All commercial citrus rootstocks are polyembryonic and propagated by seeds. Although these seeds produce uniform plant material most of the time, zygotic or polyploid citrus seedlings may arise. The aim of this study was to understand how zygotic or tetraploid rootstock could affect a citrus rootstock selection field trial. A trifoliate orange selection field trial, which was planted in 1974 and grafted with clementine, was re-investigated with respect to the presence of rootstocks that were not true-to-type. Among the 288 trees investigated, flow cytometry identified 2.4% of rootstocks as tetraploid and SSR markers indicated that 6.6% were zygotic. Yield data showed that the presence of tetraploid rootstock dramatically decreased (by about 45%) clementine fruit production. However, zygosity did not always affect fruit production and a range of effects were observed, from a slight increase in production to a 24% decrease. Exclusion of non-true-to-type genotypes from the production analysis indicated that the best candidate for rootstock was a clone previously ranked in the middle of the 32 rootstocks under evaluation. However, the presence of zygotic rootstock did not appear to cause any significant differences in fruit quality during the first 5 years of the investigation, which suggests that non-true-to-type plants cannot be identified by fruit quality parameters. This study indicates that tetraploid and zygotic rootstocks have a strong impact on citrus fruit production in orchards, and removal of off-type seedlings is required prior to planting in any agronomic trial.  相似文献

Summary

Increasing losses of trees to diseases, expansion of plantings into more marginal production areas and the need to control tree size to reduce harvesting costs have elevated the demand for new improved citrus rootstocks in Florida. A major strategy of the CREC rootstock improvement programme has been to use protoplast fusion to produce allotetraploid somatic hybrids that combine complementary rootstock germplasm. Tetraploid citrus rootstocks have been shown to have a built-in tree size control component due to polyploidy. This report focuses on the incorporation of newly identified superior rootstock germplasm into the somatic hybridization programme. Poncirus trifoliata 50–7, a selection of trifoliate orange selected for superior resistance to Phytophthora nicotianae, was hybridized with sour orange, Changsha mandarin, Navel orange, and a seedy white ‘Duncan’ type grapefruit. High-yielding Benton citrange (Citrus sinensis 3 Poncirus trifoliata) was hybridized with Changsha mandarin and sour orange. More than 200 plants of each of these six new somatic hybrids were propagated via tissue culture and rooted cuttings. To assess their horticultural performance, all the somatic hybrids were budded with commercially important scions and planted in replicated trials representing the two most important soil types in Florida. Seed trees of each somatic hybrid have also been planted in the field to determine fruiting potential and level of polyembryony.  相似文献

Summary

Simple Sequence Repeat (SSR) and Sequence-Related Amplified Polymorphism (SRAP) molecular marker systems were used to analyse four commercially important pistachio rootstocks: two species of Pistacia atlantica (cv. ‘Standard Atlantica’), P. integerrima (cv. ‘Pioneer Gold’) and two interspecific hybrids of the same, ‘Pioneer Gold II’ (‘PGII’) and ‘University of California at Berkeley 1’ (‘UCB-1’). A total of 35 putative alleles were detected by 12 SSR primer pairs with an average of 2.9 alleles per locus. The number of putative alleles ranged from 2 to 5 in the pistachio rootstocks tested. The number of bands produced by the SRAP protocol was highly variable, ranging from 11 to 38, with an average of 25.2 per primer combination. Eight primer combinations resulted in 104 (51%) polymorphic markers in these samples. SSR and SRAP markers successfully identified all pistachio rootstocks tested from their unique fingerprints. Both SSR and SRAP molecular markers confirmed that the observed variation in ‘UCB-1’ rootstock is genetic.Thus, there will always be variation among ‘UCB-1’ hybrid seedling progeny due to the segregation of alleles when propagated by seed.We also found evidence of contaminating pollen other than from P. integerrima in some hybrid ‘UCB-1’ rootstock progeny produced by closed pollination. Only alleles from the cultivar ‘Standard Atlantica’ were observed in abnormal ‘UCB-1’ rootstock in the nursery. We found that the poor performance of the scion cv. ‘Kerman’ on ‘UCB-1’ rootstock was not due to ‘UCB-1' rootstocks displaying abnormal behaviour in the nursery. We have successfully developed two efficient marker systems for genome analyses in pistachio, which can be used for identification and management in pistachio rootstock production.  相似文献

