The present study was conducted to evaluate non-return rate (NR), farrowing rate (FR), and number of total pigs born/litter (TB) of weaned sows after intra-uterine insemination (IUI) using low numbers of frozen–thawed (FT) spermatozoa. Semen from 6 boars was cryopreserved individually in a 0.5-ml straw, at a concentration of 1 × 109 spermatozoa/ml. A total of 40 multiparous sows with weaning-to-estrus interval of 3 to 7 days were included. The sows were detected for standing estrus twice daily and randomly assigned to two groups: I) spontaneous ovulation (n = 20) and II) induced ovulation (n = 20) which the sows were given 750 IU human chorionic gonadotrophin (hCG) i.m. immediately at estrus detection. Ovulation was determined every 12 h using transrectal ultrasonography. FT semen containing 1 × 109 motile spermatozoa/dose was used to IUI. In group I, the sows were inseminated at 24 h after the detection of estrus and repeated every 12 h until ovulation. In group II, the sows were inseminated at 36, 42 and/or 48 h after hCG treatment. The results showed that the interval from standing estrus to ovulation (EOI) differed significantly between group I (40.2 h) and group II (35.6 h; P = 0.01). Variation of EOI among sows within each group seemed to be lower in group II (4.5 h SD) than in group I (5.5 h SD; P = 0.5). The number of IUI per sow was 2.9 ± 0.6 times in group I and was 2.4 ± 0.5 times in group II. There were no significant differences (P > 0.05) in the NR (80 vs 85%), FR (60 vs 65%) and the TB (8.0 ± 2.8 vs 9.4 ± 3.7 piglets/litter) between the groups. These results indicated that multiple doses of IUI with a low number of FT boar spermatozoa provided a fairly good NR, and reasonable FR and TB both in spontaneous and induced ovulating sows. The number of inseminations required for attaining acceptable fertility tended to be lower in the weaned sows with induced ovulation.  相似文献

The response to the buck effect and the use of progesterone priming was studied in Saanen goats during the non-breeding season (April and May) in northern Mexico (26° N). Forty goats were assigned randomly in equal numbers (n = 10) to four treatment groups in a 2 × 2 factorial arrangement. Treatment consisted of two dosages of progesterone (P4) (0 or 5 mg) and two categories of goats, nulliparous and multiparous. Both parity groups were exposed to sexually active bucks previously exposed to a long-day photoperiodic scheme (16 h light day− 1). All multiparous goats (20/20) displayed estrous behavior within the first 10 days of buck exposure, while 70% (14/20) of the nulliparous goats showed estrus (P > 0.05). During the first five days of buck exposure, the percentage of unprimed goats in estrus was 70% points lower than the primed goats, although progesterone priming did not affect the occurrence of estrus (90% vs 80%; P > 0.05) after 10 days of buck stimulus. The time to onset of estrus was four days shorter (P < 0.05) in P4-treated goats compared with controls. Pregnancy rate did not differ between primed and unprimed goats (80 vs 60%; P > 0.05), but this variable was higher (P < 0.05) in multiparous than nulliparous goats (95 vs 45%). It was concluded that it is possible to induce fertile estrus in Saanen goats during the non-breeding season in northern Mexico using sexually active bucks. The use of progesterone priming can accelerate the response of goats to the buck stimulus although it was ineffective in improving the pregnancy rate of goats. Additionally, a higher proportion of the multiparous goats got pregnant compared with the nulliparous goats induced to estrus by the buck stimulus.  相似文献

