Reciprocal grafts were done between potato (Solanum tuberosum) and six wildSolanum species known to be resistant to the Colorado potato beetle (Leptinotarsa decemlineata (Say)). Potato or wildSolanum foliage from each reciprocal graft was fed to first instar beetles or adult potato aphids (Macrosiphum euphorbiae (Thomas)). The proportion of second instar larvae after 48 h and survival and fecundity (number of nymphs per female) of the aphids were used to i estimate the resistance level of the foliage. Survival and fecundity of potato aphid were reduced on wildSolanum foliage (except withS. circaeifolium ssp.capsicibaccatum andS. okadae) used as scion or rootstock, but were not different from the control (potato-potato graft) on potato foliage. Development of Colorado potato beetle onSolanum foliage was reduced. In some cases, potato foliage grafted to wildSolanum became resistant to the beetle. Results suggest that a chemical factor or factors providing resistance to Colorado potato beetle are translocated from foliage of some wildSolanum species to potato foliage. They also indicate that mode of resistance to the two insects differs quantitatively or qualitatively.  相似文献

Oat and barley β-glucans are polysaccharides known for their beneficial effects on glycemia and cholesterol. These physiological effects seem to be related to increased viscosity in the digestive tract. It has recently been shown that free radicals derived from the reaction of ascorbic acid and oxygen can cause a viscosity loss in β-glucan due to depolymerization through the Fenton reaction. However, recent studies have reported a synergistic effect between xanthan gum and β-glucan on viscosity in heat-treated fruit juices. Therefore, the objective of this study was to determine if xanthan gum has a protective effect against OH radical-induced depolymerization of β-glucan by ascorbic acid. Oat β-glucan (0.4%) solutions were studied in water at pH 4. Ascorbic acid concentrations and viscosity profiles were measured over a period of 56 days. The addition of ascorbic acid caused a rapid decrease in β-glucan viscosity. The presence of xanthan gum reduced the viscosity loss (p ≤ 0.0001) but did not provide complete protection for β-glucan over time. Therefore, incorporation of β-glucan in food formulations still presents a challenge in ensuring its physical and nutritional stability in the final product. The use of OH radical scavengers such as sugars and antioxidant compounds could be used in addition to xanthan gum to obtain improved protection.  相似文献

The jasmonic acid (JA) pathway is involved in the defense mechanisms induced in plants on pathogen attack and the enzyme 12-oxophytodienoate reductase (OPR3) catalyses one of the final steps of JA synthesis. A CAPS marker, OprDM, has been developed using primers designed from the tomato (Solanum lycopersicum)OPR3 cDNA sequence. Sequencing verified that the sequence of the marker derived from potato (Solanum tuberosum) genomic DNA was 98% identical to the corresponding section of that from tomatoOPR3 cDNA. The marker was mapped and examined for association with QTLs for resistance to foliage late blight in potato. OprDM mapped close to marker GP121 on chromosome VII ofS. tuberosum. No marker sequence or clone forOPR3 has previously been mapped in potato or tomato. Results indicated the presence of two putative QTLs for foliage resistance on chromosome VII close to OprDM and CP56. The marker primers could be useful in other studies involving enzymes of the JA synthesis pathway in polymorphicSolanum populations.  相似文献

Levels of total glycoalkaloids (TGA) in foliage of 10 wild, tuberbearingSolanum (Tourn.) L. species differentially resistant to infestation by the potato leafhopper,Empoasca fabae (Harris), were determined. Levels of TGA ranged from a high of 688 mg/100 g fresh wt. in a resistant species,S. polyadenium Greenm. to a low of 13 mg/100 g fresh wt. in a susceptible species,S. bulbocastanum Dun. Foliar concentration of TGA and nymphal infestation by the potato leafhopper were highly correlated (r = ?0.75, p = 0.01). The significant correlation of TGA levels and potato leafhopper resistance suggests that foliar TGA may be a significant factor in the defense of wild potato species against this pest.  相似文献

The systemic insecticides, Dasanit®, carbofuran (Furadan®), phorate (Thimet®), aldicarb (Temik®) and disulfoton (Di-Syston®) were applied in the planting furrow at 2–3 lb actual/acre for potato tuberworm (Phthorimaea operculella (Zeller)) control in Irish potato foliage. None of these compounds gave entirely satisfactory results. Dasanit was the best with slightly over 80% decrease in the number of mines and was followed closely by carbofuran; all treatments needed to be supplemented by foliage sprays for satisfactory tuberworm protection. Azinphosmethyl (Guthion®) gave consistently outstanding results as a foliage spray at 0.5 lb/acre with over 99% tuberworm reduction. Other foliar-applied insecticides giving at least 98% tuberworm control included: Monitor®, monocrotophos (Azodrin®), phosphamidon, Orthene®, Torak®, methomyl (Lannate®) and Dursban® (all at 0.5 lb/acre) and Phosvel® (1 lb/acre).  相似文献

