AIM:To investigate new methods for the expansion of human CD8+ memory T cells in vitro and provide novel means of anti-viral and anti-tumor adoptive immunotherapy. METHODS:Six kinds of stimuli, anti-CD3 antibody, anti-CD28 antibody, CD70, IL-2, IL-7 and IL-15, for the expansion of human CD8+ memory T cells in vitro were selected and arranged for their combinations, resulting in 63 kinds of stimulating combinations. Normal human CD8+ T cells were isolated and exposed to these stimuli. After 14 days of cell culture, the number and purity of CD8+ T cells, and the percentages of CD8+ central memory T cells (TCM) and CD8+ effector memory T cells (TEM) were detected. The expansion folds of CD8+ T cells, CD8+ TCM and CD8+ TEM were calculated and the relatively better stimulating combination was determined. RESULTS:The combination of anti-CD3 antibodies, IL-2 and IL-7 was a better method for the expansion of human CD8+ T cells, CD8+ TCM and CD8+ TEM in vitro, and the expansion folds were 13.19, 13.28 and 15.27, respectively. CONCLUSION:The combination of anti-CD3 antibodies, IL-2 and IL-7 is the relatively better method for the expansion of human CD8+ memory T cells in vitro.  相似文献

AIM:To investigate the effect of glucocorticoids, a kind of traditional immunosuppressive drug, on expanding central memory CD8+ T cells (CD8+ TCM) and to provide a novel method of generating CD8+ TCM for adoptive immunotherapy.METHODS:Healthy human peripheral blood mononuclear cells were isolated by density gradient centri-fugation. Naïve CD8+ T cells were further isolated with MACS microbeads and flow cytometry. After activated by cytokines, the cells were divided into experimental group and control group. Glucocorticoids at different concentrations and an identical volume of PBS were added. The phenotypic characteristics of TCM in different groups were measured by flow cyto-metry at separate time points. Furthermore, the effect of glucocorticoids on CD8+ T cell expansion was further verified using CFSE assay and Annexin V staining.RESULTS:Glucocorticoids significantly increased the proportion of CD8+ TCM in vitro. Glucocorticoids sustained CD8+ T cell expansion. Glucocorticoids had low toxicity for CD8+ T cells.CONCLUSION:Glucocorticoids effectively increase the number and proportion of CD8+ TCM in vitro, which provides novel insights into the generation of human CD8+ TCM and reveals a novel potential clinical application of glucocorticoids for immunotherapy.  相似文献

AIM: To investigate the amount and patterns of expressing CD69, IL-4 and IFN-γ on TCRVα24+ NKT cells, and compare with that of CD3+ T cells from human peripheral blood in response to in vitro stimulation. METHODS: The whole blood was stained with three-color immunofluorescence directly or after cultured with PDB+ionomycin (Ion) for 6 h, then the mononuclear cells were separated by lysing red blood cells. The expression rates of CD69, IL-4 and IFN-γ on TCRVα24+ NKT cells and CD3+ T cells were estimated by flow cytometer. RESULTS: As a proportion of mature T cells, the ratio of TCRVα24+ NKT cells to CD3+ T cells was about (1.34±0.42)%. The expression rates of CD69 on TCRVα24+ NKT cells and CD3+T cells in response to PDB+Ion for 6 h were (96.71±1.33)% and (98.60±0.47)%, respectively, while the ratio were (11.47±2.86)% and (1.07±0.45)% in the unstimulated group, and there were significant difference between them. The expression rates of IL-4 and IFN-γ on TCRVα24+ NKT cells stimulated with PDB+Ion for 6 h were (48.62±2.44)% and (46.65±8.91)%, respectively, which were significantly higher than that of unstimulated group [(31.57±3.31)%, (13.45±6.29)%] and that of stimulated CD3+ T cells, though the expression rates on stimulated CD3+ T cells were significantly higher than that of unstimulated CD3+ T cells. CONCLUSION: There is small amount of NKT cells in adult human peripheral blood. The expression rates of IFN-γ and IL-4 on these lymphocytes are higher than CD3+ T cells, suggesting that NKT cells are important immunomodulatory cells in special microvironments.  相似文献

