In our previous study, a cell wall preparation of Enterococcus faecalis strain EC‐12 (EC‐12) prevented the colonization of vancomycin‐resistant enterococci (VRE) in newly hatched broiler chicks. Relatively early prevention against VRE colonization by EC‐12 administration may be related to the enhanced innate immune system. In this study we examined the effect of EC‐12 on the kinetics of immunoglobulin A (IgA) concentration in the cecal digesta of chicks from 1 day to 14 days old to evaluate humoral immunity. We also examined the effect of EC‐12 on β‐defensin mRNA expression and lysozyme production. The IgA concentration in the cecal digesta showed a decline in chicks from 2 to 8 days old and then a recovery up to when they were 14 days old. The concentration tended to be higher in EC‐12‐administered chicks than in control chicks at 14 days old. EC‐12 administration did not stimulate the lysozyme activities in the alimentary tract. However, β‐defensin mRNA expression in the tongue and the bursa of Fabricius was higher in chicks administered EC‐12 than in the control chicks at 5 days old. These results indicate that β‐defensin may be one of the major defense mechanisms inhibiting VRE colonization in young chicks.  相似文献

Porcine edema disease (ED) is caused by Shiga toxin 2e-producing Escherichia coli (STEC). ED has become frequent in pig farms, and the use of antimicrobials has resulted in the development of antimicrobial-resistant STEC. Accordingly, the use of materials other than antimicrobials is requested for the prevention of ED. Oral administration of a heat-killed and dried cell preparation of Enterococcus faecalis strain EC-12 (EC-12) to weaning pigs was previously demonstrated to decrease animal mortality in a STEC-contaminated farm at 0.05% (w/w) dose level. In this study, pigs experimentally infected with STEC were used as a model for ED to evaluate the low dose level of EC-12 to prevent ED. Fifteen 21-day-old pigs were divided into 5 groups: STEC challenge with the basal diet, STEC challenge with EC-12 supplemented at 0.005, 0.01, or 0.05% (w/w) to the basal diet, and no STEC challenge with the basal diet. The challenge was carried out when the animals were 25, 26, and 27 days old using STEC contained in capsules resistant against gastric digestion. All pigs were euthanized at 32 days of age. The daily weight gain, feed conversion ratio, and palpebral edema were improved by supplementation with 0.05% EC-12, but not by the low dose levels. Accordingly, 0.05% level of supplementation was needed for EC-12 to improve clinical symptoms in weaning piglets infected by STEC.  相似文献

The effect of method and time of first colostrum feeding on the concentration of serum immunoglobulin (Ig) was evaluated in mithun (Bos frontalis) calves. The hypotheses were that the variable method and time of first colostrum feeding might affect the level of serum Ig and in turn the growth performance and health status of the claves during the early age. The newborn calves were randomly allotted to one of the four experimental groups - G-1: allowed to suckle the dam at own choice, G-2: separated immediately after birth and allowed to suckle the dam first at 6 h and then at own choice, G-3: bottle fed ad libitum colostrum of its own dam first at 6 h and then at 6-h intervals until 24 h, G-4: bottle fed ad libitum colostrum of its own dam within 1 h, at 6 h and then at 6-h intervals until 24 h. The concentrations of IgG, IgM, and IgA were lowest (p < 0.01) at birth and increased following colostrum feeding irrespective of the experimental group. Highest concentrations of all the Ig classes were observed at 12-24 h after birth. The concentrations then transiently decreased from day 7 to 14, and then steadily increased after day 28. The concentrations of IgG (p < 0.01) and IgA (p < 0.05) were higher in G-1 in relation to the other groups during the first week after birth. Similarly, higher concentration of IgA (p < 0.05) was found in G-1 in relation to the other groups during the rest of the experimental period. The apparent absorption efficiency of colostral IgG was higher (p < 0.05) in G-4 in relation to G-3. Growth rate and health status were not influenced by the method and time of first colostrum feeding. In conclusion, a 6-h delay in the first colostrum feeding reduced the level of serum Ig noticeably.  相似文献