The study aimed to understand the root morphological character of 1 year old seedling rootstock of citrus genotypes viz., rough lemon, Karna Khatta, Carrizo citrange, Rangpur lime, Troyer citrange, Jatti Khatti and sour orange as well as to investigate the effect of these rootstocks on the leaf mineral composition of Kinnow. Root morphological traits showed significant genotypic variation. Rough lemon seedlings had a higher root length, projected area, surface area, volume and the number of forks. Sour orange was moderate rootstock of the 7 genotypes for most of the root morphological characters, while citrange rootstocks expressed poor root morphology. The rootstocks also imparted significant influence on the leaf mineral composition. Leaf K, Ca and Mg content was recorded maximum in plants grown on rough lemon seedling, while N, P, Fe, Zn, Mn and Cu concentration was higher on sour orange rootstock. Kinnow trees on Rangpur lime rootstock accumulated more Na in their leaf tissue, where as Troyer citrange restricted the accumulation of Na. Considering the root morphological characters and nutrient accumulation pattern, rough lemon and sour orange for areas free from tristeza virus could be suggested as alternative rootstocks for the studied Kinnow mandarin under similar ecological conditions.  相似文献

Summary

Six wild species of Eriobotrya were evaluated as rootstocks for domesticated loquat. Twelve-month-old seedlings of Oak leaf loquat (an Oak Leaf loquat ‘Zaozhong No. 6’ hybrid) and wild Fragrant loquat (E. fragrans Champ) had higher root-to-shoot fresh weight (FW) ratios than other wild species. When the domesticated loquat, ‘Zaozhong No. 6’, was grafted onto seedlings of six different wild loquat species, differences in compatibility were observed. Graft compatibility was lower for all combinations compared to ‘Zaozhong No. 6’ grafted onto domesticated loquat seedlings. When wild Henry loquat was used as a rootstock, delayed incompatibility was observed. Differences in soluble sugar contents (SSC) between scion and rootstock were observed, and there was a significant negative correlation between graft compatibility and the difference in SSC values. The influence of the rootstock on anatomical changes at the graft union were observed using a scanning electron microscope (SEM). Wild rootstocks also influenced scion growth, fruit bearing, and fruit quality. Graft combinations which used wild Fragrant loquat as the rootstock had increased cold tolerance and improved scion growth. Wild Fragrant loquat therefore appears to have value as a rootstock for domesticated loquat.  相似文献

Summary

Micropropagation of Darjeeling orange (Darjeeling mandarin) was done by shoot-tip grafting on the local rootstocks rough lemon, sour orange and Rangpur lime. Seedlings of these plants were raised in MS medium in standard conditions of temperature, light duration and plant age. Microbuds (0.2 mm long), aseptically excised from the scion, were grafted onto the decapitated rootstock seedlings. The micrografts in culture medium were grown under standard lighting. Rootstock seedlings showed optimum growth at 25–30°C. Optimum growth of the micrografts resulted from treatment in complete darkness for 4 d after grafting followed by exposure to 750 W for 16 h d?1 for 15 d and 1500–2000 W for 16 h d?1 for 39 d. The success of micrografting depended on the age of the rootstock seedling which was 21 d, 14 d and 14 d for sour orange, rough lemon and Rangpur lime respectively. The micrografts were then double grafted to the two year old rootstocks seedlings of the respective species and the age of the micrografts for the successful double grafts was also standardized. The survival rate was high with 40 d, 45 d and 45 d micrografts on sour orange, rough lemon and Rangpur lime respectively. The double-grafts were then indexed for tristeza virus and greening disease after six months’ growth in an insect-proof screenhouse and found to be free from these graft transmissible diseases.  相似文献

