Sabitri is a rice variety grown in a large part of the rainfed areas of Nepal. It was originally developed for irrigated condition; hence, this variety suffers high yield decline under drought. Two QTLs, qDTY 3.2 and qDTY 12.1 , with large effects on grain yield under drought were identified in the Sabitri background in separate QTL mapping studies. The present study reports the development of Sabitri near isogenic lines (NILs) with combinations of these two QTLs and their characterization under drought. To do so, marker-assisted backcross breeding (MABB) was combined with phenotypic selection to develop high-yielding drought-tolerant NILs with Sabitri grain type. Apart from this, drought-tolerant variants for grain type with high yield under non-stress were identified among the developed NILs. Early days to flowering of up to 13 days and reduction in plant height of up to 13 cm as compared to Sabitri were observed in the developed NILs. Some of these NILs showed higher yield compared to Sabitri and relatively higher tolerance to drought, indicating the capture of positive alleles and interactions during the course of selection. The developed NILs possessed high yield potential which make them suitable materials for the testing of water-saving technologies in irrigated areas. Based on their performance, these NILs can be deployed in rainfed areas in Nepal and other countries of South Asia to increase yield stability.  相似文献

Genome-wide association studies (GWAS) are useful to facilitate crop improvement via enhanced knowledge of marker-trait associations (MTA). A GWAS for grain yield (GY), yield components, and agronomic traits was conducted using a diverse panel of 239 soft red winter wheat (Triticum aestivum) genotypes evaluated across two growing seasons and eight site-years. Analysis of variance showed significant environment, genotype, and genotype-by-environment effects for GY and yield components. Narrow sense heritability of GY (h 2  = 0.48) was moderate compared to other traits including plant height (h 2  = 0.81) and kernel weight (h 2  = 0.77). There were 112 significant MTA (p < 0.0005) detected for eight measured traits using compressed mixed linear models and 5715 single nucleotide polymorphism markers. MTA for GY and agronomic traits coincided with previously reported QTL for winter and spring wheat. Highly significant MTA for GY showed an overall negative allelic effect for the minor allele, indicating selection against these alleles by breeders. Markers associated with multiple traits observed on chromosomes 1A, 2D, 3B, and 4B with positive minor effects serve as potential targets for marker assisted breeding to select for improvement of GY and related traits. Following marker validation, these multi-trait loci have the potential to be utilized for MAS to improve GY and adaptation of soft red winter wheat.  相似文献

Enhanced root growth in plants is fundamental to improve soil water exploration and drought tolerance. Understanding of the variance components and heritability of root biomass allocation is key to design suitable breeding strategies and to enhance the response to selection. This study aimed to determine variance components and heritability of biomass allocation and related traits in 99 genotypes of wheat (Triticum aestivum L.) and one triticale (X. Triticosecale Wittmack) under drought-stressed and non-stressed conditions in the field and greenhouse using a 10?×?10 alpha lattice design. Days to heading (DTH), days to maturity (DTM), number of tillers (NPT), plant height (PH), spike length (SL), shoot and root biomass (SB, RB), root to shoot ratio (RS), thousand kernel weight (TKW) and yield (GY) were recorded. Analyses of variance, variance components, heritability and genetic correlations were computed. Significant (p?<?0.05) genetic and environmental variation were observed for all the traits except for spike length. Drought stress decreased heritability of RS from 47 to 28% and GY from 55 to 17%. The correlations between RS with PH, NPT, SL, SB and GY were weaker under drought-stress (r?≤???0.50; p?<?0.05) compared to non-stressed conditions, suggesting that lower root biomass allocation under drought stress compromises wheat productivity. The negative association between GY and RS (r?=???0.41 and ??0.33; p?<?0.05), low heritability (<?42%) and high environmental variance (>?70%) for RS observed in this population constitute several bottlenecks for improving yield and root mass simultaneously. However, indirect selection for DTH, PH, RB, and TKW, could help optimize RS and simultaneously improve drought tolerance and yield under drought-stressed conditions.  相似文献