The effects of partial substitution of fermented soybean meal (FSBM) for soybean meal (SBM) on immune‐physiological parameters, growth performance and diarrheal incidence in neonatal calves after microbial infection were investigated. Holstein calves (n = 12) were randomly assigned to two dietary treatments: the SBM and the FSBM group. The latter group received a calf starter containing 5% FSBM instead of SBM. In order to evaluate the immunocompetence against microbial infection, all calves were challenged with porcine live vaccine by intramuscular injection at 21 days of age. The incidence and severity of diarrhea were significantly less (P < 0.05) in the FSBM group compared to those in the SBM group at 21 and 42 days of age. Feeding FSBM starter resulted in a higher (P < 0.05) relative amount of bacterial‐specific IgA than those on SBM at days 5 and 14 post‐microbial infection (DPMI). Viral‐specific IgA also increased (P < 0.05) at 5 DPMI in the FSBM group when compared to the SBM group. The average concentration of haptoglobin of calves fed FSBM was generally higher at 3 DPMI. Our results indicate that FSBM plays a role in the alleviation of diarrhea and in the production of immune‐related effector cells such as IgA and haptoglobin in calves.  相似文献

Aseptic and septic inflammatory diseases often are associated with marked changes in tissue and sera trace element kinetics. Iron and zinc sequestration by the host may serve as a protective effect against microbial proliferation, but may deprive host tissues of these necessary elements as well. Conversely, systemic iron administration has been shown to increase susceptibility to, and severity of, infectious diseases, although deficient iron stores may be repleted. Escherichia coli enteritis in calves provides a model wherein the effects of enteric iron antagonism and parenteral iron supplementation may be studied simultaneously. Male calves (n = 12) were given (IM injection) 300 mg of iron-dextran after base-line blood samples were obtained, then the calves were allotted to 4 groups (each of 3 calves): group 1 (control)--orally given nonpathogenic E coli; group 2--orally given enterotoxigenic B44 E coli; group 3--orally given deferoxamine (50 mg/kg, twice a day); group 4--orally given gallium (4 mg/kg; twice a day). Calves were studied for 8 days; blood samples were obtained each day (day 1 through day 8) for hematologic and serum biochemical analyses. There were significant increases in serum iron concentration and % saturation in all calves within 24 hours of iron-dextran administration, which returned to base-line values in all but group 4 (given gallium) within 3 days. In the exceptional group (4), total iron-binding capacity decreased with time, as in the other groups, but serum iron concentration remained significantly increased, implying gallium interference with systemic iron assimilation.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

本试验选择年龄、胎次、体况评分、顸产期相近的中国荷斯坦奶牛40头,随机分成2组,一组自由舔饲维生素E富硒舔砖,一组不补饲舔砖,研究维生素E(VE)和硒对新生犊牛生长和免疫的影响.试验期90 d,从预产前60 d到产后30 d.结果表明,试验组母牛血清中VE的含量在产后12 h显著高于对照组(P<0.05),血清中硒的含量在产前30 d和产后12 h高于对照组,差异极显著(P<0.01).试验组母牛分娩后12 h,血清中IgG、IgM的含量,初乳中IgM、IgG、IgA的含量都显著高于对照组(P<0.05).试验组犊牛出生后12 h,血清中VE和硒含量极显著高于对照组(P<0.01),IgG、IgA的含量高于对照组,差异显著(P<0.05).试验组犊牛初生体质量和体尺显著高于对照组(P<0.05),30日龄体质量和体尺高于对照组,差异极显著(P<0.01).试验组犊牛的发病率比对照组降低了50%.  相似文献

