Insect-protected corn hybrids containing Cry insecticidal proteins derived from Bacillus thuringiensis have protection from target pests and provide effective management of insect resistance. MON 89034 hybrids have been developed that produce both the Cry1A.105 and Cry2Ab2 proteins, which provide two independent modes of insecticidal action against the European corn borer ( Ostrinia nubilalis ) and other lepidopteran insect pests of corn. The composition of MON 89034 corn was compared to conventional corn by measuring proximates, fiber, and minerals in forage and by measuring proximates, fiber, amino acids, fatty acids, vitamins, minerals, antinutrients, and secondary metabolites in grain collected from 10 replicated field sites across the United States and Argentina during the 2004-2005 growing seasons. Analyses established that the forage and grain from MON 89034 are compositionally comparable to the control corn hybrid and conventional corn reference hybrids. These findings support the conclusion that MON 89034 is compositionally equivalent to conventional corn hybrids. 相似文献

Insect-protected corn hybrids containing event MON 863 protect corn plants against feeding damage from corn rootworm (Diabrotica), a major North American insect pest. Corn event MON 863 contains a gene that expresses an amino acid sequence variant of the wild-type Cry3Bb1 insecticidal protein from Bacillus thuringiensis. The purpose of this study was to compare the composition of corn containing event MON 863 with that of conventional nontransgenic corn. Compositional analyses were conducted to measure proximates, fiber, amino acids, fatty acids, minerals, folic acid, thiamin, riboflavin, vitamin E, antinutrients, and certain secondary metabolites in grain and proximates and fiber content in forage collected from a total of eight field sites in the U.S. and Argentina. Compositional analyses demonstrated that the grain and forage of event MON 863 are comparable in their nutritional content to the control corn hybrid and conventional corn. These comparisons, together with the history of the safe use of corn as a common component of animal feed and human food, support the conclusion that corn event MON 863 is compositionally equivalent to, and as safe and nutritious as, conventional corn hybrids grown commercially today. 相似文献

Developments in biotechnology and molecular-assisted breeding have led to the development of a second-generation glyphosate-tolerant soybean product, MON 89788. The MON 89788 event was produced by direct transformation of a cp4 epsps (5-enolpyruvylshikimate-3-phosphate synthase) gene cassette derived from Agrobacterium sp. strain CP4 into an elite soybean germplasm known for its superior agronomic characteristics and high yielding property. The purpose of this work was to assess whether the nutrient and antinutrient levels in seed and forage tissues of MON 89788 are comparable to those in the conventional soybean variety, A3244, which has background genetics similar to MON 89788 but does not contain the cp4 epsps gene cassette. Additional conventional soybean varieties currently in the marketplace were also included in the analysis to establish a range of natural variability for each analyte, where the range of variability is defined by a 99% tolerance interval for that particular analyte. Compositional analyses were conducted on forage, seed and four processed fractions from soybeans grown in ten sites across both the United States and Argentina during the 2004-2005 growing seasons. Forage samples were analyzed for levels of proximates (ash, fat, moisture, and protein) and fiber. Seed samples were analyzed for proximates, fiber, antinutrients, and vitamin E. Defatted, toasted (DT) meal was analyzed for proximates, fiber, amino acids, and antinutrients. Refined, bleached, and deodorized (RBD) oil was analyzed for fatty acids and vitamin E. Protein isolate was analyzed for amino acids and moisture. Crude Lecithin was analyzed for phosphatides. Results of the comparisons indicate that MON 89788 is compositionally and nutritionally equivalent to conventional soybean varieties currently in commerce. 相似文献

The next generation of biotechnology-derived products with the combined benefit of herbicide tolerance and insect protection (MON 88017) was developed to withstand feeding damage caused by the coleopteran pest corn rootworm and over-the-top applications of glyphosate, the active ingredient in Roundup herbicides. As a part of a larger safety and characterization assessment, MON 88017 was grown under field conditions at geographically diverse locations within the United States and Argentina during the 2002 and 2003-2004 field seasons, respectively, along with a near-isogenic control and other conventional corn hybrids for compositional assessment. Field trials were conducted using a randomized complete block design with three replication blocks at each site. Corn forage samples were harvested at the late dough/early dent stage, ground, and analyzed for the concentration of proximate constituents, fibers, and minerals. Samples of mature grain were harvested, ground, and analyzed for the concentration of proximate constituents, fiber, minerals, amino acids, fatty acids, vitamins, antinutrients, and secondary metabolites. The results showed that the forage and grain from MON 88017 are compositionally equivalent to forage and grain from control and conventional corn hybrids. 相似文献

