Anaemic horses do not provide reticulocytes during an erythropietic response. Acute blood loss was studied in 4 ponies as a model to monitor changes in erythrocyte metabolism during active erythropoiesis. Following phlebotomy, erythrocyte adenosine-5-triphosphate, adenosine-5-diphosphate, glucose-6-phosphate, fructose-6-phosphate, and phosphoenolpyruvate increased signifcantly. The remaining glycolytic intermediates did not change. It is suggested that the determination of erythrocyte adenosine-5-triphosphate may be used to evaluate erythrocyte response following acute blood loss in the horse.  相似文献

The hematological response to acetylphenylhydrazine hemolytic anemia was studied in three standardbred horses. The lifespan of erythrocytes produced during the most severe phase of the anemia were measured with 75-selenomethionine and found to be 144 days as compared to the 139 day lifespan in response to hemorrhagic anemia or 155 days in normal standardbred horses measured previously using the same technique. The erythrocyte counts returned to initial values in 42 days (37, 34 and 54 days) a mean erythrocyte production of 6.4 times 10-12 erythrocytes/day. The mean hemoglobin production was 0.31 gm/kg body weight/day as compared to 0.11 gm Hb/kg/day previously observed in response to hemorrhagic anemia. The mean increase in erythrocyte mean cell volume was 12 mu-3 during the acute response phase to hemolytic anemia in contrast to the absence of a significant increase in the mean cell volume as previously observed during response to hemorrhagic anemia. Free Heinz bodies separated from erythrocytes during the acute phase could not be differentiated from platelets on the hemocytometer counting chamber with standard techniques.  相似文献

Heparin-induced agglutination of erythrocytes in horses   总被引:2,自引:0,他引:2  
Heparin was administered subcutaneously 2 times a day for 4 days to 5 horses. An additional group of 5 horses was used as time-matched controls. Significant decreases in PCV, erythrocyte count, and hemoglobin concentration were observed during heparin therapy. The mean corpuscular volume (MCV) of the heparin-treated horses increased to a peak value of 66.1 fl on the last day of treatment. Erythrocyte creatine concentration and glucose 6-phosphate dehydrogenase activity increased moderately during the treatment. These data indicated that the rapid, profound increase in MCV during heparin therapy was not primarily a result of release of large immature erythrocytes from the bone marrow. A second experiment was subsequently performed, using 3 horses. These horses were given heparin 2 times a day, as was done in the first experiment. Saline wet mounts of erythrocyte suspensions were examined once a day for the presence of agglutination. Cell suspensions were examined with or without exposure to a dilute trypsin solution, and erythrocyte counts were done on each suspension, using an electronic cell counter. Agglutination of erythrocytes was evident on the first day of treatment and became more pronounced as treatment progressed. Exposure to trypsin solution reversed the agglutination. The apparent erythrocyte count decreased and MCV increased sharply in the samples processed normally, but there was little change in those suspensions exposed to trypsin.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Anemia was induced in three groups of horses by moderate or severe acute hemorrhage, or by acetyl phenylhydrazine-induced hemolysis (Groups I, II, and III, respectively). Serial hemograms were done on a multichannel automated blood cell counter with histogram capability. Changes in hematocrit, mean cell volume, erythrocyte number, red cell distribution width (RDW), and standard deviation of erythrocyte volume were examined over time. Significant increases in mean cell volume were first detectable by days 17, 20, and 14 and reached maximum by days 43, 41, and 29, in Groups I, II, and III, respectively (P less than 0.05). Increased mean cell volume was interpreted as reflecting accelerated erythrocyte regeneration; however, not all horses with accelerated regeneration had changes in mean cell volume. Estimated erythrocyte production rate correlated poorly with hematocrit nadir and change in mean cell volume (r = 0.37 and r = 0.36, respectively, P greater than 0.05). In some horses effective regeneration occurs without development of macrocytosis. Mean cell volume remained increased after other parameters returned to control values, suggesting that mean cell volume values may provide retrospective evidence of altered erythrocyte turnover. Anisocytosis as indicated by significant increases in the standard deviation was greatest during the early part of the regenerative response, reaching maximum values on days 30, 28, and 21 in Groups I, II, and III, respectively, and began to decrease as homogeneous repopulation with macrocytes occurred. Red cell distribution width increased significantly only in severe hemorrhage and hemolysis groups, reaching mean maximum values of 24.3 on day 20 and of 26.4 on day 21 in Groups II and III, respectively (P less than 0.05).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In vitro erythrocyte agglutination developed in 3 hospitalized horses receiving heparin treatment. The agglutination caused artifactual decreases in erythrocyte counts and increases in mean corpuscular volume (MCV) values. Treatment of cell suspensions with trypsin eliminated the agglutination and the changes in erythrocyte count and MCV. Similar abnormalities in erythrocyte counts and MCV have been reported in healthy horses treated with heparin and have been cited as evidence of hemolysis and regenerative anemia.  相似文献

