Hardness of bovine hoof horn was tested as ball indentation hardness and as shore D hardness post mortem in different segments of the hoof wall, in the sole and the hard bulb of sound claws of 10 Austrian Holstein Friesian cows. Both methods of hardness determination showed corresponding results, with shore D hardness between 52.2 and 63.9 hardness units (hu) and ball indentation hardness between 11.2 N/mm2 and 24.3 N/mm2. Bovine hoof horn becomes significantly softer from the coronary band towards the weight bearing border (vertical decrease) and from the dorsal wall towards the heel (horizontal decrease). Decreasing hardness was associated with decreasing dry matter content. Measurements of the claw capsule showed thickness of the hoof wall increasing from the coronary border towards the sole. In dorsopalmar/-plantar direction, bovine hoof wall at the weight bearing border decreases towards the heel.  相似文献

OBJECTIVE: To determine effects of dietary cysteine on blood sulfur amino acids (SAA), reduced glutathione (GSH), oxidized glutathione (GSSG), and malondialdehyde (MDA) concentrations in cats. ANIMALS: 12 healthy adult cats. PROCEDURE: Cats were fed diets with a nominal (0.50 g/100 g dry matter [DM]), moderate (1.00 g/100 g DM), or high (1.50 g/100 g DM) cysteine content in a 3 X 3 Latin square design with blocks of 8 weeks' duration. Venous blood samples were collected after each diet had been fed for 4 and 8 weeks, and a CBC and serum biochemical analyses were performed; poikilocyte, reticulocyte, and Heinz body counts were determined; and MDA, GSH, GSSG, and SAA concentrations were measured. RESULTS: Blood cysteine and MDA concentrations were not significantly affected by dietary cysteine content. Blood methionine, homocysteine, and GSSG concentrations were significantly increased when cats consumed the high cysteine content diet but not when they consumed the moderate cysteine content diet, compared with concentrations obtained when cats consumed the nominal cysteine content diet. Blood GSH concentrations were significantly increased when cats consumed the moderate or high cysteine content diet. CONCLUSIONS: Increased dietary cysteine content promotes higher blood methionine, homocysteine, GSH, and GSSG concentrations in healthy cats. CLINICAL RELEVANCE: Supplemental dietary cysteine may be indicated to promote glutathione synthesis and ameliorate adverse effects of oxidative damage induced by disease or drugs.  相似文献

  1. An experiment with a 2 × 3 factorial design with two concentrations of dietary betaine (0 and 600 mg/kg) and three dietary concentrations of methionine (0, 600 and 1200 mg/kg) was conducted using goslings to estimate growth, nutrient utilisation and digestibility of amino acids from 21 to 70 d of age.

  2. Three hundred geese were randomised at 18 d of age into 6 groups with 5 replicates per treatment and 10 geese per replicate.

  3. Increasing dietary concentrations of methionine gave a linear increase in body weight and average daily gain. The coefficient of crude fat retention increased as dietary methionine increased and there was a significant non-linear response to increasing dietary methionine. Similarly, increasing supplemental methionine gave linear increases in the digestibility of methionine and cysteine.

  4. The results of this study indicated that optimal dietary supplementation of methionine could increase growth performance and methionine and cysteine utilisation in growing goslings. Betaine supplementation had no apparent sparing effect on methionine needs for growth performance, but did improve the apparent cysteine digestibility.

1. Two experiments were conducted with male broiler chicks from 2 to 5 weeks of age to determine the effect of dietary protein content and amino acid balance on the response to dietary sulphur‐containing amino acids (SAA) in terms of performance and carcase quality.

2. In experiment 1, 5 graded amounts of a DL‐methionine and L‐cysteine (1:1 by weight) mixture were added to basal diets containing 197 or 233 g crude protein/kg. The diets containing 197 g protein/kg were fed with or without the further addition of 36 g crude protein/kg from nonessential amino acids. The amino acid balance of all diets was kept constant for all essential amino acids except the SAA. In experiment 2, 5 graded amounts of SAA from either a crystalline source (DL‐methionine or a mixture of DL‐methionine and L‐cysteine) or from intact proteins were added to a diet containing 208 g protein/kg.

