Phylogenetic relationships among and genetic variability within 60 goats from two different indigenous breeds in Narok and Isiolo counties in Kenya and 22 published goat samples were analysed using mitochondrial control region sequences. The results showed that there were 54 polymorphic sites in a 481‐bp sequence and 29 haplotypes were determined. The mean haplotype diversity and nucleotide diversity were 0.981 ± 0.006 and 0.019 ± 0.001, respectively. The phylogenetic analysis in combination with goat haplogroup reference sequences from GenBank showed that all goat sequences were clustered into two haplogroups (A and G), of which haplogroup A was the commonest in the two populations. A very high percentage (99.90%) of the genetic variation was distributed within the regions, and a smaller percentage (0.10%) distributed among regions as revealed by the analysis of molecular variance (amova ). This amova results showed that the divergence between regions was not statistically significant. We concluded that the high levels of intrapopulation diversity in Isiolo and Narok goats and the weak phylogeographic structuring suggested that there existed strong gene flow among goat populations probably caused by extensive transportation of goats in history.  相似文献

To explore the genetic diversity and origin for genetic resource protection of Huili Black goat, the mitochondrial DNA (mtDNA) D-loop was investigated. mtDNA D-loop sequences of 41 goats were analyzed by PCR, sequencing techniques, and biological information and the phylogenetic trees were constructed. The mtDNA sequences of the Huili Black goat ranged from 1211 to 1213 bp, and 2 sequences were 1211 bp, 29 sequences 1212 bp, and 10 sequences 1213 bp. The content of A+T (60.1%) was higher than one of G+C (39.9%). There were 9 haplotypes, and the haplotype diversity was 0.842+0.00368. The nucleotide diversity was 0.01542+0.00034. The phylogenetic analysis showed that Huili Black goat was distributed in a branch, and were closed to Jianchang Black goat, Chengdu Ma goat, Jintang Black goat, Guizhou White goat, Guizhou Black goat, but they were less related to Capra falconeri. Huili Black goats had rather abundant genetic diversity, and were greatly affected by other goat breeds in history.  相似文献

To provide useful knowledge on goat breed origin and history, we studied the mitochondrial DNA (mtDNA) of 69 goats from five different breeds, Camosciata delle Alpi, Maltese, Nubian, Saanen and Sarda, and one population, the Tunisian. All goats analysed displayed a moderate haplotype and nucleotide diversity. The highest was in the Sarda – the autochthonous breed reared in Sardinia. On the basis of mtDNA control region sequences, animals showed a high genetic haplotype diversity, 35 haplotypes were each represented by a single sequence and only a few haplotypes were shared among the animals. New haplotypes of goats reared in the Mediterranean area were identified and the majority of Italian goats belonged to haplogroup A. This result confirmed worldwide distribution and diversity of haplogroup A.  相似文献

川渝部分山羊品种(类群)遗传多样性微卫星标记研究   总被引:1,自引:0,他引:1  
为分析川渝山羊品种(类群)的遗传多样性和系统发生关系,利用30个微卫星标记,对6个山羊品种(类群)进行分析。结果表明:金堂黑山羊遗传多样性最丰富,群体多态信息含量、平均杂合度和有效等位基因数分别为0.777、0.819和6.54;大足黑山羊各项指标最低,分别为0.736、0.787和5.57。对6个品种(类群)聚类分析表明,金堂黑山羊与南江黄羊的首先聚类在一起,然后依次与板角山羊、川东白山羊、大足黑山羊和波尔山羊聚类。各山羊品种(类群)的聚类关系与其来源、育成史及地理分布基本一致。  相似文献

4个引进山羊品种mtDNA控制区序列变异和系统发生关系研究   总被引:5,自引:2,他引:5  
本研究测定了四川4个引进山羊品种24个个体的线粒体控制区全序列,并从GenBank获得山羊属2个野山羊种的2每控制区序列。利用MEGA2.0软件构建分子系统发育无根树。序列分析表明:山羊控制区线粒体控制全序列长度为1 212bp或1 213 bp,A+T含量占59.9%,其中64个核苷酸位点存在变异(约占5.28%),核苷酸多样度为 1.731%,这些差异共定义了16种单倍型,单倍型多样性为0.913±0.048。安哥拉山羊、波尔山羊都有自己独特的单倍型,与其他品种间都没有共享类型。使用NJ法构建了系统发育树,结果表明:2个野山羊种中,角(?)羊与家养山羊的关系相对较近,4个家养山羊品种有2个母系来源,在4个家养山羊品种内部,安哥拉山羊的分化要比其他3个品种早。  相似文献

