the later germinating pollen tubes are arrested and burst at the stigma papillae. In the transmitting tissue inhibitional effects due to negative interactions between pollen tubes are frequently observed. Complete self-incompatibility results in total inhibition of germination. In case of pseudo-self-compatibility, some pollen germinate but germination and stigma penetration are delayed and often result in pollen bursting. There is no self-incompatibility reaction in the transmitting tract but if the pollen tubes reaching this tissue are relatively numerous, negative interactions between them occur as after compatible pollinations. An hypothesis is presented which attributes the negative interactions between pollen tubes to the diffusion of a substance from the growing pollen tubes. This substance would also provoke pollen bursting on the stigma.  相似文献

自交不亲和性是甘蓝在长期进化过程中形成的防止自交衰败、促进杂交优势的一种复杂而完善的遗传机制。克隆自交不亲和性相关基因对甘蓝自交不亲和性的深入研究和利用有重要意义。本研究通过挖掘0~60 min自花和异花授粉的甘蓝柱头转录组数据, 筛选到一个受自花授粉诱导上调表达的基因, 命名为BoSPIBoSPI开放阅读框534 bp, 编码177个氨基酸, 理论等电点为4.21, 不包含信号肽和跨膜区, 含有4个保守的EF-hand结构域。BoSPI基因起始密码子上游2000 bp的核苷酸序列中含有真菌诱导响应、代谢调节以及器官形成等应答元件。BoSPI基因在大肠杆菌中可诱导表达为17 kD的蛋白。BoSPI在柱头中表达量最高, 在花瓣、萼片、叶片、雄蕊表达量较低。BoSPI蛋白被定位在细胞膜和细胞质。自花授粉30 min后对BoSPI基因的诱导表达显著增强。表明BoSPI参与了柱头响应自花花粉刺激的分子过程, 可能是实现甘蓝自交不亲和性相关的某种新功能基因。  相似文献

Summary Variation in the frequencies of pollination and ovule fertilization was generated among faba bean (Vicia faba L.) genotypes in experiments which compared self-and open-pollination, natural and extended photoperiods, presence and absence of honey bees and a range of plant populations.There was a near-1:1 relationship between the frequencies of pollination of stigmas and the fertilization of at least one ovule within the pistil. Pollination was the major barrier to fertilization of the pistil, and ovules, since no mechanism was evident in the style to prevent pistil fertilization. The frequency with which single ovules were fertilized in pollinated pistils fell as their distance from the stigma increased, and averaged 83.5% overall.The frequency of pollination was most influenced by the level of autofertility possessed by the genotype, and the activity of pollinators. Provision of honey bees to plants in caged plots showed that high levels of pollination could be achieved under open-pollinated conditions, but this was also influenced by autofertility. Increasing plant population in the field had only a small effect on pollination, but extending photoperiod caused the levels and timing of self-pollination in three genotypes to vary.Large differences were found among genotypes in their early stigma receptivity, that is, the frequency of stigmas found to be pollinated at the erect standard stage, prior to the flower opening and being visited by bees. The implications of early stigma receptivity for producing new cultivars were discussed.  相似文献

Summary Crossability between most Paspalum species is very low. This study was undertaken to identify the impediments to hybridization. Accessions of P. intermedium Munro. ex Morong, P. jurgensii Hackel and P. dilatatum Poir were self-pollinated and crossed with one anther. Paspalum intermedium is essentially self-sterile but P. jurgensii and P. dilatatum are highly self-fertile. Following pollination, pollen germination and tube growth were studied by observing the pollinated pistils with fluorescent microscopy. Examination of self-pollinated pistils revealed that the pollen germinated shortly after contacting the stigmas. Germination was over 80% for the P. intermedium and P. dilatatum accessions but only 57% for P. jurgensii. Pollen tubes grew to the micropyle within 45 minutes after pollination in P. dilatatum and 1 hour and 15 minutes in P. jurgensii. However, in the P. intermedium accessions most tubes did not grow beyond the stigma and very few penetrated the style and ovary. Apparently stylar-incompatibility is the reason for the low selfed seed set. In the cross-pollinations, pollen germinated shortly after pollination and germination ranged from 57 to 88% for the different crosses. In all crosses the pollen tubes grew to the micropyle within 30 minutes to 2 hours after pollination indicating that a cross-incompatibility system is not the cause for low crossability among these species. By examining embryo sacs from P. intermedium × P. dilatatum, its reciprocal and P. intermedium × P. urvillei crosses, it was determined that gametes failed to unite in some crosses and this is a major reason for low crossability.  相似文献

