AIMS: To evaluate the toxicity, animal behavioural effects and animal welfare implications of 3-nitropropionic acid administration in possums. METHODS: The behavioural effects following 3-nitropropionic acid administration were recorded in 26 possums. RESULTS: The oral LD50 was between 110 and 170 mg/kg. Time to death averaged 9.5+/-1.9 h (mean+/-s.e.m.). Adverse signs from an animal welfare perspective were retching and restless behaviour that occurred in episodes during a period that averaged 1 h 51 min. Retching occurred in possums that were injected intraperitoneally or dosed orally with the 3-nitropropionic acid, suggesting that this effect was not necessarily initiated by gastric irritation. CONCLUSION: The potency of 3-nitropropionic acid as a toxin was low compared to other toxins that are used in possums, but it is relatively quick acting. It may cause some discomfort (retching and restlessness) prior to death.  相似文献

AIM: To assess the sickness behaviours of possums after eating a lethal dose of potassium cyanide. METHOD: Spontaneous behaviour and the time to loss of physical responses were examined. RESULTS: Cyanide ingestion caused a short-lasting period of mild respiratory stimulation. There was no salivation, retching or vomiting. Convulsions occurred in 73% of the possums. After the ingestion of cyanide, the average time to onset of ataxia was 3 minutes, the average time to overall loss of consciousness was 6.5 minutes, and the time to cessation of breathing was 18 minutes. CONCLUSION: Cyanide is a rapid-acting toxin with few undesirable signs from the welfare perspective.  相似文献

Possums are a wildlife vector of bovine tuberculosis in New Zealand. Vaccination of possums with BCG is being considered as a measure to control the spread of bovine tuberculosis to cattle and deer. Delivery via oral bait is feasible but BCG is degraded in the stomach. The aim was to determine whether ranitidine (Zantac) would reduce gastric acidity and enhance the efficacy of intragastrically administered BCG. A dose of 75 mg reduced gastric acidity for at least 4 h. Thus, possums were vaccinated intragastrically with BCG after receiving 75 mg ranitidine or ranitidine or BCG alone, as controls, before challenge with virulent Mycobacterium bovis. Proliferative responses of blood lymphocytes to M. bovis antigens after vaccination were significantly higher in possums given ranitidine/BCG compared to controls and seven weeks after challenge they had significantly lower lung weights and spleen bacterial counts than ranitidine alone controls. Vaccination with BCG alone only gave a reduction in loss in body weight. Agents that reduce gastric acidity may be useful in formulating BCG for oral bait delivery to wildlife for vaccination against bovine tuberculosis.  相似文献

AIMS: To describe the nature and incidence of metastatic mineralisation in brushtail possums (Trichosurus vulpecula) caught on Kawau Island. METHODS: Wild-caught possums were individually housed in captivity over a 24-month period and fed a cereal-based mash diet. Possums that became debilitated were examined post mortem for evidence of metastatic mineralisation of the heart, major blood vessels and kidneys. Serum and aortic segments were collected from affected animals and were analysed for mineral composition. Non-debilitated animals and possums that were experimentally poisoned with cholecalciferol were examined as a comparison. RESULTS: Fifteen of 42 possums (36%) captured on Kawau Island and housed in captivity were debilitated as a result of metastatic mineralisation of the walls of the aorta and/or of the kidney tubules, with four further possums having evidence of mineralisation when humanely killed. Phosphate and ammonium deposits were found in the aorta, and calcium deposits in the kidney tubules. Serum phosphorus and calcium levels were elevated in the debilitated animals. Similar lesions were found in only one of 288 possums captured from other North Island areas and housed in captivity over the same time period. CONCLUSION: Possums from Kawau Island appear to be more predisposed to metastatic mineralisation than possums from other areas of the North Island. The lesions observed in these possums were similar to those caused by cholecalciferol poisoning.  相似文献

AIMS: To determine the routes of transmission of wobbly possum disease (WPD) virus and whether or not these would favour further examination of its potential for biological control of possums. METHODS: A standard inoculum, prepared as a tissue suspension from possums which had been infected with WPD, was titrated in vivo. Possums were challenged with this inoculum by the intra-gastric, intra-tracheal and intra-dermal routes. Further possums were challenged with blood by the intra-dermal and intra-peritoneal routes, with urine by the intraperitoneal route and with homogenised mites (Trichosurolaelaps crassipes) by the intra-dermal route. Transmission was investigated between possums in closely-adjacent, individual cages and between possums in a group enclosure. RESULTS: Possums developed WPD following administration of the standard inoculum by all of the above routes, following administration of blood by the intra-peritoneal and intra-dermal routes, following administration of urine by the intraperitoneal route and following administration of homogenised mites by the intra-dermal route. Individually caged control possums did not contract WPD. All non-inoculated adult possums in the group enclosure and many joeys in direct contact with infected possums contracted WPD. CONCLUSION: WPD was efficiently transmitted by close contact. Without such contact transmission did not occur. Infectivity was demonstrated in tissue suspensions, blood, urine and mites. Given the routes by which possums are susceptible to these substances and the need for direct contact, infection may be spread in the wild by several mechanisms, including aggressive encounters in which blood is exchanged, contamination of wounds with urine, ingestion of contaminated food, transfer of mites during den-sharing, and other social encounters. WPD has potential as a biological control agent for possums on the basis that it is readily transmitted between individuals in close contact.  相似文献

