A nonsense mutation in DMRT3 (‘Gait keeper’ mutation) has a predominant effect on gaiting ability in horses, being permissive for the ability to perform lateral gaits and having a favourable effect on speed capacity in trot. The DMRT3 mutant allele (A) has been found in high frequency in gaited breeds and breeds bred for harness racing, while other horse breeds were homozygous for the wild‐type allele (C). The aim of this study was to evaluate further the effect of the DMRT3 nonsense mutation on the gait quality and speed capacity in the multigaited Icelandic horse and demonstrate how the frequencies of the A‐ and C‐ alleles have changed in the Icelandic horse population in recent decades. It was confirmed that homozygosity for the DMRT3 nonsense mutation relates to the ability to pace. It further had a favourable effect on scores in breeding field tests for the lateral gait tölt, demonstrated by better beat quality, speed capacity and suppleness. Horses with the CA genotype had on the other hand significantly higher scores for walk, trot, canter and gallop, and they performed better beat and suspension in trot and gallop. These results indicate that the AA genotype reinforces the coordination of ipsilateral legs, with the subsequent negative effect on the synchronized movement of diagonal legs compared with the CA genotype. The frequency of the A‐allele has increased in recent decades with a corresponding decrease in the frequency of the C‐allele. The estimated frequency of the A‐allele in the Icelandic horse population in 2012 was 0.94. Selective breeding for lateral gaits in the Icelandic horse population has apparently altered the frequency of DMRT3 genotypes with a predicted loss of the C‐allele in relatively few years. The results have practical implications for breeding and training of Icelandic horses and other gaited horse breeds.  相似文献

The objective of this study was to compare the feed intake and the apparent digestibilities of three different diets varying in concentrate‐to‐roughage ratios in weanling horses (n = 24) at 5 and 8 months of age. Horses were stratified by breed, gender, birth date and body weight and assigned to one of three dietary treatments containing the following concentrate‐to‐roughage ratios on an as‐fed basis: 70:30 (High Con), 50:50 (Equal) and 30:70 (Low Con). All horses were fed their respective diets for a 10‐day adaptation period and a 4‐day collection period at 5 and 8 months. There were no differences in BW or daily feed intake among treatments during both trials. The horses consuming Low Con had a greater amount of faecal output than High Con at both 5 and 8 months (p < 0.01). At 5 months, High Con had the highest crude protein (CP) digestibility (p < 0.05). At 8 months, High Con had a higher CP digestibility than Low Con (p < 0.01) and tended to be higher than Equal (p = 0.07). Acid detergent fibre (ADF) digestibility did not differ among treatments; however, horses fed the Low Con tended to digest a higher percentage of neutral detergent fibre (NDF) than both the Equal and High Con treatments (p = 0.09). Horses in the High Con treatment tended to digest a higher percentage of energy than those in the Low Con treatment (p = 0.06). Weanlings seem to digest protein more thoroughly when fed high‐concentrate diets and may digest fibre more efficiently when fed diets higher in fibre.  相似文献

Invasive procedures in animals are challenging for veterinary students who may perceive a gynaecological examination of mares as stressful. Simulator‐based training may reduce stress. In this study, students received equine gynaecology training 4 times either on horses (group H; n = 14) or a teaching simulator (group SIM; n = 13). One day and 14 days thereafter, their diagnostic skills were tested on horses (skills tests 1 and 2). During the skills tests, the students’ stress response was analysed by heart rate, heart rate variability (HRV) parameters SDRR (standard deviation of beat‐to‐beat [RR] interval) and RMSSD (root‐mean‐square of successive RR differences), and salivary cortisol. In addition, students answered a questionnaire on their perceived stress. Sympathetic activation with increased heart rate (p < 0.001) occurred in both skills tests. In test 1, this increase was more pronounced in SIM than in H students (time × group p < 0.01). HRV decreased in students of both groups (p < 0.001). In skills test 1, this decrease was more pronounced for SIM than for H students (between groups and time × group p < 0.01 for SDRR and p < 0.05 for RMSSD). High cortisol concentrations before the skills tests may indicate an anticipatory stress response. Subjective stress perception of students was higher in skills test 1 vs 2 (p < 0.01). In skills test 2, H students felt more stressed than SIM students (p < 0.01). Self‐assessment thus differed from physiological stress parameters. In conclusion, gynaecological examination of mares evoked a moderate stress response in veterinary students, which was more evident after simulator‐based than animal‐based training.  相似文献

