Limitations on the acceptable proportion (incidence) of pods with white mould may lead to the rejection of entire fields of processing snap bean. The low tolerances (no more than 2% to 6% of pods with white mould) are difficult to estimate with sufficient precision in the field when time is limited. These constraints motivated this study of white mould across three spatial hierarchical levels: pods, plants and quadrats consisting of two adjacent plants within rows. Hierarchical relationships are required when designing formal rules for estimating the incidence of pods with white mould from units higher in the spatial hierarchy. Disease assessments were made on all pods from 38 within‐row transects of 40 plants each of the snap bean cultivar Hystyle. Using probability‐based principles, equations were derived and fitted to data on the incidence of white mould on pods (ipod), plants (ipad) and quadrats (iq(2)), which led to a function directly linking ipod to iq(2). The variance of ipod increased with iq(2), but that may be of little consequence at the lower values of iq(2) likely to be associated with ipod values at processor‐set tolerances. For example, at iq(2) = 0.1 there was a 92% probability that ipod was less than 0.02. Assessing iq(2) may be more efficient than directly estimating ipod because iq(2) was about an order of magnitude higher than ipod. Results suggest it may be feasible to design sampling plans for estimating the proportion of pods with white mould from an assessment of the proportion of diseased quadrats. 相似文献

Development of common bean cultivars with partial white mould resistance through breeding techniques has been a challenge in Brazil. As yet, lines/cultivars from breeding programmes have not been investigated for resistance; therefore, this study screened 107 lines/cultivars for their reactions to white mould in 14 preliminary trials conducted under irrigation. Thirteen resistant lines/cultivars (three of Andean origin) and six Mesoamerican cultivars (three intermediately resistant and three susceptible) were selected for further investigation. These lines/cultivars and the resistant control A195 were evaluated in six advanced trials and two straw tests to assess the effectiveness of the screening procedure. In 11 preliminary trials, screenings were performed under moderate/high or higher disease pressure. These pressures occurred in two advanced trials in which, when yields were averaged across moderate/high and high pressures, 10 Mesoamerican lines/cultivars selected for resistance yielded 14%, 23%, and 38% more than intermediately resistant cultivars, A195, and susceptible cultivars, with median disease ratings (1–9 scale) of 4.5, 5.7, 5.7, and 6.7, respectively. In the straw test, three Andean lines/cultivars (A195 included) and two susceptible cultivars in the field were among those with the highest levels of physiological resistance. Thus, field rating under high disease pressure and greenhouse rating did not correlate significantly, suggesting that field trials are critical to evaluating resistance and to identifying high-yielding beans. Therefore, lines/cultivars from breeding programmes assessed in field trials may provide a low cost and fast way to identify high-yielding bean cultivars with partial resistance to white mould in the subtropical southern hemisphere. 相似文献

To improve sampling efficiency and precision in the assessment of white mould (caused by Sclerotinia sclerotiorum) disease incidence on bean (Phaseolus vulgaris), the spatial characteristics of epidemics were characterized in 54 linear transects in 18 bean fields during 2008–2010 in northern Tasmania, Australia. The incidence of diseased pods and plants was assessed prior to harvest. Distributional and correlation‐based analyses indicated the incidence of diseased pods was characterized by a largely random pattern at the individual plant scale, with some patches of similar disease levels on pods occurring at a scale of 1·5 m or greater. Collectively, these results suggested epidemics may be dominated by localized sources of inoculum. Sequential sampling approaches were developed to estimate or classify disease incidence above or below provisional thresholds of 3, 5 and 15% incidence on pods near harvest. Achieving prespecified levels of precision by sequential estimation was possible only when disease incidence on pods was greater than approximately 4% and sampling was relatively intense (i.e. 10 pods evaluated on each of at least 64 plants). Using sequential classification, correct decisions on disease status were made in at least 95% of independent validation datasets after assessment of only 10·1–15 plants, depending on classification threshold and error rates. Outcomes of this research provide the basis for implementing more efficient sampling and management strategies for this disease in Australian fields. 相似文献