3. At each protein concentration there were significant responses to the SAA addition in weight gain, food conversion efficiency, and carcase quality. Non‐linear exponential regression analyses were used to describe bird responses to SAA concentration. The broiler chick's requirement for SAA increased with increasing dietary protein concentrations ranging from 197 to 259 g protein/kg.

4. The utilisation of SAA differed also with differences in origin (crystalline or peptide‐bound), and methionine:cysteine balances. Compared to DL‐methionine, a 1:1 mixture of DL‐methionine and L‐cysteine was only 81% or 86% as effective in supporting growth or food conversion, respectively. SAA from added protein was even less effectively utilised.

5. The addition of nonessential amino acids tended to decrease food intake without affecting SAA utilisation.

6. Slaughter yield and breast meat yield were clearly increased while fat deposition was clearly decreased, by SAA addition. The response in breast meat yield suggested an important economic benefit for further meat processing. Nitrogen retention was significantly enhanced by SAA supplementation from crystalline sources, and this led to reductions of up to 30% in the amount of nitrogen excreted per kg weight gain.  相似文献

This study investigated the effects of housing primiparous lactating Holstein heifers either in cubicles with butyl rubber mats, cubicles with thicker mattresses filled with chopped rubber, or straw yards, and supplementing them with methionine for the first 13 weeks of lactation on the rates of growth of their hoof horn and the conformation of their feet. Overall, the rate of growth of the hoof horn increased significantly after calving but the rate of wear did not increase. In contrast, the type of housing had no significant effect on the growth of the hoof horn, but the heifers in straw yards had significantly lower rates of lateral horn wear. The changes in growth and wear rates resulted in changes in hoof conformation, including the toe angle and the width of the bearing surface, which suggested that heel bulb hypertrophy occurred after the heifers had been moved from soft underfoot conditions on to concrete. Providing 115 per cent of their estimated methionine requirements had no significant effect on the rates of growth and wear of the hoof horn.  相似文献

In a cross-sectional study of 6240 Danish Holstein cows in 55 herds using loose-housing systems, sole haemorrhages and heel horn erosions occurred frequently in almost all the herds. Digital dermatitis occurred in 47 of the 55 herds (85 per cent). Spearman correlation coefficients revealed relationships between lameness and sole ulcer (0.36), between heel horn erosion and sole haemorrhage (0.39), between heel horn erosion and interdigital dermatitis (0.29) and between sole ulcer and double sole (0.26). The prevalence of heel horn erosion, sole haemorrhage, interdigital dermatitis and digital dermatitis appeared to be most affected by herd-level factors. The associations between individual foot trimmers and all the foot lesions were statistically significant. The risk of interdigital dermatitis increased with decreasing amounts of bedding. Cows housed throughout the year had a marginally higher risk of sole haemorrhage.  相似文献

Cysteine is a semi-indispensable AA in neonates and is synthesized from the indispensable AA, methionine, by transsulfuration. We previously showed that the gastrointestinal tract (GIT) is a metabolically important site of methionine transsulfuration to cysteine, yet the metabolic fate of dietary cysteine in the GIT has not been established. Cysteine use by gut epithelial cells may play an important role for maintenance of glutathione synthesis and cellular redox function. Our aim was to quantify the extent of gastrointestinal first-pass cysteine metabolism in young pigs. Four-week-old weanling pigs (n = 10) were fed a liquid milk-replacer diet and given an intragastric and intravenous [1-(13)C]cysteine infusion on 2 separate days in a crossover design. Arterial and portal blood samples were collected for cysteine isotopic enrichment by gas chromatography-mass spectrometry and for (13)CO(2) enrichment by isotope ratio mass spectrometry. Our results indicated that dietary cysteine is metabolized during its first-pass splanchnic metabolism, accounting for about 40% of dietary cysteine intake. We also showed that intestinal absorption was the major metabolic fate of dietary cysteine, representing about 75% of intake, indicating that the GIT utilizes 25% of the dietary cysteine intake. Thus, utilization by the GIT represents about one-half (approximately 53%) of the first-pass, splanchnic uptake of dietary cysteine. Moreover, a substantial proportion of dietary splanchnic cysteine metabolism was consumed by the GIT via nonoxidative pathways. We conclude that the gut utilizes 25% of the dietary cysteine intake and that synthesis of mucosal epithelial proteins, such as glutathione and mucin, are a major nonoxidative metabolic fate for cysteine.  相似文献