AIM:To study the effect of adoptive transfer of CD4+ T cells with microRNA-7 (miR-7) knockdown (KD) on mouse acute liver injury model and to investigate its significance. METHODS:CD4+ CD62L+ T cells were purified from the spleen of normal wild-type (WT) mice and miR-7KD mice by magnetic bead sorting, and were stained with CFSE. These 2×106 CFSE-labeling cells were injected into normal mice via tail vein, and then the mouse acute liver injury model was induced by intraperitoneal injection of 30 mg/kg concanavalin A. After 72 h, the appearance, weight and weight index of the liver were investigated. The pathological change of the liver tissues was observed by HE staining. Real-time PCR was used to examine the mRNA expression of Bax and P53. The expression levels of CD62L, interleukin-4 (IL-4) and interferon-γ (IFN-γ) in the CD4+ T cells were analyzed by flow cytometry. RESULTS:We found that the liver tissue became lighter, and the weight (P<0.01) and weight index (P<0.05) were changed significantly in miR-7KD mice compared with control group. Moreover, HE staining showed that the liver cell damage was increased in the liver of miR-7KD mice. Meanwhile, the expression levels of Bax and P53 were significantly increased in miR-7KD group (P<0.05). The percentage of CD62L in CD4+ T cells was significantly decreased (P<0.01) in miR-7KD mice, with high expression of IFN-γ (P<0.05) and low expression of IL-4 (P<0.01) in CD4+T cells. CONCLUSION:These findings suggest that miR-7 knockdown significantly promotes the pathology of CD4+ T cell-mediated acute liver injury, which provides a preliminary experimental basis for further exploration on the mechanism of acute liver injury occurrence.  相似文献

AIM:To investigate the regulatory function of bone marrow-derived mesenchymal stem cells (MSCs) on T helper 17 cells (Th17) and regulatory T cells (Treg) in peripheral blood of severe asthmatic children. METHODS:MSCs were isolated, cultured and identified in vitro. MSCs digested with mitomycin were cocultured with T lymphocytes (TLC) at different ratios (1∶1, 1∶2, 1∶10 and 1∶20) from severe asthmatic children for 72 h. The proliferation of TLC was measured by CCK-8 method. In the coculture system of the 1∶2 ratio and the single TLC system, the supernatant levels of interleukin-17 (IL-17) and transforming growth factor-β (TGF-β) were measured by ELISA. The mRNA expression of retinoic acid-related orphan nuclear receptor C (RORC) and forkhead box protein 3 (Foxp3) in TLC was detected by qRT-PCR. RESULTS:After cocultured with MSCs, the proliferation of TLC decreased significantly in a dose-dependent manner (P<0.05). It also showed decreases in IL-17 (3 799±441 vs 4 890 ±373, P<0.05) and RORC mRNA level (1.21±0.14 vs 3.85±0.48, P<0.05), while an increase in TGF-β level (209±32 vs 117±26, P<0.05) was observed. No influence on the mRNA expression of Foxp3 was found (P>0.05). CONCLUSION:MSCs suppresses Th17 polarization of naive peripheral blood CD4+ T cells and matures Th17 cells secreting IL-17, which may effectively revise Th17/Treg imbalance of asthma.  相似文献

AIM: To investigate the synergistic effects of ampelopsin (AMP) and a chemotherapeutic drug mitomycin (MMC) on the proliferation of gastric cancer cell line SGC-7901.METHODS: SGC-7901 cells were cultured in vitro and divided into 4 groups: control group, AMP group, MMC group and AMP+MMC group. Cell proliferation was measured by MTT method. The apoptotic index was examined by flow cytometry. The expression of apoptotic proteins, Bcl-2 and survivin, was detected by Western blotting.RESULTS: AMP at the concentrations ranging from 2.2 mg/L to 14.84 mg/L exerted inhibitory effect on the growth of SGC-7901 cells. Cell proliferation in AMP (14.84 mg/L) group was inhibitory by (60.85±1.13) %, significantly higher than that in control group (P<0.05). The inhibitory rates of cell proliferation varied from (17.40±0.30) % to (72.23±1.36) % when the concentrations of MMC increased from 1×10-3 g/L to 1×10-2 g/L. AMP combined with MMC showed a synergistic effect on the growth of SGC-7901 cells. The inhibitory rates of cell proliferation varied from (21.83±2.50) % to (46.70±1.45) % when the concentrations of MMC increased from 1×10-3g/L to 5×10-3g/L. The inhibitory effect of AMP plus MMC was higher than that of MMC or AMP alone. The protein levels of Bcl-2 and survivin were inhibited in AMP group, MMC group and AMP+MMC group, and were significantly lower in AMP+MMC group than those in AMP group or MMC group.CONCLUSION: AMP enhances the inhibitory effect of MMC on the growth of SGC-7901 cells. The mechanism is related to the inhibition of apoptotic protein expression.  相似文献

