AIM:To study the effect of cilazapril on pulmonary vascular endothelial dysfunction in hypoxic rats. METHODS:The structure and function of endothelium in hypoxic rats were studied by biochemical analysis, radioimmunoassay, transmission electron microscope and correlated with hemodynamic. RESULTS:1) The change and damage of ultrastructure in endothelial cell (EC) were obsevered in hypoxic rats. 2) The contents of plasma nitric oxide (NO) and superoxide dismutase (SOD) activity in blood as well as endothelial nitric oxide synthase (eNOS) activity in the lung tissue were significantly lower in the hypoxic rat than those in contral animals. The concentrations of plasma endothelin-1(ET-1) and angiotensin converting enzyme(ACE) as well as malondialdehyde(MDA) were significantly higher in the hypoxic rat than these in contral animals. The relaxing and contracting factors had a significant positive/negative correlation with mean pulmonary artery pressure (mPAP). 3) Cilazapril significantly decreased the level of ET-1 and ACE and significantly increased the level of NO and activity of eNOS and SOD. At the same time, cilazapril extenuated hypoxia-induced injuries of EC. CONCLUSION:The results indicate that damaging structure and dysfunction of EC existes in hypoxic rats. The cilazapril effectively preventes and treates the chronic hypoxic PH by relieving the injury and improving secretion in EC.  相似文献

AIM: To study the relationship between the activity of phospholipase A2 (PLA2) and pulmonary structural remodeling with the model of chronic isobaric hypoxic pulmonary hypertension. METHODS: 29 healthy Sprague-Dawley rats were randomly divided into normal control group, chronic hypoxic group and hypoxia plus Polidatin (PD) group. By diameter, the arteries were divided into two groups: arteries of group I (30 μm-100 μm) and group II (101 μm-200μm). The mean pulmonary arterial pressure (mPAP) was measured by inserting a microcatheter into the pulmonary artery. The PLA2 activity was measured with modified microtitrimetic method. The pulmonary tissue and arterioles morphology changes were examined under light microscope. RESULTS: It was found that after 21 days hypoxia, the mean pulmonary arterial pressure (mPAP), the PLA2 activity in blood and lung homogenate increased significantly. The media thickness of group I arteries increased (P<0.01) while that of group II arteries had no significant changes. The ratio of media area and adventitia of both groups was raised. Under light microscope, it was observed that pulmonary vascular endothelium proliferated, media became thickening and adventitia matrix increased. Pretreatment with PD could attenuate the changes mentioned above. CONCLUSION:PLA2 plays an important inducing role through promotion of the pulmonary vascular structural remodeling in the formation of chronic hypoxic pulmonary hypertension.  相似文献

AIM:To investigate relationship among phosphatidylinositol 3-kinase (PI3K) and hypoxia-inducible factor 1α (HIF-1α) and vascular endothelial growth factor (VEGF) in lung of rats with hypoxia-inducible pulmonary hypertension. METHODS:Forty male adult Wistar rats were randomly divided into five groups (eight rats in each group):control group (C group) and groups with hypoxia for 3, 7, 14 and 21 days (H3, H7, H14 and H21 group). Mean pulmonary arterial pressure (mPAP), right ventric hypertrophy index (RVHI) and vessel morphometry were measured. The levels of HIF-1α mRNA expression in lung tissue was measured by in siteu hybridization (ISH). The protein expression of HIF-1α,VEGF and phosphorylated protein kinase β (P-AKT) were observed by immunohistochemistry or Western blotting. RESULTS:mPAP increased significantly 7 days after hypoxia [(23.53±1.78) mmHg], peaked 14 days after hypoxia, then remained on the high level. Pulmonary artery remodeling index (extern diameter 100 μm) and RVIH became evident 14 days after hypoxia. Expression of P-AKT protein in control group was poorly positive, but was up-regulated in pulmonary arterial tunica intima and tunica media in all hypoxia rats. HIF-1α mRNA staining was poorly positive in control,hypoxia for 3 days and hypoxia for 7 days, but began to increase significantly 14 days after hypoxia (0.305±0.104, P<0.05), then remained stable. Expression of HIF-1α protein in control group was poorly positive, but was up-regulated in pulmonary arterial tunica intima in all hypoxic rats. In pulmonary arterial tunica media, the levels of HIF-1α protein was markedly up-regulated after 3 days (0.029±0.019, P<0.05 ), reached its peak 7 days after hypoxia (0.232±0.008, P<0.05), then tended to decline 14 days and 21 days after hypoxia. Expression of VEGF protein began to increase 7 days after hypoxia (0.188±0.018, P<0.05), reached its peak 14 days after hypoxia (0.238±0.017, P<0.05), then remained on the high level in pulmonary arterial tunica intima. Linear correlation analysis showed that P-AKT, HIF-1α mRNA, VEGF and mPAP were correlated with vessel the morphometry and RVHI (P<0.01). P-AKT was positively correlated with HIF-1α and VEGF (tunica intima). CONCLUSION:P-AKT, HIF-1α and VEGF are all involved in the pathogenesis of hypoxia-induced pulmonary hypertension in rats.  相似文献

