AIM:To study the effect of fructose-1, 6-diphosphate (FDP) on adriamycin(ADM)-induced cardiomyocyte apoptosis in rats. METHODS:Twenty-four Wistar rats were randomly divided into three groups: control group, ADM treated group and FDP intervention group. The contents of malondialdehyde (MDA) and NO2-/NO3-, the activities of glutathione peroxidase (GPx) and superoxide dismutase (SOD) were determined by colorimetric method in myocardial tissue, and the cardiomyocyte apoptosis was detected by TUNEL method in myocardial tissue, and the expression of inducible nitric oxide synthase (iNOS) mRNA, Bcl-2 mRNA and Bax mRNA in myocardial tissue were detected by in situ hybridization. RESULTS:The contents of NO2-/NO3- and MDA in myocardial tissue, the expressive levels of iNOS mRNA and Bax mRNA in cardiomyocyes and its apoptotic amounts in FDP intervention group were significantly lower than those in ADM treated group (P<0.01). However, the activities of SOD and GPx in myocardial tissue, the expressive level of Bcl-2 mRNA of cardiomyocytes in FDP intervention group were significantly higher than those in ADM treated group (P<0.01). CONCLUSION:FDP antagonized the reduced expression of Bcl-2 mRNA and increased expression of Bax mRNA in myocardial tissue induced by ADM, and in turn inhibited ADM-induced cardiomyocyte apoptosis.  相似文献

AIM: To investigate the effect of intermittent hypoxia on the apoptosis of mouse pancreatic β cells and its possible mechanism. METHODS: Thirty healthy male C57BL/6J mice were randomly divided into 3 groups: control group, sustained hypoxia (SH)group and intermittent hypoxia (IH)group. Insulin tolerance test was performed immediately after experiment. The level of malondialdehyde (MDA) and superoxide dismutase (SOD) were detected by chemical colorimetry. Real-time PCR was used to measure the mRNA expression of manganese superoxide dismutase(MnSOD) and glutathione peroxidase(GPx1). The apoptosis of pancreatic β cells was determined by the method of TUNEL. RESULTS: The levels of insulin resistance and content of MDA in the pancreatic tissue in IH group were significantly higher than those in control group and SH group (P<0.01). The activity of SOD and the mRNA expression of MnSOD and GPx1 in IH group were significantly lower than those in control group and SH group (P<0.01). The apoptotic rate of IH group was significantly elevated as compared with control group and SH group (P<0.01). No significant difference of all above indexes between control group and SH groups was observed (all P>0.05). CONCLUSION: Apoptosis of pancreatic β cells induced by oxidative stress associated with IH in the pancreatic tissue may be involved in the pathogenesis of obstructive sleep apnea hyponea syndrome with insulin resistance and type 2 diabetes.  相似文献

AIM: To investigate the effect of hypoxia on the proliferation and apoptosis of pulmonary artery smooth muscle cells (PASMC); and to evaluate the role of hypoxia-inducible factor-1α(HIF-1α), iNOS, P-ERK1/2 protein expression in hypoxic pulmonary hypertension (HPH) pathogenesis.METHODS: Cultured rat PASMC were divided into normoxic group; hypoxic group; hypoxia+ADM(adrenomedulin) group, hypoxia+L-NAME(iNOS inhibitor) group; hypoxia+PD98059 group. Proliferation was investigated by MTT and PCNA. Apoptosis was examined by flow-cytometry. Westen blotting was used to measure protein expression of HIF-1α, P-ERK1/2 and iNOS. RESULTS: (1) A value of 24 h-hypoxia was significantly higher than that in the normoxic group (P<0.01). In the hypoxia+PD98059 group, ADM was significantly lower than that in hypoxia group, whereas A value of the hypoxia+L-NAME was significantly higher than that in hypoxic group and normoxic group (P<0.01). (2) PCNA was positive in PASMC after 24 h hypoxia (P<0.01). PD98059, ADM inhibited the expression of PCNA significantly (P<0.01), whereas L-NAME increased the expression of PCNA significantly (P<0.01). (3) Apoptosis index was not significantly difference among the different groups (P>0.05). (4) HIF-1α, iNOS and P-ERK1/2 expression was poorly positive in normoxic group, positive after hypoxia for 4h (P<0.01), reaching its peak at 8 h hypoxia (P<0.01), HIF-1α, P-ERK1/2 expression declined after 24 h hypoxia. L-NAME promoted the expression of HIF-1α, PD98059 inhibited the expression of HIF-1α, iNOS and P-ERK1/2 partly. ADM inhibited the expression of HIF-1α partly, promoted the expression of iNOS. CONCLUSION: (1) Hypoxia stimulates the proliferation of PASMC, and has no obvious effects on the apoptosis of PASMC. (2) HIF-1 plays an importent role in the proliferation of hypoxic PASMC.  相似文献

