为了进一步明确大豆抗感品种对根腐尖镰孢的抗病机制,通过透射电镜和形态观察分析了尖镰孢(M38)侵染及产生的毒素对抗感大豆品种的影响。研究表明大豆幼苗胚根经粗毒素处理后,抗病品种‘东农56’幼苗胚根生长没有显著差别,但显著抑制了感病品种‘黑农53’胚根的伸长和侧根的生长;尖镰孢毒素高浓度和低浓度(V粗毒素∶V无菌水=1∶0和1∶15)对抗感品种的致萎能力无显著差异,高浓度处理平均萎蔫指数为100,低浓度处理为17.9~18.1左右;但中等浓度(V粗毒素∶V无菌水=1∶1,1∶5,1∶10)对于感病品种的致萎作用更大;同时,发现尖镰孢侵染感病大豆品种根内侵染量要明显多于抗病品种,且感病品种组织中菌丝的直径明显大于抗病品种,都出现很明显的质壁分离和细胞壁加厚的现象。  相似文献

Orobanche ramosa is a parasitic Angiosperm responsible for severe yield losses in several economical crops. It is a serious threat in oilseed rape in France and Morocco and is appearing in carrot crops in Morocco. In this study, several varieties of oilseed rape and carrot were screened in order to identify resistant cultivars and to characterize the resistance mechanisms involved. All the 15 oilseed rape varieties tested were susceptible. In carrot, the varieties 'Colmar à coeur rouge' and 'Nantaise demi-longue' were susceptible, whereas 'Palaiseau' and 'Buror' were resistant. In the susceptible 'Colmar à coeur rouge' carrot no defence reactions were found and the development of the parasite inhibited carrot tap root formation. In the resistant carrot varieties, the parasite germinated, became attached to the host root but became necrotic before emergence. In 'Buror' carrot, formation of a mechanical barrier was associated with the restriction to the cortex of the parasite. In maize cv. 'Vigni', a non-host of O. ramosa, thickening of xylem vessels, cell divisions in the central cylinder and formation of an encapsulation layer were observed in association with restricted development of Orobanche haustoria.  相似文献

Cavity spot is a major disease of carrots, causing cavities on the surface of the root. Available commercial varieties show a range of susceptibility but no significant resistance. Seed progeny from 46 tissue culture-derived carrot ( Daucus carota ) somaclones were screened for viability, then 19 selected somaclone families were sown under glasshouse conditions, along with commercial cultivars (Bertan, Nandor, Bolero and Vita Longa) as controls. Mature roots were exposed to Pythium violae in a cavity spot bioassay to determine their response as measured by disease incidence and severity. Some somaclones formed fewer lesions than the least susceptible control cultivar, Vita Longa. Seven somaclonal families that showed a range of susceptibility were sown under field conditions and the assessment was repeated. Although there was little relationship between glasshouse and field-trial results, under field conditions one of the somaclones had a mean incidence of disease, as estimated by transformed data, of 1·9 compared with 37·9 for the most susceptible somaclone and 3·5 for Bolero, the most resistant commercial cultivar. The results indicated that significant genetic variation in susceptibility to cavity spot disease was present in the somaclones.  相似文献

甘蔗褐锈病是目前中国蔗区发生最普遍、危害最严重的病害之一,选育和种植抗病品种是防治该病最为经济有效的措施。为明确近年国家甘蔗体系育成的50个新品种(系)对甘蔗褐锈病的抗性,确定其应用潜力,本研究通过在甘蔗褐锈病高发蔗区云南德宏、保山2个区域化试验站,采用田间自然抗性调查与分子标记辅助鉴定抗性基因的方法,于2014年和2015年对中国近年选育的50个优良新品种(系)及2个主栽品种进行抗褐锈病基因Bru1的分子检测及自然抗性评价。田间自然发病调查结果表明,50个优良新品种(系)及2个主栽品种中,高抗至中抗的有34个,占65.38%。其中15个材料表现高抗,占28.85%,16个材料表现抗病,占30.77%,3个材料表现中抗,占5.77%;分子检测结果显示,共29个抗病材料含有抗褐锈病基因Bru1,出现频率为55.77%,表明中国近年选育的优良新品种(系)中抗褐锈病性主要由Bru1控制;其余5个抗病材料均不含抗褐锈病基因Bru1,暗示除了Bru1外,可能还有其他抗褐锈病基因存在。不同系列品种田间感病品种的频率和含抗褐锈病基因Bru1的频率不同,粤糖系列品种感病品种的频率最高达到60%,含Bru1的频率最低,只有30%,抗性最弱;云蔗系列品种感锈病品种的频率最低只有12.5%,含Bru1的频率最高,达到81.25%,抗性最好。本研究结果为深入开展甘蔗抗褐锈病育种,选育和推广优良抗病品种,有效防控甘蔗褐锈病提供了科学依据和优良抗性新品种(系)。  相似文献