AIM:To study the effects of ethyl pyruvate (EP) on brain tissues in neonatal rats with hypoxic-ischemic brain damage (HIBD) and its underlying mechanisms. METHODS:A total of 165 seven-day-old Sprague-Dawley (SD) rats were randomly divided into 3 groups:sham operation group (n=43), HIBD group (n=61) and HIBD+EP group (n=61). The rats in HIBD+EP group were intraperitoneally injected with EP (50 mg/kg) 30 min before operation, and once a day after surgery. Superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in brain homogenate, water content of brain and apoptotic cells in cortex were detected 3 days later. Ischemic and non-ischemic brain tissues were weighed to assess the extent of brain atrophy 14 days later. RESULTS:Higher level of SOD [(125.78±18.35)×103 U/(g protein)] and lower level of MDA [(4.42±1.04) μmol/(g protein)] in HIBD+EP group than that in HIBD group [(97.84±15.50)×103 U/(g protein) and (6.02±0.89) μmol/(g protein), respectively] was observed (P<0.05).In addition, the water content of ischemic hemisphere was significantly higher than that of non-ischemic one in HIBD group (P<0.05), and was indistinguishable from that of non-ischemic one after EP treatment (P>0.05), indicating the protective effect of EP against brain edema. The apoptotic cells in cortex and hippocampus in HIBD+EP group [(96.63±10.08)/field and (41.91±9.96)/field, respectively] were obviously decreased compared with HIBD group [(111.54±1.64)/field and (51.73±1.77)/field, respectively], but still higher than those in sham operation group (P<0.05). The atrophy ratio of ischemic hemisphere in HIBD+EP group was (13.25±5.19)%, significantly lower than that in HIBD group [(20.32±5.10)%, P<0.05]. CONCLUSION:Ethyl pyruvate is neuroprotective against HIBD in neonatal rats via increasing SOD level, decreasing MDA level, attenuating brain edema, decreasing apoptotic cells in cortex and alleviating atrophy of hypoxic-ischemic hemisphere.  相似文献

AIM: To study cellular and molecular mechanism involved in increasing susceptibility of infection in psychological stress persons. METHODS: Comparative studies were performed with double staining and flow cytometry analysis on immunophenotyping and in vitro expression of early activating surface molecule CD69 in response to mitogens on T cells from peripheral blood of 20 healthy college student volunteers before and after psychological stress. A series of term final examinations was defined as psychological stress. RESULTS: Immunophenotyping analysis showed no statistically significant difference in the percentage of CD2, CD3, CD4, CD8, CD19, CD20, CD16 and CD56 positive lymphocyte populations before and after psychological stress. There was a statistically significant decrease in the in vitro expression of CD69 in response to polyclonal stimulators on the T cells from persons after psychological stress than those before psychological stress. The percentage of CD69 expression (CD69+CD3+/CD3+%) in response to PHA and PDB in the whole blood culture for 72 hours decreased respectively from 28.1±4.1 and 80.7±6.8 on the T cells obtained before psychological stress to 17.6±3.8 and 65.8±7.9 on those obtained after psychological stress, while there was no statistically significant difference between the CD69 expression rates without stimulators on the T cells obtained before and after psychological stress. CONCLUSIONS: The effects of psychological stress to immune system is not on the level of changing proportions of the sub-populations within peripheral blood lymphocytes. Psychological stress can decrease the activating response of T cells in healthy persons, which may be responsible for the increase of susceptibility to infection in the psychological stress persons.  相似文献