Sugar beet hybrid varieties are produced through the crosses between male sterile lines and the multigerm pollinators. The uniformity of pollinators used for hybrid crosses depends on the presence of self-sterility (S s ) and self-fertility (S f ) genes. The aim of the study was to analyze correlation between hybrid performance and genetic distance or heterozygosity of the sugar beet pollinators. Twelve diploid pollinators classified as self-sterile (S s ) or self-fertile (S f ) and two cytoplasmic male sterile (CMS) lines were crossed in line × tester scheme, producing 24 F1 hybrids. The parents and the hybrids were evaluated for root yield and quality traits, from which F1 performance, combining abilities, mid-parent and high-parent heterosis were calculated. Parental genetic distance and diversity of the pollinators were estimated by SSR markers and, together with GCA and F1 performance, correlated with the heterosis effects. The S f hybrids had better GCA and higher values of root yield, root weight, and root circumference than the S s hybrids. Heterosis was recorded in more combinations with the S f than with the S s pollinators. Parameters of genetic diversity were higher in the S s (Na = 3.125; Ne = 2.341; He = 0.555) than in the S f pollinators (Na = 3.000; Ne = 2.188; He = 0.510). Genetic distance between the tested pollinators and the CMS lines was low (0.072–0.224) indicating that the genetic base of the investigated germplasm was narrow. Correlation of the heterosis effects with GD and heterozygosity was detected only for the root yield traits.  相似文献

Tiller number per plant (TN) and plant height (PH) are important agronomic traits related to grain yield (GY) in rice (Oryza sativa L.). A total of 30 additive quantitative trait loci (A-QTL) and 9 significant additive × environment interaction QTLs (AE-QTL) were detected, while the phenotypic and QTL correlations confirmed the intrinsic relationship of the three traits. These QTLs were integrated with 986 QTLs from previous studies by metaanalysis. Consensus maps contained 7156 markers for a total map length of 1112.71 cM, onto which 863 QTLs were projected; 78 meta-QTLs (MQTLs) covering 11 of the 30 QTLs were detected from the cross between Dongnong422 and Kongyu131 in this study. A total of 705 predicted genes were distributed over the 21 MQTL intervals with physical length <0.3 Mb; 13 of the 21 MQTLs, and 34 candidate genes related to grain yield and plant development, were screened. Five major QTLs, viz. qGY6-2, qPH7-2, qPH6-3, qTN6-1, and qTN7-1, were not detected in the MQTL intervals and could be used as newly discovered QTLs. Candidate genes within these QTL intervals will play a meaningful role in molecular marker-assisted selection and map-based cloning of rice TN, PH, and GY.  相似文献

A plant breeding program is a long-term investment. Therefore, periodic assessment of the effectiveness of a breeding strategy is essential to maximize genetic gains per unit of time and resource invested. In this work, we assessed the effectiveness of the early-generation testing (EGT) approach used in the upland rice (Oryza sativa L.) breeding program at Embrapa (Brazilian Agricultural Research Corporation), Brazil, estimating the genetic progress achieved for three traits in two distinct phases, spanning 15 years. In the first phase (from 2003 to 2010), it was used the bulk method within F3 progenies with prior testing of F2 crosses, while in the second phase (from 2010 to 2017), it was used the bulk method within F2 progenies. The dataset comprised 70 yield trials, involving 1884 F3:5 progenies (phase I) and 925 F2:4 progenies (phase II) from an elite population, and 10 check cultivars, evaluated for grain yield (GY), plant height (PH) and days to flowering (DTF). For estimating the genetic gain, we adapted a generalized linear regression method to compute bi-segmented linear regression coefficients. Desirable genetic gains were achieved only for GY in both phases of the breeding program, with 78.75 kg ha?1 year?1 (2.68%) in the first phase, and 53.78 kg ha?1 year?1 (1.54%) in the second phase. These results show the effectiveness of EGT, especially via bulk method within F3 progenies with prior testing of F2 crosses, applied to upland rice breeding. Some refinements are discussed in the method to make it more cost-effective and more efficient in achieving genetic gains.  相似文献