In some European community countries up to 8% of the agricultural area is managed organically. The aim was to obtain a metabolite profile for wheat (Triticum aestivum L.) grains grown under comparable organic and conventional conditions. These conditions cannot be found in plant material originating from different farms or from products purchased in supermarkets. Wheat grains from a long-term biodynamic, bioorganic, and conventional farming system from the harvest 2003 from Switzerland were analyzed. The presented data show that using a high throughput GC-MS technique, it was possible to determine relative levels of a set of 52 different metabolites including amino acids, organic acids, sugars, sugar alcohols, sugar phosphates, and nucleotides from wheat grains. Within the metabolites from all field trials, there was at the most a 50% reduction comparing highest and lowest mean values. The statistical analysis of the data shows that the metabolite status of the wheat grain from organic and mineralic farming did not differ in concentrations of 44 metabolites. This result indicates no impact or a small impact of the different farming systems. In consequence, we did not detect extreme differences in metabolite composition and quality of wheat grains. 相似文献

Grain is an important source of phytochemicals, which have potent antioxidant capacity. They have been implicated in the beneficial health effect of whole grains in reducing cardiovascular disease and type 2 diabetes. The aim of the present study was to identify the most important antioxidant fractions of wheat grain. It was found that the aleurone content of these fractions was highly correlated with the antioxidant capacity of the fractions (r = 0.96, p < 0.0001). Ferulic acid appeared to be the major contributor to the antioxidant capacity in fractions with higher antioxidant capacity. The contribution of protein was rather limited. It was concluded that the antioxidant potency of wheat grain fractions is predominantly determined by aleurone content, which can be attributed to the presence of relatively large amounts of phenolic compounds, primarily ferulic acid. 相似文献

On the basis of our findings that the germination of intact wheat seeds (with husks) belonging to dormancy varieties was restrained as compared with that of the dehusked seeds (grains), the germination inhibitors in the husks were explored. The water-soluble extracts from the husks were separated by the aid of inhibition assay experiments, resulting in the characterization of 2-phenylethyl alcohol 1, 4-vinylphenol 2 and its 2-methoxy derivative 3, and dihydroactinidiolide 4, all of which showed clear inhibition of germination at 500 ppm in aqueous solution. The related compounds 1-phenylethyl alcohol 5 and tetrahydroactinidiolide 6 were as active as 1 and 4, while no noticeable difference in activity was detected among both enantiomers and the DL-form of compounds 4-6. Clear synergistic relations were observed between 4 and 1 and also 4 and 3. Since the present inhibitors have been isolated from various kinds of seed plants, they may be responsible for the general germination inhibition in the seed plants. 相似文献

Glyphosate-tolerant (Roundup Ready) corn line GA21 has been developed by genetic modification to tolerate glyphosate, the active ingredient in Roundup herbicide. The purpose of this study was to evaluate the compositional and nutritional safety of corn line GA21 compared to that of conventional corn. Compositional analyses were conducted to measure proximate, fiber, amino acid, fatty acid, and mineral contents of grain and proximate, fiber, and mineral contents of forage collected from 16 field sites over two growing seasons. The nutritional safety of corn line GA21 was evaluated in a poultry feeding study conducted with 2-day old, rapidly growing broiler chickens, at a dietary concentration of 50-60% w/w. Compositional analysis results showed that, except for a few minor differences that are unlikely to be of biological significance, the grain and forage of GA21 corn were comparable in their composition to that of the control corn line and to conventional corn. Results from the poultry feeding study showed that there were no differences in growth, feed efficiency, adjusted feed efficiency, and fat pad weights between chickens fed with GA21 grain or with parental control grain. These data taken together demonstrate that Roundup Ready corn is as safe and nutritious as conventional corn for food and feed use. 相似文献