AIM: To investigate the effect of diltiazem on mean pulmonary arterial pressure (mPAP) and nitric oxide synthase (NOS) in arterioles in chronic hypoxic hypercapnic rats. METHODS: Twenty-four rats were randomly divided into three groups: control group (A), hypoxic hypercapnic group (B), hypoxic hypercapnia+ diltiazem group (C), constitutive endothelial NOS (ceNOS) were observed in arterioles of rats using the technique of immunohistochemistry, ceNOS mRNA were observed by the technique of in situ hybridization. RESULTS: (1) mPAP was significantly higher in rats of B group than that of A and C group(P<0.01). Differences of mCAP were not significant between A group and B groups (P>0.05), but mCAP was lower in rats of C group than that in B group. (2) Light microscopy showed WA/TA (vessel wall area/total area) was significantly lower in rats of C group than that of B group (P<0.01), electron microscopy showed that diltiazem inhibited the proliferation of smooth muscle cells and collageous fibers of pulmonary arterioles in chronic hypoxic hypercapnic rats. (3) Immunohistochemistry showed the average value of integral light density (LD) of ceNOS in pulmonary arterioles was significantly higher in rats of C group than that of B group (P<0.01), in situ hybridization showed LD of ceNOS mRNA in pulmonary arterioles was significantly higher in rats of C group than that of B group (P<0.01). CONCLUSION: Diltiazem inhibited pulmonary hypertension, the proliferation of smooth muscle cells and collagenous fibers of pulmonary arterioles in chronic hypoxic hypercapnic rats by incresing the expression of ceNOS in pulmonary arterioles.  相似文献

AIM: To study the effect of chronic hypoxic hypercapnia on expression of heme oxygenase-1 (HO-1). METHODS: Sprague-Dawley rats were randomly divided into three groups: control group(A),hypoxic hypercapnic group(B), hypoxic hypercapnia+hemin group(C). HO-1 and HO-1 mRNA were observed in pulmonary arterioles by the technique of immunohistochemistry and in situ hybridization. RESULTS: ① mPAP and weight ratio of right ventricle (RV) to left ventricle plus septum (LV+S) were significantly higher in rats of B group than those of A and C group (P<0.01). Differences of mCAP were not significant in three groups(P>0.05). ② Blood CO concentration was significantly higher in rats of B group than that of A group (P<0.01), it was much higher in C group than that of B group(P<0.01). ③ Light microscopy showed that vessel well area/total area (WA/TA), density of medial smooth muscle cell (SMC) and media thickness of pulmonary arterioles were much higher in rats of B group than those of A and C group (P<0.01). ④ The observation by electron microscopy showed proliferation of medial smooth muscle cells and collageous fibers of pulmonary arterioles in rats of B group, hemin could reverse the changes mentioned above. ⑤ HO-1 and HO-1 mRNA in pulmonary arterioles was significantly higher in rats of B group than those of A group(P<0.01), and they were significantly higher in rats of C group than those of B group (P<0.01). CONCLUSION: Expression of HO-1 mRNA and HO-1 in pulmonary arterioles was enhanced by hypoxic hypercapnia. Hemin partly inhibited pulmonary hypertension and pulmonary vessel remodeling by enhancing the expression of HO-1 mRNA and HO-1.  相似文献

