本试验旨在通过细菌分离鉴定,明确家养观赏地图鱼的死因,筛选敏感药物。采用常规方法分离纯化细菌后,进行细菌形态学观察,并通过小白鼠致病性试验、细菌主要生化鉴定、16SrDNA序列测定分析、药敏试验及耐药基因检测等方法对分离的细菌进行鉴定及耐药分析。分离出3株革兰氏阴性短杆菌,根据细菌形态特征及理化特性,结合16SrDNA序列测定与系统发育分析结果,判定其分别为肺炎克雷伯氏菌(Klebsiella pneumoniae)、维氏气单胞菌(Aeromonas veronii)和黏质沙雷氏菌(Serratia marcescens),其中肺炎克雷伯氏菌具有较强致病性。3株细菌均对洛美沙星、氧氟沙星、阿米卡星、卡那霉素敏感,对阿莫西林、氟苯尼考、克林霉素、甲硝唑等具有较强耐药性。结果表明,肺炎克雷伯氏菌、维氏气单胞菌、黏质沙雷氏菌的混合感染是家养观赏地图鱼的死亡原因。  相似文献

In the present study, samples representing Bunostomum trigonocephalum and Bunostomum phlebotomum from sheep and cattle in Heilongjiang Province, China, were characterized and grouped genetically by the first (ITS-1) and second (ITS-2) internal transcribed spacers (ITS) of nuclear ribosomal DNA (rDNA). The rDNA region including the ITS-1, 5.8S, ITS-2, and flanking 18S and 28S rDNA sequences was amplified by polymerase chain reaction (PCR), then sequenced and compared with that of other members of the hookworms available in GenBank?, and phylogenetic relationships between them were reconstructed using the Maximum-Parsimony method. The ITS-1, 5.8S, and ITS-2 sequences of the sheep hookworm were 381, 153, and 231 bp in length, respectively, and the corresponding sequences of the cattle hookworm were 392, 153, and 240 bp in length. The identity of ITS sequences of B. trigonocephalum and B. phlebotomum from sheep and cattle was 87.4%. A PCR-linked restriction fragment length polymorphism (PCR-RFLP) assay using restriction endonuclease Nde I was established for the unequivocal differentiation of the two hookworm species. Phylogenetic analyses based on the ITS sequences revealed that B. trigonocephalum and B. phlebotomum were closely related, but they represent two different species.  相似文献

Species of the genus Gynandropaa within the family Dicroglossidae are typical spiny frogs whose taxonomic status has long been in doubt. We used integrative methods, involving morphological and molecular analyses, to elucidate the phylogenetic relationships, and to determine identities and the geographic distribution of each valid species. We obtained DNA sequence data of 5 species of Gynandropaa (complete sequences of the mitochondrial NADH dehydrogenase subunit 2 [ND2] gene, and 890 bp of 12S rRNA and 16S rRNA partial sequences) from 37 localities (including the topotypes of 5 described species) and constructed Bayesian and maximum‐likelihood trees to examine the patterns of phylogeography. A total of 28 morphological variables were taken on 624 specimens. Three clades with clear geographic patterns were recognized: clade C (from south‐western Sichuan Province and central Yunnan Province), clade E (western Guizhou Province and eastern to central Yunnan Province) and clade W (western to southern Yunnan Province). Integrating morphological characteristics and distribution information, the clades W, E and C represent Gynandropaa yunnanensis, G. phrynoides and G. sichuanensis, respectively. We draw the following conclusions: (i) the taxon G. phrynoides, formerly evaluated as a junior synonym of G. yunnanensis, is revalidated herein at the rank of species; (ii) G. liui is a junior synonym of G. sichuanensis; and (iii) G. yunnanensis is a valid species while G. bourreti is probably a subspecies of G. yunnanensis, with the distribution range from Vietnam to southern Yunnan Province. This study clears up the taxonomic status of Gynandropaa and provides important information for understanding the evolution and conservation of these spiny frogs.  相似文献

The heat shock protein 70 (hsp70) genes of Babesia gibsoni, B. canis canis, B. canis vogeli, and B. canis rossi isolated from infected dogs were cloned by polymerase chain reaction (PCR) and sequenced. In the nucleotide sequence and the predicted amino acid sequence of the gene, the parasites were very similar to each other. The nucleotide sequences of the hsp70 gene had more variety than those of 18S nuclear subunit ribosomal DNA (18S rDNA). A phylogenetic analysis of these sequences and comparisons with sequences from other Babesia and Theileria species revealed that all canine babesial isolates analyzed in the present study were closely related to each other and formed one cluster. Additionally, a phylogenetic analysis of Babesia and Theileria species showed that these parasites could be divided into three groups: group A including canine babesial isolates, B. divergens, B. odocoilei, B. bovis, B. caballi, and B. ovis; group B including Theileria annulata, T. orientalis, and T. cervi; and group C including B. microti and B. rodhaini. These results suggested that a phylogenetic analysis of the hsp70 gene sequence might be helpful in classifying Babesia and Theileria species, and that canine babesial isolates might be closely related to each other, indicating their evolution from the same ancestry.  相似文献