The cultivation of Triticum spelta (spelt) has no tradition in Hungary. In recent years the interest towards this old species renewed in many countries. This high‐nutritional cereal, which has a high ash and fibre content, can be used in many health‐oriented grain‐based food products. Therefore, field experiments have been conducted for some years to test the performance of this species under home growing conditions. Here we report the results of analyses for some important quality parameters of grain samples from the 1996/97 season in comparison with those of older and new home‐grown bread wheat cultivars. Three common wheat cultivars and one advanced spelt line were grown on small plots fertilised with an NPK dose necessary to reach the highest yield and quality. Spikes were sampled weekly from the time of 70–77% grain moisture to full ripening. The grains were analysed for ash, N, P and K content and amino acid composition. Concentrations of 16 other macro + micro elements and in the ripe grains, baking quality parameters were also assessed. The grain development of spelt showed a remarkable time‐lag compared to that of the common wheat cultivars. However, the highest thousand‐grain‐masses, ash, N, and P concentrations were measured in this cultivar after milk ripening. The grains of spelt contained the macro‐nutrient Mg and four micro‐nutrients (Zn, Mn, Fe, Cu) in higher concentrations compared to those of the common wheat varieties. The total and essential amino acid concentrations measured in the ripe grains of spelt were also remarkably higher. Although its wet gluten content (47.5%) was considerably higher than that of the bread wheat cultivars, its breadmaking quality was poor. 相似文献

Two isoforms of catalase, CAT-1 and CAT-2, were purified from wheat germ after extraction, ammonium sulfate precipitations, hydrophobic chromatography, and two ionic-exchange chromatographies. The global yields and the purification factors were close to 3% and 50 for CAT-1 and close to 6% and 100 for CAT-2. Both isoforms exhibit an optimum activity at pH 7. When pH was decreased from 7 to 5.6, CAT-1 showed a decreasing affinity for its substrate, whereas the opposite was found for CAT-2. Both isoforms were irreversibly denaturated when exposed to acidic pH, CAT-1 being more sensitive than CAT-2. Conversely, CAT-2 appeared to be more sensitive to inhibitors. The rate as well as the extent of denaturation during incubation with 3-amino-1,2,4-triazole (AT) were higher with CAT-2 than with CAT-1. Guaiacol is a competitive inhibitor more potent with respect to CAT-2. The difference in affinity for hydrogen peroxide as well as the poor stability of CAT-1 in acidic medium suggests that this isoform could be less effective during dough mixing. 相似文献

The phenolic acid profiles of six cultivars of wheat with known tolerance to Fusarium head blight were evaluated during plant development from anthesis through maturity. Analysis by HPLC of grain at anthesis revealed that p-coumaric and ferulic acid were the two principal phenolic compounds present. The effect of these two phenolic acids on Fusarium species was evaluated in vitro. Both phenolic acids demonstrated significant reductions (p < 0.05) of Fusarium species growth at all concentrations tested. Ferulic acid is the primary phenolic acid in grain at all stages of development, and its concentration increased steadily during grain development prior to a 50% decrease during grain ripening. The accumulation of ferulic acid synthesis from anthesis until approximately 20 days after anthesis appears related with cultivar resistance to Fusarium. Concentrations of ferulic acid in the grain were similar at maturity, implying that the end-use quality would be similar for both resistant and susceptible cultivars. 相似文献

The recent drought in South Africa has reduced the production of both dryland and irrigated wheat. This study evaluated physiological traits of irrigated wheat genotypes in response to water stress (WS) imposed at different growth stages. A 8?×?2?×?3 [(genotypes)?×?(water treatmets; stresses and non-stressed)?×?(growth stages; tillering, flowering and grain filling)] factorial experiment based on a randomised complete block design with three replicates was conducetd. In general, the rate of photosynthesis was unaffected by WS except for genotypes LM43 at tillering and LM98 at grain filling. Stomatal conductance (SC) and transpiration rate (Tr) followed the same treand except for genotype LM35 which reduced its SC and Tr significantly at grain filling. Instantaneous waster use efficiency (IWUE) of genotype LM35 and LM57 was unaffected (p?>?0.05) by WS at tillering but at flowering stage it was affected. However, at grain filling IWUE was affected (p?<?0.05) in genotypes LM35, ML57, LM79 and LM 98. The relative water content was unaffected at tillering except for LM35 and LM47 genotypes whereas at flowering LM57, ML79, LM83 and LM98 were affected. These results indicate some degree of drought tolerance of these genotypes at different growth stages. 相似文献