1RS.1BL translocation in wheat, exploited for its multiple disease resistance (Lr26, Yr9, Sr31 and Pm8), has  maintained significance due to its agronomical advantages. However, this translocation exhibits serious defects in dough quality due to the presence of Sec-1 loci on 1RS arm. In the present investigation micro SDS sedimentation test (MST), high molecular weight glutenin subunits (HMWGS) and bread making analysis of 26 genotypes were studied along with their root phenotyping in the field and under hydroponic culture system. The MST values showed that genotypes having Sec-1 loci had low MST values but in the presence of Glu-D1 (5?+?10) with Glu-B1 (7?+?9) and (7?+?8) they had high MST values, thus overcoming the negative effects of secalin on dough quality. The loaf volume showed positive correlation with MST values of the genotypes. The translocation of 1RS arm led to higher root biomass and longer root length than Pavon 76 without 1RS. Better root traits in recombinant 1RS 44:38 and 1B?+?38 than Pavon 1RS.1BL suggested the role of negative epistatic effects between different QTL regions in 1RS arm. The results suggest that it should be possible to harness the useful alleles associated with good dough quality, better root traits, high yield and stress tolerance with or without secalin.  相似文献

Striga is an important parasitic weed causing substantial economic losses in cereal and legume crop production in sub-Saharan Africa. Integrated Striga management approaches such as a combined use of Striga resistant varieties and Fusarium oxysporum f.sp. strigae (FOS), a biocontrol agent of Striga, are an option to control the parasite and to boost sorghum productivity. Understanding host gene action influencing Striga resistance, with or without FOS treatment, is key to developing improved sorghum varieties with durable resistance and high yield. The objective of this study was to determine the gene action and inheritance of Striga resistance using genetically diverse populations of sorghum involving FOS treatment. Twelve sorghum parents selected for Striga resistance, FOS compatibility or superior agronomic performances were crossed using a bi-parental mating scheme. The selected male and female parents and their F1 progenies, backcross derivatives and the F2 segregants were field evaluated at three locations in Tanzania known for their severe Striga infestations using a lattice experimental design with two replications. The following data were collected and subjected to generation mean analysis (GMA): days-to-50% flowering (DFL), seed yield per plant (SYP) and number of Striga per plant (SN). GMA showed the preponderance of additive genetic action contributing to the total genetic variation in the evaluated sorghum populations. The additive genetic effect for DFL, SYP and SN, with and without FOS treatments, ranged from 72.02 to 86.65% and 41.49 to 95.44%, 75.62 to 91.42% and 71.83 to 91.89%, and 77.35 to 93.56% and 72.86 to 95.84%, in that order. The contribution of non-additive genetic effects was minimal and varied among generations. FOS application reduced DFL and SN and improved SYP in most of the tested sorghum populations. DFL of sorghum populations was reduced by a mean of 8 days under FOS treatment compared to the untreated control in families such as 675 × 654, AS435 × AS426 and 1563 × AS436. FOS treatment improved SYP with a mean of 6.44 g plant?1 in 3424 × 3993 and 3984 × 672. The numbers of Striga plants were reduced with a mean of 16 plants due to FOS treatment in the crosses of 675 × 654, 1563 × AS436, 4567 × AS424, and 3984 × 672. The study demonstrated that additive genes were predominantly responsible for the inheritance of Striga resistance in sorghum. Pure line cultivar development targeting reduced DFL, SN and high SYP in the selected populations may provide enhanced response to selection for integrated Striga management (ISM) programme.  相似文献

