Although it has been established that maternal leukocytes traffic from colostrum into the neonatal circulation, the effects of these cells on neonatal immunity are only beginning to be understood. This study examined the effects of maternal colostral leukocytes on development and maturation of neonatal antigen presenting cells. At birth, groups of neonatal calves received whole or cell-free colostrum (CFC) from their respective dams. Peripheral blood samples were obtained over the first 4 weeks of life, and expression of surface markers associated with cellular activation and physiological stress were monitored on monocyte lineage cells. Calves receiving cell-free colostrum at birth expressed elevated levels of CD11a, CD11c, and CD14, compared to calves receiving whole colostrum (C). Calves receiving cell-free colostrum had an elevated number of monocytes in the peripheral blood during the first 2 weeks of life, however, these cells expressed lower levels of expression of CD25 and MHC class I compared to calves receiving whole colostrum. The most significant differences in marker expression occurred within the first 7 days of life.  相似文献

Evidence is reviewed in support of the hypothesis that immature unkeratinized fetal skin must be present if bovine fetal mummification is to occur. The reduction in fetal and amniotic fluid is considered to be the result of intrafetal (fetal death) or prefetal (caruncular damage) effects on the normal net fluid flow from the maternal circulation through the fetal circulation and then across the fetal skin into the amniotic cavity. As the skin is keratinized permeability is reduced drastically thus limiting fluid loss from the fetus.  相似文献

Epigenetic programs controlling development of the female reproductive tract (FRT) are influenced by the effects of naturally occurring bioactive agents on patterns of gene expression in FRT tissues during organizationally critical periods of foetal and perinatal life. Aberrations in such important cellular and molecular events, as may occur with exposure to natural or manmade steroid or peptide receptor-modulating agents, disrupt the developmental program and can change the developmental trajectory of FRT tissues, including the endometrium, with lasting consequences. In the pig, as in other mammals, maternal programming of FRT development begins pre-natally and is completed post-natally, when maternal effects on development can be communicated via signals transmitted in milk. Studies involving relaxin (RLX), a prototypic milk-borne morphoregulatory factor (MbF), serve as the basis for ongoing efforts to identify maternal programming events that affect uterine and cervical tissues in the neonatal pig. Data support the lactocrine hypothesis for delivery of MbFs to neonates as a specific consequence of nursing. Components of a maternally driven lactocrine mechanism for RLX-mediated signalling in neonatal FRT tissues, including evidence that milk-borne RLX is delivered into the neonatal circulation where it can act on RLX receptor (RXFP1) -positive neonatal tissues to affect their development, are in place in the pig. The fact that all newborn mammals drink milk extends the timeframe of maternal influence on neonatal development across many species. Thus, lactocrine transmission of milk-borne developmental signals is an element of the maternal epigenetic programming equation that deserves further study.  相似文献

ABSTRACT: The presence of foreign cells within the tissue/circulation of an individual is described as microchimerism. The main purpose of the present investigation was to study if microchimerism occurs in healthy sows/fetuses and if porcine reproductive and respiratory syndrome virus (PRRSV) infection influences this phenomenon. Six dams were inoculated intranasally with PRRSV and three non-inoculated dams served as controls. Male DNA was detected in female fetal sera of all dams via PCR. Male DNA was also detected in the maternal circulation. Sex-typing FISH showed the presence of male cells in the female fetal organs and vice versa. PRRSV infection did not influence microchimerism, but might misuse maternal and sibling microchimeric cells to enter fetuses.  相似文献

Survival of 16,838 potential embryos was determined by counting corpora lutea and fetuses at 50 d of gestation for 1,081 litters by 225 sires. These data, coded as 1 or 0 depending on whether an ovulation was represented by a fetus, were used to estimate direct and maternal additive genetic variances and their covariance for embryonic survival. Data were from first-parity gilts of a Large White-Landrace composite population subdivided into two lines, one selected for an index of ovulation rate and embryonic survival for seven generations and a contemporary control line. Variance components were obtained by ANOVA and expectations of covariances among relatives and by derivative-free restricted maximum likelihood (DFREML) in an animal model. As a trait of the embryo, heritability of direct effects obtained with ANOVA was 3.8%, heritability of maternal effects was 1.5%, and the genetic correlation between them was -.51. After adjustment of embryonic survival for ovulation rate, lower estimates of each parameter were obtained with ANOVA. Heritability of embryonic survival as a trait of the dam was 9 to 10%. Estimates of heritability of both direct and maternal effects obtained with DFREML were less than 1% and the genetic correlation between them was -.64. When survival of embryos from only those dams with 15 or more ovulations was analyzed, heritability of maternal effects was 4.4%. Estimates of common environmental effects on embryonic survival ranged from 5 to 7%.  相似文献