Yaks and indigenous Qaidam cattle and cattle‐yak crosses (C × Y) graze on the Qinghai‐Tibetan Plateau (QTP) throughout the year, but yaks are raised at higher elevations than cattle. Yaks do not receive supplementary feed whereas cattle require supplementary feed during harsh winter. We hypothesized that yaks would cope with the severe conditions of the QTP better than cattle and utilize the pasture more efficiently. We also hypothesized that differences between species would be pronounced in winter, when conditions are particularly harsh. To test these hypotheses, seasonal rumen fluid parameters of yaks, C × Y and cattle (n = 3 for each) were examined. Rumen fluid was collected in summer and winter from each genotype 2, 5 and 12 hr after a day of grazing. Concentrations of total volatile fatty acid (VFA), acetate, propionate, isobutyrate and isovalerate were greater in yaks than in cattle in summer (p < 0.05), while propionate concentration was lower in yaks than in cattle in winter (p < 0.05). Concentrations of ammonia and urea were greater (p < 0.001) in yaks than in cattle (p < 0.001) in summer, whereas, concentrations of free amino acids (AA) were greater in cattle than in yaks in summer and winter (p < 0.001). Concentrations of total VFA, acetate, propionate and butyrate decreased linearly (p < 0.05), whereas concentrations of isobutyrate and isovalerate increased linearly for yak and C × Y with sampling time (p < 0.05) in summer. In summer, concentrations of isobutyrate and isovalerate were greater in yaks than in cattle (p < 0.05). In conclusion, rumen fermentation characteristics of yaks showed that they coped better than cattle or C × Y in the harsh climate as we hypothesized. However, in contrast to our hypothesis, this emerged only in summer, when pasture was plentiful and not in winter, when pasture was scarce.  相似文献

Abstract

The genetic associations between racing performance and preselection of horses considered as the binary trait racing status (trotters without or with at least one racing performance in life were classified as 0 and 1, respectively) as well as disqualified races (disqualified and non-disqualified trotters were classified as 1 and 0, respectively) were analysed in German trotters. Variance components for racing performance traits square root of rank at finish, racing time per km, and log of earnings with racing status were estimated based on an animal model using REML. Heritabilities of racing status, racing time and rank at finish were 0.30, 0.21, and 0.06, respectively. The genetic correlations between racing status and racing time or rank at finish were ?0.74 and ?0.32, indicating that horses started at least once showed a higher genetic potential in racing time or finishing ability than never started horses. This showed the high preselection of German trotters especially based on racing time. To account for this preselection, it was recommended for additional use of racing status in the German evaluation system. Breeding values of the three racing performance traits were estimated by two distinct models, in- or excluding racing status and compared by using three criteria. Racing time per km showed the highest correlation (r=0.98) between breeding values evaluated by these two distinct models. Therefore, incorrect selection rate of horses using breeding values from the model without racing status, was lowest for racing time per km (9.7%). Selection response increased about 1% for this trait after including racing status in the model. For the estimation of rank at finish, inclusion of racing status in the multiple trait model was much more important as indicated by a low correlation between breeding values (r=0.29) and high percentage of incorrectly selected stallions (97.5%). The trait disqualified races was first analysed using an univariate threshold model. Heritability of this trait was low (h 2=0.12) and repeatability (r=0.43) showed a moderate magnitude. Using a linear multiple trait animal model, disqualified races showed a low heritability (h 2=0.05) and a moderate favourable genetic correlation (r g=0.43) with racing time per km. Consequently, selection on racing time per km is expected to improve indirectly the reliability of racing performance. Combined selection of reduction in disqualified races and racing time may even further improve the reliability of racing trotters.  相似文献

