坏死性肠炎(necrotic enteritis,NE)是家禽中最重要的肠道疫病之一,呈世界性流行,严重危害养鸡业发展。人工复制鸡坏死性肠炎病例是研究该病发病机制及筛选有效药物的重要手段,然而在实际试验过程中,通常受实验动物、感染菌株、诱导病原等多方面因素影响而难以成功复制病例。鉴于此,现综述了影响鸡坏死性肠炎病例成功复制的关键因素,包括攻毒使用的实验动物、菌株毒素、菌株培养条件、诱导病因等,同时探讨了如何通过调整这些因素来改善人工复制鸡坏死性肠炎病例的严重程度,分析了鸡坏死性肠炎病例的病变评分系统,以期为鸡坏死性肠炎的实验室研究及综合防控提供理论基础。 相似文献

The objective of this study was to investigate the individual and combined effects of essential oils (EO; comprised of thymol and carvacrol) and lysozyme on experimental NE in broiler chickens. A total of 320 1-day-old chicks were randomly assigned to five treatment groups: no-challenge control (NC), NC + C. perfringens challenge (CC), CC + 120 mg/kg of EO, CC + 100 mg/kg of lysozyme, and CC + 120 mg/kg of EO + 100 mg/kg of lysozyme. The results showed that EO or lysozyme decreased the mortality, alleviated the gut lesions, inhibited the liver Enterobacteriaceae carriage, and increased the villus height of the ileum compared with CC ( p < .05), although the proliferation of C. perfringens in the ileum was not inhibited ( p > .05). Moreover, EO or lysozyme was found to decrease the ileal concentration of sialic acid and the Mucin2 mRNA expression ( p < .05). However, the blend of EO and lysozyme did not display significant effect on the NE-associated mortality or gut damage in contrast with CC ( p > .05). In conclusion, these findings suggest the similar protective effects of EO and lysozyme in NE-associated mortality and intestinal impairment, but their blend did not exhibit ameliorative effect. 相似文献

选择具有显著抗炎效果的甘草总黄酮为受试化合物,考察其抗产气荚膜梭菌所致鸡坏死性肠炎的效果,为甘草总黄酮在坏死性肠炎的治疗应用提供依据。选择AA^+肉鸡,经口给予产气荚膜梭菌ATCC13124菌悬液建立肉仔鸡坏死性肠炎模型,之后使用甘草总黄酮对造模成功的肉鸡进行治疗,通过剖检考察肉鸡肠道的病变情况、病变评分、日增重及器官指数等指标以评估甘草总黄酮对坏死性肠炎的治疗效果。结果显示,通过经口攻菌成功建立了肉仔鸡坏死性肠炎模型;剖检发现,经甘草总黄酮治疗的模型鸡其肠道病变程度明显缓解,病变损伤评分显著低于感染组,炎性因子含量显著降低;治疗组肉鸡日增质量及器官指数与健康肉鸡差异不显著。结果表明,甘草总黄酮能够缓解产气荚膜梭菌所致坏死性肠炎对肉鸡肠道的损伤,具有作为抗生素替代品治疗坏死性肠炎的潜力。 相似文献

The objective of this study was to assess the effects of a probiotic strain Clostridium butyricumMIYAIRI 588 (CBM588) on broiler and weaned piglet health and zootechnical performance. Five field studies were carried out in broilers and five in weaned piglets under European feed additive guidelines. Each study followed a randomized blocked design with two treatments: Control (basal diet) and CBM588 supplemented groups. The zootechnical performance parameters selected were body weight, daily gain, feed intake and feed efficiency (feed:gain). Broilers fed diets with CBM588 gained significantly more weight (+2%, p < .001) and exhibited significantly better feed efficiency (?1.6%, p < .001) in comparison with Controls. Similarly, analysis of pooled data of weaned piglet trials showed that CBM588‐fed piglets were significantly heavier than Controls (+2.6%, p = .014), exhibited significantly higher mean daily gain (+4.7%; p = .004), and significantly improved feed efficiency (?4.2%, p = .001). In addition to the zootechnical efficacy studies, the preventive effect of CBM588 on necrotic enteritis (NE) was assessed in a natural challenge model in broilers where CBM588 reduced the incidence and severity of NE lesions. These data indicate the potential of CBM588 to improve broiler and weaned piglet zootechnical performance, and to make a positive contribution to animal health. 相似文献