AIM:To investigate the specific anti-tumor effects of mature dendritic cells (DCs) transfected with amplified mucin 1 (MUC1) mRNA in vitro. METHODS:DCs separated and purified from the peripheral blood mononuclear cells were induced in vitro and then identified by flow cytometry. pcDNA3.1(+)-MUC1 plasmid was constructed and was able to transcribe MUC1 mRNA in vitro. The MUC1 mRNA was transfected into DCs by electroporation. MUC1-transfected DCs were used to induce T cells to be cytotoxic T-lymphocytes. Quantitative real-time PCR was performed to assess MUC1 mRNA expression in transfected DCs. The proliferation of T cells was examined by MTT assay. The proportion of CD8+ cells in the T cells was determined by flow cytometry and the specific cytotoxicity was measured by LDH assay. The secretion of IFN-γ was detected by ELISA. RESULTS:The marker gene expression in the DCs transfected with MUC1 mRNA was significantly increased compared with control group, peaking at 24 h. The transfection group showed the higher capacity to stimulate the proliferation of T cells compared with control group when the ratio of DCs to T cells was 1∶10. The proportion of CD8+ cells in transfection group was higher than that in control group. The lethal effect of special cytotoxic T-lymphocytes on target cells in transfection group was stronger than that in control group. The level of IFN-γ in the cell supernatant of transfection group was higher than that in control group. CONCLUSION:DCs plus MUC1 mRNA by electrical transfection induces specific anti-tumor effects, which provides an experiment evidence of using MUC1 as a target for immunotherapeutic strategy against non-small cell lung cancer.  相似文献

AIM:To investigate the proliferation of CD4+CD25+ T cells from PBMCs of the gastric cancer patients and the inhibitory effect on CD4+CD25- T cells in vitro. METHODS:Magnetic activated cell sorting (MACS) method was used to separate CD4+CD25+T and CD4+CD25-T cells from peripheral blood monocytic lymphocytes in the gastric cancer patients, and then the purity and activity of CD4+CD25+T cells were analyzed with flow cytometer. After stimulated with anti-CD3 Ab, anti-CD28 Ab and rh IL-2, CD4+CD25- and CD4+CD25+ T cells were cocultured. The inhibitory effect of CD4+CD25+T on CD4+CD25-T cells was assayed by [3H] thymidine proliferation experiment. RESULTS:(1)After sorting, CD4+CD25+ T cells purity in healthy control and gastric cancer patients were 83.80%±1.84% and 84.13%±2.77%, respectively. No significant difference between the two groups (P>0.05) was observed. (2)The activity of CD4+CD25+ and CD4+CD25- T cells in healthy control and the gastric cancer patients after sorting were 98.52%±0.72% and 97.80%±0.95%. There was no significantly difference between the two groups (P>0.05). (3) CD4+CD25+ T cells obviously inhibited the CD4+CD25-T cell proliferation in vitro. The inhibition achieved to maximum in coculture of CD4+CD25+ T cells together with CD4+CD25- T cells (ratio of 1∶〖KG-*2〗1). CONCLUSION:The MACS system can effectively isolate CD4+CD25+ and CD4+CD25- T cells. After sorting, CD4+CD25+T cells obviously inhibit the proliferation of CD4+CD25- T cells in vitro and the inhibitory effect display an effect-target ratio relationship.  相似文献

AIM:To study the autophagy of prostate cancer PC-3 cells induced by CD147 in vitro. ME-THODS:The method of amino acid starvation to induce autophagy was used. The expression of CD147was detected by Western blotting. To study the functional effects of CD147 on autophagy in prostate cancer PC-3 cells, the down-regulation of CD147expression was induced by the technique of RNAi. The conversion of autophagic marker protein LC3-I to LC3-II was determined by Western blotting. The cell death after starvation-induced autophagy was analyzed by trypan blue exclusion assay. RESULTS:The CD147 expression gradually increased in starvation-induced autophagy. The down-regulation of CD147 significantly increased the expression of autophagy-related protein LC3-II compared with control group. Meanwhile, the cell death rates increased from (19.3±3.1)% and (22.3±3.5)% in control groups to (38.4±3.1)% in silencing the expression of CD147in the PC-3 cells (P<0.05). CONCLUSION:CD147 inhibits starvation-induced autophgy and autophagy death in the prostate cancer PC-3 cells.  相似文献

