The pathology of acute Leptospira interrogans serotype icterohaemorrhagiae infection in the Syrian hamster was investigated up to 7 days after infection using histology, electron microscopy and an indirect fluorescence test for leptospires. The disease was characterized by the presence of many leptospires in the tissues, jaundice, leukocytosis, haemorrhages, endothelial alteration and thrombotic glomerulopathy. The leptospires were present intravascularly, in the interstitium penetrating between liver cells and tubular epithelial cells and in the tubular lumina. The presence of leptospires was not necessarily associated with lesions. These findings support both pathogenetic mechanisms suggested in the literature, namely: the ability of leptospires to penetrate actively between cells with detachment of tight junctions, without obvious lesions to the cells, and an immune-mediated process with immune complex formation and binding and activation of complement resulting in leukocytosis, thrombotic glomerulopathy, endothelial alteration and haemorrhages. 相似文献

The objective of the present study was to analyse allometrically the growth of the Syrian golden hamster pancreas during days 2 to 70 of postnatal development. Body and pancreatic mass were determined, followed by stereological determination of the absolute volume of each morphological compartment of the pancreas. The marked pancreatic growth, by 4360%, was due to an increase in the absolute volume of all morphological compartments, mainly the acini which showed an increase of 10 431%. Bivariate allometric analysis of pancreatic mass and morphological compartmental volume in relation to body mass gain showed: (1) a biphasic pattern for pancreatic mass, acinar volume, excretory duct volume and stromal volume, with the first phase being observed from 2 to 21 days of age and the second from 21 to 70 days of age, with allometry coefficients of 1.537-0.513, 1.770-0.543, 1.651-0.506 and 0.967-0.258, respectively, and (2) a monophasic pattern from 2 to 70 days for intercalated duct volume and islet volume, with allometry coefficients of 0.913 and 1.727, respectively. These results show that during the growth of the pancreas in relation to that of the body some structures - acini and excretory ducts - follow the growth pattern of the organ, while others - intercalated ducts and islets - show a different pattern. This may be related to the genetic growth characteristics of each compartment itself or to some relationship between compartments during some stage of the ontogenetic development of this organ. 相似文献

The details of the embryonic and postnatal differentiation of the olfactory epithelium (OE) and vomeronasal organ (VNO) were examined by light and electron microscopy in the Syrian hamster. At 10 days of gestation, the nasal placode is invaginated to form the olfactory pit on either side at the rostral end of the embryo. Abundant mitotic figures are observed near the free surface of the epithelium lining the olfactory pit. At 11 days of gestation, the mass of the epithelium lining a recess is separated from the medial wall of the olfactory pit to form the VNO. At 13 days of gestation, mitotic figures become observable in the basal layer of the vomeronasal sensory epithelium (VSE) in addition to the superficial to middle layers, while in the OE mitotic figures are observed mainly in the middle to basal layer. At 1 day after birth, the OE is almost complete in differentiation. On the other hand, the VSE differentiate slowly to retain some immature properties even at 10 days after birth. These findings suggest that the olfactory function seems to be solely ascribed to the OE for a while after birth. The significance of mitotic figures are discussed in the course of development with special reference to the origin of the nasal placode from the central nervous system. 相似文献

An approximately 8-week-old pet Syrian hamster (Mesocricetus auratus) with a 1-week history of dyspnea, hyporexia, and ataxia was submitted for necropsy. On gross examination, the hamster had multiple abdominal adhesions and enlargement of the mesenteric lymph node. Histologic evaluation revealed multicentric lymphoma of the liver, jejunum, mesenteric lymph node, testicular fat pad, and epididymis. Based on the hamster's age and the type and distribution of the lymphoma, a presumptive diagnosis of hamster polyomavirus-induced lymphoma was made. A specific polymerase chain reaction (PCR) was developed, which confirmed the diagnosis. An in situ PCR demonstrated hamster polyomavirus DNA within lymphocytes of the multicentric lymphoma and renal tubular epithelial cells and within clusters of enterocytes in the jejunum. These data are consistent with environmental dissemination of hamster polyomavirus virions through the renal tubular epithelium and into the urine and with fecal shedding of hamster polyomavirus virions; however, additional studies will be needed to confirm these observations. 相似文献

