Three of six ponies, 9 and 10 years of age, were each given in excess of 300 000 infective Trichonema larvae experimentally. Although all six ponies had both natural and experimental infections previously, they were virtually parasite-free during the 18-month period immediately preceding experimental reinfection.Appreciable resistance to Trichonema infections was indicated in these mature ponies by a longer prepatency and significantly smaller worm egg outputs than that which occurred when these animals were given comparable infections at a much younger age. Patent infections only developed in two of the three ponies beginning between 17 and 18 weeks post-reinfection with peak egg counts between 200 and 400 eggs per gram (epg) in one and 100 to 150 epg in the other. Within 10 days of reinfection, an abrupt rise in mean circulating eosinophils and an appreciable mean weight loss, which persisted for 2 years, occured in the reinfected but not the control ponies.The findings, particularly of unthriftiness characterized by persistent loss of weight in the reinfected but not the control ponies, demonstrated the pathogenesis of inhibited Trichonema larvae.  相似文献

A group of seven ponies naturally infected with large numbers of small strongyles and raised under conditions to minimize reinfection were treated periodically over a three year span with thiabendazole at the rate of 44 mg/kg body weight. Based on the absence of worm eggs in the feces following each treatment, thiabendazole removed the adult strongyles present with a new population subsequently developing by maturation of inhibited larvae. It took as many as four or five treatments to eliminate or reduce significantly the worm burdens present in the ponies under the conditions of this study. Strongyle eggs started to reappear in the feces about six weeks after treatment and following the first treatment the mean egg counts rose to the pretreatment level. On successive treatments the interval for worm eggs to appear in the feces lengthened and mean egg counts never rose quite as high as immediate pretreatment levels. Hematological changes were not marked, although a small steady increase in the mean hemoglobin values and an equivalent small decrease in the mean eosinophil counts occurred in all ponies following each successive treatment. The study supports the rationale of regular anthelmintic treatment of horses in that even in the absence of reinfection, new burdens of adult worms develop following treatment.  相似文献

A clinical trial was conducted with 2- to 3-year-old ponies to determine the effectiveness against strongyles of 2.64 mg pyrantel tartrate/kg body weight administered in the daily ration throughout a pasture season in Canada. Ten ponies were given the anthelmintic, and 10 were not treated and served as controls. Each group of ponies was on a separate pasture. The mean strongyle eggs/g of feces for each group, before treatment and turnout to pasture, was greater than 2,200. Thereafter, the mean eggs/g feces for the untreated group remained high (1,405−2,294) and those for the treated group decreased markedly to, and remained at, very low levels (0.2−16.8). Strongyle larval counts for the pasture with the untreated ponies rose to 26,790 larvae/kg dry herbage in August, whereas that for the treated group was 610 larvae/kg dry herbage. At the end of the season, two ponies from each group were isolated for 6 weeks and necropsied. The mean number of strongyles in the untreated and treated ponies was 69,288 and 8,452.5, respectively. In the untreated ponies, 21 species of strongyles were found, and approximately 84% of them were from eight species of cyathostomes. In the treated ponies, 14 species of strongyles were found, and approximately 77% were from one species, Cylicostephanus minutus (52.0%), and immature cythostomes (25%). Adverse reactions were not observed in any of the treated ponies. Pyrantel tartrate was highly effective in significantly reducing the strongyle egg and pasture larval counts and the transmission of strongyles during a pasture season.  相似文献

Patent infections with Strongylus vulgaris were established in 6 of 8 helminth-free ponies given 41 to 101 adult worms via nasogastric tube. The parasites were removed from the cecum and ventral colon and transferred within 1 to 2 hours of the death of the donor horses. Eggs were found in the feces of the recipients in 2 or 3 days; egg counts reached maximum, 28 eggs per gram of feces, at 4 weeks after ponies were inoculated. In 6 ponies euthanatized 3 to 7 weeks after parasitic transfers were done, 28% of the inoculated worms were found alive at necropsy. A 7th pony was maintained as a donor for establishing infections for chemotherapy trials and, although never passing more than 6 eggs per gram of feces, shed infective larvae over a period of 2 years.  相似文献

