Neglected and underutilized species often play a vital role in securing food and livestock feed, income generation and energy needs of rural populations. In spite of their great potential little attention has been given to these species. This increases the possibility of genetic erosion which would further restrict the survival strategies of people in rural areas. Ziziphus spina-christi is a plant species that has edible fruits and a number of other beneficial applications that include the use of leaves as fodder, branches for fencing, wood as fuel, for construction and furniture making, and the utilization of different parts e.g. Fruits, leaves, roots and bark in folk medicine. Moreover, the plant is adapted to dry and hot climates which make it suitable for cultivation in an environment characterized by increasing degradation of land and water resources. Lack of research in Z. spina-christi hinders its successful improvement and promotion. Therefore, studies are needed to fully exploit this species. This article aims at summarizing information on different aspects of Z. spina-christi to stimulate interest in this crop which is of importance in Sudan and other countries of the semi-arid tropics.

This study aimed to investigate the effect of inoculation with plant growth-promoting Rhizobium and Pseudomonas species on NaCl-affected maize. Two cultivars of maize (cv. Agaiti 2002 and cv. Av 4001) selected on the basis of their yield potential were grown in pots outdoors under natural conditions during July. Microorganisms were applied at seedling stage and salt stress was induced 21 days after sowing and maintained up to 50% flowering after 120 days of stress. The salt treatment caused a detrimental effect on growth and development of plants. Co-inoculation resulted in some positive adaptative responses of maize plants under salinity. The salt tolerance from inoculation was generally mediated by decreases in electrolyte leakage and in osmotic potential, an increase in osmoregulant (proline) production, maintenance of relative water content of leaves, and selective uptake of K ions. Generally, the microbial strain acted synergistically. However, under unstressed conditions, Rhizobium was more effective than Pseudomonas but under salt stress the favorable effect was observed even if some exceptions were also observed. The maize cv. Agaiti 2002 appeared to be more responsive to inoculation and was relatively less tolerant to salt compared to that of cv. Av 4001.  相似文献

Present study reported the variation in electrophoretic seed subfraction (albumins, globulin A, globulin B, glutelins) patterns in some Lathyrus species collected from their natural habitats different localities in Turkey. Electrophoretic data were documented by using a gel documentation system (Bio-Rad, USA) and analysed by using Quantity 1-D analysis software and also the dendograms were formed with 4.0% tolerance in UPGAMA (Unweighed Pair-Group Arithmetic Mean). The differences among species were observed and all seven species were clearly identifiable from the protein patterns. The dendograms obtained from cluster analysis of seed storage proteins data showed examined species have displayed a number of distinct relationships. According to our results, it seems justified to recommend a wider use of electrophoretic analysis of seed storage proteins in taxonomic investigations of leguminous plants.  相似文献

An intense exploration of the Spanish Cantabrian coast for the presence or absence of wild Brassica oleracea L., yielded 24 new localities to be added to the 21 previously known. Of the resulting 45 localities, 22 correspond to Asturias, 11 to Cantabria and 12 to the Basque Country. Data on the habitat requirements of this plant have been annotated, the conservation status of each population has been estimated and seed samples have been collected for long-term preservation. As a whole, wild B. oleracea is not threatened in Northern Spain, but some populations and/or sub-populations are at risk either because of their small size or some detectable human impact.  相似文献

This study reports for the first time the presence of diazotrophic bacteria belonging to the genera Achromobacter and Zoogloea associated with wheat plants. These bacterial strains were identified by the analysis of 16S rDNA sequences. The bacterium IAC-AT-8 was identified as Azospirillum brasiliense, whereas isolates IAC-HT-11 and IAC-HT-12 were identified as Achromobacter insolitus and Zoogloea ramigera, respectively. A greenhouse experiment involving a non-sterilized soil was carried out with the aim to study the endophytic feature of these strains. After 40 days from inoculation, all the strains were in the inner of roots, but they were not detected in soil. In order to assess the location inside wheat plants, an experiment was conducted under axenic conditions. Fifteen days after inoculation, preparations of inoculated plants were observed by the scanning electron microscope, using the cryofracture technique, and by the transmission electron microscope. It was observed that all isolates were present on the external part of the roots and in the inner part at the elongation region, in cortex cells, but not in the endodermis or in the vascular bundle region. No colonizing bacterial cells were observed in wheat leaves.  相似文献