We examined whether progesterone (P4)-induced suppression of LH release in cattle can be overcome by an increased dose of exogenous gonadotropin-releasing hormone (GnRH) or pretreatment with estradiol (E2). In Experiment 1, postpubertal Angus-cross heifers (N = 32) had their 2 largest ovarian follicles ablated 5 d after ovulation. Concurrently, these heifers were all given a once-used, intravaginal P4-releasing insert (CIDR), and they were randomly assigned to be given either prostaglandin F (Low-P4) or no treatment (High-P4) at follicle ablation, and 12 h later. Six days after emergence of a new follicular wave, half of the heifers in each group (n = 8) were given either 100 or 200 μg of GnRH i.m. Plasma luteinizing hormone (LH) concentrations were higher in the Low- vs High-P4 groups, and in heifers given 200 vs 100 μg of GnRH (mean ± SEM 15.4 ± 2.2 vs 9.1 ± 1.2, and 14.8 ± 2.1 vs 9.8 ± 1.4 ng/mL, respectively; P ≤ 0.01). Ovulation rate was higher (P = 0.002) in the Low-P4 group (15/16) than in the High-P4 group (6/16), but it was not affected by GnRH dose (P = 0.4). In Experiment 2, heifers (n = 22) were treated similarly, except that 5.5 d after wave emergence, half of the heifers in each group were further allocated to be given either 0.25 mg estradiol benzoate i.m. or no treatment, and 8 h later, all heifers were given 100 μg GnRH i.m. Both groups treated with E2 (Low- and High-P4) and the Low-P4 group without E2 had higher peak plasma LH concentrations compared to the group with high P4 without E2 (12.6 ± 1.8, 10.4 ± 1.8, 8.7 ± 1.3, and 3.9 ± 1.2 ng/mL, respectively; (P < 0.04)). However, E2 pretreatment did not increase ovulation rates in response to GnRH (P = 0.6). In summary, the hypotheses that higher doses of GnRH will be more efficacious in inducing LH release and that exogenous E2 will increase LH release following treatment with GnRH were supported, but neither significantly increased ovulation rate.  相似文献

The results of 5732 records of kids born between 1985 and 1996 at Gardel Agricultural Experiment Station (INRA) in Guadeloupe, were used in order to estimate the effect of kidding day (KD) on individual preweaning growth performances, total productivity of Creole goats and litter size. The flock was subjected to a restricted mating season for a long time, by using male effect. The results of the fixed linear model showed a highly significant (P<0.001) effect of KD on growth rate and total productivity of does. Live weights of kids born around the 21st day of the kidding period (KP) was 4% to 7% higher than those of kids born the first day of the KP. For total productivity of does, this ratio did not reached more than 4%. The optimum at 70 days of age occurred around 14th day of the KD with 3% of improvement of total productivity. No effect was observed upon litter size. The genetic (co)variance components were estimated by six different Individual Animal Models. The heritability (h2) estimated from the best model, was hD2=0.25±0.05 for genetic direct effect; hM2=0.09±0.04 for genetic maternal effect and the genetic correlation between direct and maternal effects was −0.86±0.12. The use of KD could be highly recommended in a breeding program in this population of Creole meat goats, since it is quite easy to record under commercial conditions as a character related to reproductive performance.  相似文献

The effects of body condition score of does and exposure to sexually active bucks after exposure to long-day artificial photoperiod were examined in mature anovulatory French Alpine goat in Northern Mexico. In June, goats in good (2.3 ± 0.2, scale 1 to 4; n = 10) or poor (1.6 ± 0.3; n = 10) body condition were exposed during 15 day to sexually active bucks, which had been exposed to long photoperiod (16:8-h light–dark cycle, starting in December). A third group of goats in good body condition was exposed to bucks kept under the natural photoperiod of this region (26° N). All goats in good body condition exposed to bucks treated with prolonged photoperiod exhibited estrus behavior, whereas only 50% of the does in poor body condition showed estrous behavior during the 15-day buck exposure. None of the does in good body condition showed estrus when exposed to bucks under natural photoperiod. These results revealed that a good body condition is required for maximum estrus response in anestrous Alpine goats and that exposure of bucks to long photoperiod in winter is essential for an adequate stimulus to reestablish estrus cycles in anovulatory Alpine does in Northern Mexico.  相似文献