AIM: To study the protective effect of A2a adenosine receptor (A2aAR) on hypoxic pulmonary hypertension in the rats treated with salidroside. METHODS: Sprague-Dawley rats were randomly divided into 6 groups: normal control group, hypoxia group, hypoxia+salidroside (low dose) group, hypoxia+salidroside (median dose) group, hypoxia+salidroside (high dose) group, and hypoxia+CGS-21680 (a selective agonist of A2aAR) group. Pulmonary hypertension in the rats was produced for 4 weeks. Mean pulmonary artery pressure (mPAP), mean carotid arterial pressure (mCAP) and the weight ratio of right ventricle/(left ventricle+septum)[RV/(LV+S)] were measured. The expression of A2aAR in the pulmonary arterioles was determined by immunohistochemistry and in situ hybridization. The mRNA expression of A2aAR in the lung tissues was detected by real-time RT-PCR. The protein level of A2aAR in the lung tissues was analyzed by Western blotting. RESULTS: The mPAP in hypoxia group was significantly higher than that in normal control group. The mPAP in hypoxia+salidroside (high dose) group and CGS-21680 group was significantly lower than that in hypoxia group. RV/(LV+S) in hypoxia group were significantly higher than that in normal control group. RV/(LV+S) in hypoxia+salidroside (median dose) group, hypoxia+salidroside (high dose) group and CGS-21680 group were lower than that in hypoxia group. The ratio of vessel wall area/vessel total area (WA/TA) in hypoxia group was significantly higher than that in normal control group. WA/TA in hypoxia+salidroside (low dose) group, hypoxia+salidroside (median dose) group, hypoxia+salidroside (high dose) group and CGS21680 group were obviously lower than that in hypoxia group. The expression of A2aAR was significantly higher in hypoxia group than that in normal control group. The expression of A2aAR in hypoxia+salidroside (high dose) group and CGS-21680 group was obviously higher than that in hypoxia group. CONCLUSION: The A2aAR attenuates pulmonary vessel remodeling and pulmonary hypertension induced by hypoxia. Salidroside protects the pulmonary vessel from remodeling and inhibits the development of hypoxia-induced pulmonary hypertension by up-regulation of A2aAR expression.  相似文献

AIM: To investigate the changes of 5-hydroxytryptamine(5-HT)levels and to observe the expression and distribution of 5-HT1B receptors in the lung tissues of hypoxic pulmonary hypertension(HPH) rats for exploring the mechanisms of hypoxic pulmonary hypertension.METHODS: Forty male Sprague-Dawley rats were randomly divided into 4 groups: normoxia control(control group), 3 weeks hypoxia group, 4 weeks hypoxia group and 5 weeks hypoxia group. The rats in normoxia control group stayed in normal environment. The rats in 3 weeks hypoxia group, 4 weeks hypoxia group and 5 weeks hypoxia group were kept respectively in hypoxia chamber for 3 weeks, 4 weeks and 5 weeks respectively to establish the HPH animal model. After HPH was established, the mean pulmonary pressure(mPAP) and the right ventricular systolic pressure(RVSP) were recorded by a micro-catheter. RV/(LV+S) ratio was calculated to assess the right ventricular hypertrophy. 5-HT levels in plasma and lung tissues of HPH rats were measured by ELISA. The expression and distribution of 5-HT1B receptors in the lung tissues were measured by the methods of immunohistochemistry and Western blotting. RESULTS: Compared to the normoxia controls, mPAP, RVSP and RV/(LV+S)% in 3 weeks hypoxic rats increased significantly(P<0.05), and continued to increase following prolonged hypoxia. The results of ELISA showed that 5-HT levels in plasma and lung tissues of HPH rats continued to increase following prolonged hypoxic exposure(P<0.05). The 5-HT1B receptors were localized mainly in the intima of the pulmonary arteries in normal rats. Exposed to hypoxia, the immuno-reactivity for 5-HT1B receptors increased in the media of pulmonary arteries in 3 weeks hypoxic rats, particularly those bordering the adventitia. The increase in the expression of 5-HT1B receptor was observed following prolonged hypoxic exposure. The results of Western blotting showed the same changes of 5-HT1B receptor expression in the lung tissues as that of 5-HT1B immuno-reactivity in pulmonary arteries.CONCLUSION: Hypoxia induces the high 5-HT levels and the over-expression of 5-HT1B receptors in the pulmonary arteries of HPH rats, indicating the underlying mechanism of 5-HT in the development of HPH.  相似文献