AIM:To investigate the influences of protein kinase C(PKC) inhibitors on the expression of interleukin-2(IL-2) and interferon-γ(IFN-γ) byin vitro activated T-lymphocytes. METHODS:Double fluorescent staining together with flow cytometry was adopted to detect intracellular cytokines and to analyze the effects of H7 and gossypol on IL-2 and IFN-γ expression levels of T-lymphocytes stimulated with phorbol ester (PDB)+ionomycin(I) in the presence of monensin.RESULTS:The expression rates of IL-2 and IFN-γ of CD3+ T cells stimulated with PDB+I for 4 h were 16.64±2.04 and 25.81±3.53(x±s), respectively, which were significantly higher than that of control (1.06±0.22 and 3.12±0.77)(P<0.05). Gossypol was able to inhibit the expression of IL-2 and IFN-γ significantly, with the expression rates of 2.08±0.12 and 9.01±1.90, respectively. At the presence of 50 μmol/L H7, the rates of IL-2+ and IFN-γ+ CD3+ T cells were 0.43±0.06 and 2.40±0.27, respectively. The effect of H7 was stronger than that of gossypol. CONCLUSION:PKC plays an important role in the expression of IL-2 and IFN-γ of CD3+T cells and its inhibitors H7 and gossypol exert significant inhibitory effect on the expression of these two cytokines. It is suggested that H7 and gossypol may have modulatory effect on T-cell-dependent specific immune responses by inhibiting PKC activity.  相似文献

AIM: Peripheral blood mononuclear cells (PBMC) were cultured in vitro to study the effect of gossypol, a polyphenolic antifertility agent, on the activation of normal human T cells. METHODS: Double fluorescent staining together with flow cytometry was adopted to analyze the influence of gossypol on expression of the early activation antigen CD69 on T-lymphocytes under stimulation of mitogen or phorbol ester. RESULTS:Analysis of T cell activation in vitro revealed that preincubation of PBMC with 100 μmol/L gossypol could completely inhibit the expression of early activation marker CD69 on CD3+ T cells in response to 10mg/L PHA, and block T cell activation by 10-7 mol/L PDB as well. The suppression of CD69 expression was dose-dependent and IC50 of gossypol on PDB and PHA were (35.7±2.9) μmol/L and (32.8±1.5) μmol/L(x ±s), respectively. Besides, gossypol had similar inhibitory effect on CD69 expression of CD3- lymphocytes. However, it did not have any significant effect on T cell surface molecule CD3 down-regulation. CONCLUSION: Gossypol could inhibit T cell activation in vitro in response to polyclonal activators, both PHA and PDB, suggesting that its action site may be at PKC or its downstream and that gossypol possessed potential immuno-regulatory effect.  相似文献

AIM: To investigate the basic biological characteristics of adult rat bone marrow mesenchymal stem cells(rBMMSCs), and compare to that of human BMMSCs (hBMMSCs). METHODS: rBMMSC and hBMMSCs were separated from bone marrow with the difference of adherence and Ficoll-Paque reagent, and expanded in culture medium in vitro, respectively. The proliferation and growth characteristics of the primary and different passage culture of rBMMSCs and hBMMSCs were analysed. The neural differentiation capacity of rBMMSCs with passages were observed. To detect the surface antigens of rBMMSCs, the labeled cells were analysed on a FACScan flow cytometer. The karyotype of rBMMSCs were detected by blocking cellular fission with colchicines. RESULTS: rBMMSCs and hBMMSCs have a strong self-renewal capacity. Approximately (4-8)×1012 and (3-4)×1012 cells were obtained after passage 15 in vitro, respectively. The ability of proliferation, CFU-Fs, and neural differentiation of rBMMSCs and hBMMSCs were decreased gradually with passages, but the ability of proliferation and CFU-Fs of rBMMSCs were higher than that of hBMMSCs at different passage. FACScan result showed rBMMSCs were uniformly positive for CD29 and CD44, and negative for CD11b, CD45, CD61, CD71, CD80, CD86,MHCⅠ and MHCⅡ. rBMMSCs had an normal karyotype, which had an average of 37.0±4.0 to 40.5±2.5 chromosomes. CONCLUSION: Adult rBMMSCs have strong self-renewal and neural differentiation capacity, and have an normal karyotype. So rBMMSCs can be used as the seed cells for tissue engineering.  相似文献