At birth, 24 Standardbred foals were assigned at random to 1 of 2 groups and were given a placebo supplement (group 1) or an iron supplement (248 mg of iron/treatment; group 2). Foals were given iron supplement or placebo 4 times during the second and third weeks after birth. Hematologic variables and general health were monitored until foals were 4 months old. Mean PCV in foals of both groups decreased during the first 2 weeks after birth, but values remained within adult horse reference ranges. During the first 6 weeks after birth, foal erythrocytes were smaller than adult horse erythrocytes, but foal erythrocyte glucose-6-phosphate dehydrogenase activity was greater than that in adult horses. At every measurement, indices of anisocytosis were lower in foals, compared with adult horse reference values, suggesting that foals have a homogeneous population of microcytic erythrocytes during early foalhood. In 2-week-old foals of both groups and in 4-week-old placebo-treated foals, mean serum iron concentration was lower than that in adult horses. In foals at birth and during the first 4 months, total iron-binding capacity values were above the adult reference range. In newborn foals, transferrin saturation percentage values decreased to below the reference range in foals from 2 weeks to 4 months after birth. When foals were born, serum ferritin concentration values were above the adult horse reference range, but decreased to within the reference range by the time foals were 1 day old. Immune-mediated hemolytic anemia was diagnosed in two horses on the basis of regenerative anemia, increased erythrocyte fragility in hypotonic saline, autoagglutination, and a positive direct antiglobulin (Coomb's) test. During steroid therapy partial resolution of the anemia was indicated by rising packed cell volume, macrocytosis, and bone marrow erythroid hyperplasia. Using erythrocyte volume distribution histograms (erythrograms), the regenerative response was characterized by analysis of macrocytic and normocytic erythrocyte subpopulations. In both horses, a gradual net increase of about 2 X 10(6) macrocytes/microliter occurred over a four- to five-week period. Over the same interval there was a gradual decrease in the number of normocytes. We suggest that the macrocytes remained large through this period rather than contributing to normocyte population growth. Erythrograms may provide an additional means of evaluating erythrocyte regeneration in horses.  相似文献

Lymphocyte stimulation was observed in whole equine blood in the presence of phytohaemagglutinin and M protein extracted from a typical strain of Streptococcus equi. Blood samples were collected from several healthy horses and horse and pony foals and cultured in vitro with varying concentrations of phytohaemagglutinin and M protein for several days. Phytohaemagglutinin was found to induce lymphocyte stimulation in these animals. Highest mean stimulation indices in horse foals (49.3 +/- 24.4) and pony foals (54.7 +/- 32.0) were observed with 0.625 and 1.25 micrograms/mL phytohaemagglutinin, respectively, at either 72 or 96 hours of incubation. Significantly higher radioactive counts per minute in horse and pony foals were recorded in blood cultures incubated with 0.625 and 1.25 micrograms/mL phytohaemagglutinin. M protein induced a dose related stimulation response in adult horses. Maximum stimulation indices were observed against 125 micrograms/mL M protein at 96 hours. These stimulation indices were higher in adult horses (40.0 +/- 2.2) than observed in pony foals (14.4 +/- 15.7). Higher stimulation levels in adult horses indicated either nonspecific stimulation against M protein or previous exposure of these animals to S. equi.  相似文献