The karyomorphology for eight diploid species of Arachis belonging to three sections has been described for the first time, Sect. Extranervosae: A. macedoi (2n = 20m) and A. retusa (2n = 14m + 6sm); Sect. Heteranthae: A. sylvestris (2n = 16m + 4sm); Sect. Procumbentes: A. chiquitana (2n = 18m + 2sm); Sect. Arachis: A. cruziana (2n = 18m + 2sm), A. herzogii (2n = 18m + 2sm), A. simpsonii (2n = 20m) and A. williamsii (2n = 20m). A pair of satellited chromosomes was observed in all species. A chromosomes were found in A. chiquitana, A. herzogii and A. simpsonii. Karyotypic differences between sections were observed, but not enough to establish a characteristic karyotype pattern for each section. However, the species may be differentiated by the presence of A chromosomes, the type and position of satellites, and the karyotype formulae. These results are discussed with regard to karyotype evolution in Arachis to contribute to understanding the role of chromosome changes in the evolution of the genus.  相似文献

Trigeneric hybrids may help establish evolutionary relationships among different genomes present in the same cellular-genetic background, and also offers the possibility to transfer different alien characters into cultivated wheat. In this study, a new trigeneric hybrid involving species from the Triticum, Psathyrostachys and Secale was synthesized by crossing wheat-P. huashanica amphiploid (PHW-SA) with wheat-S. cereale amphiploid (Zhongsi 828). The crossability of F1 hybrid was high with 35.13%, and the fertility was 41.95%. The morphological characteristics of F1 plants resembled the parent Zhongsi 828. The trigeneric hybrids pollen mother cells (PMCs) regularly revealed averagely 19.88 univalents, 9.63 ring bivalents, 3.97 rod bivalents, 0.60 trivalents and 0.03 tetravalents per cell. Multivalents consisted of trivalents and tetravalents can be observed in 52.7% of cells. A variation of abnormal lagging chromosome, micronuclei and chromosome bridge were formed at anaphase I and telophase II. The mean chromosomes number of F2 progenies was 2n = 46.13, and the distribution range was 42–53. GISH results revealed that most F2 plants had 6–12 S. cereale chromosomes, and only 0–2 P. huashanica chromosomes were detected. The results indicated that S. cereale chromosomes can be preferentially transmitted in the F2 progenies of trigeneric hybrid than P. huashanica chromosomes. A survey of disease resistances revealed that the stripe rust resistance from the PHW-SA were completely expressed in the F1 and some F2 plants. The trigeneric hybrid could be a useful bridge for the transference of P. huashanica and S. cereale chromatins to common wheat.  相似文献

Morphological variation of the coconut fruit measured in situ has been used to estimate genetic diversity, and generate hypotheses about the evolutionary and geographical diffusion of coconut. Some authors have questioned the validity of this methodology due to the possibly high effect of the environment on the morphological characteristics of the fruit. The general aim of this study is to validate this methodology through: (1) characterizing the pattern of morphological variation of the fruit under homogeneous growing conditions ex situ; (2) comparing this pattern with those already reported in situ; (3) estimating the heritability values for the components of fruit in coconut. Results are also discussed in comparison with ex situ leaf variation and biochemical and molecular variation patterns previously studied. Principal components and discriminant analyses indicated that the characters that best differentiate groups are basically the same in situ and ex situ. Grouping patterns obtained with principal components and cluster analysis were similar for both growing conditions. They were also similar to the grouping pattern obtained with ex situ leaf characters. No significant differences were found in the variation coefficients of fruit characters between the same populations in situ and ex situ. Consistency was found between patterns of morphological variation of fruit ex situ and in situ, and those obtained using iso-enzymatic and molecular characteristics. The results are also consistent with hypotheses on the origin and diffusion of the germplasm introduced to Mexico. High values of heritability were found in nine characters. Weight and water percentage showed the highest values (0.88 and 0.883), with a strong correlation to the mass and the roundness of fruit and seed, suggesting that human selection of these characteristics led to the differentiation of domesticated populations. We conclude that morphological characterization of the coconut fruit in situ is useful to estimate its genetic variability because of its simplicity, speed and ease of application in the field and in remote areas.  相似文献