Streptococcus dysgalactiae serogroup C, G and L strains were investigated by polymerase chain reaction (PCR) using oligonucleotide primers designed according to species‐specific parts of the 16S–23S rDNA intergenic spacer region. The oligonucleotide primers with specificity for the 16S–23S rDNA intergenic spacer region allowed a correct identification of all S. dysgalactiae serogroups C, G and L strains investigated. No cross‐reactivities could be observed with any of the control strains indicating the usefulness of PCR‐technology to identify the serologically heterogeneous species S. dysgalactiae.  相似文献

The genetic relatedness of two kidney-parasitic Sphaerospora species was studied. Although S. renicola, the causative agent of swimbladder inflammation of common carp fingerlings (Cyprinus carpio), and Sphaerospora sp. originating from goldfish (Carassius auratus auratus) were indistinguishable on the basis of spore morphology, they were found to be genetically different as their 18S rDNA sequences shared only 71.9% identical nucleotides. In the phylogenetic trees, Sphaerospora sp. from goldfish grouped with Myxidium truttae (AJ582061) within the clade of the coelozoic freshwater species. Sphaerospora renicola clustered with S. molnari (AF378345) within the group of myxosporeans histozoic in gills. The topology of the six Sphaerospora species on the phylogenetic trees implied that myxospore morphology does not correlate with the genetic relationships, and the genus seems to be polyphyletic.  相似文献

The nucleotide sequences of 18S rDNA and internal transcribed spacer (ITS) regions were used for studying the relationships of Trypanosoma evansi isolate from a buffalo. The sequences were analyzed and compared to 18S rDNA and the ITS regions of the other Trypanosoma spp. Maximum likelihood phylogenetic trees were constructed using Leishmania major as the outgroup. The tree of 18S rDNA indicated that T. evansi (buffalo B18) isolate was closely related to those of Taiwan and T. brucei stock. The ITS tree showed the genetic diversity among 32 clones of T. evansi (B18) within a single host. This data will be useful for epidemiological and dynamic studies for designing the rational control programs of the disease.  相似文献

为了确定鸡艾美耳球虫(Eimeria)不同种以及来自不同地区同种不同株之间的亲缘关系,研究其分类地位,对实验室保藏的柔嫩艾美耳球虫(Etenella)、毒害艾美耳球虫(Eneeatrix)、巨型艾美耳球虫(Emaxima)、堆形艾美耳球虫(Eaaervulina)等4种15株鸡球虫孢子化卵囊的18SrDNA基因进行克隆、测序,并与从GenBank下载的鸡球虫18SrDNA序列一起,使用软件DNAstar 5.0 MegAlign进行系统发育分析。结果显示,4种艾美耳球虫种间同源性在94.6%~99.4%之间,7株柔嫩艾美耳球虫的株间同源性在99.0%-99.9%之间,5株巨型艾美耳球虫的株间同源性在96.9%~99.8%之间。用该4种鸡球虫的18SrDNA序列与GenBank下载的另外4种鸡球虫18SrDNA序列构建系统发育树,显示这8种鸡艾美耳球虫形成2个分支,即堆形艾美耳球虫(EASH)、巨型艾美耳球虫(EMSH)、变位艾美耳球虫(Emivati)、和缓艾美耳球虫(Emitis)、布氏艾美耳球虫(Ebrunetti)、早熟艾美耳球虫(Epraecox)构成1个分支,柔嫩艾美耳球虫(ENSH)、毒害艾美耳球虫(ETAS)构成另1分支。巨型艾美耳球虫、柔嫩艾美耳球虫各株的系统发育树均根据地域关系产生2个分支。柔嫩艾美耳球虫、毒害艾美耳球虫的亲缘关系较近,不同地理区域的同种不同株的亲缘关系相对较远,种间和种内的鉴定结果与普通生物学结果一致。本研究提示18SrDNA基因可用于鸡球虫不同种/株的分类鉴定,为艾美耳球虫分子遗传学鉴定提供了理论基础。  相似文献