In-row plant densities have not been studied for common beans with type II growth habit and contrasting reactions to white mould. Advanced breeding lines with partial resistance or susceptibility to white mould were combined with 4, 7, 10 or 13 plants m−1 and with or without fungicide at a constant between-row spacing of 0.5 m in five sprinkler-irrigated field trials conducted during the autumn–winter season in Brazil. White mould pressures in the trials covered the whole range from zero to moderate/high (46–60% of white mould severity index). In all trials, means of white mould incidence, severity and yield did not vary significantly between 7 and 13 plants m−1 for the partially resistant line, regardless of the fungicide levels. For the susceptible line, 13 plants m−1 increased white mould incidence and severity under moderate disease, regardless of the fungicide levels, and decreased yield compared with 10 plants m−1 when fungicide was applied twice under moderate/high disease pressure. For the susceptible line, 7 or 10 plants m−1 maximized yield in all trials, with or without fungicide applications. The results suggest that the current recommendation of 11–13 plants m−1 could be used for type II beans with partial resistance to white mould in either a conventional or organic system. For susceptible genotypes, 7–10 plants m−1 seems to be the most appropriate in-row plant density. This study may improve the recommendation of in-row plant density for type II beans cultivated under white mould pressure. 相似文献

Microplot experiments were carried out during the autumn and winter of 1995 to determine the effects of soil moisture and levels of organic matter on the carpogenic germination of Sclerotinia sclerotiorum and subsequent infection of Phaseolus vulgaris . Soil moisture treatments were established by irrigation to field capacity: (a) when soil had dried to − 25 KPa, (b) when soil had dried to − 54 KPa, and (c) eight days after the soil had dried to − 54 KPa. Three levels of organic matter were established by mixing compost and yellow-red latosol in the following proportions (by volume): (a) 0% compost and 100% soil, (b) 50% compost and 50% soil, and (c) 100% compost and 0% soil. More apothecia and diseased plants were observed in the plots with higher moisture levels. Drier treatments resulted in a reduction in number (in the winter experiment) or elimination (in the autumn experiment) of apothecia. Other experiments were designed to examine the effect of grass mulching and organic matter levels on the carpogenic germination of S. sclerotiorum . In one experiment, grass mulching was compared with no mulching, and in another, the effects of different depths of mulch on the soil surface (0, 1.5, 3, 6 and 9 cm) were examined. Fewer apothecia were formed in mulched soil and the effect was most pronounced with 6 and 9 cm of mulch. The highest level of mulch (9 cm) impaired the vigour and yield of bean plants. In these experiments, carpogenic germination was increased by high soil organic matter content. Control of soil moisture and grass mulching may reduce the production of apothecia of S. sclerotiorum even in soils rich in organic matter. 相似文献

Cotton and snap bean were selected for a multi-year, multi-state regional (south-eastern USA) research project to evaluate the efficacy of both commercial and experimental bacterial and fungal biological control agents for the management of damping-off diseases. The goal for this portion of the project was to determine the viability and stability of biological agents after application to seed. The biological seed treatments used included: (1) Bacillaceae bacteria, (2) non-Bacillaceae bacteria, (3) the fungus Trichoderma and (4) the fungus Beauveria bassiana. Seed assays were conducted to evaluate the following application factors: short-term (< or = 3 months) stability after seed treatment; quality (i.e. isolate purity); compatibility with chemical pesticides and other biocontrol agents; application uniformity between years and plant species. For the bacterial treatments, the Bacillaceae genera (Bacillus and Paenibacillus) maintained the greatest population of bacteria per seed, the best viability over time and the best application uniformity across years and seed type. The non-Bacillaceae genera Burkholderia and Pseudomonas had the least viability and uniformity. Although Beauveria bassiana was only evaluated one year, the seed fungal populations were high and uniform. The seed fungal populations and uniformity for the Trichoderma isolates were more variable, except for the commercial product T-22. However, this product was contaminated with a Streptomyces isolate in both the years that it was evaluated. The study demonstrated that Bacillaceae can be mixed with Trichoderma isolates or with numerous pesticides to provide an integrated pest control/growth enhancement package. 相似文献