Paracetamol, the most widely and globally used analgesic and antipyretic, is easily accumulated in aquatic environments. In the present study, the biodegradation of paracetamol in different media (one for general growth, one specific for sulfate reducing bacteria, a mineral salts medium and municipal wastewater) inoculated with two types of sludge (from anaerobic lagoon and from oxidation ditch) under different oxygenic conditions (anoxic; moderate oxygenation in open flasks and high oxygenation by aeration) was investigated. In addition, bacteria with relative abundances increasing simultaneously with paracetamol degradation, when this drug was the only carbon source, thus with a putative role in its degradation, were identified using 16S rRNA gene sequences. The results show that aerobic microorganisms had a major role in the degradation of paracetamol, with 50 mg/L totally removed from municipal wastewater after 2 days incubation with aeration, and that the metabolites 4-aminophenol and hydroquinone plus one compound not identified in this work were produced in the process. The identification of bacteria with a role in the degradation of paracetamol revealed a strain from genus Pseudomonas with the highest final relative abundance of 21.2%, confirming previous works reporting strains of this genus as paracetamol decomposers. Besides, genera Flavobacterium, Dokdonella and Methylophilus were also in evidence, with initial relative abundances of 1.66%, 1.48 and 0.00% (not detected) in the inoculum and 6.91%, 3.80 and 3.83% after incubation, respectively. Therefore, a putative role of these genera in paracetamol biodegradation is suggested for the first time.

Between 1930 and 2003 with emphasis on the 1940s maize landraces (Zea mays L. ssp. mays) from all over Switzerland were collected for maintenance and further use in a new Swiss breeding program. The genetic relationship and diversity among these accessions stored in the Swiss gene bank is largely unknown. Our hypothesis was that due to the unique geographic, climatic, and cultural diversity in Switzerland a diverse population of maize landraces had developed over the past three centuries. The aims were to characterize the genetic diversity of the Swiss landraces and their genetic relationship with accessions from neighbouring regions as well as reviewing their history, collection, and maintenance. The characterization and grouping was based on analyses with ten microsatellite markers. Geographic, cultural, and climatic conditions explained a division in two distinct groups of accessions. One group consisted of landraces collected in the southern parts of Switzerland. This group was related to the Italian Orange Flints. The other group contained accessions from northern Switzerland which were related to Northern European Flints in particular German Flints. Historic evidence was found for a frequent exchange of landraces within the country resulting in a lack of region-specific or landrace-specific genetic groups. The relatively large separation between the accessions, indicated by high F ST (0.42), might be explained partly by a bottleneck during the collection and maintenance phase as well as by geographical and cultural separation of north and south of the country. Due to the high genetic diversity, the accessions here are a potential resource for broadening the European flint pool.  相似文献

Using the analysis of nine isozyme systems and cladistic analysis of 32 morphological characters of the Mexican species of the section Dutra of the genus Datura, evidence was sought on the origin of the cultivated D. metel L. The genetic similarity and the phylogenetic relationship suggest that D. metel is related more closely to D. inoxia Mill. than to the other taxa of the section Dutra based upon the small genetic distance between them. The cladistic analysis revealed two main clades: the long-lived, tuberous rooted perennials (D. inoxia, D. lanosa A.S. Barclay ex Bye, D. metel, and D. wrightii Regel) and the tap-rooted annuals (D. discolor Bernh., D. kymatocarpa A.S. Barclay, D. leichhardtii F.V. Muell. ex Benth. ssp. pruinosa (Greenm.) Barclay ex Hammer (syn.: D. pruinosa Greenm.), D. reburra A.S. Barclay). Datura inoxia is the sister taxon of D. metel next to which is D. wrightii while D. lanosa is the basal taxon of this group. The combination of genetic and cladistic data indicates that D. inoxia is most likely the progenitor of D. metel.  相似文献

The genus Aegilops L. is a very important genetic resource for the breeding of bread wheat Triticum aestivum. Therefore, an accurate and easy identification of Aegilops species is required. Traditionally, identification of Aegilops species has relied heavily on morphological characters. These characters, however, are either not variable enough among Aegilops species or too plastic to be used for identification at the species level. Molecular markers that are more stable within species, therefore, could be the alternative strategy towards an accurate identification. Since the chloroplast DNA has a lower level of evolution compared to the nuclear genome, an attempt was made in this study to investigate polymorphism in the chloroplast DNA among 21 Aegilops species (including Ae. mutica that is now known as Amblyopyrum muticum) and between the latter and T. aestivum to generate markers for the diagnosis of all targeted species. Cleaved amplified polymorphic sequence (CAPS) applied on 22 coding and non-coding chloroplast regions using 80 endonucleases and sequencing of two of those regions revealed little polymorphism between T. aestivum and the various Aegilops species examined and to a less extent was the variation among Aegilops species. Polymorphism observed among species analysed allowed the discrimination of T. aestivum and 12 Aegilops species.  相似文献