甘薯根腐病抗性在不同环境条件下的表现及遗传趋势   总被引:1,自引:0,他引:1  
结果表明,甘薯根腐病发病轻重与环境条件有很大关系,表现为干旱少雨的年份发病较重,降雨量较多的年份发病较轻;土壤瘠薄的发病较重,肥力条件较好的则发病较轻;通常情况下,年份间品种的抗性表现较为一致,但遇特殊气候则年份间品种的抗性有一定的差异。对1150份甘薯品种资源及育种材料的根腐病抗性鉴定结果表明,高抗型材料占14.6%,抗病型占15.7%,感病型占26.0%,高感型占43.7%。对754份材料及亲本的抗性分析表明,不同的抗性组合后代中均可分离出高抗至高感类型的材料,杂交后代的抗性强弱随双亲抗性水平的提高而提高。中国自1970年以来采用品种间杂交和种间杂交育种技术,先后育成了一批高产、优质的高抗型优良品种。  相似文献

Plants of the eucalypt. Eucalyptus marginata. selected through a glasshouse screening procedure for resistance or susceptibility to Phytophthora cinnamomi , were established in tissue culture and micropropagated. After inoculation with P. cinnamomi , root lesions in clonal lines selected as resistant (RR) to P. cinnamomi were restricted and became contained within four days after inoculation while lesions in roots of those lines susceptible (SS) to P. cinnamomi continued to extend rapidly. Activity of phenylalanine ammonialyase (PAL) was increased above controls in root segments of the RR lines 48 h after inoculation with P. cinnamomi while activity in unselected seedlings and the SS lines was reduced or unchanged. After inoculation, lignin concentration was increased and reached high levels compared with uninoculated control levels in roots of the two RR lines tested. Constitutive levels of phenolics in roots of the RR lines were up to 94% higher than in seedling roots and levels were further increased after inoculation. Levels of phenolics in the other lines and seedlings were unaltered by inoculation. A line derived from resistant seedlings from a susceptible family (RS) had the highest constitutive levels of lignin, which were further increased after inoculation. Resistance to P. cinnamomi in clonally propagated E. marginata seedlings is based on similar mechanisms to those of field resistant species.  相似文献

小麦旗叶和穗部吲哚生物碱含量与抗麦长管蚜关系研究   总被引:10,自引:1,他引:10  
采用蚜量比值法评价了 8个小麦品种 (系)对麦长管蚜的抗性 ,结果显示 ,高抗、抗、中抗、感虫品种 (系 )分别有 1、1、3、3个。进而研究了不同抗蚜品种 (系 )小麦旗叶和穗部的吲哚生物碱含量及其与抗蚜性的关系。结果表明 ,大多数抗蚜品种 (系 )的旗叶和穗部吲哚生物碱含量均高于感蚜品种 (系 ) ,尤其穗部。不同抗性级别的小麦品种 (系 )吲哚生物碱含量与蚜量比值的相关分析显示 ,穗部吲哚生物碱含量与蚜量比值的相关系数为-0.8633,而旗叶的相关系数为-0.3983,表明小麦旗叶吲哚生物碱含量与抗麦长管蚜具有一定的相关性 ,而穗部则呈显著的相关性  相似文献

