The effect of ACTH administration on plasma cortisol concentrations in purebred and crossbred pigs was investigated. Pigs were given either 25 IU of ACTH or physiologic saline solution IM. Blood samples were collected immediately before and 1 hour after ACTH or saline solution administration. Administration of ACTH resulted in a significant (P less than 0.01) increase in plasma cortisol concentration compared with that resulting from administration of saline solution; mean values after ACTH administration were similar in both breed groups. In contrast, a fivefold range of differences was observed among individual pigs of the same age, sex, and body weight, irrespective of breed group. The type and magnitude of the adrenocortical response was consistent and repeatable in pigs over a 3-month period, suggesting that pigs have a consistent capacity for adrenocortical response to ACTH administration. Development of a dynamic test allowed the high and low responding extremes in a population to be detected. The most suitable dose of synthetic ACTH was established to be 50 IU, and the best time for blood sample collection was 60 minutes after ACTH administration. The classification of individual pigs as high or low responders was repeatable and was not affected by prior short-term exposure to ACTH.  相似文献

Using an immunocytochemical technique, we have studied in the alpaca brainstem the distribution of immunoreactive structures containing prodynorphin (alpha‐neoendorphin)‐ and pro‐opiomelanocortin (adrenocorticotrophin hormone (18–39) (ACTH), beta‐endorphin (1–27))‐derived peptides. No peptidergic‐immunoreactive cell body was observed. Immunoreactive fibres were widely distributed, although in most of the brainstem nuclei the density of the peptidergic fibres was low or very low. In general, the distribution of the immunoreactive fibres containing the peptides studied was very similar. A close anatomical relationship occurred among the fibres containing alpha‐neoendorphin, ACTH or beta‐endorphin (1–27), suggesting a functional interaction among the three peptides in many of the brainstem nuclei. The number of fibres belonging to the prodynorphin system was higher than that of the pro‐opiomelanocortin system. A moderate/low density of immunoreactive fibres was observed in 65.11% (for alpha‐neoendorphin (1–27)), 18.18% (for ACTH) and 13.95% (for beta‐endorphin) of the brainstem nuclei/tracts. In the alpaca brainstem, a high density of immunoreactive fibres was not observed. The neuroanatomical distribution of the immunoreactive fibres suggests that the peptides studied are involved in auditory, motor, gastric, feeding, vigilance, stress, respiratory and cardiovascular mechanisms, taste response, sleep‐waking cycle and the control of pain transmission.  相似文献

An outbreak of acute, fatal, hemorrhagic pneumonia was observed in more than 1,000 mixed breed dogs in a single animal shelter. The Department of Anatomic Pathology at the University of California at Davis School of Veterinary Medicine performed necropsies on dogs that were found moribund in acute respiratory distress or found dead with evidence of nasal bleeding. All dogs had hemothorax and an acute, fibrinosuppurative pneumonia. Large numbers of gram-positive cocci were observed within the lungs of all dogs and within septic thromboemboli of remote organs in about 50% of cases. Bacterial cultures from the dogs and their environment revealed widespread beta-hemolytic Streptococus equi subspecies zooepidemicus (Lancefield Group C). Extensive diagnostic testing failed to reveal the consistent presence of copathogens in individual cases. The clinical, epidemiologic, molecular biologic, and pathologic data indicate that a single clone of S. zooepidemicus was the cause of an acutely fatal respiratory infection in these dogs.  相似文献

Five trials involving 112 individually-penned animals were conducted to determine the effects of dietary fiber additions (0 or 10% dehydrated alfalfa meal) on the rate and efficiency of growth and carcass characteristics of growing pigs housed in a cold (10 C), thermoneutral (22.5 C) or hot (35 C) thermal environment. Cold exposure (10 C) depressed efficiency of feed and energy utilization, carcass length, backfat, cold carcass dressing percentage and percentage lean cuts. Exposure to the hot environment resulted in depressed live weight gains but did not alter the efficiency of feed utilization. Dietary additions of alfalfa meal depressed daily gains by 1, 3 and 5% in pigs housed in the cold, warm and hot environments, respectively, and gain:feed ratios by 1, 7 and 10%. Ingestion of the fibrous alfalfa meal depressed cold carcass dressing percentage in the 10 and 22 C environments as compared with the basal diet. Based on these data, the nutritional value of fibrous feedstuffs (15% total dietary neutral detergent fiber or less) for growing pigs allowed to consume feed ad libitum is greater in animals housed in a moderately cold vs a warm or hot thermal environment.  相似文献