A large amount of secretory immunoglobulin A (S‐IgA) is secreted in the alimentary tract of mammals. It has been reported that S‐IgA coats a portion of commensal intestinal bacteria in human and mouse. However, S‐IgA‐coated bacteria have not been studied in pigs and calves. In this study, we evaluated the distribution of S‐IgA‐coated commensal intestinal bacteria in each portion of the gastrointestinal tracts of pigs and calves. Immunoglobulin G (IgG)‐coated bacteria were also analyzed because a considerable amount of IgG is secreted in the gastrointestinal tracts of pigs, and in particular, calves. S‐IgA‐ or IgG‐coated bacteria were detected in all the segments of the gastrointestinal tracts of pigs and calves. The proportion of S‐IgA‐coated bacteria to total bacteria (i.e. S‐IgA coating ratio) varied in the segments of the gastrointestinal tract in pigs, whereas those of calves were nearly the same throughout the gastrointestinal tract. The S‐IgA and IgG coating ratios were higher in pigs than in calves for all segments of the gastrointestinal tract.  相似文献

The study dealt with the evaluation of the contents of the total serum immunoglobulins (CS-Ig), serum albumin, and with the phagocytic activity of blood leucocytes in trichophytosis-affected calves (aged 4 months), in dependence on the doses (revaccination) of the live avirulent vaccine against trichophytosis (the calf group R) and its administration with a zinc based-injection preparation (calf group ZnR). In calf group ZnR (5 mg Zn.kg-1 l. w.) there was a potentiating effect on the percentual proportion of potential blood phagocytes (compared with data before administration: P less than 0.01 on the first day, P less than 0.05 on the third, eighth and thirtieth day; in comparison with the group of calves R: P less than 0.01 during a 24h experiment) and on the percentage of phagocytic cells (compared with data before administration: P less than 0.05 the first and 30th day; compared with calf group R: P less than 0.05 in a 24h experiment. No effect on the index of phagocytic activity was observed. In both groups positive statistical significance was stated in the level of CS-Ig, as compared with the value before administration (on the 8th, 15th, 30th and 60th day of investigation, P less than 0.05, resp. 0.01), but there was no significance when the groups were compared with each other. Significant increase was stated in serum immunoglobulin concentration on the 15th day of examination within each group of calves. Its concentration in calf group ZnR was lower at the beginning of the experiment and at the end of the experiment it was almost exactly the same as in calf group R.  相似文献

The saturation effect of a serum and colostrum immunoglobulin preparation on the humoral immunity system of calves was studied during the first days after birth. The positive effect of the preparations depended on the dose and on the immunity level acquired through colostrum. The serum immunoglobulin preparation was used at three doses (1.2 ml, 1.6 ml, 2.2 ml per kg live weight). In the calves suffering from hypogammaglobulinaemia, a significant saturation effect was recorded only at the dose of 2.2 ml per kg live weight. This dose represented an amount greater by up to 79% than the recommended maximum therapeutic dose. The administration of the recommended dose of the colostral immunoglobulin preparation (20 ml per calf) positively influenced the concentration of serum immunoglobulins in 26% of the test calves, but these suffered from hypogammaglobulinaemia. In none of the cases of calves with a normal level of gammaglobulins and calves with diarrhoea was an increase in the concentration of serum immunoglobulins recorded. An increase in the concentration of serum immunoglobulins in the calves after the administration of the above mentioned preparations was so small that it appears impossible to use them in the saturation therapy in calves affected by hypogammaglobulinaemia and agammaglobulinaemia. Some of the mechanisms of the action of gammaglobulin preparations are discussed for the cases where the results of the administration of the preparations were positive.  相似文献