A collection of 29 lines of Spanish einkorn wheat (Triticum monococcum L. ssp. monococcum) were analysed for morphological traits and waxy proteins. Four different botanical varieties were detected in the collection on the basis of the morphological traits. For the waxy proteins, no polymorphism has been described in previous works in wheat diploid species. In the current materials, two different alleles were found, namely Wx-A m 1a and Wx-A m 1a′. The allele Wx-A m 1a′ that presents less mobility than the Wx-A m 1a, has not been previously described and was only found in one line.  相似文献

DNA sequences of nuclear gene Got2 was studied in 60 accessions of Aegilops tauschii, 29 of subsp. tauschii and 31 of subsp. strangulata. It was found that Got2 allozyme polymorphism in Ae. tauschii is due to a single, unique, mutation which led to replacement of glutamic acid by isoleucine in residue 256 of the enzyme molecule, encoded by Got2. As revealed by Got2 DNA sequences variation, initially in its history Ae. tauschii was presented by subsp. strangulata, and among phylogenetic lineages of subsp. strangulata, the lineage “t-91s” (TauL3) is the most ancient, a relict one. Subspecies tauschii is relatively “young”. Initially it was presented by the lineage marked by combination of allozyme alleles Got2 105 and Acph1 100. In the past it inhabited the Continental area from Caucasia to Pakistan, but later on it was forced out by newly originated, now—a major lineage of subsp. tauschii, marked by Got2 100. This lineage extended the Continental area of the species up to Kirgizstan, but actually failed to penetrate into pre-Caspian area, occupied by subsp. strangulata. These results essentially differ from those obtained previously, using chloroplast DNA (cpDNA) sequences polymorphism. As revealed by cpDNA, the major, “usual”, subsp. strangulata (TauL2) is “younger” than subsp. tauschii, which resided on phylogenetic tree between relict lineage “t-91s”of subsp. strangulata—and major subsp. strangulata. But both cpDNA and Got2 DNA sequences indicate that the level of genetic variation in subsp. tauschii is much lower than in subsp. strangulata. According to Got2 DNA sequences variation, it was Ae. tauschii subsp. strangulata lineage “k-109″ which donated genome D to Triticum aestivum L. This lineage includes accessions: k-109 from South-Eastern Precaspian Azerbaijan; KU-2105, KU-2159 from Western Precaspian Iran; KU-2080 from Eastern Precaspian Iran.  相似文献

Cowpea landraces belonging to Vigna unguiculata (L.) Walp. subsp. unguiculata cv.-gr. unguiculata and cv.-gr. sesquipedalis collected from part of the Deccan Plateau and West Coast of India were evaluated to (i) identify the diverse source(s) of variation for improved characters like pods/peduncle and seed index (ii) study the response of landraces for adaptation to drought and heat stress and (iii) understand the breeding value of a landrace in the genetic improvement of a popular cowpea cultivar. Landraces were evaluated for various morphological characters, pods/peduncle, seed index and other economically important agronomic traits, rust resistance and drought and heat tolerance in different years and environments. Landraces were found as an important source of genetic variability for pods/plant, pods/peduncle, better pod filling ability (seed index), grain yield/plant as well as drought and heat tolerance and rust resistance. Hybridization between C 152 (cv.) and DWDCC 016 (landrace) resulted in release of new variation not present in the two parents. Thus the landrace, DWDCC 016, can be utilized to improve cultivated varieties by transferring to them the economically valuable traits like pods/peduncle and seed index thereby enhancing realisation of sink potential and ultimately grain yield in a sustainable way.  相似文献