试验选取贵州山羊4个群体(共55只),设计引物对其mtDNA细胞色素b基因片段进行测序,测序结果与GenBank报道的8个山羊品种mtDNA细胞色素b基因进行比对分析。结果发现,在55只贵州山羊中有11个变异位点,其中转换10次,T/G颠换1次;10种单倍型中有7种单倍型为1个群体独享,3种单倍型为多个群体共享。4个山羊类群间存在较近的亲缘关系,既是独立的类群,其间也存在基因交流,其中贵州黑山羊和贵州麻羊的亲缘关系最近,与贵州白山羊的亲缘关系次之,与贵州小香羊的亲缘关系最远。从山羊品种间遗传距离构建的邻接树可见,贵州4个山羊类群均起源于镰刀状角羊骨羊。  相似文献

试验对四川板角山羊、贵州白山羊、贵州黑山羊、贵州麻羊及湖南马头山羊5个山羊品种,共计69个个体的Cytb基因片段进行测序分析比较。结果显示,69个山羊Cytb基因序列均为长1140bp的同源基因,共发现17个变异位点,观察到14种单倍型;5个山羊品种的群体遗传多样性在0.442—0.889之间,核苷酸多样度从0.00145—0.00253。表明5个山羊遗传多样性属于中等;5个山羊品种可分为2大类群,即:贵州黑山羊、贵州麻羊、贵州白山羊和湖南马头山羊4个品种为一类,四川板桥山羊1个品种为另一类。  相似文献

[目的]分析我国地方培育奶山羊品种与国外引进品种的遗传进化关系。[方法]以4个国内奶山羊品种(文登奶山羊、关中奶山羊、崂山奶山羊、雅安奶山羊)和3个新西兰引进奶山羊品种(阿尔卑斯奶山羊、吐根堡奶山羊、萨能奶山羊)为研究对象,采集33只个体的外周血样本,提取血液基因组DNA,利用PCR法扩增线粒体DNA(mitochondrial DNA,mtDNA)D-loop区全长序列,对测序获得的序列进行生物信息学分析,探究不同奶山羊品种的遗传多样性及进化关系。[结果]7个品种奶山羊的mtDNA D-loop区中A、T碱基含量高于G、C碱基含量。共检测到82个多态位点,25个单一多态位点,55个简约信息位点。各品种单倍型多样度(Hd)范围为0.905~1.000,核苷酸多样度(Pi)范围为0.001 51~0.013 32;共存在26种单倍型,文登奶山羊和萨能奶山羊各有5个单倍型,阿尔卑斯奶山羊有4个单倍型,崂山奶山羊、吐根堡奶山羊、关中奶山羊、雅安奶山羊各有3个单倍型;各品种核苷酸平均差异数(KXY)范围为6.400 00~38.450 00,核苷酸歧异度(DXY)范围为0.005 79~0.034 76,遗传分化系数(GST)范围为0.000 00~0.186 05,遗传分化指数(FST)范围为0.231 56~0.971 52。品种间系统发育树表明,文登奶山羊和崂山奶山羊聚为一支;关中奶山羊与3种新西兰奶山羊遗传距离较近,从遗传学角度证实了关中奶山羊由国外奶山羊与地方品种经杂交选育而成;雅安奶山羊与其他品种遗传距离最远。[结论]中国奶山羊存在2个支系起源且未发现群体扩张;中国培育奶山羊品种含有较多的国外奶山羊血统;文登奶山羊与崂山奶山羊亲缘关系较近,雅安奶山羊在遗传进化中可能存在地域隔离。  相似文献

