极早熟梨新品系—115—3—1

115－3－1梨是湖北省农科院果茶所用伏梨和金水酥梨杂交培育而成。果实近圆形，平均单果重230g，含水溶性固形物11．5％－13．2％。在武汉地区7月初成熟。现已在南方地区推广近200hm^2。     

芜菁Br14-3-3的克隆及其在非生物胁迫下的表达分析

克隆了‘津田芜菁’（Brassica rapa subsp. rapifera‘Tsuda’）通用调节因子14-3-3基因cDNA序列,命名为Br14-3-3（GenBank登录号为MK896872）。该基因全长1 113 bp,开放阅读框全长774 bp,编码含有257个氨基酸的多肽。荧光定量PCR分析Br14-3-3在‘津田芜菁’不同组织中及其在温度、脱水、渗透、ABA和无机盐等非生物胁迫下幼苗中的表达,结果表明该基因在‘津田芜菁’的花中表达量最高,幼苗中次之;低温抑制了Br14-3-3的表达,其他非生物胁迫可诱导该基因表达,暗示Br14-3-3在非生物胁迫应答中发挥功能。     

龙眼14-3-3基因的克隆与原核表达

 以龙眼(Dimocarpus longan Lour. ) 为试材, 应用同源克隆和RACE方法从花芽中获得了调控 蛋白14-3-3的全长cDNA序列, GenBank登录号为FJ479618 (GI: 218202931) 。该cDNA全长1 121 bp, 包括一个783 bp的开放阅读框, 编码261个氨基酸, 序列比较分析显示14-3-3 cDNA具有较高的保守性。半定量RT-PCR分析结果表明, 14-3-3 mRNA在龙眼叶芽、叶片、花芽和成熟的花中都有表达, 但在花芽中表达量最大。构建了pET14-3-3原核表达系统, 将14-3-3全长cDNA在大肠杆菌中表达, 获得一个分子量约为34 kD的可溶性融合蛋白, 经Western blotting验证, 该蛋白为14-3-3蛋白。     

丰蜜3号厚皮甜瓜的选育

丰蜜3号厚皮甜瓜是以自交系M3为母本、自交系0063为父本配制而成的短椭圆形厚皮甜瓜一代杂种。中早熟,果实发育期30-35d。单果重1.4kg左右,每667m2产量约2200kg。皮色浅黄,覆10条浅绿色纵沟,果面具细网纹;果肉白色,肉厚约3.5cm,肉质结实稍脆,味清香,果实中心可溶性固形物含量17%左右。     

14-3-3σ regulates p27 and inhibits Rat1-Akt cell proliferation

AIM: The present study was designed to investigate the effect of adenovirus-mediated 14-3-3σ on the proliferation of Rat1-Akt cells and to explore whether this effect is completed regulating p27.METHODS: The effect of Ad-14-3-3σ gene transfection on Rat1-Akt cell proliferation was observed by using 5-bromodeoxyuridine (BrdU).Then immunofluorescence and kinase assay were used to detect the effect of Ad-14-3-3σ on the phosphorylated level and intracellular location of p27.RESULTS: Ad-14-3-3σ-infected cells had fewer BrdU-positive cells (45%) than control (which was set at 100%).However,Ad-β-gal-infected cells had a high percentage of BrdU-positive cells (98%).14-3-3σ gene transfection downregulated phosphorylation level of p27 and decreased Akt-mediated intracellular location of p27.CONCLUSION: Transfection of 14-3-3σ gene suppresses the proliferation of Akt overexpession cell line Rat1-Akt.14-3-3σ decreases phosphorylation of p27 by downregulating Akt kinase,blocks Akt-mediated cytoplasm dislocation of p27,and inhibits proliferation of Rat1-Akt cells.     

14-3-3σ negatively regulates Akt and inhibits Rat1-Akt cell xenografts

AIM:The present study was designed to investigate the effect of adenovirus-mediated 14-3-3σ on Rat1-Akt cell xenografts and to explore whether the effect was mediated through negative regulation of Akt. METHODS:The effect of Ad-14-3-3σ on Rat1-Akt cells xenografts was observed in nude mice ex vivo and in vivo. Western blotting was used to detect 14-3-3σ protein,phospho-Akt (Thr 308), phospho-Akt (Ser 473), and phospho-(Ser/Thr) Akt substrate in tumor tissue after transfection of 14-3-3σ gene. RESULTS:The tumor volume was dramatically decreased and its emergence was delayed, regardless of using Ad-14-3-3σ-treated Rat1-Akt cells (ex vivo) or injected Ad-14-3-3σ in vivo, of which the effect of the continuously injected group was the best. Levels of Akt protein, phosphorylated Akt and phosphorylated Akt substrates in tumors obtained from Ad-14-3-3σ-treated mice were markedly less than those in PBS or Ad-β-gal-treated mice. CONCLUSION:14-3-3σ suppressed Akt overexpession in cells of Rat1-Akt xenografts by negatively regulating Akt.     

黄皮优质小西瓜新品种泉鑫2号、3号的选育

泉鑫2号、3号系湖南博达隆科技发展有限公司最新选育的黄皮优质有籽小西瓜新品种。泉鑫2号为早熟品种,全生育期85d左右,果实发育期27d。单果重2-2.5kg,露地爬地栽培产量2000-2400kg/667m2,立架栽培3500-4600kg/667m2。果实短椭圆形,果形指数1.10左右,果皮浅黄有黄色细条纹,厚0.7cm;瓤色鲜黄有绿色纤维,瓤质沙脆,纤维少,中心可溶性固形物含量12%左右,边部7.5%左右。抗逆性强,易坐果。泉鑫3号为早熟品种,全生育期85d,果实发育期28d左右。单果重2.3kg左右,露地爬地栽培产量2200-2600kg/667m2,立架栽培3500-4600kg/667m2。植株生长势中强,抗性较强,易坐果。果实短椭圆形,果形指数1.16左右;果皮浅黄色覆浅绿线条;瓤色深红,瓤质爽脆,中心可溶性固形物含量11.5%左右,边部8.5%左右;品质和口感极佳。