OBJECTIVE: To characterize hematologic and clinical consequences of chronic dietary consumption of freeze-dried garlic at maximum voluntary intake in horses. ANIMALS: 4 healthy sex- and age-matched horses. PROCEDURE: An initial garlic dose (0.05 g/kg, twice daily) was fed to 2 horses in a molasses carrier as part of their normal ration and was gradually increased to maximum voluntary intake (0.25 g/kg, twice daily) over 41 days. Dietary supplementation then continued for a total of 71 days. Two control horses were fed molasses with no garlic with their ration. Blood samples were collected weekly and analyzed for hematologic and biochemical changes, including the presence of Heinz bodies. Recovery of affected blood values was followed for 5 weeks after termination of dietary supplementation with garlic. RESULTS: At a daily dose of > 0.2 g/kg, horses fed garlic developed hematologic and biochemical indications of Heinz body anemia, as characterized by increases in Heinz body score (HBS), mean corpuscular volume (MCV), mean corpuscular hemoglobin, platelet count, and serum unconjugated and total bilirubin concentrations and decreases in RBC count, blood hemoglobin concentration, mean corpuscular hemoglobin concentration, and serum haptoglobin concentration. Recovery from anemia was largely complete within 5 weeks after termination of dietary supplementation with garlic. Heinz body score and MCV remained high at the end of the 5-week recovery period. CONCLUSIONS AND CLINICAL RELEVANCE: Horses will voluntarily consume sufficient quantities of garlic to cause Heinz body anemia. The potential for garlic toxicosis exists when horses are chronically fed garlic. Further study is required to determine the safe dietary dose of garlic in horses.  相似文献

The mechanism of hemolysis induced by onion poisoning in dogs was studied. Six adult, clinically normal Pekingese dogs were fed cooked onions at 30 g/kg body weight/day for 2 days. Blood samples were collected on days 1, 3, 5, 8, 12, 18 and 24 after onion administration, and urine was collected the day after bleeding. Red blood cell counts, hemoglobin and hematocrit were decreased from day 1, and significantly so on day 5 (P < 0.01), contrary to the results of white blood cell counts. So the plasma bilirubin levels and urobilinogen were increased on day 3 (P < 0.01) and day 4 (P < 0.01), respectively. The Heinz body counts were increased dramatically from day 1 (P < 0.01), peaking on day 3 (P < 0.01). Reticulocyte counts were increased from day 1 and the highest value was on day 8 (P < 0.01). Besides anemia, the following erythrocyte parameters were altered: erythrocyte glucose-6-phosphate dehydrogenase was reduced from day 1 and reached the lowest value on day 5 (P < 0.01); the reduced form of nicotinamide adenine dinucleotide phosphate was decreased on day 1 (P < 0.01); reduced glutathione was decreased from day 1 (P < 0.01) and the lowest value was on day 3 (P < 0.01); glutathione-peroxidase was increased on day 1, but decreased significantly on day 3 (P < 0.01); hydrogen peroxide was increased on day 1 (P < 0.01), then went down on days 3-12 (the undermost value on day 5, P < 0.01); catalase was risen dramatically on day 5 (the peak value, P < 0.01); malondialdehyde (MDA) was increased on days 1-8, (P < 0.01), and reached the peak value on day 5 (P < 0.01). Deformity of the erythrocyte membrane was decreased on days 1-12 and fluorescence polarization (rho) and microviscosity (eta) of the erythrocyte membrane were increased on days 1-12 (P < 0.01). There were positive correlations between MDA and rho as well as eta, with correlation coefficients of 0.908 and 0.922, respectively (P < 0.01), but there was a negative correlation between MDA and deformity index, with a correlation coefficient of -0.887 (P < 0.05). This study confirmed that onion poisoning in dogs causes hemolytic anemia.  相似文献