AIM:To study the effect of safflower injection on expression of COX-2 mRNA in chronic hypoxic hypercapnic rat pulmonary arterioles.METHODS: Sprague-Dawley rats were randomly divided into normal control group, hypoxic hypercapnic group (B), hypoxic hypercapnia+ safflower injection group (C). The concentration of TXB2 and 6-Keto-PGF1α in plasma and in lung were detected by the technique of radioimmunoassay. COX-2 mRNA was observed in arterioles from rats by the technique of in situ hybridization. RESULTS: ① Mean pulmonary arterial pressure(mPAP), weight ratio of right ventricle (RV) to left ventricle plus septum (LV+S) were much higher in B group than those in control group. No significant difference of mean carotid arterial pressure(mCAP) was observed in three groups. ② The concentration of TXB2 and the ratio of TXB2/6-keto-PGF1α were significantly higher in B group than those in control group. ③ Light microscopy showed that vessel wall area/total area, the density of medial smooth muscle cells and the thickness of medial smooth cell layer were significantly higher in B group than those in control group. Electron microscopy showed proliferation of medial smooth muscle cells and collagenous fibers in pulmonary arterioles in B group. Safflower injection reversed the changes mentioned above. ④ Expression of COX-2 mRNA in pulmonary arterioles was much higher in C group than those in B group. Differences of COX-1 mRNA in pulmonary arterioles were not significant between these two groups.CONCLUSION:Safflower injection increases the expression of COX-2 mRNA in chronic hypoxic hypercapnic rat pulmonary arterioles, indicating an important mechanism that safflower injection inhibits the formation of hypoxic hypercapnia pulmonary hypertension and pulmonary vessel remodeling.  相似文献

AIM: To observe the expression of hypoxia inducible factor-1α (HIF-1α) gene and heme oxygenase-1 (HO-1) gene in pulmonary arteries in hypoxic rats. METHODS: Forty male Wistar rats were exposed to hypoxia for 0, 3, 7, 14 or 21 days. Mean pulmonary pressure (mPAP), vessel morphometry, right ventricle hypertrophy index (RVHI) were measured. Lungs were either inflation fixed for immunohistochemistry and in situ hybridization or frozen for later measurement of HO-1 enzyme activity. RESULTS: During hypoxia, mPAP increased to significantly higher values than the control values after 7-day of hypoxia,reaching its peak after 14-day of hypoxia, then remained on the high level. Pulmonary artery remodeling developed significantly after 14-day of hypoxia. Expression of HIF-1α protein in control was poorly positive, but was up-regulated in pulmonary arterial tunica intimae of all hypoxic rats. In pulmonary arterial tunica media, the levels of HIF-1α protein were markedly up-regulated after 3-day and 7-day of hypoxia, then tended to decline after 14-day and 21-day of hypoxia. HIF-1α mRNA staining was poorly positive in control, hypoxia for 3 days and hypoxia for 7 days, but began to enhance significantly after 14-day of hypoxia, then remained stable. CONCLUSION: HIF-1α and HO-1 are both involved in the pathogenesis of hypoxia-induced pulmonary hypertension in rats. Furthermore, HIF-1α may inter-regulate with HO-1 gene in this process.  相似文献