AIMS: To determine immune responses, and the localisation and persistence of Mycobacterium bovis bacille Calmette-Guérin (BCG) in gut-associated lymphoid tissues (GALT) and other organs in possums vaccinated orally with lipid-formulated BCG vaccine. To determine the duration of excretion and longevity of survival of BCG in the faeces of vaccinated animals. METHODS: Possums (n=28) were vaccinated with lipid-formulated BCG (1 x 10(8) colony forming units (cfu) of formulated BCG) by the oral route. Control possums (n=17) were fed oral bait pellets containing formulation medium only. Possums were sacrificed at 3 days and at 1, 3, 6 and 8 weeks after vaccination or ingestion of bait. Proliferation responses to bovine purified protein derivative (PPD) were measured in lymphocytes from blood and mesenteric lymph nodes (MLN) and samples of lung, spleen, liver, MLN and Peyer's patches (PP) were cultured for the presence of BCG. The number of BCG organisms excreted in faeces and the duration of excretion were determined in eight vaccinated possums and eight control possums over a 3-week period. In a separate experiment, a further six possums were vaccinated with oral BCG vaccine (5-10 x 10(8) cfu BCG/possum) and their faeces collected over 48-72 h, for culture of BCG. The longevity of survival of BCG in these faeces was determined by storing faecal samples (n=12) under three different conditions: in an incubator (22.5 degrees C), and conditions which simulated the forest floor and open pasture. A proportion (1-2 g) of these faecal samples was collected after storage for 1, 3, 5, 8 or 20 weeks, and cultured for BCG. RESULTS: Possums vaccinated orally with BCG vaccine showed strong proliferation responses to bovine PPD in peripheral blood lymphocytes at 6-8 weeks post-vaccination (p.v.). Positive lymphocyte proliferation assay (LPA) responses to bovine PPD were first evident in MLN at 3 weeks p.v. BCG was cultured from MLN and PP in a proportion of animals at 3-8 weeks p.v. BCG was not cultured from sections of spleen, lung or liver at any time p.v. BCG was recovered in low to moderate numbers from the faeces of vaccinated possums for up to 7 days, and maximal numbers were cultured in faeces collected 48-72 h p.v. After storage for 1 week, BCG was cultured from all faecal samples placed in the incubator and from a proportion of faeces exposed to conditions similar to those on the forest floor and pasture. With the exception of one faecal sample stored under forest floor conditions which was culture-positive for BCG at 3 and 5 weeks, BCG was not cultured from any other faecal sample stored for more than 1 week. CONCLUSIONS: Ingestion of oral BCG vaccine by possums was associated with the development of strong cell-mediated immunity in both blood and MLN. Following oral vaccination with BCG, the organisms were localised and persisted in GALT but did not spread to the spleen, liver or lungs. BCG was shed in low to moderate numbers in the faeces for up to 7 days p.v. The viability of BCG excreted in faeces decreased rapidly, particularly when faeces were exposed to an open pasture environment. Oral vaccination of possums with formulated BCG is unlikely to result in undue contamination of the environment with BCG.  相似文献

As part of a search for pathogens that might be useful agents for biological control of possums, the three largest offshore islands of New Zealand that still have possums were surveyed to determine the pathogens present in these isolated populations. Brushtail possums from Kawau Island (n = 158), Chatham Island (n = 214) and Stewart Island (n = 194) were examined for internal parasites. Possums from Kawau Island were infected with Eimeria spp. (16.7%), Bertiella trichosuri (5.2%) and Purustrongyloides trichosuri (15.5%). No Paraustrostrongylus trichosuri or Trichostrongylus spp. were found. Possums from Chatham Island were infected with Eimeria spp. (10.9%). Bertiella trichosuri (3.6%), T colubriformis (6.6%), T retortaeformis (1%) and T. vitrinus (0.5%). No Parastrongyloides or Paraustrostrongylus spp. were found. Possums from Stewart Island were infected only with Eimeria spp. (4.6%). Because of their paucity of some parasites, the opportunity exists to use these offshore islands to study the introduction and spread of a parasite into a possum population, and what technology would be required to bring it to hyperendemicity.  相似文献