To better understand the underlying mechanisms of agronomic traits related to drought resistance and discover candidate genes or chromosome segments for drought-tolerant rice breeding, a fundamental introgression population, BC3, derived from the backcross of local upland rice cv. Haogelao (donor parent) and super yield lowland rice cv. Shennong265 (recurrent parent) had been constructed before 2006. Previous quantitative trait locus (QTL) mapping results using 180 and 94 BC3F6,7 rice introgression lines (ILs) with 187 and 130 simple sequence repeat (SSR) markers for agronomy and physiology traits under drought in the field have been reported in 2009 and 2012, respectively. In this report, we conducted further QTL mapping for grain yield component traits under water-stressed (WS) and well-watered (WW) field conditions during 3 years (2012, 2013 and 2014). We used 62 SSR markers, 41 of which were newly screened, and 492 BC4F2,4 core lines derived from the fourth backcross between D123, an elite drought-tolerant IL (BC3F7), and Shennong265. Under WS conditions, a total of 19 QTLs were detected, all of which were associated with the new SSRs. Each QTL was only identified in 1 year and one site except for qPL-12-1 and qPL-5, which additively increased panicle length under drought stress. qPL-12-1 was detected in 2013 between new marker RM1337 and old marker RM3455 (34.39 cM) and was a major QTL with high reliability and 15.36% phenotypic variance. qPL-5 was a minor QTL detected in 2013 and 2014 between new marker RM5693 and old marker RM3476. Two QTLs for plant height (qPHL-3-1 and qPHP-12) were detected under both WS and WW conditions in 1 year and one site. qPHL-3-1, a major QTL from Shennong265 for decreasing plant height of leaf located on chromosome 3 between two new markers, explained 22.57% of phenotypic variation with high reliability under WS conditions. On the contrary, qPHP-12 was a minor QTL for increasing plant height of panicle from Haogelao on chromosome 12. Except for these two QTLs, all other 17 QTLs mapped under WS conditions were not mapped under WW conditions; thus, they were all related to drought tolerance. Thirteen QTLs mapped from Haogelao under WS conditions showed improved drought tolerance. However, a major QTL for delayed heading date from Shennong265, qDHD-12, enhanced drought tolerance, was located on chromosome 12 between new marker RM1337 and old marker RM3455 (11.11 cM), explained 21.84% of phenotypic variance and showed a negative additive effect (shortening delay days under WS compared with WW). Importantly, chromosome 12 was enriched with seven QTLs, five of which, including major qDHD-12, congregated near new marker RM1337. In addition, four of the seven QTLs improved drought resistance and were located between RM1337 and RM3455, including three minor QTLs from Haogelao for thousand kernel weight, tiller number and panicle length, respectively, and the major QTL qDHD-12 from Shennong265. These results strongly suggested that the newly screened RM1337 marker may be used for marker-assisted selection (MAS) in drought-tolerant rice breeding and that there is a pleiotropic gene or cluster of genes linked to drought tolerance. Another major QTL (qTKW-1-2) for increasing thousand kernel weight from Haogelao was also identified under WW conditions. These results are helpful for MAS in rice breeding and drought-resistant gene cloning.  相似文献

Rhizomania is one of the most devastating biotic stresses affecting sugar beet (Beta vulgaris L.). It is caused by Beet necrotic yellow vein virus (BNYVV) vectored by the plasmodiophorid Polymyxa betae K. The only means available to control the disease is the use of genetically resistant varieties. “Rizor” or “Holly” (Rz1) and WB42 (Rz2) have been the most widely used resistance sources in the commercial varieties. Recently, naturally occurring resistance-breaking (RB) rhizomania strains have been identified causing major concerns. The aim of this study was to identify SNP mutations that show associations with resistance to rhizomania in sugar beet plants grown under resistance-breaking (RB)-BNYVV soils. Rhizomania virus content was evaluated by indirect triple-antibody sandwich-ELISA within two F 2 segregating populations respectively grown on an AYPR and IV-BNYVV strain infected soils. Bulked segregant analysis (BSA) was performed. The resistant and susceptible plants were genotyped with a 384-SNPs panel. Of the 384 SNPs, SNP249 was found to associate with the resistance both to the AYPR strain (R 2 = 0.37; P = 0.0004) and to the IV-BNYVV (R 2 = 0.09; P = 0.0074). Our results suggested that the SNP249 could be readily applicable for marker-assisted breeding of resistance to AYPR strain of rhizomania.  相似文献