The reasons for this prospective experimental study were to determine a dosing scheme with loading and maintenance dose of aspirin inducing inhibition of platelet function measured by whole blood impedance aggregometry. Ten horses received aspirin orally in the morning with one loading dose of 4.7–5 mg/kg and maintenance doses of 1–1.3 mg/kg daily the following 4 days. Aggregometries (COLtest, ASPItest, ADPtest) and serum salicylic acid were measured. ASPItest showed significant difference in inhibition at 24 and 48 hr (p < .05) and 96 hr (p < .01). Significant change for ADPtest and COLtest couldn't be detected. Serum salicylic acid concentrations were significantly (p < .01) increased at 6 and 12 hr. Despite this, three horses failed any inhibitory effect of platelet function, suspecting an aspirin resistance. Regarding the other seven horses platelet aggregation induced by ASPItest was reduced between 37% and 100% from baseline at 6 and 12 hr and between 0 and 98% during the next 4 days. Correlations of serum concentration of salicylic acid and aggregometries couldn't be detected. It can be presumed that equine platelets are less susceptible to aspirin what may compromise eventually the anticoagulatory effects and efficacy in preventing and treating diseases with increased platelet activation as endotoxaemia or laminitis.  相似文献

The effect of Lippia citriodora extract on selected blood parameters and on plasma oxidative markers in Avelignese horses was evaluated. Twenty‐four horses were divided into three groups, consisting of eight animals each. Results of two experimental groups, 0.5 mg of verbascoside per kg of metabolic body weight (bw0.75) in the low‐dose group (LVB) and 1.0 mg of verbascoside per kg of metabolic body weight (bw0.75) in the high‐dose group (HVB), were compared to the control group (CON). Groups fed L. citriodora extract (HVB and LVB) showed a significant decrease in triglycerides, total cholesterol and LDL cholesterol (p < .01), bilirubin, and transaminases (p < .05), and an increase in HDL cholesterol (p < .01) compared to the CON group. Oxidative status was improved due to significant decrease in plasma concentration of ROMs and TBARS (p < .01) and increase in levels of vitamin A and vitamin E (p < .01). Based on obtained results, it is assumed that dietary supplementation with L. citriodora extract might find a useful application in horse feeding, with positive impact observed in blood parameters and plasma oxidative markers, with beneficial effects on the physiological welfare of livestock animals.  相似文献

The influence of temperament on the alteration of metabolic parameters in response to a lipopolysaccharide (LPS) challenge was investigated. Brahman bulls were selected based on temperament score. Bulls (10 months; 211 ± 5 kg BW; n = 6, 8 and 7 for Calm, Intermediate and Temperamental groups, respectively) were fitted with indwelling jugular catheters to evaluate peripheral blood concentrations of glucose, blood urea nitrogen (BUN), non‐esterified fatty acids (NEFA), insulin, epinephrine and cortisol before and after LPS administration (0.5 μg/kg BW LPS). Feed intake was also recorded. Intermediate bulls consumed more feed than the Temperamental bulls during the challenge (p = 0.046). Pre‐LPS glucose (p = 0.401) and BUN (p = 0.222) did not differ among the temperament groups. However, pre‐LPS insulin (p = 0.023) was lower, whereas pre‐LPS NEFA (p < 0.001), cortisol (p < 0.001) and epinephrine (p < 0.001) were greater in Temperamental than in Calm and Intermediate bulls. Post‐LPS glucose was increased in Calm and Intermediate bulls but not in Temperamental bulls (p < 0.001). Insulin concentrations post‐LPS were greater in Calm than in Intermediate and Temperamental bulls (p < 0.001). Concentrations of NEFA post‐LPS were greater in Temperamental than in Calm and Intermediate bulls (p < 0.001). Serum BUN concentration increased post‐LPS, with values being greater in Calm and Intermediate than in Temperamental bulls (p = 0.012). Collectively, these data demonstrate that animal temperament is related to the metabolic responses of Brahman bulls following a provocative endotoxin challenge. Specifically, Temperamental bulls may preferentially utilize an alternate energy source (i.e. NEFA) to a greater degree than do bulls of Calm and Intermediate temperaments. The use of circulating NEFA from lipolysis may reduce the negative metabolic consequences of an immune response by allowing for a prompt answer to increasing energy demands required during immunological challenge, compared with the time required for glycogenolysis and gluconeogenesis.  相似文献