Supplementary biotin for sows: effect on claw integrity   总被引:2,自引:0,他引:2  
Eighty female pigs were fed, from 25 kg liveweight, either basal diets calculated to provide 32 micrograms available biotin/kg (control diet) or basal diets supplemented with 350 micrograms biotin/kg. Their claws were examined and lesions recorded at 170 days of age and when each of their first four litters was weaned. The incidence of horn 'defects' (superficial bruises, abrasions and cuts in the soft heel) remained at a low level throughout the trial. Neither the number of claws affected with lesions nor the number of lesions per sow differed between treatments at 170 days of age. Between 170 days of age and first weaning the incidence of hoof lesions increased greatly. At first weaning and for the remainder of the trial biotin supplemented sows had significantly fewer claw lesions per sow than controls (P less than 0.05 or greater). The predominant injuries to the foot were cracks which occurred mainly in two associated regions, the heel/toe junction and the heel, and the sidewall and adjacent white-line region of the toe. The differences in foot damage did not result in differences in culling rate. It was concluded that supplemention of the diet of breeding sows with biotin from an early stage of development made a significant contribution to the maintenance of their horn integrity.  相似文献

1. Herbs, thyme essential oil (EO) and condensed tannin (CT) extracts were compared for their effects, as dietary supplements, on broiler growth performance, nutrient digestibility and volatile fatty acid (VFA) profiles in the gut. Cooked meat from the birds fed on diets with 4 herbs and an EO extract was compared by a taste panel against those fed on the control treatment, for organoleptic properties in the meat. 2. Female broiler chicks were fed on wheat-soybean meal diets from 0-42 d of age. These chicks were given either the basal diet (control), or the basal diet with one of rosemary, garlic or yarrow herbs, mimosa, cranberry or grapeseed CT's, or thyme EO supplements (8 treatments in total). Body weight (BW) and feed consumption (FC) were measured. 3. The garlic supplement tended to improve growth rate over the first 7 d, while mimosa CT and thyme EO supplements reduced weight gains. The mimosa supplement in diets significantly reduced FC to d 21. Meanwhile, the addition of a cranberry supplement reduced the digestibility of DM, OM and N, compared with the controls. Dietary thyme EO, yarrow, rosemary and garlic supplements modified caecal isovaleric and isobutyric acid proportions (presented as 'Other VFA'; p < 0.05). Dietary herb significantly affected the intensity of meat flavour, and the potential of observing both garlic and abnormal flavours. There were large differences between the consumption of red and white meat samples, while meat temperature affected several flavour attributes. 4. Broiler performance and digestibility for birds given dietary garlic and grapeseed CT supplements were similar to the controls, and these supplements appear suitable for dietary inclusion. Careful choices are necessary when selecting dietary plant extract supplements for broilers, but beneficial effects can be observed.  相似文献

The mural suspensory apparatus of third phalanx and its supportive heel cushion were examined in 19 cows with an ulcer at the 'typical' site (Rusterholz ulcer) to gain information on the pathogenesis of sole and heel ulcers. The claws of 17 healthy controls were used for comparison. The left hind claws, frozen at -20 degrees C, were sectioned in one longitudinal and four transverse planes with a band saw. The thickness of the subcutaneous tissue, the corium and the extent of displacement of the third phalanx were measured at defined sites on these sections. In addition, the suspensory apparatus, the tissue layer connecting the third phalanx to the dorsal wall of the horn capsule, was examined histologically. There was a direct relationship between the displacement of the third phalanx and ulceration of the sole or heel; in all the ulcerated claws the third phalanx had dropped and the corium and the subcutis under the bone were thinner than in the controls. The supportive cushions of the cows with ulcers contained less fat tissue. There was no histological evidence of damage to the epidermis or the corioepidermal junction in the ulcerated claws nor were the lamellae elongated. Similarly, there were no morphological changes in the connective tissue layer, the submural dermis.  相似文献