为探明花期高温胁迫对水稻花器官的影响机制,以耐热水稻品系996和热敏感水稻品系4628为材料,在人工气候室进行高温(8:00~17:00,37℃;17:00~次日8:00,30℃)和适温处理(8:00~17:00, 30℃;17:00~次日8:00,25℃), 研究高温胁迫对水稻花药抗氧化酶活性、膜透性、MDA含量及花粉性状等生理特性的影响。结果表明,高温胁迫下,水稻花药中SOD、POD、CAT、AsA-POD活性在高温胁迫初期均明显增加,尔后快速下降,耐热品系996这四种酶活性增幅大于热敏感品系4628;热敏感品系4628花药中MDA含量和膜透性在高温胁迫下增幅大于耐热性品系996;高温胁迫导致花药开裂、花粉萌发率和柱头上花粉粒数的显著下降,花粉粒直径增大。但耐热品系996的前3项参数显著高于热敏感品系4628。高温胁迫下水稻花药中保持较高抗氧化酶活性、较好的花粉散落特性和花粉萌发特性及较低的膜透性和MDA含量,是品种耐高温的生理基础。  相似文献

Summary Broccoli (Brassica oleracea L. var. italica) progenies grown from seed obtained by greenhouse budpollination (BP), open-flower self-pollination (OS), open-flower self-pollination with ether treatment of the stigma (OSE), and cross.pollination (CP) were compared in the field. For plant height, plant weight, head diameter, and head weight, BP resulted in the lowest values, OS and OSE were intermediate, and CP gave highest values. BP was highest, OS and OSE intermediate, and CP lowest for days to head emergence and days to harvest. Differences between OS and OSE were slight and all were non-significant for the 7 lines with complete data, but were significant for plant weight and head diameter when 20 lines were analyzed by the unweighted means method. The comparison BP versus OS-OSE was non-significant for plant height in both analyses and for head weight in the 7-line analysis only. In a second experiment using only BP and OS, and omitting days to head emergence, trends were similar but only the effects on head diameter were statistically significant. In the greenhouse, BP produced much more seed than either OS or OSE, and CP produced more than BP. Treatment of the stigma of open flowers with ether to overcome self-incompatibility did not result in greater seed production. Summary Experiments to determine whetherBrassica oleracea pollen could effect fertilisation following incubation in liquid culture medium are reported. Pollen was incubated in vitro, collected by centrifugation and used to pollinate compatible pistils. While retaining viability and the capacity to germinate such pollen was unable to penetrate the stigma papillae. However, incubated pollen produced functional tubes following pollination of styles from which the stigmas had been removed. These tubes grew through the style to the ovary and viable seed was obtained. The potential application of this procedure in pollen selection is discussed.  相似文献

Summary Application of heat during self-pollination of open flowers by means of an electric mini soldering iron resulted in seed set in self-incompatible Brassica oleracea varieties. Temperatures of 60, 70 and 80°C were tested. Compared with bud-pollination, in some cases the so-called thermally aided pollination (TAP) method, gave a considerably higher seed yield at 70 and 80°C. In view of the ease and rapidity of TAP it should be ascertained how far this method can replace bud-pollination in maintaining inbred lines. Summary Avocado pollen was stored at a range of temperatures and relative humidities (RH) for up to one year and the pollen was tested for viability in vivo.Pollen stored for one month was capable of germination on the stigma and penetrating the ovule when stored at 4°C with <1,23,55 and 75% r.h. and at -196°C with 0% r.h. Most pollen samples stored at 25 and -15°C at a range of r.h. would germinate on the stigma but none would penetrate the ovule.After one year of storage, pollen at 4°C and <1 and 23% r.h. would germinate on the stigma but would not penetrate the ovule. There was no germination of pollen stored at 4°C and 55 and 75% r.h. Only pollen stored at -196°C and 0% r.h. would penetrate the ovule, but thawing and refreezing once during the year destroyed the viability.  相似文献

The results of studies from 1982 to 1989 confirm that self-incompatibility (SI) of B. napus can be effectively overcome by spraying solutions of 2 — 10 % table salt (NaCl) during the flowering period. The optimum salt concentration was found to be 5 %. Spraying onto both stigma and stamen is better than onto only one of them, in particular better than spraying only onto the stamen. When self-pollination occurred at intervals of 1 to 32 hours after spraying 5 % NaCl solution upon stigma and stamen, the compatibility indexes fall slightly; they decrease considerably after 72 and 120 hours, but still then they stay higher than without spraying. After spraying 5 % NaCl solution at intervals of 4, 5, 8 and 24 hours, respectively, the compatibility indexes after self-pollination are almost equal to that of bud-pollination; this treatment is effective even at intervals of 48 or 72 hours after selfing. In practice, spraying 5 % NaCl solution every 3 to 5 days during the flowering period is enough for satisfying seed set of SI lines under field conditions.  相似文献

Summary Effects of temperature and photoperiod for donor plant growth on embryo formation in Capsicum annuum anther culture were investigated. Donor plants were grown in glasshouses at minimum temperatures between 16 and 30° C and at photoperiods between 11 and 19 h. Anthers were collected from individual plants over five to nine week periods to test the significance of donor plant age. Embryos were obtained at all temperature regimes with a calculated optimum temperature of 26.4° C. Embryo formation was unaffected by the photoperiods tested. Embryo formation varied among successive samplings.