AIM: To clarify the role of eNOS and ET-1 in development of pulmonary hypertension (PH) associated with congenital heart diseases. METHODS: 40 patients were randomly divided into three groups: severe or moderate PH group (group A, 12 cases), slight PH group (group B, 14 cases) and normal group (group C, 14 cases). ET-1 and eNOS were examined by using the technique of immunohistochemistry. RESULTS: ① Plasma ET-1 concentration was significantly higher in group A and B than that in group C (P<0.01). Serum NO concentration in group A and B was significantly lower than that in group C (P<0.01). ② The proliferation of medial smooth muscle cells and collagenous fibers of pulmonary arteriole in patients with PH were observed by electron microscopy. ③ ET-1 in pulmonary arteriole was significantly higher in group A and group B than that in group C (P<0.01). eNOS in pulmonary arteriole was significantly lower in group A and group B than that in group C (P<0.01). CONCLUSION: ET-1 and eNOS may play an important role in the pathogenesis of PH.  相似文献

AIM:To study the effects of airway and pulmonary inflammation on pulmonary arterial remodeling in rats with chronic bronchitis (CB) and emphysema.METHODS:Twenty-four male Wistar rats were divided into three groups (n=8): Group A: four-weeks CB and emphysema;Group B: six-weeks CB and emphysema group;Group C: normal control.The rat model of CB and emphysema was established by intratracheal instillation of lipopolysaccharide (LPS) and daily exposure to cigarette smog.The arterial blood gas analysis,pulmonary hemodynamics changes and cell counts in bronchoalveolar lavage fluid (BALF) were measured.The pathomorphological changes of airway inflammation,alveoli destruction and pulmonary arterial remodeling were observed by HE straining and triple straining.RESULTS:(1) The characteristic pathological changes of CB and emphysema were observed in group A and B.Neutrophils were the main cells infiltrated into the walls of airway in group A.Lymphocytes and macrophages were the main cells in group B.(2) Right ventricular systolic pressure (RVSP),mean pulmonary arterial pressure (mPAP),the ratio of the weight of right ventricle/left ventricle and septum (RV/LV+S) in group A and B were significantly higher than those in group C (P<0.05).The amount of muscular artery (MA) in group A and B were significantly higher than that in group C (P<0.05).(3) In group A and B,the levels of MA,RVSP,mPAP and RV/LV+S was correlated positively with the average alveolar area,the total cell counts and differential cell counts of neutrophils,lymphocytes and macrophages in BALF,and the level of infiltration into the walls of airway,respectively (P<0.05).The positive correlation was observed with the percentage of neutrophils,lymphocytes and macrophages between group A and B (P<0.05).The amounts of MA were also correlated positively with RVSP,mPAP and RV/LV+S (P<0.05).CONCLUSIONS:(1) The pulmonary artery hypertension,the right ventricular hypertrophy and the pulmonary arterial remodeling appeared before hypoxia.These may be related with the degree of the pulmonary inflammation.(2) The characteristic of pulmonary arterial remodeling was small artery organization,and correlated positively with the changes of hemodynamics.  相似文献

AIM:The aim of this study is to investigate the acute inhibitory effects of adrenomedullin(ADM1-52) on hypoxia pulmonary hypertension and its influence on systemic blood pressure.METHODS:Thirty-six male Wistar rats were divided into two groups. Eighteen were exposed to hypoxia for 21 days as hypoxic pulmonary hypertension group,and another eighteen were kept in ambient as control group. Each group were divided into three subgroups which were injected intravenously with ADM 0.1 nmol/kg, 0.3 nmol/kg, 1 nmol/kg, respectively,then hemodynamic parameters were recorded. Plasma cyclic adenosine 3,5,-monophosphate (cAMP) was measured by radioimmunoassay before,during and after injection of 0.3nmol/kg adrenomedullin(ADM1-52).RESULTS:Mean pulmonary arterial pressure (mPAP)in the hypoxia and control rats treated with ADM decreased, the fall in hypoxia rats is more obviously than control(P<0.05), ADM (0.1-0.3 nmol/kg) produced dose-related reductions in mPAP in hypoxia rats(P<0.05). Mean systemic blood pressure(mSBP) of the rats in both groups decreased in a does-dependent manner, and it was more obvious in control rats. Plasma cAMP is higher in hypoxia group than that in control group.CONCLUSION:ADM depresses the rat hypoxia pulmonary hypertension in short term through, at least partly, cAMP-dependent mechanism.  相似文献