This study examined the molecular mechanisms of methionine pathways in meat‐type chickens where birds were provided with a diet deficient in methionine from 3 to 5 weeks of age. The birds on the deficient diet were then provided with a diet supplemented with either D,L‐methionine or D,L‐HMTBA from 5 to 7 weeks. The diet of the control birds was supplemented with L‐methionine from hatch till 7 weeks of age. We studied the mRNA expression of methionine adenosyltransferase 1, alpha, methionine adenosyltransferase 1, beta, 5‐methyltetrahydrofolate‐homocysteine methyltransferase, 5‐methyltetrahydrofolate‐homocysteine methyltransferase reductase, betaine‐homocysteine S‐methyltransferase, glycine N‐methyltransferase, S‐adenosyl‐L‐homocysteine hydrolase and cystathionine beta synthase genes in the liver, duodenum, Pectoralis (P.) major and the gastrocnemius muscle at 5 and 7 weeks. Feeding a diet deficient in dietary methionine affected body composition. Birds that were fed a methionine‐deficient diet expressed genes that indicated that remethylation occurred via the one‐carbon pathway in the liver and duodenum; however, in the P. major and the gastrocnemius muscles, gene expression levels suggested that homocysteine received methyl from both folate and betaine for remethylation. Birds who were switched from a methionine deficiency diet to one supplemented with either D,L‐methionine or D,L‐HMTBA showed a downregulation of all the genes studied in the liver. However, depending on the tissue or methionine form, either folate or betaine was elicited for remethylation. Thus, mRNA expressions show that genes in the remethylation and transsulphuration pathways were regulated according to tissue need, and there were some differences in the methionine form.  相似文献

The associations between individual foot lesions and different types of floor were investigated in 4038 finishing pigs on 21 units. The overall prevalence of foot lesions was 93.8 per cent and the prevalence ranged from 79.4 per cent to 100 per cent on different units. Analysis showed that pigs kept on bedded floors, with either sparse or deep straw, had a lower prevalence of sole erosions, heel erosions and heel flaps and a higher prevalence of white line lesions, false sand cracks, wall separations and toe erosions than pigs kept on bare solid concrete floors. Partially slatted floors were associated with an increased prevalence of heel erosions, heel flaps, white line lesions, wall separations and false sand cracks, and totally slatted floors were associated with an increased prevalence of sole erosions and heel flaps.  相似文献

Measurements of both front feet of slaughtered German Simmental cattle (17 heifers, 13 cows) were carried out to document the normal shape and size of the forelimb claws and to gain information about function and possible predisposition to disease. Only clinically normal feet were examined, and a standardised sole thickness of 5mm at the tip and 8mm in the heel region in both claws was established as a reference. The dorsal wall length, dorsal wall angle, heel length, height and width, sole and claw length and claw width, sole circumference and sole area were measured. After removal of the horn capsules, measurements of values at the level of the corium were carried out. The lateral front claws were wider and the medial claws longer than their partner claws. Overall, the sole surface areas and circumferences of the paired claws were similar, but were statistically larger in the medial claws. The front claws were characterised by long and high heel bulbs, and had a toe length:heel bulb length ratio of approximately 1.6:1. These properties, together with the musculo-tendinous attachment of the limbs to the trunk, are believed to provide better protection for the forelimb claws when standing or walking on hard surfaces. Functional mechanical relief of the medial front claws is probably best achieved by pairing the soles of the foot to the same level, which usually entails reduction of the lateral claw. For German Simmental cattle, a dorsal wall length of 75mm can be used as a guideline when trimming front and hind feet.  相似文献