Resistance of 20 F2 Solarium chacoense Bitter clones to the Colorado potato beetle,Leptinotarsa decemlineata (Say), was measured in a field test. Levels of total glycoalkaloids (TGA) and composición of the glycoalkaloid mixtures in foliage of the clones were also determined. Clones with either commersonine or dehydrocommersonine as the major foliar glycoalkaloid were significantly more resistant (lower damage ratings, fewer larvae and adult insects) than clones with solanine and chaconine. Damage ratings were negatively correlated (r = -0.67, p = 0.01) with foliar TGA levels. The results indicate that the types of glycoalkaloids present in the foliage ofS. chacoense may be as important as the level of TGA in limiting damage and numbers of insects.  相似文献

Premature death of potato vines is widespread in irrigated potato fields of the Pacific Northwest.Verticillium dahliae is a major cause of early dying in fields with a history of potato production, but in fields new to potato production, or those that have been fumigated and have a lowV. dahliae population in the soil, premature death of vines can also occur.Erwinia carotovora pv.carotovora (Ecc) andE. carotovora pv.atroseptica (Eca) have been implicated in the early dying disease syndrome in these fields. Both bacterial pathogens are associated with symptoms similar to those caused byV. dahliae; i.e., a progressive chlorosis and necrosis of the foliage and vascular discoloration that may extend a short distance up the stem. Greenhouse pathogenicity studies on cv. Russet Burbank cuttings in which symptom development was identical to that produced byV. dahliae have verified thatEcc andEca can cause potato early dying.  相似文献

Various parameters,e.g. types of microtiter plate for DAS-ELISA (double antibody sandwich-enzyme-linked immunosorbent assay), use of fresh or frozen amplifier solutions for enzyme-amplified-ELISA, and use of sodium diethyldithiocarbamate (NaDIECA) in sample buffer in cocktail-ELISA were evaluated for the detection of potato viruses A, M, S, X, Y and leafroll from potato foliage. Dynatech Immulon immunoplates provided higher readings for all viruses. Fresh amplifier solution in amplifed-ELISA was superior to frozen solutions. Amplified ELISA gave only marginal improvement in the sensitivity over the standard DAS-ELISA. Addition of NaDIECA in sample buffer did not improve the detection of viruses in DAS-, amplified-, or cocktail-ELISA. Cocktail-ELISA can reduce antigen incubation time to as short as 15 min for PVA, PVM and PVX; 1 hr for PVY and PLRV; and 2–4 hr for PVS using pre-coated plates. Although amplified-ELISA is slightly more sensitive than DAS-ELISA for certain potato viruses, it is not suitable for large-scale indexing of potato viruses in Seed Certification Laboratories, in view of the additional steps needed in carrying out this procedure.  相似文献

Potato cultivars were assessed for their sensitivity to potato tuber necrotic ringspot disease (PTNRD) induced by two North American isolates (Tu619 and Tu660) of the tobacco veinal necrosis strain of potato virus Y (PVYN) and belonging to the tuber necrosis (PVYNTN) sub-group. Several widely grown cultivars (e.g. Norchip and Ranger Russet) were found to be highly sensitive to PTNRD, while others (e.g. Russet Burbank, Russet Norkotah and Shepody) were not. The foliage symptoms induced by these isolates were distinct from those reported to be produced by European isolates of PVYNTN, in that systemic necrotic rings and chlorophyll figures on the leaves were not produced. Instead, the symptoms were similar to those of normal strains of PVYN. The apparent correlation between resistance to the PVYO (common) strain group and cultivar sensitivity to PTNRD is discussed.  相似文献

Metronidazole, a common antibacterial drug, was incorporated into a hydrophilic polymer matrix composed of chitosan xanthan gum mixture. Hydrogel formation of this binary chitosan-xanthan gum combination was tested for its ability to control the release of metronidazole as a drug model. This preparation (MZ-CR) was characterized by in vitro, ex vivo bioadhesion and in vivo bioavailability study. For comparison purposes a commercial extended release formulation of metronidazole (CMZ) was used as a reference. The in vitro drug-release profiles of metronidazole preparation and CMZ were similar in 0.1 M HCl and phosphate buffer pH 6.8. Moreover, metronidazole preparation and CMZ showed a similar detachment force to sheep stomach mucosa, while the bioadhesion of the metronidazole preparation was higher three times than CMZ to sheep duodenum. The results of in vivo study indicated that the absorption of metronidazole from the preparation was faster than that of CMZ. Also, MZ-CR leads to higher metronidazole Cmax and AUC relative to that of the CMZ. This increase in bioavailability might be explained by the bioadhesion of the preparation at the upper part of the small intestine that could result in an increase in the overall intestinal transit time. As a conclusion, formulating chitosan-xanthan gum mixture as a hydrophilic polymer matrix resulted in a superior pharmacokinetic parameters translated by better rate and extent of absorption of metronidazole.  相似文献