 蚕豆锈病是由蚕豆单胞锈菌 [Uromyces viciae-fabae (Pers.) Schroet ] 引起的蚕豆最重要的病害之一。为提高蚕豆锈病抗性种质筛选的效率及准确性,本文采用混合菌株在温室条件下苗期接种、成株期评价的方法进行抗性鉴定。以抗病材料K0772和感病材料K90315为对照,对1416份材料(3个年度鉴定材料数量分别为290、517和609份)进行鉴定试验,其中重复鉴定材料77份。抗病对照K0772历年均表现为抗病(R);感病对照K90315分别表现为感病(S)、感病(S)和高感(HS)。不同年份间的病情指数T检验结果显示,对照品种K0772和K90315及重复鉴定材料的3年重复鉴定结果间差异并不显著,具有较好的稳定性。同时,重复鉴定材料组间分别呈极显著、显著、极显著正相关。研究结果表明年度间的重复鉴定结果高度一致,具有较好的稳定性和重复性。通过3年重复鉴定,获得高抗(HR)材料12份,抗病(R)材料47份,中抗(MR)材料18份。因此,利用混合菌源进行苗期接种和在成株期对蚕豆品种锈病抗性进行评价的方法具有较好的稳定性和准确性,该方法可以用于蚕豆锈病综合抗性材料的筛选和抗病育种研究。  相似文献

为初步探究鲜食玉米对草地贪夜蛾 Spodoptera frugiperda的抗性水平, 在田间草地贪夜蛾自然发生条件下对28个不同类型(甜玉米10个、甜糯玉米8个、糯玉米10个)的鲜食玉米品种进行了心叶期抗虫性鉴定, 研究了不同抗性水平及不同类型鲜食玉米的产量损失; 在室内研究了取食不同品种玉米苗对草地贪夜蛾幼虫发育历期的影响。经鉴定, 28个品种中无高抗及抗虫品种, 中抗品种12个, 感虫品种15个, 高感品种1个, 分别占鉴定资源总数的42.86%、53.57%和3.57%。28个品种的产量损失为8.30%~40.93%, 室内喂饲后幼虫的发育历期为11.29~15.31 d。不同类型鲜食玉米的叶部受害级别、产量损失和草地贪夜蛾幼虫发育历期无显著差异。甜玉米、糯玉米的叶部受害级别与产量损失呈正相关, 相关系数分别为0.61和0.79, 甜糯玉米的叶部受害级别与产量损失不相关。  相似文献

用浸根接种法测定了国内外622个水稻品种对水稻细菌性基腐病(Erwinia chrysanthemiBurkholder et al.)的抗性。结果表明,高抗品种有127个,中抗品种210个,中感品种189个,高感品种96个。粳稻发病重于籼稻,国际水稻(IR系统)品种较为抗病,而斯里兰卡水稻(BG系统)品种较为感病。用来自四个省的10个水稻菌株和2个玉米菌株(E.chrysanthemi pv.zeae)在20个水稻品种上人工接种测定,资料经统计分析,结果发现不同地区的水稻菌株并无致病性分化现象;但不同水稻品种间的抗性差异显著,玉米菌株对水稻的致病力相对较弱。  相似文献

对棉花不同抗感黄萎病性品种生化成分分析结果表明:棉花品种在无菌土中体内可溶性还原糖含量相应地高于接菌土中的含量。蕾期抗耐品种平均含量为2.276%,高于感性品种的1.417%,而铃期趋势相反。叶片单宁含量高于根部。在无菌土中,抗耐品种叶片单宁含量平均为3.58%,高于感病品种的3.28%;而在接菌条件下趋势相反。根部棉酚含量显著高于叶部,但两者极显著相关。在无菌土和接菌土中,抗性品种棉酚含量均高于耐性品种,而耐性品种又高于感性品种。棉株体内棉酚含量与棉花品种抗黄萎病性显著正相关。同工酶谱测定显示,过氧化物酶在抗感品种的无病症叶片中,酶带特征基本一致,叶片显症后表现主酶带区加宽,主酶带数增多,变化趋势与症状严重度正相关。在显症条件下,抗性品种酯酶同工酶活性略强于感性品种;与无症叶相比,显症叶的SOD酶带数由原来的4—6条减少至3—4条,抗性品种的酶带数少于感性品种1—2条。  相似文献