Over a period of 13 months, faecal samples were collected monthly from approximately 45 cattle over 3 months of age. Additionally, 74 calves of 1-2 months were sampled to determine the presence of Toxocara vitulorum eggs. Individual egg counts and infective strongyle larvae from pooled faecal samples were examined. Post-mortem worm counts were carried out on six groups of tracer calves (n=12) that had been kept for 4 weeks on pasture in and around the village studied. The following helminths were identified: T. vitulorum, Cooperia punctata, C. pectinata, C. oncophora, Oesophagostomum radiatum, Trichostrongylus axei, T. colubriformis, Haemonchus spp., Fasciola spp. and Paramphistomum spp. In 8% of the samples collected from young calves, individual egg counts for T. vitulorum were found indicative for pathogenic worm burdens. Strongyle egg counts and worm counts indicated that transmission is low without a distinct seasonality. In animals of 3-9 months old, a strongyle egg count peak can be demonstrated which at a higher age steadily and significantly decreased. In faecal cultures Cooperia spp. were most prominent in all age groups throughout the year with the exception of the period September-November when Haemonchus spp. and Oesophagostomum spp. were most prevalent. Fasciola spp. eggs were found in 22% of the collected faecal samples and the egg counts were low indicating that the intensity of Fasciola spp. infection is mild. Based on the present data, regular anthelmintic treatments seem not to be justified, except for a single treatment at the age of 2 weeks against toxocariosis.  相似文献

Oxibendazole (OBZ; 10 mg/kg of body weight) was administered to ponies at 8-week intervals to control strongylosis in a breeding band of Shetland-type ponies (n = 29 to 50) from October 1978 through September 1984. A similar use of cambendazole (CBZ; 20 mg/kg of body weight) in this band of ponies during the preceding 4-year period resulted in the survival of a CBZ-resistant population (S) of small strongyles. Effectiveness of OBZ treatments was monitored by pre- and posttreatment counts of the number of strongyle eggs per gram of feces (epg) and of the number of strongyle larvae per gram of feces (lpg). For the first 4 years of the 6-year period, epg and lpg counts were determined for 3 to 5 of the OBZ treatments; during the last 2 years, counts were determined for each treatment. For the 1st year, the mean reduction in epg was 96%. During the 2nd and 3rd years, mean reductions in epg were 80% and 81%, respectively. For the 4th and 5th years, mean reductions in epg were 69% and 43%, respectively. For the 6th year, the mean reduction in epg was 37%. Statistical analyses of the epg data, using the linear plateau method, indicated a significant increase in residual (surviving) strongyle infections after the periodic OBZ treatments, with a mean annual increase in survival of 10.3% for the 6 years. Changes in lpg were variable, but an overall loss of effectiveness of the OBZ treatments was evident. Sixteen nonsibling sheep, approximately 12 months old, that were raised in a helminth-free environment, were used for 2 protection studies 6 months apart. Sheep were vaccinated weekly for 5 weeks by IM injection of fibrinogen-degrading proteins derived from the intestinal tract of adult Haemonchus contortus. Ten days after the last vaccination, sheep were given 2,500 infective H contortus larvae by intraruminal injection. Vaccinated sheep produced specific antibodies, and were protected from the worm challenge. Significant differences in mean fecal worm egg counts for 56 days after worm challenge, in mean numbers of H contortus worms, and female fecundity ratios at necropsy were detected in vaccinated sheep, compared with those in control sheep. These data suggest that the fibrinogen-degrading proteins have a protective role in vaccination of sheep against H contortus.  相似文献