The objective of this study was to evaluate stomachs of 2-year-old Syrian hamsters that were naturally colonized by multiple Helicobacter species including Helicobacter aurati. A previous report on 7- to 12-month-old Syrian hamsters described chronic gastritis and intestinal metaplasia, a putative preneoplastic lesion in the stomach, without cancer. This report describes an invasive adenocarcinoma at the pyloric-duodenal junction in one of nine hamsters at a site of helicobacter-associated inflammation and marked intestinal metaplasia. Ceca of nine of nine animals were culture positive and polymerase chain reaction positive for Helicobacter spp. Furthermore, immunohistochemical analysis of the stomach using a H. pylori polyclonal antibody detected positive-staining bacteria within the pyloric region of three of nine hamsters including the neoplastic glands. However, argyrophilic bacteria were demonstrated only within the stomach of the hamster with gastric adenocarcinoma. This is a first report of gastric adenocarcinoma in helicobacter-infected hamsters. Syrian hamsters appear suitable as potential model for studying development of helicobacter-associated gastric adenocarcinomas. 相似文献

Hematological findings during the course of infection and the antibody response in Syrian hamsters infected with Babesia microti were examined. A macrocytic hypochromic anemia with an increase of the reticulocyte count was detected as a rise in the parasitized erythrocyte rate. White blood cell counts also remarkedly increased with the increases of both neutrophils and active-shaped monocytes, and thus they particularly play an important role in eliminating the parasite. In Western blotting with the sera from the hamsters infected with B. microti, a 38 kDa protozoan antigen reacted to the early-term sera, and additionally 28, 32, and 34 kDa antigens also reacted to the medium- and latter-term, and convalescent sera. These antigens were immunodominant and the antibodies against these antigens had also important roles for inhibition of this parasite. 相似文献

The Syrian hamster (Mesocricetus auratus) is the best animal model for human leishmaniosis, an emergent disease that causes 400,000 new cases every year. The main objective of this assay was to prove the relationship between female lactation, which implies hyperprolactinaemia (IFN-gamma, IL-12, IL-2 and TNF-alpha elevated), and leishmaniosis (IFN-gamma, IL-12, IL-2 and TNF-alpha diminished). The results demonstrate the clear implication of lactation in immune response. The infected lactating females did not show any symptoms of sickness, a 24 g mean increase was noticed, throughout the assay they were negative in the indirect diagnosis test (Direct Agglutination Test (DAT)), and in the direct diagnosis tests (N-N-N isolation, tissue impressions). 相似文献

The golden Syrian hamster (Mesocricetus auratus) is frequently used as a model to study virulence for several Leptospira species. Onset of an acute lethal infection following inoculation with several pathogenic Leptospira species has been widely adopted for pathogenesis studies. An important exception is the outcome following inoculation of hamsters with live L. borgpetersenii serovar Hardjo, the primary cause of bovine leptospirosis and a cause of human infections. Typically, inoculation of hamsters with L. borgpetersenii serovar Hardjo fails to induce clinical signs of infection. In this study, the authors defined LD(50) and ID(50) for 2 strains of L. borgpetersenii serovar Hardjo: JB197 and 203. Both strains infected hamsters with ID(50) values of approximately 1.5 × 10(2) bacteria yet differed in tissue invasion and interaction with leukocytes, resulting in widely divergent clinical outcomes. Hamsters infected with strain 203 established renal colonization within 4 days postinfection and remained asymptomatic with chronic renal infections similar to cattle infected with serovar Hardjo. In contrast, hamsters infected with strain JB197 developed a rapidly debilitating disease typical of acute leptospirosis common in accidental hosts (eg, humans) with an LD(50) of 3.6 × 10(4) bacteria. Evidence that strain JB197 resides in both extracellular and intracellular environments during hamster infection was obtained. Development of models that result in chronic and acute forms of leptospirosis provides a platform to study L. borgpetersenii pathogenesis and to test vaccines for the prevention of leptospirosis. 相似文献