Type 1 polysaccharide storage myopathy caused by genetic mutation in the glycogen synthase 1 gene is present in many breeds including the Noriker and Haflinger horses. In humans, EMG has already been used to document changes in the muscle activity patterns of patients affected by human glycogen storage disorders. Therefore, the aim of the present study was to describe gluteus muscle activity with surface electromyography (sEMG) in Haflinger and Noriker horses with known GYS1 mutation status during walk and trot. Thirty-two horses (11 Haflinger and 21 Noriker horses) with homozygous non-affected (GG), heterozygous affected (GA) and homozygous affected (AA) status of GYS1 mutation without overt clinical signs of any myopathy were selected for the current study. Using surface electromyography gluteus medius muscle activity at walk and at trot was measured, and muscle activity was described in relation to the maximum observed value at the same sensor and the same gait. In order to further describe the signals in detail comprising both frequencies and amplitudes, the crossings through the baseline and the 25, 50 and 75 percentile lines were determined. The result of the relative muscle activity did not show a consistent difference between affected and non-affected horses. Genetically affected (GA and AA) horses showed significantly less density of muscle activity for both gaits and horse breeds except for the crossings per second at the baseline and 75 percentile at walk in the Haflinger horses and 75 percentile at trot in the Noriker horses. The medians of all calculated density values were significantly lower in the GA Haflingers compared to the GG Haflingers (p = 0.012) and also in the AA Norikers compared to the GG Norikers (p = 0.011). Results indicate that the GYS1 mutation reduces the number of functional muscle fibres detected by sEMG measurements even in the absence of overt clinical signs.  相似文献

This study investigated the effects of amino acids (AA) supplementation in low‐crude protein (CP) diets on the growth performance and carcass characteristics in late‐finishing gilts. Ninety gilts (93.8 ± 5.5 kg) were randomly allotted to one of the five diets which consisted of a normal‐CP (137 g/kg) or four low‐CP (105 g/kg) diets for 28 days. The low‐CP diets were supplemented with lysine + threonine + methionine (LCM), LCM + tryptophan (LCT), LCT + valine (LCV) or LCV + isoleucine (LCI), respectively. Non‐significant difference in average daily gain (ADG) was obtained in gilts receiving the control and LCV diet, which was higher than that of gilts fed the LCM diet (p < .05). The additions of crystalline AA in the low‐CP diet resulted in the improvements in ADG (linear and quadratic effect, p < .05) and fat‐free lean gain (quadratic effect, p < .05) and influenced the valine concentration (linear and quadratic effect, p < .05) and proportion of saturated fatty acid (linear effect, p < .05) in longissimus muscle. The results indicated that the valine supplementation could further improve the performance in 94 to 118 kg gilts fed the 105 g/kg CP diet.  相似文献