Einkorn wheat, Triticum monococcum L. (2n = 2x = 14, AmAm genome), is a primitive, cultivated form of diploid wheat. The shortcoming of einkorn is that it lacks the free-threshing habit. Early heading and semi-dwarf traits are also required to fit modern agricultural practice. In the present study we developed T. monococcum pre-breeding germplasm having early, free threshing traits by utilizing an early heading source, two sources of soft glume (spike) and three sources of semi-dwarfism to combine their phenotypes into pre-breeding materials. We found two different genes determined free threshing of einkorn wheat. One of them was the sog (soft glume) gene from Triticum sinskajae Filat. et Kurkiev (2n = 2x = 14, AmAm genome) and another was the sos (soft spike) gene, which was completely linked or pleiotropic with the gene for semi-dwarfism. The genes sos, spd (short peduncle) and sd17654 (semi-dwarf CItr 17654) were utilized to develop semi-dwarf T. monococcum lines. Field performance of 6 early and free-threshing pre-breeding materials with sos and spd genes were tested over three crop seasons. Five semi-dwarf pre-breeding materials (PBMs) were obtained. However, these materials had slightly less grain yield than #252 (tall and hulled check) and PBM-1 (tall free-threshing check). Harvest index of the pre-breeding materials was improved due to the presence of sos and spd genes. If optimized cultivation practice is performed, these pre-breeding materials can be utilized as sources of early, free-threshing and semi-dwarf traits to produce modern T. monococcum varieties.  相似文献

Low soil nitrogen (Low N), Striga hermonthica and recurrent drought are major constraints to maize production and productivity in sub-Saharan Africa (SSA). Only a few extra-early maturing hybrids with combined tolerance to drought, Striga and low N have been commercialized in SSA. The objectives of the study were to determine the general combining ability (GCA) and specific combining ability (SCA) effects of grain yield and other traits, classify the inbreds into heterotic groups using the SCA effects of grain yield, and the heterotic group’s SCA and GCA of grain yield (HSGCA) methods, and examine the performance of hybrids under contrasting environments. Sixty-three extra-early white maize inbred lines containing genes from Zea diploperennis were crossed to four elite testers to obtain 252 single-cross hybrids and evaluated together with four checks at four locations for 2 years under drought, Striga-infested, low N and optimal environments in Nigeria. The GCA and SCA effects were significant (P ≤ 0.01) with preponderance of GCA over SCA effects for all measured traits indicating that additive genetic effects were predominant in the lines under all the contrasting environments. The HSGCA was more efficient than the SCA method in the classification of the inbreds into heterotic groups. The hybrids TZdEEI 74 × TZEEI 13 and TZdEEI 74 × TZEEI 29 were high yielding and most stable across research environments. These hybrids should be further evaluated in on-farm trials to confirm the consistency of performance for commercialization in SSA.  相似文献