We conducted a phylogenetic study through karyological data, by standard staining and Ag-NOR banding, and molecular analysis (by 12S and 16S mitochondrial rRNA genes and nuclear gene C-mos) on 11 species of Malagasy geckos, genus Paroedura, and two relatives (Ebenavia inunguis and Uroplatus phantasticus). Ebenavia inunguis and U. phantasticus had 2n = 36 telocentric elements, NORs on the first chromosome pair in E. inunguis, and on the third chromosome pair in U. phantasticus. All examined Paroedura showed NORs on the smallest chromosome pair; moreover, six of the eleven examined species show a 2n = 36 karyotype, with a pair of metacentrics and 17 telocentric pair. The remaining species exhibited karyotypes with a diploid chromosome number ranging from 2n = 31 to 2n = 38. We assume that these karyotype assemblages derived from the 2n=36 karyotype by cryptic and/or simple rearrangements, such as inversions, fissions and fusions. Furthermore, molecular and/or chromosomal data indicate that Paroedura is a monophyletic genus, in which chromosome rearrangements occurred repeatedly and independently during the specific diversification. Moreover both P. bastardi and P. gracilis in current definitions are paraphyletic assemblages of several related species, since their population proves more closely related to P. ibityensis or P. oviceps than co-specific populations.  相似文献

Purpose To investigate the possible inhibition of qPCR assays used for the diagnosis of ocular infections in cats by proxymetacaine, fluorescein, and fusidic acid, which are commonly used in veterinary ophthalmology. Methods Fluorescein, proxymetacaine, and fusidic acid were tested for possible inhibition of a triplex qPCR assay designed to detect Chamydophila felis, Feline herpesvirus 1 (FHV‐1), and the feline 28S ribosomal DNA (28S rDNA) gene by comparing threshold cycle (Ct) values of samples with and without the three products. A second experiment was carried out to measure the effects of various dilutions of fusidic acid. Results No statistically significant differences were detected between the C. felis, FHV‐1, and 28S rDNA Ct values with and without proxymetacaine or fluorescein. However, there was a statistically significant increase in FHV‐1 (P < 0.01), C. felis (P < 0.01), and 28S rDNA (P < 0.05) Ct values when fusidic acid was used. When dilutions of fusidic acid were tested, the results revealed that only the 1:2 dilution caused a statistically significant increase (P < 0.01) in the FHV‐1 Ct values. Conclusion Proxymetacaine and fluorescein did not interfere with our qPCR assays for the detection of C. felis and FHV‐1. The presence of fusidic acid caused a small inhibitory effect of doubtful clinical significance. In vivo studies are required to establish the clinical relevance of this study and to confirm our findings.  相似文献

The aim of the present research is to determine the phylogenetic position of Setaria digitata of Sri Lanka in the evolutionary tree of filarial worms. DNA sequences of portions of the mitochondrial genes cytochrome c oxidase subunit 1 (CO1) and small subunit ribosomal RNA (12S rDNA) were analysed. Intra-specific variation was observed in CO1 but not in 12S rDNA. Phylogenetic trees inferred from these two genes resembled one another in recognizing monophyly of Setaria. S. digitata and Setaria labiatopapillosa appear to be sister species.  相似文献

Babesia divergens and Babesia capreoli are closely related species with distinct host ranges, a zoonotic feature being described only for B. divergens. The two species are 99.8% similar in the 18S rDNA gene sequence and indistinguishable by morphological or serological means, leading to confusion as to their species status. The phylogenetic relatedness between the two species, and the frequent involvement of surface components in serological cross-reactions led us to postulate that an ortholog of Bd37, the merozoite surface antigen described for B. divergens, could also exist in B. capreoli. We were able to amplify a single partial PCR product from B. capreoli genomic DNA using primers specific for the B. divergens merozoite surface protein coding gene Bd37, and sequencing confirmed the presence of a Bd37 ortholog in B. capreoli, named Bcp37/41. The full sequences of the Bcp37/41 genes and their intron-exon structures were obtained for two cloned lines of B. capreoli. They suggest functional homologies between Bd37 and Bcp37/41 such as their surface localization, their role in immune escape mechanism and in the initial non-specific attachment to the erythrocyte. Restriction fragment length polymorphism (RFLP) analysis of the amplicons and partial sequencing revealed an extreme polymorphism within B. capreoli, greater than the one observed for its ortholog Bd37. Such a marker could thus be useful in epidemiological as well as phylogenetic studies.  相似文献

为探究从广东省清远的鹅体内采集的蛔虫(Ascaridia anseris)的种类,对临床采集的鹅蛔虫样品,在形态观察的基础上,用保守引物NC5和NC2扩增鹅蛔虫的ITS rDNA基因片段,将扩增产物经克隆测序后,获得大小为1 009bp的鹅蛔虫ITS rDNA基因序列(GenBank登录号为KC905082)。序列比对和系统进化分析表明,鹅蛔虫5.8SrRNA基因与GenBank收录的鸡蛔虫(登录号为AJ001508)同源性为96%,而与不同地理株的鸽蛔虫的同源性在91%~96%之间;禽蛔科的鸡蛔虫、鸽蛔虫和鹅蛔虫同一进化分支,鹅蛔虫、鸡蛔虫和鸽蛔虫处于各自的独立进化分支。上述证明,鹅蛔虫是不同于鸽蛔虫的一个独立有效种,在系统发生关系上与鸡蛔虫更亲近。  相似文献