Metamizole (MT) is an analgesic and antipyretic drug labelled for use in humans, horses, cattle, swine and dogs. MT is rapidly hydrolysed to the active primary metabolite 4‐methylaminoantipyrine (MAA). MAA is formed in much larger amounts compared with other minor metabolites. Among the other secondary metabolites, 4‐aminoantipyrine (AA) is also relatively active. The aim of this research was to evaluate the pharmacokinetic profiles of MAA and AA after dose of 25 mg/kg MT by intravenous (i.v.) and intramuscular (i.m.) routes in healthy horses. Six horses were randomly allocated to two equally sized treatment groups according to a 2 × 2 crossover study design. Blood was collected at predetermined times within 24 h, and plasma was analysed by a validated HPLC‐UV method. No behavioural changes or alterations in health parameters were observed in the i.v. or i.m. groups of animals during or after (up to 7 days) drug administration. Plasma concentrations of MAA after i.v. and i.m. administrations of MT were detectable from 5 min to 10 h in all the horses. Plasma concentrations of AA were detectable in the same range of time, but in smaller amounts. Maximum concentration (Cmax), time to maximum concentration (Tmax) and AUMC0‐last of MAA were statistically different between the i.v. and i.m. groups. The AUCIM/AUCIV ratio of MAA was 1.06. In contrast, AUC0‐last of AA was statistically different between the groups (< 0.05) with an AUCIM/AUCIV ratio of 0.54. This study suggested that the differences in the MAA and AA plasma concentrations found after i.m. and i.v. administrations of MT might have minor consequences on the pharmacodynamics of the drug.  相似文献

ObjectiveTo assess anesthetic induction, recovery quality and cardiopulmonary variables after intramuscular (IM) injection of three drug combinations for immobilization of horses.Study designRandomized, blinded, three-way crossover prospective design.AnimalsA total of eight healthy adult horses weighing 470–575 kg.MethodsHorses were administered three treatments IM separated by ≥1 week. Combinations were tiletamine–zolazepam (1.2 mg kg−1), ketamine (1 mg kg−1) and detomidine (0.04 mg kg−1) (treatment TKD); ketamine (3 mg kg−1) and detomidine (0.04 mg kg−1) (treatment KD); and tiletamine–zolazepam (2.4 mg kg−1) and detomidine (0.04 mg kg−1) (treatment TD). Parametric data were analyzed using mixed model linear regression. Nonparametric data were compared using Skillings–Mack test. A p value <0.05 was considered statistically significant.ResultsAll horses in treatment TD became recumbent. In treatments KD and TKD, one horse remained standing. PaO2 15 minutes after recumbency was significantly lower in treatments TD (p < 0.0005) and TKD (p = 0.001) than in treatment KD. Times to first movement (25 ± 15 minutes) and sternal recumbency (55 ± 11 minutes) in treatment KD were faster than in treatments TD (57 ± 17 and 76 ± 19 minutes; p < 0.0005, p = 0.001) and TKD (45 ± 18 and 73 ± 31 minutes; p = 0.005, p = 0.021). There were no differences in induction quality, muscle relaxation score, number of attempts to stand or recovery quality.Conclusions and clinical relevanceIn domestic horses, IM injections of tiletamine–zolazepam–detomidine resulted in more reliable recumbency with a longer duration when compared with ketamine–detomidine and tiletamine–zolazepam–ketamine–detomidine. Amino acid (AA) composition of body protein is considered constant although there are evidences that AA pattern in pigs may be altered by different factors. Pigs with different body composition and protein deposition rates—like fatty and lean pigs—may differ in AA composition, with possible consequences on their AA requirements. This work investigates effects of genotype and dietary lysine deficiency on AA composition of carcass and muscles of Iberian and Landrace × Large White pigs. Twenty‐eight barrows (10 kg body weight [BW]), 14 from each breed, were used. They were randomly assigned to two experimental diets according to a factorial arrangement (two breeds × two diets). Diets were isonitrogenous and isoenergetic (200 ± 1 g CP/kg dry matter (DM); 14.7 ± 0.1 MJ ME/kg DM) and with identical chemical composition except for lysine concentration (10.9 and 5.20 g lysine/kg DM, for lysine‐adequate (AL) diet and lysine‐deficient (DL) diet respectively). Pigs were individually housed, and daily feed allowance was adjusted on a weekly basis according to BW. Pigs were slaughtered at 25 kg BW. Isoleucine, valine and phenylalanine concentration were higher in carcass protein of Iberian pigs (p < .01). In longissimus muscle, higher concentration of arginine, isoleucine, phenylalanine, lysine and valine (p < .001–p < .05), and lower of methionine (p < .001) were detected in Iberian pigs, whereas phenylalanine, leucine, lysine, threonine and methionine concentration decreased and arginine increased (p < .001–p < .05) when pigs were fed DL diet. Genotype and lysine deficiency effects were moderate in the AA composition of protein of biceps femoris muscle. The results show that AA proportions in protein of carcass and longissimus muscle can be influenced by pig genotype and conditions of lysine shortage. The biceps femoris muscle, with different functional and metabolic properties, shows more constant AA composition than longissimus, which seem to prevail independent from genotype or nutritional challenges.  相似文献