本研究参阅文献挑选 80个随机引物 ,从中筛选出 3 0个重复性好的引物 ,对四川黑山羊、南江黄羊、北川白山羊、成都麻羊 4个山羊品种和藏绵羊进行RAPD分析 ,并进一步对 1 3只南江黄羊个体 ,1 8只黑山羊个体的基因组DNA进行PCR扩增。用Nei氏公式计算品种间的遗传距离 ,用UPGMA法构建树状聚类图。结果表明 :黑山羊与南江黄羊的遗传距离最小 ,亲缘关系较近 ;藏绵羊与各品种间的遗传距离都很大 ,亲缘关系远 ;RAPD技术可作为一种有效的分子标记用于山羊品种之间遗传亲缘关系的分析  相似文献

Genetic variations in chromosome Y are enabling researchers to identify paternal lineages, which are informative for introgressions and migrations. In this study, the male‐specific region markers, sex‐determining region‐Y (SRY), amelogenin (AMELY) and zinc finger (ZFY) were analysed in seven Turkish native goat breeds, Angora, Kilis, Hair, Honaml?, Norduz, Gürcü and Abaza. A SNP in the ZFY gene defined a new haplotype Y2C. All domestic haplogroups originate from Capra aegagrus, while the finding of Y1A, Y1B, Y2A and Y2C in 32, 4, 126 and 2 Turkish domestic goats, respectively, appears to indicate a predomestic origin of the major haplotypes. The occurrence of four haplotypes in the Hair goat and, in contrast, a frequency of 96% of Y1A in the Kilis breed illustrate that Y‐chromosomal variants have a more breed‐dependent distribution than mitochondrial or autosomal DNA. This probably reflects male founder effects, but a role in adaptation cannot be excluded.  相似文献

It is widely accepted that autochthonous cattle breeds can be important genetic resources for unforeseeable environmental conditions in the future. Apart from that, they often represent local culture and tradition and thus assist in the awareness of ethnic identity of a country. In Croatia, there are only three indigenous cattle breeds, Croatian Bu?a, Slavonian Syrmian Podolian and Istrian Cattle. All of them are threatened but specialized in a particular habitat and production system. We analysed 93 microsatellites in 51 animals of each breed to get thorough information about genetic diversity and population structure. We further set them within an existing frame of additional 16 breeds that have been genotyped for the same marker set and cover a geographical area from the domestication centre near Anatolia, through the Balkan and alpine regions, to the north‐west of Europe. The cultural value was evaluated regarding the role in landscape, gastronomy, folklore and handicraft. The overall results recognize Croatian Bu?a being partly admixed but harbouring an enormous genetic diversity comparable with other traditional unselected Bu?a breeds in the Anatolian and Balkan areas. The Podolian cattle showed the lowest genetic diversity at the highest genetic distance to all remaining breeds but are playing an important role as part of the cultural landscape and thus contribute to the tourist industry. The genetic diversity of the Istrian cattle was found in the middle range of this study. It is already included in the tourist industry as a local food speciality. Current and future conservation strategies are discussed.  相似文献

本研究采用PCR-RFLP方法检测生长分化因子9(growth differentiation factor 9, GDF9)基因FecTT突变在济宁青山羊、贵州白山羊、大足黑山羊、陕南白山羊、古蔺马羊、马头山羊、波尔山羊、成都麻羊、萨能奶山羊、文登奶山羊、板角山羊、金堂黑山羊、关中奶山羊、川东白山羊、安哥拉山羊、辽宁绒山羊、内蒙古绒山羊等17个山羊品种中的分布。结果表明,在17个山羊品种中都未检测到FecTT突变,提示,GDF9基因影响Thoka绵羊高繁殖力的FecTT突变对以上17个山羊品种的繁殖力均没有显著影响。  相似文献

