Heat shock proteins (HSPs) are a group of highly conserved molecular chaperones. Shrimp HSPs have recently been a topic of increasing interest because of their roles in shrimp immunity and homeostasis. In penaeid shrimp, HSP70s and the cognate forms, heat shock cognate (HSC) 70s, have been reported, but their responses towards various stimulations are different. We found a novel type HSP70 (MjHSP70-2) from the hyperexpansion of the large segmental duplication that is present in kuruma shrimp Marsupenaeus japonicus, which shows about 60 % identity with reported shrimp HSP70s. In a phylogenetic tree, MjHSP70-2 formed a sister clade with eukaryote HSP70 family while MjHSP70 was located close to the shrimp HSP70 and HSC70 group. MjHSP70-2 gene expression was not significantly increased by heat shock or pathogen challenge by Vibrio penaeicida, but it was significantly increased by infection with white spot syndrome virus. In contrast, MjHSP70 gene expression was increased by heat shock but decreased by infection with V. penaeicida. The kuruma shrimp genome was found to have 400-fold more copies of the MjHSP70-2 gene than the putative single-copy gene transglutaminase. In conclusion, our results reveal the presence of a novel type HSP70 gene, HSP70-2, from kuruma shrimp. There are multiple forms of HSP70 in crustaceans, and these HSP70s behave differently under various stressors.  相似文献

The effects of fulvic acid (FA) on survival and immune‐related gene expression were investigated in Litopenaeus vannamei challenged with Vibrio parahaemolyticus by immersion. Shrimp were fed with different dietary FA concentrations (1, 2, 4 and 6 g/kg feed) for 20 days (first bioassay) or 8 days (second bioassay, 2 g/kg feed of FA added every 2 days) and then challenged with V. parahaemolyticus. In a third bioassay, the expression of three immune‐related genes (translationally controlled tumour protein [TCTP], superoxide dismutase [SOD] and heat‐shock protein 70 [HSP70]) in haemocytes or hepatopancreas of experimental shrimp was measured by real‐time quantitative PCR at 0, 6, 12, 24, 48, 72 and 96 hr after FA (2 g/kg feed) administration. Fulvic acid increased survival at a concentration of 2 g/kg feed supplied every two days. Interestingly, TCTP gene expression was upregulated, whereas gene expression of SOD and HSP70 was downregulated. In conclusion, dietary fulvic acid improves survival in white shrimp challenged with V. parahaemolyticus and modulates the immune response. Therefore, FA merits further evaluation as prophylactic treatment in commercial shrimp farms.  相似文献

葡萄糖调节蛋白78(glucose regulated protein 78 ku,GRP78)是热休克蛋白70家族成员之一,在调节蛋白质折叠和维持内质网稳态过程中起着分子伴 侣作用。采用RT-PCR、RACE等技术,首次从拟穴青蟹获得了GRP78的cDNA全长序列。该序列全长2 284 bp,开放阅读框(ORF)为1 962 bp,编码653个氨基酸残基。 同源分析显示,该基因编码的蛋白含有HSP70家族的签名序列,C末端为内质网蛋白滞留信号KDEL,与其他物种具有很高相似性。实时荧光定量PCR结果表明,GRP78 基因在拟穴青蟹多个组织中均有表达。第一期仔蟹在不同的温度和盐度下暴露12 h后,GRP78基因表达量随环境温度升高而增加;在高盐(30)条件下GRP78表达量 较高,进而推测拟穴青蟹GRP78参与蛋白质折叠和环境胁迫的应答。  相似文献

采用RT-PCR方法扩增实验动物剑尾鱼(Xiphopohorus helleri)纯系RR-B的HSP70家族两个成员的eDNA片段,并将其克隆到PMD18-T载体中测序,将测序结果与GenBank中的核苷酸序列和推导的氨基酸序列进行同源性比较。同时利用RT-PCR半定量方法研究热应激时两个成员在剑尾鱼组织中的表达。通过克隆获得了剑尾鱼的HSP70家族两个新成员的cDNA片段,两序列均包含HSP70家族的特征性签名序列和热休克蛋白的定位序列;通过对克隆片段与已发表的青锵(Oryzias latipes)等鱼类HSP70的核苷酸序列和其编码的氨基酸序列同源性比较,发现核苷酸序列同源性较高,成员一和成员二分别为85%-89%和82%-99%;氨基酸序列同源性更高,成员一和成员二分别为97%-99%和93%-99%。一般条件下,两成员在剑尾鱼肝脏、脾脏、肾脏和心脏中不表达,但热应激能刺激成员一在剑尾鱼脾脏中表达,成员二在肝脏、脾脏、肾脏和心脏中强烈表达,并可能在参与热应激保护方面起更大作用。  相似文献