Acute leukaemia (AL) is a bone marrow malignancy of hematopoietic progenitors that historically is poorly responsive to treatment. With the widespread adoption of dose‐intense chemotherapy, more human patients attain long‐term survivals, but whether comparable progress has been made in canine AL is unknown. To investigate this question, medical records from three academic veterinary hospitals were reviewed. Fifty dogs met the criteria for AL, having excess circulating or marrow blasts, a major cytopenia(s), and no substantial lymphadenopathy. Thirty‐six dogs received cytotoxic chemotherapy; 23 achieved a complete or partial response for a median of 56 days (range, 9–218). With failure or relapse, 14 dogs were rescued. Median survival with treatment was poor at 55 days (range, 1–300). Untreated (n = 6) and palliatively‐treated (n = 8) dogs lived a median of 7.5 days. Most dogs developed chemoresistance within weeks of initiating treatment, and consequently, survival times for AL remain disappointingly short.  相似文献

The plasma cortisol response to exogenous ACTH (ACTH stimulation test) was evaluated in 22 dogs with hyperadrenocorticism caused by adrenocortical neoplasia. The mean basal cortisol concentration (6.3 microgram/dl) was high, but 7 dogs had basal cortisol concentrations that were within normal range. Administration of exogenous ACTH increased the plasma cortisol concentrations in each dog. Normal post-ACTH cortisol concentrations were found in 9 (41%) of the 22 dogs; 13 (59%) had an exaggerated increase in cortisol concentrations after ACTH administration. In 9 of 13 dogs with carcinoma and in 4 of 9 with adenoma, the cortisol response was exaggerated. The mean post-ACTH cortisol concentration in the dogs with carcinoma was approximately 4 times that of the dogs with adenoma; the 7 dogs with the highest concentrations had carcinoma. Repeat studies were performed in 6 dogs 2 to 8 weeks after initial testing. In 5 of the 6 dogs, repeat testing yielded data of similar diagnostic significance. One dog, however, had an abnormally high post-ACTH cortisol concentration at initial evaluation, but had only a minimal response to ACTH administration, with a normal post-ACTH cortisol concentration, at time of resting. Although ACTH stimulation testing is useful in diagnosing hyperadrenocorticism, it can not reliably separate dogs with hyperfunction adrenocortical tumors from clinically normal dogs or from dogs with pituitary-dependent hyperadrenocorticism (bilateral adrenocortical hyperplasia).  相似文献

Optimal ratio of lysine (Lys) to threonine (Thr) in diets for growing barrows [genotype: Piétrain × (Large White × German Landrace)] was established at 30, 50, 70, 90 and 110 kg body weight (BW). N balance studies were conducted with diets based on constant mixture of wheat, barley, soybean meal and field peas. Supplementation of crystalline amino acids (AA) provided diets with Lys (diet A) and Thr (diet B) in first limiting position. For each BW range, totally 28 individual N balance data were utilized for assessing model parameters (exponential N utilization model) of dietary efficiency of Lys and Thr, respectively. Observed maximal dietary efficiency of Lys and Thr provided the database for conclusion of optimal dietary Lys to Thr ratio. Based on applied experimental conditions, BW dependent Lys to Thr ratio could not be concluded. On average, the dietary ratio Lys to Thr = 1:0.61 ± 0.02 was obtained as optimal for growing pigs.  相似文献

Two Aujeszky's disease virus glycoprotein genes, gX and g1, have been used to produce deletion mutants which have then been developed into vaccines. These deletions then allow differentiation between pigs infected with wild type virus and those given the vaccine. It is not clear whether the glycoproteins encoded for by these genes are needed to induce a full protective immune response, in which case deletion mutants would suffer from lack of potency. To test this, commercially available Aujeszky's virus vaccines which lacked either gX or g1 were compared and isogenic constructs were made which differed only in the absence or presence of gX and, or, g1. These constructs and vaccines were used to vaccinate the natural host of Aujeszky's disease, the pig, and potency was measured using challenge with wild type virus. In all cases vaccines which lacked g1 performed significantly less well than those in which g1 was present, whereas deletions of gX had no significant effect on vaccine performance.  相似文献