AIM: To investigate the roles of regulatory T cells (Tregs) on the function of effector T lymphocytes in tumor-draining lymph nodes (TDLNs). METHODS: The number expansion of Foxp3+ Tregs and CD4+ or CD8+ T cells in the TDLNs from mouse hepatocellular carcinoma model was detected by immunohistochemical staining and flow cytometry. Foxp3 mRNA expression was determined by real-time quantitative PCR. The ability of IFN-γ secretion in CD8+ T cells in the TDLNs was measured by enzyme-linked immunosorbent spot technique(ELISPOT). RESULTS: The expansions of Tregs and effector T cells were significantly increased in the TDLNs during tumor development. Tregs diffusely distributed in the CD8+ T cells occupancy area. The level of Foxp3 mRNA expression was significantly higher in the TDLNs than that in the inguinal lymph node (P<0.01) and spleen (P<0.01) from the same mouse inoculated Hepa1-6 cells. Tregs trended to accumulate within the TDLNs exclusively, but not in other peripheral lymph nodes(LNs) of the same host. Foxp3 mRNA expression was significantly higher in the spleen from the tumor mice than that from mice injected with LPS (P<0.01). Tregs suppressed the CD8+ T cells primed in the TDLNs that retained the ability to secrete IFN-γ via anti-CD3 stimulation. CONCLUSION: Tregs play an important role in regulating the function of CD8+ T cells. Deletion of Tregs could be crucial for establishment of tumor-specific immunotherapy.  相似文献

AIM:To compare the expression levels of Toll-like receptors (TLRs) in the granulocytes isolated from umbilical cord blood and adult peripheral blood. METHODS:The granulocytes in umbilical blood and adult peri-pheral blood were isolated by the method of density gradient centrifugation combined with red blood cell splitting. The purity of the cells was evaluated by flow cytometry. The mRNA expression levels of 10 TLRs were detected by RT-qPCR, and the protein levels of some TLRs were also tested by flow cytometry. RESULTS:The populations of CD19- CD24+ cells and CD3+ cells were (95.66±3.15)% and (4.19±1.54)% in neonatal granulocytes,respectively, and were (95.36±1.74)% and (4.30±0.96)% in adult granulocytes,respectively. The relative mRNA expression levels of TLRs in the granulocytes isolated from umbilical cord blood and adult peripheral blood were as follows: TLR1 0.141±0.091 and 0.691±0.447, TLR2 0.388±0.337 and 0.901±0.508, TLR4 0.093±0.071 and 0.254±0.147, TLR6 0.056±0.045 and 0.202±0.034, TLR7 0.001±0.001 and 0.004±0.003, and TLR8 0.046±0.040 and 0.211±0.146,and the diffe-rence had statistical significance (P<0.01). However, no difference in the expression levels of TLR3, TLR5, TLR9 and TLR10 between the neonatal and adult gra-nulocytes was observed (P>0.05). Among them, the mRNA expression of TLR3, TLR7 and TLR9 was at a low level in both neonatal and adult granulocytes. The protein level of TLR2 in adult gra-nulocytes (30.50±5.69) was higher than that in neonatal granulocytes (21.40±3.09, P<0.05). CONCLUSION:Low mRNA expression of TLR1, TLR2, TLR4 and TLR6, and low protein level of TLR2 in neonatal granulocytes indicate that the ability of recognizing bacterial pathogen by neonatal granulocytes may be defective or not yet fully mature.  相似文献

AIM:To observe the effect of acupuncture on CD4+ CD25+ Foxp3(forkhead box P3)+ regulatory T-cells(Treg cells) in rats with embryo implantation failure. METHODS:One hundred and forty-four pregnant rats were randomly divided into control group(N), mifepristone treatment group(M), mifepristone+acupuncture treatment group(A) and mifepristone+progestin treatment group(W). The rats in groups M, A and W were treated with mifepristone-sesame oil solution on day 1, while the rats in group N were injected with the same amount of sesame oil. The Housanli(ST36) and Sanyinjiao(SP6) points were selected for acupuncture. From day 1 to the time of death, the rats in group A were fasten up and then the acupuncture was performed. Accordingly, the rats in group N and group M were only fixed, and the rats in group W were given progestin. Implanted embryos in each group were counted. The proportions of CD4+ CD25+ Foxp3+ Treg cells in peripheral blood and CD4+ Foxp3+ Treg cells in the endometrium were detected by flow cytometry. The mRNA and protein levels of Foxp3 were determined by real-time PCR and Western blotting, respectively. RESULTS:Compared with group N, the number of implanted embryos, the percentages of CD4+ CD25+ Foxp3+ Treg cells in peripheral blood and CD4+ Foxp3+ Treg cells in the endometrium, and the expression of Foxp3 protein and mRNA in the endometrium were significantly decreased in group M(P<005). Compared with group M, the above indexes in group A and group W were significantly increased. CONCLUSION: The effect of acupuncture in rats with embryo implantation failure may be closely correlated with the modulation of CD4+ CD25+ Foxp3+ Treg cells.  相似文献