AIM: To investigate the effects of sesamin on progression of renal injury in renal hypertensive and hyperlipidemic rats (RHHR). METHODS: RHHR was induced by 2K1C and high lipid baitvessel. After 7 weeks of intragastric administration with sesamin, the contents of serum creatinine (Scr), blood urea nitrogen (BUN), 24 h urinary protein excretion (UPE) were measured. In addition, the activity of total antioxidative capacity (T-AOC), superoxide dismutase (SOD), the concentrations of malondialdehyde (MDA), hydrogen peroxide (H2O2), and the nitric oxide synthase (NOS) and nitric oxide (NO) levels in renal homogenate were measured. RESULTS: Compared with the model group, seasamin (in 100 mg·kg-1 and 33 mg·kg-1 groups) evidently decreased the contents of Scr, BUN, UP and the concentration of MDA, iNOS, H2O2 in renal tissure. It also improved the levels of NO, cNOS and activity of SOD, T-AOC in renal tissure. CONCLUSION: Sesamin ameliorates hypertensive and hyperlipidemic-induced renal injury, probably by enhancing antioxidative activity, scavenging hydroxyl radical and restraining iNOS level.  相似文献

AIM:To investigate the effects of sevoflurane(Sevo) preconditioning on myocardial dysfunction in lipopolysaccharide(LPS)-challenged mice. METHODS:Forty male BALB/c mice were randomly allocated to 4 groups:control group, LPS group, Sevo+LPS group and Sevo group. Following pretreatment with or without 2% Sevo for 30 min and washing out for 10 min, all mice received intraperitoneal injection of LPS or normal saline(NS). The mice received an echocardiographic evaluation by a high-resolution in vivo imaging system 12 h after administration of LPS or NS. The mice were then killed and the hearts were removed for histological analysis. Serum levels of lactic dehydrogenase(LDH), creatine kinase-MB(CK-MB) were measured with an automatic biochemical analyzer. The myocardium was homogenized for detecting the activity of inducible nitric oxide synthase(iNOS) and the content of nitric oxide(NO). RESULTS:Echocardiographic evaluation demonstrated that LPS resulted in an increase in lert ventricular end-diastolic volume and significant decreases in stroke volume,cardiac output and ejection fraction. The alteration of cardiac functions was inhibited by the pretreatment with Sevo. LPS caused significant elevation of LDH and CK-MB in serum samples and severe pathological damage of the hearts. Compared with LPS group, serum levels of LDH and CK-MB were reduced and pathological damage was attenuated in Sevo+LPS group. Sevo preconditioning also significantly attenuated the increases in iNOS and NO induced by LPS. CONCLUSION:Sevo preconditioning protects against myocardial impairment and myocardial dysfunction in LPS-challenged mice. Inhibition of iNOS activity and of NO production by Sevo preconditioning may contribute to the beneficial role in the process of cardioprotection during endotoxemia.  相似文献

AIM: To study the effect of dexamethasone (DEX) on the adrenomedullin (ADM) and its gene expression in lung tissue of asthmatic rats. METHODS: 30 healthy male SD rats were randomly divided into three groups (10 for each). The asthmatic model was established by ovalbumin inhalation and injection. The mRNA expression of ADM was examined by RT-PCR and the protein expression was detected by immunohistochemical method. The airway wall thickness, the airway smooth muscle (ASM) thickness and pulmonary tissue changes were observed under light microscope. RESULTS: The expression of ADM mRNA and protein in the asthma group A were higher than those in the control group(group C) (P<0.05), indicating that the moderate expression of ADM in asthmatic rat lung tissue is compensatory. The expression was significantly higher in DEX group (group B) than that in group A (P<0.01), indicating that DEX stimulated the expression of ADM mRNA and protein in lung tissue of asthmatic rats. CONCLUSION: The remarkable expression of ADM after the therapy of dexamethasone is one of its therapeutic mechanisms.  相似文献

