The goals of the present study were to determine whether the infusion of a glucose solution into the portal vein is tolerated in cows and whether the glucose concentration differs after administration of glucose into the jugular vein and portal vein. Fifteen healthy Swiss Braunvieh cows were used. An indwelling catheter was placed in both jugular veins and a balloon-tipped indwelling catheter with a diameter of 2 mm was placed in the portal vein under the guidance of ultrasonography. Three cows received 500 ml of 20% glucose solution over 60 min via the left jugular vein. Three other cows received the same solution over 60 min via the portal vein. Blood samples were collected from the right jugular vein before and for 24 h after the infusion of glucose for the determination of the concentrations of glucose and bilirubin and the activities of glutamate dehydrogenase, sorbitol dehydrogenase and gamma-glutamyl transferase. Infusion via the portal vein did not result in abnormalities in the general condition of the cows or increases in the concentration of bilirubin or the activities of liver enzymes. The blood glucose concentration increased to the same extent after both intraportal and intrajugular infusion. Over a 12-h period, three cows received 10 l of 20% glucose solution via the left jugular vein and three others received the same solution over a 12-h period via the portal vein. Blood samples were collected from the right jugular vein before and for 30 h after the start of infusion. Infusion via the portal vein did not affect the general condition of the cows or the activities of the liver enzymes. There was no significant difference in the blood glucose concentration between the two groups throughout the study.  相似文献

An ultrasound-guided portocentesis was performed in 21 clinically healthy cows, and blood was collected for haematological, biochemical and blood-gas analyses. At the same time blood samples were collected from the left jugular vein of each cow for the same analyses. Immediately after the centesis, portal blood pressure was measured. The cows were examined daily thereafter for eight days and then slaughtered for postmortem examination of the internal organs. The mean (sd) portal blood pressure was 37.1 (5.47) mmHg, and ranged from 24 to 49 mmHg. There were small but significant differences between many of the haematological and biochemical parameters in jugular and portal blood, and the concentration of bile acids was on average 2.7 times higher, and the concentration of ammonia was 19.3 times higher in portal blood. The demeanour and behaviour of the cows, and their appetite and rectal temperature remained normal during the eight-day observation period. The haematocrit, total and differential leucocyte counts, and the concentrations of total solids and fibrinogen were not affected by portocentesis. On postmortem examination, no lesions were apparent in the peritoneum, liver and portal vein.  相似文献

The effects of barley flour on the fermentation parameters of alfalfa silage and on the productivity of dairy cows were investigated. Alfalfa forage was ensiled either with or without barley flour. Barley flour was soaked in water for 24 h before being mixed with alfalfa (12 kg: 100 kg dry matter bases) at ensiling. Eighteen multi-parous cows were assigned to three equal treatment groups using a completely randomized design. Three isocaloric and isonitrogenous total mixed rations containing alfalfa hay, ordinary alfalfa silage or barley flour mixed alfalfa silage were then prepared. The concentrations of ammonia nitrogen, acetic acid and butyric acid were lower in barley flour mixed alfalfa silage compared to that in ordinary alfalfa silage but the concentration of lactic acid was lower in the ordinary alfalfa silage. Feeding behavior, milk yield and composition, ruminal fermentation and blood metabolites were measured. Although dry matter intake and milk production were not affected, the effect of preparation of alfalfa influenced feeding behavior and rumen fermentation parameters. Cows on alfalfa silage diets spent longer ruminating compared to those fed alfalfa hay. The ruminal ammonia nitrogen and blood urea were affected by ensiling (alfalfa hay versus alfalfa silages) while both parameters were lower in cows fed on barley flour mixed alfalfa silage than those fed on ordinary silage. Although similar blood glucose was recorded for cows fed on alfalfa silages, it was higher in cows fed on alfalfa hay. It is concluded that the addition of barely flour when making alfalfa silage may improve both the fermentation process during ensilage and the ruminal ammonia nitrogen utilization with no significant effects on productivity.  相似文献