A total of 71 522 records (from 3154 horses) with the times per kilometre (TPK), recorded in Spanish Trotter horses (individual races) from racing performances held from 1991 to 2007, were available for this study. The TPK values for the different age groups (young and adult horses) and different distances (1600–2700 m) were considered as different traits, and a bi character random regression model (RRM) was applied to estimate the (co)variance components throughout the trajectory of age groups and distances. The following effects were considered as fixed: the combination of hippodrome‐date of race (404 levels); sex of the animals (3 levels); type of start (2 levels) and a fixed regression of Legendre polynomials (order 2). Those considered as random effects were the random regression Legendre polynomial (order 1) for animals (9201 animals in the pedigree); the individual environment permanent (3154 animals with data) and the driver (n = 957 levels). The residual variance was considered as heterogeneous with two classes (ages). The heritability estimated by distance ranged from 0.12 to 0.34, with a different trajectory for the two age groups. Within each age group, the genetic correlations between adjacent distances were high (>0.90), but decreased when the differences between them were over 400 metres for both age groups. The genetic correlations for the same distance across the age groups ranged from 0.47 to 0.78. Accordingly, the analysed trait (TPK) can be considered as positive genetic correlated but as different traits along the trajectory of distance and age. Therefore, some re‐ranking should be expected in the breeding value of the horses at different characteristics of the racing. The use of RRM is recommended because it allows us to estimate the breeding value along the whole trajectory of race competition.  相似文献

Objectives – To evaluate the use of citrated recalcified (nonactivated) thromboelastography (TEG) in healthy horses and horses with colitis and suspected coagulopathies. Design – Prospective, observational study conducted between October 2007 and June 2009. Setting – Veterinary Teaching Hospital. Animals – Forty‐five healthy adult horses and 12 sick adult horses with colitis and prolonged prothrombin time (PT) or activated partial thromboplastin time (aPTT). Interventions – None. Measurements and Main Results – Whole blood was collected on admission. Coagulation profile (PT, aPTT, platelet count, and fibrinogen concentration) and citrated recalcified whole blood TEG analysis (R‐time [R], K‐time [K], angle [α], maximum amplitude [MA], G value [G], lysis at 60 min [LY60]) were evaluated. Mean values (SD) for TEG parameters in healthy horses were: R=10.4 (3.1) minutes; K=3.5 (1.2) minutes; α=46.3 (11.0)°; MA=55.6 (5.1) mm; G=6,429 (1,341) dyn/cm2, and LY60=5.1 (2.4)%. Mean coefficients of variation for intra‐assay/interindividual variability in healthy horses were: R=4.7%/30.7%, K=4.8%/35.3%, α=4.4%/23.8%, MA=1.4%/9.3%, G=3.4%/20.8%, and LY60=13.1%/47.7%, respectively. Horses with colitis and prolonged PT and/or aPTT had longer mean values for R (P<0.001) and K (P<0.001), narrower mean α (P<0.001), decreased mean MA (P=0.001), and smaller mean G (P=0.02); changes consistent with hypocoagulability. Conclusions – Citrated recalcified (nonactivated) TEG demonstrated changes consistent with hypocoagulability in horses with colitis that had preidentified coagulation abnormalities. This technique has high interindividual variability and low intra‐assay variability. TEG may be useful for detecting hypocoagulable states in horses with colitis and suspected coagulopathies.  相似文献

