Effects of hydrogen peroxide (H2O2), as exogenous reactive oxygen, on browning and active oxygen metabolism in pericarp of harvested ‘Fuyan’ longan fruit were investigated. The results showed that as compared with the control fruit, there was a higher browning index in pericarp of H2O2-treated fruit. The fruit treated with H2O2 resulted in increased rate of superoxide anion (O2−) production, reduced activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), decreased amounts of ascorbic acid (AsA), glutathione (GSH) and carotenoid, and increased malondialdehyde (MDA) content. These results indicated that H2O2-induced browning in pericarp of harvested longan fruit might be due to a reducing capacity of active oxygen scavenging and an increase of accumulation of O2−, which might stimulate membrane lipid peroxidation, disrupt cellular membrane structure, and cause the loss of cellular compartmentalization, in turn, resulting in the contact of polyphenol oxidase (PPO) and peroxidase (POD) with phenolic substrates and subsequently oxidation phenolics to form brown polymers. 相似文献

Pericarp colour of litchi fruit is an important quality attribute that determines its market value and consumer acceptance. Plant growth regulators (PGR) such as abscisic acid (ABA) and ethephon are known to play important roles in peel colour development during maturation and ripening of non-climacteric fruits (e.g. grape and litchi). Our aim was to investigate the effects of preharvest application of ABA, ethephon and their combination on pericarp colour and fruit quality of litchi (cv. Calcuttia) and also to assess the potential effects on postharvest performance of fruit. Exogenous application of ABA (150 or 300 mg L−1) at the colour-break stage significantly increased the concentration of total anthocyanins and cyanidin-3-O-rutinoside, the major anthocyanin contributing ∼71–96% of the total anthocyanins, in litchi pericarp compared to ethephon (500 μL L−1). Among different anthocyanins quantified, the relative contribution of cyanidin-3,5-diglucoside to the total anthocyanins was significantly higher in all PGR-treated fruit compared to the control, but the concentration of cyanidin-3-O-glucoside was specifically enhanced by ABA. No significant effect on the concentrations of epicatechin, and quercetin-3-O-rutinoside was observed in response to PGR treatments. Ethephon (500 μL L−1) treatment did not significantly increase the anthocyanin levels in pericarp, but it caused more degradation of chlorophyll pigments than control. Aril quality with regard to firmness, soluble solids and acidity was not significantly affected by PGR treatments, except that ethephon-treated fruit showed significant softening and lower acidity. Postharvest changes in fruit quality attributes including pericarp browning during cold storage at 5 °C for 14 d were mainly related to the storage duration effect, rather than PGR treatment. In conclusion, ABA treatment (150 or 300 mg L−1) at the colour-break stage enhanced anthocyanins accumulation in litchi pericarp without adversely affecting postharvest quality and storage stability for 14 d. 相似文献

Harvested papaya fruit are perishable due to rapid ripening and softening and susceptibility to biotic or abiotic stresses. Hot water treatment (HWT) can preserve fruit quality by reducing decay. The present study investigated effects of HWT on controlling fungal pathogens of papaya fruit and the possible mechanism by which HWT induced disease resistance. HWT (54 °C, 4 min) of papaya fruit had a pronounced effect on reducing the carrier rate of Colletotrichum gloeosporioides (C. gloeosporioides) in fruit peel, significantly inhibited the incidence of anthracnose and stem-end rot, effectively delayed fruit softening, but slightly promoted the rate of fruit coloring. HWT reduced the anthracnose index and fruit ripeness to a certain extent and induced changes in the wax arrangement on the surface of treated fruit, causing the wax to melt. The cracks and most stomata appeared to be partially or completely plugged by the melted wax, thereby providing a mechanical barrier against wound pathogens. HWT induced the expression of CpPGIP and promptly induced the expression of CpNPR1, and then regulated the expression of the CpPR1 gene, which may enhance the resistance of the fruit to anthracnose disease and reduce the decay rate. Together, these results confirm that HWT could reduce disease incidence and induce resistance, and thus maintain postharvest quality during storage and prolong the shelf-life of papaya fruit. 相似文献

ROP/RAC GTPases regulate various development processes and play important roles in plant defense responses. Recently, lignification or secondary cell wall formation related ROP members were reported in rice, zinnia, cotton and Eucalyptus. The present study aimed to investigate the possible association of loquat ROPs with flesh lignification under different temperatures. Four ROP cDNA fragments, EjROP1.1, EjROP1.2, EjROP2 and EjROP3, were isolated from ‘Luoyangqing’ (LYQ) loquat fruit, and all of them shared over 80% nucleotide identity with known ROPs from other plants. Sequence analysis revealed that EjROP1.1, EjROP2 and EjROP3 might be functional while EjROP1.2, with mutated C-terminal resulted from a 65 bp deletion in the corresponding nucleotide sequence as compared with EjROP1.1, might be dominant-negative and consequently act as a negative regulator of ROP signal transduction. Increase in expression of EjROP1.1, EjROP2 and EjROP3 was observed during first 4 or 6 d of storage at 20 °C and was positively correlated with the increase in flesh firmness. Expression of EjROP1.2 was constantly low under 20 °C but was quickly, within 6 h, induced under 0 °C, and it increased by about 20 times within 24 h. The expression was induced under 5 °C as well but not so strong as that under 0 °C, and transfer of fruit from 5 °C to 0 °C re-stimulated the expression. The possible roles of EjROPs played during senescence and cold regulated lignification was discussed, and the simultaneous increase in the expression of three functional EjROPs and the negative regulator EjROP1.2 was suggested to be important for maintaining a ROP rheostat to protect cells from excessive lignification. To our knowledge, this is the first study on a dominant-negative ROP resulted from a deletion mutation, and a ROP responded to low temperature. 相似文献

The genetics of photoperiod sensitivity, flowering date, fruit size, gender, female fertility, and disease resistance were
investigated in progeny between sets of elite F. virginiana selections and F. × ananassa cultivars and selections planted at sites in Michigan, Minnesota and Ontario. Progeny means varied considerably for all the
production traits. Most notable were the large fruit and high fertility observed in crosses with High Falls 22 at all three
sites, and Montreal River 10 in Ontario and Michigan. Fragaria virginiana ssp. virginiana parents yielded progeny with much larger fruit than F. virginianassp. glauca parents. General combining ability was significant for all traits at all locations, while specific combining ability was
significant for only fruit diameter, ovule set and fruit set in Michigan. Overall, the highest number of day-neutral genotypes
were detected in Ontario (mean =44%) compared to Minnesota (31%) and Michigan (26%). In progeny populations of day-neutral
F. × ananassa × short-day F. virginiana almost all fit the 1:1 ratio expected if day-neutrality is regulated by a single dominant gene; however, only a few families
of short-day F. × ananassa ×day-neutral F. virginianacrosses fit a 1:1 ratio. Likewise, in progeny of day-neutral F. virginiana ×day-neutral F × ananassa crosses, only a few of them fit the 3:1 ratio expected if day-neutrality is regulated by a single dominant gene. These data
suggest that it should be relatively easy to useF. virginiana germplasm in strawberry cultivar improvement, and that several different sources of day-neutrality may exist in natural populations.