A vaccine for necrotic enteritis (NE) of chickens would reduce the current need to prevent or treat the disease in broiler chickens with antimicrobial drugs. The objective of this study was to understand aspects of immunity to the disease. The first experiment examined the virulence of six strains of Clostridium perfringens isolated from cases of NE in broiler chickens. Using a 5-day experimental oral infection of 2-week-old broiler chickens, four of the six strains were found to be virulent. Pulsed-field gel electrophoresis and PCR showed that virulence was not associated with a plasmid encoding the beta2 toxin gene, cpb2, since this was present in virulent and one of the two avirulent strains. In the second experiment, two virulent and one avirulent strains were tested for their ability to immunize ("infection-immunization") chickens through the oral route. The procedure used experimental infection for 5 days followed by bacitracin treatment for 9 days, and then re-challenge 2 days later with a virulent strain, CP4. Infection-immunization with the virulent isolates protected chickens from subsequent virulent challenge, whereas the infection-immunization with the avirulent isolate did not. In a third experiment, two of four alpha-toxin-negative mutants of CP4 protected birds from experimental NE after oral immunization. These two mutants were also attenuated for virulence. We conclude that it is possible to immunize chickens successfully against NE and that immunogen(s) other than alpha-toxin are important in protective immunity against oral infection. 相似文献

1. This study investigated the prebiotic properties of arabinoxylo-oligosaccharides (AXOS) produced both in situ and in vitro for their activity against the onset of necrotic enteritis in broiler chickens. 2. A 2 × 3 factorial arrangement was applied, including necrotic enteritis challenge (challenged/unchallenged) and three dietary treatments from d 10 to 21. A wheat–soy commercial-type basal-grower diet was fed with 2% of the wheat proportion replaced by the same amount of either arabinoxylan (AX), AXOS produced from hydrolysing AX with 16 000 BXU (birch xylanase unit) xylanase in vitro or AX fed with 16 000 BXU xylanase (AX + E). Necrotic enteritis (NE) challenge was induced by orally infecting birds with a vaccine strain of Eimeria oocysts at d 9 of age followed by oral gavage of a freshly prepared Clostridium perfringens broth at d 14. 3. The challenge depressed growth performance, induced gross lesions and reduced ileal viscosity at d 10–21. Birds fed on the AXOS diet had numerically less severe gross lesions, improved feed conversion at d 0–16 and lower ileal viscosity at d 16 compared to birds fed on AX. Weight gain of the unchallenged birds ranked as follows in terms of the diets: AXOS > AX + E > AX. AX + E produced a lower ileal viscosity compared to the AX treatment but only led to marginal improvements in performance and intestinal lesion scores. 4. Caecal short-chain fatty acid (SCFA) concentration was higher in birds fed on AXOS and AX + E compared to those fed on AX and was higher in the challenged birds compared to the unchallenged birds. Gizzard pH was lower in birds fed on AX + E compared to those fed on AXOS at d 16. Challenged birds had lower ileum pH compared to the unchallenged birds at d 16 and 21. 5. Results of this study suggest that AXOS appeared to be efficacious prebiotics, as highlighted by improvements in feed conversion ratio and increased SCFA production. Future studies are warranted to elucidate the types of AXOS that are most active against NE and the mechanisms by which different levels of AXOS enhance bird performance. 相似文献