Early weaning induces villous atrophy in the small intestine (SI) of piglets. Oral administration of live lactic acid bacteria (LAB) can improve villous shortening. In this study, we evaluated the oral administration of a heat‐killed and dried cell preparation of Enterococcus faecalis (a LAB) strain EC‐12 against villous atrophy in early‐weaned mice (Experiment 1) and pigs (Experiments 2 and 3). Twelve 16‐days‐old mice were divided into two groups in Experiment 1: gavage of EC‐12 (10 mg/kg body weight (BW)/day), or control. On day 21, SI was collected. Eighteen 21‐day‐old pigs were divided into two groups in Experiment 2: gavage of EC‐12 (10 mg/kg BW/day), or control. After 10 days, the villous height of jejunum was measured. Six 21‐day‐old pigs were divided into two groups in Experiment 3: the basal diet supplemented with EC‐12 at 0.05%‐fed group, or the basal diet‐fed group. After 10 days, the villous height of jejunum was measured. The villous heights in SI were significantly higher by EC‐12 administration in all experiments. EC‐12 successfully improved the villous atrophy in the early‐weaned mice and pigs when EC‐12 was administered orally.  相似文献

Biodegradable microparticles are an efficient mucosal delivery system that protect antigens from the harsh mucosal environment and facilitate their uptake by M cells at the epithelium of mucosal-associated lymphoid tissue. In this study, we determined the systemic and mucosal immune response in calves following intranasal and oral immunization with pig serum albumin (PSA) encapsulated in alginate microparticles. The size of the particles ranged from 1 to 50 microm in diameter, with 95% of the particles being smaller than 5 microm. High levels of anti-PSA IgG1 antibodies were found in the serum, nasal secretions, and to a less extent in saliva of calves vaccinated intranasally, but not orally, with PSA-microparticles. There was no significant increase of PSA-specific IgA. A weak lymphocyte proliferative immune response was observed in peripheral blood mononuclear cells (PBMCs), and few anti-PSA antibody-secreting cells (ASC) were detected in the blood of calves immunized intranasally. The combined systemic and mucosal response observed in intranasally immunized animals may be attributed to the wide variation in the size of the alginate microparticles, with smaller particles translocating to regional lymph nodes and inducing a systemic immune response, and larger particles being retained in the NALT and inducing a mucosal immune response. The procedure presented here may be useful as an intranasal vaccine against respiratory diseases in cattle.  相似文献

用复方女贞子(由女贞子、黄芪、党参组成)和益生菌对新生奶犊牛进行了增重和免疫功能影响实验。实验选用奶犊牛20头,随机分为4组,每组5头,用药30 d,共饲养120 d,测定其对犊牛增重和血清免疫球蛋白的影响。结果显示,中药组和中药益生菌联合组的犊牛平均日增重明显提高(P〈0.01),犊牛血清中IgG、IgA、IgM含量显著升高(P〈0.01)。表明复方女贞子单用或与益生菌联合使用可促进犊牛生长,明显提高犊牛的免疫功能。  相似文献

本文旨在饲粮中添加复合特异性卵黄抗体(Ig Y)探究其对18~28日龄犊牛生长、腹泻和免疫力的影响。选取初生中国荷斯坦公犊牛20头,10日龄之前饲喂鲜奶,之后随机分为2组,对照(CON)组饲喂基础饲粮;试验(IGY)组在基础饲粮中日添加45 mg/kg BW的复合特异性Ig Y。预试期为犊牛11~17日龄,期间进行代乳品过渡,正试期为18~28日龄。分别于犊牛18和28日龄晨饲前称重,每天记录犊牛采食量并进行粪便评分。试验结束当天(28日龄)晨饲前颈静脉采血,测定血清中总抗氧化能力(T-AOC)和碱性磷酸酶(AKP)活性以及免疫球蛋白、一氧化氮(NO)、溶菌酶(LZM)浓度。结果表明:饲粮添加复合特异性Ig Y虽然对犊牛平均日增重和料重比无显著影响(P0.05),却有助于降低犊牛的粪便评分和腹泻率,作用显著(P0.05);IGY组较CON组极显著提高了犊牛血清中免疫球蛋白G(Ig G)、免疫球蛋白M(Ig M)和免疫球蛋白A(Ig A)的浓度(P0.01),分别提高7.19%、5.71%和8.50%;IGY组T-AOC极显著高于CON组(P0.01),而NO浓度却显著低于CON组(P0.05);IGY组血清LZM浓度极显著低于CON组(P0.01),复合特异性Ig Y对血清AKP活性却无显著影响(P0.05)。结果提示,饲粮中添加复合特异性Ig Y通过提高犊牛免疫球蛋白浓度和抗氧化能力以及发挥Ig Y本身抗原抗体反应,增强犊牛免疫能力和抵抗外界应激能力,降低了腹泻率,促进了犊牛健康。  相似文献