Effects of grain source and dietary oil supplement on dry matter intake (DMI), feeding, chewing behavior, and production performance of lactating dairy cows were evaluated using eight multiparous Holstein cows (77±22.1 days in milk; mean±SD) in a duplicated 4×4 Latin square design with a 2×2 factorial arrangement of treatments. Experimental diets contained either ground barley or ground corn supplemented with either fish oil or soybean oil at 2% of dietary dry matter (DM). Geometric mean particle size of dietary treatments was 4.1 mm. Dry matter intake tended (P=0.09) to be greater for barley- vs. corn-based diets (23.2 vs. 22.3 kg/d), but was reduced for the fish oil compared to soybean oil supplemented diets (21.1 vs. 24.3 kg/d; P<0.001). This reduction in DMI was attributed to smaller meal size (1.24 vs. 1.55 kg of DM; P=0.004) and slower eating rate (0.082 vs. 0.098 kg of DM/min; P<0.001) for fish oil compared to soybean oil supplemented diets. Main treatment effects interacted (P=0.005) for DMI of particles retained on 19 mm sieve but not for sorting index. Eating rate (0.090 kg of DM/min) was similar between barley- and corn-based diets, however, rumination time was greater for barley- compared to corn-based diets as result of longer rumination bout duration (32.5 vs. 28.5 min/bout; P=0.01). Barley- compared to corn-based diets increased total chewing time by 71 min (709 vs. 638 min) for cows fed fish oil, but not for cows fed soybean oil. Grain source did not affect milk yield or milk composition. Compared to soybean oil, fish oil negatively affected milk yield (40.4 vs. 43.4 kg/d; P=0.01), and thereby, both milk fat (0.91 vs. 1.26 kg/d; P<0.001) and protein (1.23 vs. 1.33 kg/d; P=0.007) production. However, feed efficiency (milk yield/DMI) was greater in fish oil compared to soybean oil supplemented diets (1.94 vs. 1.80; P=0.003). Results indicated that grain source and oil supplement can interact to affect feeding and chewing behavior, but not lactational performance of lactating cows.  相似文献

The aim of this study was to evaluate the effects of methionine supplementation on energy metabolism and reproductive performance during the early post‐partum period in primiparous does. Forty nulliparous New Zealand White does were used. Females were randomized in two groups at calving: the control group (n = 20) was fed with the basal diet, and the methionine group (n = 20) was fed the basal diet plus 1 g/animal/day of methionine from the day of calving to 4 days post‐partum. Results showed that methionine supplementation increased (p = 0.032) the concentration of insulin‐like growth factor‐1 with respect to control group 4 days post‐partum. It similarly increased the prolificacy (p = 0.03), the number of kits born alive per litter (p = 0.06) and the body gain weight of the litter during supplementation (p = 0.035). These results were observed despite the does in the methionine group having a deeper negative energy balance than the does in the control group. Finally, methionine supplementation did not affect receptivity (p = 0.23), fertility (p = 0.49), the number of kits born dead per litter (p = 0.86) insulin and metabolites as glucose, non‐esterified fatty acids and triglycerides. In conclusion, our results show that methionine supplementation during the first 4 days of the post‐partum period in rabbits increases total litter size and the corporal weight of kits and is associated with an increase in blood concentration of IGF‐1.  相似文献

This study examined the influence of sex steroid hormones on lipid metabolism in horses. The group of 34 clinically healthy Standardbred trotters aged 2 to 4 years was studied during an exercise test. The horses were divided into groups according to their sex. These groups were: 11 stallions, 16 mares, and seven geldings. Concentrations of testosterone, 17-β-estradiol, leptin, ghrelin, glycerol, free fatty acids (FFA), and triacylglycerols (TG) were measured in plasma obtained from blood samples taken at rest and after the end of the exercise. At rest, plasma ghrelin concentration was significantly higher in geldings than in stallions and mares (1,541 ± 206 vs 1,280 ± 288 and 1,310 ± 267 pg/mL, respectively; P = .012). Leptin was lower in geldings than in mares (2.65 ± 0.93 vs 4.70 ± 2.31 ng/mL; P = .036). The post-exercise rise in plasma ghrelin and TG concentrations was significantly higher in mares than in geldings (+220 ± 330 vs -25 ± 206 pg/mL; P = .049 and 0.31 ± 0.14 vs 0.13 ± 0.15 mmol/L; P = .016, respectively). The increase in plasma FFA level was higher in geldings than in stallions (535 ± 178 vs 334 ± 191 μmol/L, P = .046). In conclusion, lipolysis rate in geldings is higher than in noncastrated trotters.  相似文献