Random amplified polymorphic DNA (RAPD) markers were used to estimate the genetical variability of three salt-resistant genotypes, SARC-1, SARC-5 and S-24, exposed to saline environment. High-yielding and salt-sensitive variety MH-97 was used as standard for comparison. The behavior of these genotypes under saline environment was analyzed by using the hydroponics screening methods at the seedling stage. One hundred and fifty primers were tested of which 52 primers revealed differences between SARC-1 and SARC-5, 54 revealed differences between SARC-1 and S-24 and 61 revealed differences between SARC-5 and S-24. Polymorphism differences between MH-97 and SARC-1, MH-97 and SARC-5 and MH-97 and S-24 were 53%, 64% and 42%, respectively. Four primer pairs amplified special fragments, which were located in all the three salt-resistant genotypes but none on the salt-sensitive genotype MH-97. Primer GLD-15 (5?-CCGTGGCATT-3?) generated a prominent fragment of length 1460 bp; primer GLF-18 (5?-ACCCGGAACC-3?) produced a fragment of length nearly 980 bp in the salt-resistant genotype; the primer pair GLE-5 (5?-TTCAAGCCCG-3?) located one polymorphic amplified band of 1290 bp and the primer GLH-9 (5?-ATCCAGGTCA-3?) performed as a weak polymorphic band of 640 bp, respectively. 相似文献

The composition of the microflora, N2-fixing bacteria particularly, in different soils cultivated with wheat in Egypt was investigated in some samples collected from the fields after applying the agricultural practices recommended for wheat cultivation and just before sowing. The influence of carbon sources, mineral nitrogen and water regimes on potential dinitrogen fixation (acetylene reduction assay) in soils was investigated. The bacterial population densities including-N2-fixing organisms were related to a number of environmental factors such as organic matter content. Among diazotrophs, Azotobacter spp. and Azospirillum spp. were encountered in higher densities in comparison with clostridia. Unamended soils showed a lower acetylene-reducing activity (0.5–61.5 nmoles C2H4 g?1 h?1). Addition of glucose (1% w/w) greatly enhanced such activity being the highest (86.9–2846.5 nmoles C2H4 g?1 h?1) in the clay soil with the highest organic carbon content (1.42%). Glucose amendment had no significant influence on acetylene reduction in the saline soil. N2-fixation in barley straw-amended (1%) soils was not much higher than in unamended soils. Concentrations of up to 70 ppm ammonium-nitrogen depressed N2-fixation in soils that received barley straw. Acetylene reduction in submerged soil increased after addition of cellulose. Non-flooded conditions favoured N2-fixation in the fertile clay soil amended with sucrose. 相似文献

ABSTRACTThe selection of relatively high and stable yielding genotypes is key in wheat breeding programs to improve yield performance under heat and drought-stressed environments. This study determined grain yield response and stability among elite heat and drought tolerant bread wheat genotypes under simulated drought-stressed (DS) and non-stressed (NS) environments to select promising parents for breeding. Twenty-four elite bread wheat genotypes selected from the International Maize and Wheat Improvement Centre (CIMMYT) drought and heat tolerant nursery were assessed under NS and DS conditions using a 7?×?4 alpha-lattice design under rainout shelter (RS) and glasshouse (GH) environments. Grain yield data was subjected to analysis of variance (ANOVA), the additive main effect and multiplicative interaction (AMMI) and genotype and genotype-by-environment (GGE) biplot analyses. ANOVA and AMMI revealed highly significant (p?≤?0.001) differences among test genotypes (G), environments (E) and G?×?E interaction effects suggesting differential responses for selection. The GGE biplot explained 83.76% of total variation and aided in selection of high-yielding and stable heat and drought tolerant wheat genotypes such as LM13, LM22, LM95 and LM100. Unstable breadmaking quality of wheat due to environmental influence has been a problem for Norwegian milling industries. Large variation in gluten quality was observed from field trials with Norwegian winter wheat conducted in several locations between 2005 and 2013. Moreover, extremely poor gluten quality was observed in several locations in the 2007 and 2011 season, and indicated almost complete loss of breadmaking quality. To investigate the environmental factors which cause extremely weak gluten, gluten proteins were characterized in samples selected within the 2011 season. The results revealed that the proportion of large glutenin polymers decreased in wheat samples with extremely weak gluten. Moreover, re-polymerization of large glutenin polymers, which normally occur during the resting period of a dough, did not take place in gluten prepared from these samples. Incubation of total proteins extracted from these samples in an in vitro system showed a drastic degradation of gluten proteins indicating protease activities. The origin of the proteases remains unclear; however, exogenous proteases derived from Fusarium spp. seem to play a key role for protein degradation, and thus causing severe quality deficiency. A genotypic difference was found between the two cultivars and one of them had higher resistance against the factors influencing gluten quality in negative way. 相似文献