A high level of host resistance to Trichostrongylus colubriformis was established in a group of Saanen goats infected weekly with 10,000 infective larvae for 10 weeks, treated with an anthelmintic and then challenged twice with 50,000 larvae, with a further anthelmintic treatment 28 days after the first challenge. A second group of goats exposed only to the two challenge infections developed worm burdens intermediate between, and significantly different from, the first group and a third control group given only the second challenge infection. Nematode fecundity (eggs per female) and male:female ratios were also significantly decreased in the first two groups. Faecal egg counts and worm counts were highly correlated and this relationship was dominated by the influence of the number of eggs/female nematode. Seasonal rises in mean faecal egg output were observed in grazing ponies in spring (578 eggs per gram) and in summer (930 epg) on 30 April and 2 September, respectively, in untreated ponies. Pasture infectivity reached a peak of 18,486 third stage larvae (L3)/kg on 17 September, two weeks after peak egg counts, coincidental with abundant September rainfall (103.0 mm). Differentiation of infective larvae from pasture showed the cyathostomes (small strongyles) to be predominant, but Trichostrongylus axei assumed major importance from late August to October. The large strongyles were rarely detected: Strongylus vulgaris was found only once and S edentatus only twice. The most effective parasite prophylaxis was achieved by twice weekly removal of faeces. In this group, concentrations of infective L3 on pasture reached a maximum of 1000 L3/kg, compared to 18,486 L3/kg for a control group and 4850 to 10,210 L3/kg for anthelmintic treatment groups. The removal of faeces increased the grazing area by about 50 per cent, by eliminating the characteristic separation of horse pasture into roughs and lawns. Spring and summer anthelmintic treatments of mature ponies with oxibendazole were effective in reducing the late season rise in pasture infectivity to 4850 L3/kg, but treatment of young ponies (mainly yearlings) with ivermectin every eight weeks or oxibendazole every four weeks resulted in pasture infectivity as high as 10,000 L3/kg. The efficacy of moxidectin 2 per cent equine gel against naturally acquired strongyle infections was assessed in 18 ponies which had grazed on contaminated pasture before being housed for eight weeks. Twenty-four hours before the treatment, two randomly selected ponies were euthanased and their worm burdens were determined. Eight of the remaining 16 ponies were treated with moxidectin 2 per cent gel while the other eight were given a placebo gel. Eight weeks later the 16 animals were necropsied and their worm burdens established. A 100 per cent efficacy was recorded against adult and lumenal L4 cyathostomes and adult Strongylus and Triodontophorus species. Digest recoveries of larval cyathostomes indicated a 90.8 per cent (P<0.002) reduction in early L3 and a 99.9 per cent (P<0.001) reduction in developing stages. There was a reduction in faecal egg output of between 96 and 100 per cent in the treated animals compared with the controls.  相似文献

SUMMARY Development of the free-living stages of strongylid nematodes of the horse to the infective stage occurred in faeces in all months of the year in southern Queensland, at a rate which depended on the season. Most rapid development to the infective stage occurred in the warmer months, with the hatching of strongyle eggs being completed in 2 days in summer. During the winter, egg hatching continued for over 2 weeks. Larval moults proceeded at a faster rate in summer—all larvae were infective in 7 days during the hottest months, but it was as long as 5 weeks before all were infective in winter. However, even though development was rapid in summer, survival rates varied from 1 to 10%, in contrast to the spring and autumn, when over 80% reached the infective stage. One percent of larvae in faeces survived for up to 20 weeks in autumn and winter, but for only 4 weeks in summer. These results highlight the inadequacy of short-term pasture spelling for all but the hottest months. Infective larvae were found on herbage in all months of the year, but greatest numbers were recovered in spring and early summer, and in autumn and early winter. The relationship of pasture infestation to migration of larvae from Paecal reservoirs in response to rain was clearly shown. Most infective larvae were found within 30 cm of faecal masses, and in fact 89% of all larvae isolated from herbage in this study were found within 15 cm of faeces. Migration of larvae from faeces to herbage occurred with falls of rain as small as 25 mm. Horse faecal masses dried out completely in 6–8 days in summer and in 14–16 days in winter. Strongyle larvae developed to the infective stage in faeces in the absence of rain, although many remained in the pre-infective stage and completed their development when rain fell. This study shows that massive contamination of pastures with the eggs of strongylid nematodes must be prevented in spring and autumn if susceptible young horses are not to be at serious risk.  相似文献

The faecal egg counts of groups of sheep given 5 000, 20 000 or 40 000 infective larvae of Ostertagia circumcincta reached a peak at about 28 days after infection; the height of the peak varying directly with the number of larvae given. At 35 days the ratios of total worm burdens and the total eggs in all females amongst the groups were similar. Between 35 and 70 days post infection the faecal egg counts fell to a low level which was roughly the same regardless of the number of larvae administered. At 70 days the two groups of lambs given the two higher doses of larvae had similar worm burdens, but those given 5 000 larvae harboured about half as many. At this time the total eggs in all females was approximately the same in each group. Whatever factor, in addition to worm numbers, which may have limited egg output appeared to be ineffective until some time after the peak egg count was reached.  相似文献