Compact growth is an important quality criterion in horticulture. Many Campanula species and cultivars exhibit elongated growth which is suppressed by chemical retardation and cultural practice during production to accommodate to the consumer’s desire. The production of compact plants via transformation with wild type Agrobacterium rhizogenes is an approach with great potential to produce plants that are non-GMO. Efficient transformation and regeneration procedures vary widely among both plant genera and species. Here we present a transformation protocol for Campanula. Hairy roots were produced on 26–90% of the petioles that were used for transformation of C. portenschlagiana (Cp), a C. takesimana × C. punctata hybrid (Chybr) and C. glomerata (Cg). Isolated hairy roots grew autonomously and vigorously without added hormones. The Cg hairy roots produced chlorophyll and generated plantlets in response to treatments with cytokinin (42 µM 2iP) and auxin (0.67 µM NAA). In contrast, regeneration attempts of transformed Cp and Chybr roots lead neither to the production of chlorophyll nor to the regeneration of shoots. Agropine A. rhizogenes strains integrate split T-DNA in TL- and TR-DNA fragments into the plant genome. In this study, regenerated plants of Cg did not contain TR-DNA, indicating that a selective pressure against this T-DNA fragment may exist in Campanula.  相似文献

Thorough understanding of the genetic mechanisms governing drought adaptive traits can facilitate drought resistance improvement. This study was conducted to identify chromosome regions harbouring QTLs contributing for water stress resistance in wheat. A RIL mapping population derived from a cross between W7984 (Synthetic) and Opata 85 was phenotyped for root length and root dry weight under water stress and non-stress growing conditions. ANOVA showed highly significant (p ≤ 0.01) variation among the RILs for both traits. Root length and root dry weight showed positive and significant (p ≤ 0.01) phenotypic correlation. Broad sense heritability was 86% for root length under stress and 65% for root dry weight under non-stress conditions. A total of eight root length and five root dry weight QTLs were identified under both water conditions. Root length QTLs Qrln.uwa.1BL, Qrln.uwa.2DS, Qrln.uwa.5AL and Qrln.uwa.6AL combined explained 43% of phenotypic variation under non-stress condition. Opata was the source of favourable alleles for root length QTLs under non-stress condition except for Qrln.uwa.6AL. Four stress specific root length QTLs, Qrls.uwa.1AS, Qrls.uwa.3AL, Qrls.uwa.7BL.1 and Qrls.uwa.7BL.2 jointly explained 47% of phenotypic variation. Synthetic wheat contributed favourable alleles for Qrls.uwa.1AS and Qrls.uwa.3AL. Two stable root dry weight QTLs on chromosomes 4AL and 5AL were consistently found in both water conditions. Three validation populations were developed by crossing cultivars Lang, Yitpi, and Chara with Synthetic W7984 to transfer two of the QTLs identified under stress condition. The F2.3 and F3.4 validation lines were phenotyped under the same level of water stress as RILs to examine the effect of these QTLs. There were 13.5 and 14.5% increases in average root length due to the inheritance of Qrls.uwa.1AS and Qrls.uwa.3AL, respectively. The result indicated that closely linked SSR markers Xbarc148 (Qrls.uwa.1AS) and Xgwm391 (Qrls.uwa.3AL) can be incorporated into MAS for water stress improvement in wheat.  相似文献

Fusarium verticillioides and Aspergillus flavus cause Fusarium ear rot (FER) and Aspergillus ear rot (AER) of maize, respectively. Both pathogens are of concern to producers as they reduce grain yield and affect quality. F. verticillioides and A. flavus also contaminate maize grain with the mycotoxins fumonisins and aflatoxins, respectively, which has been associated with mycotoxicosis in humans and animals. The occurrence of common resistance mechanisms to FER and AER has been reported. Hence, ten Kenyan inbred lines resistant to AER and aflatoxin accumulation were evaluated for resistance to FER, F. verticillioides colonisation and fumonisin accumulation; and compared to nine South African lines resistant to FER and fumonisin accumulation. Field trials were conducted at three localities in South Africa and two localities in Kenya. FER severity was determined by visual assessment, while F. verticillioides colonisation and fumonisin content were quantified by real-time PCR and liquid chromatography tandem mass spectrometry, respectively. Significant genotype x environment interactions was determined at each location (P ≤ 0.05). Kenyan inbred CML495 was most resistant to FER and F. verticillioides colonisation, and accumulated the lowest concentration of fumonisins across localities. It was, however, not significantly more resistant than Kenyan lines CML264 and CKL05015, and the South African line RO549 W, which also exhibited low FER severity (≤5%), fungal target DNA (≤0.025 ng μL?1) and fumonisin levels (≤2.5 mg kg?1). Inbred lines resistant to AER and aflatoxin accumulation appear to be promising sources of resistance to F. verticillioides and fumonisin contamination.  相似文献