Clones selected from the wild diploid species,Solanum berthaultii, have been shown to possess valuable levels of resistance to the Colorado potato beetle (Leptinotarsa decemlineata), as well as to insects such as aphids, flea beetles, leafhoppers and the potato tuber moth. Resistance is associated with the presence of glandular trichomes on the foliage ofS. berthaultii (2). Six generations after producing a hybrid between the diploid species and a tetraploid variety, a clone with good beetle and leafhopper resistance, as well as reasonable agronomic characteristics has been produced.  相似文献

Six hundred forty-five accessions of 70Solanum species and six natural hybrids were screened for foliar glycoalkaloids. Total foliar glycoalkaloid (TFGA) level and glycoalkaloid composition of one or more accessions of each species were determined by TLC and GLC analyses. Of the more than 12 different glycoalkaloids found in the wild species in this survey, solanine and chaconine account for more than 60% of the TFGA found in the foliage of these plants. There was wide variation in the total glycoalkaloid levels among the species.S. neocardenasii had the highest average TFGA value of the 70 species, 222 mg/100 g fresh wt (222 mg %) and an accession ofS. chacoense had the highest TFGA value recorded in the survey, 486 mg %. Of the 70 species, only 11 had average TFGA levels of more than 100 mg %. However, one or more accessions in 27 of the species synthesized more than 100 mg %. Forty-five species synthesized an average level of less than 50 mg %. The level usually found in the foliage of commercial potato cultivars is about 50 mg %. Therefore, most of theSolanum species in the potato germplasm collection can be considered low glycoalkaloid species.  相似文献

The characterization of resistance of selected potato, Solanum tuberosum L., breeding clones to the potato psyllid, Bactericera cockerelli (Sulc) (Hemiptera: Triozidae) was investigated. Antixenosis was assessed in choice tests in which a single plant of each genotype was placed inside a rearing cage, where 60 female psyllid adults were released and the number of adults and eggs on each genotype was counted 24 h later. Antibiosis was evaluated in no-choice tests in which adults (five males and five females) were confined in a cage fixed to the upper side of leaves. After 4 h of exposure, adults were removed and the number of eggs counted. The developmental time and survival of offspring were recorded until all insects became adults. All the resistant genotypes showed strong antibiotic effects to B. cockerelli. These results show promise for incorporation into an IPM program against B. cockerelli.  相似文献

Fifteen accessions ofSolarium berthaultii Hawkes andS. berthaultii xS. tarijense Hawkes were assessed for resistance to field infestations of the green peach aphid,Myzus persicae (Sulzer), potato leafhopper,Empoasca fabae (Harris), and potato fleabeetle,Epitrix cucumeris (Harris). Accessions bearing both Type A and Type B glandular trichomes were much more resistant to the green peach aphid and potato leafhopper than accessions bearing Type A hairs alone. All accessions had significantly smaller populations of these 3 pests than S.tuberosum cultivars. Total glycoalkaloid (TGA) content of foliage and tubers was not correlated with insect populations. Foliar TGA levels of field-grown plants varied among accessions, ranging from < 2–240 mg/100 g fresh wt. Solasonine and solamargine were the major foliar glycoalkaloids while solamarines predominated in tubers.  相似文献

Controlled environment experiments were used to determine the effect of light intensity on levels of glycoalkaloids and resistance to the Colorado potato beetle (CPB),Leptinotarsa decemlineata Say, inSolanum chacoense Bitter clones that synthesized acetylated glycoalkaloids, the leptines. Plants were grown under two light intensities, 50 or 250 ?mol m-2S-1, and bioassayed for CPB resistance by placing neonate larvae on excised foliage. Larval development stage and mortality were recorded after four days. The leptine-synthesizing clones responded to the increase in light intensity with a 2–4 fold increase in level of foliar leptines. Larval development rate on foliage from plants grown under high light intensity was reduced and larval mortality increased. Results showed a significant (p>0.05) effect of light on glycoalkaloid synthesis and CPB resistance.  相似文献