为明确云南籼稻主栽品种对白背飞虱Sogatella furcifera(Horváth)的抗虫性及其影响产量的水稻关键生育期,为抗虫品种选育和推广提供参考依据,本文利用苗期和分蘖盛期单株接虫鉴定、成株期(移栽期—乳熟期)田间自然种群发生鉴定等方法,测定了云南籼稻区31个水稻主栽品种对白背飞虱的抗虫性及其产量。结果表明,苗期有7个品种表现高抗,23个品种表现中抗—中感,1个品种表现高感;分蘖期有20个品种表现高抗,有11个表现中抗—中感;成株期田间自然种群鉴定,31个品种成株期白背飞虱田间自然种群的累计虫量都超过了对照品种TN1,均表现为感虫。单株接虫鉴定多数品种表现抗虫,而大田期自然种群鉴定则全部表现为感虫,一定程度说明了云南籼稻区主栽品种对白背飞虱具有耐害性而无抗生性。移栽期、孕穗期、抽穗期百丛虫量与有效穗数呈显著的负相关性;移栽期、孕穗期和抽穗期的百丛虫量是造成有效穗数少和产量低的关键因子。移栽期、孕穗期和抽穗期是防治白背飞虱的关键时期。  相似文献

 采用不同的方法接种和利用扫描、透射电镜观察,研究不同致病力的青枯菌对番茄抗病及感病品种根部的吸附、侵入与繁殖。发现番茄抗病品种与感病品种的植株体内在菌体数量上有明显差异,而与青枯菌对番茄根部的吸附关系不显著。电镜观察发现青枯菌强致病力菌株菌体能以游离的形式存在于番茄感病品种根部的细胞间隙中,并能降解植株细胞壁、破坏原生质膜;青枯菌强致病力菌株菌体在抗病品种根内和青枯菌强致病力菌株在抗病及感病品种根内均被番茄植株细胞壁吸附,并且被细胞壁周围的浓密物质所包围。  相似文献

 室内鉴定了411个水稻品种对恶苗病的抗性,没有免疫品种,高抗品种1个(Te70),抗病品种12(TR9等),其余为感病品种。籼稻比粳稻感病,杂交稻比常规稻感病。初选10个鉴别品种对湖南收集分离的50个菌株的致病性分化进行测定,表明恶苗病菌致病性存在分化,50个菌株分属4个群11个小种。小种I1为湖南的优势小种。抗病品种的锌、酚类物质含量比感病品种高,而可溶性糖/淀粉比值则感病品种比抗病品种高。  相似文献

Genes encoding an acidic wheat class IV chitinase (383), an acidic wheat β 1,3-glucanase (638) and a rice cationic peroxidase (POC1) were introduced into ‘Nantes Coreless’ carrot (Daucus carota) by Agrobacterium-mediated transformation. The genes were introduced singly or in various combinations followed by selection imposed by the herbicide phosphinothricin. Regenerated plantlets were screened for presence and expression of the three transgenes using PCR, Southern and Northern hybridisations. Eighteen transgenic lines expressing a single transgene and 2 lines each co-expressing 638/383 and 383/POC1 were assessed for resistance to the necrotrophic fungal pathogens Botrytis cinerea and Sclerotinia sclerotiorum. Percentage leaf area diseased was measured 4 and 7 days after inoculation (dai) and compared to non-transformed control plants. Six lines expressing β-1,3-glucanase 638 alone had no enhanced resistance to B. cinerea at 4 dai and only slight resistance to S. sclerotiorum; there was no effect at 7 dai. Two out of the six lines expressing 383 alone had enhanced tolerance to both pathogens with a 20–50% reduction in disease development at 7 dai. Two lines co-expressing 638/383 had slight reductions in disease by (10–20%) similar to that of the lines expressing chitinase 383 alone. Highest levels of disease resistance were seen in transgenic lines expressing POC1, alone or in combination with chitinase 383. Disease symptoms were slower to develop and symptoms were reduced by up to 90% for B. cinerea and 70% for S. sclerotiorum. The 383/POC1 co-expressing plants developed disease at levels similar to that of POC1 alone. Petioles of plants over-expressing POC1 had higher levels of lignin accumulation constitutively compared to control plants, which was greatly enhanced following inoculation with S. sclerotiorum. These results indicate that peroxidase over-expression can lead to significant disease reduction against necrotrophic pathogens in transgenic carrot plants.  相似文献