The aim of this work is to evaluate the haemodynamic characteristics of maternal and fetal vessels during normal pregnancy in bitches, using Colour and Pulsed wave Doppler ultrasonography, in order to obtain more information about maternal and fetal circulation. The blood waveforms of the uteroplacental arteries, aorta, caudal cava vein and umbilical cord of the fetuses were recorded weekly in 16 pregnant bitches. Also, the measurements of Peak Systolic, End Diastolic Velocities, Resistance and Pulsatility Indices were carried out. Uteroplacental blood flow was biphasic while the ones of the umbilical artery and aorta were first systolic and then diastolic. The cava showed a typical waveform of venous vessels. During gestation the EDV and PSV of fetal vessels increased (alpha<0.05) while the PI and RI of all vessels examined decreased (alpha<0.05) except for the IP of the Aorta. The Doppler ultrasonography was used to study the characteristics of maternal and fetal vessel flow and their progressive changes during pregnancy. This study can be considered a further contribution in diagnosing and monitoring high-risk pregnancies in Veterinary Medicine.  相似文献

It has been established that maternal leukocytes, conditioned by the mammary environment, cross the neonatal gut and circulate in the newborn calf. However, the impact of these cells on the development of neonatal immunity remains to be determined. This study examined the effects of maternal colostral leukocytes on development and maturation of neonatal adaptive immunity by examining the expression of surface markers on neonatal lymphocytes. At birth, neonatal calves were fed whole colostrum, or colostrum that had the maternal cells removed (cell-free colostrum), from their respective dams. Peripheral blood samples were collected at regular intervals over the first 4 weeks of life and lymphocytes were evaluated for surface expression of cellular markers. The results of these studies demonstrated that calves receiving whole colostrum had fewer CD11a positive lymphocytes in circulation during the first 2 weeks of life and this marker was expressed at a lower density than calves receiving cell-free colostrum. In addition, calves receiving whole colostrum also had a higher percentage of lymphocytes expressing the activation markers CD25 and CD26 by 7 days after birth. During the first week of life, lymphocytes from calves receiving whole colostrum had a higher density of MHC class I expression on their surfaces than cells from calves receiving cell-free colostrum. In general, these results indicate that transfer of maternal cells with colostrum allows for more rapid development of lymphocytes and maternal cells appeared to enhance their activation.  相似文献

Antibody responses to kappa (kappa)-light (L) chain are absent in normal (Ckappa+/+) animals because of tolerance due to the abundance of kappa-L chains expressed on more than 95% of all B cells and serum Ig.When heterozygous kappa-sufficient (Ckappa+/-) females are bred with homozygous kappa-deficient (Ckappa-/-) males, half of their offspring will become kappa-deficient but have received kappa-L chain containing maternal Ig, mainly IgG and IgA, through placental and intestinal transmission. The kappa-containing maternal Ig persists for more than 2 months in the circulation of the offspring. Starting from weeks 15 to 20 of age, a spontaneous antibody response towards the maternal kappa-L chains can be recorded. The time of onset, as well as the magnitude of the responses differ among individuals of the same litter. Invariably, once a response has been initiated, it transits into an IgG-type of response, which upon injection with kappa-containing protein shows the features of a secondary type of immune response.  相似文献