The objective of the present research was to determine the effect of increasing and decreasing natural photoperiods on selected parameters of boar ejaculates. The study material consisted of 17 boars: six Polish Large White (PLW) breed, five Polish Landrace (PL) breed, and six Duroc×Pietrain (D×P) crossbreed, all aged between 8 and 12 months at the beginning of the research. Analyses were conducted on 612 ejaculates, which were collected in two experimental periods: an increasing photoperiod (IP) (January–June) and a decreasing photoperiod (DP) (July–December). A statistically proven impact of photoperiod on the volume of semen was observed in all the studied breeds (P=0.004). During the decreasing photoperiod the mean volume of semen was 261.16±75.20 ml and this was almost 17 ml higher than that for the increasing period. For boars involved in the experiment, day length also had a significant impact on the total number of motile spermatozoa (P=0.037). In the increasing photoperiod the mean number was 3.26×109 lower. A decreasing photoperiod has a positive affect on both boars and the parameters of the collected ejaculates, which were observed in the higher number of insemination doses per ejaculate. Between the different breeds, the reactions of boars to photoperiod differed and the most significant influence of photoperiod on semen parameters was noted among D×P breed boars. Least susceptible to changes in day length were PLW breed boars.  相似文献

Background

Lighting is used during conventional broiler grow-out to modify bird behaviour to reach the goals of production and improve bird welfare. The protocols for lighting intensity vary. In a field study, we evaluated if the lighting practices impact the burden of Salmonella in broiler flocks.

Methods

Conventional grow-out flocks reared in the states of Alabama, Mississippi and Texas, USA in 2003 to 2006 were sampled 1 week before harvest (n = 58) and upon arrival for processing (n = 56) by collecting feathered carcass rinsate, crop and one cecum from each of 30 birds, and during processing by collecting rinsate of 30 carcasses at pre-chilling (n = 56) and post-chilling points (n = 54). Litter samples and drag swabs of litter were collected from the grow-out houses after bird harvest (n = 56). Lighting practices for these flocks were obtained with a questionnaire completed by the growers. Associations between the lighting practices and the burden of Salmonella in the flocks were tested while accounting for variation between the grow-out farms, their production complexes and companies.

Results

Longer relative duration of reduced lights during the grow-out period was associated with reduced detection of Salmonella on the exterior of birds 1 week before harvest and on the broiler carcasses at the post-chilling point of processing. In addition, starting reduced lights for ≥18 hours per day later in the grow-out period was associated with decreased detection of Salmonella on the exterior of broilers arriving for processing and in the post-harvest drag swabs of litter from the grow-out house.

Conclusions

The results of this field study show that lighting practices implemented during broiler rearing can impact the burden of Salmonella in the flock. The underlying mechanisms are likely to be interactive.  相似文献

Synchronization of parturition in a commercial flock of sheep has many advantages including the maintenance of the efficiency of the operation as well as optimization of care for periparturient animals. In a research setting, using an ovine model for experiments involving the induction of parturition yields the results that can be applied to other mammalian species. In sheep, as well as in a variety of other farmed species, dexamethasone is the drug used to induce synchronized parturition at the end of gestation. However, considerable variations have been noticed in the time between induction and the onset of lambing. This study set out to determine the role that the breed of dam and sire, the age and reproductive history of ewes, the month of induction and the litter/lamb characteristics played in determining the time between dexamethasone treatment and parturition. The lambing was induced in 146 ewes (4 purebred Dorset, 19 purebred Rideau Arcott and 123 Rideau Arcott × Dorset ewes) using a single i.m. dose of 20 mg dexamethasone (Unidex®, Univet Pharmaceuticals, Milton, ON, Canada) on day 140 of gestation. The ewe breed was weakly correlated (r = − 0.19, P < 0.05) whereas the sire breed was moderately correlated (r = − 0.37, P < 0.001) to the time between dexamethasone treatment and the onset of parturition (the breed in this study was defined as the % of the Rideau Arcott genotype). The time between induction and the onset of labour averaged 57.3 ± 5.3 h and 64.3 ± 8.2 h in February and April, respectively, and it was significantly longer (P < 0.05) compared with the times recorded in July, September and November (42.0 ± 2.0 h, 38.7 ± 2.8 h and 36.1 ± 5.0 h, respectively). There was a negative correlation (r = − 0.41, P < 0.001) between the proportion of ram lambs and the mean birth weight of ewe lambs. These results can be interpreted to suggest that the breed and time of the year may both affect the synchrony of dexamethasone-induced lambing. The reasons for the associations found between the month of induction and the time elapsed to the onset of parturition as well as between the proportion of ram lambs and birth weights of ewe lambs are not fully understood and warrant further studies.  相似文献