Whole-body insulin-dependent glucose utilization and insulin responses to glucose in euglycemic-hyperinsulinemic clamps (EHGC) and in hyperglycemic clamps (HGC) were evaluated in high yielding dairy cows fed rumen-protected fat (triglycerides), free fatty acids, or a starch-rich ration (n = 5 per group) in Week 9 and Week 19 of lactation. The experiment was performed under conditions of nonsignificant differences in energy and protein balances in Week 9 and Week 19 and in the three groups. Basal (pre-infusion) concentrations of glucose were lower (P < 0.05) in Week 9 than in Week 19 of lactation and were higher (P < 0.05) in Week 9 in cows fed free fatty acids than in cows fed the starch-rich ration. In EHGC, glucose infusion rates were similar in Week 9 and Week 19 and in the different groups, indicating similar insulin-dependent glucose utilization. Furthermore, because insulin concentrations in EHGC in Week 9 and Week 19 and in the three groups were very similar, metabolic clearance rates of insulin were not affected by stage of lactation and feeding. In addition, insulin responses to the same glucose increments in HGC were not different in Week 9 and Week 19 and in the three groups, indicating that insulin secretion was not affected by stage of lactation and feeding. In conclusion, insulin secretion, insulin metabolic clearance rate, and insulin-dependent glucose utilization between Week 9 and Week 19 of lactation were stable. Furthermore, feeding rumen-protected triglycerides or free fatty acids did not significantly modify insulin secretion, insulin metabolic clearance rate and glucose-dependent glucose utilization compared with starch-rich feeding.  相似文献

Although de novo glucose synthesis was 47 per cent higher in wethers fed hay plus barley than in wethers fed hay alone, the plasma glucose concentrations were similar. When two chronic jugular vein catheters were implanted, plasma glucose concentrations rose by 17 per cent and 5 per cent after 4 h in those fed hay alone and hay plus barley respectively. Levels subsided after 24 to 30 h post catheterisation.  相似文献

The objectives of the present study were (1) to establish and maintain catheters non-surgically in the portal and hepatic veins of cattle for long-term monitoring of postprandially absorbed metabolites, and (2) to assess the safety of the technique used. Aspiration and catheterization were not performed if there was risk of bleeding, as determined by hemostatic data and enlargement of portal and hepatic veins. In five Holstein cattle, before and 60-min after feeding, percutaneous ultrasound-guided aspiration of portal and hepatic blood, together with jugular blood, was performed utilizing 17G x 17.5 cm needles. Concentration of ammonia nitrogen tended to be higher after feeding, but that of urea nitrogen and glucose were not significantly changed. In five other Holstein cows liver veins were catheterized with 14 x 16.5 cm guide-needles attached with 50 cm polyethylene tubes and blood samples were collected from portal, hepatic and jugular veins at 30-min intervals for 4.5 hr. The concentration of plasma acetate and 3-hydroxybutyrate (3-HB) were significantly increased after feeding, while that of lactate was not changed. Catheters in the portal and hepatic veins enabled the collection of blood samples up to 15 days (mean, 7 days). After a 7-day observation period, five cows were euthanised and necropsied. Changes in the intercostal musculature, peritoneum and liver were negligible. During the observation period, clinical profiles, aspartate aminotransferase, alanine aminotransferase and total and differential WBC counts were within normal ranges.  相似文献

Four crossbred(Duroc×Landrace×Yorkshire)boars, weighing an average of 22.40 ± 1.08 kg and fitted with permanent catheters in their portal and mesenteric veins as well as their carotid artery, were utilized in a 4 × 4 latin square design experiment in order to investigate the effects of different starch sources(maize, brown rice, sticky rice, and resistant starch)on the absorption of energy sources through the portal vein.Portal blood flow averaged 30.68,28.40,29.99,and 30.12 mL/min per kg BW for pigs fed the maize, brown rice, sticky rice, and resistant starch diets, respectively, and did not differ(P0.05)between treatments. The absorptions of glucose, plasma ammonia, and total amino acids were significantly lower(P0.05)in pigs fed the resistant starch diet than pigs fed the other starch sources. In contrast,significantly(P0.05)higher amounts of propionate and total volatile fatty acids were absorbed from the portal vein of pigs fed resistant starch than pigs fed diets based on maize, brown rice,or sticky rice.In addition,significantly(P0.05)more lactic acid was absorbed from the portal vein by pigs fed the sticky rice diet than pigs fed the other three diets. The energy absorbed from the portal vein in the form of glucose and amino-N as well as the total energy absorbed was significantly(P0.05)lower in pigs fed the resistant starch diet than pigs fed the other three starch sources. In contrast, the amount of energy absorbed in the form of volatile fatty acids was significantly(P0.05)higher in pigs fed resistance starch than pigs fed the other three starch sources. The total energy absorbed through the portal vein as a percentage of the gross energy and digestible energy consumed in pigs fed maize were 50.8% and 54.6%,respectively,which were significantly(P0.05)higher than the values in pigs fed resistant starch(41.5% and 46.6% respectively). The results indicate that energy is absorbed in different forms and with different utilization efficiencies in the digestive tract of pigs as a result of feeding different starch sources.Starch sources supplying most of their energy in the form of glucose had higher energy utilization efficiencies, while starch sources supplying most of their energy in the form of volatile fatty acids or lactic acid had lower energy utilization efficiencies.  相似文献