Foetal sex determination using polymerase chain reaction (PCR) in mammals is based on the amplification of gender‐specific foetal DNA sequences circulating in maternal blood. The bovine synepitheliochorial placenta does not allow a direct contact between the trophoblast and the maternal blood, resulting in difficult passage of foetal DNA and, consequently, its very small amounts in maternal bloodstream. Circulating cell‐free foetal DNA (ccffDNA) encompasses short nucleotide fragments (300–600 bp) in maternal circulation. The aim of this study was to assess this non‐invasive method in accurate prenatal sexing in early and late gestational periods in comparison with ultrasound diagnostics. As various DNA isolation and amplification methods were tested, their success in obtaining reliable results was evaluated. Two groups were tested, each consisting of 20 pregnant cows. Blood of a bull and a non‐pregnant heifer was the controls. Extraction of foetal DNA was accomplished by three different methods: using tubes with silicone membranes, a single‐tube extraction without silicone membranes and phenol–chloroform extraction. Following each extraction method, foetal DNA was amplified using PCR and real‐time PCR with both bAML and TSPY primers in a separate reaction. Positive results were obtained only after amplification of foetal DNA extracted with a single‐tube extraction kit. In comparison with ultrasound examination results and foetal gender recorded at birth, the sensitivity of the PCR test was 90% in Group I, but the technique failed to detect male foetuses in Group II. The real‐time PCR test sensitivity in Group I was 90% and in Group II 91.6%.  相似文献

Reliabilities for a multiple-trait maternal model were obtained by combining reliabilities obtained from single-trait models. Single-trait reliabilities were obtained using an approximation that supported models with additive and permanent environmental effects. For the direct effect, the maternal and permanent environmental variances were assigned to the residual. For the maternal effect, variance of the direct effect was assigned to the residual. Data included 10,550 birth weight, 11,819 weaning weight, and 3,617 postweaning gain records of Senepol cattle. Reliabilities were obtained by generalized inversion and by using single-trait and multiple-trait approximation methods. Some reliabilities obtained by inversion were negative because inbreeding was ignored in calculating the inverse of the relationship matrix. The multiple-trait approximation method reduced the bias of approximation when compared with the single-trait method. The correlations between reliabilities obtained by inversion and by multiple-trait procedures for the direct effect were 0.85 for birth weight, 0.94 for weaning weight, and 0.96 for postweaning gain. Correlations for maternal effects for birth weight and weaning weight were 0.96 to 0.98 for both approximations. Further improvements can be achieved by refining the single-trait procedures.  相似文献

Ornithodoros coriaceus Koch ticks were fed on 37 pregnant cows. The fetuses were obtained from the cows at 23 to 126 days after maternal tick exposure. Characteristic lesions of epizootic bovine abortion were observed only in those fetuses recovered 100 days more or after maternal tick exposure. Fetuses collected between 50 and 100 days showed mild-to-moderate lymphoid and mononuclear cell hyperplasia. Reaction in fetuses studied less than 50 days after maternal tick exposure were mild. Lesions could not be seen in 2 of the youngest fetuses. Increases in serum immunoglobulin concentrations were present only in those fetuses examined 80 days or more after their dams had been exposed to ticks. The specificity of the immunoglobulins could not be determined. Sera from 12 fetuses tested failed to fix complement in tests for group-specific chlamydial antibodies. A wide variety of microbiological cultivation attempts were made to recover the causative agent of epizootic bovine abortion from these fetuses; however, no agent was recovered regularly, and chlamydial organisms were not recovered from any. The significance of 2 recovered agents, apparently viral, is still to be determined. Fetal tissues, both frozen and fresh, collected from fetuses of dams exposed to a feeding of ticks were capable of reproducing the disease after inoculation into pregnant cows or directly into fetuses.  相似文献