合浦珠母贝热休克蛋白hsp70基因的克隆与表达分析   总被引:10,自引:2,他引:8  
采用同源克隆和RT-PCR技术对合浦珠母贝(Pinctada fucata)热休克蛋白hsp70基因进行了克隆和表达分析。获得cDNA全长序列2 365 bp,其中3’非编码区域(UTR)为318 bp,5’UTR为88 bp,开放阅读框(ORF)为1 959 bp,编码652个氨基酸,分子量约为71.39 kD,理论等电点为5.22,并含有3个HSP70家族的签名序列IDLGTTYS、DLGGGTFD和EEVD。同源性分析表明,合浦珠母贝HSP70的氨基酸序列与太平洋牡蛎(Crassostrea gigas)等双壳贝类的相似性高达86%以上,基于氨基酸序列的聚类分析表明,合浦珠母贝与牡蛎属种类亲缘关系最近。高温、高盐刺激后,半定量RT-PCR检测发现hsp70基因的表达明显增加,高温刺激的表达量高于高盐刺激,高温刺激组不同组织的表达量由大到小依次为鳃、消化腺、外套膜、肌肉、性腺,高盐刺激组不同组织的表达量由大到小依次为鳃、外套膜、肌肉、消化腺、性腺,表明HSP70参与了机体对刺激的应答过程。该基因的克隆为进一步深入研究合浦珠母贝的抗逆机理及其遗传改良奠定了重要基础。  相似文献

This study was conducted to evaluate the effects of fulvic acid (0, 0.3, 0.6, 0.9, and 1.2%) as feed additive on growth, feed utilization, antioxidant ability, and HSP70 in hemolymph and hepatopancreas of juvenile white shrimp Litopenaeus vannamei (average weight 2.5 g) reared under experiment conditions. Shrimp were stocked at a density of 625 shrimps m?3 for 60 days in net cages submerged in recirculating tanks. At the end of the experiment, specific growth rates and survival rates of shrimp in treatment groups fed with 0.6, 0.9, and 1.2% fulvic acid were higher compared to that of the control group. Shrimp fed 0.6, 0.9, and 1.2% fulvic acid had significantly lower feed conversion rates than those fed control diet. The optimum dietary fulvic acid requirement for juvenile shrimp based on weight gain was 0.897%. Furthermore, superoxide dismutase activity and peroxidase activity increased significantly, while malonaldehyde content decreased in the hemolymph and hepatopancreas of shrimp fed 0.9 and 1.2% dietary fulvic acid. Glutathione content increased obviously in hemolymph of shrimp fed 0.6, 0.9, and 1.2% fulvic acid. In hepatopancreas, glutathione content was significantly higher in shrimp supplemented with 1.2% fulvic acid. HSP70 decreased obviously in hemolymph of shrimp fed 0.9 and 1.2% fulvic acid, while shrimp fed with 0.6 and 0.9% fulvic acid showed lower HSP70 level in hepatopancreas. The results of this study demonstrated that dietary fulvic acid could improve survival rates, growth, feed utilization, antioxidant capability, and stress resistance of juvenile L. vannamei reared under intensive stocking conditions.  相似文献

A feeding trial was conducted to study the effect of dietary lipid on growth performance and heat‐shock protein (HSP70 and HSP60) response of white seabass (WSB), Atractoscion nobilis. Five diets were formulated to contain 440 g kg?1 protein from 300 g kg?1 fish meal, 240 g kg?1 soybean meal and 100 g kg?1 soy protein concentrate with different levels of lipid: 100, 120, 140, 160 or 180 g kg?1. At the end of the trial, heat shock response based on HSP70 and HSP60 was measured in liver and white muscle from fish at ambient temperature and temperature shock conditions. Final weight and percent gain were significantly higher for fish fed the 100 g kg?1 lipid diet than for fish fed the rest of the diets (P ≤ 0.05). Feed conversion ratio was lowest for fish fed the 100 g kg?1 lipid diet. The HSP70 and HSP60 responses were positively correlated to dietary lipid levels following temperature shock. At ambient temperature, HSP60 and HSP70 responses in muscle and HSP60 response in liver increased with dietary lipid level. Temperature shock significantly increased the HSP response of fish in all treatments. Results of this study demonstrated that a moderate (110–120 g kg?1) level of dietary lipids would be recommended for production diets but a higher dietary lipid level may be required for optimal stress tolerance.  相似文献