The aim of this study was to evaluate the prevalence of gastric Helicobacter-like organisms (GHLO) and gastritis in the gastric mucosa of dogs with gastric disorders. Tissue samples of the gastric mucosa were obtained from 30 dogs with gastrointestinal symptoms (vomiting, abdominal pain or discomfort, loss of appetite) during endoscopy. Histopathological examinations were performed and occurrence of GHLO infection, gastritis and other mucosal changes were estimated. The GHLO infection and gastritis were identified in 63.3 and 36.6% of dogs respectively; other mucosal changes included fibrosis in the lamina propria, degenerative changes of the gastric glands and hyperplasia of the parietal cells. The present study has revealed that microscopically found gastritis is not frequent in dogs examined by endoscopy. GHLO infection can be responsible for some cases of gastritis and hyperplasia of parietal cells in dogs.  相似文献

Radio-tracking and direct observation were used over 18 months in 1990-92 to investigate both the use of sleeping dens and foraging activity by possums (Trichosurus vulpecula) on a 21 ha site in the Wairarapa used for a longitudinal study of bovine tuberculosis. Males had larger home ranges than females, and both sexes had larger activity areas during the autumn mating season than at other times of the year. Possums typically foraged in only a small area of their home ranges (termed an activity area) on any one night, and the areas used by individuals were commonly very similar over a series of nights. Activity areas overlapped extensively among possums. Possums used a limited number of dens, commonly in a small and in most cases a circumscribed part of their home range. No simultaneous den-sharing was found, with the exception of mother-joey pairs. The mortality of juveniles after independence was 36%. Only two of 25 juveniles under surveillance to detect dispersal dispersed more than 500 m off the study site, and both subsequently died. Grazing patterns of cattle meant that almost all accessible areas of the paddock were covered by at least some grazing cattle, and so all activity areas of possums within the paddock were covered by areas where cattle foraged. However, possums avoided contact with cattle, and when some cattle were excluded from access to the part of the paddock principally used by both tuberculous and healthy possums for denning, transmission of Mycobacterium bovis from possums to these cattle ceased, although there was subsequent transmission to deer. Cattle which grazed the area used principally for possum denning continued to become infected, and these denning areas appeared to be of importance in the transmission of tuberculosis.  相似文献

Objective   To determine the effectiveness of a topical anaesthetic formulation (Tri-Solfen) with or without the administration of a non-steroidal anti-inflammatory drug (carprofen) on the pain and distress response associated with ring or surgical castration of ram lambs.
Procedures   Merino ram lambs (n = 78) were allocated to 10 treatment groups: 4 groups of knife-castrated lambs and 4 groups of ring-castrated lambs received carprofen (4 mg/kg SC) and Tri-Solfen; 2 control groups (sham) received carprofen at 0 or 4 mg/kg SC. Measurements included plasma cortisol and haptoglobin concentrations, haematology, and behaviour, including posture.
Results   Knife-castrated lambs had higher peak cortisol and integrated cortisol responses for the first 6 h after treatment and greater concentration s of circulating acute phase proteins than ring-castrated lambs, both of which were significantly different from the sham controls. Tri-Solfen applied to the knife castration wound significantly reduced both the peak plasma cortisol concentration and the integrated cortisol response for the first 6 h and improved lying behaviour in the first 12 h. Carprofen reduced the cortisol response to knife castration at 30 min, but elevated the cortisol responses at 24 and 48 h. Carprofen nearly halved the number of acute pain behaviours associated with ring castration. There were no significant additive or synergistic effects from combining the analgesic treatments. Tri-Solfen applied to the tail wound provided no detectible benefits during ring castration + tail docking.
Conclusions   The physiological and behavioural responses suggest that ring castration has less impact on the lamb than knife castration. The specific analgesic treatments can provide modest amelioration of the pain and discomfort associated with castration. Alternative doses or application methods may enhance their efficacy.  相似文献

Experimentally induced M. bovis infection in brush-tailed possums (Trichosurus vulpecula) with a possum-adapted strain from New Zealand, produced a more rapidly progressive, severe disease than did that induced by an Australian cattle strain. Possums infected with either strain excreted large numbers of mycobacteria in urine, faeces and discharges from abscesses. Aerosol transmission of infection occurred only with the New Zealand possum strain. The infectious aerosol most probably was of respiratory origin but urine may have been the source.All possums had focal interstitial nephritis consistent with Leptospira infection. Histopathological evidence of severe stress was observed in only one possum; the remainder had either mild or no stress-associated lesions.  相似文献