The center of origin of maize is in Mexico and maize diversity is very high in many parts of the country. Here we describe and analyze the geographic distribution and diversity of maize using a database of 18,176 georeferenced observations of maize races. The number of observations by race follows the common “hollow curve” distribution, with six races comprising 54 % of the records and 37 races only 10 %. Few races are widespread, and some have a highly localized distribution. The average maximum distance (D max) between two records of the same race was 1169 km, but seven races had a D max lower than 200 km. The average circular area range size (CA25) was 51, but 14 races had a CA25 lower than 10. Some races have very few observations but were found over a large area. These races may not be valid and their classification should be reviewed. Areas of high race richness and diversity are in mountainous northwest, south central, and southern Mexico in the states of Oaxaca, Jalisco, Michoacán and Chiapas. Six contiguous maize regions were defined using hierarchical clustering. Despite the high sample size, further sampling could improve estimates of race diversity in several regions.  相似文献

Genetic diversity of populations stored ex situ or in situ can be altered due to the management practices they are subjected to. In this paper, we compare populations of two common bean (Phaseolus vulgaris L.) landraces grown on farms with material collected from the same farms and now kept in two ex situ collections (CIAT and REGEN) with the purpose to monitor any changes that have occurred due to ex situ conservation. The diversity was measured using seven bean microsatellite markers. Further phenotypic and developmental traits were registered in a field experiment. Compared with the in situ populations, the ex situ ones had a lower level of gene diversity and we suggest that this is due to the regeneration process. Most of the phenotypic traits did not differ significantly between ex situ and in situ populations, although for yield and 100-seed weight, the CIAT material showed significant lower values. We assume that these populations have gone through an adaptational change. Overall, the conservation ex situ has been successful in maintaining the majority of the adaptations found in the landraces studied, however, the probable loss of genetic diversity that we have observed, suggest that protocols for the regeneration process must be carefully worked out if the majority of alleles are to be preserved for the future. This study also highlights the complementarity of ex situ and in situ conservation methods in order to preserve landrace adaptations and to capture new, useful diversity generated in in situ populations.  相似文献

The present study was conducted to isolate and characterize rhizobial strains from root nodules of cultivated legumes, i.e. chickpea, mungbean, pea and siratro. Preliminary characterization of these isolates was done on the basis of plant infectivity test, acetylene reduction assay, C-source utilization, phosphate solubilization, phytohormones and polysaccharide production. The plant infectivity test and acetylene reduction assay showed effective root nodule formation by all the isolates on their respective hosts, except for chickpea isolate Ca-18 that failed to infect its original host. All strains showed homology to a typical Rhizobium strain on the basis of growth pattern, C-source utilization and polysaccharide production. The strain Ca-18 was characterized by its phosphate solubilization and indole acetic acid (IAA) production. The genetic relationship of the six rhizobial strains was carried out by random amplified polymorphic DNA (RAPD) including a reference strain of Bradyrhizobium japonicum TAL-102. Analysis conducted with 60 primers discriminated between the strains of Rhizobium and Bradyrhizobium in two different clusters. One of the primers, OPB-5, yielded a unique RAPD pattern for the six strains and well discriminated the non-nodulating chickpea isolate Ca-18 from all the other nodulating rhizobial strains. Isolate Ca-18 showed the least homology of 15% and 18% with Rhizobium and Bradyrhizobium, respectively, and was probably not a (Brady)rhizobium strain. Partial 16S rRNA gene sequence analysis for MN-S, TAL-102 and Ca-18 strains showed 97% homology between MN-S and TAL-102 strains, supporting the view that they were strains of B. japonicum species. The non-infective isolate Ca-18 was 67% different from the other two strains and probably was an Agrobacterium strain.  相似文献

A novel dwarf mutant of Brassica napus L. em. Metzg., named NDF-1, was derived from a high doubled haploid line ‘3529’ of which seeds were jointly treated with chemical inducers and fast neutron bombardment. The germination results showed that the germination of NDF-1 was insensitive in response to exogenous gibberellic acid 3 (GA3). The studies on growth response to exogenous GA3 showed that NDF-1 seeding has at least 10-fold insensitivity than the wild-type. Moreover, no matter what concentrations of GA3 were added to the seedlings and adult plants, the NDF-1 could not restore the wild type phenotype. These results indicated that the B. napus dwarf mutant NDF-1 was GA-insensitive mutant. The histological observations showed that the key reason of leading NDF-1 to dwarf was the reduction of hypocotyls and stems cell numbers.  相似文献