Eleven trials were conducted to collect helminthologic and pathologic data from 27 conventionally reared (CR) and 53 specific-pathogen-free (SPF) 6- to 13-week-old male New Zealand White rabbits. These rabbits were given 50,000 to 100,000 ensheathed third-stage infective Ostertagia ostertagi larvae (L3) orally. The L3 had been isolated from the feces of cattle. Fecal egg counts were conducted and worm populations were determined after euthanasia 3 to 56 days after inoculation. At necropsy, nodules were observed in a confluent pattern in the mucosa of cardiac region of the stomach in all inoculated rabbits, except in CR rabbits inoculated at 6 weeks of age, which had no nodules or worm burdens 42 days after inoculation. Confluent areas were not observed in SPF rabbits euthanatized 5 days or less after inoculation; however, small, transparent nodules were evident in the mucosa of the cardiac region of the stomach. Fourth-stage larvae (L4) were obtained from the mucosal nodules after digestion of stomach of both types of rabbits. There were more L4 in SPF than CR rabbits. In addition, greater numbers of L4 were found in SPF rabbits euthanatized 14 days or less after inoculation. Petechial hemorrhages were in the fundic area of the stomach mucosa in SPF rabbits inoculated with 100,000 L3 and euthanatized 14 days later. Mature O ostertagi or worm eggs in feces were not found in inoculated CR or SPF rabbits. An investigation of the spring rise in strongyle egg output of grazing horses on two commercial horse farms in northern USA in 1981 and 1982 revealed two distinct spring and summer rises in faecal egg counts, with peaks in May and August/September. There was a marked rise in the concentration of infective larvae on pasture two to four weeks after the peaks in egg output, so that grazing horses were at serious risk from June onwards and pasture larval counts on one farm did not fall to low levels until June of the following year. The spring and summer rises in faecal egg counts appeared to be seasonal in nature, to be derived largely from worms developing from previously ingested larvae, rather than from newly ingested larvae, and to be unrelated to the date of foaling. An epidemiological approach to strongyle control based on prophylactic treatments in the spring successfully eliminated the spring rise in egg output but was inadequate to control the summer rise or subsequent escalation of pasture infectivity in September. It was, nevertheless, superior to a conventional treatment programme at eight week intervals, using the same drug, pyrantel pamoate. Prophylactic spring/summer treatments proved to be much more effective. Both pyrantel pamoate at four week intervals and ivermectin at eight week intervals kept faecal egg counts at low levels during spring and summer. As few as two ivermectin treatments (11 May, 6 July) resulted in a sixfold reduction in pasture larval counts on 9 November and 3 January for the treated group (8872, 8416 stage three larvae [L3]/kg) compared to the control group (52,824, 50,984 L3/kg).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

OBJECTIVE: To relate trichostrongylid infections acquired by sheep during summer to prevailing weather conditions. PROCEDURE: Groups of worm-free 'tracer' sheep were put onto pastures, previously contaminated with trichostrongylid eggs, for successive periods of 2 weeks from December to March. After grazing the sheep were housed for 6 weeks. Weekly worm egg counts and worm counts were used to estimate the numbers of worms acquired and related to weather conditions during the grazing period. RESULTS: No worm eggs were detected in the faeces of sheep that grazed at the end of January when only 7 mm of rainfall was recorded. At other times rainfall between 12 and 24 mm occurred and strongyle egg counts were generally either < 50 or > 150 eggs per g (epg). Mean counts of 1,100 Ostertagia and Trichostrongylus adults gave rise to mean counts of about 350 epg whereas about 6,000 Nematodirus spp were associated with mean egg counts of about 200 Nematodirus spp epg. CONCLUSIONS: Rainfall events during summer determine the numbers of trichostrongylid larvae acquired by sheep in summer but further studies are necessary before the implications for strategic control programs in southern Australia can be fully assessed.  相似文献

Serum protein responses were examined in 52 ponies divided into five groups and subjected to various control strategies that resulted in pasture infectivity ranging from 706 to 18,486 infective third stage, cyathostome and Trichostrongylus axei larvae per kilogram of herbage (L3/kg) by 17 September 1984. Major protein changes occurred only in young ponies (Groups 4 and 5) and were observed before exposure to maximum numbers of pasture larvae (Group 4; 10,210 L3/kg, Group 5: 10,042 L3/kg) on 17 September. It appeared that a primary infection of T axei was a greater stimulus to serum beta-globulin and immunoglobulin (Ig)G(T) responses that provided by continued infection with cyathostome (small strongyle) worms. The large strongyles (Strongylus vulgaris, S edentatus and S equinus) were not detected in any larval cultures or on pastures grazed by the young ponies. A fall in beta-globulin and IgG(T) concentrations of Group 5 ponies one month after treatment with ivermectin indicated a larvicidal action against T axei and/or the cyathostomes. A subsequent rise in serum albumin concentrations of Group 5 ponies suggested that a protein-losing gastroenteropathy had been alleviated by the larvicidal action of ivermectin. Mature control ponies (Group 1) showed little beta-globulin response and only a modest IgG(T) response in six of the 10 ponies after exposure to heavily infected lawns (18,486 L3/kg) in September 1984. It was concluded that serum protein and IgG(T) responses were of limited value as an aid to diagnosis of parasitism because of numerous difficulties of interpretation.  相似文献