The aim of this study was to compare ovarian response and embryo yield of Day 0 protocol in Angora goats (AG) and indigenous Kilis goats (KG) in the non-breeding season. A total of 16 Angora goats (AG group) and 11 Kilis goats (KG group) were used in this study. In the synchronization process, after controlled internal drug release withdrawal, when estrus signs were observed, natural mating was performed. Ovarian response was determined by synchronized laparotomy 6 days after natural mating, and number of corpora lutea (CL) was recorded. Embryos were collected and morphologically evaluated by stereomicroscope. Synchronization rates did not differ between AG (88%, 14/16) and KG group (91%, 10/11). In AG and KG groups, the proportion of CL on the right (44% and 53%, respectively) and left (56% and 47%, respectively) ovaries were similar. The CL number per animal did not differ significantly between the two breeds and was determined as 4.4 ± 0.90 in AG group and 6.4 ± 1.44 in KG group. Transferable embryo yields were significantly higher in AG group (31/42, 74%) compared to KG group (16/46, 35%) in the non-breeding season (P < 0.01). In conclusion, it is suggested that the day 0 protocol can be used for goat superovulation in the non-breeding season; however, transferable embryo yields are affected by the breed.  相似文献

测定了四川7个地方山羊品种(类群)43个个体的mtDNA控制区全序列,结果表明:山羊mtDNA控制区全序列长度为1212bp或1213bp,A+T含量(59.9%)明显高于G+c含量(40.1%)。共检测到74个变异位点,序列均为中性突变,核苷酸多样度为1.686%,这些差异共定义了27种单倍型,单倍型多样度为0.966,遗传多样性较为丰富,品种(类群)间存在不同程度的遗传分化。7个地方山羊品种(类群)间的遗传距离变异范围为0.0017~0.0306。用MEGA软件的NJ法构建单倍型序列的系统发育无根树,结果表明四川地方山羊品种(类群)有两个母系来源,但是否就对应于角笋骨羊(Capra aegagrus)和捻角山羊(Capra falconeri)两个野生祖先,还有待于进一步研究。  相似文献

设计4对引物,采用PCR-SSCP技术检测骨形态发生蛋白2(bone morphogenetic protein 2,BMP2)基因外显子2、3在济宁青山羊、安哥拉、波尔、内蒙古绒山羊4个品种共191个个体中的单核苷酸多态性,分析该基因对济宁青山羊高繁殖力的影响,并对济宁青山羊BMP2基因2个外显子进行克隆测序。结果表明,4对引物的扩增片段在4个山羊品种中均无多态性;山羊扩增片段核苷酸和氨基酸序列与人、黑猩猩、牛、犬、大鼠、小鼠6个物种的同源性分别为86.5%~91.3%和88.8%~99%;与其他物种相比,山羊BMP2扩增片段推导氨基酸序列存在3处特有突变,分别是位于第131、353、365位的脯氨酸、缬氨酸、亮氨酸变为苏氨酸(P131T)、天冬氨酸(V353D)、脯氨酸(L365P)。可见,哺乳动物BMP2基因外显子2、3序列保守性强,该区域可能不是影响山羊高繁殖力的功能结构域。  相似文献

The aim of this study was to characterize the genetic diversity of domestic goat in China. For this purpose, we determined the sequence of the mitochondrial DNA (mtDNA) control region in 72 individuals of the Yangtze River delta white goat, and reanalysed 723 published samples from 31 breeds/populations across China. All goat haplotypes were classified into four haplogroups (A–D) previously described. The phylogenetic pattern that emerged from the mtDNA control region sequence was confirmed by the analysis of the entire cytochrome b sequence of eight goats representative of the four haplogroups. It appeared that in Chinese domestic goat, haplogroups A and B were dominant and distributed in nearly all breeds/populations, while haplogroups C and D were only found in seven breeds/populations. Four breeds/populations contained all four haplogroups. When grouping the breeds/populations into five geographic groups based on their geographic distributions and ecological conditions, the southern pasturing area had the highest diversity whereas the northern farming area had the lowest diversity. 84.29% and 11.37% of the genetic variation were distributed within breeds and among breeds within the ecologically geographical areas, respectively; only 4% of genetic variation was observed among the five geographic areas. We speculate that the traditional seasonal pastoralism, the annual long-distance migrations that occurred in the past, and the commercial trade would account for the observed pattern by having favoured gene flows.  相似文献