AIMS: To determine the clinical and pathological features of a neurological disease syndrome in a free-living possum population in New Zealand and to compare this syndrome with wobbly possum disease. METHODS: An outbreak of a neurological disease in possums in the Rotorua district was investigated in 1994. A variety of tissues was collected and investigated microbiologically and histopathologically. Tissues stored from clinically affected possums were homogenised, clarified and inoculated into healthy possums by the intra-peritoneal route. The clinical signs and histopathological lesions in naturally-infected and in experimentally-inoculated possums were assessed and compared with those of possums affected with wobbly possum disease. RESULTS: Histopathological investigation of three of the naturally-affected possums revealed non-suppurative encephalitis with perivascular cuffing, diffuse non-suppurative meningitis and focal non-suppurative myocarditis. These lesions were suggestive of a viral infection. No pathogenic bacteria were recovered and no viruses were isolated in tissue culture. A neurological disease, indistinguishable from wobbly possum disease, was reproduced in five out of the eight experimentally inoculated possums. In two experimental cases the clinical signs were very mild and, in most cases of the natural and experimental disease, histopathological lesions in the central nervous system were mild in comparison with wobbly possum disease. Possums which did not develop clinical signs of neurological disease or have lesions in the central nervous system did have infiltrations of mononuclear inflammatory cells in the liver and kidney. CONCLUSIONS: This neurological disease, reported for the first time in a free-living population, closely resembles and may be the same as wobbly possum disease. The milder nature of this disease could suggest there may be more than one strain of the aetiological agent.  相似文献

As part of a study to assess whether endoparasites could assist in the biological control of brushtail possums in New Zealand, we investigated the composition and distribution of possum endoparasites in the South Island. Possums were collected near five of the original release areas in the South Island : Banks Peninsula, Hokitika, Nelson, Dunedin and Invercargill. Among the nematodes, those most frequently encountered were Trichostrongylus spp., which were present in possums from all five study areas. Trichostrongylus species from possums in the Invercargill area comprised 4.5% T. colubriformis, 0.9% T. vitrinus and 11.3% T. retortaeformis. Paraustrostnmgylus trichosuri and Parastrongyloides trichosuri were found only in the Invercargill area, where they infected 1.4% and 14% of possums respectively. The cestode Bertiella trichosuri was present in possums from all locations except Dunedin. The protozoan Eimeria spp. occurred in all areas. These are the first records of Parastrongyloides trichosuri, Paraustrostrongylus trichosuri, T. vitrinus, T. retortaeformis and Eimeria spp. in South Island possums. The prevalence of endoparasites and the intensity of infection was very low compared to the lower North Island of New Zealand. Endoparasites at the existing levels in the South Island probably have very little effect on possum populations.  相似文献

Possums (Trichosurus vulpecula) captured for the first time (single-capture) had significantly lower blood lymphocyte and eosinophil counts, but higher neutrophil counts than possums which had been captured frequently. Single-capture possums also had depressed lymphocyte responses to concanavalin A (males and females) and pokeweed mitogen (males only). The haematological changes and the depressed lymphocyte reactivity to the T-cell mitogens were consistent with the animals being in a stressed state following capture for the first time. The haematological profile of single-capture possums changed to a profile similar to that of the frequently captured possums following housing in cages for 1 week, and high lymphocyte responses to T-cell mitogens were recorded following housing for 3-5 weeks. This information, together with body weight data, indicates that a minimum of 4 weeks adaptation to housing in cages should be allowed before possums are used in experimental studies.  相似文献

Surgical castration of young male piglets is now a generally accepted cause of serious distress and impairment of animal welfare. Awareness of this problem has created the moral commitment to seek for practical and more humane alternatives. As one possible alternative, the application of analgesics has been installed in Germany as an interim solution by the QS system, thus mandatory for the majority of German pig producers.Two analgesics have been authorised for this purpose. Both have been shown a significant positive impact on cortisol levels if administered pre-operatively. However, their effects on pain, stress and discomfort during castration, and on the post-castration period are conflicting. Thus, the aim of the present study was to compare the effects of Meloxicam and Flunixin on cortisol levels, behavioural indices, vocalisation, and wound healing of surgical castrated piglets in the field. There was no difference in vocalisation during castration in analgesic treated and untreated piglets. Piglets castrated under analgesia still had significantly elevated serum cortisol levels 30 min post castration, when compared to the sham castrated group. Both analgesics led to a significant impairment of behavioural indices and wound healing. It is concluded that analgesics can improve the welfare of piglets during the first part of the post-castration period. However, the benefits may be considered small and may not meet the requirements of the EU. Hence it is of high importance to prevent the interim practice of surgical castration of male piglets under analgesics from becoming implemented as a permanent condition in pig production.  相似文献

Primary gastric impaction developed in a pony as a result of the ingestion of persimmon seeds and mesquite beans. Clinical signs included mild abdominal pain, prolonged recumbency, anorexia, and lethargy. When medical therapy was unsuccessful, an exploratory laparotomy was performed. Previously, gastric impaction has been associated with signs of severe abdominal pain. Gastric impaction should be considered in cases of abdominal crisis of long duration and mild pain.  相似文献