To test the hypotheses that glucose transport capacity across the brush border membrane (BBM) of the large colon is lower than that of the small intestine in equids, and that small intestinal transport capacity in equids is lower than suids. d-glucose transport capacity (Vmax) and affinity (KM) across the BBM of the distal jejunum (DJ) and proximal large colon (PLC) of the pony and pig were measured. Mucosa was collected from the DJ and PLC of ponies (n = 4) and pigs (n = 3), flash-frozen in liquid nitrogen, and stored at −80°C. Jejunal and colonic BBM vesicles were manufactured, and d-glucose transport was determined. There was no detectable active uptake of glucose in the equine PLC. Compared with the pig DJ, d-glucose transport capacity was lower (2595 ± 331 vs. 655 ± 286 ρmol·mg protein−1·s−1, respectively, P < .01) and transport affinity tended to be lower (0.09 ± 0.07 vs. 0.27 ± 0.06 mM, respectively, P = .11) in the pony DJ. Compared with the pig DJ, d-glucose transport capacity (2,595 ± 331 vs. 571 ± 331 ρmol·mg protein−1·s−1, respectively, P < .001) and transport affinity (0.09 ± 0.07 vs. 0.54 ± 0.07 mM, respectively, P < .001) in the pig PLC were lower. Results show there is negligible d-glucose uptake across pony PLC, and capacity for d-glucose absorption across DJ BBM is fourfold lower in the pony compared with the pig. Results further exemplify, at a physiological level, the limited capacity of the equid small intestine to transport glucose relative to an omnivorous mammal, and the likely evolutionary adaptation of equids to low dietary levels of nonstructural carbohydrates.  相似文献

Sow lameness can result in decreased animal health and productivity, and is a significant animal welfare concern. Swine producers and veterinarians lack objective assessment tools to detect lameness. Objectives of this study were to evaluate pressure algometry (PA) and thermal sensitivity (TS) as objective assessment tools for changes in pain sensitivity associated with lameness and to assess analgesic drugs for mitigating lameness pain. Twelve mixed parity crossbred sows were anesthetized and injected with Amphotericin B in the distal interdigital space of both claws of one hind leg to induce transient lameness. Sows were randomly assigned to one of three analgesic treatment groups: (1) Sodium Salicylate (SS; 35 mg/kg per os q.12 h+0.04 ml/kg IM q.24 h sterile saline), (2) Flunixin meglumine (FM; 2.2 mg/kg IM q.24 h), or (3) Control (C; 0.04 ml/kg IM q.24 h sterile saline). All sows received each treatment over three trials with two-wk wash-out periods between trials. Forty-eight h post-induction, analgesic treatments were administered daily for four consecutive d. Pain sensitivity was assessed with PA and TS on each hind leg on d−1, d+1 and d+6 relative to induction (d0). Proc Glimmix of SAS 9.2 was used to analyze the difference between sound (S) and lame (L) legs on each trial day, with a simple effect comparison used to analyze effect of analgesia treatment on d+6. As predicted, S and L legs did not differ on d−1 (P=0.56) and less pressure was tolerated on L legs on d+1 (P<0.001) (Raw Means in kilograms of force: d−1 L 7.2±0.2; d−1 S 7.4±0.2; d+1 L 2.1±0.2; d+1 S 7.7±0.2 kgf). A simple effect comparison of d+6 revealed no differences between FM and C (p=0.90), FM and SS (p=0.17), or SS and C (p=0.07). The TS latency of S versus L legs differed on all trial days (d−1 p=0.02, d+1 p<0.0001, d+6 p<0.01) over all trials. (TS Raw means (s): d−1 L 7.3±0.6; d−1 S 9.1±0.6; d+1 L 3.3±0.2; d+1 S 6.8±0.6; d+6 L 6.0±0.6; d+6 S 8.4±0.7 s), including lame and sound days. In conclusion, these results support PA as an objective non-invasive method for measuring pain sensitivity in sows induced with transient lameness. Sodium salicylate or flunixin meglumine did not reduced pain sensitivity as measured by PA from d+1 to d+6 in this model of induced transient lameness.  相似文献