Plant extracts(PE)are gaining increased attention as potential alternatives to in-feed antimicrobials(AM)due to their known antimicrobial activities.This study was conducted to examine the potential of PE,a microencapsulated product composed of eugenol and garlic tincture as an alternative to AM-agent on performance and intestinal health in broilers under necrotic enteritis(NE)challenge.A total of 960 dayold mixed-sex Cobb 500 chicks were randomly distributed to 48-floor pens with 6 treatments replicated 8 times with 20 birds each.The 6 treatments were as follows:UC,unchallenged control;CC,challenged control;PE,challenged group plus PE;AM,challenged group plus AM;FAP,challenged group plus a full dose of AM with PE;HAP,challenged group plus a half dose of AM with PE in starter,grower and finisher phases.Birds in the challenged groups were inoculated with Eimeria spp.on d 9 and Clostridium perfringens on d 14.The body weight gain(BWG),feed intake(FI),feed conversion ratio(FCR),and livability of birds were compromised,and intestinal lesions and mortality were increased(P<0.05)by NE challenge,illustrating a successful clinical NE challenge.Birds fed AM had higher BWG and FI,and lower FCR,mortality,and intestinal lesions compared to the CC group(P<0.05).Birds fed PE had improved FCR(P<0.05)and livability(5.8%)in an overall period compared to the CC group.On d 16,PE supplementation reduced ileal lesion scores in only male birds(P<0.05).Birds fed PE had decreased Eimeria maxima and Eimeria acervulina oocyst counts in caecal content(P<0.05).Birds fed PE had decreased Eimeria brunetti and total oocyst counts in caecal content,and E.acervulina oocyst counts in ileal content in only female birds(P<0.05).On d 35,PE supplementation reduced variation of BW in both male and female birds and increased yellowness(b*value,14.4%)in the thigh.These findings suggest the potential of PE supplementation in diets to improve the performance and intestinal health of birds under clinical NE as indicated by improved FCR,livability,uniformity,reduced ileal lesions,oocyst counts and increased skin yellowness.However,the protective effect of PE may not be apparent in the presence of AM in the feed. 相似文献

The present study examines the responses of broiler chickens to oral administration of Clostridium perfringens freshly isolated from field cases of necrotic enteritis (NE). The challenge studies included long-term exposure and short-term exposure, factored in with dietary and management variables including high levels of dietary components such as fish meal, meat meal, abrupt change of feed, and fasting. In the long-term exposure trials, the birds were orally inoculated daily, with 1 ml (1.0 or 2 x 10(8) CFU/ml) of an overnight culture of C. perfringens for 7 days. Short-term exposure trials involved challenge with 1 ml (3 x 10(10) CFU/ml) administered as a single dose. The responses of broilers to orally administered C. perfingens under laboratory controlled conditions are presented and discussed in the context of authentic field cases of necrotic enteritis. None of the challenge trials produced overt clinical signs of NE and there were no mortalities associated with oral exposure to high doses of C. perfringens. However, many of the challenged birds showed distinctly pronounced pathological changes in the intestinal tissue. On gross examination the responses in birds challenged orally with C. perfringens could be placed into two categories: (1) no apparent pathological changes in the intestinal tissue and (2) sub-clinical inflammatory responses with focal, multi-focal, locally extensive, or disseminated distribution throughout various sections of duodenum, jejunum, ileum, and ceca. In birds that responded with intestinal lesions, hyperemia and occasional hemorrhages were the main gross changes. In some birds, the mucosa was covered with a brownish material, but typically, the mucosa was lined by yellow or greenish, loosely adherent material. Mild gross changes were seen in some control birds, but both qualitatively and quantitatively, the lesions were distinctly more pronounced in the challenged birds. Upon histological examination, none of the experimentally exposed birds showed overt mucosal necrosis typical of field cases of NE, but typically the lamina propria was hyperemic and infiltrated with numerous inflammatory cells. Most significant changes were seen at the interface of the basal domain of enterocytes and lamina propria. Multifocally, these areas were extensively edematous, allowing for the substantial disturbance of the structural integrity between the lamina propria and the enterocytes. The lesions observed in the present study were consistently reproduced in all of our challenge trials, hence these responses may signify newly emerging patterns of sub-clinical enteric disorders in contemporary strains of poultry. The pathological changes observed in broilers challenged orally with C. perfringens in the present study, differ significantly from those reported previously, and must be clearly differentiated from those described in cases of NE or ulcerative enteritis. Although no overt necrosis of the intestinal mucosa typical of field cases of NE were observed in the present study, the birds challenged with C. perfringens showed strong inflammatory reaction to the introduced pathogens. The distinct features of the microscopic lesions were changes involving apparently normal enterocytes at the interface of the basal domain of villar epithelia and lamina propria. Although the pathological changes in the intestinal tissues observed in our trials appear to be rather subtle when compared to field cases of NE, the nature of these lesions suggest a significant negative effect on the digestive physiology of intestinal mucosa. 相似文献