AIM: To investigate the effect of hydrogen sulfide (H2S) donor (NAHS) on oxidative stress of adriamycin-induced dilated cardiomyopathy rats. METHODS: Weight-matched adult male Wistar rats were randomly divided into 5 groups as follows: (1) ADR group (n=12), in which 2.5 mg/kg of adriamycin was injected intraperitoneally once a week for 10 weeks (total dose of 25 mg/kg). (2) ADR+small-dose NaHS group (n=12), in which the dosage and the use of adriamycin were as mentioned above, while NaHS solution was injected to rats at a dosage of 2.8 μmol·kg-1·d-1 at the same time. (3) ADR + large-dose NaHS group (n=12), in which the dosage and the use of adriamycin were as mentioned above, while NaHS solution was injected to rats at a dosage of 14 μmol·kg-1·d-1 at the same time. (4) Control group (n=9), in which an equivalent volume of physiological saline was administered weekly for a total of 10 weeks. (5) NaHS group (n=9), in which 14 μmol/kg of NaHS solution was injected to rats intraperitonealy once a week for 10 weeks. Hemodynamic and echocardiographic measurements were obtained 10 weeks after the treatment. Meanwhile, H2S and malondialdehyde (MDA) concentrations, the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in serum and myocardial tissues were evaluated, respectively. RESULTS: The cardiac functions in the group of ADR rats depressed obviously. H2S concentrations, SOD and GSH-Px activities in serum and myocardial tissues of ADR group rats were all significantly decreased as compared with those in the control group (P<0.01). The MDA concentrations in serum and myocardial tissues in ADR group rats were both increased significantly (P<0.01). Exogenous administration of H2S donor NaHS markedly attenuated ADR-induced cardiac dysfunction, and MDA concentration in myocardial tissues was significantly reduced (P<0.01). Serum SOD activity was obviously increased in ADR+large-dose NaHS group compared with control group (P<0.01), and GSH-Px activity in myocardial tissues was markedly increased in ADR+large-dose NaHS group compared with control group (P<0.05). CONCLUSION: H2S might play an important role in the development of adriamycin-induced dilated cardiomyopathy. Administration of exogenous H2S effectively improves myocardial contractile activity, reduces the accumulation of lipid peroxides and increases the capability of antioxidants to inhibit oxidative stress and prevents myocardial damage.  相似文献

AIM: To investigate the effects of external counterpulsation (ECP) on nitric oxide (NO) and nitric oxide synthase (NOS) and the expression of NOS gene in myocardial infarction canines. METHODS: Nineteen healthy dogs were randomly divided into three groups ie. controls, ischemia group, ischemia and ECP group. Serum NO concentrations and myocardium NO levels and NOS specific activity were determined by modified nitrate reductase method. The protein synthesis of sub-type NOS including inducible NOS (iNOS) and endothelial NOS (eNOS) of myocardial tissue were also determined by immunohistochemical method. The constitutive NOS (cNOS) mRNA was measured via in situ hybridization. RESULTS: 120 and 180 minutes after the ligating of LAD, serum NO concentration in ECP groups were higher than those in ischemic groups (P<0.05). The NO levels and NOS specific activity in myocardium of ischemic dogs were lower than those in controls and ECP group (P<0.05). Protein synthesis of iNOS increased and that of eNOS decreased in ischemic myocardium. But ECP could control the protein synthesis of iNOS, and increase that of eNOS. Further studies showed that the expression of cNOS mRNA decreased in ischemic myocardial tissue, ECP might promote the expression of it and regulate NOS in the gene level. CONCLUSION: The results suggested that it was one of the most important mechanisms through raising the NO levels to protect ischemic myocardium in ECP.  相似文献

AIM: To investigate the effects of ox-LDL on TLR2 and TLR4 expression and production of TNF-α, IL-10, IL-12, NO and MDA in macrophages and to observe intervention effect of GW1929 in above procedure. METHODS: The mouse peritoneal macrophages were pretreated with ox-LDL (50 mg/L, 100 mg/L) and GW1929 (20 μmol/L) respectively for 24 h. The concentrations of MDA, NO-2/NO-3, TNF-α, IL-10 and IL-12 in the culture fluid were detected. Flow cytometry was used to observe TLR2 and TLR4 expressions after the mouse peritoneal macrophages were pretreated with ox-LDL (50 mg/L) and GW1929 (20 μmol/L) respectively for 6 h, 12 h, and 24 h. RESULTS: The concentrations of MDA, NO-2/NO-3, TNF-α and IL-10 in ox-LDL (50 mg/L, 100 mg/L) group were higher than those in control and GW1929 group obviously, but the concentrations of above index in ox-LDL (50 mg/L, 100 mg/L)+GW1929 group were lower than those in ox-LDL (50 mg/L, 100 mg/L) group apparently. No IL-12 in every group was detected. Expressions of TLR-2 in ox-LDL+GW1929 (6 h, 12 h, 24 h) group were lower than those in ox-LDL (6 h, 12 h, 24 h) group respectively. TLR-4 expressions in ox-LDL+GW1929 (12 h) were lower than those in ox-LDL (12 h) apparently. CONCLUSION: ox-LDL up-regulates TLR2 and TLR4 expressions and promotes the production of ROX, NO, TNF-α and IL-10 in macrophages. GW1929 is capable of inhibiting the above ox-LDL effects.  相似文献