Surface and internal populations of Xanthomonas campestris pv. phaseoli, causal agent of common bacterial blight of bean, on and in flower buds, blossoms and pods of seven bean (Phaseolus vulgaris) genotypes were studied. Bean plants were grown in the field and artificially inoculated at the seedling stage (18 days old). The pathogen was recovered in high numbers from flower buds, blossoms, pods and seed of both resistant and susceptible bean genotypes. Significant differences (P = 0.05) in population levels of X. c. pv. phaseoli between stages of reproductive tissue development were observed. Infected seed from resistant bean genotypes had no visible symptoms. Such seed may play an important role in the epidemiology of common bacterial blight because they are difficult to detect and may occur at low frequency in seed lots, as was the case in the current study. 相似文献

Effects of droplet size and carrier volume on foliar uptake and transport of daminozide were investigated. A constant dose of daminozide (100 µg per leaf) was applied to both primary leaves of 10‐day‐old Phaseolus vulgaris (cv Nerina) in droplet sizes of 1–10 µl and carrier volumes of 10 to 200 µl per leaf. Decreasing droplet size or increasing carrier volume decreased daminozide penetration, but increased translocation. Plotting the logarithm of the leaf surface/droplet interface area vs daminozide uptake yielded a negative linear relationship, but for translocation an optimum quadratic type relationship was obtained. Some phytotoxicity occurred at low carrier volumes and large droplet sizes. The degree of phytotoxicity was positively related to the amount of daminozide deposited per unit wetted area above 0.7 µg daminozide mm−2. Below this threshold, there was no visual evidence of phytotoxicity. At the breakpoint, the deposit covered an area of 276 mm2 on both primary bean leaf surfaces. Since the maximum in the relationship of translocation with interface area was in close agreement with the threshold amount of deposit above which phytotoxicity occurred, the inverse relationship between daminozide uptake and translocation at low interface areas was attributed to phytotoxicity. © 2000 Society of Chemical Industry 相似文献

Cucumber mosaic virus (CMV) is a major component of the virus complex that has become more pronounced in snap bean in the midwestern and northeastern United States since 2001. Multiple-vector-transfer tests were done to estimate the CMV transmission efficiencies (p) of the main aphid species identified in commercial snap bean fields in New York and Pennsylvania. The four most efficient vectors (p > 0.05) were Aphis gossypii, A. glycines, Acyrthosiphon pisum, and Therioaphis trifolii, which were all significant species in the migratory aphid populations in snap bean. Moderately efficient vectors (0.01 < p < 0.04) were A. spiraecola, A. craccivora, Macrosiphum euphorbiae, and Rhopalosiphum maidis. Poor vectors (p < 0.01) included A. fabae, Nearctaphis bakeri, and Myzus persicae. Only one species, Sitobion avenae, failed to transmit CMV in replicated tests. Estimates of p were consistent between different clones of the same aphid species and among three different field isolates of CMV tested. Single-vector-transfer test results for a subset of the species supported those obtained via the multiple-vector-transfer approach. Our results are consistent with the notion that A. glycines is a major vector of recent CMV epidemics in snap bean, but that species is only one of several that are involved. 相似文献

采用已报道的七种油菜菌核病抗性鉴定方法,在同一条件下对两个感病和两个抗病品种进行鉴定比较。结果表明:草酸浸根(叶)法用于白菜型油菜鉴定效果不好;不同生育期的油菜对菌核病抗性有差异,应该以花期鉴定为主,苗期鉴定为辅;在花期以麦粒叶腋接种、琼脂块叶腋接种和牙签茎杆接种,接种后发病程度依次为牙签>麦粒>琼脂块;三种花期接种方法与大田接种鉴定的结果呈显著正相关(r>0.8000),这三种方法均能有效反映抗、感品种的差异,其中花期牙签茎杆接种法与大田接种鉴定法相关性达极显著水平(r=0.9140)。根据抗性鉴定方法比较的结果,选用花期牙签茎杆接种法,以中油821为对照,对四川农业大学油菜研究中心G1自交系的44份油菜育种材料进行鉴定、筛选,得到12份高抗材料,其中有5份材料的抗性极显著高于对照。 相似文献