为从分子水平对云上黑山羊遗传结构做出评价,本研究以云上黑山羊核心群母羊108只(来自7个育种核心场、10个群体)、育种素材云岭黑山羊和努比山羊各11和14只、对照组波尔山羊10只为试验动物,用Illumina公司Iselect Goat60k芯片技术在全基因组范围内进行单核苷酸多态性(SNP)分析,并运用GenAlEx 6.5、Cervus 3.0、SNPRelate、Plink 1.07、Mega 4.0进行遗传多样性参数统计计算、主成分分析和系统进化树构建。试验获得143个样本在48 116个SNPs位点上的分型结果,其中波尔山羊10个样本单独聚集,与云上黑山羊、努比山羊、云岭黑山羊存在较大的遗传距离,较小的遗传一致度和较小的基因流,在聚类进化树中显示为一个独立分支,这些结果与波尔山羊实际遗传背景完全一致,显示出它在本研究中的对照组作用,同时证明本研究方法可行、结果可靠;云上黑山羊108个样本具有高度一致性,在聚类系统树中这些样本聚为一个大簇,而努比山羊(14个)和云岭黑山羊(11个)的样本各自聚为一簇;UPGMA进化树显示,云上黑山羊与努比山羊关系较近,与云岭黑山羊关系稍远,这与云上黑山羊育种导血方案结果相一致。云上黑山羊新品种在基因组水平上可与其育种素材明显区分,在基因组层面已具备独立品种特点。  相似文献

本试验以贵州省3大地方山羊品种贵州白山羊、贵州黑山羊、黔北麻羊和2个省外地方品种关中奶山羊和内蒙古绒山羊为研究对象,采用DNA池结合PCR直接测序法对山羊前列腺素内过氧化物合酶2(prostaglandin-endoperoxide synthase 2,PTGS2)基因进行单核苷酸多态性检测。结果表明,在5个山羊品种中共检测到6个SNPs位点:A96G、C20T、A239G、T192C、T164A和G91C,其中,A96G和T164A分别位于外显子7和外显子8中,且均为同义突变,其余4个变异位点位于内含子中。生物信息学分析显示,外显子7和8中的2个SNPs位点均导致了mRNA二级结构的改变。  相似文献

A study was conducted in order to examine the possible influence of feeding behaviour on differences in the diversity and the intensity of nematode infections of the gastrointestinal tract between two breeds of goats placed in the same rangeland environment. Within the same flock bred on rangeland conditions, 10 Saanen dairy goats and 14 Angora goats were surveyed for 5 months (from May to September) and parasitological and behavioural parameters were measured. The level of nematode infection was estimated by monthly measurements of faecal egg counts whereas the feeding behaviour was assessed every fortnight. During the whole survey, nematode egg excretion was repeatedly higher in the Angora goats compared to the Saanen ones and significant differences were observed in May and September. This difference in egg excretion was related to major difference in feeding behaviour between the two breeds. In contrast with the Saanen does, Angora goats did not show any browsing and resting activities at any date of observation. Hence, Angora goats can be considered mainly as grazers whereas Saanen goats exhibit feeding activities close to browsers. The relative influence of feeding behaviour and host immune response on the susceptibility to gastrointestinal nematode infection in goats is discussed.  相似文献

【目的】试验旨在分析山羊Z-DNA结合蛋白(Z-DNA binding protein 1,ZBP1)基因3′-端非翻译区(3′-UTR)、内含子1和外显子7区域中单核苷酸多态性(single nucleotide polymorphism, SNP)位点与产羔性能之间的关联性,为高繁殖力山羊分子育种提供新的遗传标记。【方法】选取麻城黑山羊、黑头羊和波尔山羊作为研究样本,采集血样提取DNA,根据转录组数据所选SNP位点在山羊ZBP1基因序列中的位置信息设计引物,利用多重单碱基延伸SNP分型技术(SNaPshot)分析所选SNP位点的遗传多样性,采用一般线性模型(GLM)对ZBP1基因SNP位点与3个品种山羊的产羔性能进行关联分析。【结果】山羊ZBP1基因中存在12个潜在的SNPs位点,其中7个SNPs位点(g.9734 A>G、g.9772 T>C、g.352 C>T、g.955 C>T、g.1880 G>A、g.2566 T>C和g.7919 G>A)具有多态性,除g.9734 A>G在麻城黑山羊群体中仅有AA和GA 2种基因型外,其余...  相似文献