AIM: To investigate the effect of siRNA on myocarditis induced by MCMV in BALB/c mice. METHODS: One handred and twenty male BALB/c mice (4 weeks old) were divided randomly into 6 groups with 20 mice each. Among these groups, one normal control group, one viral control group and 4 experimental groups were assigned. Mice in viral control and 4 experimental groups were inoculated intraperitoneally injection (ip) with 100 μL of DMEM medium containing TCID5010-4 MCMV, and followed by 200 μL of different doses of liposomally endocapsulated siRNA in 4 experimental groups and by 200 μL of vehicle solution of siRNA in viral control group 30 min later. Mice in normal control group were first inoculated ip 100 μL of DMEM medium and followed by 200 μL of vehicle solution of siRNA 30 min later. At day 7, 14, 21, 28, 35, 42, 49, 56, 63 and 70 of postinfection, all mice were killed and blood samples were collected for detecting serum zymogram (CK, CK-MB, GOT, LDH and LDH1/LDH2) and the anti-cardiac β1 receptor autoantibody titer level. Hearts were removed aseptically for detecting cardiac histological lesions and ultrastructure changes. RESULTS: Decreased morbidity of myocarditis, lightened cardiac pathologic lesions and ultrastructural changes in 4 experimental groups were observed. Serum zymogram (CK, CK-MB, GOT, LDH and LDH1/LDH2) and β1 autoantibody titer level in viral control group were significantly higher than those in normal control group, but zymogram and β1 autoantibody titer levels in 4 experimental groups were between those in viral control group and normal control group. The activities of CK, CK-MB, GOT, LDH and LDH1/LDH2 and β1 autoantibody titer level decreased accompanied with siRNA dose increasing. These effects were in a dose dependent manner. CONCLUSION: siRNA protects myocardial tissue against MCMV infection.  相似文献

AIM:To explore the association among COX2 , iNOS and p38MAPK in late phase of preconditioning. METHODS: The expression of COX2 , iNOS and p38MAPK activity were determined by western blotting and the injury of cardiomyocyte was assessed by LDH release and trypan blue exclusion in four groups: control group, group IPC (ischemic preconditioning), group SMT (iNOS inhibitor), group NS398 (COX2 inhibitor) and group SB (p38MAPK inhibitor).RESULTS:The expression of COX2 in group IPC increased markedly in comparision with group SMT and group SB.The expression of iNOS in group SB was lower than that in group IPC and group NS398.The difference of the amount of iNOS was not significant between group IPC and group NS398.The difference of the amount of phospho-p38MAPK was not significant among group IPC, group SMT and group NS398(P>0.05).The LDH was lower, and cell viability was higher in group IPC than those in control group.The LDH was higher, and cell viability was lower in group SMT, group NS398 and group SB than those in group IPC. CONCLUSIONS:In late phase of preconditioning , the p38MAPK activity and expression of iNOS and COX2 increase significantly in rat cardiomyocytes, activited p38MAPK mediates iNOS, then promotes COX2 expression.  相似文献

AIM:To investigate the protective effect of crude Paecilomyces tenuipes polysaccharides (cPtPs) and Paecilomyces tenuipes polysaccharides (PtPs) in a rat model of acute hepatic necrosis induced by carbon tetrachloride (CCl4). METHODS:Wistar rats were divided into four groups (control group, CCl4 group, cPtPs+CCl4 group and PtPs +CCl4 group), the four groups were given intragastrically with normal saline, cPtPs and PtPs for 15 d, respectively. In the last two days, these groups except control group were injected peritoneally with CCl4. Serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL), direct bilirubin (DBIL) and indirect bilirubin (IBIL) were detected by automatic biochemistry analyzer. Pathological changes of hepatic tissue were assessed by hematoxylin-eosine (HE) staining. The levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in liver homogenate were analyzed using xanthinoxidase and thio-barbituric acid, respectively. The concentration of Ca2+ in hepatocyte mitochondria was determined by colorimetric method. The expression of α-smooth muscle actin (α-SMA) was examined in hepatic tissue by immunohistochemical method. RESULTS:Compared with control group, serum levels of ALT, AST, TBIL, DBIL and IBIL in CCl4 group increased significantly, denaturation and necrosis implicated to the whole hepatic lobules. Compared with CCl4 group, serum levels of ALT, AST, TBIL, DBIL and IBIL in PtPs +CCl4 group decreased significantly, denaturation and necrosis located in the third region of hepatic lobules, the level of SOD increased and MDA decreased (P<0.05) in endochylema. Concentration of Ca2+ in mitochondria decreased in PtPs +CCl4 group and cPtPs +CCl4 group (P<0.05, P<0.01, respectively). Expression of α-SMA was found little in PtPs+CCl4 group. CONCLUSION:PtPs, the effect is better than cPtPs, lessens CCl4-induced hepatonecrosis significantly. The role may be related with anti-lipid peroxidation.  相似文献