In good environments, cow intake is sufficient for their own growth and for milk production to support their calf. In poor environments, cows lose BW or may reduce milk supply to maintain themselves. Heritability for direct genetic and maternal components of weaning weight as well as the correlations between these components might be expected to vary according to these circumstances. The purpose of this study was to estimate heritability and genetic correlations for the direct genetic and maternal components of weaning weight classified in 2 environments according to maternal BW gain and to identify whether a single heritability estimate is appropriate for the differing environments experienced by cows from year to year. Data used in this analysis was obtained from the Red Angus Association of America and consisted of 96,064 cow BW observations and 27,534 calf weaning weight observations. A dam's change in BW from one year to the next was used to classify each calf's weaning weight into 1 of 2 environmental groups, those being good or poor. Best linear unbiased estimates of the change in cow BW with age were obtained from analysis of cow BW using a repeatability model. If the phenotypic change in cow BW exceeded this average BW change, the calf's weaning weight associated with the end of this time frame was classified as having been observed in a good environment. If not, the calf's corresponding weaning weight was classified as having occurred in a poorer than average environment. Heritability estimates of 0.24 +/- 0.03, 0.24 +/- 0.03, 0.13 +/- 0.02, and 0.14 +/- 0.02 were obtained for weaning weight good direct, poor direct, good maternal, and poor maternal, respectively. Correlations between direct genetic and maternal weaning weight components in the good and poor environments were -0.47 +/- 0.08 and -0.20 +/- 0.09, respectively. These variance components are not sufficiently distinct to warrant accounting for dam nutritional environment in national cattle evaluation.  相似文献

Ruminant placental lactogens (PL) are members of the somatotropin, prolactin gene family that are synthesized by trophectodermal binucleate cells. The structure and biology of PL has been studied in the cow, sheep, and goat. Ruminant PL have greater structural identity to prolactin than somatotropin, although they bind to both lactogenic and somatogenic receptors. The molecular weights of ovine and caprine PL are approximately 23,000, whereas bovine PL is larger (31,000 to 34,000) due to glycosylation. Placental lactogen is secreted into both the fetal and maternal circulations. The concentration of PL in the fetus decreases with advancing gestation, whereas PL concentration peaks in the maternal circulation during the last third of pregnancy then reaches a plateau. Furthermore, the maternal concentration of PL is 100- to 1,000-fold higher in sheep and goats than in cows. The precise factors that modulate secretion of PL are unknown, although placental mass and nutrition seem to play a role. Ruminant PL have both lactogenic and somatogenic biological activities and may also have unique activities mediated through a specific receptor. There is circumstantial evidence to suggest that PL plays a role in stimulating mammogenesis. Placental lactogen secreted into the fetal compartment may also help regulate fetal growth. Direct experimental data indicate that PL can regulate maternal intermediary metabolism. Thus, it may act as a partitioning agent to regulate nutrient supply for fetal growth. The precise biological function of PL in ruminants, therefore, still needs to be defined.  相似文献

The level of maternal antibody to infectious bursal disease (IBD) virus in the circulation of one-day-old layer strain chickens was found to be on average, 45% of the antibody titre in their respective dam, while the minimum ELISA titre which protected against a challenge of 1000 CID50 of virus was 400. Maternal antibody was found to disappear from the circulation of these crossbred chickens with a half-life of 6.7 days. From these data it is possible to estimate the ELISA titre necessary in vaccinated hens to provide the desired duration of passive protection; protection being assessed by an ELISA which measures IBD viral antigen in the bursa of Fabricius following challenge.  相似文献

Telegony is the belief that the sire first mated to a female will have an influence upon some of that female’s later offspring by another male. Although the reality of telegony was acknowledged by such authorities as Darwin, Spencer, Romanes and many experienced breeders, it has been met with scepticism because of Weismann’s unfavourable comments and negative results obtained in several test experiments. In this article, alleged cases of telegony are provided. A search of the literature of cell biology and biochemistry reveals several plausible mechanisms that may form the basis for telegony. These involve the penetration of spermatozoa into the somatic tissues of the female genital tract, the incorporation of the DNA released by spermatozoa into maternal somatic cells, the presence of foetal DNA in maternal blood, as well as sperm RNA‐mediated non‐Mendelian inheritance of epigenetic changes.  相似文献