Sting nematode (Belonolaimus longicaudatus Rau) has become a primary factor limiting citrus production in localized regions of the central Florida sandridge citrus production area, making the development of resistant rootstocks a new breeding objective. Osbeck) parental combinations by fusing embryogenic suspension culture-derived protoplasts isolated from selected mandarins with leaf protoplasts of pummelo seedlings previously selected for tolerance/resistance to the sting nematode (B. longicaudatus Rau) as follows: Amblycarpa mandarin + ‘Liang Ping Yau’ (seedling) pummelo seedling SN7; Amblycarpa mandarin + ‘Hirado Buntan Pink’ (HBP) pummelo seedling SN3; Murcott tangor + pummelo seedling SN3; and Shekwasha mandarin + pummelo seedling SN3. Somatic hybridization was verified by ploidy analysis (via flow cytometry) and RAPD analyses. Mandarin parents were selected for wide soil-adaptation and ability to produce friable embryogenic callus lines. Pummelo seedlings used as leaf parents were identified from a previous screen of large seed populations (200 each) from four pummelos for resistance to sting nematode as follows: ‘Hirado Buntan Pink‘; ‘Red Shaddock‘; ‘Large Pink Pummelo’ and a seedling pummelo of ‘Liang Ping Yau‘. Ten resistant/tolerant pummelo seedlings were selected from the 800 pummelo seeds planted in the screen for further study. The four new somatic hybrids have been propagated to evaluate their horticultural performance and resistance to the sting nematode. 分别以黑籽南瓜及酸黄瓜-黄瓜抗线虫渐渗系10-1为砧木,‘南水2号’黄瓜为接穗,通过LI-6400XT、微波消解ICP-AES对嫁接植株及自根苗的光合特性及矿质元素含量进行了对比分析。光合参数结果表明,黄瓜嫁接材料与自根苗材料的气孔导度、净光合速率、蒸腾速率以及胞间CO2浓度均不存在显著性差异(P≤0.05)。果实中9种矿质元素含量显示,元素Mn、Fe、P和K的含量在3种材料之间没有明显差异;而以黑籽南瓜为砧木的嫁接黄瓜果实相比自根苗果实Cu、Se元素含量升高,Mg元素含量降低;以抗线虫渐渗系10-1为砧木的黄瓜果实B元素含量升高,其他元素含量与自根苗无异。上述研究显示盆栽条件下,嫁接对黄瓜光合特性无显著影响。同时,不同砧木嫁接黄瓜果实中的矿质元素含量存在差异,证实砧木根系对土壤中矿质元素的选择性吸收影响了果实中的矿质元素含量。这为今后深入研究嫁接砧木根系影响黄瓜果实品质等相关机制奠定了基础。  相似文献

The possibility of applying grafting to improve fruit quality has been scarcely investigated. Different shoot tomato (Solanum lycopersicum) genotypes were grafted onto distinctly-different tomato rootstocks and the effect of the rootstock on two important fruit quality parameters, soluble solids content (SSC) and titratable acidity (TA), was studied under both non-saline and saline conditions. Increased SSC and TA in fruits from grafted plants grown under saline conditions were observed on different grafting combinations. One of these rootstocks (cv. Radja) was able to induce increases in both fruit yield and fruit quality traits of the scion. When recombinant inbred lines (RILs) from the crossing of the cultivated tomato and wild tomato (Solanum cheesmaniae) were tested as rootstocks, using the commercial hybrid ‘Boludo’ as scion, the rootstock also improved SSC and TA when grafted plants were grown under non-saline conditions. On the whole, these results show the effectiveness of grafting with respect to upgrading of fruit quality in tomato, which is of great importance as grafting is a rapid and efficient alternative to achieve this goal.  相似文献

Seeds were collected from a range of citrus rootstock types in China and sent to Australia for evaluation. Before evaluation proceeded, however, the seeds and their seedlings were characterised for uniformity and trueness-to-type. The seeds were cultured in vitro and/or germinated in sand beds to assess whether or not they were mono- or polyembryonic. Seedlings were then rogued on the basis of vigour and morphology before isozyme profiles were developed from leaf extracts to assess seedling uniformity within introductions. Isozyme profiles were also compared to those of the source trees, as well as hybrids generated between some of them in China, to assess and verify trueness-to-type. The degree of variability between the seedlings propagated from the imported seeds and their source trees in China differed between accessions, which highlighted some of the issues that need to be considered when importing citrus germplasm as seeds. Regardless of whether the seedlings could be shown to be identical to their source tree in China, three uniform seedlings of polyembryonic types were established in arboreta as Australian source trees to provide propagules for evaluation and possibly commercial use.  相似文献