Plasma concentrations of adiponectin and leptin were measured in 23 mature Standardbred mares (age: 10 ± 3 years) and 12 weanling fillies (10 Quarter Horse/Belgian crossbreds and two Quarter Horses; aged: 4 ± 3 months) to test the hypothesis that adipocytokines are proportional to adiposity in horses. Rump fat thickness was measured using B-mode ultrasound and percent body fat (% fat) calculated using a published formula for the estimation of fatness in horses. Plasma adiponectin and leptin were determined using radioimmunoassay. In the absence of purified equine adiponectin or leptin, results were expressed as human equivalents (HE) of immunoreactive (ir) adipocytokines.Plasma ir-leptin HE concentration was positively correlated (r = 0.543; P < 0.001) with percent body fat and (r = 0.556; P < 0.001) to fat mass in all horses. The plasma ir-leptin HE concentration was lower (P = 0.03) in weanlings (1.90 ± 0.34 ng/mL HE) than in mature mares (3.47 ± 0.50 ng/mL HE). The ratio of ir-adiponectin HE to ir-leptin HE was negatively correlated (r = −0.621; P < 0.001) to percent fat and (r = −631; P < 0.001) to fat mass in all horses. The ratio of ir-adiponectin HE to ir-leptin HE was higher (P < 0.001) in weanlings (3.04 ± 0.51) than mature mares (1.03 ± 0.13). These data suggest that leptin is proportional while adiponectin is inversely proportional to adiposity in horses.  相似文献

This study investigated the effect of L ‐theanine on carcass traits, meat quality, muscle antioxidant capacity, and amino acid (AA) profiles of broilers. Three hundred 1‐day‐old Ross 308 male broilers were randomly allotted to five groups with six replicates. Birds were fed the basal diet or basal diet with 300, 600, 900, or 1,500 mg/kg L ‐theanine for 42 consecutive days. The results showed that L ‐theanine quadratically increased dressing percentage, eviscerated percentage, and leg muscle yield (p < .05). Meanwhile, drip loss, cooking loss, shear force, L*24h, and muscle lactate content decreased quadratically in response to dietary L ‐theanine supplementation (p < .05), while pH24h and muscle glycogen content were quadratically improved by L ‐theanine (p < .05). Notably, the contents of muscle malondialdehyde and protein carbonyl, and the activities of muscle total antioxidant capacity, catalase, and glutathione peroxidase decreased quadratically in response to dietary L ‐theanine supplementation (p < .05), suggesting that the oxidative stress level of muscle was decreased quadratically. Moreover, L ‐theanine quadratically increased the concentrations of most of muscle essential AA, nonessential AA, and flavor AA (p < .05). In conclusion, L ‐theanine can be used as a valuable feed additive to modulate carcass traits, meat quality, muscle antioxidant status, and AA profiles of boilers, and its optimum addition level is 600 mg/kg based on the present study.  相似文献

In adult horses, pronounced sex differences in behavior exist, and many riders prefer male horses to mares. In the present study, we tested the hypothesis that sex differences in the response of horses to handling emerge before puberty, that is, in sexually immature animals. Warm-blooded yearling horses (6 males and 9 females) were exposed to a stationary human test (exposure to an unfamiliar person for 5 minutes) and a tolerance test (haltering and direct contact with the horse) on 5 consecutive days. The horses' behavior and heart rate were recorded. Male yearling horses displayed significantly less exploratory behavior than females during the stationary human test on days 1 and 2 (P < 0.05). Latency to halter and ease with which horses could be haltered were significantly affected by day (P < 0.05), and a significant interaction of day and sex was detected (P < 0.05; e.g., time for haltering: females, day 1: 23.8 ± 7.8 seconds [±standard error of the mean] and day 5: 21.1 ± 1.7 seconds; males, day 1: 53.6 ± 10.4 seconds and day 5: 19.4 ± 2.2 seconds). Heart rate of the horses in response to the test situations was significantly affected by day (P < 0.01) but not by sex. In conclusion, sex differences in behavior do not only exist in adult horses but are already present in young horses before puberty. When exposed to unfamiliar humans for the first time, male yearling horses display more caution than females.  相似文献