The concentrations of haptoglobin, immunoglobin (Ig)G(T) and IgG were measured in the serum of four previously parasite-free pony yearlings following a single dose of 700 (Group H) or 200 (Group L) stage three Strongylus vulgaris larvae (L3) and following a reinfection with the same doses 34 weeks later. The results are compared with an uninfected control pony. The haptoglobin concentration increased during Weeks 1 to 6 and 14 to 17 after infection in the serum of the ponies receiving 200 L3, but in only one pony dosed with 700 L3 (during Weeks 1 to 16). The serum haptoglobin also increased during the first seven weeks after the second infection, in three of the four ponies following the second dose of larvae. The serum IgG(T) concentration started to increase from Week 6 or 9 in the ponies given 700 L3, reaching peaks of 44 and 32 g/litre respectively, eight to nine weeks later, compared with a peak of 16 g/litre 20 to 22 weeks after infection in ponies dosed with 200 L3. The IgG(T) concentration increased to a maximum of 25 g/litre in the serum of only one of the four ponies after the reinfection. The serum IgG concentration in all ponies increased nearly twofold during the first eight weeks after both the primary and secondary dose of larvae. It is concluded that the measurement of specific proteins is more reliable and quicker than the electrophoretic separation and quantitation of protein bands, in tracing changes in serum proteins following the artificial infection of ponies with S vulgaris larvae.  相似文献

Faecal samples were collected from 500 cattle and 385 buffaloes belonging to backyard farmers from 24 villages in the province of Bukidnon, Mindanao, the Philippines. Strongyle nematode infections were studied on these samples by faecal egg counts and infective larvae differentiation. The effects of animal species and age on the prevalence and intensity of infections were studied. The overall prevalence of strongyle infection was 53% for cattle and 28% for buffaloes. The prevalence decreased with increasing age in both animal species. Faecal egg counts were significantly lower in buffaloes than in cattle. Adult animals had lower egg counts than calves. This age effect was most pronounced in buffaloes. Six genera of strongyle nematodes were identified from the faecal cultures in both animal species, Mecistocirrus, Haemonchus, Trichostrongylus, Cooperia, Bunostomum and Oesophagostomum. The distribution of larvae did not vary much with age.  相似文献

The small strongylid infections of two groups of three yearling female Shetland ponies and one yearling Shetland tracer pony were studied. One group was set stocked from April to November and was treated monthly with 5 mg kg-1 albendazole from two days before turnout until July. The other group grazed similar pasture until July, was treated with 5 mg kg-1 albendazole and subsequently removed to pasture grazed by sheep from April to July. The tracer ponies were added to both groups in September. The efficacy of both methods was not completely satisfactory probably because of low efficacy of anthelmintic treatment. There were no significant differences between the cyathostome burdens of the two groups. A high proportion of the cyathostome populations of all ponies consisted of inhibited early third stage larvae (L3). The finding of low numbers of immature fifth-stage worms in the tracer ponies indicated that the considerable adult burdens in the permanent ponies originated from infection picked up before the tracer ponies were added. In the group which was removed to sheep pasture after treatment in July it was likely that the majority of the adult worm burden had been ingested as infective larvae before treatment.  相似文献

The potential of the chitin synthesis inhibitor diflubenzuron (DFB) to alter the development of the parasitic nematodes (Ascaris suum and Haemonchus contortus was investigated. DFB given orally (10 mg kg-1 per day for 30 days) to sheep inoculated with H. contortus infective larvae did not prevent the establishment of adults or affect fecal egg output. However, there was a significant (greater than 90%) decrease in the number of infective larvae recovered from fecal cultures derived from lambs harboring H. contortus adults that were treated with DFB. DFB did not affect egg hatching. Oral administration (10 mg kg-1 per day for 20 days) of DFB to swine harboring adult A. suum adults had no effect on the adult worm burden or on egg morphology, but eggs removed from worms obtained from DFB-treated swine contained less chitin than eggs removed from untreated control swine. DFB also inhibited chitin synthesis in vitro in the isolated reproductive tract of A. suum adults. These results indicate that DFB at high doses can inhibit the subsequent development of H. contortus larvae in the feces. Since H. contortus larvae lack chitin, DFB may act on these larvae by a mechanism independent of a direct effect on chitin synthesis.  相似文献