Summary

Melon (Cucumis melo L.) ‘Arava’ was grafted onto 22 experimental Cucurbita rootstocks including intra- or interspecific hybrids to examine scion-rootstock compatibility and to correlate plant performance under greenhouse and field conditions. The commercial Cucurbita ‘TZ-148’ rootstock and non-grafted plants were used as controls. Numbers of leaves, stem length and plant fresh weights were recorded three weeks after planting. Vegetative growth of the grafted plants varied markedly, indicating differences in compatibility between the scion and rootstocks. A positive correlation was found between number of leaves, stem length and fresh weight of melon plants. No significant advantage in vegetative growth was found in melons grafted onto intra- or interspecific hybrid rootstocks. Stem diameter and number of vascular bundles of the rootstock did not correlate with scion plant fresh weight. Twelve selected rootstocks out of the 22 tested were grown in the field during the spring and autumn, to determine if there is an association between the plant performance in the greenhouse and fruit yield under field conditions. Positive and significant correlations were found between greenhouse growth parameters and fruit yield under field conditions. It is suggested that testing grafted melons under greenhouse conditions could be a useful tool for preliminary testing of scion-rootstock compatibility.  相似文献

Summary

Heirloom tomatoes (Solanum lycopersicum L.) such as ‘Cuore di Bue’ are highly appreciated by consumers for their outstanding quality and flavour. Nowadays, they are often grafted onto vigorous rootstocks in order to overcome several soil-borne diseases. The present study was conducted in 2007 and 2008 in Southern Italy. Plants of ‘Cuore di Bue’, an heirloom ‘oxheart’ tomato, were grown in a greenhouse either as non-grafted plants, or grafted onto two inter-specific (S. lycopersicum S. habrochaites) rootstocks (‘Beaufort F1’ and ‘Maxifort F1’) in order to evaluate their effects on vegetative growth, marketable yield, fruit quality, and the sensory properties of ‘Cuore di Bue’ tomato fruit. Growth analysis revealed that ‘Maxifort F1’ enhanced plant growth, particularly in terms of leaf area and leaf fresh weight. Grafted plants had higher leaf area ratios (by 13%) and higher leaf dry weight fractions (by 18%) compared to non-grafted ‘Cuore di Bue’ plants. Marketable yields increased by 20 – 25% in grafted plants in both years. However, total soluble solids (TSS) contents, titratable acidity (TA), and TSS/TA ratios were not significantly affected by grafting.Vitamin C contents decreased by 14 – 20% in both years in the fruit of plants grafted onto either rootstock. The sensory profiles of fruit were not modified by grafting, although taste panelists expressed a higher preference for purchasing fruit from plants grafted onto ‘Maxifort F1’. These results confirm that, when using the appropriate rootstock/scion combination, grafting can improve plant growth and the marketable yield of heirloom tomato fruit without reducing the sensory quality of the fruit or its biochemical parameters. However, grafting can reduce vitamin C contents and, thus, nutritional quality.  相似文献

Observations of respiration rate, catalase activity, peroxidase activity ("physiological activity”) and some growth characteristics of roots of citrus rootstocks were made. These were carried out on seedlings of six rootstocks (five months old) grown in a standard nutrient solution and on seven- to ten-year-old budded trees growing in the orchard. The orchard trees, budded to only one scion, were propagated from the same rootstocks as were used in the study of seedlings. A comparison was also made of five additional scions budded on one rootstock.

A technique was developed to obtain roots from orchard trees for the study of their physiological and morphological characteristics.

Significant differences were found in the “physiological activity” of the roots of the various rootstock seedlings. A significant negative correlation existed between the final length of the main root and the physiological activity of the roots.

Significant differences were found in the physiological activity of the roots of the various mature rootstocks budded with Shamouti sweet orange.

A clear and marked influence of the various scions on the physiological activity of the roots of sour-orange was observed.

An obvious positive correlation existed between respiration rate, catalase activity and peroxidase activity of the roots. The use of two different H-donors resulted in differences in the peroxidase activity measured.

The significance of the variations in physiological response of the roots is discussed.  相似文献

Water stress is one of the limiting factors for citrus production and Citrus species show great variation in their response to drought stress. Although the majority of Citrus rootstocks are sensitive to water stress, Rangpur lime (Citrus limon (L) Osbeck) (RL) shows a high degree of drought tolerance. Therefore, it has been used as a rootstock in drought-prone environments, but mechanisms of drought tolerance are not yet known. In this study, to explore the mechanisms of drought adaptation and tolerance, a subtractive cDNA library was constructed from the leaves of 14-day drought-stressed and non-stressed RL for identification of drought-induced genes. 285 cDNA sequences were obtained from randomly selected clones from the subtracted library containing the drought-induced genes. The expression analyses of 200 cDNAs in 14-day drought treated and untreated RL by macroarray hybridisation revealed that the expression of 56 cDNAs increased two to 11-fold. 30 non-redundant drought-induced genes were identified from these cDNAs and drought induction of eight selected genes was confirmed by a real-time RT-PCR assay suggesting that expressions of these genes were regulated by drought-stress. Genes identified in this study were mostly related with cell rescue and defense pathways involved in drought adaptation and tolerance of RL.  相似文献