Equine infectious anemia virus was transmitted from an acutely ill and an inapparently infected pony to uninfected ponies by the interrupted feeding of horse flies (tabanids). Transmission from acutely ill ponies was not accomplished following: (1) the interrupted feeding of a single horse fly, (2) bites of horse flies that had fed on an acutely affected pony 24 hours earlier, (3) bites of horse flies that had oviposited after feeding on an acutely affected pony, or (4) the inoculation of larval material derived from horse flies that had fed to repletion. It was concluded that horse fly transmission of equine infectious anemia virus is mechanical only and that infected horses that are free of clinical signs can be a source of virus for insect transmission.  相似文献

Oxidative stress has been correlated with pathologies that impair the performance of athlete horses. The aim of this study was to assess the effects of supplementation with a mixture of polyunsaturated oil and vitamin E on the antioxidant and haematological biomarkers of horses. Horses under maintenance care (n = 6) and horses in training (n = 10) received 100 and 300 ml of the oil mixture respectively. Supplementation was provided for a period of 8 weeks, together with isocaloric inclusion. Blood samples were collected at three time periods (pretest, after 4 weeks and after 8 weeks) to analyse the following: the red blood cell count (RBCc); haemoglobin (Hb); haematocrit (HT); leucocytes; lymphocytes; platelets; the mean corpuscular volume (MCV); the mean corpuscular haemoglobin concentration (MCHC); the standard deviation of the red blood cell distribution width (RDW‐SD); the coefficient of variation of the red blood cell distribution width (RDW‐CV); glutathione peroxidase (GPx); superoxide dismutase (SOD); uric acid (UrAc); total plasma proteins (TPP); and creatine kinase (CK). After the 8 weeks of supplementation, animals under maintenance care exhibited significant increases in SOD, UrAc, the white blood cell count (WBCc), the RDW‐SD and the RDW‐CV (p < 0.05). The animals in training exhibited increases in GPx, SOD and UrAc (p < 0.05). In conclusion, supplementation with polyunsaturated oil and vitamin E increases blood antioxidants among animals under maintenance and in training, with different trends, while contributing to the fight against oxidative stress in each group analysed.  相似文献

Objective: To report the use of ultrapurified bovine hemoglobin (PBHg) in the treatment of red maple toxicosis in horses. Summary: The use of PBHg in a miniature horse and a pony with clinical signs of red maple toxicosis is described. Initial treatment of the horses included blood transfusion in Case 1, and intravenous crystalloid fluids, nasogastrically administered mineral oil, and activated charcoal in both cases. Both cases received PBHg (16 and 11 ml/kg, respectively) when the hematocrit dropped below 10%. Clinical stabilization of the horses (reflected by improved vital signs and mucous membrane color, increase in PvO2, and increase in plasma hemoglobin) was temporary, lasting 12 and 42 hours, respectively. A whole blood transfusion was given 16 hours after the infusion of PBHg in Case 1 and 42 hours after the infusion of PBHg in Case 2. The horses were discharged on days 15 and 17, respectively. Both were stable on discharge and were reported to be doing well 1 month after discharge. New information provided: Ultrapurified bovine hemoglobin is shown to be safe in the treatment of red maple toxicosis. Ultrapurified bovine hemoglobin infusion resulted in temporary improvement in oxygen (O2) delivery as assessed by clinical and laboratory parameters. The use of PBHg may provide an ‘oxygen bridge’ in an acute hemolytic crisis in the horse. It may allow additional time in the treatment of red maple toxicity for the acute hemolysis to subside and for metabolism of the toxin to occur, thus providing a more optimal environment for an effective whole blood transfusion.  相似文献