We assessed the effects of nutrient supply and dietary bulk, both increasing with hay intake, on O2 uptake and nutrient net fluxes across the portal-(PDV) and mesenteric- (MDV) drained viscera, and the rumen in adult ewes. Four ewes, fitted with a ruminal cannula, with catheters in the mesenteric artery, the portal, mesenteric and right ruminal veins, and with a blood flow probe around the right ruminal artery, were used in a 4 x 4 Latin square design. Treatments consisted of 500 g DM/d hay (LL, low bulk and low nutrient supply), 500 g DM/d hay + infused nutrients (LH, low bulk and high nutrient supply), 750 g DM/d hay + infused nutrients (MH, medium bulk and high nutrient supply), and 1,000 g DM/d hay (HH, high bulk and high nutrient supply). Infused nutrients consisted of volatile fatty acids (VFA) and casein dissolved in salts and infused continuously in the rumen to provide the same amount of metabolizable energy (7.6 MJ/d) and digestible protein (63 g/d) for LH, MH, and HH. Both increases in bulk and nutrient supply increased O2 uptake in the MDV and PDV. Dietary bulk stimulated mainly blood flow, whereas nutrient supply stimulated mainly O2 extraction rate. The O2 uptake by the rumen was not significantly affected by hay intake, although blood flow increased due to nutrient supply. Increase in hay intake had no effects on portal net release of lactate and net uptake of glucose but increased VFA, 3-D-hydroxybutyrate, ammonia, and amino acids (AA) net release and urea net uptake across PDV. The increase in portal nutrient net fluxes with hay intake was entirely related to the increase of nutrient supply for VFA, 3-D-hydroxybutyrate, ammonia, and urea, irrespective of the amount of casein infused for AA. Dietary bulk had no effect on total energy net release in the portal vein. We conclude that despite the increase in portal O2 uptake, increasing dietary bulk had no significant impact on portal recovery of energy. In ruminal tissues, which were the main site of energy absorption, O2 uptake appeared low and was not sensitive to dietary manipulation. In contrast, in mesenteric tissues, which contribute poorly to energy absorption with forage diets, O2 uptake appeared high and very sensitive to dietary manipulation.  相似文献

Twenty-four multiparous Holstein cows were utilized in a completely randomized design to examine the effect of feeding field beans (FB, Vicia faba L. var. minor) as substitute for soybean meal (SBM) on lactation performance and metabolic response during the early lactation period. Cows were individually divided into two equal groups and fed for 16 weeks on one each of the two experimental diets. The controls were fed pelleted concentrate contained 150 g/kg dry matter (DM) of SBM as the main protein source, whereas the experimental concentrate contained 345 g/kg DM of FB. Oat hay was offered ad libitum to cows and water was freely available. Blood samples were assayed for their content of: urea, glucose, triglycerides, cholesterol, total protein, albumin, calcium and phosphorus, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), nonesterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA). Inclusion of FB had no detectable effects on DM intake (DMI), body weight (BW), or body condition score (BCS). Neither milk yield nor quality were influenced by dietary treatment, except for milk urea nitrogen (MU) that was reduced in cows fed the FB diet (P < 0.05). Clotting properties of milk were not affected adversely by added dietary FB. Concentration of blood urea (BU) was lower in cows fed the FB diet than in those fed the control SBM diet (P < 0.05). These findings indicate that feeding FB in a lactation diet supported lactation performance similar to cows fed traditional SBM-based diet, and the results may elicit great interest for countries where soybean utilization is adversely influenced by high supply costs.  相似文献