Syrian hamsters are photoperiodic rodents in which reproduction, including testicular function, is stimulated by long photoperiod exposure and curtailed by exposure to a short photoperiod. The objectives of this study were to characterize the testis histomorphometrically and to determine the role of the proliferation and apoptosis phenomena in the recovery of the seminiferous epithelium during spontaneous recrudescence after exposure to short photoperiod. The study was performed using conventional light microscopy, proliferating cell nuclear antigen and terminal deoxynucleotidyl transferase (TdT)‐mediated dUTP in situ nick end labelling staining, image analysis software, and transmission electron microscopy in three recrudescence groups: initial recrudescence (IR), advanced recrudescence (AR) and total recrudescence (TR). The results morphometrically pointed to the gradual recovery of the testicular and tubular volumes, as well as of the seminiferous epithelium. Among the IR and AR groups, the increase in testicular and tubular volumes was accompanied by an increase in tubular diameter and length, with an increase in interstitial volume. From AR to TR, there was an increase in the tubular and total volumes, but, in this case, with a gradual increase in tubular diameter. Recovery of the seminiferous epithelium was accompanied by changes in apoptosis and proliferation activities. The first decreased halfway through the process, and the second remained higher than the control levels throughout the recrudescence stage. Alterations in the spermatozoa were ultrastructurally observed, which indicated that spermiogenesis was not yet completely normal. In conclusion, spontaneous testicular recrudescence in Syrian hamster comprises two histomorphometrical phases: the first related to an increase in tubular length and diameter and interstitial volume and the second depending principally on the gradual increase in tubular diameter. The restoration of the seminiferous epithelium is due to apoptosis reaching normal values in the AR group accompanied by higher proliferative activity than that observed in the Control group. 相似文献

In pregnant Syrian hamsters (Mesocricetus auratus) used as an animal model for studying the migration of fetal trophoblasts and the associated changes in maternal blood vessels, intravascular trophoblasts migrated well beyond the blood vessels of the uterus and into the vessels of the mesometrium. They migrated beyond the decidua of the uterus, into the lumina of maternal uterine and mesometrial arteries, but not into veins. The arterial changes, which were often segmental, resembled those seen in the decidua and consisted of a replacement of normal smooth muscle cells by poorly differentiated stromal cells. Ultrastructurally, the trophoblasts were either above or below maternal endothelial cells. They occurred also as single or multiple layers within the lumina of arteries that lacked an endothelial lining. Apparent penetration of the elastic membrane by the fetal trophoblasts brought them into close apposition to maternal cells in the arterial wall. Histochemical studies showed heightened metabolic activity of the intravascular trophoblasts as suggested by strong histochemical reactions to nonspecific esterase, succinic dehydrogenase and the glycerophosphate dehydrogenase reactions. Thus, these metabolically active fetal trophoblasts actively migrate into the maternal arterial system, resulting in loss of endothelial cells and changes in the wall of the maternal arteries similar to those in the decidua at the uteroplacental junction. 相似文献

The present study's aim was to investigate change of fatty acid composition of white adipose tissue with increasing age in Syrian hamster fed continuously alfalfa (LU) or cereal based diet (F2). A total of 43 Syrian hamsters with male and female, 22 head maintained with mating between sibling in a close herd fed F2 (GN) and 21 head selected for long‐term for large number of weaning pups per a mater using GN fed LU (ALF), were used. Fatty acid composition in adipose tissues taken from the back leg subcutaneous depot, periphery of kidney and reproductive organs at 3, 8 and 13 weeks of age were determined by using a gas chromatograph. Carbon (C)18:0 in GN tended to decrease and C18:1 increased significantly with increasing age, suggesting that Δ9‐desaturase had related adaptively. C18:3 in ALF increased significantly with increasing age. The present results indicate that, although the composition in unsaturated fatty acid (USFA) in both herds was different, the ratio of USFA to saturated fatty acid increased with increasing age suggesting that USFA in GN fed F2 was at a high level compared with in ALF fed LU. 相似文献

Interleukin 10 (IL-10) genes of Djungarian, Chinese, and Syrian hamsters were cloned. The clones of IL-10 consisted of 537 bp nucleotides and 178 amino acids in full length, and the nucleotide and amino acid sequences exhibited a high degree of homology with those of the mouse and human. Since the number and position of signal sequences, N-glycosylations and cysteine sites in the IL-10 amino acid sequences of the hamsters were the same as those of the mouse, we suggest that the IL-10 molecular structures of the hamster are closer to that of the mouse than human. 相似文献