O. meridionalis is a wild species belonging to AA genome in the Oryza genus, which has a lot of beneficial genes for improvement of cultivated rice. In the present study, 99 chromosome single-segment substitution lines (SSSLs) were developed carrying donor segments of O. meridionalis in the genetic background of an indica cultivar, Huajingxian 74 (HJX74). The total lengths of the 99 substituted segments in the SSSLs were 1580.16 cM, with an average length of 15.11 cM per substituted segment, covering 873.94 cM and 54.98% of O.meridionalis genome. Phenotypic investigations of the SSSLs showed that three SSSLs had red pericarp, awn and showed seed shattering, respectively, indicating that these genes of O. meridionalis responsible for these traits have been transferred to the SSSLs. And wide variations were observed in seven quantitative traits including heading date and yield-related traits in 82 SSSLs.At P ≤ 0.001, 77 SSSLs showed significant differences compared with HJX74 in at least one trait either in the fall of 2014 or spring of 2015, and a total of 28 stable QTLs were detected in 24 SSSLs in both seasons. These results suggest that the SSSLs library of O. meridionalis developed in this study offers a good germplasm platform for the identification and transformation of beneficial genes of O. meridionalis, and facilitates the conservation of gene resources of O. meridionalis in vivo for long periods.  相似文献

Peach powdery mildew is one of the major diseases of the peach. Various sources of resistance to PPM have thus been identified, including the single dominant locus Vr2 carried by the peach rootstock ‘Pamirskij 5’. To map Vr2, a linkage map based on microsatellite markers was constructed from the F2 progeny (WP2) derived from the cross ‘Weeping Flower Peach’ × ‘Pamirskij 5’. Self-pollinations of the parents were also performed. Under greenhouse conditions, all progenies were scored after artificial inoculations in two classes of reactions to PPM (resistant/susceptible). In addition to Vr2, WP2 segregated for three other traits from ‘Weeping Flower Peach’: Rm1 for green peach aphid resistance, Di2 for double-flower and pl for weeping-growth habit. With their genomic locations unknown or underdocumented, all were phenotyped as Mendelian characters and mapped: Vr2 mapped at the top of LG8, at 3.3 cM, close to the CPSCT018 marker; Rm1 mapped at the bottom of LG1, at a position of 116.5 cM, cosegregating with the UDAp-467 marker and in the same region as Rm2 from ‘Rubira’®; Di2 mapped at 28.8 cM on LG6, close to the MA027a marker; and pl mapped at 44.1 cM on LG3 between the MA039a and SSRLG3_16m46 markers. Furthermore, this study revealed, for the first time, a pseudo-linkage between two traits of the peach: Vr2 and the Gr locus, which controls the red/green color of foliage. The present work therefore constitutes a significant preliminary step for implementing marker-assisted selection for the four major traits targeted in this study.  相似文献