Eight lactating Holstein cows were divided into two groups ( n  = 4) and used in a double reversal trial with three periods of 14 days each, to evaluate diets containing propylene glycol (PG) and ruminally undegradable protein (RUP) blend on milk yield and composition, ruminal fermentation and blood metabolism. The control diet contained 20% chopped Sudangrass hay, 20% cubed alfalfa hay, 12% corn silage, and 48% of the respective concentrate mixtures (dry matter basis). The experimental diet (PG + RUP) partially replaced the concentrate mixture from the control diet with 1.4% PG and 2.1% RUP. Both diets contained about 16% crude protein and 71% total digestible nutrients. Dry matter intake was similar between the two diets. Daily production of milk, milk lactose and milk solids-not-fat increased by 9.0%, 11.3% and 9.3%, respectively ( P  < 0.1), for cows fed diets with PG + RUP; milk composition was unchanged. Although the concentration of ruminal total volatile fatty acids was unchanged, the proportion of propionic acid increased, and the proportion of acetic acid decreased with PG + RUP. The concentration of ruminal ammonia nitrogen and number of ciliate protozoa was not significantly affected by PG + RUP. The concentration of glucose in blood plasma increased, the concentration of urea nitrogen was unaffected, and the concentration of some essential free amino acids decreased with PG + RUP. It is suggested that these changes might be caused predominantly by PG, and the addition of PG may exert a favorable effect on milk production through increased metabolism.  相似文献

In order to examine the effect of exogenous melatonin on selected biochemical variables of the blood in ruminants, dairy cows were given the pineal gland hormone in the dose of 0.1 mg/kg body weight. One and four hours after melatonin administration blood samples were collected from the cows in the control and the treated group in order to determine the levels of glucose, insulin, total cholesterol, triglycerides, free fatty acids, as well as the activities of alanine and aspartate aminotransferase. The pineal gland hormone caused a significant increase in the levels of total cholesterol and triglycerides, slight increases in glucose and insulin levels, and a significant decrease in the concentration of free fatty acids. Melatonin did not exert an effect on the activity of liver enzymes.  相似文献

The purposes of this study are to assess the responses of increased supplemental dietary fat in the cow, without upsetting rumen fermentation, on the bile acid (BA) extraction rate in the liver and to determine whether this diet would affect the postprandial lipid profiles in the portal and hepatic venous blood. Six Holstein cows were equipped with catheters fitted in the portal and hepatic veins. Two cows each were assigned randomly to a sequence of three dietary treatments of 21-day period. The methodology of this study was based on the supplementation of the basal concentrate diet with 0 (control), 5, or 10% calcium salts of fatty acids (CSFA). The total bile acids were significantly increased in the portal and hepatic veins with the 5% CSFA diet, whereas no increase occurred with the 10% CSFA diet. Data obtained in this study showed that 10% CSFA diet failed to stimulate BA secretion to exceed the values obtained with 5% CSFA-diet. Moreover, there was no change in the hepatic extraction rate of BA in animals fed either the 0 or 5% CSFA diets which ranged from 2.4 to 6.5-fold and 3.1 to 7.3-fold, respectively. However, the extraction rate increased sharply with the 10% CSFA diet (27-fold). The median portal and hepatic concentrations of total lipids, triglycerides, total cholesterol, phospholipids and non-esterified fatty acids did not show any significant increase during feeding of the control diet. Moreover, feeding either the 5 or 10% CSFA diet did not significantly increase these values in either vein.  相似文献