AIM:To observe the changes of iNOS and eNOS in lung tissue and NO in bronchoalveolar lavage fluid (BALF) in smoking rats.METHODS:80 Wistar rats were divided into control, smoking group, L-NIL group and L-NAME group (rats were exposed to smoke and injected (i.p.) with selective iNOS inhibitor L-NIL or NOS inhibitor L-NAME). iNOS and eNOS protein levels in whole lung were detected by immunohistochemical staining, and NOS mRNA was quantified using RT-PCR. In addition, NO2-/NO3- was determined using Griess assay.RESULTS:The expression of iNOS mRNA and protein in smoking rats increased, the expression of eNOS mRNA and eNOS protein decreased, and the total cell count and the level of NO2-/NO3-in BALF increased(P<0.05). In vivo, L-NIL reduced the total cell count and NO2-/NO3- in BALF (P<0.05), while L-NAME had no effect on them.CONCLUSION:Cigarette smoke increased expression of iNOS mRNA and protein and decreased expression of eNOS mRNA and protein. The large amount of NO generated by iNOS may amplify inflammation in lung tissue.  相似文献

AIM:To observe the effects of glycine on hypoxia-reoxygenation (H/R)-induced myocardial dysfunction, and to further clarify the protection of glycine (GLY) against myocardial ischemia/reperfusion injury and its mechanism. METHODS:A cardiac H/R model was established using a Langendorff isolated heart preparation. The left ventricular systolic pressure (LVSP), left ventricular end diastolic pressure (LVEDP), left ventricular developed pressure (LVDP), the maximum rising and dropping rates of left ventricular pressure (dp/dtmax and dp/dtmin) were observed. The coronary effluents at different time points were collected respectively to detect the concentration of superoxide dismutase (SOD) and malondialdehyde (MDA). RESULTS: The indexes of cardiac functions in H/R group were lower than those in other groups. After H/R, the indexes in GLY plus H/R group were higher than those in H/R group. Glycine inverted the effects of the decrease in SOD and the increase in MDA concentrations induced by H/R. CONCLUSION:Glycine ameliorates the cardiac functions under the condition of hypoxia-reoxygenation injury in isolated rat hearts. The mechanisms may be related to suppressing lipid peroxidation.  相似文献

AIM: To investigate the expression of calcium sensing receptor(CaSR) during myocardial injury induced by ischemia/reperfusion and disclose the relationship between CaSR and myocardial ischemia/reperfusion. METHODS: The experimental model was established by the 30 min ligating and 1 h, 2 h, 4 h, 6 h reperfusing the left descending coronary artery (LAD) in rats. Wistar rats were randomly divided into 5 groups: sham group, ischemia/reperfusion 1 h, 2 h, 4 h, 6 h groups (I/R 1 h, 2 h, 4 h, 6 h group). CaSR mRNA expression was detected by RT-PCR. Left ventricular function was recorded. The levels of plasma lactate dehydrogenase (LDH), alondialdehyde (MDA) and superoxide dismutase (SOD) were measured. The change of ultrastructure in the ischemia/reperfusion myocardium of rats was observed by electron microscopy. RESULTS: LVSP,±dp/dtmax and SOD activity decreased gradually with the reperfusion time prolonged. LDH and MDA peaked at 2 h. The ultramicro-structural injury at the 1 h and 2 h was more serious than that at 4 h and 6 h. The expression of CaSR increased significantly after reperfusion of 1 h and 2 h, and decreased after 4 h and 6 h. CONCLUSION: The increased expression of CaSR mRNA and serious injure of myocardium were observed. CaSR may be associated with the myocardial ischemia/reperfusion injury.  相似文献