Background: Endocarditis is a rare heart condition with variable clinical expressions in equids. Risk factors for this disease are incompletely understood. Objective: Describe risk factors for endocarditis in equids. Animals: One hundred and fifty‐three equids admitted to Liège University, 9 diagnosed with endocarditis and 144 free from endocarditis but admitted to the hospital with a differential diagnosis including this disease. Methods: Retrospective case‐control study. Results: Equids with endocarditis were significantly younger (mean age = 4.84 ± 5.74 years) than control equids (mean age = 10.8 ± 7.73 years) (P= .01). No sex or breed predisposition was observed. Animals with hyperthermia (odds ratio [OR] = 24.4; confidence interval [CI] = 1.40–428), synovial distension (OR = 13.4; CI = 3.00–59.8), lameness (OR = 6.52; CI = 1.63–26.1), hyperglobulinemia (OR = 26.4; CI = 3.03–229), hypoalbuminemia (OR = 11.4; CI = 1.34–96.8), hyperfibrinogenemia (OR = 9.81; CI = 1.16–82.7), or leukocytosis (OR = 7.12; CI = 1.40–36.4) presented a significantly higher risk of having endocarditis than control horses. The presence of two of the clinical signs mentioned above significantly increased the probability of a diagnosis of endocarditis (P≤ .05). Conclusions and Clinical Importance: Age is associated with equine endocarditis. The diagnostic value of certain clinical signs and abnormalities in blood parameters in this disease are described.  相似文献

Prenatal stress is known to affect several offspring characteristics, but its effects depend among other factors on the period of gestation in which it is applied. In the present study, oral administration of hydrocortisone-acetate (HCA) was used to elevate cortisol concentrations in pregnant sows to levels also observed after psychological stress. HCA was administered during three different periods of gestation (115 days in pigs): period 1: 21-50 (P1, n = 10), period 2: 51-80 (P2, n = 10) and period 3: 81-110 (P3, n = 10) days after insemination. Control sows (n = 11) received vehicle from 21-110 days after insemination. When P1-, P2- and P3-sows did not receive HCA, they also received vehicle. During gestation, weekly saliva samples were taken from the sows to determine salivary cortisol concentrations. Treatment effects on sow, litter and piglet characteristics were determined. In addition, two female piglets per litter were subjected to an ACTH-challenge test at 6 weeks of age to determine the adrenocortical response to ACTH. Pigs were slaughtered at 6 months of age and slaughter weight, back fat thickness and percentage of lean meat were analysed. During the period of treatment with HCA, salivary cortisol concentrations were increased in P1-, P2- and P3-sows compared to control sows (P < 0.01). The total number of piglets born per litter did not differ among treatment groups (P > 0.30), but pooled HCA-litters had a higher percentage of live born piglets (P < 0.05) and fewer mummies than control litters (P < 0.05). Gestation length did not differ among treatment groups (P = 0.21), but did affect treatment effects on birth weight. Overall, HCA-piglets weighed less at birth, and remained lighter until weaning (P < 0.05). The salivary cortisol concentrations after i.m. injection of ACTH (2.5 IU/kg) were lower in P1- and P3-piglets compared to control piglets. At slaughter, HCA-treatment indirectly decreased lean meat percentage and increased back fat thickness. In conclusion, elevated peripheral cortisol concentrations in pregnant sows affect both litter characteristics and piglet physiology, the latter depending on the period of gestation during which concentrations were elevated. Using an indirect immunoperoxidase technique, an in depth study has been carried out for the first time on the distribution of fibres and cell bodies containing neurotensin and somatostatin‐28 (1–12) (SOM) in the minipig brainstem. The animals used were not treated with colchicine. The distribution of neurotensin‐ and SOM‐immunoreactive fibres was seen to be quite similar and was moderate in the minipig brainstem: a close anatomical relationship between both neuropeptides was observed. The distribution of cell bodies containing neurotensin or SOM was quite different and restricted. Cell bodies containing neurotensin were found in four brainstem nuclei: nucleus centralis raphae, nucleus dorsalis raphae, in the pars centralis of the nucleus tractus spinalis nervi trigemini and in the nucleus ventralis raphae. Cell bodies containing SOM were found in six nuclei/regions of the brainstem: nucleus ambiguus, nucleus dorsalis motorius nervi vagus, formatio reticularis, nucleus parabrachialis medialis, nucleus reticularis lateralis and nucleus ventralis raphae. According to the observed anatomical distribution of the immunoreactive structures containing neurotensin or SOM, the peptides could be involved in sleep‐waking, nociceptive, gustatory, motor, respiratory and autonomic mechanisms.  相似文献