Six Theileria spp. from cattle, buffalo and black goat were identified in the Hubei province of China. In order to study the taxonomic status of these parasites, phylogenetic analysis of 18S rRNA genes were carried out. The 18S rRNA genes from each isolate were amplified by the polymerase chain reaction and the approximate 1.75 kb products were cloned and sequenced. Phylogenetic analysis of these gene sequences revealed that the five parasites from buffalo and cattle belonged to the Theileria sergenti/buffeli/orientalis group. The parasite from the Chinese goat (Macheng-Hubei, DQ286802) was closely related to Theileria luwenshuni isolated from sheep in the north of China. This represent the first report on the use of molecular phylogeny to classify Theileria spp. obtained in the Hubei province, showing that Theileria spp. from ruminants found in Hubei province belongs to the benign group of Theileria spp.  相似文献

The Casta Navarra lineage was one of the populations used to establish the fighting bull (FB) breed, and it has also been reproductively isolated from the others FBs. A total of 1284 individuals from two generations of 16 Casta Navarra herds were sampled to analyse their diversity, their genetic structure and the ability of 28 microsatellite markers to assign individuals to closely related populations. These animals were compared with closely related phylogenetic (FB) or geographical (Pirenaica and Monchina) populations. Hardy–Weinberg equilibrium analysis showed that 82% of the loci had a significant heterozygote deficit as a consequence of the Wahlund effect. The average proportion of genetic variation explained by farm differences was 9% by Wright's FST index. A phylogenetic tree constructed with a neighbour‐joining method based on Reynolds genetic distances and a Bayesian Markov chain Monte Carlo clustering approach revealed clear differences between farm groups that generally corresponded to historical information and could unambiguously differentiate Casta Navarra cattle from the other populations. The percentage of animals correctly assigned to the Casta Navarra population was 91.78% for a q threshold of >0.9. Admixture was only detected in 4.45% (< 0.8) of the cattle. These results are relevant for the maintenance and development of diversity and conservation in the Casta Navarra population.  相似文献

Abstract

A survey of 3–8-year-old peamouths Mylocheilus caurinus collected from the lower Fraser River, British Columbia, revealed 100%, infection by a myxosporean identified as Myxobolus cyprini based on spore morphology, site of infection, and associated pathological changes. Myxobolus cyprini is a common myxosporean parasite of several species of cyprinid fishes (family Cyprinidae) in Europe, the former USSR, and China, but this is the first report of this myxosporean in the Western Hemisphere. The source of the infection in peamouths—whether the parasite is endemic to North America or has been imported with exotic cyprinids from Europe or Asia—is unknown. It is also possible that the myxosporean from peamouths is very closely related to, but a distinct species from, M. cyprini of Eurasia. As reported from European cyprinid fishes, the myxosporean in peamouths developed in the skeletal muscle, and disseminated spores were found associated with chronic inflammation in the visceral organs.  相似文献

Molecular diversity of methanogens in the rumen of Korean black goats was investigated with 16S rRNA gene clone libraries using methanogen‐specific primers. The libraries were composed of rumen fluid‐associated methanogens (FAM) and rumen particle‐associated methanogens (PAM) from rumen‐fistulated Korean black goats. Among the 141 clones of the FAM library, the sequences were mostly related to two phyla, the Methanobacteriaceae family (77.3%) and the Thermoplasmatales family (22.7%); and among the 68 clones of the PAM library, sequences were also mainly clustered in the two phyla, the Thermoplasmatales family (63.24%) and the Methanobacteriaceae family (35.29%). Most of the sequenced clones in the two libraries were closely related to uncultured methanogenic archaeon. Quantitative real‐time PCR revealed that PAM (8.97 log 10) had significantly higher (P < 0.01) density of methanogens by the methanogenic 16S rRNA gene copies than FAM (7.57 log 10). The two clone libraries also showed difference in Shannon index (FAM library 1.70 and PAM library 1.59) and Chao 1 estimator (FAM library 18 and PAM library 17 operational taxonomic units). Apparent differences found in the microbial community from the two 16S rRNA gene libraries could be a result of such factors as the chemical and physical nature of the target material surface, types or component of diets, the interaction between the methanogens and other microbes, and age of the experimental goats.  相似文献

A new species of Echinoparyphium, E. montgomeriana, is described from Durban, South Africa. Certain unusual aspects of its morphology are discussed. The adult fluke is a parasite of the alimentary tract of birds. Its larval stages are common parasites of freshwater snails belonging to the family Planor-bidae including Buiinus africanus,the intermediate host ofSchistosoma haematobium and S. mattheei in the area. These intra-molluscan stages are shown to have a seasonal transmission cycle.  相似文献