In order to determine the maternal changes contributing to the formation of the chorioallantoic and yolk-sac placentas, rat gestation sites were examined by light and electron microscopy on days 7 through 10 of pregnancy. On day 7, the implantation chamber showed different compartments and contained the blastocyst in the antimesometrial chamber. The epithelial lining of the implantation chamber disappeared at the antimesometrial chamber, transformed into disintegrated cells in the mesometrial chamber, and showed signs of the programmed cell death in the decidual crypt. On day 8, the mesometrial chamber lumen contained red blood cells and it was continuous with subepithelial sinusoids. The endothelial cells lining the mesometrial sinusoids also showed some characteristics of the sprouting type angiogenesis such as hypertrophy and cell proliferation. While the yolk-sac placental circulation was more obvious with participation of the giant trophoblasts at the antimesometrial pole of the conceptus on day 9, the antimesometrial cells showed autophagic degeneration after the formation of the chorioallantoic placenta on day 10. The contribution of the regional cell death and angiogenesis to form both of the two placentas are discussed.  相似文献

The concentration of bovine placental lactogen (bPL) was determined in fetal placentomes, allantoic fluid, amniotic fluid, maternal and fetal plasma throughout pregnancy. In addition, chromatofocusing chromatography was used to separate the different forms of bPL found both in fetal serum and in placental homogenates in order to determine whether the different forms that have been reported to exist in the cotyledon are also found in the fetal circulation. Reproductive tracts were collected from cows between 109 and 247 days of pregnancy. The concentration of bPL in the fetal cotyledonary tissue was measured by both radioreceptor assay and radioimmunoassay, both assays showed that the concentration of bPL in the fetal portion of the placentomes remained constant throughout the period of pregnancy tested. The mass of the placenta increased approximately 10-fold during the period of study but the concentration of bPL in the maternal plasma was low (0.9±0.1 ng/ml) at all stages of pregnancy tested. The mean concentration of bPL (Mean ± S.E.M.) in amniotic and allantoic fluid was 0.4±0.1 and 1.2±0.2 ng/ml respectively. Fetal blood contained the highest concentrations of bPL, from 11.6 to 18.4 ng/ml, and the concentration tended to decrease with advancing gestation (slope = 0.07, P = 0.001). Several forms of bPL were found in the fetal circulation; however, a higher percentage of forms with more acidic isoelectric points were found in the fetal serum than in placental homogenates. These results suggest that either some forms of bPL are more stable or that the hormone isolated from placental tissue is not representative of the final secreted product.  相似文献

Summary The value of a skin test for the diagnosis of Aujeszky's disease (pseudorabies) in swine was examined. Semipurified and concentrated antigens, obtained by ether/tween 80 inactivation of Aujeszky's disease virus, were inoculated intradermally into seronegative, experimentally infected and vaccinated swine and into pigs with maternal antibodies. A specific skin reaction, characterized by a visible indurated swelling, was observed within 24 hours in animals with active immunity, No reaction was seen in pigs with maternal immunity or in seronegative uninfected and unvaccinated animals. In the infected and vaccinated groups, 89% and 58% respectively of the animals with seroneutralizing antibodies were positive by the skin test response. Positive reactions were observed as early as 8 days after the infection. All but one seronegative animals remained free of antibodies after 2 consecutive skin tests and the course of decline of maternal antibodies was not changed. An earlier skin test did not lead to sensitization for a later application. The present results, together with field experience on 8 farms, revealed that the skin test could be considered a reliable method for diagnosis of Aujeszky's disease in swine on a herd basis.  相似文献

The role of maternal antibodies is to protect newborns against acute early infection by pathogens. This can be achieved either by preventing any infection or by allowing attenuated infections associated with activation of the immune system, the two strategies being based on different cost/benefit ratios. We carried out an epidemiological survey of myxomatosis, which is a highly lethal infectious disease, in two distant wild populations of rabbits to describe the epidemiological pattern of the disease. Detection of specific IgM and IgG enabled us to describe the pattern of immunity. We show that maternal immunity attenuates early infection of juveniles and enables activation of their immune system. This mechanism associated with steady circulation of the myxoma virus in both populations, which induces frequent reinfections of immune rabbits, leads to the maintenance of high immunity levels within populations. Thus, myxomatosis has a low impact, with most infections being asymptomatic. This work shows that infection of young rabbits protected by maternal antibodies induces attenuated disease and activates their immune system. This may play a major role in reducing the impact of a highly lethal disease when ecological conditions enable permanent circulation of the pathogen.  相似文献