Yield losses due to rust and angular leaf spot (ALS) of snap beans may reach 100% in Eastern Africa. Where susceptible varieties are grown, farmers control these diseases with routine fungicide applications. To determine an optimum application rate and spray schedule for Orius® (tebuconazole 250 g/L), we sprayed 10 mL and 20 mL Orius® per 15 L spray water twice at two trifoliate leaf stage and 50% flowering, and three times at the same stages, with an additional application at pod initiation. In farmers’ fields, we tested the effect of fungicide sprays, use of resistant variety, intercropping, increased plant spacing and farmyard manure on rust and ALS diseases. Application three times of 20 mL Orius® per 15 L spray water reduced rust severity scores by 5.7 and 2.4 in 2010 and 2011, respectively. Lowest rust and ALS severities were observed when a resistant variety, fungicide or farmyard manure was used. Pod yield increments due to disease management ranged between 13% and 242%. Prophylactic fungicide application, use of resistant varieties and farmyard manure can be used to reduce disease severity and improve snap bean quality on smallholder farms. 相似文献

To study the systemic effects of active neem ingredients, the substrate of bean plants was treated with a 170 g kg(-1) azadirachtin (NeemAzal-U; Trifolio-M GmbH, Lahnau, Germany, registration pending). This product was used at a dose rate of 10 mg AZA (azadirachtin a) and 1.2 mg 3-tigloyl-azadirachtol (azadirachtin b) per treated bean plant. Afterwards, the translocation and persistence of AZA and 3-tigloyl-azadirachtol and the effects on western flower thrips, Frankliniella occidentalis (Pergande), were studied. Residues of AZA and 3-tigloyl-azadirachtol from substrates with different contents of organic matter [pure culture substrate (CS), CS-sand mixture] and from various plant parts were quantified by high-performance liquid chromatography-mass spectrometry (HPLC-MS). The dissipation trends of AZA and 3-tigloyl-azadirachtol were similar within the same substrates. A slower decline of both active ingredients was measured with CS than with CS-sand mixture. Residue analysis of the bean plants showed that only small proportions of the initial amounts of AZA and 3-tigloyl-azadirachtol applied to the substrate were present in the plant (0.3-8.1%). Variable amounts of residues of the active components in relation to plant parts and time of analysis indicated a different translocation pattern for the two active ingredients. Higher residues of the active ingredients were found in roots and stems after neem application using CS, whereas higher residues were found in leaves after CS-sand mixture treatments. Mortality of F. occidentalis after NeemAzal-U soil applications reached up to 95% on CS-sand mixture, compared with 86% in CS. 相似文献

Weeds that emerge along with or immediately after crop plants usually can reduce the yield of those crops. Two randomized complete block design experiments were conducted during 2006 and 2007 in Tabriz, Iran to determine the critical period of redroot pigweed control in the green bean hybrid “Cantander”. The treatments were weed‐infested and weed‐free plots at 2, 4, 6, 8, 10, and 14 weeks after bean emergence (WABE). The green bean biomass was affected by the early emergence of redroot pigweed, but it was not reduced when redroot pigweed emerged at 10 weeks after crop emergence, along with crop emergence, and grew with green bean until 4 WABE. The redroot pigweed biomass decreased by 2.7 g m?2 per day when weed emergence was delayed. Each 100 g m?2 of weed biomass that was produced resulted in a 1.4 kg ha?1 loss in the green bean yield. When redroot pigweed interference lasted for ≥4 weeks after green bean emergence, the green bean yield was reduced significantly. Weeds, which emerged 2 weeks after green bean and thereafter were controlled, did not decrease crop productivity significantly. The highest crop yield was obtained when the weed emerged at 14 WABE. The critical period of redroot pigweed control, considering a 10% yield loss, was between 19 and 55 days after green bean emergence. Thus, weed control practises should be begun no later than 3 WABE and should continue until at least 8 WABE in order to obtain the maximum green bean yield. 相似文献