This study was designed to monitor lipid profile in the portal and hepatic blood of cows with fasting-induced hepatic lipidosis, and to compare the results with those in the jugular blood. The work was also carried out to investigate bile acid (BA) in these vessels, and further to investigate BA extraction rate in the liver. Five cows were equipped with catheters in the portal, hepatic and jugular veins (day 0), fasted for 4 days (day 1-day 4) and then refed (day 5-day 11). Before morning feeding, blood was sampled before, during and after fasting from the catheterized vessels. In the portal blood, the concentration of non-esterified fatty acids (NEFA) showed a progressive increase and at day 5 there was an approximate twofold rise. Increased NEFA concentrations were also found similarly in the other two veins. At day 5, beta-hydroxybutyrate (BHBA) in the portal, hepatic and jugular blood rose to 197, 190 and 186% of the pre-fasting value, respectively. However, the concentrations of NEFA and BHBA in the three veins gradually returned to pre-fasting concentration during the refeeding period. Compared with the pre-fasting value at day 0, the content of liver triglyceride (TG) increased significantly at day 5 (P < 0.01). In the liver, the hepatic extraction rate of BA dropped from 3.1 times pre-fasting to 2.2 times during fasting. There were no significant differences in the concentrations of glucose, TG, total cholesterol, cholesterol esters, free cholesterol and phospholipids. The results of the current study show that metabolic alterations occur in the portal, hepatic and jugular veins during induction of hepatic lipidosis in cows, and mostly metabolites, with exception of BA concentration, run parallel. The decreased BA extraction rate in the liver of fasted cows was considered to reflect hepatic cell impairment caused by TG accumulation. Hopefully, the findings, at least in part, contribute to the explanation of the pathophysiology of hepatic lipidosis in dairy cows.  相似文献

OBJECTIVE: To determine the effect of feeding frequency and cow-specific factors on diurnal variations of blood metabolites concentrations. ANIMALS: 40 dairy cows from 2 herds. PROCEDURE: Each herd was fed concentrate at a specific feeding frequency (automatic vs twice daily). Blood samples were collected 4 times daily. Plasma concentrations of glucose, nonesterified fatty acids (NEFA), and beta-hydroxybutyrate (BHBA) were analyzed, and concentrations of urea, cholesterol, and total protein (TP) were determined in serum samples. A multivariate repeated-measures ANOVA was used to test the effect of time, feeding frequency, production group, parity, days after parturition, and daily milk yield on diurnal variations of metabolites. RESULTS: Concentrations of glucose, BHBA, and urea remained relatively constant in the herd that was fed by use of automatic distribution of concentrate; however, significant diurnal patterns were detected in the herd fed only twice daily. Only slight differences in glucose and urea concentrations were detected between high- and low-producing cows, but concentration of BHBA was significantly influenced by number of days in lactation. In contrast, diurnal variations in NEFA and cholesterol concentrations were similar between the 2 herds but differed with regard to production group. Concentrations of TP did not have relevant diurnal patterns and did not differ between production groups. CONCLUSIONS AND CLINICAL RELEVANCE: Diurnal variations and feeding frequency affect glucose, BHBA, and urea concentrations in cows. In contrast, concentrations of NEFA, cholesterol, and TP appear to be less sensitive to time of sample collection. Feeding frequency, composition of feed, or both, have a major impact on blood metabolites concentrations.  相似文献

Normal fecal samples were taken from lactating cows fed either a total mixed ration (TMR; n = 30) or pasture‐based diet (20) and from dry cows fed mainly on hay (15). Diarrheic fecal samples (n = 51) were collected from 21 sick dairy cows. Fecal analyses of ammonia, urea, lactate and volatile fatty acid (VFA) levels were used to evaluate colonic fermentation. Most normal feces had reasonably neutral pH, however, alkaline feces were observed in diarrheic cows. Although fecal lactate is higher in cows on grazing pasture, lactate levels were generally lower in the cows in the present study. Fecal VFA levels were higher in lactating cows than in dry cows. Elevated fecal urea was observed in diarrheic cows, however, many fecal samples in normal and diarrheic cows contained no urea. Fecal VFA levels in diarrheic cows were lower than in normal lactating cows, but were approximately equivalent to those in dry cows. Grazing or dry cows showed higher acetate and lower n‐butyrate proportions compared with TMR‐fed or diarrheic cows. Higher proportions of branched chain VFAs were observed in diarrheic cows, and the lowest level was observed in grazing cows. The present results indicate that intracolonic nitrogen equilibrium and proteolytic fermentation are altered by diarrheic status.  相似文献