Surface litter protects rangeland soils against wind and water erosion and provides food and nesting materials for wildlife and insects. However, the ability of grassland systems to provide these services depends on the little studied topic of seasonal surface litter decomposition. Seasonal and annual surface litter decomposition rates were determined between 2014 and 2015 in central and western South Dakota at three mixed-grass prairie locations. Residue bags containing surface litter were placed in the field in late fall (1 November) of 2014 and removed after the winter (1 April), spring (1 July), and summer + fall seasons (1 November) of 2015. The litter was analyzed for total C, total N, acid detergent fiber (ADF), and acid detergent lignin (ADL). Average winter temperatures ranged from −5oC to −15oC, while summer temperatures ranged from 10oC to 35oC. Litter decomposition was lowest during the winter (0.57−0.86 g [kg × day]−1) and greatest during the summer + fall (2.12−2.69 g [kg × day]−1). Over the entire season, 40.8−62% of the surface litter decomposed. Winter litter decomposition was positively correlated with air temperature (r = 0.62, P < 0.01) and snow depth (r = 0.61, P < 0.01), and negatively correlated with C/N ratio (r = −0.65, P < 0.01), ADF (r = −0.35, P < 0.05), and ADL (r = −0.25, P < 0.05) concentrations. These findings indicate that winter decomposition cannot be ignored and that winter surface litter decomposition increases with snow depth.  相似文献

In a 2-wk experiment, the effect of photoperiod on performance and energy metabolism of newly weaned pigs was studied. Forty 4-wk-old crossbred weanling barrows weighing 8.0 kg (SE = 0.13) were assigned to one of eight groups (five pigs per group) based on BW and litter. Groups were allotted to one of two lighting schedules: 8 h light:16 h darkness or 23 h light:1 h darkness. Each group was housed in a climate respiration chamber. Piglets had ad libitum access to feed and water. Energy and nitrogen balances, heat production, ADFI, and ADG were measured weekly. Heat production, energy metabolism, and performance were unaffected (P > 0.10) by photoperiod during wk 1. However, in the 2nd wk ADFI (418 vs 302 g/d) and ADG (381 vs 240 g/d) were higher (P < 0.05 and P = 0.05, respectively) for pigs on the 23:1 h lighting schedule than for those on the 8:16 h schedule. Furthermore, heat production (P < 0.10), total energy retention, and energy retained as protein and as fat were higher (P < 0.05) during wk 2 in pigs on the 23:1 h lighting schedule (8, 125, 41, and 350%, respectively) than in those on the 8:16 h schedule. Moreover, metabolizability of energy tended to be higher (P < 0.10) and energy requirements for maintenance were lower (P < 0.05) during wk 2 for pigs on the 23:1 h schedule compared with those on the 8:16 h schedule (P < 0.10). In conclusion, exposing pigs to a longer period of light after weaning stimulated ADFI and ADG. In addition to the feed intake, the high ADG is due to an improved metabolizability of energy and a reduced energy requirement for maintenance. This study suggests that lighting schedule can be used as a tool to stimulate feed intake after weaning.  相似文献