The enzyme-linked immunosorbent assay (ELISA) antigen-positive and agar-gel immunodiffusion test (AGID)-negative horses do not have infective equine infectious anemia (EIA) virus. The ELISA testing of horse leukocyte culture (HLC) supernatants did detect EIA virus in a HLC that was infected with the Wyoming strain of EIA virus and in HLC derived from horses in febrile, acute, or subacute stages of EIA infection. In supernatants of HLC derived from chronic and inapparent carrier horses, EIA virus was not detected with ELISA. Direct fluorescent antibody tests detected EIA virus in HLC infected with 10(6)TCID50 of the Wyoming strain of EIA virus and in 50% of the HLC from febrile acute or subacute horses. The direct fluorescent antibody testing of HLC derived from chronic and inapparent carrier horses did not detect cell-associated EIA virus. The pony inoculation test proved to be the most reliable and accurate method for detecting infective EIA virus in horses in various stages of EIA infection and accurately correlated with the AGID test.  相似文献

通过8个微卫星位点的多态性检测对宁强矮马和蒙古马共83个个体进行了遗传多样性分析。计算各微卫星位点的等位基因频率、有效等位基因数(Ne)、多态信息含量(PIC)、杂合度(H)和F统计量。结果表明:8个微卫星座位中HMS2变异最大,HTG6变异最小;蒙古马的Ne、PIC、H的平均值均高于宁强马;总群体的平均遗传分化系数Fst为0.017(P<0.01),群体内近交系数Fis为0.171(P<0.01)。说明蒙古马和宁强矮马虽外形和性能上有很大差异,但在品种形成和进化上有着较近的遗传关系。  相似文献

The clinical and ultrasonographic features of seven horses with coxofemoral subluxation are presented. Affected horses included five adult geldings (11–20 years), one large pony (6 years) and a 3-month-old filly. All were lame at the walk except for the pony with grade 3/5 lameness. Lameness was acute in all horses, but three horses progressed after initial improvement. Crepitus, muscle atrophy, and pelvic asymmetry were inconsistent findings. Ultrasonographic diagnosis of subluxation required dynamic visualization of femoral head displacement from the acetabulum while placing weight on the affected limb and subsequent replacement into its normal position upon limb resting. Acetabular rim fractures and joint effusion were visible regardless of weight bearing status in six horses each. No fractures were identified in the pony; the only patient with a good outcome. Six horses had a poor outcome with severe chronic lameness, four of which were euthanized. Postmortem ventrodorsal radiographs obtained in two horses confirmed subluxation only on extended limb projections, but not on hip-flexed projections. Acetabular rim fractures were not visible radiographically in either horse but were confirmed at necropsy. Subluxation was due to an elongated but intact ligament of the head of the femur in both horses. Osteoarthrosis was evident ultrasonographically, radiographically, and at necropsy. Dynamic ultrasonography was readily performed in the standing horse and produced diagnostic images with a low frequency curvilinear transducer. The apparent poor prognosis for horses with subluxation and acetabular fracture illustrate the importance of this imaging technique to identify affected horses.  相似文献

The haematological response of regular, repeated blood harvesting was investigated in 40 Thoroughbred and non-Thoroughbred horses that donate 8 litres of blood every 3 weeks for the purposes of commercial blood production. When this volume of blood was removed on five occasions over 12 weeks, no adverse effect on packed cell volume (PCV), haemoglobin (HB), and red blood cell count (RCC) was observed. Although PCV, RCC and Hb values decreased during the first week after blood collection, followed by a gradual increase in values until the next harvest time, all values remained within published reference ranges. Derived red cell indices [mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), and mean corpuscular haemoglobin concentration (MCHC)] also remained within reference range. We conclude that the removal of approximately 8 litres of blood (approximately 16 ml kg(-1)or 20 per cent of blood volume for a 500 kg horse) from blood donor horses every 3 weeks allows time for adequate recovery of haematological variables and does not result in adverse haematological changes.  相似文献