AIM:To study the effect of ligustrazine on pulmonary hypertensive rats induced by hypoxic hypercapnia. METHODS:Thirty rats were randomly divided into three groups:control group(A),hypoxic hypercapnic group(B), hypoxic hypercapnia+ligustrazine(lig.) group(C). RESULTS: (1) Mean pulmonary arterial pressure(mPAP)of group B was significantly higher than that of group A and mPAP of group C was significantly lower than that of group B(P<0.01),differences of mean carotid pressure(mCAP) were not significant among three groups (P>0.05); (2)Electron microscopy and immunohistochemistry showed ligustrazine could inhibit the diposition of collagenous fiber(collagen typeⅠ)in pulmonary arterioles induced by hypoxic hypercapnia; (3) Plasma endothelin level of group C was significantly lower than that of group B (P<0.01), serum (NO 2-/NO3-) of group C was significantly higher than that of group B (P<0.01). CONCLUSION:Ligustrazine can inhibit pulmonary hypertension and the diposition of collagen type Ⅰ in pulmonary arterial wall induced by hypoxic hypercapnia.  相似文献

AIM AND METHODS: To explore the effects calcitonin gene-related peptide (CGRP) and endothelin-1(ET-1) on the mechanisms of hypoxic pulmonary hypertension (HPH), the contents of CGRP and ET-1 in plasma of pulmonary artery and thoracic aorta and in extractives of lung and ventricular tissues of the chronic hypoxic rats were determined by radioimmunoassay. The changes of their hemodynamic indices and right heart hypertrophy index were monitored simultaneously. RESULTS: The level of pulmonary artery plasma CGRP was significantly higher than that of thoracic aorta plasma, but just the reverse was ET-1 or the ratio of ET-1 and CGRP in control rats( P<0.01). Compared with controls, the level of pulmonary artery plasma CGRP was gradually reduced in all hypoxic rats, but ET-1 was enhanced after 7 and 14 days of hypoxia and was decreased after 21 days of hypoxia. With prolonging time exposed to hypoxia, the level of thoracic aorta plasma CGRP was markedly increased in hypoxic animals compared with controls ( P<0.05), the positive correlation significantly with increased pulmonary arterial pressure( r = -0.896, P =0.05), but the lower level of thoracic aorta plasma ET-1 showed or negative correlation with pulmonary arterial pressure. CONCLUSIONS: These data suggest that the unbalance of effects of CGRP and ET may plays an important role in regulating the resistance of pulmonary circulation and has close relation with the formation of HPH.  相似文献

AIM: To observe the effect of Panax notoginoside (PNS) on the pulmonary artery pressure and the p38 mitogen-activated protein kinase(p38 MAPK) in lung tissues of rats treated with hypoxia. METHODS: Thirty adult male SD rats were randomly divided into 3 groups. The rats in normal control group were exposed to normal conditions, the rats in hypoxia group were exposed to isobaric hypoxia, and the rats in hypoxia+PNS group were treated with PNS under the condition of hypoxia. After 4 weeks of treatment, the mean pulmonary arterial pressure (mPAP) and the mean carotid arterial pressure (mCAP) were measured by cardiac catheterization. The heart was isolated, and the right ventricle (RV), left ventricle plus ventricular septum (LV+S) were weighed to calculate the ratio of RV/(LV+S).The quantity of phospho-p38 MAPK(p-p38 MAPK) in rat pulmonary arterioles was determined by the method of immunohistochemistry and the mRNA content of p38 MAPK was tested by RT-PCR. RESULTS: The mPAP and RV/(LV+S) in hypoxia group were higher than those in normal control group. The expression of p-p38 MAPK in rat pulmonary arterioles and p38 MAPK mRNA in the lung tissues were higher than those in normal control group (P<0.05). The mPAP, RV/(LV+S), the expression of p-p38 MAPK in rat pulmonary arterioles and p38 MAPK mRNA in the lung tissues in hypoxia+PNS group were significantly lower than those in hypoxia group (P<0.05).CONCLUSION: PNS possesses the preventive and therapeutic effect on hypoxic pulmonary hypertension by decreasing p-p38 MAPK and down-regulation of p38 MAPK mRNA in the lungs.  相似文献