Previously, we determined that a primary cause of proportional stunted growth in a line of Brahman cattle was related to an apparent refractoriness in metabolic response to GH in young animals. The objective of this study was to determine the effect of administration of GH, insulin (INS), and GH plus INS to mature miniature Brahman cows (n = 6; 9.7 ± 2.06 y; 391 ± 48.6 kg) and bulls (n = 8; 9.4 ± 2.00 y; 441 ± 54.0 kg) on circulating concentrations of metabolic hormones and metabolites, primarily IGF-I and IGF-I binding proteins. We hypothesized that IGF-I secretion could be enhanced by concomitant administration of exogenous GH and INS, and neither alone would be effective. Animals were allotted to a modified crossover design that included four treatments: control (CON), GH, INS, and GH + INS. At the start of the study, one-half of the cattle were administered GH (Posilac; 14-d slow release) and the other one-half served as CON for 7 d. Beginning on day 8, and for 7 d, INS (Novolin L) was administered (0.125 IU/kg BW) twice daily (7:00 AM and 7:00 PM) to all animals; hence, the INS and GH + INS treatments. Cattle were rested for 14 d and then were switched to the reciprocal crossover treatments. Blood samples were collected at 12-hour intervals during the study. Compared with CON, GH treatment increased (P < 0.01) mean plasma concentrations of GH (11.1 vs 15.7 ± 0.94 ng/mL), INS (0.48 vs 1.00 ± 0.081 ng/mL), IGF-I (191.3 vs 319.3 ± 29.59 ng/mL), and glucose (73.9 vs 83.4 ± 2.12 mg/dL) but decreased (P < 0.05) plasma urea nitrogen (14.2 vs 11.5 ± 0.75 mg/dL). Compared with INS, GH + INS treatment increased (P < 0.05) mean plasma concentration of INS (0.71 vs 0.96 ± 0.081 ng/mL), IGF-I (228.7 vs 392.3 ± 29.74 ng/mL), and glucose (48.1 vs 66.7 ± 2.12 mg/dL), decreased (P < 0.01) plasma urea nitrogen (13.6 vs 10.4 ± 0.76 mg/dL), and did not affect GH (13.5 vs 12.7 ± 0.95 ng/mL). In the miniature Brahman model, both the GH and GH + INS treatments dramatically increased circulating concentrations of IGF-I in mature cattle, suggesting that this line of Brahman cattle is capable of responding to bioactive GH.  相似文献

We have previously demonstrated that a constant intravenous infusion of kisspeptin (Kp) for 48 h in anestrous ewes induces a preovulatory luteinizing hormone (LH) surge followed by ovulation in approximately 75% of animals. The mechanisms underlying this effect are unknown. In this study, we investigated whether Kp-induced preovulatory LH surges in anestrous ewes were the result of the general activation of the whole gonadotropic axis or of the direct activation of central GnRH neurons required for the GnRH/LH surge. In the first experiment, a constant iv infusion of ovine kisspeptin 10 (Kp; 15.2 nmol/h) was given to 11 seasonally acyclic ewes over 43 h. Blood samples were taken every 10 min for 15 h, starting 5 h before the infusion, and then hourly until the end of the infusion. We found that the infusion of Kp induced a well-synchronized LH surge (around 22 h after the start of the Kp infusion) in 82% of the animals. In all ewes with an LH surge, there was an immediate but transient increase in the plasma concentrations of LH, follicle-stimulating hormone (FSH), and growth hormone (GH) at the start of the Kp infusion. Mean (± SEM) concentrations for the 5-h periods preceding and following the start of the Kp infusion were, respectively, 0.33 ± 0.09 vs 2.83 ± 0.49 ng/mL (P = 0.004) for LH, 0.43 ± 0.05 vs 0.55 ± 0.03 ng/mL (P = 0.015) for FSH, and 9.34 ± 1.01 vs 11.51 ± 0.92 ng/mL (P = 0.004) for GH. In the first experiment, surges of LH were observed only in ewes that also had a sustained rise in plasma concentrations of estradiol (E2) in response to Kp. Therefore, a second experiment was undertaken to determine the minimum duration of Kp infusion necessary to induce such a pronounced and prolonged increase in plasma E2 concentration. Kisspeptin (15.2 nmol/h) was infused for 6, 12, or 24 h in seasonally acyclic ewes (N = 8), and blood samples were collected hourly for 28 h (beginning 5 h before the start of infusion), then every 2 h for the following 22 h. Kisspeptin infused for 24 h induced LH surges in 75% of animals, and this percentage decreased with the duration of the infusion (12 h = 50%; 6 h = 12.5%). The plasma concentration of E2 was greater in ewes with an LH surge compared to those without LH surges; mean (± SEM) concentrations for the 5-h period following the Kp infusion were, respectively, 2.23 ± 0.16 vs 1.27 ± 0.13 pg/mL (P < 0.001). Collectively, our results strongly suggest that the systemic delivery of Kp induced LH surges by activating E2-positive feedback on gonadotropin secretion in acyclic ewes.  相似文献

Background

Exogenous melatonin could induce cashmere growth. However, induced growth of cashmere fleece by melatonin implants cannot be combined with the typical growth, resulting in earlier shedding followed by another cycle of cashmere growth. To address this issue, we examine the effects on the cashmere yield, fibre characteristics, and the growth and reproductive performance of cashmere goats of planned administration of melatonin.