In this study, we exposed black sea bream, Mylio macrocephalus (Basilewsky), fibroblast (BSF) and silver sea bream, Sparus sarba Forsskål, fibroblast (SSF) cell lines to a recombinant Vibrio harveyi haemolysin (VHH) and investigated mechanisms involved in apoptosis. A decrease in mitochondrial membrane potential, followed by an increase in caspase 3 activity, occurred within 2–8 h of VHH exposure, in both cell lines; however, VHH did not alter cellular levels of reactive oxygen species. As heat shock protein 70 (HSP70) is known to prevent the onset of apoptosis in certain mammalian cells, we aimed to test whether such a protective effect is operative in VHH‐exposed fibroblasts. The amounts of HSP70 were elevated in SSF and BSF via an acute heat shock or an acute heat shock followed by a 6 h recovery. It was found that the VHH‐mediated reduction in mitochondrial membrane potential was suppressed in cells that had a 6 h post‐heat shock recovery, and the protective effect of heat shock‐induced HSP70 was attenuated following treatment of cells with the HSP70 inhibitor, quercetin. This study demonstrates how haemolysin causes cell death via induction of apoptosis and provides evidence as to the role of HSP70 as an anti‐apoptotic factor.  相似文献

This study was conducted to determine the effects of dietary α‐ketoglutarate (AKG) supplementation on the antioxidant defense system and gene expression of heat shock protein (HSP) 70 and HSP 90 in hybrid sturgeons Acipenser schrenckii ♀ × A. baerii ♂ exposed to ammonia‐N stress. A 2 × 3 factorial experiment was arranged, in which each diet (0%, 1% AKG) was randomly assigned to 0.25 (control) 5 and 10 mg L?1 ammonia‐N groups with three replicate aquaria for each 72 h. The 10 mg L?1 ammonia‐N significantly increased serum ammonia concentrations and intestinal Gln concentrations and GS activity compared with the 0.25 or 5 mg L?1 ammonia‐N groups. The intestinal Gln concentration and GS activity increased, and the serum ammonia concentration decreased, in fish given dietary supplementation of 1.0% AKG compared with fish given diets without AKG. Superoxide dismutase (SOD) and glutathione peroxidase (GPx) activity in serum, gills and intestines decreased when fish were exposed to 5 or 10 mg L?1 ammonia‐N, and their activity increased in fish given diets with 1% AKG. Catalase in the serum and gills decreased when fish were exposed to 5 or 10 mg L?1 ammonia‐N and increased in fish given diets with 1% AKG. The 10 mg L?1 ammonia‐N or 1% AKG supplementation increased HSP 70 and HSP 90 gene expression in the liver. The increased activity of antioxidant enzymes, and increased HSP 70 and HSP 90 gene expression in fish fed diets containing 1% AKG suggested higher tolerance to ammonia‐N stress.  相似文献

Growth and digestibility trials were conducted to evaluate the use of flash dried yeast (FDY) as a supplement in practical shrimp feeds. The six‐week growth trial evaluated the effects of FDY supplementation (0, 10, 20, 40 and 60 g kg–1) in the plant‐based practical diet on the performances and proximate compositions of juvenile Pacific white shrimp, Litopenaeus vannamei. Final biomass, final mean weight, per cent weight gain (WG), feed conversion (FCR) and protein retention efficiency (PRE) of Pacific white shrimp were not significantly influenced when FDY was added up to 40 g kg–1 of the diet. However, 60 g kg–1 FDY supplementation significantly reduced growth, feed utilization and protein retention. Supplementation of FDY in the practical diets of Pacific white shrimp did not affect protein, moisture, lipid, crude fibre, and ash content of whole shrimp body. Apparent digestibility coefficients of energy, protein, and amino acid (AA) for FDY were determined using chromic oxide as an inert marker and the 70:30 replacement technique. The energy and protein digestibility of FDY were 38.20% and 53.47%, respectively, which were significantly lower than fishmeal (FM) and soybean meal (SBM) that were run at the same time. Most amino acid digestibility of FDY was lowest among the three ingredients tested. Results of this work indicated that energy, protein and amino acid digestibility of FDY are lower compared to SBM and FM, and high levels supplementation (≥60 g kg–1) of FDY can cause negative effects to the growth response of Pacific white shrimp. Based on these results, further research regarding the effects of the low level inclusion of FDY in practical diets on immune responses of Pacific white shrimp is warranted.  相似文献

Members of heat shock protein 70 (HSP70) are highly conserved proteins of about 70 kDa and play important roles in protein folding. Levels of these proteins increase when cells are under stress. Environmental temperature influences both the basal and induced levels of HSPs. However, studies on HSPs in fishes from a tropical country such as India are lacking. In the present study, Indian major carp (IMC) Cirrhinus mrigala (Ham.) acclimatized at 25±2°C had high levels of HSP70, viz., 1.2–1.3 ng μg?1 total protein in kidney and gill and 4.2–5.3 ng μg?1 total protein in liver and brain tissues, indicating the presence of biochemically significant levels of stress. However, maintenance of acclimatized fish at 17°C for up to 48 h did not lead to a significant decrease in stress protein levels. A heat shock at 37°C for up to 48 h resulted in only two to threefold increase in HSP70 levels in these organs. Although the increase in HSP70 levels was apparent from the first hour of heat stress in all these tissues, the increase was significant from the second hour in the brain, the sixth hour in liver and kidney and the 20th hour in the gills.  相似文献