Lung and alveolar macrophage studies were conducted following experimental immunostimulation of calves with Freund's complete adjuvant (FCA). Intravenous administration of FCA to calves 35 days before aerosol exposure to serratia marcescens resulted in a 53.1% reduction in the 2-hour pulmonary mean percent retention of the organism when compared to control calves. FCA treatment increased alveolar macrophage concentration (cells/g lung) by 30.9%. Lymphoid and granulomatous responses were increased in the lungs of treated calves.  相似文献

This prospective longitudinal study examined the epidemiology and disease syndrome associated with bovine coronavirus (BCV) infections in a cohort of 8 conventional calves from 0 to 120 days of age, in two dairy herds in Ohio. The periods of respiratory shedding of BCV were determined by direct immunofluorescent (DIF) staining of nasal epithelial cells and ELISA of nasal swab supernatant fluids. The periods of fecal shedding of BCV were determined by ELISA and immunoelectron microscopy (IEM). The isotype-specific antibody titers to BCV in serum (at selected intervals between 0 and 120 days of age) and the post-suckling (24 to 48 h after birth) total immunoglobulin levels were examined by ELISA and zinc sulfate turbidity tests, respectively. Of the 8 calves studied, 4 had evidence of BCV respiratory (by DIF or ELISA) or enteric infections (by IEM or ELISA) in association with diarrhea or rhinitis, even though 7 of 8 calves showed increases in one or more serum antibody isotypes to BCV and 6 of 8 calves showed BCV respiratory or enteric antigen shedding by ELISA. Serological antibody titer increases occurred in 3 calves before 30 days of age and in 4 calves after 30 days of age; two of the latter calves had a second rise in serum antibody titers to BCV after the initial rise. A serological antibody titer increase was not observed in one calf. This suggests that BCV infections may be very common in a closed herd and may occur in older calves, although many may be subclinical and some may be recurrent. There were no statistically significant correlations between total serum immunoglobulin levels or BCV antibody isotype titers in serum (24-48 h after birth) and clinical disease or infection by BCV; however, calves with low levels of IgA BCV antibodies in serum (24-48 h after birth) had a significantly greater average number of days with diarrhea than those calves having high levels of IgA BCV-specific antibodies in serum.  相似文献

Transportation exposes cattle to stress and results in increased morbidity and mortality. An investigation was made of the effects of transport and another important stressor, weaning, on the immune function of calves by determining Immoral immune responses to keyhole limpet haemocyanin (KLH). In a 2 × 2 factorial designed experiment, suckled calves were either (1) weaned at housing (day 0) and not transported, (2) weaned at housing and transported, (3) weaned while still at pasture nine to 13 days prior to housing and not transported or (4) weaned at pasture and transported. All calves were immunized with KLH at housing (day 0) and serum samples were collected subsequently to determine class and subclass anti-KLH antibody responses (IgG1, IgG2, IgA and IgM) by direct elisa. Increased anti-KLH IgG1 and IgG2 concentrations were shown in calves that were weaned prior to housing and transported on day 10 (P<0·05 and P<0·01 respectively). Transported calves had increased IgGI concentrations on day 20 (P<0·05) compared with calves that were not transported. However, calves weaned at housing and not transported had increased IgA and IgM responses on day 30 compared with the other groups of calves (P<0·05). This study shows that transportation and weaning affect the Immoral immune responses of suckler calves and that the effects persist for several weeks. However, the effects of the treatments were not consistent for all antibody classes measured.  相似文献