Summary

We have evaluated the influence of four different peach rootstocks (Prunus pumila,‘GF 655/2’ ,‘Missour’ and ‘GF 677’ as a standard) planted in medium-heavy to heavy soil on phenological events (flowering and harvest date), growth, yield, fruit quality and mortality of peach (P. persica L.) scions of cv. ‘Redhaven’. At the end of flowering, there were no significant differences between the different rootstocks in the years 1999, 2000, 2003 and 2004. In 2001, trees grafted on P. pumila reached the end of flowering 2 d before trees grafted on the other rootstocks. ‘Redhaven’ fruits ripened on the same day on trees grafted on the different rootstocks. The differences were only between years. The results showed that significantly less vigorous trees, according to their trunk cross-sectional area (TCSA) and canopy volume, with the lowest yield, were observed on P. pumila rootstock. The rootstock P. pumila had a significant negative influence on fruit dimensions and mortality was 50%. Trees grafted on ‘GF 655/2’ rootstock produced significantly lower yields than those grafted on ‘GF 677’ or ‘Missour’ rootstocks. Only the ‘GF 655/2’ rootstock had root suckers. Trees grafted on ‘GF 677’ rootstock were significantly more vigorous than on the other rootstocks, and produced the highest yield with good fruit quality. The final canopy volumes of peach trees grafted on the different rootstocks were very different. The most vigorous trees were those grafted on ‘GF 677’ rootstock, where the final tree volume was 12.8 m3. Trees grafted on P. pumila and ‘GF 655/2’ had significantly lower TCSA values than on the other rootstocks.  相似文献

为了筛选适合宁波地区秋季黄瓜嫁接耐热砧木品种,利用甬砧2号、0615、0616、1101和T1325个南瓜材料与当地主栽黄瓜品种津优1号进行秋季嫁接栽培试验,考察其嫁接亲和性、嫁接苗生长势、对果实品质和产量的影响。结果表明,T132、甬砧2号和1101综合表现较好,产量增加16.7%以上,对嫁接黄瓜品质无明显影响,且嫁接后不产生蜡粉,黄瓜外观更佳,适合作为秋季黄瓜嫁接砧木。  相似文献

Pistachio cultivation requires the use of rootstock because grafting is the only form of vegetative propagation. The main commercial rootstocks are Pistacia integerrima L., Pistacia atlantica Desf., Pistacia terebinthus L. and Pistacia vera L. Pistachio is considered to be a drought and saline-resistant crop; however, there is little information describing varietal responses of rootstocks to water stress. Some studies have suggested that P. terebinthus L. is the most drought and cold resistant rootstock. The effect of the rootstock on the water relations of the grafted plant is crucial for improving crop performance under water stress conditions and for developing the best irrigation strategy. This work studied the physiological response to water stress of pistachio plants (P. vera L. cv. Kerman) grafted onto three different rootstocks P. terebinthus L., P. atlantica Desf. and a hybrid from crossbreeding P. atlantica Desf. × P. vera L. Plant physiological responses were evaluated during a cycle of drought and subsequent recovery in potted plants. Parameters measured were soil moisture, trunk diameter, leaf area, leaf number, leaf and stem dry weight, stem water potential, leaf stomatal conductance. The results showed different responses of cv. Kerman depending on the rootstock onto which it had been grafted. The hybrid rootstock was associated with a higher degree of stomatal control and reduced leaf senescence compared to P. atlantica and P. terebinthus, despite being associated with the most vigorous shoot growth. P. terebinthus enabled very effective stomatal control but was also associated with the most rapid leaf senescence. P. atlantica was associated with less vigorous shoot growth and similar levels of water stress as occurred with the others rootstocks under conditions of high evaporative demand, which was associated with lower stomatal control. The selection of the most effective rootstock choice for different environmental conditions is discussed.  相似文献