Antioxidant activity-guided fractionation based on three in vitro antioxidant assays (Folin-Ciocalteu, TEAC, and leucomethylene blue assays) was used to identify major antioxidants in blue wheat (UC66049 Triticum aestivum L.). After consecutive extractions with solvents of various polarities and multiple chromatographic fractionations, several potent antioxidants were identified by NMR spectroscopy and mass spectrometry. Anthocyanins (delphinidin-3-glucoside, delphinidin-3-rutinoside, cyanidin-3-glucoside, and cyanidin-3-rutinoside), tryptophan, and a novel phenolic trisaccharide (β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl-(1→6)-(4-hydroxy-3-methoxyphenyl)-β-D-glucopyranoside) were the most active water-extractable constituents. However, anthocyanins were found to be major contributors to the overall blue wheat antioxidant activity only when the extraction steps were performed under acidic conditions. Alkylresorcinols were among the most active antioxidants extractable with 80% ethanol in the TEAC assay. However, this may be due to a color change instead of a bleaching of the ABTS radical. Ferulic acid was found to be the major antioxidant in alkaline cell-wall hydrolysates. The contents of the most active antioxidants were determined. 相似文献

Summary A pot experiment was conducted to study the N availability to wheat and the loss of 15N-labelled fertilizer N as affected by the rate of rice-straw applied. The availability of soil N was also studied. The straw was incorporated in the soil 2 or 4 weeks before a sowing of wheat and allowed to decompose at a moisture content of 60% or 200% of the water-holding capacity. The wheat plants were harvested at maturity and the roots, straw, and grains were analysed for total N and 15N. The soil was analysed for total N and 15N after the harvest to determine the recovery of fertilizer N in the soil-plant system and assess its loss. The dry matter and N yields of wheat were significantly retarded in the soil amended with rice straw. The availability of soil N to wheat was significantly reduced due to the straw application, particularly at high moisture levels during pre-incubation, and was assumed to cause a reduction in the dry matter and N yields of wheat. A significant correlation (r=0.89) was observed between the uptake of soil N and the dry matter yield of wheat with different treatments. In unamended soil 31.44% of the fertilizer N was taken up by the wheat plants while 41.08% of fertilizer N was lost. The plant recovery of fertilizer N from the amended soil averaged 30.78% and the losses averaged 45.55% 相似文献

Benzoxazinones are naturally occurring secondary metabolites of some Gramineae plants, responsible for their resistance to some pathogenic fungi and for their allelopathic action. Six varieties of winter wheat grown in fields under organic or conventional systems and 11 old accessions were tested for two consecutive seasons and three plant development stages for the concentration in their roots of cyclic hydroxamic acids and their degradation products. This is the first report of six benzoxazinones analyzed in plants grown in the field. An analytical technique employing LC-DAD was used for determination. It was shown that 2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one, its degradation product 6-methoxybenzoxazolin-2-one, and the lactam 2-hydroxy-7-methoxy-1,4-benzoxazin-2-one were predominant compounds in all tested samples. Their concentrations significantly differed with plant development stage and season, but no significant differences were found between varieties and between plant cultivation systems. The concentrations of 2,4-dihydroxy-1,4-benzoxazin-3-one (DIBOA) and its degradation product benzoxazolin-2-one (BOA) were much lower, ranging from 60 to 430 mg/kg of dry matter, depending on accession, stage of development, and season. There was no significant difference found between plants grown in different cultivation systems, but there were significant differences between old and new varieties; concentrations of DIBOA and its derivatives were significantly lower in old accessions. It was concluded that the concentrations of DIBOA and BOA, which are precursors of highly fungicidal 2-aminophenol, 2-amino-3H-phenoxazin-3-one, and 2-acetylamino-3H-phenoxazin-3-one, are theoretically high enough to protect plants against some soilborne pathogens. 相似文献