AIM:The expression of CD28, CD56 and CD57 on CD8+T cells in the peripheral blood of young (age range 20-35) and elderly(age range 60-75) healthy donors were compared to explore the change of the cellular immune function with aging.METHODS:Three-color fluorescent flow cytometry was performed to analyze the differences in percentage of CD8+CD28+, CD8+CD56+ and CD8+CD57+T cells in the peripheral blood between the young and elderly groups.RESULTS:CD8+CD28+T cells in the peripheral blood of the elderly group was significantly lower than those in the young group, with percentage of 34.07±5.28 and 49.84±7.43,respectively (P<0.05). Conversely, CD8+CD56+T cells and CD8+CD57+T cells in the peripheral blood of the elderly group were significantly higher than those in the young group, and the percentage was 6.60±2.40 vs 2.10±0.35,41.82±6.01 vs 22.89±2.80, respectively(P<0.05).CONCLUSION:The expression of CD28, CD56 and CD57 on the CD8+T cells in the peripheral blood are changed significantly with aging. The decrease in CD28 expression may play an important role in the immunosenescence, while the increase in CD56 and CD57 expression seems to be a compensatory adaptation for the immune dysfunction.  相似文献

AIM: Gene transduction with a recombined murine stem cell retroviral vector has been investigated to find an effective way of gene transduction and to offer theory and experimental basis for the recombined murine stem cell retroviral vector used for gene transduction. METHODS: 1. Construction of retrovirus vector: EC1-4 gene (repeats 1-4 of cadher in-5 extracellular domain) and mutant (Ser 222A) MEK1 gene were cloned into retrovirus vector pMSCV after cut by Bgl Ⅱ and EcoR 1 restriction endonuclease. 2. Obtaining CD41+ cells and cell culture: Cells expressing CD34+ from cord blood were isolated. The inducement of cells expressing CD41 from CD34+ cells was performed by using TPO and cells CD41+ were selected by FACS. NIH 3T3 cells were cultured in high sugar DMEM medium and U937 in RPMI 1640 medium. UT7 cells which is cytokine-dependent cell line were grown in Iscove's modified Dulbeco's medium supplemented by GM-CSF. 3. Determination of viral titers: Retroviral vectors were transferred to packaging cell line 293. Retroviral containing supenatant was collected after transfection. The viral titers was tested on infection of NIH 3T3 cells by FACS analysis. 4. Western blot: Transduced CD41+, UT7, U937 and MDA-MB-435 cells were analyzed by western blot to examine expression of transduced genes. RESULTS: A packaging cell line 293 produces high-titer MEK1 pMSCV retroviruses (3.1×107) and EC1-4 pMSCV retroviruses (1.0×108). With 8-folds dilution retroviruses, 60.73% GFP positive cells have been obtained in MEK1 pMSCV transduced UT7 cells, 72.56% in U937 cells and 30.57% in CD41+ cells, respectively. GFP positive cells have reached up 97.54 % in EC1-4 pMSCV transducted MDA-MB-435 cells. Phosphorylated MEK1 has been decreased in experiment group when TPO has stimulated CD41+ and UT7 cells or serum has stimulated U973 cells. This indicates that is a dominant negative effect of mutation MEK gene. EC1-4 gene transduced MDA-MB-435 cells have expressed EC1-4. CONCLUSION:The recombined murine stem cell retrovirus can effectively mediate gene transduction of CD41+,UT7,U937 and MDA-MB-435 cells,and transduced genes can be stably expressed.  相似文献