Fat‐tailed sheep breeds can tolerate periods of negative energy balance without suffering from elevated concentration of plasma non‐esterified fatty acid (NEFA). This ability was attributed to unique metabolism of fat‐tailed adipose depot, whereas role of liver as an influential organ in fatty acid metabolism was not evaluated yet. Hence, current study was conducted to evaluate the effects of negative and positive energy balances on liver expression of genes related to fatty acid metabolism in fat‐tailed and thin‐tailed lambs. Lambs experienced negative (21 days) and positive (21 days) energy balances and were slaughtered at the beginning and end of negative energy balance and at the end of positive energy balance. Real‐time quantitative polymerase chain reaction (RT‐Q‐PCR) was conducted to evaluate changes in gene expression. Expression of diglyceride acyltransferase 1 (DGAT1), 3‐hydroxy‐3‐methylglutaryl‐CoA synthase 2 (HMGCS2) and apolipoprotein B (APOB) was not affected by genotype, energy balance and their interaction. Expression of carnitine palmitoyltransferase 1 (CPT1) was significantly higher in liver of fat‐tailed comparing to thin‐tailed lambs regardless of energy balance (p < 0.02). Catalase mRNA abundance was increased in response to negative energy balance (p < 0.02), and severity of this enhancement was higher in fat‐tailed lambs (p < 0.06). Expression of CPT1 was positively correlated with expression of HMGCS2 in both fat‐tailed (p < 0.05) and thin‐tailed lambs (p < 0.002); however, the correlation was weaker in fat‐tailed lambs (0.72 vs. 0.57, respectively, for thin‐tailed and fat‐tailed lambs). There was a positive correlation between DGAT1 and APOB genes expression in fat‐tailed lambs (0.94; p < 0.001), whereas this correlation was not observed in thin‐tailed lambs. Results demonstrate that liver of fat‐tailed lambs has higher capacity for metabolism of mobilized NEFA exposed to liver during negative energy balance.  相似文献

The objective of this study was to examine markers of whole‐body and muscle protein metabolism in aged horses fed a diet typical for North American aged horses, supplemented with amino acids. In a replicated Latin square design, six aged horses (20 ± 1.1 years) were studied while receiving each of three isocaloric, isonitrogenous diets, a control treatment concentrate (CON; 100 mg/kg?1 BW day?1 lysine, 84 mg kg?1 day?1 threonine, 51 mg kg?1 day?1 methionine), LYS/THR (134 mg kg?1 BW day?1 lysine, 110 mg kg?1 BW day?1 threonine, 52 mg kg?1 BW day?1 methionine) and LYS/THR/MET (132 mg kg?1 BW day?1 lysine, 112 mg kg?1 BW day?1 threonine, 62 mg kg?1 BW day?1 methionine). In each 15‐days period, urine and faeces were collected for assessment of nitrogen balance. Blood samples were collected before and after feeding for analysis of plasma urea nitrogen (PUN), glucose, insulin and plasma amino acid concentrations. Skeletal muscle samples were collected for measurement of proteins associated with muscle protein synthesis and degradation, and horses underwent stable isotope infusion procedures for comparison of differences in whole‐body rates of protein synthesis and degradation. There was no effect of treatment on relative abundance of proteins involved in protein synthesis, nitrogen retention or phenylalanine kinetics. PUN concentrations tended to be higher for LYS/THR (p = 0.054) and were higher for LYS/THR/MET (p = 0.0056) than for CON. Atrogin‐1 abundance tended to be higher in the post‐absorptive state for the CON treatment (p = 0.07), indicating that amino acid supplementation resulted in less muscle protein degradation when horses were in the post‐absorptive state. However, lack of differences in nitrogen retention and phenylalanine kinetics indicated that whole‐body protein metabolism was not improved, and higher PUN concentrations in the supplemented diets suggest that the supplemented amino acids may have been catabolized. Amino acid availability was not limiting protein synthesis in the sedentary aged horses in this study when fed the CON diet.  相似文献