Serum concentrations of chloramphenicol were determined after oral doses (55 mg/kg body weight) were administered to 7–9 day old Holstein-Friesian calves. Chloramphenicol in an oral solution produced greater serum concentrations than did an equivalent dose of chloramphenicol in capsules ( P <0.005). A second dose of each formulation administered 12 h after the first dose elevated serum chloramphenicol concentrations significantly ( P <0.001). The average serum chloramphenicol concentration exceeded 5 μg/ml of serum 1 h after administration of the solution compared with 4 h for the capsules. Average serum chloramphenicol concentration was greater than 5 μg/ml for at least 12 h after the dose was administered for both formulations. Of the eight calves receiving repeat doses of chloramphenicol, seven (87.5%) developed diarrhea in 76 ± 8.6 h. Six of the eight calves (75%) died during or shortly after the period of chloramphenicol administration.  相似文献

OBJECTIVE: To determine whether single-fraction and combination modified-live bovine respiratory syncytial virus (BRSV) vaccines commercially licensed for parenteral administration could stimulate protective immunity in calves after intranasal administration. DESIGN: Randomized controlled trial. ANIMALS: 39 calves. PROCEDURES: Calves were separated from dams at birth, fed colostrum with a minimal concentration of antibodies against BRSV, and maintained in isolation. In 2 preliminary experiments, 9-week-old calves received 1 (n = 3) or 2 (3) doses of a single-component, modified-live BRSV vaccine or no vaccine (8 control calves in each experiment), and were challenged with BRSV 21 days after vaccination. In a third experiment, 2-week-old calves received combination modified-live virus (MLV) vaccines with or without BRSV and calves were challenged with BRSV 8 days later. Calves were euthanized, and lung lesions were measured. Immune responses, including serum and nasal antibody and nasal interferon-alpha concentrations, were assessed. RESULTS: BRSV challenge induced signs of severe clinical respiratory tract disease, including death and pulmonary lesions in unvaccinated calves and in calves that received a combination viral vaccine without BRSV. Pulmonary lesions were significantly less severe in BRSV-challenged calves that received single or combination BRSV vaccines. The proportion of calves that shed virus and the peak virus titer was decreased, compared with control calves. Protection was associated with mucosal IgA antibody responses after challenge. CONCLUSIONS AND CLINICAL RELEVANCE: Single and combination BRSV vaccines administered intranasally provided clinical protection and sparing of pulmonary tissue similar to that detected in response to parenteral delivery of combination MLV and inactivated BRSV vaccines previously assessed in the same challenge model.  相似文献

A trial involving 1,803 feedlot calves was conducted under commercial feedlot conditions in western Canada to compare the relative effectiveness of a new oxytetracycline formulation, administered either intramuscularly (BMI) or subcutaneously (BMS), to a currently available oxytetracycline formulation, administered intramuscularly (LAB), for the prevention of bovine respiratory disease (BRD) in feedlot calves. All experimental treatments were administered upon arrival at the feedlot and again on the third day after arrival.

Over the entire feeding period, there were no significant differences (p≥0.05) in the BRD treatment rates or the BRD relapse rates between either the BMI or BMS groups compared to the LAB group. Similarly, there were no significant differences (p≥0.05) in the BRD treatment rates in the BMI or BMS groups from days 8-14, days 15-90, or days 1-90 of the feeding period compared to the LAB group. However, during the first seven days of the feeding period the BRD treatment rate in the BMI group was 1.55 times (p<0.05) higher than in the LAB group. From days 1-90 and day 1 to the end of the feeding period, the overall mortality rates, BRD mortality rates, and BRD case fatality rates were two to six times lower in the BMS and BMI groups as compared to the LAB group; however, these differences were not statistically significant (p≥0.05).

These data indicate that both the intramuscular and subcutaneous administration of a new oxytetracycline formulation are comparable to the intramuscular administration of a currently available oxytetracycline formulation when given to calves upon arrival at the feedlot.