The main goal of the present work was to characterize the rust resistance (RR) gene present in the Mesoamerican common bean cultivar Ouro Negro, temporarily named Ur‐OuroNegro or Ur‐ON, which is the main RR source used in Brazil. The RR spectrum presented by cv. Ouro Negro was compared with those of other bean lines harbouring known RR genes when inoculated with nine selected races of Uromyces appendiculatus, the causal agent of bean rust. In addition, all bean lines were screened with molecular markers linked to Ur‐ON in order to identify additional evidence for the presence of alleles for this locus in the screened RR sources. The allelic relationships of Ur‐ON were tested with previously characterized RR genes from lines resistant to at least one race of the pathogen. Allelism tests were also carried out between cv. Ouro Negro and cvs CNC and CSW 643, important RR sources in Brazil harbouring unnamed RR genes. The results showed that the major dominant gene conditioning RR in cv. Ouro Negro is positioned at a locus distinct from those with which it was compared. It is proposed that this gene – or complex gene locus – is unique and be designated Ur‐14. 相似文献

BACKGROUND: A point mutation often confers resistance of organisms against medical drugs and agricultural pesticides. Allele‐specific nucleotide polymerase chain reaction (ASPCR) and allele‐specific quantitative real‐time PCR using SYBR Green (ASQPCR) are widely and effectively applied to detect and monitor this type of resistance. However, the former is unsuitable for high‐throughput detection, and the latter often reduces the accuracy of detection. RESULTS: In order to decrease background amplification, a rapid and high‐throughput genotyping method with mismatch primers was developed (ASQPCR‐MP) and applied specifically to survey the frequency of the highly benzimidazole‐resistant MBCHR mutation (E198A) in the β‐tubulin gene of Sclerotinia sclerotiorum (Lib.) de Bary populations. Genomic DNA from 223 sclerotia was analysed. Similar genotype results were also obtained using ASPCR with mismatch primers and a mycelial growth inhibition assay. It was found that ASQPCR‐MP clearly differentiated MBCHR and benzimidazole‐sensitive MBCS phenotypes. Moreover, ASQPCR‐MP took less than 6 h to complete. CONCLUSION: ASQPCR‐MP appears suitable for large epidemiological studies involving resistant genotypes and requiring high‐throughout formats. Copyright © 2009 Society of Chemical Industry 相似文献

Meta‐analytic modelling of the incidence–yield and incidence–sclerotial production relationships in soybean white mould epidemics 下载免费PDF全文
White mould (Sclerotinia sclerotiorum) is a destructive disease of soybean worldwide. However, little is known of its impact on soybean production in Brazil. A meta‐analytic approach was used to assess the relationship between disease incidence and soybean yield (35 trials) and between incidence and sclerotia production (29 trials) in experiments conducted in 14 locations across four seasons. Region, site elevation and season included as moderators in random‐effects and random‐coefficients models did not significantly explain the variability in the slopes of the incidence–yield relationship. The Pearson's r, obtained from back‐transforming the Fisher's Z estimated by an overall random‐effects model, showed that incidence of white mould was moderately and negatively correlated with yield (r = ?0.76, P < 0.0001). A random‐coefficients model estimated a slope of ?17.2 kg ha?1%?1, for a mean attainable yield of 3455 kg ha?1, indicating that a 10% increase in white mould incidence would result in a mean yield reduction of 172 kg ha?1. White mould incidence and production of sclerotia were strongly and positively correlated (r = 0.85, P < 0.0001). For every 10% increase in white mould incidence, 1 kg ha?1 of sclerotia was produced. The relationship between disease incidence and production of sclerotia was stronger in southern regions and at higher elevation. In the absence of management, economic losses associated with white mould epidemics, assuming 43% incidence in 22% of the soybean area, were estimated at approximately US $1.47 billion annually within Brazil. 相似文献