Poultry production has undergone a substantial increase compared to the livestock industries since 1970.However, the industry worldwide is now facing challenges with the removal of in-feed antibiotics completely or gradually, as the once well-controlled poultry diseases have re-emerged to cause tremendous loss of production. Necrotic enteritis(NE) is one of the most important diseases which costs the industry over two billion dollars annually. In this paper, we review the progress on the etiology of NE and its control through dietary modifications, pre-and probiotics, short chain fatty acids, and vaccination. The other likely measures resulted in the most advances in the toxin characterization are also discussed. Vaccine strategies may have greater potential for the control of NE mainly due to clearer etiology of NE having been elucidated in recent years with the identification of necrotic enteritis toxin B-like(NetB) toxin. Therefore, the use of alternatives to in-feed antibiotics with a better understanding of the relationship between nutrition and NE, and limiting exposure to infectious agents through biosecurity and vaccination, might be a tool to reduce the incidence of NE and to improve gut health in the absence of in-feed antibiotics. More importantly, the combinations of different measures may achieve greater protection of birds against the disease. Among all the alternatives investigated, prebiotics, organic acids and vaccination have shown improved gastrointestinal health and thus, have potential for the control of NE. 相似文献

Considering market demands concerning the decreased use of growth promoters and anticoccidial drugs in feed formulations, the poultry industry has been trying to reduce or eliminate the inclusion of subtherapeutic doses of antimicrobials into feed. Formulating diets not only to meet birds’ nutrient requirements for growth but also for gastrointestinal health parameters is increasingly important. Maintenance and enhancement of intestinal integrity is essential for bird performance when antimicrobials are not included in feed, as commercial poultry face numerous enteric pathogen challenges. Necrotic enteritis has reemerged as an important disease of poultry in recent years. The reduction in the use of antimicrobials in poultry feeds has been attributed as one of the main contributing factors for the increasing incidence of necrotic enteritis (NE) in commercial poultry. Mortality due to NE is extremely high (1% daily mortality), which results in great economic losses. Economic losses due to NE are not only associated with high mortality, but also associated with decreases in bird performance and FE, particularly in subclinical cases of NE. Birds that survive NE outbreaks usually have a reduced ability to digest and absorb nutrients due to extensive damage to the mucosal lining, which ultimately results in reduced profitability. 相似文献

The efficacy of narasin in the control of necrotic enteritis (NE) was investigated in a floor pen study of 2000 broiler chickens using a Clostridium perfringens feed inoculum challenge model. Treatments were 1) nonmedicated, nonchallenged; 2) nonmedicated, challenged; 3) narasin, nonchallenged; 4) narasin, challenged. Narasin was administered at 70 ppm in the feed from day 0 to trial termination on day 41. Challenge inoculum contained approximately 1 x 10(8) colony-forming units CP/ml and was administered from day 14 to day 16. In the unmedicated groups, challenged birds had significantly (P < 0.05) lower mean body weight and reduced feed efficiency at day 21 and significantly (P < 0.01) higher cumulative NE mortality at day 41 compared with unchallenged. Similarly, among unmedicated birds, those challenged had a significantly (P < 0.01) higher mean NE score on day 17 and significantly (P < 0.05) higher mean huddling scores on days 15-17 than unchallenged. Among challenged birds, those fed narasin had significantly (P < 0.05) higher mean body weight and improved feed efficiency at days 21 and 41 and significantly (P < 0.01) lower cumulative NE mortality at day 41 than unmedicated. Similarly, among challenged birds, those receiving narasin had a lower mean NE score on day 17 (P > 0.05) and significantly (P < 0.05) lower huddling scores on days 16 and 17 than unmedicated. Coccidiosis lesion scores were zero for birds euthanatized from all treatment groups on day 17, suggesting that the beneficial effects of narasin were not due to prevention of coccidiosis. This study thus provides evidence that narasin is effective in the prevention of necrotic enteritis in broiler chickens. 相似文献