In northwest Washington State, fungicidal control of late blight caused byPhytophthora infestons was evaluated for three years. During 1977, a high rainfall year with supplemental sprinkler irrigations, all metalaxyl and chlorothalonil treatments reduced tuber rot at harvest, but only metalaxyl extended control of tuber rot after 4 months’ storage. During 1978, a high rainfall year, one application of metalaxyl sprayed over or dusted on potato seed pieces at planting or applied five times to foliage at 2 or 4 wk intervals gave excellent blight control equal to that with chlorothalonil applied to foliage every 2 wk. In 1979, a low rainfall year,P. infestons did not kill plants in untreated plots until October. Five applications of metalaxyl sprayed every 2 wk on foliage did not control foliage blight. However, metalaxyl reduced foliage blight when dusted on cut potato seed pieces. Mancozeb sprayed every 2 wk also gave excellent control. The possibility that metalaxyl applied to foliage is effective in late blight control only when washed into the soil by rainfall or irrigation for uptake by roots is discussed. Yields and percent U.S. No. 1 grade tubers were not significantly affected by any treatment in these 3-year trials.  相似文献

Six insecticides were evaluated in a replicated field experiment at Gonzales, California, for control of the potato tuberworm,Phthorimaea operculella (Zeller), and the green peach aphid,Myzus persicae (Sulzer). Azodrin®, methomyl, Stauffer N 4543 (O-isobutyl ethylphosphonodithioate S-ester with N-(mercaptomethyl) phthalimide) and azinphosmethyl were applied as foliage sprays by ground sprayer at 2-week intervals. The systemic insecticides, aldicarb (Temik®) and carbofuran (Furadan®) formulated as 10% granules, were applied to the soil as side dress treatments on the same day that the spray treatments were begun. Aldicarb granules were also applied topically so that the granules fell on the young plants and on the soil surface. Azinphosmethyl, Stauffer N 4543, Azodrin® and methomyl effectively controlled the tuberworm larvae in the foliage but effective control of tuberworms in the harvested tubers occurred only where azinphosmethyl, methomyl and Stauffer N 4543 were used. The granule applications of carbofuran and aldicarb were totally ineffective in controlling the potato tuberworm. Effective green peach aphid control was obtained with Azodrin® spray and the two granular aldicarb treatments. The side dressed application of aldicarb appeared to provide better green peach aphid control than did the soil surface application.  相似文献

Potato cultivars Norland, Kennebec, and Russet Burbank were inoculated with a Minnesota isolate of potato virus S (PVS) in the field. The incidence of secondary infection, measured by enzyme-linked immunosorbent assay (ELISA), indicated that significantly more PVS infection occurred in Russet Burbank than Norland or Kennebec, and that Norland had significantly more infection than Kennebec (P≤ 0.01). Mature plant resistance was not observed. Rapid reinfection of PVS-free seedlots resulted from inoculum point-sources when routine cultivation practices were followed. Reinfection rates were significantly greater for seedlots grown at Grand Forks, ND, when compared with seedlots grown at Becker, MN, during both years of the study (P≤ 0.01). After two years in the field, reinfection rates for Norland (71.8%) and Russet Burbank (73.0%) did not differ significantly, but were significantly greater than the reinfection rate for Kennebec (29.5%) (P≤ 0.01). In greenhouse-grown plants, PVS moved out of rub-inoculated leaves within 24 hr, however, 13 and 20 days were required before PVS was detected with ELISA in foliage above and below the inoculated leaf, respectively. The frequency of PVS detection was significantly greater in foliage above the inoculated leaf compared to foliage below the inoculated leaf (P≤ 0.05). Translocation of PVS from inoculated leaves to daughter tubers occurred within 13 days for Russet Burbank and Norland and within 20 days for Kennebec.  相似文献

When five varieties of virus-free potatoes were introduced and multiplied for three years at an elite seed farm in Prince Edward Island where virus-infected potatoes were also grown, approximatly 10% of each variety became infected with both potato virs S (PVS) and potato virus X (PVX). However, when eight varieties of similar stocks were multiplied for 2 years at private farms in the absence of virus-infected potatoes, all remained free from PVX. Four of the varieties also remained free from PVS and four became infected with amounts of less than 5%. In other tests on these stocks, no potato virus Y (PVY) was detected. Post harvest tuber testing onNicotiana debneyi Domin. was found to be more reliable in detecting recent infections of PVS than foliage tests done during the growing season. This was so whether the foliage tests were done on the indicator plant, serologically or doubly-tested by both methods.  相似文献