Striga gesnerioides (Willd) Vatke, is a major destructive parasitic weed of cowpea (Vigna unguiculata (L.) Walp.) which causes substantial yield reduction in West and Central Africa. The presence of different virulent races within the parasite population contributes to significant genotype × environment interaction, and complicates breeding for durable resistance to Striga. A 3-year study was conducted at three locations in the dry savanna agro-ecology of Nigeria, where Striga gesnerioides is endemic. The primary objective of the study was to identify cowpea genotypes with high yield under Striga infestation and yield stability across test environments and to access suitability of the test environment. Data collected on grain yield and yield components were subjected to analysis of variance (ANOVA). Means from ANOVA were subjected to the genotype main effect plus genotype × environment (GGE) biplot analysis to examine the multi-environment trial data and rank genotypes according to the environments. Genotypes, environment, and genotypes × environment interaction mean squares were significant for grain yield and yield components, and number of emerged Striga plants. The environment accounted for 35.01%, whereas the genotype × environment interaction accounted for 9.10% of the variation in grain yield. The GGE biplot identified UAM09 1046-6-1 (V7), and UAM09 1046-6-2 (V8), as ideal genotypes suggesting that these genotypes performed relatively well in all study environments and could be regarded as adapted to a wide range of locations. Tilla was the most repeatable and ideal location for selecting widely adapted genotypes for resistance to S. gesnerioides.  相似文献

The germplasm of valuable for breeding wild allotetraploid potato species Solanum stoloniferum is rarely used because of pre- and postzygotic reproductive barriers with cultivated potatoes. One of the factors that complicate crosses between S. stoloniferum and S. tuberosum is unilateral incompatibility (UI). Here, we present the results of application of S. verrucosum and S v S v -lines for overcoming UI in crosses with S. stoloniferum and of generating male fertile hybrids derived from this species. S v S v -lines are F2 S. tuberosum dihaploid × S. verrucosum that are male fertile and have D/γ-type cytoplasm. Since they are homozygous for S v gene from S. verrucosum, they were expected to have the same ability for elimination of prezygotic incompatibility as this species. Three accessions of S. verrucosum and seven S v S v -lines were pollinated by 26 accessions of S. stoloniferum. The crosses with S. verrucosum failed or had low efficacy (1.5–2.4 seeds per pollination). On the other hand, use of S v S v -lines was more efficient: 15.8 seeds per pollination. In spite of low percentage of germination (1.9%), 40 seedlings of interspecific hybrids were produced. The experiment on hybridization between S v S v -lines and S. stoloniferum has been reproduced with the accession PI 205522 of the wild species, which had DNA markers of PVY and LB resistance genes and W/γ cytoplasm: 950 hybrid seeds and 12 viable seedlings were produced. The genome of the seedlings was doubled by colchicine treatment, which generated hexaploids that formed highly fertile pollen and set seeds from self-pollination. Forsythia suspensa and F.Courtaneur’ were used as female parents to cross with Abeliophyllum distichum in 2011 and an intergeneric hybrid of F. suspensa × A. distichum was obtained, though with very low seed set. The morphological characteristics, flower fragrance and volatile organic compounds of flowers were analysed. The intergeneric hybrid had intermediate morphological characteristics of both parents and flower fragrance and was confirmed as a true intergeneric hybrid by amplified fragment length polymorphism (AFLP) markers. Compared with its mother parent (F. suspensa), flowers of the intergeneric hybrid are pale yellow with delicate fragrance. Volatile organic compounds of flowers were retrieved by purge-and-trap techniques, and determined by gas chromatography and mass spectrometry (GC–MS). The main volatile organic components of F. suspensa were isoprenoids, while the main volatile organic components of A. distichum and the hybrid of F. suspensa × A. distichum were aliphatics. To determine the time and the site of intergeneric hybridizing barriers occured, the pollen tubes’ behavior after pollination was observed under fluorescence microscopy. It was found that significant pre-fertilization incompatibility existed in intergeneric crossing combinations [F. ‘Courtaneur’ (Pin) × A. distichum (Thrum) and F. suspensa (Pin) × A. distichum (Thrum)], and only a few pollen tubes of A. distichum penetrated into the ovaries of Forsythia. In our research, an intergeneric hybrid between Forsythia and Abeliophyllum was obtained for the first time, which will provide a solid foundation for expanding the flower color range of Forsythia and breeding fragrant-flowered cultivars.  相似文献