Methods

Eighteen half-sib, female goats were assigned to two treatments (n = 9) including a control and a treatment where melatonin (2 mg/kg BW) was implanted at the end of April and end of June. Cashmere growth and shedding were observed for approximately 1 year following implantation. Fibre samples were collected monthly to determine cumulative cashmere length. Initiation and cessation dates for cashmere growth as well as the rate of cashmere growth were calculated. Cashmere yield, weight gain of dam, kidding date, litter size, and birth weight were also recorded.

Results

Melatonin implantation increased cashmere yield by 34.5 % (control 553.7 g vs. melatonin 745.0 g; P < 0.01), cashmere length by 21.3 % (control 95.2 mm vs. melatonin 115.4 mm; P < 0.01), and decreased fibre diameter by 4.4 % (control 14.6 μm vs. melatonin 14.0 μm; P < 0.03). In melatonin-treated goats, the average initiation date was earlier than in control goats (May 18, 2013 vs. July 2, 2013; P < 0.01) but there was a similar cessation date (March 22, 2014 vs. March 27, 2014). Consequently, the duration of cashmere growth was longer in melatonin-treated goats than in control goats (307 vs.270 days; P < 0.01). The final BW, average daily gain, kidding date, litter size, and birth weight were not influenced by melatonin implantation.

Conclusions

These data indicate that melatonin implantation (2 mg/kg BW) on two occasions (late April and June) increased cashmere yield by combining the induced growth of cashmere fleece with the typical growth and decreased the fibre diameter without changing dam growth rate or reproductive performance.  相似文献

This study was conducted to investigate the effects of parity and litter size on gestation length in beagle bitches. The mean duration of the initial elevation (>2 ng/mL) in progesterone concentrations after the onset of proestrus was shorter (P < 0.05) in bitches without (nulliparous) whelping experience than in bitches with (multiparous) whelping experience (6.9 d versus 8.0 d). When calculated as the interval between the day of initial elevation in progesterone concentrations and the day of whelping, the gestation length in the nulliparous bitches was noted to be similar to that in the multiparous bitches (64.3 d versus 64.2 d). No significant correlation between gestation length and litter size was observed in any of the bitches. Our results indicate that the gestation length in beagle bitches is not affected by parity or litter size.  相似文献

The objective of this study was to improve the quality of cryopreserved–thawed equine sperm using single-layer density centrifugation (SLC). Sperm quality was assessed by DNA integrity, motility, morphology, mitochondrial membrane potential, viability, and plasma membrane alteration. The percentage of DNA-damaged sperm (expressed in % COMP) was lower (P = .001) after SLC (1.6 ± 0.5% vs. 6.8 ± 0.5%). Total sperm motility (80 ± 2.4% vs. 41.7 ± 2.4%) and progressive sperm motility (69.5 ± 2.9% vs. 31.5 ± 2.9%) (P < .001), as well as the percentage of morphologically normal sperm (45 ± 3.9% vs. 27.7 ± 3.9%), increased after SLC compared with control sample. In addition, the proportion of sperm with high mitochondrial membrane potential increased (81.6 ± 1.8% vs. 42.1 ± 1.8%), as did the viability of sperm (71.1 ± 2.4% vs. 39.5 ± 2.4%), after SLC compared with the control sperm. The proportion of sperm with alteration in plasma membrane structure was lower after SLC compared with control sample (6.4 ± 1.1% vs. 18.9 ± 1.1%). Overall, sperm recovery was 72.7 ± 3.6% in the control sample compared with 14.8 ± 3.6% after SLC (P < .001). We conclude that based on the sperm parameters evaluated, SLC improves the quality of cryopreserved–thawed equine spermatozoa.  相似文献