In order to understand the blood physiological and biochemical reactions after long-distance transport, 23 Akhal-teke horses (male horse 14, mare 9, aged 2 to 6 years old), were selected and transported from Russian to Alashankou of Xinjiang (5 d road transport). 7 and 35 d after reaching the destination, the jugular vein blood were collected for the determination of physiological and biochemical indicators and stress-related hormone levels. The remote control horses without transport (15 local Akhal-teke horses,male horse 10, mare 5, aged 6 to 10 years old) were used for comparative study. The results showed that on the 7th and 35th day after transport, the number and proportion of granulocytes (Gran), hematocrit (HCT), blood platelet (PLT) and alkaline phosphatase (ALKP) were significantly or extremely significantly higher than those of the local Akhal-teke horses (P<0.05; P<0.01), while the number and percentage of intermediate cells (Mid) and the level of blood glucose (GLU) were significantly or extremely significantly decreased (P<0.05; P<0.01); On the 35th day after transport, the mean red blood cell volume (MCV), phosphorus (PHOS) and total protein (TP) were significantly or extremely signifcantly increased (P<0.05; P<0.01), and the percentage of lymphocytes (LY) was significantly decreased (P<0.05); On the 7th day after transport, the levels of antidiuretic hormone (ADH), growth hormone (GH), UREA and insulin (INS) were significantly or extremely signifcantly decreased (P<0.05; P<0.01), and the total bilirubin (TBIL) and triiodothyronine (T3) were signigicantly increased (P<0.05), but there was no significant difference between the two groups (P>0.05) on the 35th day after transport. Based on the above results, after the long-distance transport, there had a corresponding transient stress response in Akhal-teke horses, which required some adjustment after the recovery period (35 d) to let most of the indicators were consistent with those without long-distance transport. So the horses could not be used until they were in a normal state, in order to avoid long-term irreversible lesions.  相似文献

To explore the prevalence of Theileria equi (T.equi) infection in horse in Guizhou province, the antibody level and 18S rRNA gene were detected from blood samples of Guizhou pony, Southwest horse and Yili horse using competitive ELISA and PCR methods.Giemsa-stained blood smear was prepared to observe T.equi in red blood cells.Intact protozoans of T.equi were observed in red blood cells of horses at Giemsa-stained slide smears with a detection rate of 12.5%.The 18S rRNA gene fragment of T.equi was detected in Guizhou pony, Southwest horse and Yili horse, and the consistent rates with the known nucleotide sequence were 97% to 100%.The PCR result indicated that the positive rates of T.equi in Guizhou pony (76.62%) and Yili horse (73.81%) were similar, which were higher than that in Southwest horse (33.33%).Furthermore, the antibody levels against T.equi in Guizhou pony (24.68%) and Southwest horse (12.12%) were lower than that in Yili horse (31.71%).A weak correlation between the antibody level and the blood physicochemical indexes was calculated from Guizhou pony and Southwest horse, including weak positive correlations with neutrophils numbers, gamma-glutamyl transferase and creatine kinase levels, and weak negative correlations with the numbers of red blood cell, white blood cell, platelet and lymphocyte and contents of hemoglobin.It suggested that a higher proportion of T.equi infection present in three herds.  相似文献

试验采用显微镜观察、PCR和竞争性酶联免疫吸附试验(competitive enzyme-linked immunosorbentassay,cELISA)等方法对贵州矮马、西南马和伊犁马的马泰勒虫病的感染状况进行研究。结果显示,从32份新鲜的血液涂片中,观察到形态完整的马泰勒虫(Theileria equi)虫体,检出率12.5%。从贵州矮马、西南马及伊犁马3个马群血液总DNA中都检测到马泰勒虫的18S rRNA基因片段,与已知序列的同源性为97%~100%;相比之下,贵州矮马与伊犁马的阳性率相近,分别为76.62%和73.81%,西南马较低,仅为33.33%。另外,经cELISA检测,与伊犁马(31.71%)相比,贵州矮马和西南马血液中抗马泰勒虫抗体的阳性率较低,分别为24.68%和12.12%,并与两个马群的血液理化指标存在一定的联系:与中性细胞数量、γ-谷氨酰转移酶和肌酸激酶的含量呈弱正相关;与红细胞、白细胞、血小板、淋巴细胞数量及血红蛋白含量呈弱负相关。这些研究结果提示3个马群中均存在较高比例的马泰勒虫感染。  相似文献