Previous experiments have shown that increasing the dietary crude protein (CP) of cats does not increase urea cycle enzymes or alanine amino transferase as occurs in rats. Also when an essential amino acid (EAA) is limiting in a diet for growing kittens, the kittens do not exhibit an amino acid imbalance when other EAAs are added to the diet. To study the metabolic basis for these observations which are different from that found in omnivores and herbivores, the hypothesis that increased dietary CP decreases methionine catabolism, so more is spared for growth, was tested. Fifteen male kittens were randomly assigned to one of three dietary treatments. Each diet contained 2.5 g l-methionine/kg diet and 200, 300 or 500 g CP/kg diet. The livers and kidneys were removed and assayed for methionine transaminase (MTA), cystathionase (CASE) and cystathionine synthase (CS). Free amino acid concentrations were determined in liver, kidney and plasma. The 300 and 500 g CP/kg groups had significantly greater kidney weights and body weight gains than the 200 g CP/kg group. Hepatic MTA activity was lower in the 300 than the 200 or 500 g CP/kg groups (p < 0.05). Renal MTA and CASE activities were 35% and 50% greater, respectively, for the 500 g CP/kg group than for the 200 g CP/kg diet group (p < 0.05). Renal CS activities for the 300 and 500 g CP/kg groups were 29% (p > 0.05) and 38% (p < 0.05) greater, respectively, than the 200 g CP/kg group. Cyst(e)ine concentrations were lower in the livers of the 500 g CP/kg group than the 200 g CP/kg group (p < 0.05). Cystathionine was lower in plasma and kidney from the 500 g CP/kg diet group than from the 200 g CP/kg diet group (p < 0.05). It was concluded that the metabolic basis for the increased growth of kittens fed diets marginally limiting in methionine, with increasing concentrations of dietary CP, was not mediated through decreased enzyme activity associated with the catabolism of methionine, but was the result of an increase in food (methionine) intake.  相似文献

New techniques involving extraction with organic solvents and lithium carbonate solution have been used to fractionate nitrogenous components of the excreta from laying hens fed diets in which the supplementary protein was groundnut meal, poor‐quality fish meal or good‐quality fish meal. The effect of adding methionine or lysine or both to the groundnut diet was also studied.

The proportion of ingested nitrogen excreted as uric acid or urate was significantly higher when the dietary protein supplement was groundnut meal than when it was good‐quality fish meal or groundnut meal supplemented with both lysine and methionine. Nitrogen retention, egg production and the apparent digestibility of nitrogen were higher on diets containing good‐quality fish meal than on diets containing groundnut meal alone. Intermediate effects were obtained when feeding diets supplemented with poor‐quality fish meal of groundnut meal containing either lysine or methionine. The implications of these findings are discussed. The main differences in protein utilisation associated with variations in dietary protein quality were reflected in the proportions of ingested nitrogen excreted as urates and as protein. Other nitrogenous components of excreta examined showed little variation with dietary protein quality. Benefits derived from the addition of methionine and lysine to the groundnut diet were obtained mainly by a more efficient use of absorbed amino acids.  相似文献

The aim of this study was to describe the microstructure of hoof capsules of the buffalo. In addition, the study emphasized the morphometric aspects of the horn tubules, the Vickers nanohardness of the dorsal and abaxial walls and sole of the digits of the thoracic and pelvic limbs of the buffalo. The abaxial wall in the thoracic and pelvic digits showed larger diameter of the horn tubules when compared to all dorsal wall and sole. In addition, the abaxial wall of the thoracic digits showed larger diameter of the horn tubules when compared with the pelvic digits. According to the three‐dimensional microtomography, the dorsal wall was higher in density compared with the abaxial wall. The latter exhibited an intermediate density, while the sole showed the lowest density. The Vickers nanohardness test showed that there was no difference in hardness and resistance between the experienced regions. However, the elastic modulus was greater on the transversal section of the hoof capsule. In conclusion, the results of the current study show that modern technologies such as microtomography and subsequent imaging can be used to investigate details of the basic morphology in different regions of the buffalo's hoof.  相似文献

Effects of dietary biotin supplementation on serum biotin levels and physical properties of sole horn of 40 Holstein cows were evaluated. The mean serum biotin level in biotin-supplemented cows after 10 mo of biotin supplementation (1163.2 +/- 76.2 pg/mL) was significantly higher (P = 0.007) than that in control cows (382.0 +/- 76.2 pg/mL). The sole horn of biotin-supplemented cows was significantly harder (P = 0.026) and had a significantly lower moisture content (P = 0.021) than that of control cows. No morphologic differences in horn tubules or intertubular horn were found between the biotin-supplemented and control cows. The total lipid content of sole horn was significantly higher (P = 0.030) in the biotin-supplemented cows than in the control cows. These results suggest that dietary biotin supplementation causes increases in serum biotin levels and changes in physical properties and fat content of sole horn.  相似文献