OBJECTIVE: To quantify glutamine use by viscera drained by the portal vein in neonatal calves and to determine whether uptake could be stimulated by long-term IV infusion or long-term use of oral supplements. ANIMALS: 4 healthy neonatal calves. PROCEDURE: A femoral artery, jugular vein, and the portal vein were surgically cannulated in each calf. Blood flow in the portal vein was measured, using an ultrasonic transit-time flow probe. Calves were given an IV infusion of glutamine on days 6, 8, and 10 after surgery. Before the first infusion, calves were fed a diet of milk only. The diet was supplemented with glutamine for the second and third infusions. Glutamine was administered via the jugular vein during a 5-hour period. Venous and arterial blood samples were collected every hour for 5 hours. RESULTS: During glutamine infusion, uptake of glutamine by viscera drained by the portal vein increased in association with increased production of ammonia. Glutamine supplementation of the diet did not alter glutamine uptake. Glutamine infusion did not increase viscera uptake of indispensable amino acids. Long-term use of glutamine supplements or infusion of glutamine for periods of more than 1 hour increased glutamine uptake by viscera. Arterial leucine concentration and uptake of leucine by the viscera decreased during glutamine infusion, indicating that leucine became the limiting factor. CONCLUSION: Glutamine administration (supplements or infusions) to calves may require that a mixture of amino acids be provided to improve effectiveness. CLINICAL RELEVANCE: Glutamine may be beneficial in treatments designed to promote intestinal healing in diarrheic calves.  相似文献

A replicated trial was conducted with suckled Angus and Polled Hereford cows (110 d postcalving) to determine metabolic and endocrine responses to an energy-restricted diet after cows had re-established postpartum estrous cyclicity. Cows were individually fed 26.5 Mcal ME (H) or 15.2 Mcal ME (L) for a 30-d preliminary period and fitted with an indwelling jugular cannula at synchronized estrus. Average daily weight change during the estrous cycle was .60 +/- .25 and -1.37 +/- .30 kg/d for H and L, respectively (P less than .05). Blood concentrations of cortisol, progesterone and LH during the estrous cycle were not affected by diet, nor did diet affect frequency or amplitude of LH pulses (P greater than .05). No dietary differences were observed for daily concentrations of total protein, glucose, nonesterified fatty acids or acetate. Mean blood concentrations of propionate and butyrate were not different between diets; however, L cows had lower concentrations of propionate and butyrate on d 11 of the cycle (P less than .05). Cows fed L had higher concentrations of blood urea nitrogen (P less than .05), but they had lower concentrations of cholesterol (P less than .05) on d 4, 11, 18 and subsequent estrus (E). Insulin was not different on d 4 and 11; however, cows fed L had lower insulin concentrations on d 18 and d E (P less than .05). Dietary energy restriction in these cyclic cows caused no change in endocrine responses. Of metabolic responses measured, only blood urea nitrogen, cholesterol and insulin showed consistent changes.  相似文献

The purpose of this study was to elucidate the effects of medium-chain fatty acids (MCFAs) on plasma ghrelin concentration in lactating dairy cows. Five early-lactating Holstein cows were randomly assigned to 2 dietary treatments in a crossover design with 2-wk periods. Treatments consisted of diets supplemented or not (control) with calcium salts of MCFAs (MCFA-Ca; 1.5% dry matter). Plasma hormone and metabolite concentrations in blood samples taken from the jugular vein were measured on the morning of feeding on day 14 of each period. Dry matter intake, milk protein, and lactose content of cows fed the MCFA-Ca diet were decreased compared with controls, but with no change in milk yield. Plasma ghrelin concentrations were higher in cows fed the MCFA-Ca diet; however, no significant effect was found on glucagon-like peptide-1 concentrations in plasma. Plasma insulin concentrations decreased, but plasma glucagon concentrations remained unchanged in cows fed the MCFA-Ca diet. The concentrations of nonesterified FAs, total cholesterol, and β-hydroxybutyrate in plasma increased in these cows. In conclusion, dietary MCFAs increase the plasma ghrelin concentrations in lactating dairy cows.  相似文献