Four crossbred barrows, weighing an average of 26 kg each, were fitted with simple T-cannulas in the terminal ileum and placed in metabolism cages to evaluate the effect of dietary supplements of NaHCO3 or KHCO3 on the short-term metabolism of sodium (Na), potassium (K), calcium (Ca), magnesium (Mg) and chloride (Cl). A control diet containing 2.8 g/kg Na and 4.1 g/kg K was compared with similar diets supplemented with either 13 g/kg or 26 g/kg NaHCO3 or 30 g/kg KHCO3. All diets contained 4 g/kg Cr2O3 as an external marker and were offered twice daily (1,100 g X pig-1 X d-1) in a 4 X 4 Latin square arrangement. Feces and total urine output were collected for 24 h on the fifth day after introducing a new diet; digesta was collected for 12-h periods on d 6 and 7. Sodium and K concentrations at the terminal ileum were unaffected by dietary treatment. Apparent ileal digestibility of Na was increased by NaHCO3 supplements. Over the total gastrointestinal tract, diet had no affect on apparent Na digestibility. Urinary Na clearance was increased by NaHCO3 in the diet in a dose-dependent manner. Net Na retention (g/d) was increased by NaHCO3. Apparent ileal digestibility of K was increased by KHCO3. Apparent fecal digestibility of K was increased by KHCO3 and NaHCO3. Urinary K clearance was elevated by KHCO3, but not enough to overcome the increased K intake; net K balance (g/d) rose in response to dietary KHCO3 supplements. The mechanical properties of horn samples from 22 hind claws with chronic laminitis were determined in adult Austrian Fleckvieh cows. The resistance to deformation was quantified as the modulus of elasticity (E). Tension tests revealed mean E values of 520MPa for the dorsal wall, 243MPa for the abaxial wall, 339MPa for the axial wall and 97MPa for the sole. E tended to be lower in laminitic horn than in sound horn in all segments tested, with the difference being largest in the abaxial wall. The mean dry-matter content (DMC) of the laminitic claws was 75.8% in the dorsal wall, 75.86% in the abaxial wall, 71.15% in the axial wall and 69.28% in the sole. These values are generally comparable to those for sound claws except in the axial wall. Further, E and DMC were only correlated in the axial wall. Chronic laminitis leads to a low resistance of claw horn to mechanical insults in the dorsal wall, abaxial wall and sole, and to the loss of a correlation between the E and DMC in these segments. The reason for these alterations is therefore not increased ingress of moisture, but must be due to changes in the microstructure, biochemical components and/or horn formation by the diseased dermis.  相似文献

The influence of supplementing 0 (NB) or 440 (SB) micrograms biotin/kg to corn- or wheat-based diets on toe lesions, hair characteristics and structural soundness in 116 crossbred female swine was studied from selection (100 kg) until completion of four parities. Gilts that had been previously fed corn-soybean meal diets with 0 or 220 micrograms supplemental biotin/kg diet during growth and development, remained on either the biotin unsupplemented or supplemented diet. Females were housed in buildings containing partially slatted and solid concrete floors. Toe, hair and soundness evaluations were made at a mean age of 240, 521, 732, 916 and 1,090 d. Type of grain fed did not influence (P greater than .10) any response criteria evaluated. Six types of toe lesions were observed across all dietary treatments and varied in severity from minor to very severe. The percentage of females with heel cracks, heel-horn junction cracks and side-wall horn cracks was reduced (P less than .01) when females were fed SB diets. Females fed SB diets had fewer (P less than .001) total lesions, heel cracks, heel-horn junction cracks, side-wall horn cracks and white-line horn cracks (P less than .03) compared with females consuming NB diets. In general, biotin supplementation was more effective in reducing the number and percentage of toe lesions in multiparous sows compared with gilts and primiparous sows. Biotin supplementation increased (P less than .001) the number of hairs/cm2 skin and improved (P less than .001) hair scores.
